1. Male chicks were deprived of feed for 48 hr to study the effect of metabolic stress on hepatic membrane and lipoprotein fluidity and binding of radioiodinated lipoproteins to hepatic membranes. 2. Plasma levels of low density lipoprotein (LDL) and high density lipoprotein (HDL) were markedly and slightly elevated, respectively. 3. There was a reduction in lipoprotein and hepatic membrane fluidity. 4. Binding of [125I]LDL, but not [125I]HDL, to hepatic membranes was decreased. 5. It is suggested that a reduction in the fluidity of LDL and/or hepatic membranes impedes LDL catabolism in vivo.  相似文献

1. Plasma high density lipoprotein (HDL), the principal carotenoid carrier in broiler chicks, was analyzed at zero time and 3, 5 and 7 days after inoculation of chicks with the intestinal coccidial parasite, Eimeria acervulina. 2. Infection caused significant decreases in total HDL mass from 3 through 7 post-inoculation (PI), and decreases in relative percents of protein and carotenoids, and relative increases in phospholipid and cholesterol esters by 7 days PI. 3. Carotenoid content correlated best with protein content (principally apolipoprotein A-I) throughout the experiment. 4. Findings emphasize the importance of the small intestinal mucosa as a source of plasma HDL in the broiler chick, and suggest that apolipoprotein A-I is the binding site for HDL carotenoids.  相似文献

1. Thermal Pulse Decay (TPD) methodology was used to monitor hepatic tissue blood flow (hepatic perfusion) in anesthetized birds prior to and during hemorrhagic hypotension. 2. Hemorrhage was accomplished by periodic removal of blood through a carotid cannula. Reducing the estimated blood volume (BV) from 100 to less than 50% decreased hepatic perfusion from 4.36 +/- 0.7 to 1.88 +/- 0.7 ml/min/g. 3. Changes in hepatic perfusion during the experiment were related to mean arterial blood pressure (MABP) by the following linear regression equation: hepatic perfusion = -1.79 +/- 0.0807x (r2 = 0.57).  相似文献

Explants of pig small intestine were maintained at 37 degrees C in organ culture for periods up to 24 h in a system using Trowell T-8 medium supplemented with 10% foetal-calf serum. The mucosal morphology was well preserved during culture, as judged by light and electron microscopy. The explant contents of protein and two brush-border enzymes, microvillus aminopeptidase (EC 3.4.11.2) and dipeptidyl peptidase IV (EC 3.4.14.5), were not significantly modified during culture compared with controls, but a moderate, continuous release of both protein and enzyme activities into the medium was observed. Continuous labelling with [35S]methionine resulted in an even incorporation of radioactivity in the protein components, and the rate of labelling only moderately decreased over the 24 h period. The polypeptide compositions of sucrase (EC 3.2.1.48)--isomaltase (EC 3.2.1.10), maltase--glucoamylase (EC 3.2.1.20) lactase (EC 3.2.1.23)--phlorizin hydrolase (EC 3.2.1.62), microvillus aminopeptidase and aspartate aminopeptidase (EC 3.4.11.7) synthesized during culture were studied, and some were found to be similar to those of the pro-forms of the enzymes isolated from animals that had had their pancreatic duct disconnected 3 days before being killed. These results confirmed earlier findings of the existence of pro-forms of some of the microvillar enzymes and thus indicate a low activity of pancreatic proteinases in the culture system.  相似文献

1. The effect of dietary protein and fat content on tri-iodothyronine (T3) induced thermogenesis was studied in young chicks (Gallus domesticus). 2. Irrespective of dietary protein and fat contents employed, energy and fat retained were consistently decreased and heat production was increased, but protein retained was not affected by the T3 administration throughout the experiments. 3. Little or no effect of dietary protein and fat contents on T3-induced thermogenesis was observed. 4. Increased heat production by the T3 administration was primarily brought about by a reduction in retained fat which served as fuel to give rise to extraneous heat.  相似文献

1. Lipoprotein lipase activity of the abdominal fat pad and hepatic triacylglyceride synthetase activity were determined at 2, 7.5, 9, 12.5 and 20 weeks of age. 2. Specific activity of lipoprotein lipase in fat pad was constant at 2-9 weeks, decreasing until 20 weeks. Hepatic triacylglyceride synthetase activity decreased from 2 to 12.5 weeks, increasing by 20 weeks. 3. Lipoprotein lipase activity was higher throughout 20 weeks and triacylglyceride synthetase activity was higher at 7.5 weeks with energy restricted diet. 4. An inverse relationship existed between fat pad weight and specific activity of lipoprotein lipase; higher activity in adipose tissue of energy restricted pullets may have been a function of smaller fat pad size.  相似文献

The effects of a 24-h fast on serum lipids and lipoprotein profiles in commercial laying hens were investigated. Blood was analyzed at 34 and 46 weeks of age from Single Comb White Leghorn hens that had been either fed ad libitum or had been fasted for 24 h prior to collection. At 12 weeks, birds were divided into 16 biological isolation units, with 8 replicate units assigned to each treatment group. Four birds out of 10 in each unit were tagged for bleeding. Parameters evaluated included total serum cholesterol and triglycerides, mean diameters of very low density lipoproteins (VLDLs) for the 10th, 50th, and 90th percentiles of serum total VLDL, mean total population VLDL particle diameter (MPD), and percentage serum cholesterol recovered in VLDL, low density lipoprotein (LDL), and high density lipoprotein (HDL) fractions. Fasting led to decreases in total serum cholesterol and triglycerides, and a decrease in mean serum VLDL particle diameter in the 90th population percentile. At Week 34, percentage serum cholesterol recovered from LDL was increased, whereas percentage serum cholesterol recovered from HDL was decreased due to fasting. At Week 46, MPD and percentage serum cholesterol recovered from VLDL were decreased, whereas percentage serum cholesterol recovered from HDL was increased due to fasting. It was concluded that a 24-h fast decreased serum lipids (cholesterol and triglycerides) and the size of VLDL particles in the 90th population percentile in commercial laying hens. Furthermore, bird age influenced the effects of a 24-h fast on MPD and the redistribution of serum cholesterol among VLDL, LDL, and HDL particles.  相似文献

The effect of dietary amino acids and protein on cholecystokinin (CCK) release into plasma was investigated in chicks by feeding a meal through a stomach tube, followed by the CCK determination with specific CCK-8 antibody. 2. The results showed that both isolated soya protein and an amino acid mixture simulating the amino acid composition of the soya protein increased the release of CCK, though to a lesser extent with a delayed response in the former, when added to a protein-free diet. 3. Among amino acids added singly to the protein-free diet, phenylalanine was more efficient than arginine and valine, exerting a response almost identical to the complete amino acid mixture.  相似文献

The effects of dietary cholesterol and fatty acids on the plasma cholesterol level and rates of very low density lipoprotein (VLDL) cholesterol secretion and low density lipoprotein (LDL) transport through LDL receptors in the liver of the hamster were investigated. Increases of plasma VLDL- and LDL-cholesterol levels and VLDL-cholesterol secretion from hepatocytes were observed in animals fed a diet enriched with 0.1% cholesterol for 2 weeks in comparison with animals fed a control diet. The addition of dietary palmitic acid accelerated the effect of dietary cholesterol on plasma VLDL- and LDL-cholesterol levels and VLDL-cholesterol secretion from hepatocytes. Dietary linoleic acid accelerated the effect of dietary cholesterol on VLDL-cholesterol secretion from hepatocytes and diminished the effect on the plasma LDL-cholesterol level. Hepatic LDL receptor activity was considerably suppressed by a control diet containing 0.05% cholesterol and a further small suppression was induced by a diet enriched with 0.1% cholesterol with or without 5% palmitic acid. However, dietary linoleic acid diminished the effect of dietary cholesterol on the suppression of hepatic LDL receptor activity. These results suggest that dietary palmitic acid augments the effect of dietary cholesterol in elevating the plasma LDL-cholesterol level through acceleration of VLDL-cholesterol secretion from the liver, and that dietary linoleic acid diminishes the effect of dietary cholesterol in elevating the plasma LDL-cholesterol level by preventing the suppression of hepatic LDL receptor activity induced by cholesterol.  相似文献

1. Twenty-eight day old broiler chicks gavage fed for 22 days consumed 40% more feed and gained 100-110 g more than control birds. 2. Normal feeding resumed on day 41, by day 63 feed intakes and body weights of overfed and control birds were equal. 3. Moderate differences in lipid compositions of LDL and VLDL were observed at 41 and 50 but not at 63 days. 4. The oral aversion behaviour of the chicken (head shaking, beakwiping and tongue/beak movements) was measured following oralstimulation with 0.1 M quinine hydrochloride, 40% sucrose, 3M sodium chloride, 5 M acetic acid and pure methyl anthranilate.Section of the lingual and laryngolingual nerves did not affectthe oral aversion behaviour and therefore demonstrates the presenceof functional extra-lingual chemoreceptors. This study was undertaken to investigate whether the gelatinous membranes (GMs) of the chick (Gallus domesticus) inner ear contain glycoproteins and proteoglycans, as is the case in mammals, and whether or not the relative concentrations of glycoproteins and proteoglycans change during development. We used electron-microscopic histochemistry with tannic acid, Alcian blue and ruthenium red and immunoperoxidase with monoclonal antibodies to fibronectin and keratan sulfate. Both techniques were applied to material from newly hatched chicks in various developmental stages. In hatchlings, tannic acid, which precipitates mainly proteoglycans, stained the mineralizing GMs of the utricle, saccule and lagenar macula darker than it did the nonmineralizing GMs of the cristae and tectorial membrane of the basilar papilla, while Alcian blue and ruthenium red stained all GMs in the vestibule and basilar papilla almost to the same degree. Antikeratan sulfate (a proteoglycan marker) stained the GMs intensively, while antifibronectin (glycoprotein monoclonal marker) stained them less intensively. In the mineralizing GMs of the embryos, the immunoreactivity (density) of keratan sulfate almost doubles between days 7 (stages 31) and 21 (stage 46), while the density of fibronectin does not seem to change as much. Different concentrations of glycoproteins and proteoglycans in each GM may impart to it the capacity to remain unmineralized, as is the case for cupulae of the vestibule and tectorial membrane of the basilar papilla, or mineralized, as is the case for the statoconial membrane of the maculae.  相似文献

This trial examined effects of lutein supplied from maternal (i.e., in ovo) and dietary routes on cutaneous basophil hypersensitivity responses in chickens. Chicks hatched from one of two in ovo carotenoid levels (n=100/level; carotenoid-replete or carotenoid-deplete eggs) were fed one of two diet lutein levels (0 or 40 mg diet lutein/kg diet). At 14 d post-hatch, phytohemagglutinin (PHA) was injected into the wing web, and thickness and leukocyte populations were measured at 0 to 48 h. PHA increased wing web thickness at 4 to 48 h post-PHA (p<0.05), and the swelling response was most related to macrophage numbers in the wing web. Swelling occurred more rapidly in chicks from carotenoid-deplete eggs (p<0.05), but eventually reached a greater thickness in chicks from carotenoid-replete eggs (p<0.05). Differences in leukocyte infiltration occurred due to diet and in ovo carotenoid exposure, and indicate that pre- and post-hatch carotenoid exposure had additive or synergistic effects on the PHA-induced wing web response. Evaluation of the cellular contents of the injection site is a much better indicator of the immunomodulatory effects of lutein than measurements of the amount of swelling.  相似文献
