A study is made of the excitation of waves in the whistler frequency range by a given ring electric current in a collisional magnetoplasma in the presence of a cylindrical duct with enhanced density. It is shown that, under certain conditions, dissipative losses due to electron collisions in plasma can lead to a substantial redistribution of the source radiation power over the spatial spectrum of the modes guided by the duct, as compared to the case of a collisionless plasma. Numerical results are presented that illustrate these changes in the excitation efficiency of whistler modes.  相似文献

Propagation of whistler-mode waves in magnetized plasma in the presence of small-scale field-aligned irregularities with enhanced or depressed plasma density is simulated numerically. The numerical experiments have demonstrated the effect of guided propagation of whistler-mode waves in plasma regions occupied by irregularities with transverse dimensions smaller than the whistler wavelength in uniform plasma. It is shown that not only individual irregularities but also the entire modified region, which serves as a specific guiding structure, exhibit waveguide properties.  相似文献

The structure of the RF magnetic field in the vicinity of a loop antenna operating in the whistler frequency range has been studied experimentally and theoretically. The experiments were performed over a wide frequency range at different values of the plasma density, electron temperature, and ambient magnetic field strength. It is shown that, when a loop antenna is smaller than the wavelength of a quasi-longitudinal whistler, the structure of the magnetic field of such an antenna is nearly the same as that of the field of a current-carrying loop in vacuum; otherwise, the RF field is localized near the antenna wire. The results of numerical calculations agree with the measured field distributions. The antenna field is calculated by expanding it in the eigenmodes of a magnetized plasma with allowance for not only propagating but also nonpropagating (exponentially decaying) waves, which make the main contribution to the near field. An analytic estimate of the depth to which the RF magnetic field of a loop antenna penetrates into the plasma is obtained.  相似文献

The minimal sufficient set of plasma parameters is presented to describe the dispersion properties of electron whistler waves (helicons) in a wide frequency range above the ion cutoff frequency, provided that the wave frequency is significantly lower than the electron plasma frequency. When the gyrofrequency of the lightest ions is much higher than those of heavier ions, it is sufficient to know the relative content of the lightest ions, the highest ion cutoff frequency, the lower hybrid resonance frequency, and the electron gyro- and plasma frequencies. In this case, the frequency of electron whistler waves is determined by the upper root of the biquadratic equation derived, whereas the lower root corresponds to a resonant mode with its refractive index increasing when the frequency tends toward the highest ion gyrofrequency from below. The developed approach is also efficient in plasmas containing a substantial amount of negative ions and/or heavy dust particulates. The accuracy of the approximate solution of the total cold plasma dispersion relation is illustrated graphically.  相似文献

Bone fatigue fracture is a progressive disease due to stress concentration. This study aims to evaluate the long bone fatigue damage using the ultrasonic guided waves. Two-dimensional finite-difference time-domain method was employed to simulate the ultrasonic guided wave propagation in the long bone under different elastic modulus. The experiment was conducted on a 3.8 mm-thick bovine bone plate. The phase velocities of two fundamental guided modes, A1 and S1, were measured by using the axial transmission technique. Simulation shows that the phase velocities of guided modes A1 and S1 decrease with the increasing of the fatigue damage. After 20,000 cycles of fatigue loading on the bone plate, the average phase velocities of A1 and S1 modes were 6.6% and 5.3% respectively, lower than those of the intact bone. The study suggests that ultrasonic guided waves can be potentially used to evaluate the fatigue damage in long bones.  相似文献

The fundamental ultrasonic guided modes are consistently observed in long bones ex vivo and in vivo. However, the responses of ultrasonic guided waves to the changes of cortical thickness, cortical elastic parameters, and thickness of the overlying soft tissues are not comprehensively understood. This paper systematically presents a sensitivity analysis of leaky Lamb modes to the geometry and material characteristics of layered bone model by means of semi-analytical finite element modeling. The stratified bone model is consisted of a transversely isotropic cortex with an overlying soft tissue and underlying marrow. The study is important as it offers guidance to the parameter inversion process about the optimal selection of guided modes and regions of sensitivity for better inversion results.  相似文献

The ability to rationally engineer secondary metabolic pathways is a promising approach to novel therapeutics. Atomic resolution structures of biosynthetic enzymes provide information on active site architecture and macromolecular assembly that can aid in the engineering of new compounds. Assembly of a bipolar mitotic spindle requires the action of class 5 kinesins, and inhibition or depletion of this motor results in mitotic arrest and apoptosis. S-Trityl-l-cysteine is an allosteric inhibitor of vertebrate Kinesin Spindle Protein (KSP) that has generated considerable interest due to its anti-cancer properties, however, poor pharmacological properties have limited the use of this compound. We have modified the triphenylmethyl and cysteine groups, guided by biochemical and cell-based assays, to yield new cysteinol and cysteamine derivatives with increased inhibitory activity, greater efficacy in model systems, and significantly enhanced potency against the NCI60 tumor panel. These results reveal a promising new class of conformationally-flexible small molecules as allosteric KSP inhibitors for use as research tools, with activities that provide impetus for further development as anti-tumor agents.  相似文献

In this study, the role of all-trans retinoic acid (RA) on the proliferation of rat embryonic pancreas ducts and on the proportion of insulin cells was investigated. All-trans RA (10-6 m) was added to Ham's F12.ITS serum-free medium in which 12.5 day rat dorsal pancreatic buds were cultured on Matrigel. Control explants were cultured on Matrigel in Ham's F12.ITS alone or in Ham's F12.ITS containing ethanol (the diluent for RA). After a 7 day culture period, explants were incubated with bromodeoxyuridine (BrdU) for assessment of cell proliferation. Explants were processed for both morphometry and immunocytochemistry. The length density and volume density of the pancreatic ducts were assessed using an image analysis system. Cells positive for insulin, BrdU and glucagon were localized on adjacent serial sections. RA treatment caused a statistically significant increase in the volume density (P < 0.007) and length density (P < 0.008) of the ducts, as well as a 1.2-fold increase (P < 0.0001) in the proportion of insulin to glucagon cells, compared to both control groups. Few insulin cells were BrdU positive, indicating that cells had a low proliferation rate. The increased proportion of insulin cells may relate to the increased volume density and length density of the ducts in RA-treated explants. It is suggested that RA stimulated the production of additional progenitor cells and not proliferation of existing insulin cells.  相似文献

Collective cell migration is an essential process in embryo development, wound healing, inflammatory response, and cancer invasion. Although cell motions in two-dimensional (2D) monolayers have been studied previously, three-dimensional (3D) collective cell migration, which constantly occurs during embryogenesis such as the establishment of ducts and acini in vivo, remains elusive. In this paper, we develop a cell-based model incorporating cell mechanics and cell motility to address coherent cell motions in a spherical acinus-like lumen with different cell populations. It is found that the interplays between cell persistence, random fluctuation, and geometrical confinement may engender rich and novel migratory modes. In a 3D spherical lumen, two cells may undergo stripe-like or cross-circular coherent rotations, whereas multiple cells can form dynamic twisting or circulating bands, leaving sparse cells at the center or even a hollow cavity in the cell aggregate. The cell density is found to profoundly influence the collective cell migration modes. Our model can reproduce the fundamental features observed in experiments and highlight the role of mechanics in steering 3D collective cell dynamics during mammary acinar morphogenesis.  相似文献

Statins have both cholesterol lowering and anti-inflammatory activities, whether mechanisms underlying their activities are independent remains unclear. The ATP-gated P2X(4) receptor is a pro-inflammatory mediator. Here, we investigate the action of fluvastatin and other cholesterol depleting agents on native and recombinant human P2X(4) receptor. Fluvastatin and mβCD suppressed P2X(4)-dependent calcium influx in THP-1 monocytes, without affecting P2Y receptor responses. mβCD or filipin III suppressed the current density of recombinant human P2X(4) receptors. Human P2X(2) was insensitive to cholesterol depletion. Cholesterol depletion had no effect on intrinsic P2X(4) receptor properties as judged by ATP concentration-response relationship, receptor rundown or current decay during agonist occupancy. These data suggest fluvastatin suppresses P2X(4) activity in monocytes through cholesterol depletion and not by modulating intrinsic channel properties.  相似文献

The bile ducts in the liver of larval sea lamprey, Petromyzon marinus, undergo programmed degeneration during metamorphosis. The degenerative process is most dramatic in the middle metamorphic stages (3-5), and is asynchronous, occurring more rapidly in small peripheral biliary components than in larger, medial ducts. All classes of bile ducts within the biliary tree exhibit similar histological changes during regression. The initial evidence of degeneration in the epithelium is a folding of the basal lamina, and this is accompanied by cell shrinkage and disruption of cell order. "Shedding" of microvilli and cytoplasmic constituents then takes place at the apical surface resulting in the accumulation of periodic acid-Schiff positive membranous debris in the Lumen. The appearance of "hyalin bodies" in the lumen coincides with the depletion of intermediate-sized filaments from the cytoplasmic matrix. Numerous, large dense bodies, myelin figures, and autophagic vacuoles are consistently observed in necrotic cells. Following cytolysis, bile duct remnants become ensheathed within regions of fibrosis. Ultimately, these fibrous regions are replaced with cords of hepatocytes. By stage 7, all bile ducts have disappeared. The events of biliary atresia in lampreys are comparable to tissue regression which is associated with normal development and pathological conditions in other vertebrates but are particularly reminiscent of those in human biliary atresia. The unique ability of the adult lamprey to service without bile ducts enhances the value of this organism as an experimental model for studying human biliary atresia.  相似文献

Zebrafish primordial germ cells (PGCs) are guided toward their targets by the chemokine SDF-1a. PGCs were followed during three phases of their migration: when migrating as individual cells, while remaining in a clustered configuration, and when moving as a cell cluster within the embryo. We found that individually migrating PGCs alternate between migratory and pausing modes. Pausing intervals are characterized by loss of cell polarity and correlate with subsequent changes in the direction of migration. These properties constitute an intrinsic behavior of PGCs, enabling erasure of prior polarity and re-sampling of the environment. Following migration arrest at a site of high SDF-1a levels, PGCs resume migration as a cluster. The seemingly coordinated cluster migration is a result of single-cell movement in response to local variations in SDF-1a distribution. Together, these behavioral modes allow the cells to arrive at specific destinations with high fidelity and remain at their target site.  相似文献

Insulin-like growth factor-1 (IGF-1) plays a critical role in the development of the growing skeleton by establishing both longitudinal and transverse bone accrual. IGF-1 has also been implicated in the maintenance of bone mass during late adulthood and aging, as decreases in serum IGF-1 levels appear to correlate with decreases in bone mineral density (BMD). Although informative, mouse models to date have been unable to separate the temporal effects of IGF-1 depletion on skeletal development. To address this problem, we performed a skeletal characterization of the inducible LID mouse (iLID), in which serum IGF-1 levels are depleted at selected ages. We found that depletion of serum IGF-1 in male iLID mice prior to adulthood (4 weeks) decreased trabecular bone architecture and significantly reduced transverse cortical bone properties (Ct.Ar, Ct.Th) by 16 weeks (adulthood). Likewise, depletion of serum IGF-1 in iLID males at 8 weeks of age, resulted in significantly reduced transverse cortical bone properties (Ct.Ar, Ct.Th) by 32 weeks (late adulthood), but had no effect on trabecular bone architecture. In contrast, depletion of serum IGF-1 after peak bone acquisition (at 16 weeks) resulted in enhancement of trabecular bone architecture, but no significant changes in cortical bone properties by 32 weeks as compared to controls. These results indicate that while serum IGF-1 is essential for bone accrual during the postnatal growth phase, depletion of IGF-1 after peak bone acquisition (16 weeks) is compartment-specific and does not have a detrimental effect on cortical bone mass in the older adult mouse.  相似文献

Biliary ducts collect bile from liver lobules, the smallest functional and anatomical units of liver, and carry it to the gallbladder. Disruptions in this process caused by defective embryonic development, or through ductal reaction in liver disease have a major impact on life quality and survival of patients. A deep understanding of the processes underlying bile duct lumen formation is crucial to identify intervention points to avoid or treat the appearance of defective bile ducts. Several hypotheses have been proposed to characterize the biophysical mechanisms driving initial bile duct lumen formation during embryogenesis. Here, guided by the quantification of morphological features and expression of genes in bile ducts from embryonic mouse liver, we sharpened these hypotheses and collected data to develop a high resolution individual cell-based computational model that enables to test alternative hypotheses in silico. This model permits realistic simulations of tissue and cell mechanics at sub-cellular scale. Our simulations suggest that successful bile duct lumen formation requires a simultaneous contribution of directed cell division of cholangiocytes, local osmotic effects generated by salt excretion in the lumen, and temporally-controlled differentiation of hepatoblasts to cholangiocytes, with apical constriction of cholangiocytes only moderately affecting luminal size.  相似文献

The sperm of the shiner surfperch are packaged into high density aggregations which are introduced into the female genital tract at insemination. Germ cell differentiation occurs within cysts formed by nongerminal Sertoli cells. In late spermiogenesis, spermatozoa within the cysts come to lie parallel to each other and become more densely packed. These sperm packets (spermatophores), containing approximately 600 spermatozoa, then are released into the efferent sperm ducts. The exact nature of the spermatophore binding material is not known, but a major component is proteinaceous and is synthesized in the rough endoplasmic reticulum of the efferent sperm duct epithelial cells. The mechanism by which the spermatophores pass from cysts into ducts is not clear. It appears that whereas many Sertoli cells degenerate causing the cyst wall to break down, many Sertoli cells do not degenerate, but rather assume the configuration of columnar duct cells. The spermatophores remain intact within the testicular ducts, but rapidly dissolve within the female ducts in response to increased pH.  相似文献

Summary Rats were given a lithium-containing diet (40 mmol/kg) to Study the effect of lithium on the structure of collecting ducts from the inner stripe of the outer medulla. The results show that there is a significant increase in the volume density of collecting ducts already after one week on this diet. The volume density of both intercalated and principal cells increases, whereas the volume density of mitochondria in the cytoplasm increases in the intercalated cells only. The increased volume of both principal and intercalated cells seems to be part of a general hyperplasia and hyperactivity of the collecting duct, which may in some way be related to the effects of lithium on vasopressinmediated water transport. The specific changes in the intercalated cells may be a consequence of the effects of lithium on distal nephron potassium and hydrogen ion transport in the distal nephron.  相似文献

The compound eyes of most diurnal butterflies have a reflecting tapetum below the retina. Light that enters the eye is guided down the rhabdom, reflected by the tapetum, and then guided back up the rhabdom. The light that is not absorbed by the rhabdom is reemitted and gives rise to an eyeshine. We have measured the fraction of the incident light that is re-emitted, and also the degree to which this light retains its original polarization. The following conclusions are drawn:
1.  Even at the wavelength where the eyeshine is most intense, only a few percent of the incident photons are re-emitted.
2.  The tapetum acts as a plane mirror that preserves polarization.
3.  The light that passes through the rhabdom in second-order waveguide modes is depolarized to a greater extent than the light contained in first-order modes. The depolarization is expected to decrease only slightly the polarization sensitivity of the retina.
4.  Theoretical modelling of the waveguide properties of the rhabdom provided a way of using depolarization measurements for estimating the refractive index of the rhabdom. The measured amount of depolarization is consistent with the dispersion of phase velocities of different second-order modes propagating in a rhabdom of refractive index 1.363.

The parotid glands of the pika and the volcano rabbit were examined by light and transmission electron microscopy. The acinar cells of the pika consisted of light cells containing basophilic granules of low density, while in the volcano rabbit the acinar cells consisted of light and dark cells containing acidophilic granules of moderate density. Intercalated duct cells were composed of light cells containing a few granules of moderate density. These segments of the two animals were similar in morphology. The striated duct cells in both species were composed of light and dark cells. Most of those in the pika contained a few moderately dense granules. In both animals, no myoepithelial cells were detected around the acini, intercalated ducts or striated ducts, while nerve terminals were observed among the adjacent acinar cells.  相似文献

The golden‐mangrove whistler complex Pachycephala pectoralis/melanura is, with 66 named populations, described as the most complex avian example of geographic variation in the World. It represents a well‐known example of niche differentiation where one species P. pectoralis is found on faunistically rich larger islands (and mainland) and the other P. melanura is confined to adjacent, faunistically poor islets. Except for a comparison of some Solomon Island populations, the relationships among these taxa have not been tested using molecular data. Here we combine a dense taxon sampling (22 populations of 12 named taxa distributed east of New Guinea and in Australia), with molecular data (two nuclear and two mitochondrial markers) to unravel more of the evolutionary history behind the golden whistler complex. In particular we examine relationships between Pachycephala melanura, and the very similar‐looking P. citreogaster (Bismarcks Islands) and P. pectoralis (Australia). We demonstrate that a well‐supported group of golden whistlers inhabit the Bismarck Islands and that several small islets in the immediate vicinity are inhabited by another monophyletic group consisting of P. melanura, mangrove golden whistler.  相似文献
