Muramylpeptides from bacteria cell wall are strong stimulators of immune system and phagocytic cells are main effectors. Dimer containing glucoseaminylmuramylpentapeptide (di-GMPP) was obtained from cell wall of Salmonella typhi bacteria. Di-GMPP decrease the phagocytic activity of macrophages obtained from peripheral blood of healthy donors and increase intracellular killing. Also di-GMPP resulted in decrease of expression of macrophages' receptors which play role in phagocytosis (CD16, CD64, CD11b) and detection of bacterial molecular patterns (TLR2, TLR4, CD206), as well as in increase of expression of antigen-presenting (HLA-DR) and costimulatory molecules (CD86, CD40) which involved in formation of immunological synapse and presentation of antigens to T- and B-lymphocytes. 相似文献

We are studying the enzymatic modification of polycyclic aromatic hydrocarbons (PAHs) by the laccase from Coriolopsis gallica UAMH 8260. The enzyme was produced during growth in a stirred tank reactor to 15 units ml−1, among the highest levels described for a wild-type fungus; the enzyme was the major protein produced under these conditions.
After purification, it exhibited characteristics typical of a white rot fungal laccase. Fifteen azo and phenolic compounds
at 1 mM concentration were tested as mediators in the laccase oxidation of anthracene. Higher anthracene oxidation was obtained
with the mediator combination of ABTS and HBT, showing a correlation between the oxidation rate and the mediator concentration.
Reactions with substituted phenols and anilines, conventional laccase substrates, and PAHs were compared using the native
laccase and enzyme preparations chemically modified with 5000 MW-poly(ethylene glycol). Chemically modified laccase oxidized
a similar range of substituted phenols as the native enzyme but with a higher catalytic efficiency. The k
cat increase by the chemical modification may be as great as 1300 times for syringaldazine oxidation. No effect was found of
chemical modification on mediated PAH oxidation. Both unmodified and PEG-modified laccases increased PAH oxidation up to 1000
times in the presence of radical mediators. Thus, a change of the protein surface improves the mediator oxidation efficiency,
but does not affect non-enzymatic PAH oxidation by oxidized mediators.

A study was made of the dynamics of the lysozyme activity of the serum in dogs which sustained clinical death on the background of a 2-hour hypovolemic hypotension due to blood loss. There was revealed a progressive increase in the lysozyme activity of the serum during the hypotension and during the first 30 minutes of the postreanimation period. Lysozyme activity of the serum was increased in the course of 4 days after the revival. The significance of the lysozyme activity of the serum as an index of hypoxic injury of the internal organs in terminal conditions is discussed. 相似文献

Characteristics of the mismatch negativity (MMN) were studied by presenting the subjects with four blocks of stimuli containing standard series of clicks (90%) simulating a stationery sound image located in the head midline, and one of three different deviant series of clicks (10%) simulating either a stationary sound image located near the left ear or a moving sound image which shifted from the head midline to the left ear or in the opposite direction. All the deviant stimuli elicited the MMN with the minimal peak amplitude and the greatest latency evoked by the deviant series of clicks simulating the sound image moving from the head midline to the left ear. These findings suggest that the MMN may be considered as a pre-perceptual physiological measure of the discrimination accuracy for the sound signals with various spatial locations. 相似文献

Most of the biological processes in the living organisms of both animals and man are known to be of rhythmical nature. Variability of enzymatic activity in circadian cycle depends on many factors among other on age, sexual maturity, diet as well as medication. The results obtained in our studies indicate, that the activity changes of acid phosphatase and ATP-ase Mg++ dependent in the liver of all the examined age groups were of 24 hour circadian rythm. As to the acid phosphatase activity the results of this experiments showed that in circadian cycle in all examined age groups there is only one peak of elevated activity. ATP-ase Mg++ dependent showed two activity peaks appearing at the same hour both in 30 and 60 days old animals. It should be noticed that the activities of ATP-ase Mg++ dependent in 100 day old animals were two times higher than in 30 and 60 days old rats. 相似文献

The influence of Calacarus heveae Feres on physiological processes was evaluated in two rubber tree clones. Experiments were conducted in a greenhouse with 5-month-old potted seedlings of RRIM 600 and GT 1 clones, that were either infested with C. heveae or not (non-infested control). The level of photosynthetic pigments, net photosynthetic rate, stomatal conductance, transpiration rate, changes in relative humidity between leaf surface and ambient air (Δw) and intercellular CO(2) concentration (Ci CO(2)) were evaluated. Infested plants showed significant reductions in the rate of transpiration, the rate of photosynthesis, stomatal conductance and Δw. RRIM 600 seedlings showed more pronounced physiological damage than GT 1 seedlings, indicating a lower physiological tolerance of the former clone to the mite. However, carotenoid levels were reduced only in GT 1 seedlings. Photosynthesis was probably reduced due to a decrease in stomatal opening, as indicated by reductions in transpiration rate and stomatal conductance and by the absence of differences in chlorophyll levels between treatments. Our results indicate that populations of C. heveae reduce the productivity of rubber trees. Thus, farmers must to be aware to control this mite pest in rubber tree plantations. 相似文献

The transesterification activity, autolysis, thermal stability and conformation of subtilisin Carlsberg, made soluble in dioxane by covalent linking to methoxypoly(ethylene glycol) (PEG), were investigated as a function of the concentration of water in the medium. Electrospray mass spectrometry showed that the modified enzyme preparation was a mixture of proteins containing from 2 to 5 covalently linked PEG chains per subtilisin molecule. PEG-subtilisin catalyzed transesterification between vinyl butyrate and 1-hexanol was optimum at 0.55 MH(2)O, while hydrolysis prevailed above 2 MH(2)O. There was a decrease in the overall enzyme activity with increasing water concentration because of autolysis and denaturation of the enzyme. Subtilisin powder and celite-immobilized subtilisin were more stable and less susceptible to autolysis than the PEG-modified enzyme. Circular dichroism and intrinsic protein-fluorescence studies showed that the conformation of PEG-subtilisin did not change as a function of water concentrations between 0 and 9 M. The K(m,app) value of PEG-subtilisin for 1-hexanol was highly influenced by water, which behaved as a competitive inhibitor in the transesterification reaction with an affinity for the enzyme similar to that of the alcohol. The K(m,app) for the acylating agent was not significantly modified by water. Lyoprotectants such as sorbitol and free PEG did not influence the activity of PEG-subtilisin but notably increased the activity of subtilisin powder and celite-immobilized subtilisin. The addition of 1.7-5.5 M water, however, rendered enzyme preparations containing no additives as active as those containing the lyoprotectants. (c) 1997 John Wiley & Sons, Inc. Abstract. Two groups of rabbits were infested twice with different numbers of Ixodes ricinus adults: one group (high infestation) with twenty-five females and twenty-five males and the other group (low infestation) with five pairs. A third infestation was performed in both groups with fifteen adult pairs. Tick biology was monitored for resistance effects. At the second infestation, the feeding and the egg production were more perturbed in ticks fed on high infestation rabbits. The embryogenesis was only affected in ticks from high infestation rabbits. At the third infestation, resistance was increased only in low infestation rabbits, which became more resistant than high infestation rabbits. The blastogenic response of peripheral blood lymphocytes and antibody production against ticks were assessed. A salivary gland extract and an integumental antigen from Lricinus adult females were able to initiate lymphocyte proliferation. The response was significantly higher in high infestation rabbits, especially at the end of the second infestation, and higher in low infestation rabbits during the third infestation. Non-specific proliferation with concanavalin A was temporarily decreased in both rabbits groups during the first and the second infestations. Specific antibody response to salivary and integument antigens were always the highest in high infestation rabbits. The involvement of tick-induced immunosuppression is discussed. 相似文献
