EF-1a binds aminoacyl-tRNA to the ribosome with the hydrolysis of GTP; the complex facilitates the exchange of GDP for GTP to initiate another round of elongation. To examine the subunit structure of EF-1 and phosphorylation by protein kinase CKII, recombinant , , and subunits from rabbit were expressed in E. coli and the subunits were reconstituted into partial and complete complexes and analyzed by gel filtration. To determine the availability of the and subunits for phosphorylation by CKII, the subunits and the reconstituted complexes were examined as substrates for CKII. Formation of the nucleotide exchange complex increased the rate of phosphorylation of the subunit and reduced the Km, while addition of to or the complex inhibited phosphorylation by CKII. However, a had little effect on phosphorylation of . Thus, the and subunits in EF-1 were differentially phosphorylated by CKII, in that phosphorylation of was altered by association with other subunits, while the site on was always available for phosphorylation by CKII. From the availability of the subunits for phosphorylation by CKII and the composition of the reconstituted partial and complete complexes, a model for the subunit structure of EF-1 consisting of (22)2 is proposed and discussed.  相似文献

Summary Lymphokine production by human melanoma tumor-infiltrating lymphocytes (TIL) was studied. Uncultured TIL produced interferon (IFN), but not interleukin-2 (IL-2) or IL-4, in response to anti-CD3 mAb or IL-2. In bulk cultures, IL-2-activated TIL displaying autologous tumor-specific cytotoxicity (CTL-TIL) produced IFN in culture with medium alone, whereas IL-2-activated noncytotoxic TIL did not. Addition of anti-CD3 mAb or autologous tumor cells up-regulated IFN production in IL-2-activated TIL from 10 of 12 or 6 of 12 cases respectively. Those from 4 of 12 cases (2 CTL-TIL and 2 noncytotoxic TIL) produced IL-2 in culture with medium alone. At the clonal level, 5 (4 CD4+ and 1 CD8+) of 7 autologous tumor-specific CTL clones derived from TIL and 3 (2 CD4+ and 1 CD8+) of 7 noncytotoxic TIL clones produced IFN in culture with medium alone, which was up-regulated by adding anti-CD3 mAb. Two IFN-producing CTL clones tested produced IL-2 in 4 ×-concentrated supernatants from a 3.5-h culture with medium alone. Furthermore, 2 IFN-producing CTL clones tested expressed mRNA for both IFN and IL-2. IL-2 production and its mRNA expression were up- or down-regulated, respectively, by adding anti-CD3 mAb or autologous tumor cells. IL-4 production was not observed in culture either with medium alone or with IL-2 in any of the cells described above. Anti-CD3 mAb was required for IL-4 production in 3 of 12 IL-2-activated TIL, 2 of 6 CTL clones, and none of 5 noncytotoxic TIL clones. In summary, IFN production was characteristic of melanoma TIL. Some autologous tumor-specific CTL in TIL are suggested to be productive of IL-2 and IFN under unstimulated conditions, both being required for self-activation in an autocrine loop.This work was supported in part by grant CA-47891 from the National Cancer Institute  相似文献

In cyclically fluctuating vole populations, strong intraspecific competition and intense predation simultaneously or separately increase the costs of reproduction and so may set the framework for the optimal breeding tactic of voles. In a factorial experiment, we manipulated two factors in the breeding environment of bank vole (Clethrionomys glareolus) pairs, each with two treatment levels: no predation risk or high risk of specialist predators and low or high density of voles. In the manipulation, we used odours of conspecific voles and/or mustelid predators. Both over-wintered and young, summer-born, wild-caught bank vole females and males in breeding condition were used. Each female—male pair of voles was placed in a cage and the cages were distributed in large outdoor enclosures. All animals were fed ad libitum. Under predation risk, both old and young females suppressed breeding significantly. The density of conspecific voles did not affect overall breeding. However, there was some evidence that population density stimulated breeding of old females but suppressed breeding of young ones. Both risk factors appeared to increase litter sizes of those individuals who decided to breed. Our results indicate that the risk of predation may be an important factor determining reproductive tactics of bank vole females. In risky environments, females seemed to choose between two totally opposite tactics: they suppressed breeding, which may increase their own survival to the next breeding event, or they continued to breed in spite of expected high survival costs. Females seemed to compensate the latter costs with a higher effort to the current and probably the last reproduction.  相似文献

Growth rings (GR) in vertebral centra of 15 whale sharks, Rhincodon typus, four female (418–750cm precaudal length), 10 male (422–770cm), and one of unknown sex (688cm), were examined using x-radiography. GR counts were made from scanned images and count precision was determined using the average percentage error index (4.19%) and the index of precision D (3.31%). In females, counts ranged from 19 GR (418cm) to 27 GR (750cm); in males from 20 GR (670cm) to 31 GR (770cm). Three mature males had 20 GR (670cm), 24 GR (744cm) and 27 GR (755cm). A female with 22 GR (445cm) was adolescent. There was a linear relationship between centrum dorsal diameter and body length, and back-calculated body lengths at number of GR are presented. A linear relationship between body length and number of GR prevented the calculation of von Bertalanffy parameters from either observed or back-calculated values.  相似文献

We have developed a new method for the large scale preparation of pyridylaminated (PA-) oligosaccharides from glycoproteins. Phenol/chloroform extration was adapted for the removal of protein and excess 2-aminopyridine, improving the efficiency of preparation. From a 2.5 g sample of human apo-transferrin, 25–30 mol of agalacto biantennary PA-oligosaccharide could be obtained. Summary ATP-inhibited potassium channels (K(ATP)) were studied in excised, inside-out patches from cultured adult mouse pancreatic -cells and HIT cells. In the absence of ATP, ADP opened K(ATP) channels at concentrations as low as 10 m and as high as 500 m, with maximal activation between 10 and 100 m ADP in mouse -cell membrane patches. At concentrations greater than 500 m, ADP inhibited K(ATP) channels while 10 mm virtually abolished channel activity. HIT cell channels had a similar biphasic response to ADP except that more than 1 mm ADP was required for inhibition. The channel opening effect of ADP required magnesium while channel inhibition did not. Using creatine/creatine phosphate solutions with creatine phosphokinase to fix ATP and ADP concentrations, we found substantially different K(ATP)-channel activity with solutions having the same ATP/ADP ratio but different absolute total nucleotide levels. To account for ATP-ADP competition, we propose a new model of channel-nucleotide interactions with two kinds of ADP binding sites regulating the channel. One site specifically binds MgADP and increases channel opening. The other, the previously described ATP site, binds either ATP or ADP and decreases channel opening. This model very closely fits the ADP concentration-response curve and, when incorporated into a model of -cell membrane potential, increasing ADP in the 10 and 100 m range is predicted to compete very effectively with millimolar levels of ATP to hyperpolarize -cells.The results suggest that (i) K(ATP)-channel activity is not well predicted by the ATP/ADP ratio, and (ii) ADP is a plausible regulator of K(ATP) channels even if its free cytoplasmic concentration is in the 10–100 m range as suggested by biochemical studies.We would like to thank Mr. Louis Stamps for expert technical assistance and Dr. Wil Fujimoto and Ms. Jeanette Teague for generously providing HIT cells obtained from Dr. Robert Santerre at Eli Lilly. The reproductive behavior of horned rainbow scarab beetles,Phanaeus difformis, was studied to determine the influence of morphological traits on intersexual and intrasexual interactions.Phanaeus difformis is a sexually dimorphic dung beetle in which males possess much larger horns than females, and males can be grouped into major and minor male morph categories based on horn size. Male-female pairs cooperated in nest construction and provisioning. In the laboratory, males of both morphs assisted females and were equally successful at copulating. However, in the field larger individuals had a pairing advantage due to greater success in intrasexual competition. Some males used an alternative mating tactic which involved sneaking copulations with paired females. In most cases the sneak male was smaller than the paired male.  相似文献

A novel syrup containing neofructo-oligosaccharides was produced from sucrose (Brix 70) by whole cells of Penicillium citrinum. The efficiency of fructo-oligosaccharides production was more than 55% and those of the main carbohydrate components, 1-kestose (Fruf 21Fruf 21 Glc), nystose (Fruf 21Fruf 21 Fruf 21 Glc) and neokestose (Fruf 26 Glc12 Fruf), were 22, 14 and 11%, respectively.  相似文献

A model of associative memory for time varying spatial patterns is proposed and simulated on a digital computer. This is a network composed of many neuron-like elements, and shows an ability for associative memory similar to that of the brain.Suppose a number of sequences of spatial patterns are presented to this network, for example, 12345, ABC, and so on. Then, these patterns are memorized in the network. After that, if any part of one of these sequences, say 23, is presented to the circuit, the rest of the sequence, 45, is recalled following to it. It resembles to such a situation — if we hear a part of a melody which we have memorized in the past, the rest of the melody is recalled even after it is stopped half-way. Although the recalled patterns are not always 100% correct, they are not completely destroyed even if the presented patterns are imperfect.  相似文献

Zusammenfassung 1974–1981 wurden in einem Lärchen-Versuchsforst (320 ha; 600 künstliche Nisthöhlen) in Niedersachsen (52.27 N/7.15 E) 529 Brutvögel des Trauerschnäppers beringt. Geburtsortstreue — auf das Versuchsgebiet bezogen — konnte bei 78 Individuen (=5,2 % der ausgeflogenen Nestlinge) festgestellt werden. Die geburtsortstreuen wurden in der Regel schon im ersten Jahr als Brutvogel nachgewiesen, während die Hälfte der geburtsortstreuen bei der ersten Erfassung ein Alter von mindestens 2 Jahren besaß. In der mittleren Entfernung zwischen Geburts- und erstbekannter Bruthöhle ergab sich zwischen den Geschlechtern kein gesicherter Unterschied ( : 883 m, : 798 m). Geschwister verhielten sich bezüglich Ansiedlungsalter und Ansiedlungs-entfernung nicht einheitlich. Von den als Brutvögel beringten Trauerschnäppern konnten 38,2 % der und 24,0 % der in einem späteren Jahr wiedergefangen werden. Bei den lagen die in aufeinanderfolgenden Jahren besetzten Nisthöhlen näher beieinander als bei den (Mittelwerte: 371 m bzw. 534 m). In der untersuchten Population war in allen Jahren bei den der Prozentsatz brutortstreuer Individuen höher und der Anteil an Zuwanderern geringer als bei den (Mittelwerte: 48 % gegenüber 30 % bzw. 45 % gegenüber 64 %). Diese Befunde wurden unter anderem im Hinblick auf die Lage des Untersuchungsgebietes am Rande des Trauerschnäpper-Areals diskutiert. Die Ortstreue war geringer ausgebildet als in mehreren anderen Populationen.
On place-fidelity behaviour in the Pied Flycatcher(Ficedula hypoleuca) in the western border area of its Central European range
Summary The evaluation is based on findings from 1974 to 1981, in a 320 ha. experimental larch forest (52.27N, 7.15E), with 600 nestboxes, in western Lower Saxony, from a total of 529 breeding Pied Flycatchers. Fidelity to birthplace — recovery in the study area — was recorded for 78 individuals (=5.2 % of the fledged nestlings). faithful to birthplace were traced, as a rule, in the first year, while half of the who showed this fidelity were at least 2 years old when first recorded. There was no significant difference between sexes in the mean distance between the hole in which they were hatched and the first known breeding hole ( : 883 m, : 798 m). There was no uniformity among siblings in relation to age of settlement and distance of settlement. 38.2 % of and 24.0 % of Pied Flycatchers ringed as breeding birds were recaptured in a later year. Nest boxes occupied in successive years were closer together in the case of than (mean: 371 m and 534 m respectively). The percentage of individuals showing fidelity to birthplace was higher, in all years, and the number of immigrants smaller, in the than in (mean: 48 % against 30 % and 45 % against 64 % respectively). These findings are discussed in relation to the border situation of the study area. The rate of recoveries was lower than in some other populations.

Feulgen and silver-stained karyotypes and meiosis of two triploid viviparous onion forms (Allium cepa var.viviparum), the Croatian Ljutika and the Indian Pran, were comparatively analyzed. The results of chromosome measurements show that Ljutika and Pran are karyologically not identical, although significant similarities were found in the morphology of their chromosomes. Five geographically distant clones of Ljutika showed good agreement in the number and gross morphology of the chromosomes and in the number and position of NORs and interphase nucleoli. Heterotrivalents were predominant in meiosis of Ljutika but a relatively high frequency of higher multivalents together with univalents and bivalents were also observed. The relationship between Ljutika and Pran and their possible origin are discussed.  相似文献

Four glycosidases were analyzed in 10 mm apical segments prepared from growing roots (15 mm) of Zea mays L. The pH optima were found to be 5.8 for -glucosidase, 4.4 for -galactosidase, 6.4 for -glucosidase and 6.0 for -galactosidase. The -glucosidase showed 4-fold higher activity than the -galactosidase. Zusammenfassung In der vorliegenden Arbeit werden Beobachtungen ausgewertet, die in den letzten Jahren in einer Seggenrohrsängerpopulation der Mark Brandenburg gesammelt wurden. Brutbiotop ist eine Großseggenwiese. und haben getrennte Reviere, wobei sich -Reviere, -Reviere und auch - und -Reviere zum Teil sehr erheblich überlagern. Keinem konnte ein bestimmtes zugeordnet werden, sie leben also in Keinehe. versuchen, aus der Nestnähe zu vertreiben. Aggressive Handlungen zwischen benachbarten konnten nicht festgestellt werden. Ihre Singflüge unterscheiden sich von denen der Schilfrohrsänger. 28 Eier maßen im Durchschnitt 17,76×13,14 mm. Nur die brüten und Füttern. Die durchschnittliche Fütterungsfrequenz liegt bei 19,7 Fütterungen pro Stunde, die tägliche Aktivitätszeit der ist im Juli etwa 1 Stunde kürzer als im Juni. 13–14 Tage nach dem Schlüpfen verlassen die Jungen das Nest. In der Nestlingsnahrung ist bei Julibruten der hohe Heuschreckenanteil auffällig. Nachdem die Jungen selbständig sind, streunen sie im Brutbiotop umher. Zweitbruten sind sehr wahrscheinlich, konnten aber noch nicht nachgewiesen werden.
Summary Observations made during the last years on a population of the Aquatic Warbler (Acrocephalus paludicola) in Brandenburg, Germany, gave the following results:In a sedge meadow, withCarex elata the dominant species, males and females ofAcrocephalus paludicola held separate territories, those of males as well of females sometimes considerably overlapping territories held by other individuals of the same or the opposite sex. In no case a certain male could be considered as having formed a firm pair bond with a certain female: on the contrary, the relations between the sexes were very loose, and probably the birds are even promiscuous. Females try to chase away males from the neighbourhood of their nests. Aggressiveness between neighbouring males has not been observed. The display flights of the males are different from those of the Sedge Warbler(A. schoenobaenus). 28 eggs measure on the average 17,76×13,14 mm. Females alone incubate and feed the young, at an average rate of 19,7 times per hour. The young leave the nest 13–14 days after hatching. Young reared in July are fed a great amount of grasshoppers. Second broods probably occur, but could not be proved.

Zusammenfassung In zunehmendem Maße werden anisoploideBeta-Rübensorten angebaut, deren zytologische Kontrolle zwecks Feststellung der Genomstufenprozentanteile recht arbeitszeitaufwendig ist. Übereinstimmend mit polnischen Autoren wurde festgestellt, daß die Hypokotylfarbe ein geeigneter Markierungsfaktor für die einzelnen Genomstufen darstellt. Kreuzt man tetraploide Pflanzen, die ein grünes Hypokotyl besitzen, mit diploiden Pflanzen, die ein rosa Hypokotyl aufweisen, so erhält man von dem tetraploiden Partner tetraploide grüne und triploide hellbraune, von dem diploiden Partner diploide rosa und triploide hellbraune Nachkommenschaften. Die in bezug auf die Hypokotylfarbe heterozygoten Pflanzen kann man demnach von den homozygot grünen und homozygot rosa Individuen unterscheiden. Die Kreuzung diploid grünxtetraploid rosa ist für diese Zwecke nicht brauchbar, da sich die triploiden Heterozygoten mit einem grünen und zwei rosa Allelen in der Hypokotylfarbe nicht deutlich von den homozygoten rosa Pflanzen abheben. Auf die Bedeutung dieser Markierungsmöglichkeit für bestimmte Forschungsprobleme, die Züchtung und die Saatgutkontrolle wird hingewiesen.  相似文献

Identification of estrogenresponsive genes is important to understand the molecular mechanisms of estrogen action. Suppression subtractive hybridization was employed to screen estrogenresponsive genes in chick liver. A single injection of estrogen into 6weekold chick induced upregulation of several known genes encoded for yolk proteins, such as Vitellogenin I and II and very low density lipoprotein II (apo-VLDL II). One novel sequence displayed a dramatic change (3fold increase) in response to estrogen treatment. This cDNA fragment was extended and the resultant sequence was analyzed. Translated amino acid sequence was 90, 88, 83 and 87% identical to the Larginine:glycine amidinotransferase of pig, rat, frog and human, respectively. The sequence has a conservative catalytic site of Larginine:glycine amidinotransferase. The expression pattern of this gene in organs is consistent with previous reports of Larginine:glycine amidinotransferase in chick. Thus, this clone represented the chicken Larginine:glycine amidinotransferase. It appeared that estrogeninduced alteration of arginine:glycine amidinotransferase was not dependent on protein synthesis, because concurrent administration of cycloheximide did not affect the estrogenmediated expression pattern. This is the first study demonstrating that Larginine:glycine amidinotransferase is a target of the estrogen receptor.  相似文献

Those standard historiographic themes of evolution and revolution need replacing. They perpetuate mid-Victorian scientists history of science. Historians history of science does well to take in the long run from the Greek and Hebrew heritages on, and to work at avoiding misleading anachronism and teleology. As an alternative to the usual evo-revo themes, a historiography of origins and species, of cosmologies (including microcosmogonies and macrocosmogonies) and ontologies, is developed here. The advantages of such a historiography are illustrated by looking briefly at a number of transitions the transition from Greek and Hebrew doctrines to their integrations by medieval authors; the transition from the Platonist, Aristotelian, Christian Aquinas to the Newtonian Buffon and to the no less Newtonian Lamarck; the departures the early Darwin made away from Lamarcks and from Lyells views. Issues concerning historical thinking about nature, concerning essentialism and concerning classification are addressed in an attempt to challenge customary stereotypes. Questions about originality and influence are raised, especially concerning Darwins tree of life scheme. The broader historiography of Darwinian science as a social ideology, and as a worldview, is examined and the scope for revisions emphasised. Throughout, graduate students are encouraged to see this topic area not as worked out, but as full of opportunities for fresh contributions.  相似文献

Zusammenfassung 1980–1984 wurden in Südfrankreich und Portugal Untersuchungen zum agonistischen Verhalten, Territorialverhalten und zur Balz der ZwergtrappeTetrax tetrax durchgeführt.Während der Fortpflanzungsperiode kommt es zwischen Zwergtrappen- häufig zu agonistischen Auseinandersetzungen. Ausdruck aggressiver Erregung ist enges Anlegen des Halsgefieders, Langstrecken des Halses, und nur beim Aufrichten und dachartiges Falten der Schwanzfedern. Aggressive Verhaltensweisen sind Drohen durch Hinlaufen oder -fliegen zu Reviereindringlingen, seitliches Drohen gleichstarker , und nur, wenn Verhaltensweisen niedrigerer Intensität nicht zur Unterlegenheit eines der Rivalen führen, Schnabelkämpfe.Rufen dient der Markierung eines Reviers. Die Intensität des Rufens wird durch die Anwesenheit von nicht beeinflußt, die Anwesenheit fremder nahe der Reviergrenze führt jedoch zu einem deutlichen Anstieg. Die Rufbewegung macht eine gewisse optische Signalwirkung, vor allem auf kurze Distanz, zusätzlich zur akustischen Wirkung, wahrscheinlich.Fliegende erzeugen mit den Schwingen (besondere Struktur der 7. Handschwinge) ein pfeifendes Geräusch, das während der Fortpflanzungszeit Bedeutung in der innerartlichen Kommunikation hat. Es zeigt sowohl als auch den Anflug eines weiteren an und löst damit territoriale bzw. aggressive Verhaltensweisen oder Flucht aus. Weitere Bedeutung erlangt es als Element des Territorialen Flügelschlagens und der Sprungbalz.Über ihrem Revier fliegen territoriale stets mit leicht hochgebogenem Kopf und verhaltenen, flachen Flügelschlägen. Deutungen dieses Fluges als Imponierflug zur besseren Darstellung des auffällig gefärbten Halsgefieders bzw. als Suchflug zum leichteren Auffinden und Verjagen von Reviereindringlingen werden diskutiert.Territoriales Flügelschlagen beginnt mit Fußtrampeln, das sich beschleunigt und in einen Ruf überleitet. Gleichzeitig schlägt das dreimal sehr schnell und flach mit den Flügeln, hebt jedoch nicht vom Boden ab. Alle Elemente des Verhaltens sind deutlich zu hören. Der Verstärkung der beim Fußtrampeln erzeugten Klopfgeräusche dienen Balzplätze, die entweder auf akustisch besonders geeignetem Boden angelegt oder durch das Fußtrampeln der sekundär verbessert werden.Territoriales Flügelschlagen wird ausschließlich in niedrigen Lichtintensitäten während kurzer Zeit in der Morgen- und Abenddämmerung gezeigt. Die Anwesenheit von hat keinen Einfluß auf seine Intensität. Es ist eine territoriale Verhaltensweise mit akustischem Signal und wird als ritualisiertes Anlaufen gegen einen Reviereindringling bzw. ritualisiertes Auffliegen eines Revier- zum Eindringling hin gedeutet. Sie ähnelt zwar in der Ausführung dem Territorialen Flügelschlagen, der Vogel hebt sich jedoch während der langsameren Flügelschläge durch einen Sprung vom Boden ab, und das Fußtrampeln ist wesentlich weniger intensiv. Charakteristische Flügelbewegungen während des Balzsprunges exponieren schwarzweiße Gefiederpartien bis in 65 cm Höhe. Sprungbalz erhöht im Vergleich zu Territorialem Flügelschlagen stark die optische Auffälligkeit eines ; die Sprunghöhe garantiert zusammen mit der Lage der Balzplätze bei geringstmöglichem Energieaufwand eine maximale Sichtbarkeit des über der umgebenden Vegetation. Sowohl die zeitliche Korrelation der Sprungbalz mit den Aktivitäten der als auch die Art der Interaktionen mit während Sprungbalzphasen machen deutlich, daß diese Verhaltensweise ins Paarungsrevier zieht.Das Hennenjagen dient der Stimulierung der zur Kopulation. Das läuft in charakteristischer Körperhaltung schnell hinter einem her, hat dabei den Kopf tief in die aufgerichtete Halskrause eingezogen, hält oft ruckartig an und ruft unter Zurseitewerfen des Kopfes. Kopulationen sind sehr kurz. Offensichtlich kann das Hennenjagen die sonst sehr ausgeprägte Abwehr des gegen das unterdrücken und zur Kopulationsbereitschaft führen. Die Kopulation selbst jedoch hebt diesen Effekt wieder auf, so daß nach 1–2 Sekunden das abwehrt oder flieht.
Agonistic behaviour, territorial behaviour and courtship display of the Little Bustard (Tetrax tetrax)
Summary From 1980 to 1984 studies were made of the agonistic, territorial and courtship behaviour of the Little Bustard in Southern France and Portugal. The behaviour patterns are described in detail, the signals involved are analysed, and factors which could have led to their evolution are discussed.In the reproductive period agonistic encounters between males happen remarkably often. Aggressive excitement is expressed in both sexes by compression of the neck-feathers and stretching of the neck, and in the female by the erection and lateral folding of the tail-feathers. There are several kinds of threat behaviour in territorial encounters: running or flying towards intruders; lateral threat behaviour (if both males are of similar strength); and bill-fighting which only occurs if aggressive behaviour of a lower intensity has not led to the submission of one of the rivals. Calling is a territorial behaviour and serves to mark a territory. The frequency of calls is not affected by the presence of females, but the presence of other males close to the territory borders can result in a marked increase in calling frequency. In addition to the acoustic signal, the call involves a sharp neck-jerk which may act as an optical signal, at least over short distances.In flight males produce a whistling sound by means of the specially adapted 7th primary feather on their wings. This sound has important meaning in intraspecific communication during the reproductive period: it announces a flying male to other males as well as to females, and causes territorial and aggressive behaviour, or fleeing of these birds. The flight sound is also an important element of the behaviour patterns territorial wingbeat display and jumping display. Within their territories males always fly with the neck raised at a slight angle and with suppressed, shallow wing-beats. Two interpretations of this flight are discussed: that it serves to advertise the conspicuously coloured neck-feathers in a display; or that it is a search flight for locating and chasing-off intruders.Territorial wingbeat display begins with an accelerating foot-stamping and leads into a call. During the call the male performs three very fast, whistling wing-beats but remains on the ground. The combined elements, foot-stamping, calling and whistling wings, produce a unique and distinctive acoustic signal. The foot-stamping is only performed at special display sites where the sound is amplified by the substrate structure. At such sites, the soil typically has trapped pockets of air below a compacted surface which may result from the repeated defecating and stamping of the male on the same spot. Territorial wingbeat display behaviour is only performed for short periods in low light intensities at dawn and dusk. The presence of females has no effect on the intensity of the behaviour which serves a territorial function. It is interpreted to be a ritualisation of the aggressive running or flying of a territorial male towards an intruder. Optical elements of this behaviour cannot have much importance because the body and wings of the bird are rarely visible above the vegetation. Territorial wingbeat display behaviour cannot be seen over long distances and from this reason cannot serve to attract females into a males territory.Jumping display is only performed at much higher light intensities than territorial wingbeat display so the two never occur at the same time of day. The intensity of the behaviour increases markedly in the presence of females in sharp contrast to the territorial wingbeat display. The jumping display is performed in a similar way to the territorial wingbeat display except that the foot-stamping is much less intensive, the wing-beats are slower, and the bird jumps off the ground during the wing-beats. During the jumping display the black and white patterns on the body and wings are clearly visible and the behaviour increases the conspicuousness of the male markedly. The jump, advertising the wing-pattern up to a height of 65 cm, together with the specific location of the display site, ensures that maximum visibility of the male above the vegetation is achieved at minimum energetic cost. Activities of the females and their interactions with males during the jumping periods indicate that this behaviour serves to attract females to the males territory.The chasing of females is also a courtship behaviour and serves in stimulating and preparing females for copulation. In a characteristic posture with the head retracted into the neck-collar, the male rapidly runs behind the female, repeatedly stopping abruptly and calling whilst throwing its head and body sideways. Copulations are performed very quickly and only happen after a female has been chased for some time. Under certain preconditions chasing suppresses the aggressive and defence behaviour of the female which normally characterises encounters with males, and thus leads to readiness for copulation. Copulation itself removes this effect and after only one or two seconds aggression leads to the escape of the female.

Summary Two specific -N-acetylglucosaminyltransferases involved in the branching and elongation of mucin oligosaccharide chains, namely, a 1,6 N-acetylglucosaminylsaminyltransferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3GalNAc-Mucin to yield Gal3(GlcNAc6)GalNAc-Mucin and a 3-N-acetylglucosaminyl transferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3(GlcNAC6)GalNAc-mucin to yield GlcNAc3Gal3 (GlcNAc6)GalNAc-Mucin were purified from the microsomal fraction of swine trachea epithelium. The 1,6-N-acetylglucosaminyltransferase was purified about 21,800-fold by procedures which included affinity chromatography on DEAE columns containing bound asialo Cowper's gland mucin glycoprotein with Gal1,3GalNAc side chains. The apparent molecular weight estimated by gel filtration was found to be about 60 Kd. The purified enzyme showed a high specificity for Gal1,3GalNAc chains and the most active substrates were mucin glycoproteins containing these chains. The apparent Km of the 6-glucosaminyltrans-ferase for Cowper's gland mucin glycoprotein containing Gal1,3GalNAc chains was 0.53 µM; for UDP-N-acetylglucosamine, 12 µM; and for Gal 1,3GalNAc NO2ø, 4 mM. The activity of the 6-glucosaminyltransferase was dependent on the extent of glycosylation of the Gal3GalNAc chains in Cowper's gland mucin glycoprotein.The best substrate for the partially purified 3-Glucosaminyltransferase was Cowper's gland mucin glycoprotein containing Gal1,3(GlcNAc6)GalNAc side chains. This enzyme showed little or no activity with intact sialylated Cowper's gland mucin glycoprotein or derivatives of this glycoprotein containing GalNAc or Gal1,3GalNAc side chains.The radioactive oligosaccharides formed by these enzymes in large scale reaction mixtures were released from the mucin glycoproteins by treatment with alkaline borohydride, isolated by gel filtration on Bio-Gel P-6 and characterized by methylation analysis and sequential digestion with exoglycosidases. The oligosaccharide products formed by the 6- and 3-glucosaminyltransferases were shown to be Gal3(GlcNAC6) GalNAc and GlcNAc3 Gal3(GlcNAC6)GalNAc respectively.Taken collectively, these results demonstrate that swine trachea epithelium contains two specific N-acetylglucosaminyltransferases which catalyze the initial branching and elongation reactions involved in the synthesis of O-linked oligosaccharide chains in respiratory mucin glycoproteins. The first enzyme a 6-glucosaminyltransferase converts Gal3GalNAc chains in mucin glycoproteins to Gal3(GlcNAc6)GalNAc chains. This product is the substrate for a second 3-glucosaminyltransferase which converts the Gal3(GlcNAc6)GalNAc chains to GlcNAc3Gal(GlcNAc6)GalNAc chains in the glycoprotein. Amyloid beta-protein (A), in its soluble form, is known to bind several circulatory proteins such as apolipoprotein (apo) E, apo J and transthyretin. However, the binding of A to intracellular proteins has not been studied. We have developed an overlay assay to study A binding to intracellular brain proteins. The supernatants from both rat and human brains were found to contain several proteins that bind to A 1–40 and A 1–42. No major difference was observed in the A binding-proteins from brain supernatants of patients with Alzheimer's disease and normal age-matched controls. Binding studies using shorter amyloid beta-peptides and competitive overlay assays showed that the binding site of A to brain proteins resides between 12–28 amino acid sequence of A. The presence of several intracellular A-binding (AB) proteins suggests that these proteins may either protect A from its fibrillization or alternatively promote A polymerization. Identification of these proteins and their binding affinities for A are needed to assess their potential role in the pathogenesis of Alzheimer's disease.  相似文献
