Field experiments were conducted in Sicily (south Italy) during two seasons to characterize by chlorophyll (Chl) fluorescence four genotypes (Spunta, Sieglinde, Daytona, and Ninfa) of potato (Solanum tuberosum L.) for off-season production during plant aging and to analyse the possible relation between Chl parameters and tuber yield. Chl fluorescence parameters [initial fluorescence (F0), maximum fluorescence (Fm), Fv/Fm, time in which maximal fluorescence occurs (Tmax)] gained from Kautsky kinetics and Chl content were measured weekly, from 5th to 6th leaf appearance to beginning of plant senescence in the first season and to full plant senescence in the second season. F0 and Fv/Fm were the most reliable Chl fluorescence parameters for the definition of genotypic differences while Chl content and Tmax were the most reliable Chl parameters to predict plant aging. Tuber yield was highly correlated with Chl content, Tmax, F0, and Fm.

Changes in stomatal and chlorophyll fluorescence characteristics were analyzed in the course of leaf expansion in European beech (Fagus sylvatica L.) cultivars Aurea Pendula, Cristata, Rohanii, Rotundifolia and Viridivariegata. Stomatal length increased gradually from the second to the fifth phenological stage. Rotundifolia reached the highest mean stomatal length whereas Aurea Pendula and Cristata had the lowest values. Stomatal density for all cultivars decreased from the second to the fifth stage. Aurea Pendula reached the highest stomatal density in all phenological stages. The highest values of variable to maximum fluorescence ratio (Fv/Fm) were recorded in Rotundifolia, Rohanii, and the wild type, whereas Viridivariegata showed the lowest Fv/Fm. Similar trend was found in maximum to initial fluorescence ratio (Fm/F0), but extremely low Fm/F0 values were recorded in Viridivariegata in the last phenological stage. The highest potential electron capacity was found in Rohanii, Viridivariegata and the wild type and lowest in Cristata. This parameter increased in the course of early leaf development.

Due to anthropogenic influences, solar UV-B irradiance at the earth’s surface is increasing. To determine the effects of enhanced UV-B radiation on photosynthetic characteristics of Prunus dulcis, two-year-old seedlings of the species were submitted to four levels of UV-B stress, namely 0 (UV-Bc), 4.42 (UV-B1), 7.32 (UV-B2) and 9.36 (UV-B3) kJ m−2 d−1. Effects of UV-B stress on a range of chlorophyll (Chl) fluorescence parameters (FPs), Chl contents and photosynthetic gas-exchange parameters were investigated. UV-B stress promoted an increase in minimal fluorescence of dark-adapted state (F0) and F0/Fm, and a decrease in variable fluorescence (Fv, Fv/Fm, Fv/F0 and F0/Fm) due to its adverse effects on photosystem II (PSII) activity. No significant change was observed for maximal fluorescence of dark-adapted state (Fm). Enhanced UV-B radiation caused a significant inhibition of net photosynthetic rate (P N) at UV-B2 and UV-B3 levels and this was accompanied by a reduction in stomatal conductance (g s) and transpiration rate (E). The contents of Chl a, b, and total Chl content (a+b) were also significantly reduced at increased UV-B stress. In general, adverse UV-B effects became significant at the highest tested radiation dose 9.36 kJ m−2 d−1. The most sensitive indicators for UV-B stress were Fv/F0, Chl a content and P N. Significant P<0.05 alteration in these parameters was found indicating the drastic effect of UV-B radiation on P. dulcis.

Based on PAM and PEA measurements of fluorescence of bark chloroplasts, we have compared the information capacity of the methods for assessing the physiological state of Tilia cordata Mill. by the maximal quantum efficiency of PS II photochemistry (F v/F m) and by the performance index (PI). The measurements were made on annual shoots of linden trees growing in different environs. It was shown that the chlorophyll content in the bark of shoots growing near a busy urban street was twice less than in controls growing out of town. For the trees in the unfavorable environment, a small decrease in (F v/F m) was registered, and there was a significant statistical scatter in these values as compared with controls. The PI and its constituent parameters calculated from fluorescence induction curves (PEA method) are more informative and allow recognizing changes in the primary energy conversion processes in PS II when they are still small. Thus, PI can be used as a sensitive, robust, and rapid test to evaluate the physiological state of trees and other plant objects even under minor environmental changes.

Adult trees of Quercus petraea were submitted to controlled water shortage in a natural stand near Nancy, France. Diurnal course of net CO2 assimilation rate (A) was measured in situ together with chlorophyll a fluorescence determined on dark adapted leaves. In 1990, trees experienced a strong water stress, with predawn and midday leaf water potentials below –2·0 and –3·0 MPa, respectively. Diurnal course of A of well-watered trees exhibited sometimes important midday decreases in A related to high temperature and vapour pressure deficit. Decreases in initial (Fo) and maximal (Fm) fluorescence and sometimes in photochemical efficiency of photosystem II (Fv/Fm) were observed and probably revealed the onset of mechanisms for thermal de-excitation. These mechanisms were shown to be sensitive to dithiothreitol. All these effects were reversible and vanished almost completely overnight. Therefore, they may be considered as protective mechanisms adjusting activity of photosystem II to the electron requirement for photosynthesis. Water stress amplified these reactions: A was strongly decreased, showing important midday depression; diurnal reductions in Fm and Fv/Fm were enhanced. The same trends were observed during summer 1991, despite a less marked drought. These protective mechanisms seemed very effective, as no photoinhibitory damage to PS II could be detected in either water stressed or control trees.

Photosynthetic rates of green leaves (GL) and green flower petals (GFP) of the CAM plant Dendrobium cv. Burana Jade and their sensitivities to different growth irradiances were studied in shade-grown plants over a period of 4 weeks. Maximal photosynthetic O2 evolution rates and CAM acidities [dawn/dusk fluctuations in titratable acidity] were higher in leaves exposed to intermediate sunlight [a maximal photosynthetic photon flux density (PPFD) of 500–600 μmol m−2 s−1] than in leaves grown under full sunlight (a maximal PPFD of 1 000–1 200 μmol m−2 s−1) and shade (a maximal PPFD of 200–250 μmol m−2 s−1). However, these two parameters of GFP were highest in plants grown under the shade and lowest in full sun-grown plants. Both GL and GFP of plants exposed to full sunlight had lower predawn Fv/Fm [dark adapted ratio of variable to maximal fluorescence (the maximal photosystem 2 yield without actinic irradiation)] than those of shade-grown plants. When exposed to intermediate sunlight, however, there were no significant changes in predawn Fv/Fm in GL whereas a significant decrease in predawn Fv/Fm was found in GFP of the same plant. GFP exposed to full sunlight exhibited a greater decrease in predawn Fv/Fm compared to those exposed to intermediate sunlight. The patterns of changes in total chlorophyll (Chl) content of GL and GFP were similar to those of Fv/Fm. Although midday Fv/Fm fluctuated with prevailing irradiance, changes of midday Fv/Fm after exposure to different growth irradiances were similar to those of predawn Fv/Fm in both GL and GFP. The decreases in predawn and midday Fv/Fm were much more pronounced in GFP than in GL under full sunlight, indicating greater sensitivity in GFP to high irradiance (HI). In the laboratory, electron transport rate and photochemical and non-photochemical quenching of Chl fluorescence were also determined under different irradiances. All results indicated that GFP are more susceptible to HI than GL. Although the GFP of Dendrobium cv. Burana Jade require a lower amount of radiant energy for photosynthesis and this plant is usually grown in the shade, is not necessarily a shade plant.

The possibility to improve the recovery of sugar beet plants after water stress by application of synthetic cytokinins N6-benzyladenine (BA) or N6-(m-hydroxybenzyl)adenosine (HBA) was tested. Relative water content (RWC), net photosynthetic rate (PN), transpiration rate (E), stomatal conductance (gs), chlorophyll (Chl) a and Chl b contents, and photosystem 2 efficiency characterized by variable to maximal fluorescence ratio (Fv/Fm) were measured in control plants, in water-stressed plants, and after rehydration (4, 8, 24, and 48 h). Water stress markedly decreased parameters of gas exchange, but they started to recover soon after irrigation. Application of BA or HBA to the substrate or sprayed on leaves only slightly stimulated recovery of PN, E, and gs in rehydrated plants, especially during the first phases of recovery. Chl contents decreased only under severe water stress and Fv/Fm ratio was not significantly affected by water stress applied. Positive effects of BA or HBA application on Chl content and Fv/Fm ratio were mostly not observed.

In a previous study, we characterized a high chlorophyll fluorescence Ipal mutant of Arabidopsis thallana, in which approximately 20% photosystem (PS) Ⅱ protein is accumulated. In the present study, analysis of fluorescence decay kinetics and thermoluminescence profiles demonstrated that the electron transfer reaction on either the donor or acceptor side of PSII remained largely unaffected in the Ipa1 mutant. In the mutant, maximal photochemical efficiency (Fv/Fm, where Fm is the maximum fluorescence yield and Fv is variable fluorescence) decreased with increasing light intensity and remained almost unchanged in wildtype plants under different light conditions. The Fv/Fm values also increased when mutant plants were transferred from standard growth light to low light conditions. Analysis of PSll protein accumulation further confirmed that the amount of PSll reaction center protein is correlated with changes in Fv/Fm in Ipal plants. Thus, the assembled PSll in the mutant was functional and also showed increased photosensitivity compared with wild-type plants.

Field experiments were conducted in Sicily (south Italy) to assess chlorophyll (Chl) fluorescence parameters in response of potato crop to nitrogen dose, to variation in genotype and in plant age, and to detect relationships between Chl content, fluorescence parameter Fv/Fm, and tuber yield. The experiment included five nitrogen doses (0, 10, 20, 30, and 40 g m−2) and four genotypes (Spunta, Sieglinde, Daytona, and Igea). Chl fluorescence parameters (initial fluorescence, F0, maximum fluorescence, Fm, variable fluorescence, Fv, Fv/Fm, Tmax (the time required to reach Fm), and Chl content were measured weekly between the appearance of the fifth and sixth leaves and the onset of plant senescence. A positive linear relationship was established between nitrogen supply and Chl content, F0, and Tmax. Nitrogen supply up to 10 g m−2 also had a positive effect on Fm and Fv, but above this rate it reduced Fv/Fm. Spunta had the highest Chl content, Fm, Fv, and Fv/Fm, but the lowest F0, whereas Sieglinde had the lowest Chl content, Fv, Fv/Fm, and Tmax and the highest F0. The cvs. Igea and Daytona exhibited intermediate Chl fluorescence parameters. Chl content and Tmax decreased with increasing plant age, whereas F0, Fm, and Fv increased until complete canopy development and thereafter declined until crop maturity. Tuber and plant dry matter yield were significantly correlated with Chl content, F0, and Tmax. Thus Chl fluorescence and content detect differences in the response of potato to N supply, can discriminate between genotypes, predict plant age, and yield performance under field conditions.

High temperature generally constrains plant growth and photosynthesis in many regions of the world; however, little is known about how photosynthesis responds to high temperature with regard to different leaf ages. The synchronous changes in gas exchange and chlorophyll fluorescence at three leaf age levels (just fully expanded, mature, and older leaves) of maize (Zea mays L.) were determined at three temperatures (30°C as a control and 36 and 42°C as the higher temperatures). High temperature significantly decreased the net CO2 assimilation rate (A), stomatal conductance (g s), maximal efficiency of photosystem II (PSII) photochemistry (F v/F m), efficiency of excitation energy capture by open PSII reaction centers ( F\textv /F\textm F^{\prime}_{\text{v}} /F^{\prime}_{\text{m}} ), photochemical quenching of variable chlorophyll fluorescence (q P), and the electron transport rate (ETR), whereas minimal fluorescence yield (F 0) and nonphotochemical quenching of variable chlorophyll fluorescence (q N) were increased. The youngest fully expanded leaves had higher A, ETR, and q P compared with older leaves. Higher temperature with old leaves led to significant malondialdehyde (MDA) accumulation, a proxy for lipid peroxidation damage from active oxygen species (AOS). MDA content was significantly negatively correlated with A, F v/F m, F\textv /F\textm F^{\prime}_{\text{v}} /F^{\prime}_{\text{m}} , and q P. Thus, the results suggest that photosynthetic potentials, including stomatal regulation and PSII activity, may be restricted at high temperature, together with increasing cell peroxidation, which may be closely associated with leaf age.

Chronic photoinhibition in seedlings of tropical trees
Seedlings of five canopy species of tropical trees from Costa Rica and Puerto Rico were grown in full shade (midday range of photosynthetic photon flux density [PPFD], 100–140 μmol m?2 s?1), partial shade (midday PPFD, 400–600 μmol m?2 s?1) and full sun (midday PPFD, 1 500–1 800 μmol m?2 s?1) for 3 months. The species were Ochroma lagopus (Bombacaceae), a pioneer species; Inga edulis (Fabaceae), found in secondary forest; and Dipteryx panamensis (Fabaceae), Hampea appendiculata (Malvaceae), and Manilkara bidentata (Sapotaceae), three species characteristic of primary forest. After the plants were placed in the dark overnight, chlorophyll fluorescence characteristics were measured for recently expanded and mature leaves. The ratio of variable fluorescence to maximum fluorescence (Fv/Fm) was used to estimate the degree of chronic photoinhibition. Only individuals of one species, Dipteryx panamensis, showed significant depression of Fv/Fm after long-term exposure to full sun. The depression was highly correlated with quantum yield of O2 evolution which also declined after exposure to full sun. The decline may have been related to foliar N concentration. Although all plants were supplied with ample nutrients, foliar N did not increase significantly for Dipteryx seedlings in full sun, whereas it did for Ochroma and Inga. Leaf age affected Fv/Fm only in the cases of Manilkara, where it was slightly lower in recently expanded leaves, and of Dipteryx where it interacted with the effects of light regime. We conclude that chronic photoinhibition is not common in seedlings of canopy trees of tropical rain forests except when availability of mineral nutrients may be limiting.

In biological oceanography, it has been widely accepted that the maximum quantum yield of photosynthesis is influenced by nutrient stress. A closely related parameter, the maximum quantum yield for stable charge separation of PSII, (φ PSII )m, can be estimated by measuring the increase in fluorescence yield from dark-adapted minimal fluorescence (Fo) to maximal fluorescence (Fm) associated with the closing of photosynthetic reaction centers with saturating light or with a photosynthetic inhibitor such as 3′-(3,4-dichlorophenyl)-1′,1′-dimethyl urea (DCMU). The ratio Fv/Fm (= (Fm− Fo)/Fm) is thus used as a diagnostic of nutrient stress. Published results indicate that Fv/Fm is depressed for nutrient-stressed phytoplankton, both during nutrient starvation (unbalanced growth) and acclimated nutrient limitation (steady-state or balanced growth). In contrast to published results, fluorescence measurements from our laboratory indicate that Fv/Fm is high and insensitive to nutrient limitation for cultures in steady state under a wide range of relative growth rates and irradiance levels. This discrepancy between results could be attributed to differences in measurement systems or to differences in growth conditions. To resolve the uncertainty about Fv/Fm as a diagnostic of nutrient stress, we grew the neritic diatom Thalassiosira pseudonana (Hustedt) Hasle et Heimdal under nutrient-replete and nutrient-stressed conditions, using replicate semicontinuous, batch, and continuous cultures. Fv/Fm was determined using a conventional fluorometer and DCMU and with a pulse amplitude modulated (PAM) fluorometer. Reduction of excitation irradiance in the conventional fluorometer eliminated overestimation of Fo in the DCMU methodology for cultures grown at lower light levels, and for a large range of growth conditions there was a strong correlation between the measurements of Fv/Fm with DCMU and PAM (r2 = 0.77, n = 460). Consistent with the literature, nutrient-replete cultures showed consistently high Fv/Fm (∼0.65), independent of growth irradiance. Under nutrient-starved (batch culture and perturbed steady state) conditions, Fv/Fm was significantly correlated to time without the limiting nutrient and to nutrient-limited growth rate before starvation. In contrast to published results, our continuous culture experiments showed that Fv/Fm was not a good measure of nutrient limitation under balanced growth conditions and remained constant (∼0.65) and independent of nutrient-limited growth rate under different irradiance levels. Because variable fluorescence can only be used as a diagnostic for nutrient-starved unbalanced growth conditions, a robust measure of nutrient stressed oceanic waters is still required.

Experimental ecology methods and chlorophyll fluorescence technology were used to study the effects of different concentrations of manganese (10−12– 10−4 mol L−1) on the growth, photosystem II and superoxide dismutase (SOD) activity of Amphidinium sp. MACC/D31. The results showed that manganese had a significant effect on the growth rate, fluorescence parameters (maximal photochemical efficiency of PSII (F v /F m ), photochemical quenching (qP) and non-photochemical quenching (NPQ)) in the exponential stage (days 1–3) and SOD activity of Amphidinium sp. (P < 0.05). F v/F m in the exponential stage in 10−12 mol L−1 manganese concentration was significantly lower whilst qP and NPQ significantly higher than those in the other concentrations. F v /F m (days 6–9) in 10−4 mol L−1 manganese was significantly higher than those in the other concentrations. F v /F m (days 3–6) increased with increased concentration of manganese from 10−12 to 10−4 mol L−1. The values of qP and NPQ decreased with decreased concentrations of manganese, except for those in days 4–6. F v /F m under each concentration increased earlier and decreased later with culture stage whilst NPQ decreased earlier and increased later. The SOD activity increased with increased concentration of manganese from 10−12 to 10−8 mol L−1. The SOD activity in 10−4 mol L−1 manganese was significantly higher than those in the other concentrations and in 10−12 mol L−1 manganese, it was significantly lower than those in the other concentrations.

We measured the responses of pigments and chlorophyll a fluorescence parameters of the Antarctic leafy liverwort Cephaloziella varians to snowmelt during austral spring 2005 at Rothera Point on the western Antarctic Peninsula. Although no changes to the concentrations of UV-B photoprotective pigments were detected during snowmelt, chlorophyll and carotenoid concentrations and maximum photosystem (PS)II yield (F v /F m) were respectively 88, 60 and 144% higher in the tissues of the liverwort that had recently emerged from snow than in those under a 10 cm depth of snow. A laboratory experiment similarly showed that effective PSII yield increased rapidly within the first 45 min after plants sampled from under snow were removed to an illuminated growth cabinet. The pigmentation and PSII yields of plants during snowmelt were also compared with those of plants in January, during the middle of the growing season at Rothera Point. During snowmelt, plants had lower F v /F m values, chlorophyll a/b ratios and concentrations of UV-B photoprotective pigments and carotenoids than during mid-season, suggesting that although there is some recovery of PSII activity and increases in concentrations of photosynthetic pigments during snowmelt, the metabolism of C. varians is restricted during this period.

Changes in pigment composition and chlorophyll (Chl) fluorescence parameters were studied in 20 year-old Scots pine (Pinus sylvestris L.) trees grown in environment-controlled chambers and subjected to ambient conditions (CON), doubled ambient CO2 concentration (EC), elevated temperature (ambient +2−6 °C, ET), or a combination of EC and ET (ECT) for four years. EC did not significantly alter the optimal photochemical efficiency of photosystem 2 (PS2; Fv/Fm), or Chl a+b content during the main growth season (days 150–240) but it reduced Fv/Fm and the Chl a+b content and increased the ratio of total carotenoids to Chl a+b during the ‘off season’. By contrast, ET significantly enhanced the efficiency of PS2 in terms of increases in Fv/Fm and Chl a+b content throughout the year, but with more pronounced enhancement in the ‘off season’. The reduction in Fv/Fm during autumn could be associated with the CO2-induced earlier yellowing of the leaves, whereas the temperature-stimulated increase in the photochemical efficiency of PS2 during the ‘off season’ could be attributed to the maintenance of a high sink capacity. The pigment and fluorescence responses in the case of ECT showed a similar pattern to that for ET, implying the importance of the temperature factor in future climate changes in the boreal zone. This revised version was published online in August 2006 with corrections to the Cover Date.

Summary Recovery from winter depression of photosynthesis was studied in Pinus sylvestris, Pinus conforta and Picea abies by means of chlorophyll fluorescence and gas exchange measurements. During the winter 1986–1987 the fluorescence yield was low and no variable fluorescence was detectable before the end of March. In the field recovery of variable fluorescence/maximum fluorescence (Fv/Fm) during spring was slow for all three species studied. The temperature dependence of recovery was confirmed from measurements of the potential rate of recovery of Fv/Fm at different temperatures in the laboratory. At 20° C, Fv/Fm increased from 0.1 to 0.8 within 3 days. Recovery of Fv/Fm was paralleled by an increase in apparent photon yield. No significant differences could be demonstrated between the studied tree species in potential rate of recovery in the laboratory or in actual recovery in the field.

A portable instrument for measuring chlorophyll fluorescence induction kinetics is described and examples of measurements are given. The instrument is centered around a statistically-mixed bifurcated optical fiber. One fiber branch guides the actinic light to the sample, whereas the other branch carries the emitted chlorophyll fluorescence to the photodetector. Scattered actinic light is cut out from the detector by a red interference filter. The instrument measures fast as well as slow fluorescence induction kinetics, but is particularly well designed for analyzing fast kinetics. The high time resolution and strong, variable actinic light mean that both Fo (non-variable fluorescence) and Fm (maximal fluorescence at the P-peak) are well defined. A built in microprocessor unit with attached memory stores the fluorescence induction curve and calculates key fluorescence parameters such as Fo, Fm, Fv (variable fluorescence equals Fm?Fo), Fv/Fm (the photochemical efficiency of photosystem II) and t1/2 (half rise time from Fo, to Fm). These values are digitally displayed after each recording and they (or the whole induction curve) can be stored in a memory and later retrieved. Because of a flexible setting of the instrument it can be used with high accuracy both for optically thick leaves and for diluted suspensions of algae or chloroplasts. A simple, light weight clamp cuvette for dark adaptation of leaves has been developed. It is equipped with a gate allowing the optical fiber to be inserted without daylight reaching the dark adapted portion of the leaf. The instrument has been developed for rapid monitoring of changes in activities and organization of the photosynthetic apparatus in vivo when plants are exposed to environmental stress both in the field and in the laboratory. Examples of measurements are given for differently treated leaves of Pinus sylvestris, Salix sp., Betula verrucosa, Zea mays, Epilobium angustifo-lium and for chloroplast thylakoids isolated from Spinacia oleracea.

Measurements of Sorbus stomata size and density, maximal photochemical efficiency of photosystem II (Fv/Fm), variable-to-initial fluorescence ratio (Fv/F0) and potential electron acceptor capacity (‘area’) were performed during leaf development in four parental diploid species, S. aria, S. aucuparia, S. chamaemespilus, S. torminalis, and two hybrid species, S. hazslinszkyana and S. intermedia. In fully expanded mature leaves, stomata lengths and densities were significantly larger in the shrub S. chamaemespilus than in the five tree species. The best performance of both the Fv/Fm and the Fv/F0 ratio was recorded in S. intermedia, whereas S. chamaemespilus had the highest value of ‘area’. From a physiological point of view, the results of this study showed that the photosystem II reaction centers remained intact functionally through all phenological stages of leaf expansion for all examined species of Sorbus.
