Four type-1 (single-chain) ribosome-inactivating proteins (RIPs), with isoelectric points between 9.5 and 9.7, were isolated from leaves of Phytolacca dioica L. The purification procedure furnished the four proteins with an overall yield of about 16% and separated them from a protein of 29 407 ± 2 Da, as determined by electrospray mass spectrometry, whose N-terminal amino acid sequence differed from that of pokeweed (Phytolacca americana L.) leaf chitinase (PLC-B) by only one amino acid (R17I). The four RIPs (PD-L1 to PD-L4) inhibited protein synthesis by a rabbit reticulocyte lysate with 50% inhibition at the picomolar level, and produced the β-fragment, diagnostic of the specific enzymatic action of RIPs, on yeast ribosomes. Comparison of their N-terminal sequences, up to residue 45, showed that PD-L1 is identical to PD-L2 [designated the isoleucine (Ile) form from the N-terminal residue] and PD-L3 is identical to PD-L4 [designated the valine (Val) form from the N-terminal residue] and that there are 35 identical residues between the two forms. Furthermore, the Val form presents the same number of identical residues as PD-S2, an RIP isolated from the seeds of the same plant. With the exception of PD-L4, the purified RIPs gave a positive reaction when stained for sugars on SDS-PAGE gels and, when analyzed by electrospray mass spectrometry, had Mr values of 32 715 ± 1 (PD-L1), 31 542 ± 1 (PD-L2), 30 356 ± 1 (PD-L3) and 29 185 ± 1 Da (PD-L4). The 1171 kDa difference in Mr, within the same RIP form, could be due to glycosylation. Like leaf saporins and many other RIPs, the four RIPs released several adenines from poly(A), herring sperm DNA and rRNA 16S + 23S, thus acting as polynucleotide:adenosine glycosidases. This property was less pronounced in PD-L1 and PD-L3 than in PD-L2 and PD-L4, respectively. The proteins PD-L1 and PD-L4 showed 3.7% reactivity with the antiserum anti-dianthin 32 and no reactivity with antisera to PAP-R saporin-S6, momordin I and even PD-S2, an RIP isolated from the seeds of the same plant. The influx of glucose into the brain and plasma glucose disappearance were estimated in rainbow trout (Oncorhynchus mykiss) intravenously injected (1 ml · kg−1 body weight) with a single dose (15 μCi · kg−1 body weight) of 3-O-methyl-D-[U-14C]glucose ([U-14C]-3-OMG) at different times (2–160 min), and after intravenous injection at 15 min of increased doses (10–60 μCi · kg−1 body weight) of [U-14C]-3-OMG. Brain and plasma radiotracer concentrations were measured, and several kinetic parameters were calculated. The apparent brain glucose influx showed a maximum after 15–20 min of injection then decreased to a plateau after 80 min. Brain distribution space of 3-OMG increased from 2 min to 20 min reaching equilibrium from that time onwards at a value of 0.14 ml · g−1. The unidirectional clearance of glucose from blood to brain (k1) and the fractional clearance of glucose from brain to blood (k2) were estimated to be 0.093 ml · min−1 · g−1, and 0.867 min−1, respectively. A linear increase was observed in brain and plasma radiotracer concentrations when increased doses of [U-14C]-3-OMG were used. All these findings support a facilitative transport of glucose through the blood-brain barrier of rainbow trout with characteristics similar to those observed in mammals. The injection of different doses of melatonin (0.25–1.0 mg · kg−1) significantly increased brain glucose influx suggesting a possible role for melatonin in the regulation of glucose transport into the brain. We used tritium-labeled water to measure total body water, water influx (which approximated oxidative water production) and water efflux in free-flying tippler pigeons (Columba livia) during flights that lasted on average 4.2 h. At experimental air temperatures ranging from 18 to 27 °C, mean water efflux by evaporation and excretion [6.3 ± 1.3 (SD) ml · h−1, n = 14] exceeded water influx from oxidative water and inspired air (1.4 ± 0.7 ml · h−1, n = 14), and the birds dehydrated at 4.9 ± 0.9 ml · h−1. This was not significantly different from gravimetrically measured mass loss of 6.2 ± 2.1 g · h−1 (t = 1.902, n = 14, P>0.05). This flight-induced dehydration resulted in an increase in plasma osmolality of 4.3 ± 3.0 mosmol · kg−1 · h−1 during flights of 3–4 h. At 27 °C, the increase in plasma osmolality above pre-flight levels (ΔP osm = 7.6±4.29 mosmol · kg−1 · h−1, n = 6) was significantly higher than that at 18 °C (ΔP osm = 0.83±2.23 mosmol · kg−1 · h−1, (t = 3.43, n = 6, P < 0.05). Post-flight haematocrit values were on average 1.1% lower than pre-flight levels, suggesting plasma expansion. Water efflux values during free flight were within 9% of those in the one published field study (Gessaman et al. 1991), and within the range of values for net water loss determined from mass balance during wind tunnel experiments (Biesel and Nachtigall 1987). Our net water loss rates were substantially higher than those estimated by a simulation model (Carmi et al. 1992) suggesting some re-evaluation of the model assumptions is required. The effect of ambient osmolality on the height of lateral ciliated cells from the gills of two freshwater bivalve species (Dreissena polymorpha, Toxolasma texasensis) was directly observed microscopically. The addition of 1 mmol · l−1 KCl to an artificial pondwater (APW) superfusion medium resulted in an increase in cell height. When the superfusion solution was made hyperosmotic (∼90 mmol · kg−1 H2O) by the addition of 45 mmol · l−1 NaCl to APW, the cell height decreased by about 20–30% and there was no evidence of a regulatory volume increase over 20–30 min. In contrast, when 1 mmol · l−1 KCl was added to the hyperosmotic medium the cell height always partially (40–50%) recovered. When the gill tissue was returned to APW following the hyperosmotic treatment the cells returned to the original cell height. Bivalve gills superfused with the hyperosmotic NaCl and KCl solution in the presence of 1 mmol · l−1 ouabain experienced a similar 25% decrease in cell height. When the ouabain-treated tissues were returned to APW the cells swelled, overshooting the original cell height. These results indicate these freshwater bivalves have a limited ability for cellular volume regulation using inorganic ions, but depend on a suitable balance of Na+ and K+ in the environment to effect regulatory volume changes. A laboratory study investigated the metabolic physiology, and response to variable periods of water and sodium supply, of two arid-zone rodents, the house mouse (Mus domesticus) and the Lakeland Downs short-tailed mouse (Leggadina lakedownensis) under controlled conditions. Fractional water fluxes for M. domesticus (24 ± 0.8%) were significantly higher than those of L. lakedownensis (17 ± 0.7%) when provided with food ad libitum. In addition, the amount of water produced by M. domesticus and by L. lakedownensis from metabolic processes (1.3 ± 0.4 ml · day−1 and 1.2 ± 0.4 ml · day−1, respectively) was insufficient to provide them with their minimum water requirement (1.4 ± 0.2 ml · day−1 and 2.0 ± 0.3 ml · day−1, respectively). For both species of rodent, evaporative water loss was lowest at 25 °C, but remained significantly higher in M. domesticus (1.1 ± 0.1 mg H2O · g−0.122 · h−1) than in L. lakedownensis (0.6 ± 0.1 mg H2O · g−0.122 · h−1). When deprived of drinking water, mice of both species initially lost body mass, but regained it within 18 days following an increase in the amount of seed consumed. Both species were capable of drinking water of variable saline concentrations up to 1 mol · l−1, and compensated for the increased sodium in the water by excreting more urine to remove the sodium. Basal metabolic rate was significantly higher in M. domesticus (3.3 ± 0.2 mg O2 · g−0.75 · h−1) than in L. lakedownensis (2.5 ± 0.1 mg O2 · g−0.75 · h−1). The study provides good evidence that water flux differences between M. domesticus and L. lakedownensis in the field are due to a requirement for more water in M. domesticus to meet their physiological and metabolic demands. Sodium fluxes were lower than those observed in free-ranging mice, whose relatively high sodium fluxes may reflect sodium associated with available food. Accepted: 16 August 1999  相似文献

Despite the availability of many mutants for signal transduction, Arabidopsis thaliana guard cells have so far not been used in electrophysiological research. Problems with the isolation of epidermal strips and the small size of A. thaliana guard cells were often prohibiting. In the present study these difficulties were overcome and guard cells were impaled with double-barreled microelectrodes. Membrane-potential recordings were often stable for over half an hour and voltage-clamp measurements could be conducted. The guard cells were found to exhibit two states. The majority of the guard cells had depolarized membrane potentials, which were largely dependent on external K+ concentrations. Other cells displayed spontaneous transitions to a more hyperpolarized state, at which the free-running membrane potential (Em) was not sensitive to the external K+ concentration. Two outward-rectifying conductances were identified in cells in the depolarized state. A slow outward-rectifying channel (s-ORC) had properties resembling the K+-selective ORC of Vicia faba guard cells (Blatt, 1988, J Membr Biol 102: 235–246). The activation and inactivation times and the activation potential, all depended on the reversal potential (Erev) of the s-ORC conductance. The s-ORC was blocked by Ba2+ (K1/2 = 0.3–1.3mM) and verapamil (K1/2 = 15–20 μM). A second rapid outward-rectifying conductance (r-ORC) activated instantaneously upon stepping the voltage to positive values and was stimulated by Ba2+. Inward-rectifying channels (IRC) were only observed in cells in the hyperpolarized state. The activation time and activation potential of this channel were not sensitive to the external K+ concentration. The slow activation of the IRC (t1/2 ≈ 0.5 s) and its negative activation potential (Vthreshold = −155 mV) resemble the values found for the KAT1 channel expressed in Saccharomyces cerevisiae (Bertl et al., 1995, Proc Natl Acad Sci USA 92: 2701–2705). Drinking in Atlantic salmon (Salmo salar) juveniles was investigated in fresh water and following transfer to sea water. There was a significant effect of fish size on drinking, and smolts (20–30 g) imbibed about ten times less water than alevins of 0.2–0.3 g. Freshwater smolts drank at a rate of 0.15 ± 0.03 ml · kg−1 · h−1 and administration of doses of 10 or 20 mg · kg−1 of papaverine (stimulator of the renin- angiotensin system RAS) or [Asn1, Val5]-Angiotensin II (0.4 μmol · kg−1) resulted in significant increases in drinking, while administration of the angiotensin converting enzyme inhibitor, enalapril (50 mg · kg−1) had no effect on drinking. Transfer of Atlantic salmon smolts to 1/3, 2/3 and full strength sea water resulted in significant increases in drinking to 1.06 ± 0.12, 1.24 ± 0.0.16 and 3.89 ± 0.28 ml · kg−1 · h−1, respectively. In sea water, stimulation of the endogenous RAS by administration of papaverine (20 mg · kg−1) resulted in a 20% increase in drinking, while administration of enalapril to doses of 50 and 200 mg · kg−1 lowered drinking to 1.99 ± 0.48 and 0.32 ± 0.06 ml · kg−1 · h−1, respectively. All treatments were without effect on blood plasma levels of Na+ and Cl in fresh water, while in sea water smolts both stimulation and inhibition of drinking resulted in hemoconcentration of Na+ and Cl. A mixed microbial culture was immobilized by entrapment into silica gel (SG) and entrapment/ adsorption on polyurethane foam (PU) and ceramic foam. The phenol degradation performance of the SG biocatalyst was studied in a packed-bed reactor (PBR), packed-bed reactor with ceramic foam (PBRC) and fluidized-bed reactor (FBR). In continuous experiments the maximum degradation rate of phenol (q s max) decreased in the order: PBRC (598 mg l−1 h−1) > PBR (PU, 471 mg l−1 h−1) > PBR (SG, 394 mg l−1 h−1) > FBR (PU, 161 mg l−1 h−1) > FBR (SG, 91 mg l−1 h−1). The long-term use of the SG biocatalyst in continuous phenol degradation resulted in the formation of a 100–200 μm thick layer with a high cell density on the surface of the gel particles. The abrasion of the surface layer in the FBR contributed to the poor degradation performance of this reactor configuration. Coating the ceramic foam with a layer of cells immobilized in colloidal SiO2 enhanced the phenol degradation efficiency during the first 3 days of the PBRC operation, in comparison with untreated ceramic packing. We measured the osmotic water permeability (P os) of vacuoles isolated from onion (Allium cepa L.), rape (Brassica napus L.), petunia (Petunia hybrida Hook.) and red beet (Beta vulgaris L.). For all the vacuolar types investigated, P os values were in the range 200–1000 μm s−1. The change in membrane surface area induced by an osmotic gradient was smaller than 2–6%. The vacuolar P os values for red beet and onion were reduced by 1 mM HgCl2, to 14% and 30% of the control values, respectively, but were partially restored to 51% and 76% by 5 mM β-mercaptoethanol. These results suggest that aquaporins were present in all the vacuoles tested. In HgCl2-treated onion vacuoles, the reduced P os (56 μm s−1) had a low activation energy (approx. 6 kJ mol−1), indicating that water permeation was still occurring mainly via aquaporins, and that the water permeability of the lipid part of the vacuolar membrane is probably very low. Whereas with advancing age, peak heart rate (HR) and cardiac index (CI) are clearly reduced, peak stroke index (SI) may decrease, remain constant or even increase. The aim of this study was to describe the patterns of HR, SI, CI, arteriovenous difference in oxygen concentration (C a-vO2), mean arterial pressure (MAP), systemic vascular resistance index (SVRI), stroke work index (SWI) and mean systolic ejection rate index (MSERI) in two age groups (A: 20–30 years, n = 20; B: 50–60 years n = 20. After determination of pulmonary function, an incremental bicycle exercise test was performed, with standard gas-exchange measurements and SI assessment using electrical impedance cardiography. The following age-related changes were found: similar submaximal HR response to exercise in both groups and a higher peak HR in A than in B[185 (SD 9) vs 167 (SD 14) beats · min−1, P < 0.0005]; increase in SI with exercise up to 60–90 W and subsequent stabilization in both groups. As SI decreased towards the end of exercise in B, a higher peak SI was found in A [57.5 (SD 14.0) vs 43.6 (SD 7.7) ml · m−2, P < 0.0005]; similar submaximal CI response to exercise, higher peak CI in A [10.6 (SD 2.5) vs 7.2 (SD 1.3) l · min−1 · m−2, P < 0.0005]; no differences in C a-vO2 during exercise; higher MAP at all levels of exercise in B; higher SVRI at all levels of exercise in B; lower SWI in B after recovery; higher MSERI at all levels of exercise in A. The decrease in SI with advancing age would seem to be related to a decrease in myocardial contractility, which can no longer be compensated for by an increase in preload (as during submaximal exercise). Isolated perfused gills of stenohaline crabs Cancer pagurus adapted to seawater, brackish water-adapted euryhaline shore crabs Carcinus maenas and freshwater-adapted extremely euryhaline Chinese crabs Eriocheir sinensis were tested for their capacity to excrete ammonia. Gills were perfused with haemolymph-like salines and bathed with salines equal in adaptation osmolality. Applying 100 μmol · l−1 NH4Cl in the perfusion saline and concentrations of NH4Cl in the bath that were stepwise increased from 0 to 4000 μmol · l−1 allowed us to measure transbranchial fluxes of ammonia along an outwardly as well as various inwardly directed gradients. The gills of all three crab species were capable – to different extents – of active excretion of ammonia against an inwardly directed gradient. Of the three crab species, the gills of Cancer pagurus revealed the highest capacity for active excretion of ammonia, being able to excrete it from the haemolymph (100 μmol · l−1 NH+ 4) through the gill epithelium against ambient concentrations of up to 800 μmol · l−1, i.e. against an eightfold gradient. Carcinus maenas and E. sinensis were able to actively excrete ammonia against approximately fourfold gradients. Within the three crab species, the gills of E. sinensis exhibited the greatest capacity to resist influx at very high external concentrations of up to 4000 μmol · l−1. We consider the observed capacities for excretion of ammonia against the gradient as ecologically meaningful. These benthic crustaceans protect themselves by burying themselves in the sediment, where, in contrast to the water column, concentrations of ammonia have previously been reported that greatly increase haemolymph levels. Electrophysiological results indicate that the permeabilities of the gill epithelia are a clue to understanding the species-specific differences in active excretion of ammonia. During the invasion of brackish water and freshwater, the permeabilities of the body surfaces greatly decreased. The gills of marine Cancer pagurus exibited the greatest permeability (ca. 250 mS cm−2), thus representing practically no influx barrier for ions including NH+ 4. We therefore assume that C. pagurus had to develop the strongest mechanism of active excretion of ammonia to counteract influx. On the other hand, freshwater-adapted E. sinensis exhibited the lowest ion permeability (ca. 4 mS cm−2) which may reduce passive NH+ 4 influxes at high ambient levels. Our objective was to quantify the potential variability in remotely sensed chlorophyll a (Chl a) and primary productivity in coastal waters of the Southern Ocean. From data collected throughout the springs/summers of 1991–1994, we calculated the proportion of water column Chl a and primary productivity within the upper optical attenuation length (K−1 par) and the satellite-weighted depth. The temporal variability was resolved every 2–3 days and was observed to be greater within years than between years. Three-year averages (n=223) revealed that 10.2 ± 3.6% of total Chl a and 14.8 ± 6.5% of production occurred within satellite-weighted depth in predominantly Case I waters. The average values were twice as high within K−1 par, 24.1 ± 8% of total Chl a and 34 ± 9% of production respectively. Masked in these long-term averages are very large changes occurring on short time scales of seasonal blooms. We observed that the patterns of Chl a vertical distribution within blooms are also subject to taxonomic influence and dependent upon the physiological state of the phytoplankton. Highest proportions of water column Chl a in the first optical depth were measured during the rapid onset of surface cryptophyte blooms each year, i.e. 50% within K−1 par and 30% above the satellite-weighted depth. Lowest fractions, 6% and 2% of biomass within K−1 par and satellite-weighted depth respectively, were associated with peak bloom conditions independent of taxonomy. Our analyses suggest that satellite-dependent models of Chl a and subsequent chlorophyll-dependent primary production will be challenging to develop for the near-shore Southern Ocean, especially given the potentially high natural variability in the vertical distribution of Chl a driven by physical forcing, the photoadaptive abilities of polar phytoplankton, and taxonomic influences. 1,3-Propanediol inhibition during glycerol fermentation to 1,3-propanediol by Clostridium butyricum CNCM 1211 has been studied. The initial concentration of the 1,3-propanediol affected the growth of the bacterium more than the glycerol fermentation. μ max was inversely proportional to the initial concentration of 1,3-propanediol (0–65 g l−1). For glycerol at 20 g l−1, the growth and fermentation were completely stopped at an initial 1,3-propanediol concentration of 65 g l−1. However, for an initial 1,3-propanediol concentration of 50 g l−1 and glycerol at 70 g l−1, the final concentration (initial and produced) of 1,3-propanediol reached 83.7 g l−1(1.1 M), with complete consumption of the glycerol. Therefore, during the fermentation, the strain tolerated a 1,3-propanediol concentration higher than the initial inhibitory concentration (65 g l−1). The addition of 1,2-propanediol or 2,3-butanediol (50 g l−1) in the presence of glycerol (50–100 g l−1), showed that 2-diols reduced the μ max in a similar way to 1,3-propanediol. The measurement of the osmotic pressure of glycerol solutions, diols and diol/glycerol mixtures did not indicate any differences between these compounds. The hypothesis of diol inhibition was discussed. Taking into account the strain tolerance of highly concentrated 1,3-propanediol during fermentation, the fermentation processes for optimising production were considered. The influence of (NH4)2SO4 concentration and dilution rate (D) on actual and potential H2 photoproduction has been studied in ammonium-limited chemostat cultures of Rhodobacter capsulatus B10. The actual H2 production in a photobioreactor was maximal (approx. 80 ml h−1 l−1) at D = 0.06 h−1 and 4 mM (NH4)2SO4. However, it was lower than the potential H2 evolution (calculated from hydrogen evolution rates in incubation vials), which amounted to 100–120 ml h−1 l−1 at D = 0.03–0.08 h−1. Taking into account the fact that H2 production in the photobioreactor under these conditions was not limited by light or lactate, another limiting (inhibiting) factor should be sought. One possibility is an inhibition of H2 production by the H2 accumulated in the gas phase. This is apparent from the non-linear kinetics of H2 evolution in the vials or from its inhibition by the addition of H2; initial rates were restored in both cases after the vials had been refilled with argon. The actual H2 production in the photobioreactor at D = 0.06 h−1 was shown to increase from approximately 80 ml h−1 l−1 to approximately 100 ml h−1 l−1 under an argon flow at 100 ml min−1. Under maximal H2 production rates in the photobioreactor, up to 30% of the lactate feedstock was utilised for H2 production and 50% for biomass synthesis. Sophorolipids, obtained by a two-stage process starting from deproteinized whey concentrate using Cryptococcus curvatus ATCC 20509 and Candida bombicola ATCC 22214, were compared to products from one-stage processes, using different lipidic compounds as substrates. Results showed that above all carbon source and not cultivation conditions had a distinct influence on the composition of the crude product mixture and therefore on the physicochemical and biological properties of the sophorolipids, such as, for example, surface activity, cytotoxicity and stability against hydrolases. The results were completed by corresponding data for purified mono- and diacetylated (17-hydroxyoctadecenoic)-1′,4′′-lactonized sophorolipids. Crude sophorolipid mixtures showed moderate to good surface active properties (SFTmin 39 mN m−1, CMC 130 mg l−1), water solubilities (2–3 g l−1) and low cytotoxicities (LC50 300–700 mg l−1). In contrast, purified sophorolipids were more surface active (SFTmin 36 mN m−1, CMC 10 mg l−1), less water soluble (max. 70 mg l−1) and showed stronger cytotoxic effects (LC50 15 mg l−1). Incubation of crude sophorolipid mixtures with different hydrolases demonstrated that treatment with commercially available lipases such as from Candida rugosa and Mucor miehei distinctly reduced the surface active properties of the sophorolipids, while treatment with porcine liver esterase and glycosidases had no effect. In caulonemal filaments of the moss, Physcomitrella patens, which had been incubated in darkness, 3 s irradiation with blue light (λmax 450 nm) at fluence rates of 100 μmol m−2 s−1 and above caused a transient␣increase in cytosolic calcium ion concentration, [Ca2+]cyt, which was both intensity- and time-dependent. Measurements of [Ca2+]cyt were made using moss transformed with the cDNA for apoaequorin and reconstituting the Ca2+-dependent photoprotein aequorin in the cytosol by incubation in coelenterazine.␣In response to blue light at fluence rates of 100–1000 μmol photons m−2 s−1, [Ca2+]cyt increased transiently from a basal level of approximately 50 nM to between 200 and 700 nM. Irradiation with red light did not evoke any measurable change in [Ca2+]cyt. The presence of calcium in the incubating medium was not required for the increase in [Ca2+]cyt to occur. A mutant strain, gad-139, was identified which required an irradiance of only 1 s to evoke a response. Unidirectional flux rates of Ca2+ across gastrointestinal tissues from sheep and goats were measured in vitro by applying the Ussing-chamber technique. Except for the sheep duodenum, mucosal to serosal Ca2+ flux rates (J ms) exceeded respective flux rates in the opposite direction (J sm) in both species and in all segments of the intestinal tract. This resulted in net Ca2+ flux rates␣(J net = J ms − J sm) ranging between −2 and 9 nmol · cm−2 · h−1 in sheep and between 10 and 15 nmol cm−2 · h−1 in goats. In sheep, only J net in jejunum, and in goats, J netin duodenum and jejunum were significantly different from zero. Using sheep rumen wall epithelia, significant J net of Ca2+ of around 5 nmol · cm−2 · h−1 could be detected. Since the experiments were carried out in the absence of an electrochemical gradient, significant net Ca2+ absorption clearly indicates the presence of active mechanisms for Ca2+ transport. Dietary Ca depletion caused increased calcitriol plasma concentrations and induced significant stimulations of net Ca2+ absorption in goat rumen. J net of Ca2+ across goat rumen epithelia was significantly reduced by 1 mmol · l −1 verapamil in the mucosal buffer solution. In conclusion, there is clear evidence for the rumen as a main site for active Ca2+ absorption in small ruminants. Stimulation of active Ca2+ absorption by increased plasma calcitriol levels and inhibition by mucosal verapamil suggest mechanistic and regulatory similarities to active Ca2+ transport as described for the upper small intestines of monogastric species. Isochorismate hydroxymutase (i.e. isochorismate synthase, EC 5.4.99.6) was purified from an anthraquinone-producing cell-suspension culture of Galium mollugo L. Although attempts to stabilize the labile enzyme met with little success, a substantial increase in enzyme activity was observed in the presence of glycine betaine (500 mM). Column chromatography on solid supports other than diethylaminoethyl (DEAE)-Sephacel, Phenylsepharose Cl-4B or Cibacron Blue 3G-A did not give active enzyme preparations. In spite of these drawbacks the enzyme was purified 573-fold. Enzyme activity depended strictly on the presence of Mg2+. Kinetic data for chorismate in the forward reaction (K m = 807 μM, V max = 6.2 pkat · mg−1) and for isochorismate in the reverse reaction (K m = 675 μM, V max = 5.9 pkat · mg−1) were determined. Castor bean (Ricinus communis L.) plants were grown for 5–7 weeks in a controlled environment at 350 μl l−1 or 700 μl l−1 CO2. Carbon assimilation, assimilate deposition, dark respiration and assimilate mobilization were measured in leaves 2, 3 and 4 (counted from the base of the plant), and a balance sheet of carbon input and export was elaborated for both CO2 concentrations. Carbon dioxide assimilation was nearly constant over the illumination period, with only a slight depression occurring at the end of the day in mature source leaves, not in young source leaves. Assimilation was ca. 40% higher at 700 μl l−1 than at 350 μl l−1 CO2. The source leaves increased steadily in weight per unit area during the first 3 weeks, more at 700 μl l−1 than at 350 μl l−1 CO2. On top of an irreversible weight increase, there was a large gain in dry weight during the day, which was reversed during the night. This reversible weight gain was constant over the life time of the leaf and ca. 80% higher at 700 μl l−1 than at 350 μl l−1. Most of it was due to carbohydrates. The carbon content (as a percentage) was not altered by the CO2 treatment. Respiration was 25% higher in high-CO2 plants when based on leaf area, but the same when based on dry weight. The rate of carbon export via the phloem was the same during the daytime in plants grown at 350 μl l−1 and 700 μl l−1 CO2. During the night the low-CO2 plants had only 50% of the daytime export rate, in contrast to the high-CO2 plants which maintained the high export rate. It was concluded that the phloem loading system is saturated during the daytime in both CO2 regimes, whereas during the night the assimilate supply is reduced in plants in the normal CO2 concentration. Two-thirds of the carbon exported from the leaves was permanently incorporated as plant dry matter in the residual plant parts. This “assimilation efficiency” was the same for both CO2 regimes. It is speculated that under 350 μl l−1 CO2 the growing Ricinus plant operates at sink limitation during the day and at source limitation during the night. Genes encoding the mosquitocidal binary toxin of Bacillus sphaericus 2362 were introduced into Synechococcus PCC6301, a cyanobacterium that can tolerate a number of potential variations in the mosquito breeding environment, and can serve as a food source for mosquito larvae. The toxin genes, preceded by a Synechococcus rbcL promoter, were located on a mobilizable Escherichia coli Synechococcus shuttle vector, which was introduced into Synechococcus PCC6301 at frequencies of 10−5–10−7 exconjugants/recipient, depending on the selective conditions used. Recombinant Synechococcus exhibited significant toxicity against 2-day-old and 6-day-old Culex quinquefasciatus larvae, the concentration required to kill 50 % of larvae (LC50) being 2.1 × 105 and 1.3 × 105 cells/ml respectively.
