In certain toxicological experiments with laboratory animals, the outcome of interest is the occurrence of dead or malformed fetuses in a litter. Previous investigations have shown that the simple one-parameter binomial and Poisson models generally provide poor fits to this type of binary data. In this paper, a type of correlated binomial model is proposed for use in this situation. First, the model is described in detail and is compared to a beta-binomial model proposed by Williams (1975). These two-parameter models are then contrasted for goodness of fit to some real-life data. Finally, numerical examples are given in which likelihood ratio tests based on these models are employed to assess the significance of treatment-control differences.  相似文献

When faced with proportion data that exhibit extra-binomial variation, data analysts often consider the beta-binomial distribution as an alternative model to the more common binomial distribution. A typical example occurs in toxicological experiments with laboratory animals, where binary observations on fetuses within a litter are often correlated with each other. In such instances, it may be of interest to test for the goodness of fit of the beta-binomial model; this effort is complicated, however, when there is large variability among the litter sizes. We investigate a recent goodness-of-fit test proposed by Brooks et al. (1997, Biometrics 53, 1097-1115) but find that it lacks the ability to distinguish between the beta-binomial model and some severely non-beta-binomial models. Other tests and models developed in their article are quite useful and interesting but are not examined herein.  相似文献

The response of mosquito larvae to plant toxins found in their breeding sites was investigated by using Aedes aegypti larvae and toxic arborescent leaf litter as experimental models. The relation between larval tolerance to toxic leaf litter and cytochrome P450 monooxygenases (P450s) was examined at the toxicological, biochemical and molecular levels. Larvae pre-exposed to toxic leaf litter show a higher tolerance to those xenobiotics together with a strong increase in P450 activity levels. This enzymatic response is both time- and dose-dependent. The use of degenerate primers from various P450 genes (CYPs) allowed us to isolate 16 new CYP genes belonging to CYP4, CYP6 and CYP9 families. Expression studies revealed a 2.3-fold over-expression of 1 CYP gene (CYP6AL1) after larval pre-exposure to toxic leaf litter, this gene being expressed at a high level in late larval and pupal stages and in fat bodies and midgut. The CYP6AL1 protein has a high level of identity with other insect's CYPs involved in xenobiotic detoxification. The role of CYP genes in tolerance to natural xenobiotics and the importance of such adaptive responses in the capacity of mosquitoes to colonize new habitats and to develop insecticide resistance mechanisms are discussed.  相似文献

Rank approaches are very common in the analysis of ordered categorical data but can only be interpreted on an experiment‐wise level. Therefore, parametric tests from linear models, although based on metric structures, are used frequently to analyze this type of data. So the questions arise 1. what parametric tests measure in this context and 2. whether the rank approach could be modified to achieve a global level of interpretation. A possible solution to question 2. offers the so called ridit approach, which is based on known reference distributions. In this paper we discuss a global view that shows how rank analysis and ridit analyses are related and how parametric procedures fit into the same framework. The use of the uniform distribution as a reference in the ridit approach gives an explanation to question 1. The asymptotic multivariate normality of the effect estimators is shown and robust test statistics are discussed. The identification of environmental compounds that have adverse effects on reproductive health and animal development is particularly challenging. Fenarimol, a systemic fungicide, is considered non or weakly genotoxic. However, its available toxicological data are controversial and incomplete. This study was conducted in rat in vivo to determine whether this compound (150 and 300 mg/kg) had adverse effects on DNA integrity in dams and pups after maternal subcutaneous exposure. The animals were exposed during early gestation (1-6 days), late gestation (last 6 days), or first 6 days of lactation. Findings on fenarimol genotoxicity showed an adverse effect when detected by the Comet assay, both in dams and pup, and state that animal sensitivity to fenarimol is higher during postnatal period. Since the DNA damage increases during the time of exposure (2 h to 6 days after the birth), our data on pups suggest that fenarimol can mainly act on cell DNA through direct exposure of litter via milk.  相似文献

In certain experiments with laboratory animals like rodents, it is common to observe the binary littermate data that the fetuses in the same litter provide a set of independent Bernoulli responses conditional on the litter specific abnormal probability. For testing homogeneity of abnormal proportions of several groups, five methods are discussed in the frame of the one‐way layout with litter effects. The 1983 book, Risk Assessment in the Federal Government: Managing the Process, recommended developing consistent inference guidelines for cancer risk assessment. Over the last 15 years, extensive guidance have been provided for hazard assessment for cancer and other endpoints. However, as noted in several recent reports, much less progress has occurred in developing consistent guidelines for quantitative dose response assessment methodologies. This paper proposes an approach for dose response assessment guided by consideration of mode of action (pharmacodynamics) and tissue dosimetry (pharmacokinetics). As articulated here, this systematic process involves eight steps in which available information is integrated, leading first to quantitative analyses of dose response behaviors in the test species followed by quantitative analyses of relevant human exposures. The process should be equally appropriate for both cancer and noncancer endpoints. The eight steps describe the necessary procedures for incorporating mechanistic data and provide multiple options based upon the mode of action by which the chemical causes the toxicity. Given the range of issues involved in developing such a procedure, we have simply sketched the process, focusing on major approaches for using toxicological data and on major options; many details remain to be filled in. However, consistent with the revised carcinogen risk assessment guidance (USEPA, 1996c), we propose a process that would ultimately utilize biologically based or chemical specific pharmacokinetic and pharmacodynamic models as the backbone of these analyses. In the nearer term, these approaches will be combined with analysis of data using more empirical models including options intended for use in the absence of detailed information. A major emphasis in developing any harmonized process is distinguishing policy decisions from those decisions that are affected by the quality and quantity of toxicological data. Identification of data limitations also identifies areas where further study should reduce uncertainty in the final risk evaluations. A flexible dose response assessment procedure is needed to insure that sound toxicological study results are appropriately used to influence risk management decision-making and to encourage the conduct of toxicological studies oriented toward application for dose response assessments.  相似文献

After a brief discussion of the threshold concept in toxicology, we consider models for the estimation of thresholds in the case where the observed response is binary or quantal. A generalization of the four-parameter Tukey-lambda family of distributions is proposed as a useful class of models for threshold estimation. Properties of these models are discussed and the process of model fitting and evaluation is illustrated using a number of data sets. The discussion includes consideration of background or spontaneous response, and comparison with other models. One of these is the linear-plateau or hockey-stick model, which has been used in a number of toxicological studies.  相似文献

There are compelling reasons to search for alternatives to the use of animals in medical and pharmaceutical research. Aside from the obvious animal welfare issues, both the well-established differences between animal models and humans, and the inherent inter-individual variability in human biological responses, indicate that human-based alternatives are urgently required. However, any such alternative must out-perform the animal-based alternative, otherwise there will be little or no uptake and adoption by end-users. Data obtained from inbred animal models is often highly reproducible, and is therefore attractive to researchers in the fields of biomedical and pharmaceutical research. The inter-individual variability observed during human volunteer and human tissue-based studies is often considered to be problematic, and has been highlighted further with the advent of the 'omics' technologies, which generate large biological datasets. However, the variability in both baseline data and response to pharmacological or toxicological challenge observed in human tissues potentially contains a veritable gold mine of information, which may be critical for the advancement of drug discovery.  相似文献

Despite years of intensive work, the biochemical defect responsible for the pathogenesis of inherited muscular dystrophy has not been identified either in humans or animal models. This review examines evidence in support of the hypothesis that free radicals may be responsible for muscle degeneration in this disorder. A variety of cellular abnormalities noted in dystrophic muscles can be accounted for by free radical mediated damage. In addition, chemical by-products associated with free radical damage are found in dystrophic muscle tissue from humans and animals with this disease. Various enzymatic antioxidant systems can be enhanced as a normal cellular response to oxidative stress, and such changes are seen both in dystrophic muscle cells and certain other tissues of dystrophic animals. An increased level of free radical damage would follow from either: enhanced production of free radical species, or a deficient component of the cellular antioxidant system, such as vitamin E. The free radical hypothesis of muscular dystrophy can account for data supporting several alternative theories of the pathogenesis of this disease, as well as other observations which have not previously been explained.  相似文献

The present experiment compared the anxiety- and activity-related behaviour of sub-adult females from a mouse strain selected for over 106 generations for high litter size with that of a randomly selected control strain, to illuminate possible differences in their ability to cope with exposure to a novel environment. Selected for large litter size, the H-strain has an average litter size of 21 pups, whereas the randomly bred C-strain has an average litter size of 10 pups. The elevated plus-maze was used to measure the behaviour of the mice expressed in response to novelty. The results are described and discussed in relation to the ability to cope with novel environmental challenges. In the elevated plus-maze, the H-strain was significantly more anxious (having a lower percentage of entries into and percentage of time spent on open arms) and less active (having a lower number of entries into closed arms, and a lower total number of arm entries) than the C-strain. Thus, there were clear anxiety- and activity-related differences between the strains, which may be related to selection for large litter size. A tentative hypothesis is presented whereby selection for large litter size may accelerate adaptation to the home environment, but decrease the ability of selected animals to cope with exposure to novelty.  相似文献

A mathematical model for describing the cancer growth dynamics in response to anticancer agents administration in xenograft models is discussed. The model consists of a system of ordinary differential equations involving five parameters (three for describing the untreated growth and two for describing the drug action). Tumor growth in untreated animals is modelled by an exponential growth followed by a linear growth. In treated animals, tumor growth rate is decreased by an additional factor proportional to both drug concentration and proliferating cells. The mathematical analysis conducted in this paper highlights several interesting properties of this tumor growth model. It suggests also effective strategies to design in vivo experiments in animals with potential saving of time and resources. For example, the drug concentration threshold for the tumor eradication, the delay between drug administration and tumor regression, and a time index that measures the efficacy of a treatment are derived and discussed. The model has already been employed in several drug discovery projects. Its application on a data set coming from one of these projects is discussed in this paper.  相似文献

Climate and litter quality are primary drivers of terrestrial decomposition and, based on evidence from multisite experiments at regional and global scales, are universally factored into global decomposition models. In contrast, soil animals are considered key regulators of decomposition at local scales but their role at larger scales is unresolved. Soil animals are consequently excluded from global models of organic mineralization processes. Incomplete assessment of the roles of soil animals stems from the difficulties of manipulating invertebrate animals experimentally across large geographic gradients. This is compounded by deficient or inconsistent taxonomy. We report a global decomposition experiment to assess the importance of soil animals in C mineralization, in which a common grass litter substrate was exposed to natural decomposition in either control or reduced animal treatments across 30 sites distributed from 43°S to 68°N on six continents. Animals in the mesofaunal size range were recovered from the litter by Tullgren extraction and identified to common specifications, mostly at the ordinal level. The design of the trials enabled faunal contribution to be evaluated against abiotic parameters between sites. Soil animals increase decomposition rates in temperate and wet tropical climates, but have neutral effects where temperature or moisture constrain biological activity. Our findings highlight that faunal influences on decomposition are dependent on prevailing climatic conditions. We conclude that (1) inclusion of soil animals will improve the predictive capabilities of region‐ or biome‐scale decomposition models, (2) soil animal influences on decomposition are important at the regional scale when attempting to predict global change scenarios, and (3) the statistical relationship between decomposition rates and climate, at the global scale, is robust against changes in soil faunal abundance and diversity.  相似文献

Protective forms of animal color, such as crypsis, are thought to reduce the probability of detection by visual predators. However, because crypsis is ostensibly intuitive, the working hypothesis of cryptic coloration is seldom tested. Additionally because crypsis is a background-specific adaptation, events which alter habitat structure and substrate composition are likely to affect rates of predation on cryptic animals; animal colors that are cryptic against one visual background may be conspicuous against different visual backgrounds. Populations of Sceloporus woodi, a cryptic diurnal lizard, occupy clear-cut stands of sand pine scrub and prescribe-burned longleaf pine habitat within the Ocala National Forest. Here, we used a combination of clay models resembling S. woodi, and spectral analysis, to examine the effects of spatial heterogeneity and model-substrate contrast on rates of predation. The rate of attack on clay models differed between substrate types and habitats, and was highest when clay models were conspicuous against the local visual background. The dorsal color of models greatly contrasted open sand and dead wood, but had similar reflectance values to leaf litter, suggesting that models were most cryptic on leaf litter. We conclude that crypsis is adaptive in this species, and that variation in rates of attack between sampling locations is related to changes in substrate composition due to management history. For instance, the data suggest that the rate of attack on clay models would decrease in response to succession in sand pine scrub, because aging in sand pine scrub results in increased amounts of leaf litter and decreased amounts of open sand. Overall, the results of this study support the theory of protective coloration.  相似文献

The relationships between quantitative and reproductive fitness traits in animals are of general biological importance for the development of population genetic models and our understanding of evolution, and of great direct economical importance in the breeding of farm animals. Two well investigated quantitative traits--body weight (BW) and litter size (LS)--were chosen as the focus of our review. The genetic relationships between them are reviewed in fishes and several mammalian species. We have focused especially on mice where data are most abundant. In mice, many individual genes influencing these traits have been identified, and numerous quantitative trait loci (QTL) located. The extensive data on both unselected and selected mouse populations, with some characterized for more than 100 generations, allow a thorough investigation of the dynamics of this relationship during the process of selection. Although there is a substantial positive genetic correlation between both traits in unselected populations, caused mainly by the high correlation between BW and ovulation rate, that correlation apparently declines during selection and therefore does not restrict a relatively independent development of both traits. The importance of these findings for overall reproductive fitness and its change during selection is discussed.  相似文献

Attention is focussed on two frequently occurring complications in medical applications: censoring and tail alternatives. A review is given of the author's recent work on a new and simple class of censored rank tests. Background The purpose of this study is to better characterize the hematological, biochemical, respiratory, cardiovascular and electroneurophysiological parameters in young adult Cercopithecus aethiops sabaeus of both sexes. The rhesus and cynomolgus monkeys are widely used as experimental primate models. However, only few articles have been published testing toxicological effects of pharmaceuticals on African green monkey. Methods The present study was carried out with the recompilation of all parameters recorded before the first drug administration in five sub‐chronic or chronic toxicological studies performed on 66 Cercopithecus aethiops sabaeus, born in Cuba. Results This study provides hematological, biochemical, respiratory, cardiovascular and electroneurophysiological data for both choosing animals to be included into experiments and monitoring these parameters during the study. Conclusions We conclude that this study provides valuable integrated data for determining the health status, including electroneurophysiological parameters, data not previously reported for this species, of the African green monkey.  相似文献

The toxicity of iv injected hydrophilic aluminum complex tris(maltolate)aluminum(III) was studied in New Zealand white rabbits for a period of time ranging from 5 to 63 wk. Animals were injected 3–5 times a week with 1 mL of 7.5 mM Al(malt)3 and one rabbit with a dose 10 times higher after 14 wk of treatment. Autopical examination was performed on all animals. Chemoclinical analysis (glucose, urea, creatinine, cholesterol, bilirubin, alanin aminotransferase, aspartate aminotransferase, alkaline phosphatase, γ-glutamyltransferase, LDH, CK, total protein, triglycerides, and Ca2+) gave no variation in treated animals with respect to the control. The toxicological data show a moderate systemic general toxicity at doses far higher than those used in similar previous experiments using Al(acac)3 (acac=2,4 pentanedionate), a hydrolytically stable and more lipophilic aluminum(III) complex (1). The diversity of behavior is discussed in terms of metal speciation as well as respect to the thermodynamic and kinetic properties of the two complexes in aqueous solution. The toxicological model presented here emphasizes that neutral, water compatible aluminum(III) complexes are to be considered as promising tools for toxicological experiments providing biological models of human pathologies.  相似文献

In this paper the problem of "impulsive" and "self-control" behaviour in animals and the experimental models for their assessment are discussed. It is pointed out that a widely accepted model for assessment of impulsive behaviour in animals predominantly measures their ongoing properties of impulsivity/self-control independently on belonging them to the certain types of higher nervous activity. In contrary, our model at the first instance assesses the typological characteristics of animals, including impulsivity and self-control, by their abilities to perform or not to perform an appropriate behaviour. The differences in the data obtained by these two different models and the possible reasons for it are discussed.  相似文献

Human exposure to endocrine disrupters (EDs) is widespread and is considered to pose a growing threat to human health. Recent advances in molecular and genetic research and better understanding of mechanisms of blastic cell transformation have led to efforts to improve cancer risk assessment for populations exposed to this family of xenobiotics. In risk assessment, low dose extrapolation of cancer incidence data from both experimental animals and epidemiology studies has been largely based on models assuming linear correlation at low doses, despite existence of evidence showing otherwise. Another weakness of ED risk assessment is poor exposure data in ecological studies. Those are frequently rough estimates derived from contaminated items of local food basket surveys. Polyhalogenated hydrocarbons are treated as examples. There is growing sense of urgency to develop a biologically based dose response model of cancer risk, integrating emerging data from molecular biology and epidemiology to provide more realistic data for risk assessors, public, public health managers and environmental issues administrators.  相似文献
