Cryoconite holes have biogeochemical, ecological and biotechnological importance. This communication presents results on culturable psychrophilic bacterial diversity from cryoconite holes at Midre Lovénbreen (ML), Austre Brøggerbreen (AB), and Vestre Brøggerbreen (VB) glaciers. The culturable bacterial count ranged from 2.7 × 103 to 8.8 × 104 CFUs/g while the total bacterial numbers ranged from 5.07 × 105 to 1.50 × 106 cells at the three glaciers. A total of 35 morphologically distinct bacterial isolates were isolated. Based on 16S rRNA gene sequence data, the identified species belonged to eight genera namely Pseudomonas, Polaromonas, Micrococcus, Subtercola, Agreia, Leifsonia, Cryobacterium and Flavobacterium. The isolates varied in their growth temperature, NaCl tolerance, growth pH, enzyme activities, carbon utilization and antibiotic sensitivity tests. Fatty acid profiles indicate the predominance of branched fatty acids in the isolates. To the best of our knowledge, this is the first record of culturable bacterial communities and their characterization from glacier cryoconites from High Arctic. High amylase and protease activities expressed by Micrococcus sp. MLB-41 and amylase, protease and lipase activities expressed by Cryobacterium sp. MLB-32 provide a clue to the potential applications of these organisms. These cold-adapted enzymes may provide an opportunity for the prospect of biotechnology in Arctic.  相似文献

A total of 132 yeast strains were characterised from 4 sediment samples collected from small puddles in the vicinity of Midre Lovénbreen glacier, Arctic. Based on the D1/D2 domain sequence similarity, the isolates could be categorised into 6 groups. The nearest phylogenetic neighbour of groups I to VI were identified as Cryptococcus gastricus, Cryptococcus terricolus, Rhodotorula muscorum, Mrakia psychrophila, Mrakia gelida and Rhodotorula glacialis, respectively. Strains representative of the six groups were psychrophilic and salt tolerant but varied in their ability to produce cold-active extracellular enzymes such as lipase, protease, pectinase, cellulase and amylase. C18:1 (w9C) and C18:2 (w9,12C) were the only two fatty acids common to all the yeasts and branched and (or) unsaturated fatty acids increased in yeasts growing at 8°C compared to 22°C, probably as an adaptation to low temperature. The present study establishes that psychrophilic yeasts are predominant in Arctic and could be used as work horses to produce cold-active enzymes and poly unsaturated fatty acids which have been implicated in low temperature adaptation and also for their use in biotechnology.  相似文献

Seeds ofKarwinskia humboldtiana obtained from a 1997 collection in the locality of Villa de García Nuevo (León, Mexico) were contaminated with spores of filamentous fungi, bacteria and yeasts. The concentration of microorganisms in unscarified seeds ranged from 3.0×103 to 7.5×103 CFU/g. Predominant were bacterial isolates of the generaAeromonas sp.,Bacillus, andPseudomonas; from filamentous fungi were identifiedAlternaria, Aspergillus niger, Cladosporium sp.,Fusarium sp.,Mucor sp.,Penicillium commune, Trichothecium sp.; from yeastsRhodotorula sp. andSaccharomyces cerevisiae. Seed scarification significantly reduced the microbial contamination. Of the original fungal isolates, only two were identified on scarified seeds,viz. Cladosporium sp. andSaccharomyces cerevisiae; although a relatively high incidence of a unidentifiable ofPenicillium sp. was found, the bacterial spectrum was not altered. Treatment of scarified seeds with Vitavax 200 WP and Pomarsol Forte 80 WP (3 mg/g seeds) augmented germination by 10–19% compared to treated unscarified seeds, and by 16–31% compared to untreated unscarified seeds.  相似文献

Isolation of Brazilian marine fungi capable of growing on DDD pesticide   总被引:1,自引:0,他引:1  
The fungi Aspergillus sydowii Ce15, Aspergillus sydowii Ce19, Aspergillus sydowii Gc12, Bionectria sp. Ce5, Penicillium miczynskii Gc5, Penicillium raistrickii Ce16 and Trichoderma sp. Gc1, isolated from marine sponges Geodia corticostylifera and Chelonaplysylla erecta, were evaluated for their ability to grow in the presence of DDD pesticide. Increasing concentrations of DDD pesticide, i.e., 5.0 mg (1.56 × 10−12 mmol), 10.0 mg (3.12 × 10−2 mmol) and 15.0 mg (4.68 × 10−2 mmol) in solid and liquid culture media were tested. The fungi Trichoderma sp. Gc1 and Penicillium miczynskii Gc5 were able to grow in the presence of up to 15.0 mg of DDD, suggesting their potential for biodegradation. A 100% degradation of DDD was attained in liquid culture medium when Trichoderma sp. Gc1 was previously cultivated for 5 days and supplemented with 5.0 mg of DDD in the presence of hydrogen peroxide. However, the quantitative analysis showed that DDD was accumulated on mycelium and biodegradation level reached a maximum value of 58% after 14 days.  相似文献

Two strains of a basidiomycetous yeast were derived from an insect trypanosomatid culture isolated from the intestine of a plant bug, Collaria oleosa (Heteroptera: Miridae), collected in Costa Rica. The yeast did not form ballistoconidia but reproduced only by budding. Teliospores were not observed in individual and crossed cultures of each strain. Morphological and other taxonomic characteristics of the yeast were similar to those of the species in the polyphyletic genus Rhodotorula. However, molecular phylogeny inferred from the internal transcribed spacers and D1/D2 region of the large subunit rRNA gene showed that the strains represent a new species placed among the smut fungi in the family Ustilentylomataceae, which includes Aurantiosporium subnitens, Fulvisporium restifaciens, Ustilentyloma fluitans, and Rhodotorula hordea. Given the well distinguished phylogenetic position of this novel species within the Ustilentylomataceae, we propose Microbotryozyma collariae gen. nov., sp. nov. to accommodate the yeast isolated from C. oleosa, with strain American Type Culture Collection MYA-4666T (= PRA303-1S = CBS 12537) designated as the type strain.  相似文献

Natural environmental isotopes of nitrate, sulphate and inorganic carbon are discussed in conjunction with major ion chemistry of subglacial runoff from a High Arctic glacier, Midre Lovénbreen, Svalbard. The chemical composition of meltwaters is observed to switch in accordance with subglacial hydrological evolution and redox status. Changing rapidly from reducing to oxidizing conditions, subglacial waters also depict that 15N/14N values show microbial denitrification is an active component of nutrient cycling beneath the glacier. 18O/16O ratios of sulphate are used to elucidate mechanisms of biological and abiological sulphide oxidation. Concentrations of bicarbonate appear to be governed largely by the degree of rock: water contact encountered in the subglacial system, rather than the switch in redox status, although the potential for microbiological activity to influence ambient bicarbonate concentrations is recognised. Glaciers are therefore highlighted as cryospheric ecosystems supporting microbial life which directly impacts upon the release of solute through biogeochemically mediated processes.  相似文献

The search for new compounds with antifungal activity is accelerating due to rising yeast and fungal resistance to commonly prescribed drugs. Among the molecules being investigated, plant lectins can be highlighted. The present work shows the potential of six plant lectins which were tested in vitro against yeasts of medical importance, Candida albicans, Candida tropicalis, Candida parapsilosis, Cryptococcus gattii, Cryptococcus neoformans, Malassezia pachydermatis, Rhodotorula sp. and Trichosporon sp. Broth microdilution susceptibility testing was performed in accordance with standard protocols to evaluate antifungal activity. Minimum inhibitory concentration (MIC) was determined at 80 % yeast growth inhibition, whereas the minimum fungicidal concentration (MFC) was evaluated after making the subcultures of each dilution. Only C. parapsilosis growth was inhibited by the lectins tested. Abelmoschus esculentus lectin showed the highest MIC (0.97 μg ml?1). Lectins from Canavalia brasiliensis, Mucuna pruriens and Clitoria fairchildiana presented the highest MFC at (3.90 μg ml?1). These results encourage further studies with wider yeast strain selections, and open new perspectives for the development of pharmacological molecules.  相似文献

Many working environments are predisposed for larger than average amounts of fungi and other microorganisms often due to organic material being handled. From 2003 to 2007, the area used for strawberry production in Denmark increased by 62%. The purpose of this study was to determine the levels of exposure to microorganisms, endotoxin, (1→3)-β-d-glucan (β-glucan), and pollen in a field of strawberries. The study was carried out in eastern Denmark from the middle of June to the beginning of August 2008. The strawberries were grown organically, and microbiological pest control agents (MPCAs) were applied during this and former growth seasons. In order to measure exposure to inhalable bioaerosol components, we used stationary filter samplers. Bioaerosol sampling was performed during 4 working days, and a total of 57 samplings were performed. The filters were analysed for contents of fungi, MPCAs, endotoxin, β-glucan, and pollen. The mean exposure was 6,154 CFU Cladosporium sp. m−3, 1.0 × 105 fungal spores m−3, 4.1 × 104 hyphal fragments m−3, 5.8 × 103 pollen m−3, 57.3 ng β-glucan m−3, and 8.9 endotoxin units (EU) m−3. A significant and positive correlation was found between β-glucan and fungal spores and between CFU of Cladosporium sp. and CFU of fungi. We selected specifically for Metarhizium anisopliae, Beauveria bassiana, and the applied MPCAs Trichoderma harzianum, T. polysporum, and Bacillus thuringiensis but found none of these species. In conclusion, our study shows that berry pickers in this organic strawberry field were potentially subjected to higher levels of fungal spores, Cladosporium sp., hyphal fragments, pollen, and thus also β-glucan than is usually seen in outdoor air. Exposure to MPCAs was not seen. The exposure to endotoxin was only slightly higher than e.g. in a town.  相似文献

The High Arctic glacial ecosystem: new insights from nutrient budgets   总被引:3,自引:2,他引:1  
This paper describes detailed budgets of water, Cl, dissolved Si and both inorganic and organic forms of nitrogen and phosphorus for two small glacier basins in Arctic Svalbard (Midre Lovénbreen and AustreBrøggerbreen). Rates of nutrient deposition are modest, dominated by inorganic nitrogen and episodically enhanced by extreme events. Hence deposition rates are also variable, ranging from 20 to 72 kg NO3-N km−2 a−1 and 10–37 kg NH4-N km−2 a−1 over just two consecutive years. Deposition of dissolved organic and particulate forms of nitrogen (DONand PN respectively) also appears significant and therefore requires further investigation (3–8 kg DON-N km−2 and 7–26 kg PN-N km−2 during winter – no summer data are available). Evidence for microbially mediated nutrient cycling within the glacial system is clear in the nutrient budgets, as is the release of large phosphorus, Si and organic/particulate nitrogen fluxes by subglacial erosion. The latter is entirely dependent upon the presence of subglacial drainage, promoting silicate mineral dissolution and the erosion of largely unweathered apatite. The large DON and PN fluxes are surprising and may relate to young organic nitrogen associated with microbial life within the glaciers. This is because wide spread assimilation of NH4+ and perhaps even nitrification occurs on the glacier surface, most likely within abundant cryoconite holes. Further microbial activity also occurs at the glacier bed, where denitrification and sulphate reduction is now known to take place. Thus a two component ‘glacial ecosystem’ is proposed that is highly sensitive to climate change.  相似文献

This study investigates the exposure of workers to biological particles in a poultry litter burning plant in operation. The microorganism concentrations were examined at different workplaces during procedures leading to increased emissions. The concentrations of culturable airborne mesophilic, xerophilic and thermophilic microorganisms in the ambient air were tested inside and outside of the burning plant using two different methods of measuring. The focus of this study was on the quantitative evaluation of culturable bacteria as well as the quantitative and qualitative evaluation of gram-negative bacteria, fungi and thermophilic actinomycetes. The maximum airborne concentrations were found in the delivery hall. Mesophilic bacteria concentrations reached up to 1.7 × 106 CFU/m3; gram-negative bacteria up to 9.1 × 102 CFU/m3. Fungal propagule concentrations for xerophilic fungi were between 1.2 × 103 and 2.9 × 104 CFU/m3 and for mesophilic fungi between 4.4 × 102 and 2.9 × 104 CFU/m3. Among fungi, Aspergillus niger, Eurotium herbariorum and Scopulariopsis brevicaulis species were dominant. Thermophilic actinomycetes reached airborne concentrations of 8.7 × 104 CFU/m3, with increased concentrations of the pathogens causing extrinsic allergic alveolitis. The high concentrations of airborne microorganisms in poultry litter burning plants and the potential hazard of the intake of microorganisms including potential pathogens require the introduction of consistent measures in both technical areas and personnel management.  相似文献

The diversity of highly active bacterial communities in cryoconite holes on three Arctic glaciers in Svalbard was investigated using terminal restriction fragment length polymorphism (T-RFLP) of the 16S rRNA locus. Construction and sequencing of clone libraries allowed several members of these communities to be identified, with Proteobacteria being the dominant one, followed by Cyanobacteria and Bacteroidetes. T-RFLP data revealed significantly different communities in holes on the (cold) valley glacier Austre Brøggerbreen relative to two adjacent (polythermal) valley glaciers, Midtre Lovénbreen and Vestre Brøggerbreen. These population compositions correlate with differences in organic matter content, temperature and the metabolic activity of microbial communities concerned. No within-glacier spatial patterns were observed in the communities identified over the 2-year period and with the 1 km-spaced sampling. We infer that surface hydrology is an important factor in the development of cryoconite bacterial communities.  相似文献

Airborne yeasts were collected with 6‐stage Andersen samplers indoors and outdoors in rural and urban areas in southern Finland in 1982–1983, usually twice a month (but once a month only during the period of snow cover). The predominant yeast genus was Sporobolomyces followed by Rhodotorula and Cryptococcus. The most frequently isolated species were Sporobolomyces roseus, Sp. holsaticus, Rhodotorula minuta, Rh. glutinis, Rh. rubra, Cryptococcus albidus and Cr. laurentii. Ascomycetous yeasts were caught infrequently; for example Candidas occurred in samples only occasionally. Numbers of yeasts indoors were significantly correlated with those out of doors in the country but not in town although numbers outdoors in town and country were correlated. Species found indoors were similar to those found out of doors. Numbers of yeast cells in the air were always small, never exceeding 50 CFU m‐3 and clearly below estimated threshold values for airborne fungi causing allergic reactions.  相似文献

To survey the effects of endophytic Enterobacter sp. CBSB1 and Rhodotorula sp. CBSB79 resistant to Cd2+, Pb2+, Zn2+, and Cu2+ on the growth and phytoextraction of Brassica, the endophytes were isolated by surface- sterilized methods and characterized. The CBSB1 significantly increased 44.2% of the dry weight of Brassica napus in the multimetal contaminated soil (P < 0.05) and showed no effect or declined the dry weight of B. alboglabra, B. campestris ssp. chinensis var. cummunis, B. campestris ssp. chinensis var. utilis cv. Youqing12, B. campestris ssp. chinensis var. utilis cv. Lvbao701 plants. The dry weights of B. napus, B. campestris ssp. chinensis var. utilis, and B. alboglabra showed a significant increase when the CBSB79 was inoculated (P < 0.05). In general, inoculation with bacteria and yeast did not greatly alter the metal concentration in plant tissues. Compared to Enterobacter sp. CBSB1, the yeast Rhodotorula sp CBSB79 showed higher potentials to improve extraction efficacy of Cd, Pb, Zn, and Cu by Brassica seedlings in the field.  相似文献

Fructooligosaccharides (FOSs) are functional food ingredients with prebiotic properties, and a recent increase in the use of oligosaccharides in the food industry has led to the search for “new” microorganisms and enzymes for the production of oligosaccharides. This paper focuses on the screening of yeasts obtained from fruits and flowers (from Brazilian tropical forests), and capable of secreting extra-cellular enzymes with high fructosyl transferase activity (FTA). The screening and isolation procedures resulted in four potentially interesting yeast strains: Candida sp. (LEB-I3), Rhodotorula sp. (LEB-U5.), Cryptococcus sp. (LEB-V2) and Rhodotorula sp. LEB-V10. All were able to produce more then 100 g l−1 of FOS from a 500 g l−1 sucrose solution, but only the last one, (LEB-V10), showed no hydrolytic activity with respect to the FOS produced, giving a continuous increase in FOS content up to the end of the reaction, when it was about 50% of the total carbohydrates.  相似文献

In this study, enumeration and identification of total aerobic heterotrophic bacteria and petroleum-utilizing bacteria as well as the degradative potential of petroleum-utilizing bacterial isolates were carried out. The average counts of total aerobic heterotrophic bacteria in cow dung and poultry manure were 74.25 × 105 c.f.u. g−1 and 138.75 × 105 c.f.u. g−1 respectively. Acinetobacter sp, Bacillus sp, Pseudomonas sp, and Serratia spp. occurred as aerobic heterotrophs in both cow dung and poultry manure. However, Alcaligenes spp. occurred only in cow dung while, Flavobacterium sp, Klebsiella sp, Micrococcus sp, and Nocardia spp. occurred only in poultry manure as aerobic heterotrophs. The average counts of petroleum-utilizing bacteria in cow dung and poultry manure were 9.25 × 105 c.f.u. g−1 and 17.25 × 105 c.f.u. g−1 respectively. Pseudomonas spp. occurred as petroleum utilizer in both cow dung and poultry manure. However, Bacillus spp. occurred only in cow dung while Acinetobacter spp. and Micrococcus spp. occurred only in poultry manure as petroleum utilizers. Relative abundance of petroleum utilizers in total aerobic heterotrophs ranged from 6.38% to 20.00% for cow dung and from 9.38% to 17.29% for poultry manure. Introduction of pure cultures of petroleum-utilizing bacteria from cow dung and poultry manure into sterile oil-polluted soil revealed oil degradation in one week period.  相似文献

Uncertainty surrounds estimates of microbial cell and organic detritus fluxes from glacier surfaces. Here, we present the first enumeration of biological particles draining from a supraglacial catchment, on Midtre Lovénbreen (Svalbard) over 36 days. A stream cell flux of 1.08 × 107 cells m?2 h?1 was found, with strong inverse, non‐linear associations between water discharge and biological particle concentrations. Over the study period, a significant decrease in cell‐like particles exhibiting 530 nm autofluorescence was noted. The observed total fluvial export of ~ 7.5 × 1014 cells equates to 15.1–72.7 g C, and a large proportion of these cells were small (< 0.5 μm in diameter). Differences between the observed fluvial export and inputs from ice‐melt and aeolian deposition were marked: results indicate an apparent storage rate of 8.83 × 107 cells m?2 h?1. Analysis of surface ice cores revealed cell concentrations comparable to previous studies (6 × 104 cells ml?1) but, critically, showed no variation with depth in the uppermost 1 m. The physical retention and growth of particulates at glacier surfaces has two implications: to contribute to ice mass thinning through feedbacks altering surface albedo, and to potentially seed recently deglaciated terrain with cells, genes and labile organic matter. This highlights the merit of further study into glacier surface hydraulics and biological processes.  相似文献

The distribution and presence of hygiene indicator and pathogenic micro‐organisms in 375 samples of attieke marketed in Côte d'Ivoire, and their roles in the food poisoning were evaluated. Microbiological analyses were carried out, which included the total viable bacteria, coliforms, Escherichia coli, Staphylococcus aureus, Salmonella, Bacillus spores, fungi and Clostridium perfringens. The results revealed that the viable bacteria counts ranged from 2·2 ± 1·2 × 105 to 3·4 ± 1·4 × 106 CFU g?1, while the yeasts and the moulds counts ranged, respectively, from 2·4 ± 0·12 × 104 to 9·8 ± 0·4 × 105 CFU g?1 and 1·3 ± 0·7 × 101 to 1·7 ± 0·7 × 102 CFU g?1. Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Citrobacter freundi, Enterobacter amnigenus, Citrobacter youngae, Enterobacter aerogenes, Klebsiella pneumoniae, Serratia marcescens, Enterobacter agglomerans and Klebsiella oxytoca were the bacteria isolated, and Rhizopus spp., Mucor spp., Thamnidium spp., Fusarium spp., Moniliella spp. the fungi. Escherichia coli, Clostridium perfringens and Salmonella spp. were not found. The occurrence of some bacteria and fungi illustrate that attieke collected in Côte d'Ivoire markets may act as a reservoir of pathogenic micro‐organisms for human.

Significance and Impact of Study

This study demonstrates the great need to carry out microbiological tests frequently on attieke and even more the need to apply correct HACCP system during the production. Attieke is especially a well‐known product in West Africa; hence, it is extremely important to ensure an adequate microbiological quality to guarantee consumers health. Overall, the study highlighted the need for effective communication on microbiological food risks, proper instruction and supervision in food‐handling procedures, greater education on food safety risks.  相似文献

Thailand border market is where the local Thais, Cambodians, Laotians, and Burmeses exchange their goods and culture at the border checkpoints. It is considered to be the source of aerial disease transmission especially for foreigners because it is always very crowded with people from all walks of life. Unhealthy air quality makes this area high risk of spread of airborne diseases. This study assessed airborne concentrations of bacteria and fungi in a border market to improve exposure estimates and develop efficient control strategies to reduce health risk. The density and distribution of airborne bacteria and fungi were investigated in the Chong Chom border market in Surin Province, Thailand. Eighteen air sampling sites were taken from outdoors and various work environments including indoor footpaths, wooden handicraft shops, electronic shops, the secondhand clothing shops, and fruit market areas. Exposed Petri plate method and liquid impinger sampler were used for sampling at the breathing zone, 1.5 m above the floor level, during weekend and holiday. Meteorological factors such as relative humidity, temperature, and light intensity were collected by portable data logger. The relative humidity was 67–73%, and temperature 29–33°C, and light varied between 18 and 270 Lux m−2. Gram-positive and Gram-negative bacteria were found at a mean value of 104 CFU m−3, and airborne fungi of 103 CFU m−3 were recorded. The highest concentration of culturable airborne microorganisms was found along the indoor footpath (9.62 × 104 CFU m−3 and 750.00 CFU/plate/h for impingement and sedimentation methods, respectively), the fruit market area (7.86 × 104 CFU m−3 and 592.42 CFU/plate/h for impingement and sedimentation methods, respectively), and the secondhand clothing shop (4.59 × 103 CFU m−3 and 335.42 CFU/plate/h for impingement and sedimentation methods, respectively) for Gram-positive bacteria, Gram-negative bacteria, and fungi, respectively. The lowest concentration of Gram-positive bacteria, Gram-negative bacteria, and fungi was found only at the outdoor area at 1.53 × 104 CFU m−3, 0.93 × 104 CFU m−3 and 0.80 × 103 CFU m−3 by means of impingement method and 136.67 CFU/plate/h, 69.25 CFU/plate/h, and 62.00 CFU/plate/h by means of sedimentation methods for Gram-positive bacteria, Gram-negative bacteria, and fungi, respectively. The most frequently present airborne bacteria were identified as Bacillus, Corynebacteria, Diplococcus, Micrococcus, Acinetobacter, Alcaligenes, Enterobacter, and spore former rods. Acremonium, Aspergillus, Cladosporium, Penicillium, and Sporotrichum were the most frequently found aerosol fungi genera. The distribution of airborne microorganisms correlated with relative humidity and light factors based on principal component analysis. In conclusion, the border market is a potential source of aerial disease transmission and a various hazards of bioaerosols for workers, consumers, sellers, and tourists. The bioaerosol concentration exceeded the standard of occupational exposure limit. Many major indicators of allergenic and toxigenic airborne bacteria and fungi, Acinetobacter, Enterobacter, Pseudomonas, Cladosporium, Alternaria, Aspergillus, and Penicillium, were found in the various market environments.  相似文献

Twenty-five psychrophilic yeasts were isolated from the soil of Roopkund Lake, Himalayas, India. Two colony morphotypes were identified and representatives of ‘morphotype 1’ were identified as Cryptococcus gastricus. Representatives of ‘morphotype 2’, namely 3AT, 4A, 4B and Rup4B, showed similar phenotypic properties and are identical with respect to the nucleotide sequence of the ITS1-5.8S rRNA gene-ITS2 region and D1/D2 domain of the 26S rRNA gene. The sequence of D1/D2 domain of 3AT shows 97.6–98.8% similarity with Rhodotorula psychrophila CBS10440T, Rhodotorula glacialis CBS10437T and Rhodotorula psychrophenolica CBS10438T and in the neighbour-joining phylogenetic tree strains; 3AT, 4A, 4B and Rup4B form a cluster with Rhodotorula glacialis and Rhodotorula psychrophila. Strains 3AT, 4A, 4B and Rup4B also differ from their nearest phylogenetic relatives in several biochemical characteristics such as in assimilation of d-galactose, l-sorbose, maltose, citrate, d-glucuronate and creatinine. Thus, based on the phylogenetic analysis and the phenotypic differences 3AT, 4A, 4B and Rup 4B are assigned the status of a new species of Rhodotorula for which the name Rhodotorula himalayensis sp. nov. is proposed with 3AT as the type strain (=CBS10539T =MTCC8336T). Acinetobacter sp. XMZ-26 (ACCC 05422) was isolated from soil samples obtained from glaciers in Xinjiang Province, China. The partial nucleotide sequence of a lipase gene was obtained by touchdown PCR using degenerate primers designed based on the conserved domains of cold-adapted lipases. Subsequently, a complete gene sequence encoding a 317 amino acid polypeptide was identified. Our novel lipase gene, lipA, was overexpressed in Escherichia coli. The recombinant protein (LipA) was purified by Ni-affinity chromatography, and then deeply characterised. The LipA resulted to hydrolyse pNP esters of fatty acids with acyl chain length from C2 to C16, and the preferred substrate was pNP octanoate showing a k cat = 560.52 ± 28.32 s−1, K m = 0.075 ± 0.008 mM, and a k cat/K m = 7,377.29 ± 118.88 s−1 mM−1. Maximal LipA activity was observed at a temperature of 15°C and pH 10.0 using pNP decanoate as substrate. That LipA peaked at such a low temperature and remained most activity between 5°C and 35°C indicated that it was a cold-adapted enzyme. Remarkably, this lipase retained much of its activity in the presence of commercial detergents and organic solvents, including Ninol, Triton X-100, methanol, PEG-600, and DMSO. This cold-adapted lipase may find applications in the detergent industry and organic synthesis.  相似文献
