Tropical island stream ecosystems continue to be threatened by increasing anthropogenic demands for freshwater, with many streams dammed or diverted. Stream flow amendments can have substantial effects on aquatic insect populations of tropical archipelagoes. In Hawaiian streams, an endemic Diptera community of the following genera dominates cascades and other torrential habitats: Telmatogeton Schiner (Chironomidae), Procanace Hendel (Canacidae), Scatella Robineau-Desvoidy (Ephydridae). Larval densities, standing stock biomass (SSB, as ash-free dry mass [AFDM]), and monthly secondary production of Telmatogeton and Procanace were measured during two summers of significantly different stream discharge in Iao Valley, Maui, Hawaii. Very few Scatella larvae were collected (<20 larvae for both years combined), so they were omitted from subsequent analyses. Stream discharge decreased approximately 40% from 1994 to 1995, providing a ‘natural test’ of the effects of reduced stream flow on these torrenticolous populations between two years. Combined Telmatogeton and Procanace SSB (total torrential community SSB) was 3176 and 1683 mg AFDM m−2 for 1994 and 1995, respectively, with Telmatogeton accounting for >95% in both years due to significantly larger body size and high density. The SSB of Telmatogeton significantly decreased from 3138 to 1622 mg AFDM m−2 from 1994 to 1995 but increased for Procanace (37.6–60.9 mg AFDM m−2, respectively). Total torrential community secondary production was 31% lower in 1994 (12,833 mg AFDM m−2 mo−1) compared to 1995 (8855 mg AFDM m−2 mo−1), reflecting the Telmatogeton proportion of total community production (99%); however, Procanace production increased by 40%. Monthly P/B ratios indicated that biomass turnover was generally high and increased for Telmatogeton from 1994 (3.8) to 1995 (5.1), whereas it remained lower and did not change between years for Procanace (1.7). A natural drought of the Iao Stream valley was associated with structural and functional changes in two endemic aquatic insects; these results are a conservative indication of permanent stream flow reductions from anthropogenic withdrawal (e.g., dams and diversions).  相似文献

Previous work in a young Hawaiian forest has shown that nitrogen (N) limits aboveground net primary production (ANPP) more strongly than it does decomposition, despite low soil N availability. In this study, I determined whether (a) poor litter C quality (that is, high litter lignin) poses an overriding constraint on decomposition, preventing decomposers from responding to added N, or (b) high N levels inhibit lignin degradation, lessening the effects of added N on decomposition overall. I obtained leaf litter from one species, Metrosideros polymorpha, which dominates a range of sites in the Hawaiian Islands and whose litter lignin concentration declines with decreasing precipitation. Litter from three dry sites had lignin concentrations of 12% or less, whereas litter from two wet sites, including the study site, had lignin concentrations of more than 18%. This litter was deployed 2.5 years in a common site in control plots (receiving no added nutrients) and in N-fertilized plots. Nitrogen fertilization stimulated decomposition of the low-lignin litter types more than that of the high-lignin litter types. However, in contrast to results from temperate forests, N did not inhibit lignin decomposition. Rather, lignin decay increased with added N, suggesting that the small effect of N on decomposition at this site results from limitation of decomposition by poor C quality rather than from N inhibition of lignin decay. Even though ANPP is limited by N, decomposers are strongly limited by C quality. My results suggest that anthropogenic N deposition may increase leaf litter decomposition more in ecosystems characterized by low-lignin litter than in those characterized by high-lignin litter. The warty cabbage Bunias orientalis is an invasive pest in much of central Europe, including much of Germany since the 1980s, whereas in other countries, such as The Netherlands, it is a less common exotic species. Here, healthy larvae of Mamestra brassicae, which has been found feeding on B. orientalis plants in Germany, and larvae parasitized by one of its major larval endoparasitoids Microplitis mediator, were reared on both herbivore-induced and noninduced leaves of B. orientalis originating from single large populations growing in The Netherlands and central Germany. Herbivore performance was less negatively affected than parasitoid performance by differences in plant quality. Development times in both M. brassicae and Mi. mediator were shorter on Dutch than German plants and also shorter on noninduced than induced plants. Moreover, survival and body size of the parasitoid was more strongly affected by plant population and induction than survival of healthy M. brassicae. Chemical analyses of defensive secondary metabolites [glucosinolates (GS)] revealed that concentrations of the major GS sinalbin were constitutively expressed in German plants whereas they were induced in Dutch plants. However, in separate feeding bioassays in which preference for induced and noninduced leaves was compared separately, L3 instars of M. brassicae preferred noninduced German plants over Dutch plants but induced Dutch plants over German plants, revealing that changes in primary metabolites or an unidentified non-GS compound mediates population-related differences in plant quality. The results reveal asymmetric effects of plant quality in exotic plants on organisms in the second and third trophic level.  相似文献

Hyphae and vesicles of arbuscular mycorrhizal fungi (AMF) were found within the decomposing leaves of Myrica parvifolia, M. pubescens and Paepalanthus sp. at three montane sites in Colombia. Hyphae, vesicles, and arbuscule-like structures were also found within scale-like leaves of the rhizomes of Paepalanthus sp. The litter found in the vicinity of the roots was divided into three decomposition layers. The highest AMF colonization occurred in the most decomposed leaves, which were in close association with roots. In contrast, there were no differences in AMF colonization of roots present in the different decomposition layers. Colonization of decomposing leaves by AMF did not differ between the two closely related species M. parvifolia and M. pubescens, nor between two sites (Guatavita and Zipacón, Colombia) differing in soil fertility. Occurrence of vesicles in decomposing leaves was correlated with abundant AMF extraradical hyphae among the leaves. We propose that AMF enter decomposing leaves mechanically through vascular tissue. As a consequence, AMF are well positioned to obtain and efficiently recycle mineral nutrients released by decomposer microorganisms before their loss by leaching or immobilization in soil.  相似文献

Brassica nigra is a newly found invasive species in Zhejiang Province, China. It distributes alongside the roads, in vegetable fields and on riversides. When it blooms, some natives there will suffer from allergic rhinitis. We designed gene-specific primer pairs according to reported profilin genes and successfully isolated their homolog from flower bud cDNA of B. nigra. The gene, designated BnPFN, was submitted to GenBank under accession number EU004073. BnPFN was 405 bp in length encoding 134 amino acids. Expression analysis of BnPFN gene was carried out by means of RT-PCR. The results showed that BnPFN express only in anthers and pollens, and there was no detection in roots, leaves, stems, sepals, petals and pistils. We suggest that BnPFN is a pollen-specific gene and may be responsible for pollen anaphylactic reactions in those invading areas when B. nigra blooms.  相似文献

In this experiment we investigate how chemical and physical attributes affect the grazing of 14 species of Hakea (Proteaceae) seedlings by western grey kangaroos (Macropus fuliginosus). Needle-leaved Hakeas were preferred over broad-leaved congeners, although differences between species within these groups were sometimes large. Needle-leaved species had smaller, thicker and spinier leaves with lower phenolic contents, but similar nitrogen contents (except for high levels in H. platysperma). The three tallest needle-leaved species, with longest, most upright and thickest leaves (H. adnata, H. obliqua and H. platysperma) were most preferred. Principal components analysis on attributes of the 14 species showed that the relative volume of shoot material consumed was significantly related (inversely) to phenolic content. Chemical defences therefore appear to have a significant deterrent role at this stage in the plant’s life history. Physical defences however were ineffective deterrents against kangaroo herbivory raising the possibility that other ecophysiological factors contribute to the possession of structures such as spines and sclerophylly in young seedlings.  相似文献

Less than a decade after being observed off Florida, the invasive Indo-Pacific lionfish is now widely distributed off the southeast coast of the United States. As a step towards measuring invasion impacts to native communities, we examine the magnitude and extent of this invasion by first, compiling reports of lionfish to provide range information and second, estimate lionfish abundance from two separate studies. We also estimate native grouper (epinepheline serranids) abundance to better assess and compare lionfish abundances. In the first study we conducted SCUBA diver visual transect surveys at 17 different locations off the North Carolina coast in water depths of 35–50 m. In the second study, we conducted 27 Remote Operated Vehicle (ROV) transect surveys at five locations from Florida to North Carolina in water depths of 50–100 m. In both studies, lionfish were found to be second in abundance only to scamp (Mycteroperca phenax). Lionfish were found in higher abundance in the shallower North Carolina SCUBA surveys ( ha−1) than in the deep water ROV surveys ( ha−1). Lionfish reports continue to expand most recently into the Bahamas, raising the specter of further spread into the Caribbean and Gulf of Mexico. The potential impacts of lionfish to native communities are likely to be through direct predation, competition and overcrowding. The high number of lionfish present in the ecosystem increases the potential for cascading impacts throughout the food chain. Within the southeast region the combined effects of climate change, overfishing and invasive species may have irreversible consequences to native communities in this region. The Chinese mitten crab, Eriocheir sinensis is a successful invader. Whilst non-breeding adult E. sinensis have been associated with the destruction of riverbanks, little is known about the ecology of this species in its invaded areas. This is especially true of the juveniles which are a key migratory stage. Intertidal surveys along the Thames estuary indicated an increase in the population since the 1990s. Juvenile E. sinensis were abundant in the sampled upper tidal region of the Thames, refuging under boulders in the intertidal at low tide. Seasonal differences in sampled populations were observed, with a significantly lower abundance of crabs found during winter compared to summer. Mark-recaptures indicated movement in the intertidal occurring during high tide, with an influx of new crabs evident after a single tidal cycle. Endogenous rhythms were also apparent, with peaks in activity occurring corresponding with night-time high tide conditions. E. sinensis juveniles can successfully exclude similar sized native Carcinus maenas from shelters in the laboratory, regardless of which species originally inhabited the shelter. This may have implications for native estuarine Carcinus populations, which can rely on such intertidal shelters for refuges.  相似文献

A reproducible plant regeneration and an Agrobacterium tumefaciens-mediated genetic transformation protocol were developed for Perilla frutescens (perilla). The largest number of adventitious shoots were induced directly without an intervening callus phase from hypocotyl explants on MS medium supplemented with 3.0 mg/l 6-benzylaminopurine (BA). The effects of preculture and extent of cocultivation were examined by assaying -glucuronidase (GUS) activity in explants infected with A. tumefaciens strain EHA105 harboring the plasmid pIG121-Hm. The highest number of GUS-positive explants were obtained from hypocotyl explants cocultured for 3 days with Agrobacterium without precultivation. Transgenic perilla plants were regenerated and selected on MS basal medium supplemented with 3.0 mg/l BA, 125 mg/l kanamycin, and 500 mg/l carbenicillin. The transformants were confirmed by PCR of the neomycin phosphotransferase II gene and genomic Southern hybridization analysis of the hygromycin phosphotransferase gene. The frequency of transformation from hypocotyls was about 1.4%, and the transformants showed normal growth and sexual compatibility by producing progenies.  相似文献

In this article we describe the identification of endophytic bacteria belonging to three groups isolated from shoot tip cultures of banana cv. Grand Naine in a recent study (Thomas et al. 2008) based on partial 16S rRNA gene sequence homology analysis. The first group included banana stocks that displayed obvious colony growth on MS based tissue culture medium during the first in vitro passage. The second group constituted stocks that were tissue index-negative for cultivable bacteria initially but turned index-positive after a few to several (4–8) in vitro passages while the third group formed one sub-stock that turned index-positive after about 18 passages. The organisms belonged to about 20 different genera comprising of α, β, γ-proteobacteria, Gram-positive firmicutes and actinobacteria. Visibly expressing easily cultured organisms during the first in vitro passage included Enterobacter, Klebsiella, Ochrobactrum, Pantoea, Staphylococcus and Bacillus spp. Organisms of second group that were not detected or non-culturable originally constituted Brevundimonas, Methylobacterium, Alcaligenes, Ralstonia, Pseudomonas, Corynebacterium, Microbacterium, Staphylococcus, Oceanobacillus and Bacillus spp. while the third group that turned cultivable after extended in vitro culturing included mostly non-filamentous actinobacteria (Brachybacterium, Brevibacterium, Kocuria and Tetrasphaera spp.). The identification results suggested that the endophytes of second and third groups were not strictly obligate or fastidious microbes but those surviving in viable but-non-culturable (VBNC) state and displaying gradual activation to cultivable form during continuous tissue culturing. Several of the organisms isolated are known as beneficial ones in agriculture while some organisms have possible implications in human health. The use of tissue cultures for isolating uncommon endophytes is discussed. We compared the abundance, population structure and palatability of bilberry ramets on vole-free islands, islands with voles but no predators (predator-free islands) and mainland sites with both voles and predators. As expected, bilberry biomass was strongly correlated with the herbivory pressure exerted by the voles, since it was significantly lower on the mainland, and much (>80%) lower on the predator-free islands, than on the vole-free islands. However, another finding, which conflicts with hypotheses postulating that herbivory generally induces plant defenses, was that voles preferred ramets from predator-free islands. Bilberry plants were fairly tolerant to grazing since they compensated for some of the lost tissue by producing more new ramets. This response should promote stability in the plant–herbivore interaction by reducing the impact of past grazing on current food production and thus minimizing time delays in the interactions that could potentially generate population cycles.  相似文献

The present paper reports a protocol for minimum growth conservation of Drosophyllum lusitanicum (L.) Link. in vitro. Double-node cuttings were maintained for 4, 8 and 12 months at 5 or 25 °C in the dark. The effects of sucrose either alone at 5, 20, 30, 40 and 60 g dm−3 or at 20, 40 and 60 g dm−3 in combination with 20 g dm−3 mannitol, on survival and post-storage shoot multiplication efficiency were investigated. The cultures could effectively be conserved under minimum growth at 5 °C for 8 months on Murashige and Skoog’s medium supplemented with 60 g dm−3 sucrose, 20 g dm−3 mannitol and 0.91 μM zeatin. Following extended conservation, the cultures could be successfully regenerated into new shoots, and they were morphologically similar to those of non-stored controls.  相似文献

An effective protocol was developed for in vitro regeneration of the Melothria maderaspatana via indirect organogenesis in liquid and solid culture systems. Organogenesis was achieved from liquid culture calluses derived from leaf and petiole explants of mature plants. Organogenic calluses (98.2?±?0.36 and 94.8?±?0.71%) were induced from both leaf and petiole explants on Murashige and Skoog (MS) liquid medium containing 6.0 µM 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 µM thidiazuron (TDZ); and 6.0 µM 2,4-D and 1.0 µM benzyladenine (BA) combinations, respectively. Adventitious shoot regeneration (68.2?±?0.06 shoots per explant) was achieved on MS medium supplemented with 2.0 µM BA, 4.0 µM TDZ, 10% v/v coconut water and 0.06 mM glutamine from leaf-derived calluses. Petiole-derived calluses produced adventitious shoots (45.4?±?0.09 shoots per explant) on MS medium fortified with 2.0 µM BA, 4.0 µM TDZ, 10% v/v coconut water, and 0.08 mM glutamine. Elongation of shoots occurred in MS medium with 2.0 µM gibberellic acid (GA3). Regenerated shoots (2–3 cm in length) rooted (74.2?±?0.38%) and hardened (85?±?1.24%) when they were transferred to 1/2-MS medium supplemented with 3.0 µM indole-3-butyric acid (IBA) followed by garden soil, vermiculate, and sand (2:1:1 ratio) mixture. The elongated shoots (4–5 cm in length) were exposed simultaneously for rooting as well as hardening (100%) in moistened [(1/8-MS basal salt solution with 5 µM IBA and 100 mg l?1 Bavistin® (BVN)] garden soil, vermiculate, and sand (2:1:1 ratio) mixture. The N2-fixing shrub Alnus viridis is currently encroaching on montane grasslands in the Alps as a result of reduced land management and complete abandonment. Alnus introduces large amounts of nitrogen (N) into these formerly N-poor grasslands and restricts the succession to montane forests. We studied pools and fluxes of N and the associated C pools in pastures (controls) and adjacent Alnus shrublands at two elevations (1650 versus 1950 m a.s.l.) in three valleys in the Swiss central Alps. The total N and C pools stored in 50-year-old Alnus shrubland did not exceed those in adjacent pastures with a total of approximately 610 g N m?2 in phytomass plus soil (down to 30 cm) at both elevations. In Alnus stands, reduced soil N pools balanced the gain in phytomass N pools, a likely result of a faster turnover of soil N. The soil solution under Alnus was continuously enriched with nitrate, with a total N leaching of 0.79 g N m?2 season?1 (June–October) under 50-year-old stands at both elevations and the highest flux of 1.76 g N m?2 season?1 in 25-year-old shrubland at low elevation, clearly indicating an excess of available N in Alnus shrubland. In contrast, N leaching across all pastures was close to zero (0.08 g N m?2) throughout the season. At the catchment scale, streamlet water showed increased nitrate concentrations with typical flushing peaks in spring and autumn, provided more than one fifth of the catchment area was covered by Alnus shrubs. We conclude that the expansion of Alnus rapidly converts centuries-old, N-poor grassland into N saturated shrubland, irrespective of elevation, and it reduces the C storage potential of the landscape because the Alnus dominance constrains re-establishment of a natural montane forest.  相似文献

Floral organ identity B class genes are generally recognized as being required for development of petals and stamens in angiosperm flowers. Spinach flowers are distinguished in their complete absence of petals in both sexes, and the absence of a developed stamen whorl in female flowers. As such, we hypothesized that differential expression of B class floral identity genes is integral to the sexual dimorphism in spinach flowers. We isolated two spinach orthologs of Arabidopsis B class genes by 3 and 5 RACE. Homology assignments were tested by comparisons of percent amino acid identities, searches for diagnostic consensus amino acid residues, conserved motifs, and phylogenetic groupings. In situ hybridization studies demonstrate that both spinach B class genes are expressed throughout the male floral meristem in early stages, and continue to be expressed in sepal primordia in reduced amounts at later stages of development. They are also highly expressed in the third whorl primordia when they arise and continue to be expressed in these tissues through the development of mature anthers. In contrast, neither gene can be detected in any stage in female flowers by in situ analyses, although northern blot experiments indicate low levels of SpAP3 within the inflorescence. The early, strong expressions of both B class floral identity genes in male floral primordia and their absence in female flowers demonstrate that B class gene expression precedes the origination of third whorl primordia (stamen) in males and is associated with the establishment of sexual floral dimorphism as it initiates in the first (sepal) whorl. These observations suggest that regulation of B class floral identity genes has a role in the development of sexual dimorphism and dioecy in spinach rather than being a secondary result of organ abortion.Electronic Supplementary Material Supplementary material is available for this article at Edited by G. Summary  The new species Eugenia pallidopunctata, E. caducibracteata, E. tenuiflora and E. breviracemosa from the Brazilian Amazon, all belonging to Eugenia sect. Racemosae (Myrtaceae), are described, illustrated and compared with their putative close relatives.
