Quantitatively evaluated C-banded and conventional karyograms are presented for the related speciesScilla bifolia subsp.danubialis Speta,S. drunensis (Speta)Speta, andS. vindobonensis Speta. On the basis of banding patterns and karyotype structureS. bifolia subsp.danubialis (2n = 18, 2×) andS. drunensis (2n = 36, 4×) are quite similar, whileS. vindobonensis (2n = 18, 2×) is entirely different. There is a moderate degree of karyotypic variation withinS. bifolia subsp.danubialis andS. drunensis. However, inS. vindobonensis karyotypes and banding patterns are almost completely stable over a geographical range of about 500 km. The present results confirm the recent taxonomic separation ofS. vindobonensis fromS. bifolia, and suggest a considerable phylogenetic distance between these two diploid species. Phylogeny of the genusAgaricusinferred from restriction analysis of enzymatically amplified ribosomal DNA.Fungal Genetics and Biology20,243–253. The 26S and 5S ribosomal RNA genes and the intergenic region between the 26S and the 5S rRNA genes of the ribosomal DNA repeat of 21 species ofAgaricuswere amplified using PCR and then digested with 10 restriction enzymes. Restriction fragment length polymorphisms were found among the 21 putative species ofAgaricusinvestigated and used to develop a phylogenetic tree of the evolutionary history ofA. bisporus.The 5′ end of the 26S gene showed more variability than the 3′ end.A. excellens, A. chionodermus,andA. carolirepresented the species most distantly related toA. bisporus.We present here the first comprehensive attempt at systematically resolving the entire genusAgaricususing modern techniques for molecular genetic analysis. A taxonomic treatment ofSalvia sect.Ekmania, a group of tall shrubs endemic to the Caribbean island of Hispaniola, is provided. Morphology, phenology, pollination biology, habitats, and distributions are discussed. Species boundaries are examined using phenetic analysis of morphological data. Eight species are here accepted as belonging to the section. Two recently described species,S. lavendula andS. paryskii, are considered for inclusion in the section, but only the latter appears to be a member.Salvia lachnaiclada andS. ottoschulzii are treated as conspecific.Salvia bahorucona is recognized as a distinct species.  相似文献

Using the AFLP technique highly informative DNA fingerprints were generated from 19 taxa ofSolanum sect.Petota (potatoes) and three taxa ofSolanum sect.Lycopersicum (tomatoes). Both phenetic and cladistic analyses were conducted from the individual genotypic level to the species level. An AFLP fingerprint, using a combination of suitable AFLP primers, generated 12 to 71 scorable fragments per genotype which was sufficient for taxonomic interpretation. The classifications based on the molecular markers were generally in agreement with current taxonomic opinions. Unexpectedly,S. microdontum was associated with ser.Megistacroloba rather than with ser.Tuberosa, andS. demissum (ser.Demissa) and species of ser.Acaulia appeared closely affiliated. AFLP is an efficient and reliable technique to generate biosystematic data and therefore a promising tool for evolutionary studies.  相似文献

We have used total genomic DNA as a probe to size-fractionated restriction enzyme digests of genomic DNA from a range ofTriticeae species from the generaLeymus Hochst.,Psathyrostachys Nevski, andHordeum L., and hybrids betweenHordeum andLeymus to investigate their taxonomic relationships. Genomic Southern hybridization was found to be an effective and simple way to assess the distribution and diversity of essentially species-specific and common, repetitive DNA sequences, and is hence especially useful in evolutionary studies. The DNA sequences ofH. vulgare seem to diverge substantially from those ofH. brachyantherum, H. lechleri, H. procerum, andH. depressum. The genome ofThinopyron bessarabicum shows little homology to those of theLeymus species investigated, confirming thatT. bessarabicum is not an ancestral genome inLeymus. Although the genomes ofLeymus andPsathyrostachys share substantial proportions of DNA sequences, they include divergent repeated sequences as well. Hybridization with a ribosomal DNA probe (pTa 71) showed that the coding regions containing structural genes encoding the 18 S, 5.8 S, and 26 S ribosomal RNA were conserved among the species investigated, whereas the intergenic spacer region was more variable, presenting different sizes of restriction fragments and enabling a classification of the species. The rye heterochromatin probe pSc 119.2 hybridized to DNA fromH. lechleri andT. bessarabicum, but not to DNA from the other species investigated.  相似文献

A study of morphometric karyotype characters was carried out in 14 species ofPeucedanum s. lat. (Umbelliferae — Apioideae) and in one species of the related genusLomatium. The differences of the species in their karyotype characters are correlated only to a limited degree with their similarities of dissimilarities in morphological (i. a. carpological) characters, and thus with their taxonomic position. The evolution of these two sets of characters seems to have proceeded not synchronously or even in different directions in the group of platycarpousUmbelliferae united in the genusPeucedanum. Therefore, it is unlikely that morphometric chromosome characters revealed by monochrome staining can be used appropriately in the taxonomic revision of the polymorphous genusPeucedanum.  相似文献

Five taxa ofSolanum sect.Basarthrum were studied in an effort to clarify their taxonomic position and to determine the effective evolutionary mechanisms. Methods included an analysis of chromosome number and behavior, artificial hybridizations and a study of herbarium material. The data suggest thatS. canense andS. suaveolens are closely related and that 5.suaveolens may have been the progenitor ofS. canense. The only successful interspecific cross involving one of these two species was vigorous but highly sterile. Evidence from both morphology and crossing studies indicates a close relationship betweenS. basendopogon andS. caripense. Hybrids between these two species with relatively high fertility through the F3 generation were secured. The status ofS. basendopogon f.obtusum remains a problem since there is but one collection of the typical form. The placement ofS. sanctae-marthae in sect.Basarthrum is considered problematic. Virtually none of 170 interspecific crosses with this species were successful. Seed size and the presence and size of a seed wing are proposed as useful morphological characters in sect.Basarthrum. There is apparently a correlation between short styles and self-compatibility. All species are diploid (n = 12) and no chromosomal or meiotic aberrations were noted in the species or hybrids. Most of the more than 1,000 interspecific crosses failed. Most of the hybrid fruits bore no seeds or seeds which did not germinate. The primary barriers separating species are considered to be strong prefertilization isolating mechanisms and ecogeographic factors.  相似文献

The systematics ofScleromitrula andVerpatinia of the family Sclerotiniaceae is reevaluated on the basis of morphological, cultural and molecular criteria.Scleromitrula shiraiana, Verpatinia species andCiborinia candolleana share gross morphological, microanatomical and cultural features which clearly distinguish them from the closely relatedCiborinia andRutstroemia species. Phylogenetic analyses of sequences from the internal transcribed spacer region (ITS1, ITS2, and the 5.8S gene) of nuclear ribosomal DNA demonstrate that the stipitate-capitate specimens ofScleromitrula andVerpatinia species plus the stipitate-cupulateCiborinia candolleana constitute a monophyletic clade separate from a clade including the type species ofCiborinia. Scleromitrula is emended to includeS. shiraiana, the new speciesS. rubicola, C. candolleana, and specimens formerly assigned toVerpatinia. Three new species ofRelbunium, R. humilioides, R. catarinense, andR. longipedunculatum (Rubiaceae), endemic in the southern states of Brazil, are described. Flavonoid patterns from two-dimensional thin layer chromatography and peroxidase patterns from gel electrophoresis are presented for two of them. We used morphological and molecular characteristics to perform a taxonomic reappraisal ofSuaeda australis (Brown) Moquin-Tandon from Korea. Populations of this species are dispersed at the bottoms of sand zones, and usually exhibit a depressed habit. Except for their total heights and leaf lengths, the morphology of these plants does not differ from that ofS. maritima. Molecular traits were examined based on ITS and psbB-psbH spacer region sequences. The former region included ITS-1, 5.8S, and ITS-2, which were 629 nucleotide bases long. Pair-wise distances (p-distance) among KoreanSuaeda species ranged from 1.12 to 17.84. The psbB-psbH spacer region sequences were 618 nucleotide bases long, and were conserved in the alignment of KoreanSuaeda species. In our ML and MrBayesian analysis of ITS sequences aligned with other sequences from GenBank, the plants of KoreanSuaeda made three clades: 1)S. japonica;S. australis, andS. maritima;2)S. malacosperma; and 3) S.glauca. However, the psbB-psbH region sequences could not be resolved amongS. japonica, S. maritima, andS. australis from Korea. Molecular and vegetative characteristics indicated that the plants now classified asS. australis from Korea should instead be named as S.maritima (L.) Dumont.  相似文献

Summary The existence of bristles, the hair-like appendages in several species of the genus Scenedesmus has been confirmed.The ultrastructure of bristles in Scenedesmus helveticus Arlet, Sc. acutus Meyen and Sc. acuminatus/Lagerh./Chodat has been studied by negative staining. While bristles of Sc. helveticus are composed of several microstrands with a regular pattern of cross-striation, the bristles of spineless species Sc. aculus and Sc. acuminatus seem to be composed only of individual microstrands.These investigations of bristles suggest their protein composition.  相似文献

Guatteria, Guatteriopsis, Guatteriella andHeteropetalum share the same conspicuous pollen type which is new for theSpermatophyta. It is zonoaperturate with a folded aperture region and an extremely reduced exine. First chromosome counts and karyotype analyses forGuatteriopsis (4 species investigated) andGuatteriella (1 species) are identical with those ofGuatteria (19 species seen): 2n = 28. The genome is characterized by diploidization and partly telocentric chromosomes. Sequentially Giemsa C- and fluorochrome banded chromosomes and interphase nuclei are described. The cuticular folding pattern is distinct forHeteropetalum only. Growth forms and ecology are reported for many species. The evolutionary pattern of theGuatteria group is discussed and compared with other genera and families.  相似文献

The roles of hybridization and of introgression as potential threats to small native populations are current topics of interest both for evolutionary and conservation biologists. Two factors are mainly involved in the threat to natural populations through hybridization: demographic swamping and genetic assimilation. At prealpine locations, twoSaxifraga species with very different growth habits,S. aizoides andS. mutata, form a hybrid,S. ×hausmannii. Investigation of meiosis and crossing experiments revealed regular pairing of the 26 chromosomes and development of viable seeds in the hybrid. Analysis of hybrid indices, GPI allozymes, germination experiments and seed sizes showed an intermediate position ofS. ×hausmannii between its parental species.Saxifraga aizoides andS. mutata were separated by only weak hybridization barriers. Introgression occurred mostly in direction ofS. aizoides, although to a small amount. There is no actual threat to small, prealpine populations ofS. aizoides andS. mutata through interspecific gene flow because hybridization is localized, resulting from different flowering phenologies of the parental species and from the narrow habitat requirements ofS. ×hausmannii, which are similar to those ofS. aizoides. The major threat to local, prealpine populations ofS. aizoides andS. mutata is the loss of landscape dynamics. Both species need open erosion slopes for successful recruitment.  相似文献

This work examines the cytogeographical distribution, the morphological characters, and the karyotypes of threeCrepis species endemic to Greece (C. sibthorpiana, C. incana, andC. heldreichiana). C. sibthorpiana is diploid (2n = 2x = 8),C. incana is diploid (2n = 2x = 8) and tetraploid (2n = 4x = 16, 17), andC. heldreichiana is always dekaploid (2n = 10x = 40). The Giemsa positive bands, usually pairs of dots, are mainly centromeric inC. incana, while they are terminal inC. sibthorpiana (on the short arm of all chromosomes) and inC. heldreichiana (on both arms of all chromosomes). Intercalary C-bands are scarce and usually variable within karyotypes, individuals, and species. The most variable karyotype both in Feulgen and Giemsa preparations is that ofC. incana, in which also supernumerary chromosomes were observed, which are polysomic to standard set members. On the basis of morphological and karyological data the evolutionary relationships among the threeCrepis taxa are discussed.  相似文献

Chloroplast DNA of seven species belonging toAnemone (sectt.Omalocarpus, Anemonidium, andAnemonanthea),Hepatica, andPulsatilla have been analyzed by restriction enzymes. According to the dendrogram constructed, the sections ofAnemone and the generaHepatica andPulsatilla seem to be evolutionary approximately equidistant to each others. This supports the concept that these groups should be treated on a similar taxonomic level, either as genera or subgenera.  相似文献

Karyomorphological comparisons were made of 16 native and cultivated species ofSelaginella in Japan. The somatic chromosome numbers are 2n=16 inS. boninensis; 2n=18 inS. doederleinii, S. helvetica, S. limbata, S. lutchuensis, S. nipponica, S. selaginoides, S. tama-montana, andS. uncinata; 2n=20 inS. biformis, S. involvens, S. moellendorffii, S. remotifolia, andS. tamariscina; 2n=30 inS. rossii; and 2n=32 inS. heterostachys. The interphase nuclei of all species examined are uniformly assigned to the simple chromocenter type. The metaphase karyotype of 2n=16 (x=8) is 8 m (=median centromeric chromosomes)+8(st+t)(=subterminal and terminal). The group of the species having 2n=18 (x=9) is heterogeneous karyomorphologically: The karyotype ofS. nipponica is 2n=18=6 m+12(st+t),S. tama-montana 10 m+2 sm(=submedian)+6(st+t), andS. uncinata 6 m+7 sm+5(st+t). Although the remaining five species have the common karyotype 8 m+4 sm+6(st+t), the values of mean chromosome length are variable. Another group of the specles having 2n=20 (x=10) is homogeneous, since all species have the same karyotypes 8 m+4 sm+8(st+t) and have similar chromosome size. The karyotype of 2n=30 is 12 m+6 sm+12(st+t) and is suggested to be a triploid of x=10, and 2n=32=16m+16(st+t), a tetraploid of x=8. Thus, three kinds of basic chromosome numbers, x=8, 9, 10 are present in JapaneseSelaginella examined, and their karyomorphological relationships are discussed.  相似文献

A total of 86 adult bluethroats (Luscinia svecica) from nine different localities, covering the full length of the Fennoscandian mountain range, were screened for blood parasites of the three genera Haemoproteus, Plasmodium and Leucocytozoon using a recently developed polymerase chain reaction method. The overall occurrence of infection was 59.3%. Prevalence of Leucocytozoon spp. (47.7%), Plasmodium spp. (23.3%) and Haemoproteus spp. (1.2%) was detected. Of the infected birds, 15.1% carried mixed infections. Five different mitochondrial DNA-lineages of Leucocytozoon spp., eight lineages of Plasmodium spp. and one lineage of Haemoproteus spp. were found. Due to large sequence divergence these corresponded to at least five different species, but with the possibility of all 14 being independent evolutionary units with the potential of evolving different effects on the host. Of the lineages of Leucocytozoon spp., the most common was found throughout the range. The occurrence of the second most common lineage of Leucocytozoon spp. showed significant variation in prevalence between sites. The data also showed molecular evidence of one lineage of Leucocytozoon sp. existing in more than one species of avian host, thus challenging the use of host taxon as a taxonomic character when distinguishing between different species leucocytozoids.Communicated by F. The term house-dust mite usually refers to those species of the mite family Pyroglyphidae, that are known to commonly occur, although sometimes regionally, in the dust of human dwellings. These species belong to five genera:Dermatophagoides, Euroglyphus, Hirstia, andMalayoglyphus. Related species ofDermatophagoides have the most world-wide occurrence, the commonest being:D. farinae, D. microceras, andD. pteronyssinus. A correct taxonomic identification of house-dust mites is very important, not only from a biological stand point, but also regarding the consequences of their respective allergenical properties. Several immuno-chemical studies revealed differences between the products of two hard to distinguish sibling species. A preliminary practical taxonomic key for the most common and important house-dust mites is presented.  相似文献

The karyotype analysis ofPeumus boldus reveals a remarkable differentiation of chromosomes in size and form. C-banding pattern and interphase nuclei are discribed and compared with other genera. The great variation in leaf-shape and growth-form is continuous and correlated with the very diverse habitats of the species. The recognition of infraspecific taxa is therefore not recommended. The lower leaf surface is very uniform within the monotypic genus and ± similar with that ofSiparuna. The different evolutionary strategies ofPeumus and related genera are discussed and show a different eco-systematic situation than inMollinedia andSiparuna.
