The role of gill chloride cells (CCs) and pavement cells (PVCs) in acid-base regulation was evaluated in brown bullhead catfish (Ictalurus nebulosus) subjected to acute hypercapnia (water Pco2=15 torr). Chronic (10 day) cortisol treatment was used as a tool to cause CC proliferation to permit a comparison of the regulatory capacities in groups of fish with widely different gill CC populations. Cortisol (4mg kg?1 day?1) caused a pronounced increase (170%) in the surface area of CCs exposed to the water based on scanning and transmission electron microscope analysis. The density of PVC apical membrane microvilli was significantly increased (20%) by cortisol treatment. Exposure of either group of fish to hypercapnia caused similar changes in gill epithelial morphology including: (i) a marked reduction in the surface area of exposed CCs (52 and 78% reduction in the control and cortisol-treated fish, respectively); and (ii) pronounced increases in PVC apical membrane microvilli density (21 and 27% in the control and cortisol-treated fish, respectively). The rates of Cl? uptake (Jincl?) and Na+ uptake (JinNa+) were elevated (150 and 262%, respectively) in the cortisol-treated fish. Regardless of treatment, Jincl? was markedly reduced to approximately the same levels after 6 h of hypercapnia, JinNa+ was stimulated in the control group and reduced in the cortisol-treated group and thus, after 6 h of hypercapnia, JinNa+ was equal in each group. The similar morphological responses in fish possessing different initial populations suggests that the predominant mechanism of acid-base regulation during hypercapnia, reduction of C1?/HCO3? exchange, is accomplished by removal of the CC-associated C1-/HCO3? exchange sites from the water. The increase in PVC microvilli density during hypercapnia suggests a role for the PVC in acid-base regulation.  相似文献

To evaluate the role of the gill chloride cells in regulating metabolic alkalosis in rainbow trout (Oncorhynchus mykiss), the surface area of branchial chloride cells was altered experimentally using combined cortisol/ovine growth hormone injections. Long-term (10-day) treatment of fish with cortisol/ovine growth hormone caused an increase in the two-dimensional chloride cell fractional surface area when compared to uninjected fish (from 8.4 to 29.7%). This was the combined result of an increase in the size of individual cells (from 34.6 to 59.2 m2) and increased numbers of cells (from 2368 to 5006 cells · mm-2). Metabolic alkalosis was induced by intra-arterial infusion of 140 mmol · l-1 NaHCO3; control fish were infused with 140 mmol · l-1 NaCl. Blood pH and plasma [HCO3 -] increased in both the untreated and the cortisol/ovine growth hormone-treated fish. However, the increases in pH (from 8.05 to 8.53) and [HCO3 -] (from 5.9 to 22.2 mmol · l-1) in the untreated fish were significantly greater than in the cortisol/ovine growth hormone-treated fish (pH increased from 7.78 to 8.11; [HCO3 -] increased from 5.5 to 13.9 mmol · l-1). In all fish, NaHCO3 infusion elicited an increase in the rate of branchial basic equivalent excretion (acidic equivalent uptake) which, in turn, was caused by decreases and increases in branchial Na+ uptake and Cl- uptake, respectively. In the untreated fish, there was a pronounced increase (75%) in chloride cell surface area during NaHCO3 infusion. The attenuation of the metabolic alkalosis during HCO3 - infusion in the cortical/ovine growth hormone-treated fish was caused, at least in part, by an enhancement of branchial basic equivalent excretion. In these fish that already displayed a proliferation of chloride cells, there was no further increase in chloride cell surface area. The changes in Na+ influx and Cl- influx were quantitatively similar during NaHCO3 infusion in both groups. Whole body calcium influx, branchial calcium efflux, and renal Ca2+ excretion were measured in rainbow trout (Oncorhynchus mykiss) exposed to hypercapnia. These experiments were performed to assess the potential impact on Ca2+ balance of the changes in gill morphology known to accompany respiratory acidosis in this species. After 48 h of hypercapnia, gill filamental chloride cell fractional area was significantly reduced. Despite this reduction and the presumed involvement of the chloride cell in calcium influx, whole body calcium influx was increased after 12 h of hypercapnia and remained elevated for 48 h. Branchial calcium efflux was unaltered during hypercapnia exposure, whereas renal Ca2+ excretion was elevated over preflux values only at 6 h of hypercapnia. Measurement of the kinetics of whole body calcium influx after 48 h of hypercapnia revealed a significant increase in the maximal uptake rate of Ca2+, yet the affinity constant of Ca2+ uptake was unaffected. Measurements of high-affinity Ca2+ -ATPase activities and ATP-dependent Ca2+ transport of gill basolateral membrane vesicles revealed that the ATP-dependent Ca2+ extrusion mechanism of the gills was not affected by hypercapnia. The results of the present study clearly show that the reduced chloride cell surface area that accompanies hypercapnia in trout does not impair calcium homeostasis. Although adjustments to the basolateral membrane high affinity Ca2+ transporter do not appear to play a role, the mechanism(s) underlying the maintenance of calcium homeostasis under hypercapnic conditions are unresolved. Summary Daily intramuscular injection of cortisol (4 mg kg–1 body weight) in rainbow trout,Salmo gairdneri, for 10 days caused significant increases in the number and individual apical surface area of gill chloride cells per mm2 of filament epithelium. Concomitantly, whole body influxes of sodium (Na+) and chloride (Cl) increased. Acute (3 h) intra-arterial infusion of cortisol did not affect whole body Na+ or Cl influx. A significant correlation was observed between both Na+ and Cl influxes and the fractional apical surface area of filament chloride cells in control, sham (saline-injected) and experimental (cortisol-injected) fish. The chloride cells displayed similar ultrastructural modifications in trout undergoing cortisol treatment as in trout transferred to ion-deficient water. These findings suggest the existence of structure/function relationships in which branchial chloride cell morphology is an important determinant of Na+ and Cl transport capacity. We conclude that chronic cortisol treatment enhances whole body Na+ and Cl influxes by promoting proliferation of branchial chloride cells. The results of correlation analysis indicate that the chloride cell is an important site of NaCl uptake in freshwater rainbow trout.  相似文献

This review examines the branchial mechanisms utilized by freshwater fish to regulate internal acid-base status and presents a model to explain the underlying basis of the compensatory processes. Rainbow trout, Oncorhynchus mykiss, and brown bullhead, Ictalurus nebulosus, were examined under a variety of experimental treatments which induced respiratory and metabolic acid-base disturbances. Acid-base regulation was achieved by appropriate adjustments of Na+ and Cl- net fluxes across the gills which, in turn, were accomplished by variable contributions of three different branchial mechanisms: 1) differential changes in Na+ and Cl- diffusive effluxes, 2) changes in internal substrate (H+, HCO3-) availability, and 3) morphological adjustments to the gill epithelium. Differential diffusive efflux of Na+ over Cl- was involved only during periods of metabolic alkalosis. The importance of internal substrate availability was demonstrated using a two-substrate model. According to the model, ionic flux rates (J(in)Cl-, J(in)Na+) are determined not only by the concentration of the external ion (Na+, Cl-) but also by the concentration of the internal counterion (H+, HCO3-). This system provides for an "automatic negative feedback" to aid in the compensation of metabolic acid-base disturbances. Morphological alteration of the gill epithelia and the associated regulation of chloride cell (CC) fractional area is an essential third mechanism which is especially important during respiratory acid-base disturbances. Specifically, fish vary the availability of the CC associated Cl-/HCO3- exchange mechanism by physical covering/uncovering of CCs by adjacent pavement cells.  相似文献

Summary Bioelectrical parameters and unidirectional sodium and chloride fluxes were measured under voltageclamp conditions in groups of lizards submitted to single or chronic aldosterone treatment. Both acute (AT) and chronic (CT) treatment induced significant increases in the short-circuit current (I sc), as well as in the mucosa-to-serosa (J m-s Na ) and net sodium flux (J net Na ). In AT tissues, aldosterone did not change net chloride flux (J net Cl ) but did so in CT tissues. Amiloride reduced the aldosterone-increased I sc in AT and CT tissues, inhibited J net Na in AT tissues and abolished it in CT colons. J net Cl was also reduced by the diuretic in the group of AT colons, whereas no changes were observed in the CT tissues. Addition of luminal DIDS reduced Na+ absorption and totally inhibited Cl- absorption in the AT tissues, but did not change I sc. However, in CT tissues neither Na+ nor Cl- transport were affected by DIDS. A good relationship between I sc and J m-s Na was apparent after DIDS treatment in AT tissues. In this group, simultaneous addition of DIDS and amiloride totally abolished J net Na and reduced I sc to untreated control values. Addition of serosal ouabain abolished I sc and Na+ absorption in AT and CT colons, but Cl- absorption was only altered in AT tissues. Summary The responses of net and unidirectional fluxes of Na+ and acid-base balance to the drug amiloride were assessed during normocapnia and hypercapnia in larval salamanders, Ambystoma tigrinum. Isotope flux measurements demonstrated that 10-4 M amiloride in the external medium inhibits Na+ influx during normocapnia and reverses the usual increase in influx of this ion during hypercapnia, causing a significant decrease instead. Ion and acid–base balance were examined in the freshwater-adapted mummichog (Fundulus heteroclitus) using a series of treatments designed to perturb the coupling mechanisms. Unidirectional Cl uptake (JClin) was extremely low whereas JNain was substantial (three- to sixfold higher); comparable differences occurred in unidirectional efflux rates (JClout, JNaout). JClin was refractory to all treatments, suggesting that Cl/base exchange was unimportant or absent. Indeed, no base excretion or modulation of ion fluxes occurred for acid–base balance for up to 8 h after NaHCO3 loading (injections of 1000 or 3000 nequiv.·g−1). Acute environmental low pH (4.5) and amiloride (10−4 M) treatments caused concurrent inhibition of JNain and net H+ excretion (JH+net), indicating the presence of Na+/H+ exchange. JNain was elevated and JH+net restored during recovery from both treatments, but this exchange did not appear to be dynamically adjusted for acid–base homeostasis. High external ammonia exposure (1 mmol·l−1) initially blocked ammonia excretion (JAmmnet) but had no effect on JNain, whereas high pH (9.4) reduced both JAmmnet and JNain. Inhibition of JNain by the low pH and amiloride treatments had no effect on JAmmnet. These results indicate that ammonia excretion is entirely diffusive and independent of both Na+uptake and the protons that are transported via the Na+/H+ coupling. In addition, ureagenesis served as a compensatory mechanism during high external ammonia exposure, as a marked elevation in urea excretion partially replaced the inhibited JAmmnet. In all treatments, changes in the Na+–Cl net flux differential were consistent with changes in JH+net measured by traditional water titration techniques, indicating that the former can be used as an estimate of the acid–base status of the fish. Overall, the results demonstrate that the freshwater-adapted F. heteroclitus does not conform to the ion/acid–base relationships described in the standard model based on commonly studied species such as trout, goldfish, and catfish.  相似文献

Rainbow trout that were held under control conditions, at pH8·0, in moderately hard Hamilton tap water, had Cl? and Na+ influx rates (JCLin and JNa, respectively) of 270 and 300 μmol kg?1 h?1, respectively. Exposure to pH 9·5 water led to an immediate 67% decline in JCLin and a 45% reduction in JNain at 0–1 h. Influx rates declined further and by 4–5 h the net decreases in both JCLin and JNain approximated 80%. By 24 h JCLin had recovered to rates not significantly different from those at pH 8·0; while JNain only partially recovered and remained about 50% lower than control measurements through 72 h. The complete recovery of JCLin and partial recovery of JNain may have been related to a fourfold greater branchial chloride cell (CC) fractional surface area observed in rainbow trout exposed to pH 9·5 for 72 h. Ammonia excretion (JAmm) was about 170 μmol N kg?1 h?1 at pH 8·0 but was initially reduced by 90% over the first hour of high pH exposure. JAmm rapidly recovered and by 24 h it had returned to pre-exposure levels. This recovery tended to parallel the partial recovery of JNain. However, subsequent addition of amiloride (10?4M) to the water at 75 h led to no change in JAmm, despite a 50% reduction in JNain. Thus, it does not appear that there is a linkage between Na+ influx and the recovery of ammonia excretion under highly alkaline conditions.  相似文献

In most reptiles, the ventilatory response to hypercapnia consists of large increases in tidal volume (V(T)), whereas the effects on breathing frequency (f(R)) are more variable. The increased V(T) seems to arise from direct inhibition of pulmonary stretch receptors. Most reptiles also exhibit a transitory increase in ventilation upon removal of CO(2) and this post-hypercapnic hyperpnea may consist of changes in both V(T) and f(R). While it is well established that increased body temperature augments the ventilatory response to hypercapnia, the effects of temperature on the post-hypercapnic hyperpnea is less described. In the present study, we characterise the ventilatory response of the agamid lizard Uromastyx aegyptius to hypercapnia and upon the return to air at 25 and 35 degrees C. At both temperatures, hypercapnia caused large increases in V(T) and small reductions in f(R), that were most pronounced at the higher temperature. The post-hypercapnic hyperpnea, which mainly consisted of increased f(R), was numerically larger at 35 compared to 25 degrees C. However, when expressed as a proportion of the levels of ventilation reached during steady-state hypercapnia, the post-hypercapnic hyperpnea was largest at 25 degrees C. Some individuals exhibited buccal pumping where each expiratory thoracic breath was followed by numerous small forced inhalations caused by contractions of the buccal cavity. This breathing pattern was most pronounced during severe hypercapnia and particularly evident during the post-hypercapnic hyperpnea.  相似文献

Summary The novel application of a two-substrate model (Florini and Vestling 1957) from enzymology to transport kinetics at the gills of freshwater trout indicated that Na+/acidic equivalent and Cl-/basic equivalent flux rates are normally limited by the availability of the internal acidic and basic counterions, as well as by external Na+ and Cl- levels. Adult rainbow trout fitted with dorsal aortic and bladder catheters were chronically infused (10–16 h) with isosmotic HCl to induce a persistent metabolic acidosis. Acid-base neutral infusions of isosmotic NaCl and non-infused controls were also performed. Results were compared to previous data on metabolic alkalosis in trout induced by either isosmotic NaHCO3 infusion or recovery from environmental hyperoxia (Goss and Wood 1990a, b). Metabolic acidosis resulted in a marked stimulation of Na+ influx, no change in Cl- influx, positive Na+ balance, negative Cl- balance, and net H+ excretion at the gills. Metabolic alkalosis caused a marked inhibition of Na+ influx and stimulation of Cl- influx, negative Na+ balance, positive Cl- balance, and net H+ uptake (=base excretion). Mean gill intracellular pH qualitatively followed extracellular pH. Classical one-substrate Michaelis-Menten analysis of kinetic data indicated that changes in Na+ and Cl- transport during acid-base disturbance are achieved by large increases and decreases in Jmax, and by increases in Km. However, one-substrate analysis considers only external substrate concentration and cannot account for transport limitations by the internal substrate. The kinetic data were fitted successfully to a two-substrate model, using extracellular acid-base data as a measure of internal HCO 3 - and H+ availability. This analysis indicated that true Jmax values for Na+/acidic equivalent and Cl-/basic equivalent transport are 4–5 times higher than apparent Jmax values by one-substrate analysis. Flux rates are limited by the availability of the internal counterions; transport Km values for HCO 3 - and H+ are far above their normal internal concentrations. Therefore, small changes in acid-base status will have large effects on transport rates, and on apparent Jmax values, without alterations in the number of transport sites. Summary The action of poly-l-lysine (PLL) on Na and Cl transport across freshwater fish gills was studied. Low concentrations (10–6M) were added to the external medium for brief periods (1–5 min), then removed. During the next 20 min there was a rapid net loss of Na (117±17 Equiv[100 g]–1 hr–1) and Cl (129±17 Equiv[100 g]–1 hr–1). Both values are an order of magnitude larger than unidirectional effluxes in control fish. The efflux of both ions decreased to control values within 60 min after application and removal of PLL. In contrast, unidirectional influxes (J in Cl andJ in Na ) were inhibited by about 40% and showed no sign of returning to the original rates for 3 hr. Thus, PLL has two independent actions, causing a large increase in gill permeability which is reversible within an hour and a partial inhibition of influx which showed no sign of reversing for 2–3 hr. When PLL was applied for a longer period (60 min, the results were qualitatively similar but the permeability change was larger and persisted longer. These effects were compared with those of the small organic amines, amiloride and methyl-l-lysine. The latter inhibitedJ in Na , but there was no other similarity to PLL. Neither affected sodium efflux, nor did they have any effect on Cl movements, in or out, across the gill. Inhibition ofJ in Na , was rapidly and completely reversible, amd amiloride was shown to act by competing with Na for an entry site.  相似文献

Summary In a previous study we presented evidence that chloride transport across the basolateral membrane inNecturus proximal tubule cells occurs predominantly via exchange for both Na+ and HCO 3 . In this study the regulation of intracellular chloride was further examined in the doubly-perfused kidney preparation using conventional and chloride-sensitive microelectrodes. Application of hypertonic basolateral solutions containing 80mm raffinose stimulated an efflux of chloride such that chloride activity remained unchanged at control levels. Membrane potential did not change in these experiments. Inhibition of Cl exit across the basolateral cell membrane by removal of either HCO 3 or Na+ from the perfusion solution resulted in a significant increase in intracellular chloride activity,a Cl i , when basolateral osmolarity was raised. Hypertonic basolateral solutions also produced a significant rise ina Cl i in the presence of SITS.This study provides further evidence that chloride is transported across the basolateral cell membrane in exchange for both Na+ and HCO 3 . Since this exchange mechanism is activated in response to hypertonic solutions, these studies suggest a functional role for this exchanger in the regulation ofa Cl i in theNecturus proximal tubule cell during volume changes.  相似文献

Summary Two methods, the measurement of the response of the basolateral membrane potential (V bl) of proximal tubule cells ofNecturus to step changes in basolateral K+ concentration, and cellular cable analysis, were used to assess the changes in basolateral potassium conductance (G K) caused by a variety of maneuvers. The effects of some of these maneuvers on intracellular K+ activity (a K i ) were also evaluated using double-barreled ion-selective electrodes. Perfusion with 0mm K+ basolateral solution for 15 min followed by 45 min of 1mm K+ solution resulted in a fall in basolateral potassium (apparent) transference number (t K),V bl anda K i . Results of cable analysis showed that total basolateral resistance,R b , rose. The electrophysiological effects of additional manipulations, known to inhibit net sodium reabsorption across the proximal tubular epithelium ofNecturus, were also investigated. Ouabain caused a fall int K accompanied by large decreases ina K i andV bl. Lowering luminal sodium caused a fall int K and a small reduction inV bl. Selective reduction of peritubular sodium, a maneuver that has been shown to block sodium transport from lumen to peritubular fluid, also resulted in a significant decrease int K. These results suggest thatG K varies directly with rate of transport of the sodium pump, irrespective of the mechanism of change in pump turnover.Part of this material has been presented at the 10th International Conference on Biological Membranes (Cohen & Giebisch, 1984).  相似文献

Summary Segments of descending colon obtained from rabbits, that had been maintained on drinking water containing 25mm NaCl and an artificial diet which contains 1% Na and is nominally K-free, respond to aldosterone in vitro (after a 30 to 60-min lag period) with a marked increase in the short-circuit current (I sc ), an equivalent increase in the rate of active Na absorption (J net Na ) and a decline in tissue resistance (R t ). Aldosterone also brings about a marked increase in the unidirectional influx of Na into the cells across the mucosal membrane (zero-time rate of uptake) which does not differ significantly from the increase inI sc . Treatment of control tissues with amphotericin B brings about sustained increases inI sc andJ net Na to levels observed in aldosterone-treated tissues. However, addition of amphotericin B to the mucosal solution of aldosteronetreated tissues does not result in a sustained increase inI sc orJ net Na and these values do not differ markedly from those observed in control tissues treated with amphotericin B. These findings, together with other evidence that Na entry in the presence of amphotericin B is sufficiently rapid to saturate the active Na extrusion mechanism at the baso-lateral membrane, are consistent with the notion that the aldosterone-induced protein increases the permeability of the mucosal membrane to Na but does not increase the saturation level of the active Na pump within the time-frame of these studies (3 hr).Finally, aldosterone has no effect on the bidirectional or net transepithelial movements of K under short-circuit conditions, suggesting that the enhanced secretion of K observed in vivo is the result of increased diffusion of K from plasma to lumen via paracellular pathways in response to an increased transepithelial electrical potential difference (lumen negative).  相似文献

Summary The unidirectional influx of Na from the mucosal solution into the epithelium ofin vitro descending rabbit colon (J me Na ) determined under short-circuit conditions, is comprised of two components: one represents entry of Na into transporting epithelial cells and is abolished by amiloride which also abolishes Na absorption (J net Na ). The other represents diffusional Na entry into paracellular pathways traversing the epithelium. In all instances, exposure of the mucosal surface to amphotericin B increased tissue conductance andJ me Na and elicited K secretion. Tissues showing a spontaneousI sc of approximately 4 eq/cm2hr did not respond to amphotericin B with increasedI sc andJ net Na . However, in tissues characterized by a lowerI sc under control conditions, amphotericin B increasedI sc andJ net Na to approximately 4eq/cm2hr. These findings suggest that amphotericin increasesJ net Na and elicits K secretion by disrupting the normal permselectivity of the mucosal membrane. Under these conditions the extrusion of Na from cell-to-serosal solution becomes the rate limiting step in transepithelial Na transport. Finally, a close correlation betweenJ me Na andJ net Na was observed when the rate of Na absorption varied either spontaneously or experimentally with amiloride, suggesting that the backflux of Na from cell-to-mucosal solution is undetectably small.  相似文献

Summary The Na/K/Cl-dependent component of the binding of the loop diuretic bumetanide to basolateral membrane vesicles from the rabbit parotid is studied. A Scatchard analysis indicates that this binding is due to a single high-affinity site withK D =3.2±0.3 m (n=9) at 100mm sodium, 100mm potassium and 5mm chloride. When KCl-dependent22Na transport and tracer [3H]-bumetanide binding are monitored simultaneously as a function of (unlabeled) bumetanide concentration it is found that theK 0.5 for bumetanide inhibition of both processes are identical indicating that the high-affinity bumetanide binding site studied here is identical with a bumetanide-inhibitory site on the Na/K/Cl cotransport system previously identified in this preparation (R.J. Turner, J.N. George and B.J. Baum,J. Membrane Biol. 94:143–152, 1986). High-affinity bumetanide binding exhibits a hyperbolic dependence on both [Na] and [K] consistent with Na/bumetanide and K/bumetanide binding stoichiometries of 11 andK 0.5 values of approximately 33mm for sodium and 23mm for potassium. In contrast, the dependence on [Cl] is biphasic, with bumetanide binding increasing from 0 to 5mm chloride and decreasing toward baseline levels thereafter. Scatchard analysis of this latter inhibitory effect of chloride indicates a competitive interaction with bumetanide in agreement with earlier indications that bumetanide inhibits Na/K/Cl cotransport at a chloride site. However, studies of the effects of various anions on bumetanide binding and22Na transport show a poor correlation between the specificities of these two processes, suggesting that the inhibitory chloride site is not a chloride transport site.  相似文献

Summary Na and Cl fluxes and short-circuit current (I sc) in rabbit ileum have been studied as a function of ionic concentrations in HCO3-free solutions. Both net Na flux (J net Na ) andI sc show similar saturation functions of [Na] at fixed [Cl]. They show no significant difference between zero and 112mm Na but at 140mm NaI sc is significantly greater than theJ net Na . Net Cl transport, secretion, is observed only at 140mm Na and is approximately equivalent to the difference between theI sc andJ net Na . The transcellular mucosa-to-serosa Na fluxes measured at 140 and 70mm Na do not differ significantly from the correspondingI sc. The net Cl flux varies with [Cl] at fixed [Na] whileI sc is virtually not affected by [Cl]. These results suggest that the absorptive Na transport process is electrogenic and responsible for theI sc and that the secretory fluxes of Na and Cl are coupled, require high [Na], vary with [Cl], and do not contribute toI sc. K-free solution abolishes theI sc after a prolonged lag. Crayfish in which sodium absorption was maximally stimulated had elevated levels of both cAMP and Na+-K+-ATPase activity in gill tissue. The concentration of cAMP and activity of Na+-K+-ATPase in gill tissue were monitored following transfer of crayfish from water containing 125 mmol.l−1 Na to Na-free media. Both parameters were significantly elevated within 10 min of transfer to Na-free media and [cAMP] peaked between 1 and 2 h before falling transiently to the control level at 3 h. A second peak of [cAMP] and a further rise in Na+-K+-ATPase activity were evident 6 h after transfer and elevated levels were then maintained. The pattern observed was consistent with the existence of two separate mechanisms for the control of sodium absorption both of which stimulated the activity of Na+-K+-ATPase via elevation of the intracellular concentration of cAMP. The initial response was very rapid (<10 min) but of brief duration (1–2 h) and this mechanism appeared to be sensitive to changes in external ion levels. The second mechanism exhibited a much longer response time (3–6 h) and duration and was likely to be sensitive to changes in internal ion concentrations.  相似文献

The branchial osmoregulatory response of gilthead sea bream (Sparus auratus L.) to short-term (2-192 hr) and long-term (2 weeks) exposure to different environmental salinities (5 per thousand, 15 per thousand, 25 per thousand, 38 per thousand and 60 per thousand) was investigated. A "U-shaped" relationship was observed between environmental salinity and gill Na+,K+ -ATPase activity in both long- and short-term exposure to altered salinity, with the increase in activity occurring between 24 and 96 hr after the onset of exposure. Plasma osmolality and plasma ions (sodium, chloride, calcium and potassium) showed a tendency to increase in parallel with salinity. These variables only differed significantly (P<0.05) in fish adapted to 60 per thousand salinity with respect to fish adapted to full-strength sea-water (SW). Plasma glucose remained unchanged whereas plasma lactate was elevated at 5 per thousand and 60 per thousand. Muscle water content (MWC) was significantly lower in fish adapted to 60 per thousand. Chloride cells (CC) were only present on the surface of the gill filaments and absent from the secondary lamellae. CC distribution was not altered by external salinity. However, the number and size of CC were significantly increased at salinity extremes (5 per thousand and 60 per thousand), whereas fish exposed to intermediate salinities (15 per thousand and 25 per thousand) had fewer and smaller cells. Furthermore, the CC of fish exposed to diluted SW became rounder whereas they were more elongated in fish in full-strength and hypersaline SW. This is consistent with previous reports indicating the existence of two CC types in euryhaline fish. At likely environmental salinities, gilthead sea bream show minor changes in plasma variables and the effective regulation of gill Na+,K+ -ATPase. However, at very low salinities both haemodilution and up-regulation of gill Na+,K+ -ATPase predict a poor adaptation most likely related to deficiency or absence of specific components of the CC important for ion xuptake.  相似文献
