A method is described for the qualiitative and quantitative determination of phenylbutazone and oxyphenbutazone in horse urine and plasma samples viewing antidoping control. A horse was administered intravenously with 3 g of phenylbutazone. For the qualitative determination, a screening by HPLC was performed after acidic extraction of the urine samples and the confirmation process was realized by GC-MS. Using the proposed method it was possible to detect phenylbutazone and oxyphenbutazone in urine for up to 48 and 120 h, respectively. For the quantitation of these drugs the plasma was deproteinized with acetonitrile and 20 gml were injected directly into the HPLC system equipped with a UV detector and LiChrospher RP-18 column. The mobile phase used was 0.01 M acetic acid in methanol (45:55, v/v). The limit of detection was 0.5 μg/ml for phenylbutazone and oxyphenbutazone and the limit of quantitation was 1.0 μg/ml for both drugs. Using the proposed method it was possible to quantify phenylbutazone up to 30 h and oxyphenbutazone up to 39 h after administration.  相似文献

The circadian variation of edema produced by carrageenin (carr.) administration into plantar tissue was studied in rats kept under a 12 light - 12 dark regimen. Three doses were used (125, 250 and 500 micrograms per rat) injected at different time (02.00, 08,00, 14.00 and 20.00 h). With the high doses, the level of edema for the four hour period after carr. administration was similar whatever the hour of injection. In contrast, with the lower dose (125 micrograms) a circadian rhythm in the intensity of the edema produced was observed, showing a maximum of susceptibility during the light span. Repetitive experiments performed at different periods of the year validated this finding. Comparing mean mesors, analysis of this data showed two distinct levels of inflammation, with the lower level observed in autumn and winter indicating evidence for a circannual variability.  相似文献

The effects of 10(-6) and 10(-9) M of progesterone were documented on isoproterenol-stimulated melatonin release by perifused pineal glands removed from female rats in diestrous at two different times of a 12 : 12 h light/dark cycle, 7 and 19 h after light onset (which corresponds to daytime and nighttime, respectively), to look for the existence of a circadian stage-dependence of the hormone effects. Three weeks before the experiment, the rats were synchronized with a 12 : 12 lighting regimen. Progesterone decreased by approximately 50% the release of melatonin during the light span, but not during the dark span. These results show the direct effects of this ovarian hormone on pineal melatonin release and strongly suggest a time-related effect of progesterone on pineal function.  相似文献

Available cytosolic estradiol binding sites (EBS) were estimated at 6 h intervals over a 24 h period in individual extracts of uteri, pituitaries and hypothalami from intact, ovariectomized or hypophysectomized 28-day old rats, maintained from birth in a 12 h light-12 h dark photoperiod. Plasma estrogen levels were also recorded, in intact rats, they displayed a 20% rise during daytime. In the uterus and pituitary of ovariectomized rats EBS concentrations exceeded by one third those of intact controls. In intact animals, significant diurnal variations in EBS were observed in both these tissues, with increased daytime levels. These variations appeared to depend on the amount of estrogen in the plasma, as they were not observed in spayed rats. No diurnal variations in EBS were apparent in the hypothalamus of intact animals, but they did occur in ovariectomized rats, and even more significantly in hypophysectomized rats, although the latter had lower EBS concentrations than spayed animals. These variations were presumed to result from the interference of pituitary-controlled hormones other than estrogens in the regulation of EBS levels.  相似文献

The effect of an n-3 (fish) and n-6 (soybean) fatty acid-rich diet on carrageenin paw oedema in rats, and the participation of adrenal gland, corticosterone and alpha(2)-macroglobulin (alpha(2)-M) in this process were studied. A significant inhibition of carrageenin oedema was observed not only in rats fed a diet rich in fish oil but also in the soybean group. alpha(2)-M was not detectable before carrageenin injection, suggesting that this putative antiinflammatory factor does not participate in the observed anti-inflammatory effect. Corticosterone levels were higher in fat-fed than in control rats, before carrageenin stimulus and adrenalectomy abolished the anti-inflammatory response in fat-fed animals, showing the important role of the adrenocortical hormones in this process.  相似文献

Abstract

We investigated the dosing-time dependency of acute resveratrol administration on lipoperoxidation level found in the heart, liver and kidney of male rats synchronized with a 12-h dark-light cycle. Resveratrol was administered by the i.p. route at the middle of the dark (6 h after dark onset, HADO) or light span (18 HADO) and thiobarbituric acid reactive species (TBARS) measured 4 h later at 10 and 22 HADO, respectively. Basal TBARS levels in the three organs were higher during the night span when compared to day span. Resveratrol effect on tissues TBARS was also dosing-time dependent. When administered during the dark phase, resveratrol decreased TBARS levels whereas at the light span, the polyphenol increased TBARS in the three organs. Resveratrol behaved as an antioxidant during the dark span and as a pro-oxidant during the light span. These data suggested a day/night rhythm in basal lipoperoxidation and in resveratrol antioxidant effect.  相似文献

Abstract

The daily variations of locomotor activity, plasma and adrenal corticosterone levels and cholesterol‐LDL were studied in male Wistar rats with food ad libitum and feeding restricted to the first 4 hours of the light phase in LD 12:12..

Under LD 12:12 (light on from 9:00 to 21:00h) rats with food ad libitum were eating and moving during the dark period and the locomotor activity clearly showed a biphasic pattern with three harmonic components. Plasma and adrenal corticosterone levels increased during the light period and reached a maximum value just before the dark period whereas the acrophase of cholesterol‐LDL is found at the beginning of the light phase.

The acrophases of activity, plasma and adrenal corticosterone levels in the restricted feeding schedule rats occurred in the first three hours of lighting and the cholesterol‐LDL acrophase at the beginning of the dark phase.

These results confirm a previous report that the shift of feeding to the light phase seems to cause a concomitant phase‐shift in all the variables measured.  相似文献

A high-performance liquid chromatographic method was developed for the simultaneous determination of phenylbutazone and its metabolites, oxyphenbutazone and γ-hydroxyphenylbutazone, in plasma and urine. Samples were acidified with hydrochloric acid and extracted with benzene—cyclohexane (1:1, v/v). The extract was redissolved in methanol and chromatographed on a μBondapak C15 column using a mobile phase of methanol—0.01 M sodium acetate buffer (pH 4.0) in a linear gradient (50 to 100% methanol at 5%/min; flow-rate 2.0 ml/min) in a high-performance liquid chromatograph equipped with an ultra-violet absorbance detector (254 nm). The detection limit for phenylbutazone, oxyphenbutazone and for γ-hydroxyphenylbutazone was 0.05 μg/ml.A precise and sensitive assay for the determination of phenylbutazone and its metabolites was established.  相似文献

To assess to what extent the presence of an ectopic pituitary differentially affected circulating prolactin (PRL) and gonadotropin levels at different times of the year, rats kept under 12h light, 12h dark (12:12 LD) photoperi-ods and receiving a pituitary graft or a sham operation in summer or winter were examined 3 months later. In both male and female sham-operated rats, a circadian variation in serum PRL levels was found, with an acrophase varying from 21:53h to 00:54h and the mesor and amplitude higher in spring than autumn in males and higher in autumn than in spring in females. After grafting a pituitary, changes in serum PRL related to time of day were no longer observed. In pituitary-grafted male rats killed during spring, serum PRL levels were higher than controls at only a few time points throughout the 24h cycle, whereas in rats killed during autumn, there were no significant differences in PRL levels between grafted and control rats. Pituitary-grafted female rats killed during spring showed serum PRL levels significantly higher than those of sham-operated rats, while in female rats killed in autumn, PRL levels of pituitary-grafted and sham-operated rats did not differ. Significant variations of luteinizing hormone (LH) related to time of day were found in sham-operated male rats only, with acrophases at 23:52h and 00:24h for spring and autumn, respectively, and the mesor and amplitude of the rhythm significantly higher in autumn. Pituitary transplants suppressed 24h variations in circulating LH and depressed its levels during the two seasons examined. As far as follicle-stimulating hormone (FSH), pituitary grafts decreased circulating levels, with the extent of decrease higher during autumn than in spring. The results indicate that some endocrine consequences of the grafting of an ectopic pituitary are dependent on time of year.  相似文献

Daily and circadian variations of melatonin contents in the diencephalic region containing the pineal organ, the lateral eyes, and plasma were studied in a urodele amphibian, the Japanese newt (Cynops pyrrhogaster), to investigate the possible roles of melatonin in the circadian system. Melatonin levels in the pineal region and the lateral eyes exhibited daily variations with higher levels during the dark phase than during the light phase under a light-dark cycle of 12 h light and 12 h darkness (LD12:12). These rhythms persisted even under constant darkness but the phase of the rhythm was different from each other. Melatonin levels in the plasma also exhibited significant day-night changes with higher values at mid-dark than at mid-light under LD 12:12. The day-night changes in plasma melatonin levels were abolished in the pinealectomized (Px), ophthalmectomized (Ex), and Px+Ex newts but not in the sham-operated newts. These results indicate that in the Japanese newts, melatonin production in the pineal organ and the lateral eyes were regulated by both environmental light-dark cycles and endogenous circadian clocks, probably located in the pineal organ and the retina, respectively, and that both the pineal organ and the lateral eyes are required to maintain the daily variations of circulating melatonin levels.  相似文献

To study the anti-inflammatory mechanisms of glucocorticoids, we have compared the effects of intratracheal carrageenin (2.5 mg) on control rats and those in which inflammation was subdued by prior dexamethasone treatment (10 mg/l in drinking water). Inflammation was maximal 48 h post-carrageenin. After dexamethasone, carrageenin caused tittle inflammation or oedema (wet lung (mg), n = 6, mean +/- S.E.M.; control, 995 +/- 51; carrageenin + dexamethasone, 1144 +/- 83; compared with carrageenin alone, 1881 +/- 198), but rats had more lung lavage neutrophils than those given carrageenin alone (PMN x 10(6) /lung, mean +/- S.E.M.; control, 0.055 +/- 0.003; carrageenin + dexamethasone, 8.54 +/- 1.52; compared with carrageenin alone, 6.30 +/- 1.71). Proteolysis and partial inactivation of the anti-inflammatory mediator, lipocortin 1 (Lcl), in carrageenin-instilled rats was offset in those also given dexamethasone, by increased Lc1 levels (intact Lc1 ng/ml lavage fluid, n = 4, mean +/- S.E.M.; control 24 +/- 6; carrageenin 15 +/- 4; carrageenin + dexamethasone, 40 +/- 15). Maintenance of sufficient intact (fully active) extracellular Lc1 may contribute to the actions of glucocorticoids.  相似文献

This work analyzes the effect of social isolation of growing male rats on 24-h changes of plasma prolactin, growth hormone, ACTH and leptin, and on plasma and adrenal corticosterone concentrations. At 35 days of life, rats were either individually caged or kept in groups (6-8 animals per cage) under a 12:12 h light/dark schedule (lights on at 08:00 h). A significant arrest of body weight gain regardless of unchanged daily food intake was found in isolated rats after 2 weeks of isolation. On the 4th week, rats were killed at 6 time intervals during a 24-h cycle, beginning at 09:00 h. In isolated rats the 24-h pattern of all parameters tested became distorted, as assessed by Cosinor analysis. When analyzed as a main factor in a factorial analysis of variance, isolation decreased plasma prolactin and growth hormone, increased plasma leptin and corticosterone while decreased adrenal corticosterone. Plasma corticosterone levels correlated significantly with plasma ACTH and with adrenal corticosterone levels in group-caged rats only. These changes can be attributed to an effect of mild stress on the endogenous clock that modulates the circadian hormone release.  相似文献

Membrane‐bound pyroglutamyl‐2‐naphthylamide‐hydrolyzing enzyme activity was analyzed fluorometrically in the anterior hypothalamus, pituitary, and retina of adult male rats to investigate day–night differences. Six groups (n=6 per group) were assessed—three during the light span and three during the dark span—under a standard 12 h–12 h light–dark cycle (light on from 07:00 to 19:00 h) and controlled temperature environment, with food and water available ad libitum. In the hypothalamus, enzyme activity levels were higher for time points of the dark than the light period. In contrast, the pituitary and retina exhibited the highest levels at the time points of the light period. The pituitary and retina also exhibited significant differences between the clock‐hour means of the light period. Day–night differences in membrane‐bound pyroglutamyl‐2‐naphthylamide‐hydrolyzing activity may reflect differences in its susceptible endogenous substrates.  相似文献

Effect of non-steroidal anti-inflammatory drugs on fertility of male rats   总被引:1,自引:0,他引:1  
In an attempt to study the influence of seminal prostaglandin reduction on male fertility, the effect of prolonged treatment with 4 non-steroidal anti-inflammatory drugs (acetylsalicylic acid, indomethacin, naproxen and phenylbutazone) on fertility was determined in male rats. Before the fertility experiments, the pharmacokinetics of the drugs were determined to find dosage regimens by which drug concentrations known as active from human anti-inflammatory therapy could be reached and maintained in the animals. Except for phenylbutazone, all drugs decreased prostaglandin E-2 level in seminal fluid by 80-90%, but only indomethacin reduced fertility significantly. The results suggest that reduction of prostaglandin synthesis in male rats does not affect fertility, which might be related to the very low seminal prostaglandin levels in rats compared to those in animals of other species.  相似文献

Some metabolic aspects of seromucoids in inflammation were elucidated by the use of oxyphenbutazone (OPB), a potent anti-inflammatory drug whose mode of action is fundamentally different from that of corticosteroids. Inflammation was induced in rats by intraperitoneal injection of 3 ml. of 1% formaldehyde. This resulted in an immediate and remarkable increase in the plasma seromucoid level, which persisted for 23 days. The subcutaneous injection of OPB (100 mg./kg.), six hours after formal-dehyde treatment, significantly lowered plasma seromucoids. The decrease was more pronounced when the same dosage of OPB was administered 24 hours before formal-dehyde injection. However, no effects were detected with single or repeated injections of OPB in a dosage of 50 mg./kg. Furthermore, the plasma seromucoid levels of normal rats were uninfluenced by OPB. From these results, it was postulated that some mediating or stimulating factor was released at the site of injury that was responsible for the increased synthesis of seromucoids and that was inhibited by OPB. The lowering effect of OPB was in contrast to the action of ACTH and cortisone, which latter do not alter the increased plasma level of seromucoids. These results further confirm that plasma seromucoid regulation is independent of the pituitary-adrenal mechanism.  相似文献

The effects of colchicine and its analogs on the carrageenin-induced footpad edema in rats were investigated. The anti-inflammatory effects of colchicine analogs were measured at 3 and 5 hr after the carrageenin injection. Colchicine, 1-demethylcolchicine and 3-demethylcolchicine markedly inhibited the carrageenin edema whereas 2-demethylcolchicine was much less active. Thiocolchicinoids, having a thiomethyl group at C-10 instead of a methoxy group, were considerably less potent. These results suggest that the presence of methoxy groups at C-2 and C-10 in colchicine is necessary to maintain anti-inflammatory activity. Inactivity of deacetylcolchicine indicates that substitution of the amino group at C-7 with electron withdrawing groups is also important. Significant inhibition of carrageenin edema and strong binding to tubulin in vitro were manifested by colchicine, 3-demethylcolchicine, N-butyryldeacetylcolchicine and colchifoline. On the other hand, N-carbethoxydeacetylcolchicine which did bind well to tubulin, did not show much effect on the carrageenin edema. These results suggest that the anti-inflammatory action of colchicinoids may not be regulated through the microtubule system.  相似文献

In a series of experiments, male and female Sprague Dawley rats, kept in light (L) from 06(00) to 18(00) alternating with darkness (LD 12:12) inhaled different concentrations of carbon monoxide (50-1,700 ppm) at each of two test times, 12 h apart. A decrease in flow of CO2 (VCO2) resulting from CO inhalation was greater in the active dark (D) than resting light (L) span. Experimental hypoxic mortality of male and female mice also shows circadian variations, being greater in the D than in the L span. Moreover, a difference of mortality was observed betwen hypoxic exposures performed at 12(00) (in LD or DL) and hypoxic exposures performed at 00(00) (in LD or DL). Such results await tests of any extent to which they model responses of human beings to air pollution. In human beings any external environmental circadian, circaseptan and circannual variations in air pollution as such may serve to variable extent as socioeconomic synchronizers of innate rhythms with a corresponding frequency, rather than as solely generators of time patterns in any physiopathologic response to air pollution.  相似文献

This study evaluates the local levels of proinflammatory cytokine, tumor necrosis factor alpha (TNF-alpha), and anti-inflammatory cytokine, interleukin-10 (IL-10), in an experimental buccal abscess of a diabetic rat model. We prepared a buccal cavity induced by injection of carrageenin in a diabetic rat (blood glucose, 460.6 +/- 54.7 mg/dl, mean +/- SE) induced by streptozotocin (STZ). The buccal abscess was formed by the direct inoculation of Streptococcus pyogenes S-8 (2 x 10(7) cfu) into the buccal cavity at day 5 after carrageenin injection. Cytokine levels in the exudate of the buccal abscess were measured by enzyme-linked immunosorbent assay for 48 h after infection. Bacterial counts, weighing of exudate, and histological analysis were also performed. The mean TNF-alpha levels in the buccal abscess exudate of the diabetic group, which were generally higher than those of the control group, tended to increase over time until 48 h after infection, while the TNF-alpha levels in the control group peaked at 24 h after infection and then decreased. The IL-10 levels in the diabetic group remained almost unchanged until 48 h after infection, while the IL-10 levels in the control group were significantly higher than in the diabetic group at 12-24 h after infection. The mean ratio of TNF-alpha to IL-10 levels was 1.17-1.67 in the diabetic group, which was higher than the 0.26-0.69 of the control group. The bacterial counts in the buccal abscess and the weight of exudate were significantly higher in the diabetic group compared to the control group at 36-48 h. Histological findings showed that inflammatory cell infiltration was remarkable in the diabetic group compared to that of the control group. These results suggest that the elevated proinflammatory TNF-alpha levels and decreased anti-inflammatory IL-10 levels, which are produced at local infection sites, may at least in part be related to the severity of inflammation in this rat model with diabetes induced by STZ.  相似文献
