The proper development of fruits is important for the sexual reproduction and propagation of many plant species. The fruit of Arabidopsis derives from the fertilized gynoecium, which initiates at the center of the flower and obtains its final shape, size, and functional tissues through progressive stages of development. Hormones, specially auxins, play important roles in gynoecium and fruit patterning. Cytokinins, which act as counterparts to auxins in other plant tissues, have been studied more in the context of ovule formation and parthenocarpy. We recently studied the role of cytokinins in gynoecium and fruit patterning and found that they have more than one role during gynoecium and fruit patterning. We also compared the cytokinin response localization to the auxin response localization in these organs, and studied the effects of spraying cytokinins in young flowers of an auxin response line. In this addendum, we discuss further the implications of the observed results in the knowledge about the relationship between cytokinins and auxins at the gynoecium.

The reproductive development of the Christmas rose (Helleborus niger L.) is characterized by an uncommon feature in the world of flowering plants: after fertilization the white perianth becomes green and photosynthetically active and persists during fruit development. In the flowers in which fertilization was prevented by emasculation (unfertilized) or entire reproductive organs were removed (depistillated), the elongation of the peduncle was reduced by 20?C30%, and vascular development, particularly lignin deposition in sclerenchyma, was arrested. Chlorophyll accumulation in sepals and their photosynthetic efficacy were up to 80% lower in comparison to fertilized flowers. Endogenous auxins were investigated in floral and fruit tissues and their potential roles in these processes are discussed. Analytical data of free indole-3-acetic acid, indole-3-ethanol (IEt), and seven amino acid conjugates were afforded by LC-MS/MS in floral tissues of fertilized as well as unfertilized and depistillated flowers. Among amino acid conjugates, novel ones with Val, Gly, and Phe were identified and quantified in the anthers, and in the fruit during development. Reproductive organs before fertilization followed by developing fruit at post-anthesis were the main source of auxin. Tissues of unfertilized and depistillated flowers accumulated significantly lower levels of auxin. Upon depistillation, auxin content in the peduncle and sepal was decreased to 4 and 45%, respectively, in comparison to fruit-bearing flowers. This study suggests that auxin arising in developing fruit may participate, in part, in the coordination of the Christmas rose peduncle elongation and its vascular development.

An intriguing phenomenon in plant development is the timing and positioning of lateral organ initiation, which is a fundamental aspect of plant architecture. Although important progress has been made in elucidating the role of auxin transport in the vegetative shoot to explain the phyllotaxis of leaf formation in a spiral fashion, a model study of the role of auxin transport in whorled organ patterning in the expanding floral meristem is not available yet. We present an initial simulation approach to study the mechanisms that are expected to play an important role. Starting point is a confocal imaging study of Arabidopsis floral meristems at consecutive time points during flower development. These images reveal auxin accumulation patterns at the positions of the organs, which strongly suggests that the role of auxin in the floral meristem is similar to the role it plays in the shoot apical meristem. This is the basis for a simulation study of auxin transport through a growing floral meristem, which may answer the question whether auxin transport can in itself be responsible for the typical whorled floral pattern. We combined a cellular growth model for the meristem with a polar auxin transport model. The model predicts that sepals are initiated by auxin maxima arising early during meristem outgrowth. These form a pre-pattern relative to which a series of smaller auxin maxima are positioned, which partially overlap with the anlagen of petals, stamens, and carpels. We adjusted the model parameters corresponding to properties of floral mutants and found that the model predictions agree with the observed mutant patterns. The predicted timing of the primordia outgrowth and the timing and positioning of the sepal primordia show remarkable similarities with a developing flower in nature.

Floral size is an ecologically important trait related to pollination success and genetic fitness. Independently of the sexual reproduction strategy, in many plants, floral size seems to be controlled by several genetic programs that are to some extent independent of vegetative growth. Flower size seems to be governed by at least two independent mechanisms, one controlling floral architecture that affects organ number and a second one controlling floral organ size. Different organ-dependent growth control may account for the final proportions of a flower as a whole. Genes controlling floral organ identity, floral symmetry and organ polarity as well as auxin and gibberellin response, also play a role in establishing the final size and architecture of the flower. The final size of an organ seems to be controlled by a systemic signal that might in some cases overcome transgenic modifications of cell division and expansion. Nevertheless, modification of basic processes like cell wall deposition might produce important changes in the floral organs. The coordination of the direction of cell division and expansion by unknown mechanisms poses a challenge for future research.

A mathematical model simulating spatial pattern formation (positioning) of floral organs is proposed. Computer experiment with this model demonstrated the following sequence of spatial pattern formation in a typical cruciferous flower: medial sepals, carpels, lateral sepals, long stamens, petals, and short stamens. The positioning was acropetal for the perianth organs and basipetal for the stamens and carpels. Organ type specification and positioning proceed non-simultaneously in different floral parts and organ type specification goes ahead of organ spatial pattern formation. Computer simulation of flower development in several mutants demonstrated that the AG and AP2 genes determine both organ type specification and formation of the zones for future organ development. The function of the AG gene is to determine the basipetal patterning zones for the development of the reproductive organs, while the AP2 gene maintains proliferative activity of the meristem establishing the acropetal patterning zone for the development of the perianth organs.

Hypothetically, a species with both cleistogamous (CL) flowers and delayed selfing chasmogamous (CH) flowers should display high levels of reproductive assurance because, over time, obligate selfing by CL flowers should reduce inbreeding depression and delayed selfing in CH flowers should compensate for the absence of outcross pollen. We used pollinator-exclusion experiments to investigate reproductive assurance in the CH flowers of Polygala lewtonii, an herb with a mixed mating system. We followed CH flowers from bud-break to flower/fruit abscission to quantify fruit initiation and maturation and rates of floral development. We also evaluated the efficacy of the selfing mechanism, conducted pollinator watches to assess the likelihood of pollinator limitation, and performed regression analysis to determine the effect of flower position on fruit production. Pollinator exclusion significantly reduced fruit initiation and maturation. Investigation of floral development demonstrated that the selfing mechanism is largely dysfunctional in CH flowers, indicating the failure of reproductive assurance. Low observed rates of insect visitation appear to contradict high rates of CH fruit production in open-pollinated plants, particularly given the rarity of delayed selfing. In both treatments, flower position significantly affected fruit initiation, suggesting a role for resource limitation in both pollinator-excluded and open-pollinated flowers.

In the zucchini squash, Cucurbita pepo, a well coordinated abscission of the female flower during fruit set is essential to obtain a fruit of commercial value. In Spain zucchini is mainly produced in greenhouses in Almería, where high temperatures during the spring-summer period provoke a cultivar-dependent defect in fruits known as the “sticky flower” syndrome. This disorder is characterised by an arrest in growth and maturation of floral organs, and a lack of female floral abscission, thus diminishing fruit shelf-life, commercial quality and value. The aim of the present work was to improve knowledge of the abscission process in C. pepo to better understand the fundamental causes of this disorder. The anatomical analysis of abscission shows a well defined male floral abscission zone (AZ), few hours after anthesis, which differs from the female zone which is not differentiated from the adjacent tissue until the abscission process has begun, and which occurs as a consequence of AZ cell enlargement and the dissolution of their cell walls. To evaluate the role of ethylene and auxins in the regulation of floral abscission in zucchini we performed several treatments, with: ethylene, added as 0.25% ethrel solution; AVG, the inhibitor of ethylene synthesis, at 100 μM; indol-3-acetic acid, 100 μM; and TIBA, the inhibitor of auxin polar transport, at 10 mM. These treatments show that ethylene is an accelerator of zucchini floral abscission, and also promotes abscission in isolated AZs of sticky flowers. On the other hand, IAA delays abscission of the female flowers, whilst the inhibitor of auxin polar transport promotes it. The activity of the cell wall hydrolytic enzymes, polygalacturonase and cellulase, sharply increased just before the shedding of zucchini floral organs (72 h after anthesis). Moreover, both enzyme activities were induced by ethylene, which partly explains the ethylene promoting effect.

Undoubted lines of evidence point out that members of CYCLOIDEA (CYC) 2 clade are essential players to control flower symmetry and, amusingly, also are determinants of capitula architecture (pseudanthium). In several species, CYC-like genes influence the androecium patterning, but to date, the function of these genes in the development of gynoecium organs is less clear. In this review, we first reported details about floral symmetry and an overview of genes and molecular mechanisms regulating the development of zygomorphism in different angiosperm lineages (e.g., basal and core eudicots and monocots). Then, we paid emphasis on the role of CYC-like genes in the development of heterogamous inflorescence of sunflower as well as other Asteraceae and some species within the Dipsacaceae family. Helianthus annuus is particularly attractive because it represents a useful model to study the role of CYC-like genes on shaping floral corolla as well as the differentiation of reproductive organs in different flowers of pseudanthia. A special attention was reserved to inflorescence morphology mutants of sunflower (i.e., Chrysanthemoids2 and tubular ray flower) because they provide useful information on the role of CYC-like genes in the radiate capitulum evolution. Finally, we discuss data from literature to suggest that CYC-like genes are also co-opted to regulate stamen and carpel differentiation likely throughout their interaction with the cell cycle and flower organ identity genes. The recruitment of reproductive organs in ray flowers also supports the phylogenetic origin of a radiate inflorescence of sunflower from a discoid capitulum and suggests that in sterile zygomorphic ray flower primordia the latent identity to differentiate both microsporangium and macrosporangium was conserved.

A Role for Auxin in Flower Development
Auxin has long been implicated in many aspects of plant growth and development including flower development. However, the exact roles of auxin in flower development have not been well defined until the recent identification of auxin biosynthesis mutants. Auxin is necessary for the inltiation of floral primordia, and the disruption of auxin biosynthesis, polar auxin transport or auxin signaling leads to the failure of flower formation. Auxin also plays an essential role in specifying the number and Identity of floral organs. Further analysis of the relationship between the auxin pathways and the known flower development genes will provide critical information regarding mechanisms of organogenesis and pattern formation in plants.
