The rapid erosion of the genetic diversity of both wild and cultivated plants has recently attracted more and more international concern. As a consequence, germplasm conservation techniques have become accessible to protect the third world's germplasm diversity. Several approaches are used for germplasm conservation and utilization. The evaluation and use of these techniques has to be focussed with one important objective; to preserve with as much integrity as possible, the genetic variability of the selected species.Questions have risen about the best way to preserve genetic diversity of the humid tropics, since it is being recognized that these conditions hold much of the world's gene pool of plants. Seed banks are considered the best system for orthodox seed storage. For many tropical species that cannot support dehydration, tissue culture and cryoconservation are the best alternative. Important considerations regarding the adaptation of modern techniques to tropical species have to be carefully analyzed. Quantitative studies of genetic indices of somaclonal variation, or types or mutant plants can be observed.Consideration of plant germplasm as a base for genetic improvement has come a long way, but much remains to be done, particularly, with the plant diversity that exists in the tropics. All efforts to conserve and use genetic resources will contribute to the benefit of future human generations. 相似文献

Conservation of plant genetic resources attracts more and more public interest as the only way to guarantee adequate food supplies for future human generations. However, the conservation and subsequent use of such resources are complicated by cultural, economical, technical and political issues. Over the last 30 years, there have been significant increases in the number of plant collections and in accessions in ex situ storage centres throughout the World. The present review is of these ex situ collections and the contribution biotechnology has made and can make to conservation of plant germplasm. The applications and limitations of the new, molecular approaches to germplasm characterization are discussed.
In vitro slow growth is used routinely for conserving germplasm of plants such as banana, plantain, cassava and potato. More recently, cryopreservation procedures have become more accessible for long-term storage. New cryopreservation techniques, such as encapsulation-dehydration, vitrification and desiccation, lengthen the list of plant species that can not only tolerate low temperatures but also give normal growth on recovery. Extensive research is still needed if these techniques are to be fully exploited.V.M. A comparative assessment of molecular marker assays (AFLP, RAPD and SSR) for white yam (Dioscorea rotundata) germplasm characterization 总被引:1,自引:0,他引:1
Several DNA‐based marker systems are available for genetic fingerprinting of plants but information on their relative usefulness for yam germplasm characterisation is lacking. The efficiency of RAPD, AFLP and SSR markers for the assessment of genetic relationships, and for cultivar identification and discrimination among 45 West and Central African white yam cultivars belonging to 22 morphotypes/cultivar groups was investigated. Dendrograms were produced based on band pattern scores using the UPGMA method. Results showed that each of the three techniques could unequivocably identify each cultivar, but that techniques differed in the mean number of profiles generated per primer (or primer pair) per cultivar, referred to as genotype index (GI). The order of merit based on this criterion in this study was AFLPs (GI = 2.56), SSRs (GI = 0.39) and RAPDs (GI = 0.35). Yam genotypes classified in the same cultivar group based on morphology were often genetically different, emphasising the need for molecular fingerprinting in yam germplasm characterisation. AFLPs showed the highest efficiency in detecting polymorphism and revealed genetic relationships that most closely reflected morphological classification. 相似文献

The need for conservation of biotic diversity is well recognized. However, improved techniques for the efficient, cost effective-preservation of plant germplasm are needed. The conservation and distribution of plant germplasm in vitro is gaining acceptance. However, increased usage is dependent upon the ability of curators to minimize culture maintenance requirements. This report examines the effect of various levels of sucrose, photoperiod, temperature, sorbitol and mannitol on minimal growth storage of Ipomoea batatas (L.) Lam. Growth was reduced 50% with a temperature reduction of from 21.1 to 15.6°C. Sucrose concentrations of 15 and 20 g l-1 resulted in reduced plant stature with few adverse effects on plantlet viability or morphology. Reduction of photoperiod from 16 to 4 h produced smaller, slightly chlorotic, but otherwise normal plants. The addition of sorbitol or mannitol to culture media generally produced undesirable effects on gross plant morphology and loss of apical dominance. Arachis retusa Krapov. et W. C. Gregory et Valls is endemic in the West-central region of Brazil, occurring In areas endangered by human actions. The establishment of in vitro preservation methods for wild species of Arachis is an alternative to seed banks for germplasm storage, multiplication and distribution. The risk of genetic changes Induced by tissue culture and the monitoring of the genetic stability of the biological material before, during and after storage must be considered In the context of conservation. Random amplified polymorphlc ONA (RAPO) and amplified fragment length polymorphism (AFLP) fingerprinting were used to evaluate the genetic stability of in vitro plants originated from cotyledons and embryo axes of A. retusa. Cotyledons originated shoots through direct organogenesls and embryo axes displayed muItishoot formation Induced by 110 mmol/L and 8.8 mmol/L BAP, respectively. Ninety genomlc regions (loci) generated from RAPO and 372 from AFLP analyses were evaluated. All amplified fragments detected by both techniques in plants derived from the two explant types were monomorphic. 应用SCoT和ISSR标记对36份龙眼资源和1份近缘种龙荔的遗传多样性进行分析。结果表明:12对SCoT引物共扩增出127条带,平均每条引物扩增10.58条带;15条ISSR引物共扩增出117个条带,平均扩增7.8条带。UPGMA聚类结果表明:SCoT标记和ISSR标记分别在相似系数0.672和0.685水平上,均可将37份材料分成6大类群,SCoT和ISSR标记均适用于龙眼材料的遗传多样性分析,如果将两种标记的数据进行综合分析,可以缩小单一标记的误差。研究结果为龙眼种质资源的保存和利用提供了重要的依据。 相似文献

Summary Cryopreservation (liquid nitrogen, −196°C) represents the only safe and cost-effective option for long-term conservation of
germplasm of non-orthodox seed species, vegetatively propagated species, and of biotechnology products. Classical cryopreservation
techniques, which are based on freeze-induced dehydration, are mainly employed for freezing undifferentiated cultures and
apices of cold-tolerant species. New cryopreservation techniques, which are based on vitrification of internal solutes, are
successfully employed with all explant types, including cells suspensions and calluses, apices, and somatic and zygotic embryos
of temperate and tropical species. The development of cryopreservation protocols is significantly more advanced for vegetatively
propagated species than for recalcitrant seed species. Even though its routine use is still limited, there are a growing number
of examples where cryopreservation is employed on a large scale for different types of materials, including seeds with orthodox
and intermediate storage behaviour, dormant buds, pollen, biotechnology products, and apices sampled from in vitro plantlets of vegetatively propagated species. Cryopreservation can also be employed for uses other than germplasm conservation,
such as cryoselection, i.e., the selection through freezing of samples with special properties, or cryotherapy, i.e., the
elimination of viruses from infected plants through apex cryopreservation. Because of its high potential, it is expected that
cryopreservation will become more frequently employed for long-term conservation of plant genetic resources. 相似文献

以香菇L808菌株基因组序列为参照,开发了300份插入/缺失(InDel)标记。通过5个菌株的初筛,选出均匀分布于基因组的82份多态标记对42份香菇种质资源进行遗传背景分析。同时在设施化栽培条件下考察了种质资源的生育期、菌棒转色、菌棒硬度、现蕾期和产量等表型。结果表明:供试香菇种质资源的遗传多样性丰富,栽培菌株与野生菌株的遗传距离较远,遗传相似性系数平均值为0.51。群体结构分析将种质资源分为6个亚群,与基于遗传距离的系统聚类结果较为一致。亚群间菌株具有较远的亲缘关系,遗传分化指数平均值为0.290,适合用于杂交育种。表型结果显示,设施化栽培条件下的种质资源农艺性状分化程度高,亚群间菌株的生育期、现蕾期和产量均有显著差异。种质资源多样性分析结果为香菇杂交育种工作奠定了基础。 相似文献

The seaweed industry worldwide uses 7.5–8.0 million tonnes of wet seaweeds annually with a majority of it derived from cultivated
farms, as the demand for seaweed based-products exceeds the supply of seaweed raw material from natural stocks. The main advantage
of cultivation is that it not only obviates overexploitation of natural populations but also facilitates the selection of
germplasm with desired traits. To enhance the economic prospects of seaweed cultivation, varied practices, such as simple
and cost effective cultivation methods, use of select germplasm as seed stock coupled with good farm management practices,
etc., are adopted. Nevertheless, in vitro cell culture techniques have also been employed as they facilitate development and
propagation of genotypes of commercial importance. There are more than 85 species of seaweeds for which tissue culture aspects
have been reported. Although the initial aim of these techniques focuses mostly on genetic improvement and clonal propagation
of seaweeds for mariculture, recently the scope of these techniques has been extended for use in bioprocess technology for
production of high value chemicals of immense importance in the pharmaceutical and nutraceutical sectors. Recently, there
has been a phenomenal interest in intensifying seaweed tissue and cell culture research to maximize the add-on value of seaweed
resources. This paper deals with the status of seaweed micropropagation techniques and their applications in the context of
the marine biotech industry. Further, it also provides an analysis of the problems to be resolved for removing the barriers
that are impeding the true realization of potentials offered by these techniques for sustainable development and utilization

In vitro culture techniques are usually employed for ex situ conservation of endangered plant species. However, encapsulation
to preserve threatened bryophytes is scarcely used, and only as a pretreatment prior to cryopreservation. In our study, two
different methods of germplasm conservation, involving calcium-alginate encapsulation of moss material, were assessed. The
plant material used was gametophyte buds (gametophores) of Splachnum ampullaceum Hedw., a rare species of moss. Moss regeneration was evaluated at different periods of time to examine the efficacy of the
technique for moss germplasm conservation. The effects of encapsulation and cold storage on developmental parameters such
as protonematal colony diameter, bud length, and number of buds were also studied. The results suggest that moss encapsulation
with no prior treatment may be a suitable method for germplasm conservation during long periods of time. With our method survival
rates as high as 50% could be reached after 2.5 years of cold storage versus 0% of 24-h cryopreserved beads. This technique
together with cold storage, avoiding freezing, may be especially important in desiccation intolerant mosses. 相似文献

【目的】燕麦种质资源遗传多样性研究不仅有助于种质资源的收集和评价,而且对燕麦生产和育种具有重要指导意义。【方法】研究对260份燕麦种质资源的20个形态学性状多样性、变异及聚类进行分析,评价其形态学性状的遗传变异水平,明确燕麦种质资源的性状特点与遗传多样性,以期为燕麦种质创新和品种改良提供依据。【结果】260份燕麦种质资源形态学性状间存在广泛的遗传多样性,质量性状的遗传多样性指数以粒色最大(1.53),芒色最小(0.76);12个数量性状呈正态分布,数量性状的遗传多样性指数以千粒重最大(2.03),有效分蘖数最小(1.22),变异系数最大的是有效分蘖数(89.02%),最小的是株高(11.19%)。根据燕麦品种(系)间各性状的遗传差异,聚类分析将供试的260份燕麦种质资源分为6类,其中种质群Ⅰ包括42份材料,可作为种用型育种目标的亲本材料;种质群Ⅱ包括31份材料,可作为选育高产饲草品种的亲本材料;种质群Ⅳ包括41份材料,可作为选育大粒专用型品种的育种材料;种质群Ⅴ包括46份材料,可作为燕麦矮化的亲本材料;而种质群Ⅲ包括46份材料,种质群Ⅵ包括54份材料,这两类种质群材料的综合性状表现不突... 相似文献

Actually, the germplasm of Jatropha spp. is conserved as whole plants in field collections. Under this storage method, the genetic resources are exposed to disease, pest and natural hazards such as human error, drought and weather damage. Besides, field genebanks are costly to maintain and with important requirements of trained personnel. Thus, the development of efficient techniques to ensure its safe conservation and regeneration is therefore of paramount importance. In this work we describe a method for Jatropha curcas seeds cryoexposure and seedling recovery after thawed. In a first experiment, an efficient protocol for in vitro plant recovery was carried out using zygotic embryo or seeds with or without coat. In a second experiment, desiccated seeds with or without coat were exposed to liquid nitrogen and evaluated after cryoexposure. Germination percentages were variable among treatments, and seeds demonstrated tolerance to liquid nitrogen exposure under certain conditions. Seeds of J. curcas presented up to 99.6% germination after seed coat removal. Seeds with coat cultured in vitro did not germinate, and were 60% contaminated. The germination of the zygotic embryos was significantly higher in the 1/2 MS medium (93.1%) than in WPM medium (76.2%), but from zygotic embryo, abnormal seedlings reached up to 99%. Seeds with coat exposed to liquid nitrogen showed 60% germination in culture after coat removal with good plant growth, and seeds cryopreserved without coat presented 82% germination, but seedlings showed a reduced vigor and a significant increase in abnormal plants. Seeds cultured in vitro with coat did not germinate, independently of cryoexposure or not. This study reports the first successful in vitro seedling recovery methodology for Jatropha curcas seeds, after a cryopreservation treatment, and is recommended as an efficient procedure for in vitro plant recovery, when seeds are conserved in germplasm banks by low or cryotemperatures. 相似文献

Rice genetic resources, comprising landrace varieties, modern and obsolete varieties, genetic stocks, breeding lines, and the wild rices, are the basis of world food security. The International Rice Genebank at the International Rice Research Institute in the Philippines conserves the largest and most diverse collection of rice germplasm. The facilities of the genebank ensure the long-term preservation of this important diversity. In field research, factors that affect long-term viability of rice seeds have been identified, leading to the introduction of modified practices for germplasm multiplication and regeneration. The value of conserved germplasm can be assessed in terms of useful traits for rice breeding and the economic impact that germplasm utilization has on rice production and productivity. The application of molecular markers is changing perspectives on germplasm management. International policies affecting access to and use of rice germplasm are discussed. 相似文献
