The effects of liming and inoculation with the arbuscular mycorrhizal fungus, Glomus intraradices Schenck and Smith on the uptake of phosphate (P) by maize (Zea mays L.) and soybean (Glycine max [L.] Merr.) and on depletion of inorganic phosphate fractions in rhizosphere soil (Al-P, Fe-P, and Ca-P) were studied in flat plastic containers using two acid soils, an Oxisol and an Ultisol, from Indonesia. The bulk soil pH was adjusted in both soils to 4.7, 5.6, and 6.4 by liming with different amounts of CaCO3.In both soils, liming increased shoot dry weight, total root length, and mycorrhizal colonization of roots in the two plant species. Mycorrhizal inoculation significantly increased root dry weight in some cases, but much more markedly increased shoot dry weight and P concentration in shoot and roots, and also the calculated P uptake per unit root length. In the rhizosphere soil of mycorrhizal and non-mycorrhizal plants, the depletion of Al-P, Fe-P, and Ca-P depended in some cases on the soil pH. At all pH levels, the extent of P depletion in the rhizosphere soil was greater in mycorrhizal than in non-mycorrhizal plants. Despite these quantitative differences in exploitation of soil P, mycorrhizal roots used the same inorganic P sources as non-mycorrhizal roots. These results do not suggest that mycorrhizal roots have specific properties for P solubilization. Rather, the efficient P uptake from soil solution by the roots determines the effectiveness of the use of the different soil P sources. The results indicate also that both liming and mycorrhizal colonization are important for enhancing P uptake and plant growth in tropical acid soils.  相似文献

Mycorrhizoplane-associated actinomycetes were isolated using an enrichment technique from red pine (Pinus resinosa Ait.) roots of seedlings recently outplanted onto cleared northern hardwood sites in the Upper Peninsula of Michigan, USA. Interactions were assessedin vitro between actinomycete isolates and three commonly occurring ectomycorrhizal fungi (Laccaria bicolor (Maire) Orton,L. laccata (Scop.: Fr.) Berk. and Br., andThelephora terrestris Fr.). Most actinomycete isolates exerted a range of effects on the growth of the three fungus isolates during the four week test period, inhibiting some while stimulating others; several inhibited growth of all three fungus isolates. Mycorrhizoplane-associated actinomycetes show potential for use as coinoculants with selected ectomycorrhizal fungi to optimize the soil microflora for developing seedlings.  相似文献

Transformed hairy roots were induced at the excised site ofthe epicotyl of dry mature seed of a Spanish type peanut(Arachishypogaea)cv. Java 13 2 weeks after inoculation with a wild typestrain ofAgrobacterium rhizogenes,MAFF-02-10266. Composite plantsconsisting of transformed roots with non-transformed shootswere cultured using pouches. Forty days after inoculation, thecomposite plant showed a root system with abundant root mass,more lateral branching and high fractal dimension compared tothe control. No differences were observed in production of rosette-typeroot hairs or the cross sectional structure between transformedand non-transformed roots. The inoculation ofBradyrhizobiumsp.A2R1 strain to the composite plants led to the induction oftransformed root nodules. Seeds of Sesbania punicea (Cav.) Benth. are largely impermeable,and the natural, low, percentage which germinate are permeableat the lens. Scarifying seed in acid stimulates germinationand causes various types of damage to the testa, including thelens. Placing seeds in boiling water stimulates germinationby rendering the seeds permeable at the lens. Seeds were mechanicallyscarified at precise sites on the testa to assess influenceof the site and degree of damage on germination. Results indicatethat both the site of initial water entry and the rate of waterentry are important in determining seed and seedling vigour.Damaging the lens is more effective in stimulating germinationand reducing seedling abnormalities than complete excision ofthe lens. This effect can be reversed if seeds from which thelens was excised are allowed to imbibe gradually in inert osmotica.The leguminous lens thus acts as the site of initial water entryand regulates the rate of water uptake, thereby increasing seedand seedling vigour. The sympton of dwarfing in the sea campion, Silene vulgaris(Moench) Garcke sub sp. maritima(With.) A. & D. Löveinfected with the anther smut fungus Ustilago violacea (Pers.)Fuckel, a systemic, perennial parasite, has been investigated. Extracts of both healthy and diseased plants contain IAA andGA3, but diseased plants contain less gibberellins than healthyplants. Neither IAA nor gibberellins were detected in significantquantities in the medium when U. violacea was grown in pureculture, but IAN was present. IAN was also found m extractsof diseased plants and it is tentatively suggested that it isformed by the fungus and may accumulate in the host owing tothe inability of the plant to convert IAN to IAA The exogenousapplication of IAN to healthy plants does not produce any diseasesymptoms. The dwarfing symptom of the diseased plant may be due to thelower levels of gibberellins which it contains compared withhealthy plants since the exogenous application of GA3, restoresthe diseased plant to normal growth  相似文献

Symbiotic interactions of the tripartite association of soybeanplant, vesiculararbuscular (VA) mycorrhizal fungus and Rhizobiumjaponicum were shown. Mycorrhizal plants absorbed more P, Ca and Mg and had higherP, Ca and Mg contents in their stems or leaves than non-mycorrhizalplants. Phosphorus concentration was also higher in the nodulesof mycorrhizal plants. VA mycorrhizae increased nodule number, nodule weight and acetylenereduction activity of nodules. Concomitantly seed productionand N content of leaves were enhanced. Both nodulating (A62-1) and non-nodulating (A62-2) cultivarsof soybean plants [Glycine max (L.) The three-horned stock, Matthiola tricuspidata (L.) R. Br. isa widespread annual plant of the Mediterranean sandy shores.Its seeds are dark germinating and negatively photosensitive,in accordance with our previous findings for a number of othermaritime plants. Full germination was obtained at a wide rangeof temperatures (5-25 °C) in the dark. Inhibition of germinationunder light of various spectral qualities could be generallycorrelated, negatively and positively, respectively, with phytochromephotostationary state (ø) and relative cycling rate ofphytochrome (H). The inhibition of germination by white (fluorescent),blue and far-red light, applied either continuously or intermittently,consistently showed a linear dependence upon the logarithm ofthe flux density of the irradiation. The resulting photoinhibitioncurves had parallel slopes and, compared to those of other maritimeplants, they were shifted to higher flux densities, Continuousblue or far-red irradiations, both establishing a similar øvalue (0·26), resulted in statistically similar regressioncurves, thus favouring the hypothesis that phytochrome is thesingle photoreceptor in the photoinhibition of seed germination.Copyright1994, 1999 Academic Press Matthiola tricuspidata (L.) Recent improvements in the management of oil sand tailings used by the Canadian oil sand industry have resulted in the production of composite tailing sands (CT): a new challenging material for reclamation work. Jack pine (Pinus banksiana Lamb.), hybrid poplar (Populus deltoides Bartr. ex Marsh. ×Populus nigra L.) and red clover (Trifolium pratense L.) plants were used in an 8-week greenhouse bioassay to evaluate the mycorrhizal inoculum potential of CT. This inoculum potential was compared with that of three other reclamation materials [common tailing sands (TS), deep overburden (OB) and muskeg peat (MK)], and with three sites reclaimed in 1982 (R82), 1988 (R88) and 1999 (R99). CT was devoid of active mycorrhizal propagules while all other materials showed some level of inoculum potential. Arbuscular mycorrhizal fungi were observed on roots of clover or poplar grown in TS, OB, and all substrates containing peat (MK, R82, R88 and R99). Pine roots were also colonized by vesicle-forming hyphae of an unidentified fine endophyte and by dark septate fungi. Ectomycorrhizas (ECM) were observed on pine and poplar grown in OB, MK, and in soils from the two older reclaimed sites (R82 and R88). Using morpho- and molecular typing, six ECM fungi were identified to the genus or species level: Laccaria sp., Thelephora americana, Wilcoxina sp. (E-strain), Tuber sp. (I-type), a Sebacinoid, and a Pezizales species. Laccaria sp. and Wilcoxina sp. were the most frequently observed ECM species.  相似文献

In Mediterranean ecosystems vegetation overlying shallow, transient aquifers is often dominated by woody phreatophytes, trees and shrubs that have been shown to be dependent on groundwater for their water requirements. Natural and anthropogenic alterations of groundwater tables (abstraction) are of clear importance to phreatophytic vegetation as reduction of water tables may sever these plants from their natural water sources. Seasonal water sources were determined for species growing on a coastal dune system that overlies a shallow sandy aquifer in south-western Australia. The plants studied grew over groundwater that ranged in depth from 2.5 to 30 m. The naturally occurring stable isotope of hydrogen (deuterium, '2H) was used to distinguish potential water sources. Isotopic ratios from vascular water of the dominant species of the study area (Banksia ilicifolia R. Br. and Banksia attenuata R. Br. trees) were compared with those of potential sources of precipitation, soil moisture and groundwater. A relatively shallow-rooted perennial shrub, Hibbertia hypericoides Benth., was also included as an isotopic reference. The results suggest that both B. attenuata and B. ilicifolia are phreatophytic as they derived some of their water from groundwater throughout the dry-wet cycle, with the exception of B. attenuata at the site of greatest depth to groundwater (30 m) which did not use groundwater. A high proportion (>50%) of groundwater use was not maintained throughout all seasons. With the onset of the hot Mediterranean summer, progressive drying of the surface soils resulted in increased use of groundwater and deep soil moisture. During the wet winter plants used proportionately more water from the upper layers of the soil profile. The degree to which groundwater was utilised by the study species was dependent on the proximity of groundwater, availability of moisture in shallower horizons of the soil profile, root system distribution and maximum root depth.  相似文献

Negative linear relations were detected (P < 0·005)between the rate of progress from sowing to panicle initiationand CO2 concentration (210-720 µmol CO2 mol-1 air) fortwo genotypes of sorghum [Sorghum bicolor (L.) Moench]. Relationsbetween CO2 concentration and the rate of progress from sowingto first flowering were also negative in soyabean [Glycine max(L.) Merrill] (P < 0·025), but positive in cowpea[Vigna unguiculata (L.) Walp.] (P < 0·025), albeitthat in both grain legumes sensitivity was much less than insorghum. Thus CO2 elevation does not delay flowering in allshort-day species. The considerable effect of CO2 concentrationon times to panicle initiation resulted in large differencesamong the sorghum plants at this developmental stage; with increasein CO2 concentration, plants were taller with slightly moreleaves and more pronounced apical extension. At the same timeafter sowing however, sorghum plants were heavier (P < 0·05)at 210 than at 360 µmol CO2 mol-1 air. In contrast, relationsbetween the dry masses of the soyabean and cowpea plants andCO2 concentration were positive and curvilinear (P < 0·05).It is suggested that the impact of global environmental changecould be severe for sorghum production in the semi-arid tropics.Copyright1995, 1999 Academic Press Sorghum bicolor (L.) The survival of seeds of lemon (Citrus limon L.), lime [C. arantifolia(Christm.) Swing.] and sour orange (C. aurantium L.) was examinedunder a wide range of constant moisture contents and temperatures.Seed longevity was increased by decreasing the moisture contentand temperature of the storage environment. Maximum viabilitywas maintained in the combination of storage conditions includingthe lowest moisture content (5 per cent) and lowest temperature(–20 °C) investigated. The practicality of dry storageof citrus seed for genetic conservation is discussed. Seed swelling, germination, root extension, lateral root initiationand shoot growth were studied in soils of different water contents,using non-destructive, serial neutron radiography. Seeds fromthree varieties of soya beans (Glycine max L.) and one varietyeach of maize (Zea mays L.) and vetch (Vicia sativa L.) wereused. The seeds germinated when they had increased in size bya certain amount, if germination is taken as the time when theradicle first appears.  The isolation of vesicular-arbuscular mycorrhizal (VAM) fungi from natural (savanna, rocky hill, wetland and rainforest) and disturbed (minesite) habitats in a seasonally-dry tropical region in the Northern Territory of Australia into open-pot cultures was undertaken to supplement knowledge about the diversity of these fungi. This experiment considered factors affecting the diversity of fungi obtained in trap cultures started using diluted soil from field sites and two host plants. A range of soil phosphorus and other nutrient levels from severely deficient to sufficient for maximal growth was used to determine the impact of nutrition on mycorrhizal associations of sorghum (Sorghum sp.) and clover (Trifolium subterraneum). Soil cores taken from pots at 6-week intervals provided roots and soil to assess mycorrhiza formation and sporulation without substantial damage to plants. The identification of VAM fungi to genus by observing morphological patterns within clover roots revealed substantial differences in fungus populations between soils and a moderate effect of nutrient levels on fungal diversity. Changes in the proportion of different fungi in roots over the 31 weeks of the experiment were also observed. Glomus spp. were initially the most abundant fungi within roots, but Scutellospora spp. gradually became more dominant at later harvests, while colonisation by Acaulospora spp. was limited at all times. For both clover and sorghum, sporulation was limited and was dominated by single species of Scutellospora and Acaulospora. This contrasted with the much higher diversity of spore types in the original field soils.  相似文献

Translocation of 14C-photosynthates to mycorrhizal (+ +), half mycorrhizal (0+), and nonmycorrhizal (00) split-root systems was compared to P accumulation in leaves of the host plant. Carrizo citrange seedlings (Poncirus trifoliata [L.] Raf. × Citrus sinensis [L.] Osbeck) were inoculated with the vesicular-arbuscular mycorrhizal fungus Glomus intraradices Schenck and Smith. Plants were exposed to 14 CO2 for 10 minutes and ambient air for 2 hours. Three to 4% of recently labeled photosynthate was allocated to metabolism of the mycorrhiza in each inoculated root half independent of shoot P concentration, growth response, and whether one or both root halves were colonized. Nonmycorrhizal roots respired more of the label translocated to them than did mycorrhizal roots. Label recovered in the potting medium due to exudation or transport into extraradical hyphae was 5 to 6 times greater for (+ +) versus (00) plants. In low nutrient media, roots of (0+) and (+ +) plants transported more P to leaves per root weight than roots of (00) plants. However, when C translocated to roots utilized for respiration, exudation, etc., as well as growth is considered, (00) plant roots were at least as efficient at P uptake (benefit) per C utilized (cost) as (0+) and (+ +) plants. Root systems of (+ +) plants did not supply more P to leaves than (0+) plants in higher nutrient media, yet they still allocated twice the 14C-photosynthate to the mycorrhiza as did (0+) root systems. This indicates there is an optimal level of mycorrhizal colonization above which the plant receives no enhanced P uptake yet continues to partition photosynthates to metabolism of the mycorrhiza.  相似文献

Seedlings ofAcacia auriculiformis A. Cunn. ex. Benth.,Albizia lebbeck (L.) Benth.,Gliricidia sepium (Jac.) Walp andLeucaena leucocephala (Lam.) de Wit. were inoculated with an ectomycorrhizal (Boletus suillus (l. ex. Fr.) or indigenous vesicular-arbuscular mycorrhizal (VAM) fungi in a low P soil. The plants were subjected to unstressed (well-watered) and drought-stressed (water-stressed) conditions. InGliricidia andLeucaena, both mycorrhizal inoculations stimulated greater plant growth, P and N uptake compared to their non-mycorrhizal (NM) plants under both watering regimes. However, inAcacia andAlbizia, these parameters were only stimulated by either ectomycorrhiza (Acacia) or VA mycorrhiza (Albizia). Growth reduction occurred as a result of inoculation with the other type of mycorrhiza. This was attributed to competition for carbon betweenAcacia and VA mycorrhizas and parasitic association betweenAlbizia and ectomycorrhiza. Drought-stressed mycorrhizal and NMLeucaena, and drought-stressed mycorrhizalAcacia tolerated lower xylem pressure potentials and larger water losses than the drought-stressed mycorrhizal and NMAlbizia andGliricidia. These latter plants avoided drought by maintaining higher xylem pressure potentials and leaf relative water content (RWC). All the four leguminous plants were mycorrhizal dependent. The higher the mycorrhizal dependency (MD), the lower the drought tolerance expressed in terms of drought response index (DRI). The DRI may be a useful determinant of MD, as they are inversely related.  相似文献

The fine lateral roots ofLysinema ciliatum R. Br., an epacridfrom habitats subject to periodic drought in Western Australia,are hair roots resembling those of Ericaceae. The finest (ultimate)hair roots have a cortex consisting only of an endodermis andan exodermis. Both layers have Casparian strips on the radialwalls. The exodermis develops to state III very close to theroot tip, showing wall thickening and a suberized lamella encirclingeach cell. In many roots collected after tip-growth had ceasedand the tip had fully differentiated this suberized exodermiscompletely encircled the apex. In older hair roots the epidermiscollapses or is sloughed off leaving the suberized exodermisas the outermost layer. The very fine hair roots have a verysmall stele containing only one xylem tracheid, and phloem consistingof a single sieve element with companion cell. The very smalldiameter of the single tracheid indicates a high resistanceto water flow along the hair roots. This may tend to conservesoil moisture in the region of the hair roots, leading to improvedsurvival and prolonged function of mycorrhizas in the field. Lysinema ciliatum R. Understanding how environmental factors affect the longevityof roots is essential if root mortality linked nutrient cyclingprocesses within ecosystems are to be understood, and the impactof natural and anthropogenic climate change properly evaluated.In this study the longevity of roots at two geographically andclimatically distinct sites were compared to identify the scaleof change that can occur due to environmental differences. Minirhizotrontubes were inserted into swards sown with the same variety ofLolium perenne and Trifolium repens at sites in the UK and Italy.Roots were viewed using a video camera and digitized imagesof roots generated at intervals. From these images the lifehistory of individual roots was determined and compared. Therewere few differences in patterns of longevity between differentspecies at the same site. Major differences, however, were observedbetween roots of the same species at different sites. For example,73% of Trifolium repens roots failed to survive for 21 d inItaly compared to only 29% at the UK site. Similarly, over 84%of Lolium perenne roots failed to survive for more than 21 din Italy compared to 38% in the UK. Five to 7 per cent of plants of Trifolium repens L. and T.pratenseL. and 100 per cent of plants of T. subterraneum L. were nodulatedby Rhizobium leguminosarum but none of T. hybridum L., T. glomeratumL. or T parvifirum Ehrh. The frequency of nodulation of T. pratenseby R. leguminosarum was much increased by breeding from susceptibleplants. Such plants were not nodulated by bacteria isolatedfrom any other cross-inoculation group, but remained fully susceptibleto R. trifolii. The nodules formed by R. leguminosarum are generallyassociated with lateral roots and are ineffective.  相似文献
