Summary A brief review is presented of the Vester-Ulbricht -decay Bremsstrahlen hypothesis for the origin of optical activity, and of subsequent experiments designed to test it. Certain of our experiments along these lines, begun in 1974 and involving the irradiation of racemic and optically active amino acids in a 61.7 KCi90Sr–90Y Bremsstrahlen source, have now been completed and are described. After 10.89 years of irradiation with a total Bremsstrahlen dose of 2.5×109 rads, crystallinedl-leucine, norleucine, and norvaline suffered 47.2, 33.6, and 27.4% radiolysis, respectively, but showed no evidence whatsoever of asymmetric degradation.d- andl-Leucine underwent about 48% radiolysis and showed 2.4–2.9% radioracemization. Other samples in solution were too severely degraded to analyze. Probable intrinsic reasons for the failure of the Vester-Ulbricht mechanism to afford asymmetric radiolysis in the present and related experiments involving -decay Bremsstrahlen are enumerated.A portion of this material was presented at the 7th International Conference on the Origins of Life, Mainz, FRG, July 10–15, 1983  相似文献

Conclusions The sexual theory of cell evolution has clear corollaries. All higher life would be initially sexual, and of two types (A- or Eu-karyotes). Relations would be dynamic with alternative unions and dissolution of the union of the two constituent moneric species, even as their viability as free-living species tended to decline in subsequent evolution. Superior bioenergetics meant eukaryotes were retained in multicellular species. However, some a-karyotes would be phylogenetically older than eukaryotes, a point which if verified, would render such biota of fundamental significance.  相似文献

As a contribution to the still open debate on the multiple african origins of the domesticated donkey, this work focuses on the wild ass of the Maghreb. It follows the recent paleontological review of the equids from Lac Karâr, a middle Pleistocene site in Northern Algeria, where most of the faunal remains were attributed to wild asses (Equus africanus Heuglin and Fitzinger, 1866). The morphometric resemblance between the equid of Lac Karâr, and Equus melkiensis Bagtach, Hadjouis and Eisenmann, 1984 which might correspond to the equid commonly referred to, as the Atlas wild ass (vernacular name) and, by descent, to Equus tabeti Arambourg, 1970, the only subspecies of ass from the early Pleistocene in Northern Africa, highlights the need for extensive biomolecular and radiometric studies on this wild and robust ass, endemic to the Maghreb. This is especially important given that progress in extracting ancient DNA from equids has been demonstrated by several recent studies and that remains of wild asses are still being uncovered in many late Pleistocene to Holocene (Neolithic) deposits in the Maghreb. The results from such studies could substantially improve the knowledge on the origin of asses. In particular, these analyses could shed light on the, as yet, undetermined clade 2 which may originate from the wild ass of the Maghreb as recently hypothesized by several authors. The archaeological and genetic data reviewed in this paper focuses on the ancient range of the Atlas wild ass mainly from sites in the northern part of the Maghreb; it contributes to current debates regarding this subspecies as one of the possible ancestors of modern asses. In addition, the paper identifies directions for further genetic researches on the extant donkey of the Maghreb.  相似文献

Plicatamide is a modified octapeptide from the ascidian Styela plicata having the structure Phe-Phe-His-Leu-His-Phe-His-decarboxyDeltaDOPA (where decarboxyDeltaDOPA = decarboxy-(E)-alpha,beta-dehydro-3, 4-dihydroxyphenylalanine). Edman sequencing, tandem mass spectrometry, and proton NMR have been used to characterize 100 microg of the compound. Plicatamide represents an important link between two classes of biomolecules: the tunichromes which share an oxidatively decarboxylated C-terminus and higher molecular weight DOPA-polypeptides. The 8-residue sequence provides the first opportunity to investigate the biosynthetic origins of the tunichrome family by molecular biological techniques.  相似文献

Macroautophagy is a transport pathway to the lysosome/vacuole that contributes to the degradation of numerous intracellular components. Despite the recent advances achieved in the understanding of the molecular mechanism underlying macroautophagy, the membrane origin of autophagosomes, the hallmark of this process is still a mystery. It has been suggested that mitochondria may be one of the lipid sources for autophagosome formation and that possibly this organelle provides the phosphatidylethanolamine (PE) that covalently links to the members of the ubiquitin-like Atg8/microtubule-associated protein 1 light chain 3 (LC3) protein family. These lipidated proteins are inserted into the outer and inner surface of autophagosomes and are essential for the biogenesis of these large double-membrane vesicles. However, because PE is an integral component of all cellular membranes, designing appropriate experiments to determine the origin of the autophagosomal PE is not easy. In this review, we discuss the idea that mitochondria provide the pool of PE necessary for the autophagosome biogenesis and we propose some possible experimental approaches aimed to explore this possibility.  相似文献

The set of these two theoretical papers offers an alternative to the hypothesis of a primordial RNA-world. The basic idea of these papers is to consider that the first prebiotic systems could have been networks of catalysed reactions encapsulated by a membrane. In order to test this hypothesis it was attempted to list the main obligatory features of living systems and see whether encapsulated biochemical networks could possibly display these features. The traits of living systems are the following: the ability they have to reproduce; the fact they possess an identity; the fact that biological events should be considered in the context of a history; the fact that living systems are able to evolve by selection of alterations of their structure and self-organization. The aim of these two papers is precisely to show that encapsulated biochemical networks can possess these properties and can be considered good candidates for the first prebiotic systems. In the present paper it is shown that if the proteinoids are not very specific catalysts and if some of the reactions of the network are autocatalytic whereas others are not, the resulting system does not reach a steady-state and tends to duplicate. In the same line, these biochemical networks possess an identity, viz. an information, defined from the probability of occurrence of these nodes. Moreover interaction of two ligands can increase, or decrease, this information. In the first case, the system is defined as emergent, in the second case it is considered integrated. Another property of living systems is that their behaviour is defined in the context of a time-arrow. For instance, they are able to sense whether the intensity of a signal is reached after an increase, or a decrease. This property can be mimicked by a simple physico-chemical system made up of the diffusion of a ligand followed by its chemical transformation catalysed by a proteinoid displaying inhibition by excess substrate. Under these conditions the system reacts differently depending on whether the same ligand concentration is reached after an increase or a decrease.  相似文献

For decades, HDL and HDL-cholesterol (HDL-C) levels were viewed as synonymous, and modulation of HDL-C levels by drug therapy held great promise for the prevention and treatment of cardiovascular disease. Nevertheless, recent failures of drugs that raise HDL-C to reduce cardiovascular risk and the now greater understanding of the complexity of HDL composition and biology have prompted researchers in the field to redefine HDL. As such, the focus of HDL has now started to shift away from a cholesterol-centric view toward HDL particle number, subclasses, and other alternative metrics of HDL. Many of the recently discovered functions of HDL are, in fact, not strictly conferred by its ability to promote cholesterol flux but by the other molecules it transports, including a diverse set of proteins, small RNAs, hormones, carotenoids, vitamins, and bioactive lipids. Based on HDL''s ability to interact with almost all cells and transport and deliver fat-soluble cargo, HDL has the remarkable capacity to affect a wide variety of endocrine-like systems. In this review, we characterize HDL''s unique cargo and address the functional relevance and consequences of HDL transport and delivery of noncholesterol molecules to recipient cells and tissues.  相似文献

Microbial hydantoinases – industrial enzymes from the origin of life?   总被引:9,自引:0,他引:9  
Hydantoinases are valuable enzymes for the production of optically pure d- and l-amino acids. They catalyse the reversible hydrolytic ring cleavage of hydantoin or 5′-monosubstituted hydantoins and are therefore classified in the EC nomenclature as cyclic amidases (EC 3.5.2.). In the EC nomenclature, four different hydantoin-cleaving enzymes are described: dihydropyrimidinase (3.5.2.2), allantoinase (EC 3.5.2.5), carboxymethylhydantoinase (EC 3.5.2.4), and N-methylhydantoinase (EC 3.5.2.14). Beside these, other hydantoinases with known metabolic functions, such as imidase and carboxyethylhydantoinase and enzymes with unknown metabolic function, are described in the literature and have not yet been classified. An important question is whether the distinct hydantoinases, which are frequently classified as l-, d-, and non-selective hydantoinases depending on their substrate specificity and stereoselectivity, are related to each other. In order to investigate the evolutionary relationship, amino acid sequence data can be used for a phylogenetic analysis. Although most of these enzymes only share limited sequence homology (identity<15%) and therefore are only distantly related, it can be shown (i) that most of them are members of a broad set of amidases with similarities to ureases and build a protein superfamily, whereas ATP-dependent hydantoinases are not related, (ii) that the urease-related amidases have evolved divergently from a common ancestor and (iii) that they share a metal-binding motif consisting of conserved histidine residues. The difference in enantioselectivity used for the classification of hydantoinases on the basis of their biotechnological value does not reflect their evolutionary relationship, which is to a more diverse group of enzymes than was assumed earlier. This protein superfamily probably has its origin in the prebiotic conditions of the primitive earth. Understood as a philosophy counselling group rights and concomitant public policies undertaken by governments to preserve and enhance group identities, multiculturalism has little purchase in European immigrant integration debates. However, based on interviews with various actors engaged in policies that affect immigrants in the Netherlands and Germany, we argue that multiculturalism remains a going concern if it is understood as a problem-solving practice through which governments acknowledge and work with ethnic and religious groups to address issues of mutual concern, particularly on issues related to gender. We label this ‘multicultural governance’ and argue that it supports the principal ends of integration policy: the socialization of immigrants into prevailing national languages, norms and institutions.  相似文献

The origin of life is an extraordinary problem that leads back to the structure and dynamics of the cosmos and early development of organic molecules. Within that wider question lies an unsolved problem that has troubled biologists for 150 years. What is the origin of the dominant presence of left-handed stereoisomers of amino acids in nature even though their synthesis normally results in an equal mixture of the right- and left-handed molecular forms? We propose that asymmetric Earth rotation caused at dawn and dusk circularly polarized UV light (CPUVL) of opposite polarity and reversed temperature profiles in the oceans. Destruction of the d-isomer by CPUVL at dusk in a sea surface hotter than at dawn created a daily l-isomer excess protected from radiation by nightfall, preserved by down-flow (diffusive, mechanical) into cold, darker regions, eventually initiating an l-amino-acid excess embodied in early marine forms. Innumerable mechanisms have been proposed for the origin of l-chiral dominance in amino acids and none proven. Since the thalidomide tragedy, homochirality of amino acids has been a growing practical issue for medicine. Understanding its origin may bring further and unexpected benefits. It may also be a modest pointer to the possibility of positive answers to whether intelligent life will have the capacity to continue to protect itself from conditions inimical to survival.  相似文献

The complex pathway of tetrapyrrole biosynthesis can be dissected into five sections: the pathways that produce 5-aminolevulinate (the C-4 and the C-5 pathways), the steps that transform ALA to uroporphyrinogen III, which are ubiquitous in the biosynthesis of all tetrapyrroles, and the three branches producing specialized end products. These end products include corrins and siroheme, chlorophylls and hemes and linear tetrapyrroles. These branches have been subjects of recent reviews. The similarity of the louse faunas of flamingos and ducks has been used as evidence that these two groups of birds are closely related. However, the realization that ducks actually are more closely related to Galliformes caused many workers to reinterpret this similarity in parasite faunas as host switching from ducks to flamingos. Recent unexpected phylogenetic results on the relationships of waterbirds and their lice call for a reinterpretation of the origins of the lice of the enigmatic flamingos. Here, we bring together new evidence on the phylogenetic relationships of flamingos and their lice and show that the lice of flamingos and grebes are closely related because their hosts share a common ancestor (cospeciation). We also demonstrate that the similarity of the louse faunas of flamingos and ducks is a result of host switching from flamingos to ducks, rather than from ducks to flamingos.  相似文献
