Three phases of cohesion were observable during the development of compatible autografts in Solanum pennellii. Phase I cohesion 1) lasted 4–5 d after grafting, 2) was characterized by an average increase in tensile strength of 4 g breaking weight (BW) mm–2 graft area (GA) d–1, and 3) correlated positively with cellular interdigitation at the graft interface. The fresh weight of the scion increased by approximately 5% d–1 during the first 2 d after grafting. Phase II cohesion occurred 5–15 d after grafting, during which time 1) the tensile strength of the graft union increased by 14 g BW mm–2 GA d–1, 2) vascular differentiation across the graft interface was completed, and 3) the fresh weight of the scion increased by 9% d–1. Phase III cohesion occurred subsequent to 15 d after grafting, during which time 1) the tensile strength of the graft union leveled off at a value similar to that of an ungrafted internode, and 2) the fresh weight of the scion increased by 14% d–1. These results are discussed relative to mechanisms underlying the formation of compatible grafts.  相似文献

The total electrical responses and action potentials of the neurons in the medial geniculate bodies in Vespertilionidae and Rhinolophidae were investigated. Maximum sensitivity to ultrasonic stimuli was recorded inMyotis oxygnathus (Vespertilionidae) in the range 10–40 kc/sec and 65–80 kc/sec, and in Rhinolophidae in the ranges 10–70 kc/sec and 81–86 kc/sec. Low thresholds were observed inMyotis oxygnathus for the frequencies covered by their echo-location cries, whereas the thresholds recorded in Rhinolophidae in the 80 kc/sec band (the principal frequency of their echo location cries) were 15–30 dB higher than those for adjacent frequencies. Minimum thresholds of off-responses were observed inMyotis oxygnathus in the range 50–60 kc/sec, and in Rhinolophidae in the range 78–80 kc/sec. The regions of neuronal response in both species of bat were generally similar in form to those of responses recorded in the medial geniculate bodies of other mammals. Some of the neurons in Rhinolophidae with a characteristic frequency of about 80 kc/sec were also sensitive to stimuli with one-half and one-third of the principal frequency. In Rhinolophidae the greatest selectivity for frequencies was possessed by neurons that responded within the range from 80 to 90 kc/sec.A. A. Zhadanov Leningrad State University. The response was investigated of neurons composing the cerebral ganglia inAchatina fulica (the Giant African snail) to application of acetylcholine (ACh), gamma-aminobutyric acid (GABA), and glycine (Gly). Chloride-dependent currents induced by these transmitters in 1 1/2-month old siblings were inhibited by dibutyryl-cAMP and strychnine. Inhibition of ACh response produced 10–8 M GABA was mimicked by application of dibutyryl-cAMP and isobutylmethylxanthine. Complete cross-desensitization was characteristic of both GABA- and Bly-induced response, but this effect did not occur when ACh and GABA (or Gly) were applied. A conclusion was reached on the basis of the pharmacological relationship between GABA- and Gly-induced response that these amino acids act on a single receptor — channel complex in the neurons of infant snails, whereas ACh-, GABA-, and Gly-induced chloride currents were not so related in cells of 4 year-oldAchatina.A. The ionic mechanisms of hyperpolarization produced by applying oxytocin (OT) were investigated at the membrane of identifiedHelix pomatia neurons. Two types of neuron were known to exist, in one of which hyperpolarization is produced by a reduction in chloride ions at the membrane and a rise in membrane permeability to potassium ions in the other. In the first of these, response to OT had a reversal potential of –40 mV and decreased when furosemide and tolbutamide were added to the external medium. In the second case, the potential of the reversal of the response to OT was –70 mV. Upon doubling of potassium ion concentration in the external solution it was shifted towards depolarization by 15 mV. It is sugested thatHelix pomatia neurons have different types of OT receptors, some of which, when activated, manifest reduced chloride permeability at the membrane (probably through the cell cyclase system) with a rise in potassium permeability at the membrane in others.A. Protoplasts were isolated from female gametophytes of Chondrus crispus (Stackh.) using commercial cellulase and various carrageenases prepared from marine bacteria. Depending on the nature of the donor tissue (apices or whole thallus, wild or cultivated strains), yields ranged from 1.0–8.5×108 protoplasts per gram of fresh tissue. Preincubating the tissue with a potassium chelator, Kryptofix 222, enhanced protoplast yields by 30–50 %. Based on staining with fluorescein diacetate most protoplasts were viable. A few protoplasts regenerated a cell wall and divided.  相似文献

Currents passing through individual potassium channels with anomalous (inward) rectification were recorded at the neuronal membrane ofPlanorbarius corneus using the patch clamp technique. These currents could be detected, whether in "right side out" or "inside out" configurations in the presence of 50 mM potassium ions or one of the potassium channel blockers: tetraethylammonium (TEA), barium, or cesium (2–20 mM) on the external side of the membrane. Inward currents were observed in individual channels at potentials more negative than level of potassium equilibrium potential (Ek); conductance of these measured 81±12 pS (n=11). At more positive potentials than Ek, conductance fell to zero. Potassium channels with anomalous (inward) rectification inPlanorbarius corneus resemble equivalent channels in other cells in their kinetics: time scale of the open state may be described by a single exponential function. This would imply that the ionic channel has a single open state. Time scale of the closed state was biexponential, thus indicating the possible existence of two kinetically different nonconducting states of the potassium channel with anomalous (inward) rectification at the neuronal membrane ofPlanorbarius corneus.I. Treatment of the roots of 24–48 h old seedlings of the forage legumeLotus corniculatus with 1.0% Cellulase YC, and 0.1% Pectolyase Y-23 in 4.2% mannitol solution released protoplasts from the tips of root hairs within 30–40 sec of enzyme incubation. Roots from approximately 1000 seedlings yielded 1.7×105 protoplasts. Ten percent of protoplasts divided to form cell colonies when cultured at 1.0×105 ml–1 in droplets of KM8P medium with 0.6% Sea Plaque agarose. Colonies formed callus on UM agar medium; protoplast-derived tissues produced shoots on B5 medium containing 0.05 mg 1–1 of BAP. Regenerated plants were phenotypically and cytologically normal (2n=2x=24±2), and produced nitrogen fixing root nodules following inoculation withRhizobium. These results confirm the totipotency of protoplasts isolated from specialised epidermal cells of seedling roots ofLotus corniculatus.  相似文献

Summary The fermentation of large sugar cane chips (1.0–1.5 in) to ethanol by Zymomonas mobilis CP4 (Z. mobilis) was studied in two glass fermentors operating with culture circulation for agitation (the EX-FERM type): a. A laboratory scale(2.5 liter) cylindrical vessel; b. A bench scale (8 liter) wide vessel. Z. mobilis cultures consumed 89–96% of the cane sucrose, converting it to ethanol by 90–97% of the theoretical yield in the laboratory scale fermentor and by 83–90% in the bench scale fermentor culture. Comparative Saccharomyces spp. cultures in laboratory fermentor consumed 96–98% of the cane sucrose, with ethanol conversion of only 75–79% of the theoretical yield.These preliminary results indicated that sucrose in agricultural size sugar cane chips was ethanol fermentable as compared to small size sugar cane chips or to sugar cane juice. Z. mobilis CP4 cultures converted sucrose more efficiently to ethanol than Saccharomyces spp. as shown in the laboratory scale fermentor studies.The ethanol yields in a wide bench scale fermentor cultures were slightly lower than in a laboratory fermentor.  相似文献

Protoplasts were isolated at high yields from actively growing callus and cell suspensions of cotyledons and needles of mature trees. The best protoplast growth response was obtained from cell suspensions of cotyledon and needle callus. Lower protoplast yields were obtained directly from young needles of flushing buds on explants from mature shoots (30-year-old trees) growing in vitro. In all cases, the first divisions, promoted by dimethyl sulfoxide, were observed in 10–45% of the protoplasts by 7–10 days. After 25–30 days, colonies of 8–10 cells were established. Summary The morphogenetic potential of the shoot tip explants ofEnsete superbum (Roxb.) Cheesman, a wild relative of the cultivated bananas, was investigated and an effective clonal propagation method devised. Shoot tip explants grown in modified MS medium containing 1.5 mg l–1 BAP and 1 mg l–1 KIN developed corms which on transfer to medium containing 3 mg l–1 IBA and 1.5 mg l–1 BAP, regenerated a large number of shoots from the surface of the corm, the origin of which was traced to single hypodermal cells. Shoots were rooted on a half-strength MS medium salts containing 3 mg l–1 IBA and 0.1 mg l–1 BAP. The rooted plantlets were hardened and planted in the field where the plants looked normal.  相似文献

Summary A 2.9 kb DNA fragment encoding s-(2-aminoethyl)-l-cysteine (AEC) resistant aspartokinase ofCorynebacterium glutamicum was cloned on theC. glutamicum/E. coli shuttle vector pECCG117. Summary A novel procedure employing mild sonication for transformation of plant protoplasts is described. Transient expression of a chloramphenicol acetyltransferase (CAT) gene in protoplasts of sugar beet (Beta vulgaris L.) and tobacco (Nicotiana tabacum L.) was obtained by a brief exposure of the protoplasts to 20 kHz ultrasound in the presence of plasmid DNA. Maximum levels of CAT activity were achieved by sonication for 500–900 ms at 30–70 W electric power (0.65–1.6 W/cm2 acoustic power). This reduced the viability to 15–20 % and 60 % for sugar beet and tobacco protoplasts, respectively. Up to 12 % (sugar beet) and 81 % (tobacco) of maximum transient expression could be achieved with no significant loss of viability. Protoplasts surviving exposure to ultrasound were found to have a similar long-term viability and to regenerate to micro-calli as untreated protoplasts. Plasmid DNA concentrations of 80–110 g/ml and sucrose concentrations of 21–28 % in the sonication medium were found to be optimal for transient expression.Abbreviations CAT chloramphenicol acetyltransferase  相似文献

Summary Several amino acid requiring auxotrophs have been isolated from unsupplemented protoplast cultures of haploid Nicotiana plumbaginifolia following incubation with BUdR (1-5x10-5, 2 days) and recovery on complete medium. The auxotrophic lines required the following amino acid(s) for growth: his, ile, leu, ile+val, met or try. Met is a new type isolated in higher plants. The same absolute amino acid requirement was observed in plants regenerated from auxotrophic cultures. Precursor feeding tests, enzyme assays, and/or metabolic complementation through protoplast fusion were used to identify the genetic lesion leading to auxotrophy. Mutant seeds were obtained from supplemented Met plants. Seeds were also collected from selfed plants regenerated from various complementing fusion products, and a His revertant. Summary Nine species of fungi viz.,Aspergillus niger,A. flavus,A. terreus,Fusarium solani,Mucor sp.,Neurospora crassa,Penicillium janthinellum,Trichoderma harzianum andTrichothecium roseum were evaluated for their potential to remove NH3–N from domestic waste water. Of the fungi tested,A. flavus was found to be the most effective in the removal of NH3–N. Maximum reduction (92%) of NH3–N by this organism was observed at pH 8.0 at 20°C.  相似文献

We have examined a variety of common mutagens in producing auxotrophic mutants in cowpea rhizobia strains JRC23 and IRC256. While NTG (N-methyl-N-nitro-N-nitrosoguanidine), EMS (ethylmethane sulfonate), NA (nitrous acid), and UV (ultraviolet) irradiation were mutagenic with the strain JRC23, these mutagenic agents did not mutate strain IRC256. On the contrary, transposon mutagenesis with Tn5 yielded auxotrophs in strain IRC256 but not in strain JRC23, while only methionine (Met) auxotrophs from strain JRC23, histidine (His), and adenine plus thiamine (Ade+Thi) auxotrophs from strain IRC256 were isolated.  相似文献

Summary AnEscherichia coli/Streptomyces bifunctional cosmid vector, pJP3, which selects for clones with insert size 23–33 kilobase pairs (kb), has been constructed. A genomic DNA library fromS. kanamyceticus has been stably maintained inE. coli using pJP3. Advanced intercross lines (AIL) and interval–specific congenic strains (ISCS) were used to fine map previously coarsely defined quantitative trait loci (QTL) on Chromosomes 1,10, and 19, influencing behaviors in the open Field (OF) and light–dark (LD) paradigms in mice. F12(A × B) AIL mice (N = 1130) were phenotyped, genotyped, and mapped. The ISCS were studied only in the telomeric Chromosome 10 region of interest, containing the exploratory and excitability QTL1 (Exq1). The Chromosome 10 Exq1 and Chromosome 19 Exq4 loci mapped robustly in the AIL. The most significant QTL findings (2.0 LOD score intervals; peak; LOD score) came from the TD15 and LD transitions traits, yielding estimated intervals of 2.2 cM for Exq1 (71.3–73.5 cM; peak 72.3 cM; LOD 11.9) and 9.0 cM for Exq4 (29.0–38.2 cM; peak 34 cM; LOD 4.2). The replicated QTLs on Chromosome 1 failed to map in this AIL population. The ISCS data confirmed Exq1 loci in general. However, the ISCS data were complex and less definitive for localizing the Exq1 loci. These exploratory and fear-like behaviors result from inheriting “many small things,” namely, QTL explaining 2%–7% of the phenotypic variance. These results highlight the challenges of positionally cloning loci of small effect for complex traits. A procedure was developed which demonstrates the expression of differential resistance in pine callus tissues to the fungal pathogen Phytophthora cinnamomi Rands. Callus tissues were maintained on a modified Murashige and Skoog medium with 10–5M 2,4-D and inoculated with hyphae of P. cinnamomi at 26°C in the dark. The number of intracellular hyphae was used as an index of resistance. Summary A turbidostatic and oxystatic fermentation system was used to study the growth kinetic ofMethylococcus capsulatus (Bath). Dissolved oxygen and methane concentrations were measured continuously with membrane inlet mass spectrometry. The specific growth rate was found to increase from 0.25 h–1 to 0.37 h–1 and the saturation constant for methane was found to decrease from 71 M to 1.3 M as the copper content of the medium was varied from a very low to a high value.  相似文献

A method is described for rapid multiplication from axillary buds of six Mentha species. Nodal segments from one-year old plants were grown on Murashige and Skoog medium (BMS), supplemented with BAP (1.0; 2.0 mg/l) and KIN (1.0 mg/l) and kept at 28 ± 1°C under fluorescent light for 30 days. After this period, several shoots (15–20 shoots per explant) with roots were produced which were placed in soil for further growth.  相似文献
