Nitrogenase activity measured by acetylene reduction, was detected when surface-sterilized spores of vesicular-arbuscular (VA) mycorrhizal fungi (Glomus fasciculatum, G. intraradices, G. scientillans, G. mosseae, Gigaspora gilmorei andEndogone dusii) were inoculated into nitrogen-free liquid medium containing malic acid and incubated under microaerophilic conditions (99% N2+1% O2) at 30°C.Azospirillum species were isolated from the nitrogenase-active cultures.  相似文献

The ability of Hawaiian volcanic soils to nodulate actinorhizal Myrica cerifera, Casuarina equisetifolia, and Alnus glutinosa was determined using a host-plant bioassay. Myrica-nodulating Frankia occurred in five volcanic deposits with depositional ages ranging from 20 to 162 years before present. The oldest deposit had a mean estimated nodulation capacity from 450 to 1200 times greater than those of the younger deposits. Only the oldest deposit had high moisture content, high organic matter content, and increased vegetative cover, including an abundance of actinorhizal M. faya. Casuarina- and Alnus-nodulating Frankia were not detected in any of these volcanic deposits.  相似文献

The phosphate-solubilizing bacteria (PSB) were enumerated in 52 soil samples collected from agricultural areas at Baghdad. The results revealed that more than 90% of the samples were inhabited with indigenous PSB. The number varied and ranged from 0.012–28.4×104 cell g–1 soil. The correlations between PSB counts and electrical conductivity, available phosphorus, cation-exchange capacity, soil moisture, organic matter and pH were insignificant. Both abundance and numbers of PSB were more pronounced in descending order under vegetables, legumes, grasses, cereal and orchard trees.  相似文献

Soil samples from 30 fields in neighbouring districts of Varanasi were screened for rhizobiophages on 60 Rhizobium strains. Plaques were observed on five strains: P1, P5, SU391 (R. leguminosarum), CB756 and 32H1 (Rhizobium sp.). Rhizobiophages infective on one or more of the five strains were present in all fields. There seems to be no correlation between presence of phage and the standing crop or the pH (7.1-8.2) of the soil. Eight distinct rhizobiophages have been isolated and characterized for host specificity, plaque morphology and maximum titer in broth.  相似文献

The occurrence of Clostridium perfringens was estimated in 750 samples originated from a variety of soils bearing various bulb crops: Brawnica oderacea (vegetable), Olea europaea, Daucus carota (carote), Solanum tuberosum (potato), Phaseolus vulgaris (green haricot), Beta vulgaris var. rapaceum (beetroot), Cucurbita pepo (squash), Allium cepa (onion), Cucumis sativus (cucumber) and Capsicum annum (pepper). All isolated strains were tested for their antimicrobial activities to amoxicillin, penicillin G, kanamycin, tetracycline, streptomycin, erythromycin, chloramphenicol and metronidazole.When considering the type of the bulb production, it was observed increased number of C. perfringens spore densities in the most undersurface bulb soils. Moreover, C. perfringens spore are likely to occur in particularly large numbers in soil contaminated by fecal matter. Additionally, there is a close relationship between the spore amount and nature of organic content. Presence of C. perfringens was associated with acidic soil. Most of our strains showed resistance to the studied antibiotics applied usually for human and veterinary care. A systematic monitoring of the cultivated soil ecosystems must include bacteriological parameters together with chemical indices of organic pollution in order to obtain information adequate for assessing their overall quality.  相似文献

The pathways for catabolism of fructose were investigated in the type strains of Azospirillum lipoferum and Azospirillum brasilense grown aerobically with (NH4)2SO4 as the nitrogen source. When grown on fructose, the former species possessed a complete Entner-Doudoroff pathway, whereas the latter species lacked activity for glucose-6-phosphate dehydrogenase. Both species possessed a complete catabolic Embden-Meyerhof-Parnas pathway. Neither species possessed the key enzyme of the hexose monophosphate pathway, 6-phosphogluconate dehydrogenase. Both species could phosphorylate fructose to fructose-1-phosphate by means of a phosphoenolpyruvate-phosphotransferase system, and high activities of 1-phosphofructokinase occurred. Both species possessed glucokinase activity, but only A. lipoferum had hexokinase activity; moreover, the cells of A. brasilense were nearly impermeable to glucose, accounting for the inability of this species to grow on glucose. Both species possessed pyruvate dehydrogenase, a complete tricarboxylic acid cycle, a glyoxylate shunt, and malic enzyme. Analysis of the acidic end products for both species indicated the formation of only small amounts of various organic acids, and most of the titratable acidity was due to utilization of the ammonium ions of the medium. Gluconic acid was not formed during growth of either species on fructose but was detected during growth of A. lipoferum on glucose; this species also possessed an NADP-linked glucose dehydrogenase and gluconokinase.  相似文献

Occurrence of natural organic chlorine in soils for different land uses   总被引:1,自引:0,他引:1  
Consideration of natural formation of organochlorine compounds in soils is necessary in radioecology in order to understand chlorine radioisotope (36Cl) cycling in various environments for safety assessment purposes, but also in ecotoxicology because certain chlorinated organics in soils are toxic compounds. On the other hand, occurrence of organic chlorine in soils is poorly documented, especially in non-forest ecosystems. We measured total and organic chlorine concentrations in 51 French surface soils sampled from grassland, arable and forest sites on a national scale (French soil quality monitoring network) in order to characterize the variability of organic chlorine concentrations for these different land uses. While previous studies reported that the chlorination of soil organic matter is responsible for chlorine retention in temperate forest ecosystems, this study shows that the non-extractable organohalogen pool accounts for the majority (>80 % on an average) of the total measurable chlorine in grassland and agricultural soils. This suggests that natural chlorination is a widespread phenomenon in all kinds of soils. A multiple linear regression analysis performed on the dataset indicated that retention of organochlorine in soils is related to the organic carbon content, Cl input and soil pH.  相似文献

Application of a peat-based powder inoculant of Azospirillum brasilense, as well as a granular inoculant (each containing 0.5–1.0×107 Azospirillum/g moist peat), in the seed furrows of Zea mays resulted in significantly increased yields (11 to 14%) in light soils at low rates of N fertilization. In general, there was no effect of inoculation on plant yields in heavier soils nor when N fertilization was high. Pre-emergence application of granular inoculant and inoculation associated with irrigation were more efficient in increasing yield than inoculation post-emergence or seed coating.E. Summary Nitrogen fixation by strains of Azospirillum isolated from several rice soils and rice cultivars was investigated by15N2 incorporation and C2H2 reduction. C2H2 reducing ability markedly varied among the strains obtained from soils differing widely in their physico-chemical properties. Large variations in15N2 incorporation by Azospirillum isolated from the roots of several rice cultivars were also noticed. The present study reveals that rice cultivars harbour Azospirillum with differential N2-fixing ability and that plant genotype is of importance for optimal associations.  相似文献

Summary The occurrence of phosphate-solubilizing fungi in coconut plantation soil types was investigated. The laterite, alluvial and clayey soils harboured more of the P-solubilizing fungi than the sandy soils. The isolated P-solubilizing fungi solubilized 26 to 74 per cent of the tricalcium phosphate in 5 to 15 days. The competitive saprophytic ability of the active P-solubilizing fungi in soil varied between the isolates. Eight fungi with high P-solubilizing capacity and high competitive saprophytic ability were recognised. Azospirillum brasilense, A. amazonense, and A. lipoferum strains were screened for restriction endonucleases using phage lambda DNA. The extract of A. brasilense 29711 cleaved lambda DNA into specific fragments. It was concluded that this strain possesses a class II restriction endonuclease which was named AbrI. AbrI has a single recognition site on lambda DNA at position of approx. 33 500 bp. AbrI was characterized as an isoschizomer of XhoI, which cuts lambda DNA at 33 498 bp and cleaves double-stranded DNA at the sequence 5'-C TCGAG-3'. From other Azospirilla strains only A. amazonense QRZ42 extracts (AamI activity) cleaved DNA into specific fragments under certain conditions.  相似文献
