Artibeus lituratus is one of the most recognized bat species in the neotropics, probably due to its high abundance in urban areas. Despite its wide distribution, the morphology and physiology of its reproductive organs remain unclear. Thus, the aim of the present study was to describe the morphology and histology of the female reproductive organs of this species, and evaluate morphological variations during its different reproductive phases (nonreproductive, pregnant, lactating, etc.). Thirty adult females were collected, divided into six (6) sample groups, according to the reproductive status, and submitted to anatomical and histological analyses. We show that the ovaries of A. lituratus are polarized, present a low number of interstitial glands, have large and well-developed Graafian follicles and a persistent corpus luteum, and may present a functional everted corpus luteum. Ovulation is simple, unilateral and nonpreferential, possibly alternated in successive ovulations, and may be sequential, with the occurrence of postpartum estrus. The uterus is simplex, the implantation is fundic and interstitial and the placenta is chorioallantoic, discoidal, and hemochorial. The female reproductive organs of A. lituratus share similarities with the pattern observed in human, which indicates that A. lituratus may be an interesting animal model in reproductive studies.  相似文献

Lowland Amazon is climatically one of the least seasonal regions on the planet, but little is known about how this is reflected in ecological seasonality. The central objective of this study was to determine whether seasonal fluctuations in the availability of fruit resources in Neotropical forests are sufficiently marked to affect the ecology and physiology of frugivorous bats. Seasonal variations in the overall bat abundance and in the captures, body condition, and reproductive activity of the two most abundant species, Carollia perspicillata and Artibeus planirostris, were studied within a region of central Brazilian Amazonia dominated by a mosaic of nonflooded (terra firme) and seasonally flooded forests (várzea and igapó). Concurrent seasonal changes in fruit availability were measured. The abundance of fruits was markedly seasonal, with far fewer resources available during the low-water season. There was a positive correlation between fruit and bat abundance. Overall, bats did not increase the consumption of arthropods during the period of fruit shortage. In A. planirostris, the body condition declined when fruits were scarcer. In both C. perspicillata and A. planirostris, foraging and reproductive activity were positively correlated with fruit availability. Consequently, the results suggest that resource seasonality is sufficiently marked to affect frugivorous bats and force them to make important eco-physiological adjustments.  相似文献

The microscopic structure of theAlouatta caraya ovary is studied in different ages and reproductive stages. The most significant feature seems to be the presence in adult ovaries of abundant glandular interstitial tissue which occupies both the cortex and medulla. It seems to be derived from the theca interna of atretic follicles. Discrete luteinized masses are present in the medulla in all the ovaries observed. Invaginations of the surface epithelium are seen only in infant and juvenile ovaries. The development of cystic follicles seems to be a common pathway of atresia.  相似文献

In this work we studied the structures of 51 foetal and 14 adult ovaries obtained from slaughtered Kano brown does in Nsukka abattoir. The ages of the adult does were determined by dentition and foetuses by crown rump length method. The foetal and adult ovaries were divided into five different groups using specific age intervals as Gestation day (GD) 50–65, 66–95, 96–125 and 126–145 and adults. For histological studies the ovaries were fixed, processed and routinely stained with H&E. The ovarian follicles were classified into 5 types according to granulosa cell layers surrounding the oocytes. The number of ovarian follicles per microscopic field, number of granulosa cells surrounding type 1 and 1A follicles and diameter of the ovarian follicles were determined for each group at 400× magnification. Grossly the foetal ovaries were like pin head, oval in shape, uniformly smooth and creamy in colour. The adult ovaries had follicles with different sizes. The adult mean ovarian weights were significantly higher (P < 0.01) than those of the foetuses. Microscopically, the GD 50–65 ovaries had no distinct cortex and medulla. Oogonia were numerous among other stromal cells toward the periphery of the ovary. By GD 66–95 the ovaries contained types 1, 1a, 2 and 3 follicles. GD 96–125 ovaries contained type 4 follicles with early antrum formation and those of GD 126–145 comprised type 5 among other follicles. The adult ovaries comprised all the ovarian follicle types. The number of type 1 follicles increased significantly (P < 0.01) with foetal age. It was least in the adults. The diameter of adult follicles was significantly higher (P < 0.01) than those of the foetuses. This result provides baseline information on the morphological development of ovaries in Kano brown goats.  相似文献

The ovary of the seahorse, Hippocampus erectus, is a cylindrical tube bounded by an outer layer consisting of a mesothelium and muscular wall and by an inner luminal epithelium, with a single row of developing follicles sandwiched between the two layers. Follicles are produced by a germinal ridge, which contains oogonia, early oocytes, and prefollicle cells, and which runs along the length of the ovary. The germinal ridge is an outpocketing of the luminal epithelium, as indicated by a continuous underlying basal lamina. Prefollicle cells invest diplotene oocytes and the complex eventually pinches off the germinal ridge as a primordial follicle surrounded by a basal lamina derived from the germinal ridge. Subsequent investment of the primordial follicle by elements of the theca complete the process of folliculogenesis. H. erectus has two ovaries and each ovary has two dorsally located germinal ridges. Thus, in each ovary the derived follicular lamina is bilaterally symmetrical: two temporally and spatially arranged sequences of developing follicles are produced, with the largest follicles found along the ventral midline of the ovary. The advantages of developmental, kinetic, and systemic analyses of these unusual ovaries are indicated.  相似文献

The mammalian ovary is composed of ovarian follicles, each follicle consisting of a single oocyte surrounded by somatic granulosa cells, enclosed together within a basement membrane. A finite pool of follicles is laid down during embryonic development, when oocytes in meiotic arrest form a close association with flattened granulosa cells, forming primordial follicles. By or shortly after birth, mammalian ovaries contain their lifetime’s supply of primordial follicles, from which point onwards there is a steady release of follicles into the growing follicular pool.The ovary is particularly amenable to development in vitro, with follicles growing in a highly physiological manner in culture. This work describes the culture of whole neonatal ovaries containing primordial follicles, and the culture of individual ovarian follicles, a method which can support the development of follicles from an immature through to the preovulatory stage, after which their oocytes are able to undergo fertilization in vitro. The work outlined here uses culture systems to determine how the ovary is affected by exposure to external compounds. We also describe a co-culture system, which allows investigation of the interactions that occur between growing follicles and the non-growing pool of primordial follicles.  相似文献

Summary Sympathetic neurotransmitters have been shown to be present in the ovary of the rat during early postnatal development and to affect steroidogenesis before the ovary becomes responsive to gonadotropins, and before the first primordial follicles are formed. This study was undertaken to determine if development of the ovarian innervation is an event that antedates the initiation of folliculogenesis in the rat, Rattus norvegicus. Serial sections of postnatal ovaries revealed a negligible frequency of follicles 24 h after birth (about 1 primordial follicle per ovary). Twelve hours later there were about 500 follicles per ovary, a number that more than doubled to about 1300 during the subsequent 12 h, indicating that an explosive period of follicular differentiation occurs between the end of postnatal days 1 and 2. Electron microscopy demonstrated that before birth the ovaries are already innervated by fibers containing clear and dense-core vesicles. Immunohistochemistry performed on either fetal (day 19) or newborn (less than 15h after birth) ovaries showed the presence of catecholaminergic nerves, identified by their content of immunoreactive tyrosine hydroxylase (TH), the rate-limiting enzyme in catecholamine biosynthesis. While some of these fibers innervate blood vessels, others are associated with primordial ovarian cells, thereby suggesting their participation in non-vascular functions. Since prefollicular ovaries are insensitive to gonadotropins, the results suggest that the developing ovary becomes subjected to direct neurogenic influences before it acquires responsiveness to gonadotropins.  相似文献

Estrogen inhibits cyst breakdown and primordial follicle assembling of germ cells, but little is known about the underlying mechanisms. We aimed to analyze the effects of estrogen on the early development of mouse follicles using an in vitro ovary culture system and in vivo injection. Newborn mouse ovarian tissues were cultured in vitro for 2 or 4 days with estrogen of 0 M, 10−8 M and 10−4 M, respectively, and neonatal mice were injected with 5 mg/kg/day estrogen. We found that the percentages of different-stage follicles significantly varied between the control and estrogen-treated groups. In vitro experiments showed that the unassembled follicles accounted for 70.5 ± 2.7% and the primordial follicles accounted for 29.5 ± 2.7% in the treatment group, but in the control group, ovaries had 61.7 ± 8.4% unassembled follicles. In vivo experiments showed that the percentages of unassembled follicles and primordial follicles were 37.1 ± 5.2% and 51.6 ± 2.4% in the control group, while they were 72.6 ± 5.2% and 25.1 ± 5.5% in the treatment group. Moreover, we analyzed the expression of Kit ligand in mouse ovaries treated by estrogen with real-time PCR and western blot technology, and found that compared with the control group, both mRNA and protein expression levels were decreased in the treatment group (P < 0.05). These results indicate that estrogen inhibits the development of mouse ovarian follicles by regulating the expression of Kit ligand.  相似文献

In mammals, the primordial follicle pool represents the entire reproductive potential of a female. The transforming growth factor-β (TGF-β) family member activin (ACT) contributes to folliculogenesis, although the exact mechanism is not known. The role of FST288, the strongest ACT-neutralizing isoform of follistatin (FST), during cyst breakdown and primordial follicle formation in the fetal mice ovary was assessed using an in vitro culture system. FST was continuously expressed in the oocytes as well as the cuboidal granulosa cells of growing follicles in perinatal mouse ovaries. Treatment with FST288 delayed germ cell nest breakdown, particularly near the periphery of the ovary, and dramatically decreased the percentage of primordial follicles. In addition, there was a dramatic decrease in proliferation of granulosa cells and somatic cell expression of Notch signaling was impaired. In conclusion, FST288 impacts germ cell nest breakdown and primordial follicle assembly by inhibiting somatic cell proliferation.  相似文献

The reserve of primordial follicles, which serves all oocytes for the female reproductive lifespan, is established a few days after birth in mice. During this process, more than half of the oocytes are primarily eliminated by apoptosis. Autophagy, the conserved intracellular process maintaining cellular homeostasis, serves as a protective mechanism for oocyte survival. In the current study, we speculate a new role for autophagy during primordial folliculogenesis. Active autophagy was observed in perinatal ovaries from 16.5 days post coitus to 3 days post parturition. The inhibition of autophagy by 3-methyladenine (3-MA) increased the number of cyst oocytes and delayed follicle formation in vivo and in organ cultures. Furthermore, the reactive oxygen species (ROS) level was elevated in ovaries treated with 3-MA, while N-acetylcysteine, an oxidant, alleviated the inhibitory effect of 3-MA on primordial folliculogenesis. Additionally, the expression of growth differentiation factor 9 and transforming growth factor β1, which regulates follicle activation, was decreased after 3-MA treatment. These data suggest that the physiological level of autophagy in perinatal ovaries regulates germ cell cyst breakdown and primordial follicle assembly by ROS clearance and exerts extensive effects on further follicular development.  相似文献

Previous studies have reported that only primordial follicles and empty follicles can be found in 7.5 days postparturition (dpp) Sohlh1?/? mouse ovaries and females are infertility. There appears to be a defect in follicle development during the primordial‐to‐primary follicle transition in Sohlh1?/? mouse ovaries. However, detailed analyses of these phenomena have not been performed. In this study, we used Sohlh1?/? transgenic mice to explore the role of Sohlh1 in folliculogenesis. The results showed that only primordial follicles and empty follicles can be observed in Sohlh1?/? ovaries from 0.5 to 23.5 dpp. The expression of Foxo3 and FOXO3 was downregulated; nucleocytoplasmic shuttling of FOXO3 was normal in 7.5‐dpp Sohlh1+/+ but not Sohlh1?/? ovaries; and primordial follicle activation (PFA) was not observed in 7.5‐dpp Sohlh1?/? mice. The expression levels of KIT, AKT, and P308‐AKT were downregulated (p < 0.05), whereas that of P473‐AKT was not significantly changed (p > 0.05). The KIT/PI3K/AKT pathway was inhibited. Furthermore, we conducted a dual luciferase assay and chromatin immunoprecipitation. The results showed that SOHLH1 can upregulate the Kit gene by binding to the ?3698 bp E‐box motif. The absence of Sohlh1 may affect PFA in mouse ovaries via downregulation of Kit and inhibition of the KIT/PI3K/AKT pathway.  相似文献

Summary A method for superficial pinealectomy of the adult white-footed mouse,Peromyscus leucopus, is presented. Histological examination of the brain of pinealectomized mice showed that the deep pineal gland was left intact. The survival rate of pinealectomized mice was 80%. Pinealectomized mice were exposed to a short day photoperiod (8L:16D) at 15°C for 7 weeks. After this time male mice maintained active gonads with a testicular index (TI, testis width×length/body weight) of 2.0±0.1. Testis weight was 202±35 mg, and the seminiferous tubules contained abundant spermatozoa (spermatogenic index [SI]=4.5±0.2). Sham operated animals had regressed testes. TI was 1.2±0.2, testis weight was 97±26 mg, and the SI was 2.7±0.7 (allP<0.05 relative to pinealectomized mice). Pinealectomized females were reproductively competent in that all of the mice had a perforate vagina, the reproductive tract weight (vagina, uterus, oviducts, and ovaries) was 111±15 mg, and the ovaries from each animal contained preovulatory follicles. Sham operated mice had an imperforate vagina, reproductive tract weights were 34±5 mg, and in only 1 out of 5 mice did the ovaries contain a preovulatory follicle (allP<0.05). The weight of the lipid-free interscapular brown fat was 28% less in pinealectomized mice relative to sham operated animals (P<0.01). These results support the role of the pineal gland as regulator of short day, cold induced reproductive regression and brown fat hypertrophy.  相似文献

目的研究大鼠卵巢中卵泡生长发育的增龄性变化,为生殖生物学研究寻找动物模型.方法 用组织学方法,观察了0天~15月大鼠卵巢的形态结构.结果 0天见卵母细胞聚集呈团索状,未见卵泡;4天见大量原始卵泡;1周出现大量初级卵泡;2周见次级卵泡;3周次级卵泡数量增加,卵泡腔扩大;1月可见不同发育阶段的卵泡和闭锁卵泡;45天出现黄体;2月~5月,有大量黄体和生长卵泡;6月~8月,卵巢体积缩小,卵泡数量减少;12月萎缩,未见正常卵泡和黄体;15月,纤维化,可见囊泡.结论 大鼠从6月开始,生殖功能随月龄增长而进行性衰减,机体逐渐衰老.大鼠可用作研究女性生殖与衰老关系的动物模型.  相似文献

As the first long noncoding RNA to be discovered, H19 has gained substantial attention as a key regulator of several biological processes and its roles in female reproductive biology are gradually getting revealed. Herein, we have summarized the current evidence regarding H19 expression pattern and involvement in the developmental and pathological processes associated with the ovary and the placenta. The findings indicate that within the ovaries, H19 is expressed in the antral and cystic atretic follicles as well as in the corpora lutea but absent in the primordial, primary, and secondary follicles. Its normal expression promotes the maturation of antral follicles and prevents their premature selection for the ovulatory journey while its aberrant induction promotes polycystic ovary syndrome development and ovarian cancer metastasis. In the placenta, H19 is highly expressed in the cytotrophoblasts and extravillous trophoblasts but weakly expressed in the syncytiotrophoblast layer and potentially controls trophoblast cell fate decisions during placenta development. Abnormal expression of H19 is observed in the placental villi of pregnancies affected by pre-eclampsia and fetal growth restriction. Therefore, dysregulated H19 is a candidate biomarker and therapeutic target for the mitigation of ovarian and placenta-associated diseases.  相似文献

This study was performed to gain insight into the maturation of the reproductive system of Echinostoma paraensei worms grown in an early infection of Mesocricetus auratus. Hamsters were infected with 100 metacercariae and necropsied on days 3, 5, 7, 10 and 14 post infection (dpi). Recovered flukes stained with hydrochloric carmine were preserved as whole mounts and analyzed by light and confocal scanning laser microscopy. The average worm recovery was 43.7 per host. Images of the male and female reproductive systems were taken. The ovary and anterior and posterior testis were evidenced on day 3, while the ootype and cirrus sac were present on day 5. Confocal imaging showed primordium testis and ovary as a cluster of primordial cells from day 3 onward. The testes, ovary, cirrus sac and uterus organs were already present during the first week of life. The two testes were seen as individual structures on 7 dpi while the cirrus sac and vitelline glands were in development. The ovary was connected to the uterus while the ootype was adjacent to it. Both testes were larger than the ovary, showing cells at different stages of development, but with few bundles of functional spermatozoa in 10 day-old worms. On day 14, eggs and spermatozoa were seen in the uterus and seminal vesicle, respectively, while oocytes appeared in the ootype as fertilized eggs. We conclude that the reproductive system of E. paraensei was functional on 14 dpi in the hamsters.  相似文献

Age-related changes in ovarian morphology were studied in female Japanese monkeys,Macaca fuscata fuscata. A total of 47 nonlactating females of various ages ranging from new-born to >28 yr old were used. Ovarian size increased during the first decade of life, reached a plateau at around 10 yr. This was followed by a gradual decline throughout the remaining life span. The ovarian cortex of new-born animals consisted of numerous clusters of mitotic primordial germ cells. Such mitotic germ cells were observed even in the ovary of 28-day-old animal, but were not found in any animal after 1.5 yr of age. Numbers of primordial follicles decreased exponentially with the advance of age, and only a few primordial follicles were observed after about 16 yr of age. The numbers of primary and tertiary follicles increased from ages 4 to 16 yr, with a peak at 8 to 10 yr, and then decreased gradually. Developing tertiary follicles were observed as early as 1.5 yr of age. About 40% of tertiary follicles were atretic follicles throughout life, and their size was similar to that of developing tertiary follicles. Corpora lutea or corpora albicantia were found in ovaries more than 4 yr old. Remnants of corpora lutea and corpora albicantia, together with thick-walled blood vessels and fibrosis, became apparent in ovaries after 16 yr, and were observed characteristically in ovaries over 26 yr of age. There was no significant difference in the number or in the size of tertiary follicles between the breeding and nonbreeding seasons.  相似文献

ABSTRACT: BACKGROUND: Follicle numbers and developing ovarian morphology, particularly with reference to the presence of interstitial tissue, are intimately linked within the ovary of the African elephant during the period spanning mid-gestation to puberty. These have not been previously quantified in any studies. The collection of 7 sets of elephant fetal ovaries between 11.2 and 20.2 months of gestation, and 29 pairs of prepubertal calf ovaries between 2 months and 9 years of age during routine management off-takes of complete family groups in private conservancies in Zimbabwe provided an opportunity for a detailed study of this period. RESULTS: The changing morphology of the ovary is described as the presumptive cortex and medulla components of the fetal ovary settled into their adult form. Interstitial tissue dominated the ovary in late fetal life and these cells stained strongly for 3beta-hydroxysteroid dehydrogenase. This staining continued postnatally through to 4.5 years of age suggesting continued secretion of progestagens by the ovary during this period. The considerable growth of antral follicles peaked at 28% of ovarian volume at around 16.7 months of fetal age. The numbers of small follicles (primordial, early primary and true primary), counted in the cortex using stereological protocols, revealed fewer small follicles in the ovaries of animals aged 0 to 4.5 years of age than during either late fetal life or prepubertal life. CONCLUSIONS: The small follicle populations of the late-fetal and prepubertal ovaries of the African elephant were described along with the changing morphology of these organs. The changes noted represent a series of events that have been recorded only in the elephant and the giraffe species to date. The expansion of the interstitial tissue of the fetal ovary and its continued presence in early post natal life may well contribute to the control of follicle development in these early years. Further research is required to determine the reasons behind the variation of numbers of small follicles in the ovaries of prepubertal calves.  相似文献

The synergetic process of folliculogenesis is mainly regulated by GDF-9 and BMP-15 as well as their receptors, such as BMPR2, TβR1 and BMPR1B. Expressions of these factors and the receptors are significant different among species. This study was designed to detect expression of GDF-9, BMP-15 and their receptors in mouse, porcine and human healthy follicles by immunohistochemistry. Three ages of human ovary were studied according to ovarian developmental schedule, i.e. gestational week (GW) 16, puberty (14 year-old) and adult (40 year-old). The results showed that both GDF-9 and BMP-15 were detectable in oocytes from primary follicles onward, besides, BMP-15 also presented in granulosa cells (GCs) and follicular follicle of mature follicles in mouse. However, they were maintained in oocytes and GCs from primordial to mature follicles in porcine except that GDF-9 was undetectable in GCs of mature follicles. For human ovary, GDF-9 presented in oocytes of primordial follicles in all samples, whereas BMP-15 was only observed in primordial follicle of adult ovary. Receptors, BMPR2, TβR1 and BMPR1B were found in oocytes and GCs of all follicles in mouse and porcine. In human, they were stained in oocytes from primordial follices but BMPR1B was not expressed in pubertal primordial follicles. Furthermore, we found that GDF-9, BMP-15 and three receptors distributed in adult corpus lutea. Collectively, our studies suggested that GDF-9, BMP-15 and their receptors might correlate with primordial follicular recruitment in pig and human. Positive expression of the receptors (BMPR2, TβR1 and BMPR1B)in primordial follicles of mouse ovaries indicated that these receptors might interact with others ligands besides GDF-9 and BMP-15 to regulate primordial follicular activity in mouse. Moreover, presence of GDF-9 in oocytes and BMP-15 in oocytes and GCs of mature follicles from mice and porcine elucidated coordinated roles of GDF-9 and BMP-15 in cumulus oophorus expansion. Additionally, expression of these factors in adult human corpus lutea suggested they play roles in corpus luteum activity.  相似文献
