Toxic aspergilli from pistachio nuts   总被引:2,自引:0,他引:2  
Pistachio nut samples taken during various stages of development from orchards in Iran, showed that contamination with fungi occurred mainly during the later stages of nut development. Members of the genera Aspergillus and Penicillium occurred most frequently. Of the Aspergilli, the species A. niger, A. flavus and A. fischeri var. spinosus occurred most frequently, followed by A. terreus, A. tamarii and A. nidulans. Twenty-two isolates comprising 13 species were tested for toxicity to ducklings. Isolates of known toxic fungi included A. flavus, A. niger, A. parasiticus, A. ochraceus, A. versicolor, A. nidulans and A. terreus. The toxicity of A. fischeri var. spinosus is reported. Chemical analysis showed that all isolates of A. flavus and A. parasiticus produced aflatoxin B1, the isolates of A. versicolor and A. nidulans produced sterigmatocystin while the toxic isolate of A. ochraceus did not produce ochratoxins.Toxic fungi have been shown to occur in a variety of nuts (4), (5), (11), (12), (13), (15), (18), (20), (21). Aflatoxin contamination of pistachio nuts has been reported and has in the past led to the rejection of consignments of Iranian pistachio nuts (1). In Iran, pistachio nuts are produced mainly in the south eastern provinces (Kerman & Zahedan) and to a limited extent in the Northern part (Kazvin & Damghan). In 1975 it was estimated (7) that there were some 24 million pistachio trees in Iran, of which 60% were situated in Rafsanjan, Kerman (Table 1). Economic considerations as well as the potential health hazard posed by aflatoxin-contaminated nuts, prompted the University of Isfahan to initiate a study of various aspects of the mycotoxin problem in pistachio nuts.  相似文献

A successful micrografting technique was developed for Pistacia vera. High levels of graft union were achieved when shoots from Stage II cultures of four-year-old P. vera. cv. Mateur were grafted onto in vitro-raised seedling rootstocks. Proteolytic activity in the digestive system of the pistachio green stink bug, Brachynema germari, was investigated. The maximum total proteolytic activity in the midgut extract was observed at pH 5, suggesting the presence of cysteine proteases. Hydrolyzing the specific substrates for cysteine proteases revealed the presence of cathepsin B and cathepsin L activities in the midgut extract. The presence of cysteine proteases was confirmed by their noticeable inhibition and activation due to specific inhibitors and activators, respectively. The significant inhibition of chymotryptic activity by the inhibitors showed the presence of chymotrypsin in the midgut. No considerable tryptic activity was observed in the midgut extract. There was no detectable total proteolytic activity in the salivary gland extract. Tryptic activity of the salivary gland extract was also inhibited by the specific inhibitors. The substrates for cysteine proteases were also slightly hydrolyzed by the salivary gland extract. Zymogram analysis showed at least one distinct band due to cysteine protease activity in the midgut extract, and the cysteine protease inhibitor caused almost complete disappearance of the band. Cathepsin B and L activities were mainly detected in midgut divisions m1 and m3, respectively, and maximum chymotrypsin and trypsin activities were observed in m3. In general, the results revealed the significant presence of cathepsin B, cathepsin L, and chymotrypsin proteases in the midgut extract. The major proteolytic activity in the salivary glands seems to be conducted by trypsin-like proteases.  相似文献

The parasitoid complex of the pistachio twig borer moth, Kermania pistaciella Amsel (Lepidoptera: Tineidae), a native pest of pistachio trees, was investigated at 27 pistachio plantation sites in Kerman province, the major pistachio growing area of Iran. The present study was conducted to document the naturally established parasitoid complex and to assess the need for improving the biological control of this species. In total, 22,390 moth cocoons were collected from 186 samples collected from commercial orchards during 2006–2008 and kept singly in controlled conditions to rear immature insects. An average of 2.8% of moth cocoons had been attacked by predators at time of sampling. Of the collected cocoons, on average 46.7% completed development and emerged as adult moths, no insects emerged from 8%, suggesting that the moth or wasp died before maturing, and parasitoids emerged from the remaining 42.5%. The overall percentage of host cocoons from which wasps emerged ranged from 25.6 to 59%. Fifteen hymenopterous parasitoid species were recovered from cocoons, of which three species were primary parasitoids, two were obligatory hyperparasitoids and the remaining 10 species were facultative hyperparasitoids. The primary parasitoid, Chelonus kermakiae (Tobias) (Hymenoptera: Braconidae), was the most abundant comprising almost 85% of the total emerging parasitoids. In addition, a further four species of larval parasitoid developed within the PTBM's larval tunnels in pistachio fruit cluster-stem tissue. Conservation of these parasitoids in the pistachio growing areas is recommended since a high level of parasitized moths’ cocoons was found at the majority of experimental sites.  相似文献

Elasmus nudus (Nees) is known as a synovigenic, gregarious ectoparasitoid of the pistachio fruit hull borer moth, Arimania komaroffi Ragonot in pistachio plantations in southern Iran. The biological parameters of this parasitoid were studied using the Mediterranean flour moth, Ephestia kuehniella Zeller as a host and the resulting data were compared with that of its native host, A. komaroffi. The parasitoid developed successfully from egg to adult over a temperature range of 17–35°C. The developmental times of the parasitoid on A. komaroffi and E. kuehniella were not significantly different. The threshold value at which the development rate of E. nudus was zero was estimated to be 9.2°C and the estimated number of degree days (°D) for development from egg to adult was 238 above this threshold. The reproductive potential of E. nudus was similar on both hosts at 30°C. The intrinsic rate of natural increase (r m ) for E. nudus did not vary much between 25°C and 32.5°C. In addition, the clutch size of the parasitoid on the two hosts was not statistically different. The present study provides some basic information on the biological characteristics of E. nudus.  相似文献

The effects of shoot girdling on stomatal conductance (g s), leaf photosynthesis (P N), concentrations of carbohydrates, nitrogen and chlorophyll (Chl) in leaves, areal leaf mass (ALM), the diameter and length of shoots, and bud abscission in pistachio were investigated. Girdling individual shoots at the base of the current year’s shoot (girdle I), separating inflorescent buds on the terminal current year’s shoot from the developing fruits on the previous year’s shoot, reduced inflorescent bud abscission by 70% in comparison to nongirdled controls. Girdle I significantly reduced concentrations of nitrogen in leaves but increased those of nonstructural carbohydrates particularly of starch. Shoot diameter increased by 13.1% and 26.4% at 33 and 81 days after girdling (DAG), respectively, compared to 1% and 3.4% in the control, respectively. Both the leaf dry mass/fresh mass ratio and ALM were increased significantly by girdle I from 12 DAG. The concentrations of Chl a, Chl b, Chl (a+b), as well as the ratio of Chl a/b, all decreased with girdle I. The greatest negative effect of girdle I was on g s and P N. P N was reduced by 55% of its initial value and was 44% less than in the control leaves at 10 DAG, and fell to approximately 30% that of the control from 21 DAG. In contrast, girdling at the base of one-year-old shoots (girdle II), thus not separating fruits from the inflorescent buds, did not significantly affect g s or P N. The effect of girdling on P N and the possible factors that are involved in the reduction of photosynthesis in pistachio are discussed.  相似文献

The parasitoids associated with the common pistachio psylla, Agonoscena pistaciae Burckhardt and Lauterer, were investigated at three pistachio plantations in Rafsanjan, Iran. Of the 6504 wasps emerging from mummified psyllids, 46% were the primary parasitoid Psyllaephagus pistaciae Ferrière, and the remaining 54% represented six species of hymenopterous hyperparasitoids, including Chartocerus kurdjumovi (Nikol’skaja), Marietta picta (André), Pachyneuron aphidis (Bouché), Pachyneuron muscarum (Linnaeus), Psyllaphycus diaphorinae (Hayat), and Syrphophagus aphidivorus (Mayr). Lysiphlebus fabarum Marshall, the parasitoid of Aphis gossypii Glover and Aphis craccivora Koch present on weeds, was found to be an alternative host for three major hyperparasitoids of A. pistaciae. The most abundant hyperparasitoid was S. aphidivorus, appearing during the growing season in all trial locations on psyllids and aphids in pistachio orchards. The weed-infesting aphids, along with their primary parasitoid, can act as a reservoir of A. pistaciae secondary parasitoids. Therefore, parasitized aphids allow populations of secondary parasitoids to increase and consequently to apply higher pressure on P. pistaciae. We detected that two primary parasitoid species, including P. pistaciae and L. fabarum, attacking different species of hosts interact indirectly through shared secondary parasitism. It is suggested that the community structure of A. pistaciae may be influenced by apparent competition, although more work is needed to provide firm evidence.  相似文献

The agronomic profitability of nut production in pistachio (Pistacia vera L.) is hampered by several factors such as the production of empty nuts, the small size and dry weight of the edible kernel and the low percentage of endocarp dehiscence. The causes underlining these problems are still unknown. The effect of pollinations with pollen from P. atlantica and six different P. vera genotypes on several fruit, nut and embryo characters was compared to study possible xenic and/or metaxenic effects. The use of P. atlantica pollen affected some traits, but there were no clear differences when the pollen of the different P. vera genotypes was used suggesting that, at the intraspecific level, the main limitation to pistachio fruit production problems might not be due to pollen factors.  相似文献

Abstract:  Two distinct seasonal forms were found for the common pistachio psylla, Agonoscena pistaciae , the most serious insect pest of cultivated pistachio trees, Pistacia vera in Iran. The two forms are different morphologically as well as biologically. The summer form is of light colour and readily mates and starts egg laying 1–2 days after emergence, whereas, the winter form is larger, generally darker and in reproductive diapause. The first form is observed under long photoperiod, while the second form is induced under a combination of short photoperiod and low temperature. The combination of long photoperiod and warm temperature enhanced post-emergence ovarian development in the winter form, showing a close inverse correlation between pre-oviposition period and increasing temperatures. Both forms of A. pistaciae are adapted to a wide temperature range of 20–30°C for reproduction, and the fecundity of psyllid adults of either summer- or winter forms was found to be very high under experimental conditions.  相似文献

The common pistachio psylla, Agonoscena pistaciae (Hemiptera: Aphalaridae) is one of the most detrimental pests to pistachio trees. Nowadays various pesticides are used to control the common pistachio psylla, but extensive pesticide spraying against this pest over a period of several years has overpowered its natural enemies. The plant’s byproducts come as an alternative ecologically more compatible in substitution to the synthetic insecticides. This work aims to evaluate the insecticidal potential of herbal extracts of fruit peels of Citrus sinensis and Citrus reticulata, seeds of C. sinensis and Rubia tinctorum, and leaves of Lawsonia inermis against common pistachio psylla. Five concentration 40, 47, 56, 67 and 80 µl/ml, of these plant extracts were sprayed on the fifth instar nymphs of common pistachio psylla by Potter Spray Tower under laboratory conditions (26?±?2?°C, 60–70% RH and a photoperiod of 16L: 8D h). Results showed significant differences among the treatments of the plant extracts and between the concentrations. The LC50 values of C. sinensis peels, C. reticulata peels, C. sinensis seeds, R. tinctorum seeds and L. inermis leaves were 62.04, 38.84, 43.60, 59.01 and 33.99 µl/ml, respectively. Also, all extracts showed the greatest mortality effect in 80 µl/ml concentration. According to these results, these plant extracts may use as herbal pesticides chemical control programme of common pistachio psylla.  相似文献

As genetic erosion of pistachio (Pistacia vera L.) has been occurring in the Mediterranean, Central and West Asia and North Africa, experiments were conducted to conserve two cultivars (‘Atl?’ and ‘Siirt’) of mature pistachio germplasm by assessing both medium- and long-term conservation techniques. In medium-term conservation, our results showed that it was feasible to conserve both cultivars in the form of either microshoots or encapsulated shoot apices up to 12 months at 4°C in the dark. As regards long-term conservation, encapsulation-dehydration and droplet-vitrification techniques were assessed for cryopreservation of cold-hardened and osmoprotected shoot apices of mature ‘Atl?’ cultivar. Among the methods tested, 13.6% of regrowth was achieved with incubation of explants in the droplets of vitrification solution for 150 min at 0°C followed by direct immersion in liquid nitrogen (LN), rapidly thawed and then cultured on Murashige and Skoog’s (MS) medium containing 1 mg L?1 BA and 0.5 mg L?1 GA3. The developed droplet-vitrification technique appeared as a promising procedure for long-term preservation of shoot apices of mature pistachio germplasm. Moreover, assesment of genetic fidelity by Random Amplified Polymorphic DNA analysis (RAPD) revealed out high levels of genetic stability between donor plant and cryopreserved plants (similarity indexes between 0.959 and 0.973) after they were subcultured for at least 3 months. The detected low level of genetic instability could be due to the toxic effect of PVS2 and regeneration phase. The optimized conservation techniques, especially slow growth storage, could be applied to preserve other Pistacia species.  相似文献

In the present study, effects of salinity stress were evaluated in the leaves and roots of two pistachio cultivars (Badami-Rize-Zarand (BZ) and Badami-e-Sefid (BS)). In overall, salinity negatively affects growth of both cultivars with more pronounced effects on BS. The physiological reason of the reduction could be attributed to some extent to more depletion of photosynthetic pigment in BS. In both cultivars, salinity increased proline content. Moderate and high salinities increased the soluble sugar contents in BZ. In both cultivars, Na+ content increased in plant organs with increasing Na+ in the media. Salinity treatment decreased the Fe and Pi contents in BS cultivar, while they remained unchanged in BZ. These results show that BZ cultivar exhibits more tolerance to salinity stress than BS cultivar possibly by better growth performance, accumulating more osmolytes, lesser accumulation of toxic sodium ion and lower Na+/K+ in the shoots as well as maintaining nutrient contents.  相似文献

Abstract

Pistachio (Pistacia vera L.) is a salt tolerant species that is considered an alternative crop for cultivation in salinzied orchard soils. In this work, 12-week-old pistachio seedlings cultivated in soil under greenhouse conditions were treated with five levels of salinity including control (0.63 dSm?1), low (2 and 4 dSm?1) and high (8 and 10 dSm?1) salt concentrations for further 12 weeks. Plant growth parameters were not affected by mild salinity; a significant reduction was only observed from 8 dSm?1. Considerable differences were observed between the young and mature leaves regarding osmotic and ionic stress effects of salt. Main compatible solutes were proline in mature leaves, proline and soluble sugars in young leaves, and soluble sugars and amino acids, other than proline, in roots. Concentration and content of Na in the leaves were not significantly increased at low levels of salinity and the K:Na and Ca:Na ratio of leaves were affected only by higher salt concentrations. Using the sequential extraction procedure for cell wall isolation, we observed that both absolute and relative amounts of Na in the cell wall fraction increased under low salinity, while decreased under higher levels of salt supply. Stable water relations, photochemistry and CO2 assimilation rates particularly of young leaves, as well as ion homeostasis were mechanisms for maintenance of plants growth under mild salinity. Under severe saline conditions, the impaired ability of mature leaves for synthesis of assimilates, preferent allocation of carbohydrates to roots for maintenance of osmotic homeostasis and finally, reduction of protein synthesis caused growth inhibition in pistachio.  相似文献

A series of laboratory and field studies were conducted using two lines of navel orangeworm, reared on different stages of new crop and mummy pistachios, Pistacia vera L. This study demonstrated the potential importance of malformed pistachios (pea splits) to the population dynamics of navel orangeworm, because these nuts, which are available as early as two months before mature nuts, supported navel orangeworm development and survival. Overall, the developmental rate on new crop pistachios is fastest on mature nuts, 422.3 +/- 123 degree-days (DD, degrees C), but other factors such as exposure to insecticide residue also sped development, although survival decreased. Development took the longest on unharvested nuts (mummies) dried at 90 degrees C for 24 h, 2664.7 +/- 131.4 DD. In most trials development was variable and two generations could develop at the fastest rate before the slowest individual completed development, which in turn calls into question the concept of discrete generations. Generally, survival was highest on mature pistachios and other stages of new crop nut and lowest on mummies collected in May. Survival was also higher on the new varieties 'Lost Hills' and 'Golden Hills' (24.7 and 32.0%, respectively) than on the most extensively planted variety 'Kerman' (13.3%). In our trials, both the rate of development and survival were dependent on nut stage, age, variety, and quality, indicating that pistachios, like almonds, Prunus dulcis (Mill.) D. A. Webb, are a dynamic rather than a static nutrient source for navel orangeworm.  相似文献

The rodents are a series of mammalians that have a high regeneration power. Their population increases rapidly. These animals comprise important agricultural pests, especially in pistachio orchards. In recent years, there is an increase in rodent population in pistachio orchards, which leads to enormous damage to these orchards. In this study, the effects of different poisons (phostoxin, simag, zinc phosphide and klerat) in rat control were compared. Based on the tests conducted, it was concluded that the poisons simag and zinc phosphide were the best. The suitability of Agonoscena pistaciae Burckhardt & Lauterer as a prey for Adalia bipunctata (Linnaeus) was evaluated by studying the predator’s development and reproduction on this species in comparison to reference diets of nymphs of Myzus persicae (Sulzer) and eggs of Ephestia kuehniella Zeller. Both larval and adult A. bipunctata fed voraciously on A. pistaciae. The development time of the ladybird was shorter when fed on M. persicae and A. pistaciae and longer on E. kuehniella. The survival rate was 87% on M. persicae and 85% on A. pistaciae, both significantly greater than on E. kuehniella eggs (63%). The ladybirds were heaviest at eclosion and their fecundity was higher on M. persicae than on A. pistaciae or E. kuehniella. Both the aphid and psyllid diets yielded higher R0, rm values and lower T and DT values than E. kuehniella eggs. Based on total development time, immature survival and adult fecundity, the relative suitability for A. bipunctata was M. persicae > A. pistaciae > E. kuehniella. When ladybirds were reared on psyllids, they showed no preference between aphid or psyllid diets, but when raised on M. persicae, they preferred an aphid diet. A field experiment showed that A. bipunctata preferred to oviposit on pistachio trees at a height of 150–200 cm, i.e. towards the top. The results indicate that A. pistaciae constitutes a complete food for A. bipunctata, supporting both completed development and successful reproduction.  相似文献
