The spatial and temporal variability of bacterial communities were determined for the nearshore waters of Lake Michigan, an oligotrophic freshwater inland sea. A freshwater estuary and nearshore sites were compared six times during 2006 using denaturing gradient gel electrophoresis (DGGE). Bacterial composition clustered by individual site and date rather than by depth. Seven 16S rRNA gene clone libraries were constructed, yielding 2717 bacterial sequences. Spatial variability was detected among the DGGE banding patterns and supported by clone library composition. The clone libraries from deep waters and the estuary environment revealed highest overall bacterial diversity. Betaproteobacteria sequence types were the most dominant taxa, comprising 40.2–67.7% of the clone libraries. BAL 47 was the most abundant freshwater cluster of Betaproteobacteria , indicating widespread distribution of this cluster in the nearshore waters of Lake Michigan. Incertae sedis 5 and Oxalobacteraceae sequence types were prevalent in each clone library, displaying more diversity than previously described in other freshwater environments. Among the Oxalobacteraceae sequences, a globally distributed freshwater cluster was determined. The nearshore waters of Lake Michigan are a dynamic environment that experience forces similar to the coastal ocean environment and share common bacterial diversity with other freshwater habitats.  相似文献

Dietary sodium intake for freshwater salmonids feeding in the wild (invertebrate diet) or in captivity (pellet diet) was calculated and compared with published branchial sodium influx values. Dietary sodium intake (mmol kg−1 per month) increases from winter minimum values of 5 and 30-40 to reach maximum values in summer of 175 and 240 for invertebrate and pellet diet, respectively. In summer, dietary sodium intake for fish feeding in the wild was of the same magnitude as branchial sodium influx. The implications of dietary sodium intake for sodium balance in freshwater fish are discussed.  相似文献

Flavobacterium psychrophilum (F. psychrophilum) is the causative agent of bacterial cold-water disease (BCWD) that occurs in ayu Plecoglossus altivelis. Formalin-killed cell of F. psychrophilum has long been studied as an immersion vaccine for BCWD. In this study, we explored the possibility of F. psychrophilum collagenase (fpcol) for use as the immersion vaccine. BCWD convalescent ayu sera contained specific IgM antibodies against somatic F. psychrophilum and fpcol, meaning that fpcol is a promising antigen for the vaccine development. The recombinant fpcol was successfully expressed in Escherichia coli and Brevibacillus chosinensis (B. chosinensis). The culture supernatant of the B. chosinensis was used as an immersion vaccine solution. The vaccinated ayu were then challenged by soaking into F. psychrophilum culture. In two experimental groups, the relative percentages of survivals were 63 and 38%, respectively, suggesting that fpcol is promising as the immersion vaccine for ayu-BCWD.  相似文献

The phylogenetic composition of a bacterial community from a hypertrophic freshwater lake in China was investigated by sequencing cloned 16S rRNA genes. Three hundred and thirty-six bacterial clones from four clone libraries in different months (March, May, July and September in 2004) were classified into 142 operational taxonomic units, most of which were affiliated with bacterial divisions commonly found in freshwater ecosystem, e.g. Alpha-, Beta-, Gamma- and Deltaproteobacteria, Bacteriodetes and Actinobacteria. The results showed that the composition of bacterial community in the July library was the most diverse one. Actinobacteria was the most significant lineage in Lake Taihu, with dominant numbers of operational taxonomic units in the May, July and September libraries. Phylogenetic analysis suggested that 53 sequences were grouped into six novel clusters which may represent specific populations indigenous to the environment. Coverage analyses indicated that the clone libraries could provide a fine inventory of bacterial diversity in the lake.  相似文献

Previous studies have reported wide distribution of anaerobic ammonia oxidation (anammox) bacteria in various ecosystems. However, little is known about the distribution of anammox bacteria under varying environmental conditions in intensive aquaculture systems. In Yangcheng Lake, a famous crab farm situated in the Yangtze River Delta, sediment samples were collected in October (feeding period) and January (nonfeeding period) to analyze the distribution and diversity of anammox bacteria and their relationships with environmental factors. Based on the functional biomarker of Anammox bacteria, hzo gene, anammox bacterial clone libraries were constructed and their abundances were determined by quantitative PCR (qPCR). The Anammox bacteria were detected in the lake with the abundances ranging from 0.70 × 105 to 6.05 × 105 copies per gram of sediment. Sequences from eight clone libraries yielded seven unique operational taxonomic units (OTUs), distantly related to the Candidatus Jettenia genera with a similarity of about 91%. The Anammox bacterial community structures, diversities and abundances varied spatiotemporally with environmental conditions. In October, the level of the nitrogen compounds, the diversity, evenness and abundance of Anammox bacteria were higher than in January. The predominant OTU of samples changed from HZO-OTU-1 (34.25%) in January to HZO-OTU-2 (28.90%) in October. Moreover, the site (SW) nearing to sewage inlet was lack of HZO-OTU-7 in January. Canonical correspondence analysis (CCA) showed that the pore water NO2? concentration, ammonium to nitrogen oxides ratio (NH4+/NOx?) and total organic carbon to total nitrogen ratio (TOC/TN) contributed most to Anammox bacterial community structures variances. Pearson correlations analysis revealed that the Anammox bacteria abundance had positive co-relationships with TN, NH4+, NO3? concentrations, and negative correlation with TOC/TN in porewater.  相似文献

The Cape Horn Biosphere Reserve, one of the last wild areas of the planet, is not exempt from the pressures of global change, such as non-native species introductions. During 2018 and 2019 we studied the Róbalo river basin in order to update the diversity and distribution of fishes. Here, we report for the first time the native and endangered “Peladillas” Aplochiton taeniatus and the non-native coho salmon Oncorhynchus kisutch. The coexistence of native and non-native fishes poses a challenge for the management and conservation of aquatic biota from the Cape Horn Biosphere Reserve.  相似文献

The stress response of Oncorhynchus mykiss in high‐altitude farms in central Mexico was investigated over two seasons: the cool (9·1–13·7° C) dry winter season, and the warmer (14·7–15·9° C), wetter summer season. Fish were subjected to an acute stress test followed by sampling of six physiological variables: blood cortisol, glucose, lactate, total antioxidant capacity, haemoglobin concentration and per cent packed cell volume (VPC%). Multivariate analyses revealed that lactate and total antioxidant capacity were significantly higher in the summer, when water temperatures were warmer and moderate hypoxia (4·9–5·3 mg l?1) prevailed. In contrast, plasma cortisol was significantly higher in the winter (mean ± s.e .: 76·7 ± 4·0 ng ml?1) when temperatures were cooler and dissolved oxygen levels higher (6·05–7·9 mg l?1), than in the summer (22·7 ± 3·8 ng ml?1). Haemoglobin concentrations (mg dl?1) were not significantly different between seasons, but VPC% was significantly higher in the summer (50%) than in the winter (35%). These results suggest that in summer, effects of high altitude on farmed fish are exacerbated by stresses of high temperatures and hypoxia, resulting in higher blood lactate, increased total antioxidant capacity and elevated VPC% levels.  相似文献

【目的】 黄河入海口地处河流-陆地-海洋的交汇地带,是淡水水体与海水水体相互作用的混合区域,也是多样化和生产力较强的河口生态系统。本研究以黄河入海口为研究对象,探究其淡水与海水水体细菌群落特征。【方法】 采用高通量绝对定量技术,获得细菌群落绝对丰度信息,在此基础上比较分析2种水体细菌群落优势物种组成、α和β多样性、网络共现模式、构建机制及潜在功能,并探究优势物种与水体环境因子的相关性。【结果】 淡水水体细菌的绝对拷贝数为2.61×106 copies/mL,是海水的1.8倍。2种水体共同优势菌门为放线菌门、假单胞菌门、蓝细菌门和拟杆菌门等,其各自绝对丰度有明显差异。淡水中放线菌门数量最高,约等于海水中所有优势菌门数量之和,而海水中假单胞菌门数量最高。淡水水体细菌群落α多样性高于海水,2种水体细菌群落结构存在较明显的差异,主要源于各自优势物种丰度的差异。淡水水体细菌共现网络较海水更复杂和稳定,随机性过程主导2种水体细菌群落构建机制。2种水体细菌群落功能结构存在差异,但拥有共性功能。新陈代谢是2种水体细菌群落丰度最高的功能,其在淡水中的相对丰度显著高于海水。5种环境因子[pH、氧化还原电位(oxidation-reduction potential, ORP)、电导率(electrical conductivity, EC)、总有机碳(total organic carbon, TOC)和总氮(total nitrogen, TN)]与水体优势物种分别具有不同程度的相关性。除EC外的4种环境因子间均存在共线性关系,与pH、TOC和TN呈正相关关系的优势菌属均与ORP呈负相关,反之亦然。放线菌门和假单胞菌门分别与pH呈正相关和负相关关系。【结论】 黄河入海口淡水和海水水体细菌群落特征存在较大差异,主要体现在细菌数量、多样性、功能结构和共现网络上,但2种水体具有相似的优势物种组成和群落构建机制。本研究结果可为黄河入海口水体微生物生态学研究及开发利用微生物资源提供数据支持。  相似文献

[目的]研究连作条件下番茄青枯病不同发病时期的非根际土壤微生物群落差异,明确土壤微生物对青枯病发病时期的响应机制。[方法]本研究对16S rRNA V4-V5区进行实时荧光定量PCR和高通量测序,综合分析了连续种植第1、3、5和7季的发病高峰期和发病末期的番茄非根际土壤的病原菌数量、细菌群落多样性、群落组成、标志细菌类群和群落构建机制。[结果]发病高峰期的青枯菌数量(1.28×107 copies/g)要高于发病末期(1.77×106 copies/g)。随着连作时间的增加,发病时期对细菌群落多样性的影响逐渐增加。第3季和第5季不同发病时期之间的细菌群落alpha和beta多样性存在显著差异。LEfSe分析发现,番茄青枯病发病高峰期和发病末期的非根际土壤样品有其各自不同的标志细菌类群。此外,随连作时间的延长协助青枯菌致病的细菌逐渐累积。发病高峰期是微杆菌属(Microbacterium)和亚硝化螺菌属(Nitrosospira)协助青枯菌致病,而在发病末期,则由鞘脂菌属(Sphingobium)、norankf...  相似文献

The study examined the density‐mediated effects on growth, survival and dispersal of wild and farmed Atlantic salmon Salmo salar offspring in the period immediately following emergence, using a substitutive design. In small confined stream channels, wild parr coexisting with farmed parr had a significantly poorer survival, than wild parr alone. Density did not affect this relationship. In larger unconfined stream channels, wild parr coexisting with farmed parr entered a downstream trap in higher numbers than wild parr in allopatry. The results suggests that during the earliest life stages, farmed S. salar can outcompete wild S. salar, resulting in a reduced survival of wild S. salar.  相似文献

In recent decades, the decline of coastal water quality has promoted the birth of a new industrialized aquaculture mode in China, which involves the cultivation of organisms using underground seawater extracted from various depths below the intertidal zone. In view of the special physicochemical characteristics of underground seawater, the microbial community in this environment has attracted interest. In this study, the microbial community in the underground seawater of an intertidal area of the Qingdao coast of China was investigated. Compared with the upper coastal water, the underground seawater displayed lower numbers of microorganisms (2.7?±?0.3?×?105 cells mL?1 in underground seawater vs. 5.3?±?0.4?×?105 cells mL?1 in upper coastal seawater) but displayed much higher microbial diversity. At the phyla level, Proteobacteria, Bacteroidetes, Cyanobacteria, and Actinobacteria inhabited both environments, whereas bacteria in the phyla Planctomycetes, Deferribacteres, and Nitrospirae were recovered only from the underground seawater. Eighty-nine percent of the OTUs in the underground seawater were environmental specific. Furthermore, compared with coastal water, underground seawater displayed significant lower (p?<?0.05) concentration of NH3-N, NO2-N, PO4-P, and DOC-C, and contained fewer potentially harmful pathogens (e.g., Verrucomicrobia/Opitutae) and more denitrifying bacteria (e.g., Shewanella denitrificans), thus making it more suitable for aquaculture.  相似文献

Differences in sperm metabolism and morphology between wild and non‐local farmed Atlantic salmon Salmo salar were assessed by measuring metabolic enzyme activities and length of sperm flagella. No differences were observed between wild and farmed S. salar sperm with regards to cell counts or any of the biochemical variables assessed. Flagella of sperm cells were significantly longer in wild than farmed S. salar; however, this did not result in higher energy levels or different fertilization rates.  相似文献

Understanding the genetic basis of variation in traits related to growth and fillet quality in Atlantic salmon is of importance to the aquaculture industry. Several growth‐related QTL have been identified via the application of genetic markers. The IGF1 gene is considered a highly conserved and crucial growth‐regulating gene in salmonid species. However, the association between polymorphisms in the IGF1 gene and growth‐related traits in Atlantic salmon is unknown. Therefore, in this study, regions of the Atlantic salmon IGF1 gene were sequenced, aligned and compared across individuals. Three SNPs were identified in the putative promoter (SNP1, g.5763G>T; GenBank no. AGKD01012745 ), intron 1 (SNP2, g.7292C>T; GenBank no. AGKD01012745 ) and intron 3 (SNP3, g.4671A>C; GenBank no. AGKD01133398 ) regions respectively. These SNPs were genotyped in a population of 4800 commercial Atlantic salmon with data on several weight and fillet traits measured at harvest (at approximately 3 years of age). In a mixed model, association analysis of individual SNPs, SNP1 and SNP3 were both significantly associated with several weight traits (< 0.05). The estimated additive effect on overall harvest weight was approximately 35 and 110 g for SNPs 1 and 3 respectively. A haplotype analysis confirmed the association between genetic variation in the IGF1 gene with overall body weight (< 0.05) and fillet component traits (< 0.05). Our findings suggest the identified nucleotide polymorphisms of the IGF1 gene may either affect farmed Atlantic salmon growth directly or be in population‐wide linkage disequilibrium with causal variation, highlighting their possible utility as candidates for marker‐assisted selection in the aquaculture industry.  相似文献

We used two analyses to test the hypothesis that planktivore abundances contribute to the residual variations of Secchi depth or chlorophyll-a plotted with respect to mean summer epilimnetic total phosphorus. The first analysis involved 15 lake years of data from six lakes. The data set comprised mark-recapture assessments of piscivore and planktivore numbers and estimates of mean summer chlorophyll-a, total phosphorus and Secchi depth. We found that residual chlorophyll-a variation was not significantly (p>0.05) correlated with planktivore densities, but that planktivore densities did contribute (p<0.02) to the residual variation of Secchi depth on mean total phosphorus. The second analysis included all of the data used in the first plus an additional 13 lake years of data from the literature. These data showed that the percentage of the total fish community comprising planktivores did not significantly (p>0.05) contribute to the residual variation in chlorophyll-a with respect to mean summer total phosphorus. Together, our results suggest that planktivore abundance has a significant cascading impact on water clarity, but no long term statistically significant impact on mean summer chlorophyll-a concentration.  相似文献

Massive mortality of planktivorous fish had a dramatic impact on plankton community dynamics of Lake Mendota, Wisconsin, USA. After fish mortality, the largerDaphnia pulicaria replaced the smallerDaphnia galeata mendotae, resulting in greater grazing pressure on phytoplankton. This was accompanied by a much longer spring clear-water period and lower summer phytoplankton biomass compared to years before the fish mortality. Analysis of historical data (from the mid-1970's) showed that previous fluctuations in planktivorous abundance had similar effects onDaphnia abundance and species composition, and on spring phytoplankton biomass. However, the mid-1970's fish fluctuations had no detectable effect on summer phytoplankton. Concentrations of phosphorus were much higher in the 1970's (spring P 80–135 μg 1−1) than in the 1980's (spring P 19–36 μg 1−1) and it is possible that high P concentrations may reduce trophic cascade effects on summer phytoplankton communities. This suggests that the success of biomanipulation programs may be dependent on lake nutrient status.  相似文献

【背景】碳汇是指从大气中消除二氧化碳的过程、活动或机制,我国最先提出碳汇渔业概念。【方法】捕捞鱼类的碳均来自天然饵料,故以其平均碳含量估算碳移出量。而养殖鱼类中,一般假定不考虑施肥养鱼的碳输入;鲢和鳙是滤食性鱼类,主要摄食浮游生物,鳜属鱼类以其他种鱼类为食物,而这些鱼类主要摄食天然饵料,故可以认为其碳均来自天然饵料。此外,假设草鱼、鲫和鲤等产量的20%来自天然饵料,而河蟹产量的50%来自天然饵料。基于渔业统计年鉴(2011—2015年),估算了我国近5年来淡水渔业碳汇强度。【结果】2010—2014年,全国淡水养殖碳移出量逐年稳步增长,分别为136.2万、140.5万、146.0万、153.0万和164.5万t,平均每年的碳移出量为148.0万t。2010—2014年全国淡水捕捞碳移出量分别为29.3万、28.7万、29.6万、29.7万和29.6万t,平均每年的碳移出量为29.4万t。【结论与意义】在自然资源日益减少的情况下,淡水养殖渔业碳汇的发展必然会成为淡水渔业经济发展的主体。  相似文献

Renibacterium salmoninarum (Rs) is common in wild Arctic charr Salvelinus alpinus and brown trout Salmo trutta in Iceland. Of 22 charr and nine trout populations none were free of Rs antigens. In two charr populations only one fish exceeded the Rs antigen detection limit and in one of these cases the ELISA value was within uncertainty limits of the infection criterion. Mean prevalence of infection was 46% for Arctic charr (range: 3–100%) and 35% for brown trout (range: 6–81%). No infected fish showed gross pathological signs of bacterial kidney disease (BKD). The ubiquity and high prevalences of infection indicated that the bacterium has been endemic for a long time, and is probably a normal, low density resident in the fish. A lack of correlation in mean intensity of Rs antigen and prevalence of infection between sympatricpopulations of Arctic charr and brown trout suggests that the dynamics of infection and internal proliferation of bacteria can be quite independent in the two species even if they live in the same lake. Rs intensity and its coefficient of variation decreased with age in older fish, suggesting a connection between Rs intensity and host mortality. However, this can be caused by other ecological factors that decrease survival, especially low food availability, which simultaneously increase the susceptibility to Rs infection and internal proliferation.  相似文献
