The male gonopores, male reproductive apparatus, spermatophore and spermatozoa of the Mediterranean hermit crab Paguristes eremita are described, using interference phase microscopy, scanning electron microscopy and transmission electron microscopy. A correlation is made between the gonopore morphology and the different kinds of setae accompanying them, and the reproductive biology of these crabs. Each testes merges into a tubular duct made up of four zones: (1) the collecting tubule with free spermatozoa; (2) the proximal zone, where the ampulla of the spermatophores starts to be formed; (3) the medial zone, where the ampulla is completed, the stalk lengthens and the pedestal is formed; (4) the distal zone, where the mature spermatophores are stored. The sizes of the different parts of the spermatophore and of the sperm are given and their exterior morphology and ultrastructure described and compared to congeners. The morphology of the gonopore, male reproductive system, spermatophore and spermatozoa of P. eremita are species-specific, clearly distinguishing the species from the other members of the family. The available spermatozoal and spermatophore data is used to place P. eremita within a sperm phylogeny of the hermit crab family Diogenidae.  相似文献

This study aims to analyze the functional anatomy of the male reproductive system in Neocaridina davidi, a very popular ornamental species of caridean shrimp. Mature males were cold‐anaesthetized and their reproductive systems were dissected for histological and histochemical analysis, while the spermatozoa and spermatophore wall ultrastructure were analyzed under transmission electron microscopy. The male reproductive system consisted of two coiled testes, which were continuous with the vasa deferentia. Testes were positioned on the dorsal side of the cephalothorax above the hepatopancreas, and comprised seminiferous tubules where spermatogenesis occurred. Each vas deferens (VD) was a long tube dorsolaterally positioned with respect to the hepatopancreas, and increased in diameter at the distal end. Three regions could be recognized in the VD: proximal, middle, and distal. The proximal region had a cylindrical epithelium with secretory cells. The middle region (or typhlosole) had a dorsal fold (or typhlosole) with a thick columnar epithelium and high secretory activity. The spermatophore was a continuous cord with three acellular layers, which were mainly characterized by the presence of neutral glycoconjugates and proteins. The sperm morphology was distinct from the inverted cup‐shaped spermatozoa observed in the majority of caridean shrimps. The spermatozoa in specimens of N. davidi were spherical in shape, with a cross‐striated, single, short spike, and arranged in clusters of three or four sperm cells. The composition of the spermatophore, and the arrangement and form of the spermatozoa, seem to be unique in comparison to other species of Caridea.  相似文献

Summary

Light and electron microscopy were used to analyze the morphology and ultrastructure of the spermatozoon of Fissurella nigra and Fissurella picta, two sympatric species of keyhole limpets from the southern Chilean coast. The spermatozoa of both species are of the aquasperm type, typical of species with external fertilization. Each species had its own distinctive spermatic morphology, particularly in relation to size. Proceeding from anterior to posterior, the spermatozoa of both species each consists of an elongated head with a conical acrosome having a deep subacrosomal space and truncated conical nucleus, followed by a midpiece containing five mitochondria associated in a compact ring around the proximal and distal centrioles, and, at the end, a flagellum. The spermatozoon head of F. nigra is almost twice as long as that of F. picta. This difference constitutes the first morphological evidence that the size of the spermatozoon could represent a candidate for the maintenance of reproductive isolation between these two sympatric species.  相似文献

The comparative ultrastructure of ejaculated boar, bull and ram spermatozoa is studied by scanning electron microscopy. After washing, the spermatozoa are fixed in glutaraldehyde or im picric acid-formaldehyde-glutaraldehyde mixture. Samples are prepared either by critical point drying (Freon) on Millipore filters or by air drying on glass cover slips. In all the species studied, three regions may be distinguished in the paddle-shaped head of the sperm: an anterior segment (surrounded by the marginal thickening) and an equatorial segment constituting together the acrosome, and the postacrosomal region. Most of the feature of the postacrosomal lamina described in transmission electron microscopy are visible through the plasma membrane, particularly after air drying. The surface morphology of the neck and of the different segments of the flagellum is also evident. Some species differences are encountered, e.g. rough surface of acrosome and absence of serrations in postacrosomal lamina of boar spermatozoa only. The techniques employed result in good general morphology and fine resolution of surface detail of the sperm samples; they also permit analysis of spermatozoa treated by freezing or submitted to acrosomal extraction.  相似文献

The families Aethridae and Calappidae were originally considered as part of the same family; however, their morphology and molecular biology separate them into two families. In this context, we describe the ultrastructure of spermatozoa of species of the Calappidae, Aethridae and Menippidae to elucidate the relationships among taxa. The vasa deferentia were submitted to routine protocols for transmission electron microscopy. Our results indicate that the morphology of the spermatozoa of Hepatus pudibundussupports its exclusion from the Superfamily Calappoidea due to the presence of the apical striated layer. The spermatozoa of Menippe nodifrons is very similar to H. pudibundus and corroborates the recent phylogenetic analysis using sequence data of nuclear genes. Moreover, our results evidence two morphological patterns of spermatozoa within Calappidae. Calappa ocellata and C. cinerea show spermatozoa with a wide acrosome vesicle, a thick operculum shaped as a shallow “W” and a large thickened ring. Calappa gallusand C. hepatica show spermatozoa with a longer acrosome vesicle, a pointed operculum and a slender thickened ring. Our ultrastructure results conform with previous molecular proposal and show that spermatozoa ultrastructure can be an effective tool to adjust phylogenetic relationship when used in association with molecular data.  相似文献

Studies of sterile mutants in Caenorhabditis elegans have uncovered new insights into fundamental aspects of gamete cell biology, development, and function at fertilization. The genome sequences of C. elegans, Caenorhabditis briggsae and Caenorhabditis remanei allow for informative comparative studies among these three species. Towards that end, we have examined wild-type sperm morphology and activation (spermiogenesis) in each. Light and electron microscopy studies reveal that general sperm morphology, organization, and ultrastructure are similar in all three species, and activation techniques developed for C. elegans were found to work well in both C. briggsae and C. remanei. Despite important differences in the reproductive mode between C. remanei and the other two species, most genes required for spermiogenesis are conserved in all three. Finally, we have also examined the subcellular distribution of sperm epitopes in C. briggsae and C. remanei that cross-react with anti-sera directed against C. elegans sperm proteins. The baseline data in this study will prove useful for the future analysis and interpretation of sperm gene function across nematode species.  相似文献

Pea crabs of the subfamily Pinnotherinae (Pinnotheridae) have a high investment in reproduction and an outstanding reproductive output, probably as an adaptation to the required increase in reproductive rate due to the pinnotherids small size and their parasitic, host‐dependant way of life. In the present study, we investigate the male internal reproductive structures and the ultrastructure of spermatozoa of Pinnotheres pisum and Nepinnotheres pinnotheres by histological methods and both scanning‐ and transmission electron microscopy. In the Brachyura, the male internal reproductive systems generally consist of paired testes and corresponding vasa deferentia where spermatozoa develop and mature. Spermatozoal ultrastructure of the investigated pinnotherids conforms to the thoracotreme type, however, N. pinnotheres has an accessory opercular ring and a periopercular rim, neither of which are present in spermatozoa of P. pisum. Spermatozoa are enclosed within spermatophores in the secretory proximal vas deferens. Two types of secretions were observed in P. pisum and N. pinnotheres: an electron dense substance secreted in the proximal vas deferens involved in spermatophore formation, and large electron‐luscent vesicles constituting the seminal plasma in the medial and distal vas deferens. The medial vas deferens is strongly widened compared to other brachyurans to purpose storing spermatophores embedded in seminal plasma. Tubular appendices, which produce and store large amounts of seminal plasma, arise from the distal region of the vas deferens. The appendices extend into the ventral cephalothorax and also in the first pleomere. The latter being an exceptional location for reproductive structures among male brachyurans. J. The need for information on the reproductive physiology of different wildlife species is important for ex situ conservation using such methods as in vitro fertilization (IVF). Information on species reproductive physiology and evaluation of sperm quality using accurate, objective, repeatable methods, such as computer-assisted sperm analysis (CASA) for ex situ conservation has become a priority. The aim of this study was to evaluate motility patterns of antelope epididymal spermatozoa incubated for 4 h under conditions that support bovine IVF using CASA. Cauda epididymal spermatozoa were collected postmortem from testicles of springbok (N = 38), impala (N = 26), and blesbok (N = 42), and cryopreserved in biladyl containing 7% glycerol. Spermatozoa were thawed and incubated in Capacitation media and modified Tyrode lactate (m-TL) IVF media using a protocol developed for domestic cattle IVF. The study evaluates 14 motility characteristics of the antelope epididymal sperm at six time points using CASA. Species differences in CASA parameters evaluated under similar conditions were observed. Several differences in individual motility parameters at the time points were reported for each species. Epididymal sperm of the different antelope species responded differently to capacitation agents exhibiting variations in hyperactivity. Motility parameters that describe the vigor of sperm decreased over time. Spermatozoa from the different antelope species have different physiological and optimal capacitation and in vitro culture requirements. The interspecies comparison of kinematic parameters of spermatozoa between the antelopes over several end points contributes to comparative sperm physiology which forms an important step in the development of species specific assisted reproductive techniques (ARTs) for ex situ conservation of these species.  相似文献

Spermatozoa from 16 species of primates, representing Families Cercopithecidae Cebidae, Pongidae, and Hominidae, were viewed using scanning electron microscopy (SEM). Various techniques of specimen preparation were employed, especially critical point drying using CO2, but also critical point drying using N2O as well as air drying using amyl acetate. Details of ultrastructure and identifying features of the various families are presented, with both relationships and differences being evident. Accompanying light micrographs of several of the species illustrate size relationships of the head, midpiece, and principal piece of the tail. Dimensions of these parts of spermatozoa were calculated from such photographs, and add to the interpretation of overall morphology. SEM is of considerable value in understanding many details of structure in spermatozoa, and should contribute to the study of taxonomic affinities among the various primate groups.  相似文献

 Spermatophore development and ultrastructure of the mature sperm of Craterostigmus tasmanianus were studied using light and electron microscopy. In C. tasmanianus, as in the Scolopendromorpha, the spermatophore develops within the vas deferens. The latter consists of three parts, each with a different morphology. The first may be involved in guiding the sperm to roll up into typical ring-like structures, while the other two, which show an evident secretory activity, secrete the acellular wall of the spermatophores. The ultrastructure of mature spermatozoa showed that a very close similarity exists between Craterostigmomorpha and Lithobiomorpha, especially regarding the organization of the connecting piece. Based on this similarity, we consider the Craterostigmomorpha together with the Scolopendromorpha, Geophilomorpha and Lithobiomorpha (=Pleurostigmophora) to be the sister group of the Scutigeromorpha. The spermatozoa ultrastructure of the pink shrimp Farfantepenaeus paulensis was investigated in this morphological study. Spermatophores and spermatozoa were analyzed by electron microscopy. The pink shrimp spermatophore is divided into two regions: the appendage and the spermatophore main body, where spermatozoa are grouped in a spermatic mass. Pink shrimp spermatozoa are unistellate and are composed of main body and single spike. The spermatozoa body comprises a perinuclear cytoplasmic band, nucleus, acrosomal cap, and subacrosomal region. The spermatozoa cell mean total length was 10.71 μm, the mean body diameter was 5.56 μm, and the mean spike length and diameter were 5.15 μm and 0.85 μm, respectively.  相似文献

The predatory ladybird beetle Tenuisvalvae notata (Coleoptera: Coccinellidae) (Mulsant) is a polygamous species and its morphology, as well as the storage capacity of seminal fluid in the spermatheca, may affect its reproductive performance. Thus, the present study evaluates the spermatheca morphology of virgin and mated T. notata females using light and scanning microscopy. The results show that the spermatheca of T. notata is kidney shaped and consists only of the receptacle and spermathecal duct, being morphologically similar in virgin and mated females. There is no secretion in the spermathecae of virgin females and, in mated females, only once it was not possible to observe the presence of spermatozoa. By contrast, females mate multiple times it is possible to observe spermatozoa in the lumen of the spermatheca surrounding the secreted material. Polygamy in T. notata might be related to the maintenance of viable spermatozoa in the spermatheca, in which case the female would prefer to copulate more times during its adult life than to store spermatozoa for a longer period of time.  相似文献

In this transmission electron microscopy study, we describe the ultrastructure of the spermatozoon of Heterotis niloticus (Osteoglossiformes), which is distinguished by having two flagella. Our investigation also highlights the great diversity of sperm cell structures observed across osteoglossiform families, such as aflagellate (Gymnarchidae, Mormyridae), monoflagellate (Notopteridae, Pantodontidae) and biflagellate spermatozoa. As biflagellate spermatozoa are rare in vertebrates, we also summarize the orders and families known to possess this ultrastructural character, most of which are fishes.  相似文献

The aim of this study was to describe and illustrate the morphology of the spermatozoon of the Western Atlantic shrimp, Hippolyte obliquimanus. Individuals were sampled from Itaguá Beach (Ubatuba, southern Brazil). The male reproductive system was dissected and morphological analysis was undertaken using a stereomicroscope, a light microscope, and transmission electron and scanning electron microscopes. When viewed from the nuclear or acrosomal poles, each spermatozoon has many translucent radiating arms (about 20) from a denser cell body, while laterally the cell body and arms resemble a “cnidarian medusa”, with all the arms projecting away from the bell-like cell body. This sperm morphology is distinct from the “thumbtack”-shaped spermatozoa observed in the majority of carideans but has similarities to the spermatozoa of Rhynchocinetes spp. The morphology of sperm of several species of the genus Hippolyte resembles the spermatozoon of H. obliquimanus with the presence of posterior nuclear arms, but it is necessary to study other Hippolyte species to place these arms in the context of the genus.  相似文献

The morphology and function of the female reproductive organs in 6Protodrilus species are investigated by light- and transmission electron microscopy. Possible ways in which spermatozoa may enter the female coelom after leaving the spermatophore are discussed for species with and without special female reception organs. Only femaleP. rubropharyngeus andP. flavocapitatus have “dorsal organs” for spermatophore reception. The structure and function of these organs are described, as well as those of the oviduct found in 3 of the species investigated. The possible phylogenetic origin of gonoducts and different modes of oviposition within the genus are discussed. Finally, the high taxonomic significance of female traits such as dorsal organs, oviducts, cocoon glands and lateral ciliary rows in this genus is stressed.  相似文献

The morphology and ultrastructure of the olfactory sensilla on the antennae and maxillary palps were investigated through scanning electron microscopy (SEM) and transmission electron microscopy (TEM), and their responses to five volatile compounds were measured using electroantenogram (EAG) and electropalpogram (EPG) techniques in the pumpkin fruit fly, Bactrocera depressa (Shiraki; Diptera: Tephritidae). Male and female B. depressa displayed distinct morphological types of olfactory sensilla in the antennae and maxillary palps, with predominant populations of trichoid, basiconic, and coeloconic sensilla. Basiconic sensilla, the most abundant type of olfactory sensilla in the antennae, could be further classified into two different types. In contrast, the maxillary palps exhibited predominant populations of a single type of curved basiconic sensilla. High‐resolution SEM observation revealed the presence of multiple nanoscale wall‐pores on the cuticular surface of trichoid and basiconic sensilla, indicating that their primary function is olfactory. In contrast, coeloconic sensilla displayed several longitudinal grooves around the sensillum peg. The TEM observation of individual antennal olfactory sensilla indicates that the basiconic sensilla are thin‐walled, while the trichoid sensilla are thick‐walled. The profile of EAG responses of male B. depressa was different from their EPG response profile, indicating that the olfactory function of maxillary palps is different from that of antennae in this species. The structural and functional variation in the olfactory sensilla between antennae and maxillary palps suggests that each plays an independent role in the perception of olfactory signals in B. depressa.  相似文献

Previous studies established the existence of morphologically highly similar amphiatlantic populations of the predominantly interstitial genus Microphthatmus (European coastline - from the German Bay to the Bay of Biscaya; American coastline - from Maine to Massachusetts and at the coast of North Carolina). Originally these three populations were seen as belonging to a single species. By using a broad spectrum of different methodologies for character investigations (especially through the use of electron microscopy) three, distinct species can now be distinguished: M. listensis Westheide 1967, M. nahantensis n. sp. and M. carolinensis n. sp. While habitat structure and external morphology - generally important in polychaete systematics - were found to be rather similar between the three species, this study uncovered distinct differences between the species in their internal organization. Such sharp differences could even be followed down to the ultrastructure of single cells (e. g. copulatory stylets, position and number of ovaries, sperm and secretory granules of male accessory glands). Within population character variability is significantly lower in several features of M. carolinensis than in the same characters of the other two species. Similarities as well as differences have been noted in behavioural features and population dynamics between the species.  相似文献

How alternative reproductive tactics (ARTs) are maintained in wildlife populations is one of the major questions in evolutionary biology. As a dominant status, territoriality is typically linked to increased mating opportunities, and one explanation why this behaviour coexists with other tactics is that dominance implies survival costs. Such a trade-off may occur in the Northern chamois Rupicapra rupicapra, as reproductive advantages of territorial males over non-territorial males could be counterbalanced by a reduction in survival mediated through energy expenditure, stress and parasitic infections, ultimately favouring ART coexistence. Here, we analysed age-dependent survival probabilities of territorial (n = 15) and non-territorial (n = 16) adult chamois using information collected over 12 years between 2010 and 2021 in the Gran Paradiso National Park (Western Italian Alps). Survival rates were estimated with a CMR approach using Burnham's joint modelling of live encounter and dead recovery data. The model selection procedure, based on AICc value minimisation, supported a linear decrease of survival with age but the results did not match our predictions, as territorial chamois did not have lower survival rates than non-territorial chamois. In contrast, territorial males appeared to enjoy reproductive success at lower survival costs. This, in turn, supports the role of other factors, such as snow-dependent environmental stochasticity, in the maintenance of ARTs in chamois populations. The limited sample size, however, calls for caution in interpretation, and long-term studies of lifetime reproductive success and survival are necessary to clarify the mechanisms underlying the expression and coexistence of different reproductive behaviours in this species.  相似文献
