American ginseng (Panax quinquefolius L.) is one of the most valuable herbs in the world. Its major active components are ginsenosides. In order to produce ginsenoside heterogeneously, somatic hybridization, a novel approach for genetic introgression, was employed in this study. Protoplasts derived from respective calli of carrot (Daucus carota var. sativus Hoffm.) and American ginseng (P. quinquefolius L.) were used as the fusion partners. Hybrid calli derived from single cell lines containing chromatin of American ginseng were confirmed by the analyses of isozyme, Random amplified polymorphic DNA (RAPD) and genomic in situ hybridization (GISH). High performance liquid chromatography (HPLC) results showed that the ginseng monomer Rb1 was synthesized in seven of the hybrid calli identified as well as in the parent American ginseng calli but not in the parent carrot calli. Results indicated that hybrid introgression lines could produce ginsenoside Rb1 and the ginsenoside Rb1 biosynthesis pathway has been introgressed into carrot cells via somatic hybridization. From the point of biosafety view concerning the consumer acceptance, the potential predominance to produce ginsenosides with somatic hybridization other than with genetic transformation is discussed. Over the past several decades, the pharmacological effects of ginsenosides in Panax ginseng roots have been extensively investigated. Here, we developed a method for producing specific ginsenosides (F1 and F2) with good yields (F1:162 mg/g, F2:305 mg/g) using ??-glycosidase purified from Aspergillus niger. In addition, each ginsenoside (at least 25 species) was separated and purified by high performance liquid chromatography (HPLC) using five different types of solvents and different purification steps. In addition, the Rg3:Rh2 mixture (1:1, w/w) was shown to inhibit a specific lung cancer cell line (NCI-H232) in vivo, displaying an anticancer effect at a dose lower than achieved using treatments with single Rg3 or Rh2. This finding suggests that the combination of ginsenosides for targeting anticancer is more effective than the use of a single ginsenoside from ginseng or red ginseng.  相似文献

Rg3 and Rh2 ginsenosides are primarily found in Korean red ginseng root (Panax ginseng C.A. Meyer) and valued for their bioactive properties. We quantified both Rh2 and Rg3 ginseng leaf and Rg3 from root extracts derived from North American ginseng (Panax quinquefolius). Quantification was obtained by application of HPLC with ion fragments detected using ESI-MS. Ginseng leaf contained 11.3+/-0.5 mg/g Rh2 and 7.5+/-0.9 mg/g Rg3 in concentrated extracts compared to 10.6+/-0.4 mg/g Rg3 in ginseng root. No detectable Rh2 was found in root extracts by HPLC, although it was detectable by ESI-MS analysis. Ginsenosides Rg3 and Rh2 were detected following hot water reflux extraction, but not from tissues extracted with 80% aqueous ethanol at room temperature. Therefore ginsenosides Rg3 and Rh2 are not naturally present in North American ginseng, but are products of a thermal process. Using ESI-MS analysis, it was found that formation of Rg3 and Rh2, among other compounds, were a function of heating time and were breakdown products of the more abundant ginsenosides Rb1 and Rc. Our findings that heat processed North American ginseng leaf is an excellent source of Rh2 ginsenoside is an important discovery considering that ginseng leaf material is obtainable throughout the entire plant cycle for recovery of valuable ginsenosides for pharmaceutical use.  相似文献

Ginseng (Panax ginseng) is one of the most medically important plants in the world. Dammarane-type ginsenosides, which mainly include protopanaxatriol-type (PPT-type) and protopanaxadiol-type (PPD-type) ginsenosides, are the major pharmacologically relevant compounds that are produced by ginseng. Dammarenediol-II synthase (DDS) is the first committed enzyme in the ginsenoside biosynthetic pathway for dammarane-type ginsenosides, and PPD-type and PPT-type ginsenosides are catalyzed by protopanaxadiol synthase (PPDS) and protopanaxatriol synthase (PPTS), respectively. Ginseng cells are often used in stress studies. During their growth and development, ginseng plants are often exposed to cold stress. This study evaluated the effects of different chilling stresses on the accumulation of ginsenosides and the expressions of the DDS, PPDS and PPTS genes in ginseng cells. The results showed that continuous chilling (5 °C for 12 h) induced the PPT-type ginsenosides; whereas intermittent chilling (25 °C for 12 h and 5 °C for 12 h) stimulated the accumulation of PPD-type ginsenosides. The expression levels of DDS, PPDS and PPTS were clearly consistent with the accumulation pattern for PPT-type ginsenosides under continuous chilling stress or PPD-type ginsenosides under intermittent chilling stress, as was their order of involvement in the PPT-type or PPD-type biosynthetic pathway. These results indicate that different chilling treatments stimulated the accumulation of different types of ginsenosides, suggesting that cold stress may be one of the reasons for ginsenoside accumulation in ginseng cells.  相似文献

An HPLC-MS method using an atmospheric pressure chemical ionisation (APCI) source has been developed to assist in the differentiation of three ginseng species: Panax quinquefolium (American ginseng), P. ginseng (Chinese ginseng) and P. notoginseng (sanqi) species. The differentiation method relies on the identification of ginsenosides Rf and F11 and notoginsenoside R1. R1 is observed in both P. notoginseng and Chinese ginseng, whilst F1, is found exclusively in the American species. The presence of these compounds permits the definitive identification of the species to be made. The APCI ionisation source has been employed to tackle the matrix interference in analysing Chinese medicinal materials and to minimise the associated matrix effects that are commonly encountered with other ionisation modes. Moreover, the method allows direct interface to conventional HPLC systems. More importantly, chemical reference standards of ginsenosides are not required in this method. This technique provides an alternative approach to analysing high molecular weight polar compounds that typically encountered in complex matrices of Chinese medicinal materials.  相似文献

In order to determine the active ingredients in root extracts of Panax quinquefolius (American ginseng), a gradient HPLC method involving UV photodiode array detection was applied to separate and quantify simultaneously the ginsenosides Rb1, Rb2, Rc, Rd, Re, Rf and Rg1. All ginseng saponins were baseline-resolved under the selected conditions, and the detection limits were 1.0 microg/mL or less. The method has been applied to analyse ginsenosides extracted from American ginseng cultivated in both Wisconsin and Illinois. Ginsenosides Re and Rb1 were the two main ginseng saponins in the root. The amounts of Re in 5- and 7-year Illinois-cultivated samples were greater than those found in ginseng cultivated for 3 or 4 years in Wisconsin, whereas the levels of Rb1 were greater in the younger Wisconsin samples.  相似文献

Ginseng saponins (ginsenosides) were isolated from soil associated with the roots of commercially grown American ginseng (Panax quinquefolius L.), identified via LC-MS and quantified via analytical HPLC. The ginsenosides, including F(11), Rb(1), Rb(2), Rc, Rd, Re and Rg(1), represented between 0.02 and 0.098% (average 0.06%) of the mass of the soil collected from roots annually between 1999 and 2002. The same ginsenosides were also isolated from run-off of undisturbed plants grown in pots in a greenhouse using a root exudate trapping system. To investigate (1) whether these saponins could influence the growth of pythiaceous fungi pathogenic to ginseng, and (2) whether soil levels of ginsenosides were sufficient to account for any effects, bioassays were completed using a crude saponin extract and an ecologically relevant concentration of purified ginsenosides. Thus, when cultured on media containing crude saponins, the colony weight of both Phytophthora cactorum and Pythium irregulare was significantly greater than that of control, indicating a strong growth stimulation by ginsenosides. The growth of Pythium irregulare was also significantly stimulated after addition of an ecologically relevant, low concentration (i.e. 0.06%) of purified ginsenosides to culture medium. By contrast, growth of the saprotrophic fungus Trichoderma hamatum was slightly (but not significantly) inhibited under the same conditions. These results imply that ginsenosides can act as allelopathic stimulators of the growth of pythiaceous fungi in the rhizosphere, and this may contribute to the disease(s) of this crop.  相似文献

Ginsenosides are the principal components responsible for the pharmaceutical activities of ginseng. The minor ginsenosides, which are also pharmaceutically active, can be produced via the hydrolysis of the sugar moieties in the major ginsenosides using acid hydrolytic, heating, microbial, and enzymatic transformation techniques. The enzymatic method has a profound potential for ginsenoside transformation, owing to its high specificity, yield, and productivity, and this method is increasingly being recognized as a useful tool in structural modification and metabolism studies. In this article, the transformation methods of ginsenosides, the characterization of microbial glycosidases with ginsenoside hydrolyzing activities, and the enzymatic production of minor ginsenosides are reviewed. Moreover, the conversions of ginsenosides using cell extracts from food microorganisms and recombinant thermostable β-d-glycosidases are proposed as feasible methods for use in industrial processes.  相似文献

环境条件影响中药材活性成分的形成和积累.利用各种数学统计分析方法探讨影响人参皂苷积累的生态因子,提高人参品质.人参样品采自人参道地产区(主产区)吉林、辽宁、黑龙江三省5年生栽培人参,同时采集采样点处的土壤样品.超高效液相(UPLC)色谱法分析了不同产区9种人参皂苷(Rg1、Re、Rf、Rg2、Rb1、Rc、Rb2、Rb3、Rd)的含量;利用“中药材产地适宜性分析地理信息系统”的生态因子空间数据库,获得采样区包括温度、水分、光照等10个生态因子数据;按土壤理化性质常规方法测定土壤样品中的有效硼、有效铁等微量元素和速效氮、速效钾等有效养分.对人参有效成分含量与土壤养分进行典型相关性分析发现,土壤中的有效硼、有效铁、速效氮与人参皂苷含量呈显著正相关,即适当提高土壤中有效硼、有效铁和速效氮的含量可以促进人参皂苷成分的积累,土壤水分与所测人参皂苷含量(Rb3除外)呈显著正相关,速效磷(P)、pH、速效锌(Zn)与各人参皂苷含量呈弱相关;人参皂苷与气候因子相关分析表明,温度(年活动积温、年平均气温、7月最高气温、7月平均气温、1月最低气温、1月平均气温)与人参皂苷含量呈显著负相关,其中与药典中人参含量测定项下的人参皂苷Rg1、Re、Rb1负相关尤为显著(r>0.6),说明在一定温度范围内,人参皂苷是随着温度的降低而升高的,即适当低温有利于人参皂苷有效成分的积累;海拔与人参皂苷Rc、Rb2、Rb3含量呈显著正相关(r>0.6),即相对较高的海拔可以促进这3种成分的积累;而年均降水量、年相对湿度和年均日照时数与人参皂苷相关不显著.通过主成分分析(PCA)、典型相关分析、排序等统计方法,考察不同产地样品中人参皂苷含量与生态因子间的相关性,研究结果揭示了温度在人参的主要活性成分-皂苷类形成中起决定性作用,在一定的温度范围内,温度越低越有利于人参皂苷的积累;阐明了土壤中的有效硼、有效铁、速效氮与人参皂苷含量成正相关.研究结果提示在人参实践生产中可以通过适当低温处理,增施硼、铁、氮肥等农艺措施来调控人参皂苷含量.  相似文献

Ginseng, a traditional medical herb, has been reported having beneficial effects in fatigue, heart diseases, diabetes, immune function and erectile dysfunction. In recent years, increasing investigations have been conducted on ginseng in preventing and treating of obesity, one of the major worldwide escalating public health concerns. However, the effect and the relevant mechanisms behind how ginseng works as an antiobesity treatment are still controversial. In this review, we briefly discussed the chemical structures, metabolism and pharmacokinetics of ginseng and its major bioactive components ginsenosides. The major focus is on the antiobesity effects and the physiological, cellular and molecular mechanisms of ginseng and its ginsenosides in cultured cells, animal models and humans. We particularly compared the ginsenosides profiles, the antiobesity effects and the mechanisms between Asian ginseng (Panax ginseng) and American ginseng (Panax quinquefolius), the two major ginseng species having opposite medical effects in traditional Chinese medicine. Our unpublished data on the ginseng antiobesity in cultured cells and mice were also included. We further addressed the current problems and future directions of the ginseng antiobesity research.  相似文献

BackgroundGinseng has therapeutic potential for treating obesity and the associated gut microbiota dysbiosis. However, whether white ginseng and red ginseng, the two kinds of commonly used processed ginseng, possess different anti-obesity effects remains unknown.PurposeAnti-obesity effects of water extracts of white ginseng and red ginseng (WEWG and WERG) were compared, and the potential mechanisms were discussed.MethodsChemical profiles of WEWG and WERG were characterized by ultra-high performance liquid chromatography-tandem triple quadrupole mass spectrometry (UHPLC-QqQ-MS/MS) and high performance liquid chromatography coupled with evaporative light scattering detector (HPLC-ELSD). Anti-obesity effects of WEWG/WERG were examined by determining fat accumulation, systemic inflammation, enteric metabolic disorders and gut microbiota dysbiosis in high-fat diet (HFD)-fed obese mice.ResultsBoth WEWG and WERG exerted anti-obesity effects, with WEWG stronger than WERG. Compared to WERG, WEWG contained less contents of carbohydrates (polysaccharides, oligosaccharides, free monosaccharides) and ginsenosides, but chemical structures or compositions of these components in WEWG were characteristic, i.e. narrower molecular weight distribution and higher molar ratios of glucose residues of polysaccharides; higher content ratios of oligosaccharides DP2–3 (di-/tri-saccharides)-to-oligosaccharides DP4–7 (tetra-/penta-/hexa-/hepta-saccharides), sucrose-to-melibiose, maltose-to-trehalose and high-polar-to-low-polar ginsenosides. WEWG better ameliorated fat accumulation, enteric metabolic disorders and gut microbiota dysbiosis in HFD-fed obese mice than WERG.ConclusionThe stronger anti-obesity effect of white ginseng appears to correlate with differences in its chemical profile as compared to red ginseng. The carbohydrates and ginsenosides in WEWG potentially present more structural and compositional specificity to the obesity-associated gut bacteria, allowing more beneficial effects of WEWG on the gut microbiota dysbiosis. This consequently better alleviates the enteric metabolic disorders and systemic inflammation, thereby contributing to the stronger anti-obesity effect of WEWG as compared to WERG.  相似文献

Asian ginseng (Panax ginseng) and American ginseng (Panax quinquefolium), are thought to be representative plant of Panax species, have important commercial value and are used in worldwide. Panax species produces triterpene saponins called ginsenosides, which are classified into two groups by the skeleton of aglycones, namely dammarane-type and oleanane-type. Dammarane-type ginsenosides dominate over oleanane-type not only in amount but also in structural varieties. Researches shows that the saponins content in American ginseng is higher than that in Asian ginseng, the higher part of ginsenosides is from dammarane-type biosynthesis. It has been proposed that protopanaxadiol derived from dammarenediol-II, is a key hydroxylation by cytochrome P450 for the biosynthesis of ginsenosides, and the gene number of protopanaxadiol synthase has been published independent in Asian ginseng (PgCYP716A47). However, little is known about genes involved in hydroxylation and glycosylation in American ginseng ginsenoside biosynthesis. Here, we first cloned and identified a P450 gene named PqD12H encoding enzymes catalyzed dammarenediol-II to protopanaxadiol by RT-PCR using degenerate primers designed based on sequence homology. In vitro, the ectopic expression of PqD12H in recombinant WAT21 yeast resulted in protopanaxadiol production after dammarenediol-II was added to the culture medium. In vivo, we established both PgCYP716A47 and PqD12H RNAi transgenic. The RT-PCR and HPLC analysis of the final products of protopanaxadiol and protopanaxatriol showed a result that declined level of protopanaxadiol-type and protopanaxatriol-type ginsenosides. It suggested that the P450 synthase content or expression in American ginseng exceed than in Asian ginseng. The result elucidated the evolution relationship of P450s and the reason of different saponins content among Panax species.  相似文献

Most of the known pharmacological effects of Panax ginseng on the central nervous system are due to its major components - ginsenosides. Although the antioxidant ability of ginseng root has already been established, this activity has never been evaluated for isolated ginsenosides on astrocytes. The activity of protopanaxadiols Rb(1), Rb(2), Rc and Rd, and protopanaxatriols Re and Rg(1) was evaluated in vitro on astrocytes primary culture by means of an oxidative stress model with H(2)O(2). The viability of astrocytes was determined by the MTT reduction assay and by the LDH release into the incubation medium. The effects on the antioxidant enzymes catalase, superoxide dismutase (SOD), glutathione peroxidases (GPx) and glutathione reductase (GR) and on the intracellular reactive oxygen species (ROS) formation were also investigated. Exposure of astrocytes to H(2)O(2) decreased cell viability as well as the antioxidant enzymes activity and increased ROS formation. Oxidative stress produced significant cell death that was reduced by previous treatment with the tested ginsenosides. Ginsenosides Rb(1), Rb(2), Re and Rg(1) were effective in reducing astrocytic death, while Rb(1), Rb(2), Rd, Re and Rg(1) decreased ROS formation, ginsenoside Re being the most active. Ginsenosides from P. ginseng induce neuroprotection mainly through activation of antioxidant enzymes.  相似文献

20(S)-protopanaxadiol (PPD(S)) and 20(R)-protopanaxadiol (PPD(R)), the main metabolites of ginsenosides Rg3(S) and Rg3(R) in black ginseng, are potential candidates for anti-cancer therapy due to their pharmacological activities such as anti-tumor properties. In the present study, we report the preparation of PPD(S, R) by a combination of steaming and biotransformation treatments from ginseng. Aspergillus niger was isolated from soil and showed a strong ability to transform Rg3(S, R) into PPD(S, R) with 100% conversion. Furthermore, the enzymatic reactions were analyzed by reversed-phase HPLC, showing the biotransformation pathways: Rg3(S) → Rh2(S) → PPD(S) and Rg3(R) → Rh2(R) → PPD(R), respectively. In addition, 12 ginsenosides including 3 pairs of epimers, namely Rg3(S), Rg3(R), Rh2(S), Rh2(R), PPD(S) and PPD(R), were simultaneously determined by reversed-phase HPLC. Our study may be highly applicable for the preparation of PPD(S) and PPD(R) for medicinal purposes and also for commercial use.  相似文献

Almost half of the drugs in use today are chiral. It is well established that the pharmacological activity is mostly restricted to one of the enantiomers (eutomer). There can be qualitative and quantitative differences in the activity of the enantiomers. In many cases, the inactive enantiomer (distomer) shows unwanted side effects or even toxic effects. Even if the side effects are not that drastic, the distomer has to be metabolized and this represents an unnecessary burden for the organism. Therefore, the development of methods for the separation of enantiomers, both on analytical and preparative scale, has become increasingly important. Chromatographic techniques such as thin layer chromatography (TLC), gas chromatography (GC), supercritical fluid chromatography (SFC), and above all high-performance liquid chromatography (HPLC) have been used for enantiomer separation for about two decades. More recently, electromigration techniques, such as capillary electrophoresis and capillary electrochromatography, have been shown to be powerful alternatives to chromatographic methods. This review gives a short overview of different chiral separation principles and their application. Several new developments are discussed.  相似文献

为了扩大人参(Panax ginseng)栽培面积, 解决人参资源日益短缺的问题, 研究了人参皂苷与生态因子之间的相关性。利用超高效液相(UPLC)色谱法, 测定了辽宁、吉林和黑龙江三省不同产区人参样品中3种人参皂苷(Rg1、Re和Rb1)的含量, 并基于“中药材产地适宜性分析地理信息系统”(TCMGIS)平台, 获得采样区域10个生态因子(包括活动积温、年平均气温、海拔、相对湿度、年日照时数、年降水量、7月最高气温、7月平均气温、1月最低气温和1月平均气温等)数据; 利用因子分析法对16个人参基地进行因子得分评价, 得分最高的是吉林和辽宁的人参基地, 故将吉林和辽宁的人参基地作为人参生态适宜性分析的最佳区域; 通过偏最小二乘回归法建立3种人参皂苷成分与上述10个生态因子间的回归方程并获取其相应的权重, 结果发现多个温度因子与人参皂苷含量呈强负相关关系, 说明热量因子对人参皂苷活性成分的累积起主要作用, 而水分因子、地理因子和光照因子与人参皂苷含量呈弱相关关系; 以因子得分最高的吉林和辽宁人参基地为基点区域, 分别对3种人参皂苷进行单成分生态适宜性区划以及综合区划, 得知3种人参皂苷成分积累的最佳区域主要集中在长白山脉, 而燕山山脉和太行山脉只有少量分布区域。  相似文献

人参皂苷是我国传统中药人参的主要活性物质,稀有人参皂苷相较人参皂苷具有更好的生物活性,也更利于身体吸收,具有镇静催眠、促进细胞分化增殖、抗肿瘤、降血糖、提升免疫力等作用。然而,稀有人参皂苷结构复杂且在人参中含量极低,限制了其开发利用。随着生物技术的发展,利用生物法合成稀有人参皂苷成为本领域的研究热点。因此,对近年来生物合成稀有人参皂苷研究进行汇总梳理,总结稀有人参皂苷的主要种类结构及近年来生物转化法和异源合成法合成稀有人参皂苷的研究进展,生物转化法汇总了以人参皂苷为底物的转化生物,异源合成法总结人参皂苷的生物合成途径及形成结构多样化人参皂苷的酶。对生物合成稀有人参皂苷存在的问题进行了讨论,同时展望了其前景以及未来研究方向,以期为从事人参研究者提供更多生物线索和制备策略。  相似文献
