Eight isolates of Cucumber mosaic virus (CMV) isolated from seven different host species were tested for their virulence on potato cv. Desirée. Three isolates caused a systemic infection, of which one isolate from Asiatic lily (CMV-P26) and one from cucumber (CMV-J) appeared to be highly virulent, in contrast to the third isolate (CMV-M) that originated from cucumber and caused mild symptoms only. These three isolates were transmitted to 26 additional potato cultivars by mechanical inoculation in a greenhouse. All cultivars were infected with at least one CMV isolate and developed local chlorotic symptoms, but only 17 cultivars (including Desiree) developed primary systemic symptoms including necrosis, mosaic and/or malformation of leaves. Furthermore, in only five cultivars (including Desiree) CMV was transmitted to tubers and was subsequently detected in plants of the first and second vegetative progeny, the secondary symptoms of these plants being severe. The observed phenotypic responses of potatoes to CMV were not associated with the maturity type (early or late) or resistance to other viruses. Results of this study indicate a high level of biological variability among CMV isolates and that infection in potato depends on CMV isolate and potato cultivar.  相似文献

The Mi resistance gene in tomato reduces the feeding, fecundity, and survival of certain isolates of the potato aphid (Macrosiphum euphorbiae Thomas). This study compared the performance of two potato aphid isolates, WU11 and WU12, on nearly isogenic susceptible (Mi-) and resistant (Mi+) tomato cultivars. Although Mi significantly reduced the population growth of both aphids, WU12 numbers decreased by only 15% compared with 95% for isolate WU11. These results show that there are quantitative differences in virulence among potato aphid isolates. Compared with WU11 aphids, isolate WU12 caused more necrosis on both resistant and susceptible plants, and this increased damage may play a role in the partial virulence of isolate WU12. However, infestation with aphid isolate WU12 did not compromise plant defenses against isolate WU11 in resistant plants. Prior inoculation with either aphid isolate caused a modest reduction in the survival of WU12 adults, but this form of induced resistance was observed on both resistant and susceptible cultivars. Thus, Mi did not play a role in acquired resistance or mediate any indirect interactions between the two aphid isolates. Notably, the mode of action of Mi-mediated resistance seemed to differ depending on the aphid isolate tested. Mi dramatically deterred feeding by WU11 aphids, whereas the effects of resistance on isolate WU12 seemed to be caused primarily by antibiosis. Tolerance did not seem to be a major component of Mi-mediated responses, although resistant plants showed a modest reduction in the amount of foliar necrosis induced per aphid compared with susceptible plants.  相似文献

18 PLRV isolates differed in severity of symptoms induced on Physalis floridana and on three potato cultivars (Osa, Pola, and Giewont). Reactions of potato cultivars to individual isolates were not always related. Some isolates induced more severe symptoms than others in all host plants. Isolates producing more severe symptoms reached a higher concentration in potato cultivars (evaluated with ELISA test) and were more virulent to Capsella bursa-pastoris. An interaction between PLRV isolates and host plants could be demonstrated. In general, the severity of the reaction of cv. Pola was related to that of P. floridana but not to the reaction of cvs. Osa and Giewont. The reactions of cvs. Osa and Giewont were closely related. All six isolates selected for a more detailed study could infect C. bursa-pastoris with obvious disease symptoms if potato cv. Osa served as source of the virus. Some isolates failed to infect this plant when other cultivars were used.  相似文献

In 1998, Verticillium sp. (CE98Vt1 and CE98Vt2) were isolated from discolored vascular structures of potato tubers sold at a market in Chiba Prefecture. These isolates were identified as Verticillium tricorpus on the basis of cultural and morphological characteristics and PCR diagnosis. This observed vascular discoloration of the potato tuber was demonstrated in three cultivars (Touya, Toyoshiro, and Waseshiro) among eight cultivars by inoculation to seedlings. External and internal symptoms of these isolates were not distinct in potato plants. The virulence of these isolates to potato was very low as compared with Verticillium dahliae. These two isolates were not pathogenic to Chinese cabbage, eggplant, green pepper, larkspur, parsley, snapdragon, soybean, tobacco, and tomato. This is the first report of V. tricorpus from potato in Japan.  相似文献

Differential gene expression was analyzed after infection with Phytophthora infestans in six potato cultivars with different levels of resistance to late blight. To verify the infection of the potato leaflets, the amount of phytopathogen mRNA within the plant material was quantified by real-time quantitative PCR. The expression of 182 genes selected from two subtracted cDNA libraries was studied with cDNA array hybridization using RNA from non-infected and infected potato leaflets. Gene up- and down-regulation were clearly detectable in all cultivars 72 h post inoculation. Gene expression patterns in susceptible cultivars differed from those in potato varieties with a higher level of resistance. In general, a stronger gene induction was observed in the susceptible cultivars compared to the moderately to highly resistant potato varieties. Five genes with the highest homology to stress and/or defence-related genes were induced specifically in the susceptible cultivars. Four genes responded to pathogen attack independently of the level of resistance of the cultivar used, and three genes were repressed in infected tissue of most cultivars. Even in the absence of P. infestans infection, six genes showed higher expression levels in the somewhat resistant cultivars Bettina and Matilda. Possible reasons for the different levels of gene expression are discussed.  相似文献

Potato virus X (PVX) isolates were obtained from a simple seed potato production scheme or from ware potatoes produced by seed potatoes obtained from it. In this scheme, PVX infection is widespread in seed stocks and most of the potatoes grown lack PVX resistance genes. Thirteen PVX isolates were typed to strain group by inoculation to potato cultivars containing different combinations of hypersensitivity genes Nx and Nb. Six failed to overcome either gene and therefore belonged to strain group 1, four overcame Nb only and were placed in strain group 3 and three were mixtures of the two. All 13 isolates failed to overcome extreme resistance/immunity gene Rx. Naturally infected cultivars of genotype nx.nb contained strain group 1 alone or strain groups 1 and 3, while those of genotype nx:Nb contained only strain group 3. The widespread occurrence of strain group 1 contrasts with the predominant occurrence of strain group 3 in potatoes in the UK. However, it resembles the UK situation before sophisticated seed potato production schemes were introduced and before PVX hypersensitivity genes Nx and Nb were deliberately exploited in potato breeding. Prior infection with potato leafroll virus (PLRV) did not affect expression of hypersensitivity to PVX in inoculated leaves of an nx:Nb genotype.  相似文献

Prevalence of tan spot of wheat caused by the fungus Pyrenophora tritici-repentis has become more prevalent in Oklahoma as no-till cultivation in wheat has increased. Hence, developing wheat varieties resistant to tan spot has been emphasized, and selecting pathogen isolates to screen for resistance to this disease is critical. Twelve isolates of P. tritici-repentis were used to inoculate 11 wheat cultivars in a greenhouse study in split-plot experiments. Virulence of isolates and cultivar resistance were measured in percent leaf area infection for all possible isolate x cultivar interactions. Isolates differed significantly (P < 0.01) in virulence on wheat cultivars, and cultivars differed significantly in disease reaction to isolates. Increased virulence of isolates detected increased variability in cultivar response (percent leaf area infection) (r = 0.56, P < 0.05) while increased susceptibility in cultivars detected increased variance in virulence of the isolates (r = 0.76, P < 0.01). A significant isolate × cultivar interaction indicated specificity between isolates and cultivars, however, cluster analysis indicated low to moderate physiological specialization. Similarity in wheat cultivars in response to pathogen isolates also was determined by cluster analysis. The use of diverse isolates of the fungus would facilitate evaluation of resistance in wheat cultivars to tan spot.  相似文献

One hundred and four Swedish isolates of Bremia lactucae were tested between 1971 and 1976 for their virulence phenotypes. Eighty of these isolates showed patterns of virulence previously recorded in other countries while the remainder conformed to seven previously unreported virulence factor combinations. The simplest resistance factor combination that could give resistance to all isolates collected was R2 plus Rl 1. This combination now exists in some recently bred lettuce cultivars. Most of the possible virulence factor combinations occurred in the population at the expected frequency. However, several combinations, which had not been directly selected (by the growing of cultivars carrying the comparable resistance factor combinations) were present at higher or lower than expected frequencies.  相似文献

Samples of lettuces infected with Bremia lactucae were collected from seven areas of England during 1973-5. These were tested under controlled environmental conditions on a range of differentially resistant cultivars to determine the frequency of virulence (V) factors in the pathogen population. In most cases the results could be accommodated by the relationship advanced by Crate & Johnson (1976). A virulence phenotype was determined for each isolate and individual virulences within isolates were identified as present at high or low frequencies. The most common V factors were V3, V4, V6 and V8 which reflected the resistance (R) factor complement of lettuce cultivars currently in commercial production. There were few gross differences in the geographical distribution of virulence factors between the seven areas. In the West Central region, however, which encompasses the Evesham and Lancashire production areas, results indicated that the use of cultivars containing resistance (R) factors 2 and 8 respectively would have provided protection from the disease during 1974. There were differences in virulence combinations at the holding or locality level within each large area, which could be exploited by growers providing testing was done in their particular locality. Universally susceptible cultivars and those with only limited resistance factors were often found to be infected with B. lactucae populations which contained large numbers of V-factors. It might have been expected that these pathogen populations would have been restricted to cultivars with large numbers of R-factors. This finding conflicts with the suggestion that pathogen populations with complex virulence patterns are ‘unfit’ as compared with those with more simple virulence components.  相似文献

Eight isolates from different potato growing regions in Vietnam were characterized. All were highly pathogenic in some potato cultivars, but did not overcome the extreme resistance of Solanum stoloniferum and Solanum demissum. RT‐PCR analysis revealed that all of these isolates are recombinants. Sequence data for 4 isolates were obtained, and their reaction in potato cultivars harbouring specific N genes was determined. Different phylogenetic analyses of viral sequences confirmed previous results that the recombinant isolates evolved from different parental sequences. One of the Vietnamese isolates investigated had a specific structure. The need for a clear classification of PVYNWi isolates is discussed.  相似文献

The virulence spectrum of 23 monopycnidiospore isolates of Mycosphaerella graminicola was determined using wheat genotypes that carried different resistance genes (Stb1Stb8 and Stb15). Disease severity was measured as the percentage of necrotic leaf area. The isolates used in the experiments were of diverse origin: eight from Poland, seven from Germany, and eight from other countries around the world. Analysis of variance revealed significant differences in the virulence of the isolates. Using multiple regression and Cook’s D statistic, 26 significant cultivar × isolate interactions were detected. The Israeli isolate IPO86036 showed the widest spectrum of specific reactions. It expressed specific virulence on at least four cultivars and specific avirulence on at least three. The other isolates showed specific interactions with 1–6 different cultivars. Despite the limited number of isolates that were tested, we recommend that a number of resistant lines, namely cultivars Veranopolis (Stb2), Cs/Synthetic 7D (Stb5), Arina (Stb15, Stb6 and partial resistance), and Liwilla (unknown resistance factors), could be incorporated into central European wheat breeding programmes that are aimed at developing resistance against septoria tritici blotch. In contrast, resistance gene Stb7, which is carried by cultivar Estanzuela Federal, was ineffective against most of the isolates that were used. These results on the virulence spectrum of M. graminicola isolates provide valuable information for effective wheat breeding programmes to develop resistance to the pathogen.  相似文献

Reactions to two subgroup I isolates (Fny-CMV and Pf-CMV) and two subgroup II isolates (A9-CMV and LS-CMV) of cucumber mosaic virus (CMV) were studied in three non tuber-bearing wild potato species (Solanum spp.) of the series Etuberosa, and in two tuber-bearing interspecific potato hybrids and four potato cultivars using graft-inoculation. Three classes of phenotypic reactions (susceptible, hypersensitive, extreme resistance) were observed in the tuber-bearing genotypes. Susceptible genotypes developed mosaic or severe mosaic with leaf malformation and had high CMV titres. Hypersensitive genotypes developed either top necrosis or vein necrosis and/or necrotic spots on apical leaves, and had low CMV titres. Extremely resistant genotypes had no symptoms and no CMV was detected. The hybrid 87HW13.7 (S. tuberosum×S. multidissectum) developed top necrosis specific to infection with Fny-CMV. The hybrid ‘A6’ (S. demissum×S. tuberosum cv. Aquila) was hypersensitive to all CMV isolates tested. Extreme resistance was not functional against all CMV isolates. Neither hypersensitivity nor extreme resistance were related to the CMV subgroup.  相似文献

The interaction between sunflower plants showing a high level of quantitative resistance and five Plasmopara halstedii (the causal agent of downy mildew) isolates of several races were studied using five single zoosporangium isolates per pathogen isolate. Aggressiveness criteria were analyzed for 25 P. halstedii single zoosporangium isolates. Based on the reaction for the P. halstedii isolates to four sunflower hybrids H1–H4 varying only in their downy mildew resistance genes, there were differences in virulence spectrum in pathogen isolates. Analysis of five single zoosporangium isolates for P. halstedii isolates showed significant variability within pathogen isolate for all aggressiveness criteria but not for all pathogen isolates. The hypothesis explaining the interaction between P. halstedii and its host plant was discussed on the level of pathogenicity.  相似文献

Migrations or introduction of new genotypes of Phytophthora infestans to a specific region imposes a different perspective for potato production. During 2009–2010, a late blight epidemic affected the Northeastern United States, which quickly spread through several states. The epidemic was characterized by the appearance of a new genotype of P. infestans designated US‐22, which was isolated from tomato and potato. Potato tubers are an essential component of late blight epidemics where the pathogen cannot overwinter on Solanaceous plants. Six potato cultivars were inoculated with 12 isolates of P. infestans (five different genotypes), including isolates of the genotype US‐22. Tuber blight development was characterized in terms of tissue darkening expressed as area under the disease progress curve values and lenticel infection. The responses indicated that US‐8 was more aggressive than US‐22, but US‐22 isolates obtained from potato were more aggressive on potato than those acquired from tomato. Tuber periderm responses to infection were limited, yet US‐8 isolates infected the periderm more often than US‐22 isolates. There were significant differences among the cultivars tested but cv. Jacqueline Lee was the most resistant overall. Although isolates of P. infestans genotype US‐22 were less aggressive in comparison with US‐8 isolates, US‐22 isolates still infected potato tubers and were as aggressive us US‐8 isolates on some cultivars. Management of late blight caused by isolates of US‐22 through host resistance may be feasible but imposes a different set of criteria for consideration from those that US‐8 imposed.  相似文献

Abstract

Aphids have serious damage and well known of the most important pests, which may transmit different viruses on potato. Potato has immense significance as a food crop throughout the world. In the last research, the most important and different aspects of predominant potato varieties; Agria, Arinda, Fontane, Banba and Sante were studied during 2015–2016 in Karaj, Iran. This research has been conducted into several disparate categories: the effect of environmental conditions on the physiological trends, aphid characteristics and virus infection. The results showed that potato fields had an increasing trend to the end of season in the aspect of infestation by Aphis gossypii Glover and Myzus persicae Sulzer. The potato cultivars showed the same reaction in the lab and no host preference was observed between them against cotton aphid (A. gossypii). Finally, Arinda, and Agria varieties had the most photosynthesis activity, chlorophyll concentration, and leaf dry weight values and had the least electrical conductivity and susceptibility to some potato virus diseases. These results may help to provide better potato varieties by considering to the important physiological characteristic related to some destructive pests.  相似文献

Quantitative powdery mildew resistance in compatible host-pathogen-combinations was measured by the number of pastules/cm2 leaf area. Spring barley cultivar ‘Proctor’ was significantly less infected than ‘Golden Promise”. Using these two cultivars (having no effective major resistance gene) as controls, MO- and AR-resistant cultivars were inoculated with virulent mildew isolates. ‘Mona”, ‘Grit’ and ‘Nudinka’ had a higher or, at least, the same level of quantitative resistance as ‘Proctor”. None of the remaining cultivars showed the high susceptibility expressed by ‘Golden Promise”. Ranking of host genotypes was nearly constant while that of mildew isolates varied considerably. Only a small portion of the observed variance was due to interaction between host cultivars and pathogen isolates. ‘Triesdorfer Diva’ gave a resistant infection type after inoculation with different AR-virulent isolates, indicating that this cultivar has major resistance other than that conditioned by gene Ml-a12.  相似文献
