A lithotrophic freshwater Beggiatoa strain was enriched in O2-H2S gradient tubes to investigate its ability to oxidize sulfide with NO3 as an alternative electron acceptor. The gradient tubes contained different NO3 concentrations, and the chemotactic response of the Beggiatoa mats was observed. The effects of the Beggiatoa sp. on vertical gradients of O2, H2S, pH, and NO3 were determined with microsensors. The more NO3 that was added to the agar, the deeper the Beggiatoa filaments glided into anoxic agar layers, suggesting that the Beggiatoa sp. used NO3 to oxidize sulfide at depths below the depth that O2 penetrated. In the presence of NO3 Beggiatoa formed thick mats (>8 mm), compared to the thin mats (ca. 0.4 mm) that were formed when no NO3 was added. These thick mats spatially separated O2 and sulfide but not NO3 and sulfide, and therefore NO3 must have served as the electron acceptor for sulfide oxidation. This interpretation is consistent with a fourfold-lower O2 flux and a twofold-higher sulfide flux into the NO3-exposed mats compared to the fluxes for controls without NO3. Additionally, a pronounced pH maximum was observed within the Beggiatoa mat; such a pH maximum is known to occur when sulfide is oxidized to S0 with NO3 as the electron acceptor.  相似文献

Experiments demonstrated that Beggiatoa could induce a H2S-depleted suboxic zone of more than 10 mm in marine sediments and cause a divergence in sediment NO3 reduction from denitrification to dissimilatory NO3 reduction to ammonium. pH, O2, and H2S profiles indicated that the bacteria oxidized H2S with NO3 and transported S0 to the sediment surface for aerobic oxidation.  相似文献

The interactions between colorless sulfur bacteria and the chemical microgradients at the oxygen-sulfide interface were studied in Beggiatoa mats from marine sediments and in Thiovulum veils developing above the sediments. The gradients of O2, H2S, and pH were measured by microelectrodes at depth increments of 50 μm. An unstirred boundary layer in the water surrounding the mats and veils prevented microturbulent or convective mixing of O2 and H2S. The two substrates reached the bacteria only by molecular diffusion through the boundary layer. The bacteria lived as microaerophiles or anaerobes even under stirred, oxic water. Oxygen and sulfide zones overlapped by 50 μm in the bacterial layers. Both compounds had concentrations in the range of 0 to 10 μmol liter−1 and residence times of 0.1 to 0.6 s in the overlapping zone. The sulfide oxidation was purely biological. Diffusion calculations showed that formation of mats on solid substrates or of veils in the water represented optimal strategies for the bacteria to achieve a stable microenvironment, a high substrate supply, and an efficient competition with chemical sulfide oxidation. The continuous gliding movement of Beggiatoa cells in mats or the flickering motion of Thiovulum cells in veils were important for the availability of both O2 and H2S for the individual bacteria.  相似文献

Zero-valent sulfur is a key intermediate in the microbial oxidation of sulfide to sulfate. Many sulfide-oxidizing bacteria produce and store large amounts of sulfur intra- or extracellularly. It is still not understood how the stored sulfur is metabolized, as the most stable form of S0 under standard biological conditions, orthorhombic α-sulfur, is most likely inaccessible to bacterial enzymes. Here we analyzed the speciation of sulfur in single cells of living sulfide-oxidizing bacteria via Raman spectroscopy. Our results showed that under various ecological and physiological conditions, all three investigated Beggiatoa strains stored sulfur as a combination of cyclooctasulfur (S8) and inorganic polysulfides (Sn2−). Linear sulfur chains were detected during both the oxidation and reduction of stored sulfur, suggesting that Sn2− species represent a universal pool of bioavailable sulfur. Formation of polysulfides due to the cleavage of sulfur rings could occur biologically by thiol-containing enzymes or chemically by the strong nucleophile HS as Beggiatoa migrates vertically between oxic and sulfidic zones in the environment. Most Beggiatoa spp. thus far studied can oxidize sulfur further to sulfate. Our results suggest that the ratio of produced sulfur and sulfate varies depending on the sulfide flux. Almost all of the sulfide was oxidized directly to sulfate under low-sulfide-flux conditions, whereas only 50% was oxidized to sulfate under high-sulfide-flux conditions leading to S0 deposition. With Raman spectroscopy we could show that sulfate accumulated in Beggiatoa filaments, reaching intracellular concentrations of 0.72 to 1.73 M.  相似文献

Benthic cyanobacterial mats with the filamentous Microcoleus chthonoplastes as the dominant phototroph grow in oxic hypersaline environments such as Solar Lake, Sinai. The cyanobacteria are in situ exposed to chemical variations between 200 μmol of sulfide liter−1 at night and 1 atm pO2 during the day. During experimental H2S to O2 transitions the microbial community was shown to shift from anoxygenic photosynthesis, with H2S as the electron donor, to oxygenic photosynthesis. Microcoleus filaments could carry out both types of photosynthesis concurrently. Anoxygenic photosynthesis dominated at high sulfide levels, 500 μmol liter−1, while the oxygenic reaction became dominant when the sulfide level was reduced below 100 to 300 μmol liter−1 (25 to 75 μmol of H2S liter−1). An increasing inhibition of the oxygenic photosynthesis was observed upon transition to oxic conditions from increasing sulfide concentrations. Oxygen built up within the Microcoleus layer of the mat even under 5 mmol of sulfide liter−1 (500 μmol of H2S liter−1) in the overlying water. The implications of such a localized O2 production in a highly reducing environment are discussed in relation to the evolution of oxygenic photosynthesis during the Proterozoic era.  相似文献

Hydrogen sulfide (H2S) has proved to be a multifunctional signaling molecule in plants and animals. Here, we investigated the role of H2S in the decay of fresh-cut pears (Pyrus pyrifolia). H2S gas released by sodium hydrosulfide (NaHS) prolonged the shelf life of fresh-cut pear slices in a dose-dependent manner. Moreover, H2S maintained higher levels of reducing sugar and soluble protein in pear slices. H2S significantly reduced the accumulation of hydrogen peroxide (H2O2), superoxide radicals (•O2 ) and malondialdehyde (MDA). Further investigation showed that H2S fumigation up-regulated the activities of antioxidant enzymes ascorbate peroxidase (APX), catalase (CAT), and guaiacol peroxidase (POD), while it down-regulated those of lipoxygenase (LOX), phenylalanine ammonia lyase (PAL) and polyphenol oxidase (PPO). Furthermore, H2S fumigation effectively inhibited the growth of two fungal pathogens of pear, Aspergillus niger and Penicillium expansum, suggesting that H2S can be developed as an effective fungicide for postharvest storage. The present study implies that H2S is involved in prolonging postharvest storage of pears by acting as an antioxidant and fungicide.  相似文献

The behavioral response of single Beggiatoa sp. filaments moving on a gas-permeable membrane was studied by the combined use of microscopy and oxygen microelectrodes during controlled oscillations of oxygen tension. The bacteria reacted to increasing oxygen by reversing the direction of movement. The same step-up phobic response to oxygen was observed when a filament tip or loop glided into a stable microgradient of increasing oxygen. The response was sensitive to a change in oxygen tension of <5% of air saturation min−1. The response time was 20 to 50 s. Frequently, only part of the filament responded, which led to the formation of sharp bends, loops, and coils. This partial response facilitated the positioning of the long filaments within the narrow O2-H2S interface. The structure of whole Beggiatoa mats on sediment surfaces varied from loose to dense in relation to shallow or steep oxygen gradients in the 0.3- to 2-mm-thick, unstirred boundary layer. In an illuminated sediment Beggiatoa spp. lived together with photosynthetic organisms and migrated vertically in accordance with light/dark variations. The combined effect of phobic responses to light and oxygen can explain this migration.  相似文献

Zusammenfassung Axenische Kulturen von Beggiatoa wurden zur Prüfung der Frage verwendet, ob autotrophe Ernährung möglich ist. Dazu wurde eine neue Technik ausgearbeitet, bei der FeS als H2S-Quelle dient.Es bestätigte sich, daß entweder heterotrophe Ernährung oder neben H2S kleinste Konzentrationen organischer Energiesubstrate nötig sind. Es liegt dann eine Sonderform von Mixotrophie vor, anders als bei chemoautotrophen Bakterien.Der aus H2S entstehende S ist ein Reservestoff, der aber nicht nur, etwa wie Stärke, der Überschuß-Speicherung dient, sondern eine Zwischenstufe in der Verwertung der Oxydationsenergie darstellt, denn es wird sogleich H2SO4 gebildet.Unter Bedingungen, die in Reinkulturen die stärkste Vermehrung bewirken, kann Beggiatoa wegen des Wettbewerbes in der Natur nicht leben. Die ökologisch besten Lebensumstände sind von besonderer Art und von den physiologisch optimalen verschieden.
The mixotrophy of Beggiatoa
Summary Axenic cultures of Beggiatoa were used to answer the question whether permanent autotrophic growth is feasible, so that oxidation of sulfur compounds would supply energy for all living processes, and organic substances were not needed.For that purpose a new technique was devised with FeS as a source of H2S. Results confirmed the assumption that for Beggiatoa either heterotrophic nutrition or in addition to H2S minute quantities of an organic substrate are required.The colorless Cyanophycean Beggiatoa represents therefore nutritionally a special mode of mixotrophy differing from the nutrition of chemo-autotrophic bacteria.Sulfur as a reserve material does not only serve as a surplus deposit but also represents a step in the utilization of oxidation energy from H2S to H2SO4.Ecologically its living conditions are characterized by low tensions of O2 and H2S, a supply of CO2 and low concentrations of organic nutrients. Under optimum conditions of nutrition. Beggiatoa would not stand competition by other organisms.

White and orange mats are ubiquitous on surface sediments associated with gas hydrates and cold seeps in the Gulf of Mexico. The goal of this study was to determine the predominant pathways for carbon cycling within an orange mat in Green Canyon (GC) block GC 234 in the Gulf of Mexico. Our approach incorporated laser-scanning confocal microscopy, lipid biomarkers, stable carbon isotopes, and 16S rRNA gene sequencing. Confocal microscopy showed the predominance of filamentous microorganisms (4 to 5 μm in diameter) in the mat sample, which are characteristic of Beggiatoa. The phospholipid fatty acids extracted from the mat sample were dominated by 16:1ω7c/t (67%), 18:1ω7c (17%), and 16:0 (8%), which are consistent with lipid profiles of known sulfur-oxidizing bacteria, including Beggiatoa. These results are supported by the 16S rRNA gene analysis of the mat material, which yielded sequences that are all related to the vacuolated sulfur-oxidizing bacteria, including Beggiatoa, Thioploca, and Thiomargarita. The δ13C value of total biomass was −28.6‰; those of individual fatty acids were −29.4 to −33.7‰. These values suggested heterotrophic growth of Beggiatoa on organic substrates that may have δ13C values characteristic of crude oil or on their by-products from microbial degradation. This study demonstrated that integrating lipid biomarkers, stable isotopes, and molecular DNA could enhance our understanding of the metabolic functions of Beggiatoa mats in sulfide-rich marine sediments associated with gas hydrates in the Gulf of Mexico and other locations.  相似文献

A catalase-negative mutant of the yeast Hansenula polymorpha consumed methanol in the presence of glucose when the organism was grown in carbon-limited chemostat cultures. The organism was apparently able to decompose the H2O2 generated in the oxidation of methanol by alcohol oxidase. Not only H2O2 generated intracellularly but also H2O2 added extracellularly was effectively destroyed by the catalase-negative mutant. From the rate of H2O2 consumption during growth in chemostat cultures on mixtures of glucose and H2O2, it appeared that the mutant was capable of decomposing H2O2 at a rate as high as 8 mmol · g of cells−1 · h−1. Glutathione peroxidase (EC 1.11.1.9) was absent under all growth conditions. However, cytochrome c peroxidase (CCP; EC 1.11.1.5) increased to very high levels in cells which decomposed H2O2. When wild-type H. polymorpha was grown on mixtures of glucose and methanol, the CCP level was independent of the rate of methanol utilization, whereas the level of catalase increased with increasing amounts of methanol in the substrate feed. Also, the wild type decomposed H2O2 at a high rate when cells were grown on mixtures of glucose and H2O2. In this case, an increase of both CCP and catalase was observed. When Saccharomyces cerevisiae was grown on mixtures of glucose and H2O2, the level of catalase remained low, but CCP increased with increasing rates of H2O2 utilization. From these observations and an analysis of cell yields under the various conditions, two conclusions can be drawn. (i) CCP is a key enzyme of H2O2 detoxification in yeasts. (ii) Catalase can effectively compete with mitochondrial CCP for hydrogen peroxide only if hydrogen peroxide is generated at the site where catalase is located, namely in the peroxisomes.  相似文献

The coexistence of sulfate-reducing bacteria (SRB) and methanogenic archaea (MA) in anaerobic biofilms developed in sewer inner pipe surfaces favors the accumulation of sulfide (H2S) and methane (CH4) as metabolic end products, causing severe impacts on sewerage systems. In this study, we investigated the time course of H2S and CH4 production and emission rates during different stages of biofilm development in relation to changes in the composition of microbial biofilm communities. The study was carried out in a laboratory sewer pilot plant that mimics a full-scale anaerobic rising sewer using a combination of process data and molecular techniques (e.g., quantitative PCR [qPCR], denaturing gradient gel electrophoresis [DGGE], and 16S rRNA gene pyrotag sequencing). After 2 weeks of biofilm growth, H2S emission was notably high (290.7 ± 72.3 mg S-H2S liter−1 day−1), whereas emissions of CH4 remained low (17.9 ± 15.9 mg COD-CH4 liter−1 day−1). This contrasting trend coincided with a stable SRB community and an archaeal community composed solely of methanogens derived from the human gut (i.e., Methanobrevibacter and Methanosphaera). In turn, CH4 emissions increased after 1 year of biofilm growth (327.6 ± 16.6 mg COD-CH4 liter−1 day−1), coinciding with the replacement of methanogenic colonizers by species more adapted to sewer conditions (i.e., Methanosaeta spp.). Our study provides data that confirm the capacity of our laboratory experimental system to mimic the functioning of full-scale sewers both microbiologically and operationally in terms of sulfide and methane production, gaining insight into the complex dynamics of key microbial groups during biofilm development.  相似文献

Although homoacetogenic bacteria are generally considered to be obligate anaerobes, they colonize the intestinal tracts of termites and other environments that are not entirely anoxic in space or time. In this study, we investigated how homoacetogenic bacteria isolated from the hindguts of various termites respond to the presence of molecular oxygen. All strains investigated formed growth bands in oxygen gradient agar tubes under a headspace of H2-CO2. The position of the bands coincided with the oxic-anoxic interface and depended on the O2 partial pressure in the headspace; the position of the bands relative to the meniscus remained stable for more than 1 month. Experiments with dense cell suspensions, performed with Clark-type O2 and H2 electrodes, revealed a large capacity for H2-dependent oxygen reduction in Sporomusa termitida and Sporomusa sp. strain TmAO3 (149 and 826 nmol min−1 mg of protein−1, respectively). Both strains also reduced O2 with endogenous reductants, albeit at lower rates. Only in Acetonema longum did the basal rates exceed the H2-dependent rates considerably (181 versus 28 nmol min−1 mg of protein)−1). Addition of organic substrates did not stimulate O2 consumption in any of the strains. Nevertheless, reductive acetogenesis by cell suspensions of strain TmAO3 was inhibited even at the lowest O2 fluxes, and growth in nonreduced medium occurred only after the bacteria had rendered the medium anoxic. Similar results were obtained with Acetobacterium woodii, suggesting that the results are not unique to the strains isolated from termites. We concluded that because of their tolerance to temporary exposure to O2 at low partial pressures (up to 1.5 kPa in the case of strain TmAO3) and because of their large capacity for O2 reduction, homoacetogens can reestablish conditions favorable for growth by actively removing oxygen from their environment.  相似文献

Aureobasidium pullulans (de Bary) Arnaud isolated from the phylloplane of sycamore exposed to heavy atmospheric pollution oxidized S0 to S2O32−, S4O62−, and SO42− in vitro. The intermediates S2O32− and S4O62− were also oxidized to SO42−. Cell-free extracts of A. pullulans also oxidized reduced forms of S, the oxidation increasing linearly with increasing protein concentration, showing that the process is enzymatic. The possible role of fungi in S oxidation in soils is discussed.  相似文献

Many industrial activities produce H2S, which is toxic at high levels and odorous at even very low levels. Chemolithotrophic sulfur-oxidizing bacteria are often used in its remediation. Recently, we have reported that many heterotrophic bacteria can use sulfide:quinone oxidoreductase and persulfide dioxygenase to oxidize H2S to thiosulfate and sulfite. These bacteria may also potentially be used in H2S biotreatment. Here we report how various heterotrophic bacteria with these enzymes were cultured with organic compounds and the cells were able to rapidly oxidize H2S to zero-valence sulfur and thiosulfate, causing no apparent acidification. Some also converted the produced thiosulfate to tetrathionate. The rates of sulfide oxidation by some of the tested bacteria in suspension, ranging from 8 to 50 µmol min?1 g?1 of cell dry weight at pH 7.4, sufficient for H2S biotreatment. The immobilized bacteria removed H2S as efficiently as the bacteria in suspension, and the inclusion of Fe3O4 nanoparticles during immobilization resulted in increased efficiency for sulfide removal, in part due to chemical oxidation H2S by Fe3O4. Thus, heterotrophic bacteria may be used for H2S biotreatment under aerobic conditions.  相似文献

The active transport of CO2 in the cyanobacterium Synechococcus UTEX 625 was inhibited by H2S. Treatment of the cells with up to 150 micromolar H2S + HS at pH 8.0 had little effect on Na+-dependent HCO3 transport or photosynthetic O2 evolution, but CO2 transport was inhibited by more than 90%. CO2 transport was restored when H2S was removed by flushing with N2. At constant total H2S + HS concentrations, inhibition of CO2 transport increased as the ratio of H2S to HS increased, suggesting a direct role for H2S in the inhibitory process. Hydrogen sulfide does not appear to serve as a substrate for transport. In the presence of H2S and Na+ -dependent HCO3 transport, the extracellular CO2 concentration rose considerably above its equilibrium level, but was maintained far below its equilibrium level in the absence of H2S. The inhibition of CO2 transport, therefore, revealed an ongoing leakage from the cells of CO2 which was derived from the intracellular dehydration of HCO3 which itself had been recently transported into the cells. Normally, leaked CO2 is efficiently transported back into the cell by the CO2 transport system, thus maintaining the extracellular CO2 concentration near zero. It is suggested that CO2 transport not only serves as a primary means of inorganic carbon acquisition for photosynthesis but also serves as a means of recovering CO2 lost from the cell. A schematic model describing the relationship between the CO2 and HCO3 transport systems is presented.  相似文献

The succession of sulfur-oxidizing bacterial (SOB) community structure and the complex internal sulfur cycle occurring in wastewater biofilms growing under microaerophilic conditions was analyzed by using a polyphasic approach that employed 16S rRNA gene-cloning analysis combined with fluorescence in situ hybridization, microelectrode measurements, and standard batch and reactor experiments. A complete sulfur cycle was established via S0 accumulation within 80 days in the biofilms in replicate. This development was generally split into two phases, (i) a sulfur-accumulating phase and (ii) a sulfate-producing phase. In the first phase (until about 40 days), since the sulfide production rate (sulfate-reducing activity) exceeded the maximum sulfide-oxidizing capacity of SOB in the biofilms, H2S was only partially oxidized to S0 by mainly Thiomicrospira denitirificans with NO3 as an electron acceptor, leading to significant accumulation of S0 in the biofilms. In the second phase, the SOB populations developed further and diversified with time. In particular, S0 accumulation promoted the growth of a novel strain, strain SO07, which predominantly carried out the oxidation of S0 to SO42− under oxic conditions, and Thiothrix sp. strain CT3. In situ hybridization analysis revealed that the dense populations of Thiothrix (ca. 109 cells cm−3) and strain SO07 (ca. 108 cells cm−3) were found at the sulfur-rich surface (100 μm), while the population of Thiomicrospira denitirificans was distributed throughout the biofilms with a density of ca. 107 to 108 cells cm−3. Microelectrode measurements revealed that active sulfide-oxidizing zones overlapped the spatial distributions of different phylogenetic SOB groups in the biofilms. As a consequence, the sulfide-oxidizing capacities of the biofilms became high enough to completely oxidize all H2S produced by SRB to SO42− in the second phase, indicating establishment of the complete sulfur cycle in the biofilms.  相似文献

Four bacteria isolated from peat biofilters, Thiobacillus thioparus DW44, Thiobacillus sp. HA43, Xanthomonas sp. DY44 and Hyphomicrobium sp. I55, were selected to enhance the removal ratios of hydrogen sulfide (H2S), methanethiol (MT) and dimethyl sulfide (DMS) in a mixed gas system. Two bacteria, DW44 and I55, which degrade H2S, MT, DMS and dimethyl disulfide (DMDS), were mixed with DY44 or HA43 which degrade only H2S and MT. Although DMS removal was significantly inhibited by the presence of H2S and MT in a peat biofilter inoculated with the single bacterium, enhanced removability of H2S, MT and DMS was observed by mixing Hyphomicrobium sp. I55 either with Thiobacillus sp. HA43 or Xanthomonas sp. DY44. The removal rate (g-S-kg-dry peat−1·d−1) by I55 after 8 d was 0.664 in total sulfur load, 0.827 g-S·kg-dry g-S·-kg-dry peat−1·d−1, but the rates by the mixed cultures of I55 plus HA43, and I55 plus DY44 were 0.760 and 0.801, respectively. In particular, DMS removability in mixed gases by a mixed culture of I55 and DY44 was almost equivalent to that by I55 when only DMS was supplied, suggesting that removal of H2S and MT, which inhibited DMS removal, was preferentially conducted by DY44 and led to improved DMS removability by I55.  相似文献

Polysulfides formed through the breakdown of elemental sulfur or other sulfur compounds were found to be reduced to H2S by the hyperthermophilic archaebacterium Pyrococcus furiosus during growth. Metabolism of polysulfides by the organism was dissimilatory, as no incorporation of 35S-labeled elemental sulfur was detected. However, [35S]cysteine and [35S]methionine were incorporated into cellular protein. Contact between the organism and elemental sulfur is not necessary for metabolism. The sulfide generated from metabolic reduction of polysulfides dissociates to a strong nucleophile, HS, which in turn opens up the S8 elemental sulfur ring. In addition to H2S, P. furiosus cultures produced methyl mercaptan in a growth-associated fashion.  相似文献

Hydrogen sulfide (H2S), which is produced endogenously from L-cysteine, is an irritant with pro-nociceptive actions. We have used measurements of intracellular calcium concentration, electrophysiology and behavioral measurements to show that the somatic pronociceptive actions of H2S require TRPA1. A H2S donor, NaHS, activated TRPA1 expressed in CHO cells and stimulated DRG neurons isolated from Trpa1+/+ but not Trpa1−/− mice. TRPA1 activation by NaHS was pH dependent with increased activity at acidic pH. The midpoint of the relationship between NaHS EC50 values and external pH was pH 7.21, close to the expected dissociation constant for H2S (pKa 7.04). NaHS evoked single channel currents in inside-out and cell-attached membrane patches consistent with an intracellular site of action. In behavioral experiments, intraplantar administration of NaHS and L-cysteine evoked mechanical and cold hypersensitivities in Trpa1+/+ but not in Trpa1−/− mice. The sensitizing effects of L-cysteine in wild-type mice were inhibited by a cystathionine β-synthase inhibitor, D,L-propargylglycine (PAG), which inhibits H2S formation. Mechanical hypersensitivity evoked by intraplantar injections of LPS was prevented by PAG and the TRPA1 antagonist AP-18 and was absent in Trpa1−/− mice, indicating that H2S mediated stimulation of TRPA1 is necessary for the local pronociceptive effects of LPS. The pro-nociceptive effects of intraplantar NaHS were retained in Trpv1−/− mice ruling out TRPV1 as a molecular target. In behavioral studies, NaHS mediated sensitization was also inhibited by a T-type calcium channel inhibitor, mibefradil. In contrast to the effects of NaHS on somatic sensitivity, intracolonic NaHS administration evoked similar nociceptive effects in Trpa1+/+ and Trpa1−/− mice, suggesting that the visceral pro-nociceptive effects of H2S are independent of TRPA1. In electrophysiological studies, the depolarizing actions of H2S on isolated DRG neurons were inhibited by AP-18, but not by mibefradil indicating that the primary excitatory effect of H2S on DRG neurons is TRPA1 mediated depolarization.  相似文献
