Saccharomyces cerevisiae GL7 cells require exogenous sterol and unsaturated fatty acid for growth. When grown in the presence of cholesterol or 7-dehydrocholesterol, the cells incorporated less saturated fatty acid into phospholipids than cells grown with ergosterol, stigmasterol, or beta-sitosterol as the sterol source. This lower saturated fatty acid content was most pronounced in phosphatidylethanolamine, slightly less so in phosphatidylcholine, and least evident in phosphatidylserine and phosphatidylinositol. Growing the cells with the various sterols did not affect the ratios of individual phospholipids. The ability of strain GL7 to use 7-dehydrocholesterol as the only sterol supplement for growth was dependent upon the nature of the unsaturated fatty acids added to the growth medium. In the presence of linoleic, linolenic, or a mixture of palmitoleic and oleic acids, excellent growth was observed with either ergosterol, cholesterol, or 7-dehydrocholesterol. However, when the medium was supplemented with either oleic or petroselenic acid, the cells grew more slowly (oleic) or much more poorly (petroselenic) with 7-dehydrocholesterol than with ergosterol. A specific relationship between sterol structure and membrane fatty acid composition in yeast cells is implied.  相似文献

Major glyco- and phospholipids as well as betaine lipid 1,2-diacylglycero-O-4'-(N,N,N-tri-methyl)-homoserine (DGTS) were isolated from five species of marine macrophytes harvested in the Sea of Japan in summer and winter at seawater temperatures of 20-23 and 3 degrees C, respectively. GC and DSC analysis of lipids revealed a common increase of ratio between n-3 and n-6 polyunsaturated fatty acids (PUFAs) of polar lipids from summer to winter despite their chemotaxonomically different fatty acid (FA) composition. Especially, high level of different n-3 PUFAs was observed in galactolipids in winter. However, the rise in FA unsaturation did not result in the lowering of peak maximum temperature of phase transition of photosynthetic lipids (glycolipids and phosphatidylglycerol (PG)) in contrast to non-photosynthetic ones [phosphatidylcholine (PC) and phosphatidylethanolamine (PE)]. Different thermotropic behavior of these lipid groups was accompanied by higher content of n-6 PUFAs from the sum of n-6 and n-3 PUFAs in PC and PE compared with glycolipids and PG in both seasons. Seasonal changes of DSC transitions and FA composition of DGTS studied for the first time were similar to PC and PE. Thermograms of all polar lipids were characterized by complex profiles and located in a wide temperature range between -130 and 80 degrees C, while the most evident phase separation occurred in PGs in both seasons. Polarizing microscopy combined with DSC has shown that the liquid crystal - isotropic melt transitions of polar lipids from marine macrophytes began from 10 to 30 degrees C mostly, which can cause the thermal sensitivity of plants to superoptimal temperatures in their environment.  相似文献

Saccharomyces cerevisiae is an attractive chassis for the production of medium-chain fatty acids, but the toxic effect of these compounds often prevents further improvements in titer, yield, and productivity. To address this issue, Lem3 and Sfk1 were identified from adaptive laboratory evolution mutant strains as membrane asymmetry regulators. Co-overexpression of Lem3 and Sfk1 [Lem3(M)-Sfk1(H) strain] through promoter engineering remodeled the membrane phospholipid distribution, leading to an increased accumulation of phosphatidylethanolamine in the inner leaflet of the plasma membrane. As a result, membrane potential and integrity were increased by 131.5% and 29.2%, respectively; meanwhile, the final OD600 in the presence of hexanoic acid, octanoic acid, and decanoic acid was improved by 79.6%, 73.4%, and 57.7%, respectively. In summary, this study shows that membrane asymmetry engineering offers an efficient strategy to enhance medium-chain fatty acids tolerance in S. cerevisiae, thus generating a robust industrial strain for producing high-value biofuels.  相似文献

The influence of the physical state of membrane on L-alanine uptake has been investigated in Saccharomyces cerevisiae KD115, an unsaturated fatty acid auxotrophic mutant. By monitoring the unsaturation index and steady state fluorescence polarization of 1,6 diphenyl hexatriene (DPH), it was observed that at mid log phase the membrane fluidity increased with an increase in the number of double bonds of supplemented fatty acid. Arrhenius plots of the velocities for L-alanine transport in cells grown on palmitoleate, oleate, linoleate and linolenate were biphasic and dependent on supplemented unsaturated fatty acid. Results illustrate a correlation between membrane fluidity and shift in transition points. Further, results confirm the role of fatty acyl milieu in regulation of transport activity of S. cerevisiae.  相似文献

Summary The lipid composition of S. cerevisiae cells was studied in different conditions of pH and of HCO3 - and CO2 concentration.Increasing the concentration of CO2 in the broth determines an increase of total lipids, of total fatty acids, and of the relative amount of unsaturated fatty acids.Changing the concentration of bicarbonates at constant pCO2 does not affect the lipid composition of the cells. Two types of lipid nutritional mutants have been induced in this organism. One of these classes of mutants requires an unsaturated fatty acid and is associated with a locus on chromosome VII. The other class of mutants needs either an unsaturated fatty acid or ergosterol for growth. Experiments involving identification and characterization of these mutants are presented.  相似文献

The fatty acid composition of yeast lipid was manipulated by using auxotrophic strain of S.cerevisiae, KD115, which requires unsaturated fatty acid (UFA) for its growth. It was possible to specifically enrich the yeast with different fatty acyl residues. As compared to wild type strain (S288C), the uptake of amino acids viz., L-alanine, glycine, L-glutamic acid, L-valine in KD115 was drastically reduced, however, the uptake of L-leucine and L-lysine was not affected by the change in lipid unsaturation. Kinetic studies revealed that KT and Jmax values for L-alanine were altered whereas for L-lysine they remained unaffected by UFA modification. Furthermore, unsaturation index for wild type cells was found to be fairly constant while it was variable in KD115 supplemented with different UFAs. It is observed that the variation in amino acid permeases activity which was affected by fluctuations in fatty acyl composition corresponds more to degree of unsaturation rather than growth stage of KD115.  相似文献

One major mechanism of copper toxicity towards microorganisms is disruption of plasma membrane integrity. In this study, the influence of plasma membrane fatty acid composition on the susceptibility of Saccharomyces cerevisiae to Cu2+ toxicity was investigated. Microbial fatty acid composition is highly variable, depending on both intrinsic and environmental factors. Manipulation was achieved in this study by growth in fatty acid-supplemented medium. Whereas cells grown under standard conditions contained only saturated and monounsaturated fatty acids, considerable incorporation of the diunsaturated fatty acid linoleate (18:2) (to more than 65% of the total fatty acids) was observed in both whole-cell homogenates and plasma membrane-enriched fractions from cells grown in linoleate-supplemented medium. Linoleate enrichment had no discernible effect on the growth of S. cerevisiae. However, linoleate-enriched cells were markedly more susceptible to copper-induced plasma membrane permeabilization. Thus, after addition of Cu(NO3)2, rates of cellular K+ release (loss of membrane integrity) were at least twofold higher from linoleate-supplemented cells than from unsupplemented cells; this difference increased with reductions in the Cu2+ concentration supplied. Levels of cellular Cu accumulation were also higher in linoleate-supplemented cells. These results were correlated with a very marked dependence of whole-cell Cu2+ toxicity on cellular fatty acid unsaturation. For example, within 10 min of exposure to 5 microM Cu2+, only 3% of linoleate-enriched cells remained viable (capable of colony formation). In contrast, 100% viability was maintained in cells previously grown in the absence of a fatty acid supplement. Cells displaying intermediate levels of linoleate incorporation showed intermediate Cu2+ sensitivity, while cells enriched with the triunsaturated fatty acid linolenate (18:3) were most sensitive to Cu2+. These results demonstrate for the first time that changes in cellular and plasma membrane fatty acid compositions can dramatically alter microbial sensitivity to copper.  相似文献

Yeast plasma membrane vesicles were obtained by the fusion of liposomes with purified yeast membranes by means of the freeze thaw-sonication technique. Beef heart mitochondria cytochrome-c oxidase was incorporated into the vesicles. Addition of substrate (ascorbate/TMPD/cytochrome c) generated a membrane potential negative inside, and an alkaline pH gradient inside the vesicle, that served as the driving force for leucine transport. Both delta pH and delta psi could drive leucine transport. When delta pH was increased in the presence of valinomycin and potassium, at the expense of delta psi, leucine uptake increased by 10%.  相似文献

Peroxisomes play a major role in human cellular lipid metabolism, including fatty acid β-oxidation. The most frequent peroxisomal disorder is X-linked adrenoleukodystrophy, which is caused by mutations in ABCD1. The biochemical hallmark of X-linked adrenoleukodystrophy is the accumulation of very long chain fatty acids (VLCFAs) due to impaired peroxisomal β-oxidation. Although this suggests a role of ABCD1 in VLCFA import into peroxisomes, no direct experimental evidence is available to substantiate this. To unravel the mechanism of peroxisomal VLCFA transport, we use Saccharomyces cerevisiae as a model organism. Here we provide evidence that in this organism very long chain acyl-CoA esters are hydrolyzed by the Pxa1p-Pxa2p complex prior to the actual transport of their fatty acid moiety into the peroxisomes with the CoA presumably being released into the cytoplasm. The Pxa1p-Pxa2p complex functionally interacts with the acyl-CoA synthetases Faa2p and/or Fat1p on the inner surface of the peroxisomal membrane for subsequent re-esterification of the VLCFAs. Importantly, the Pxa1p-Pxa2p complex shares this molecular mechanism with HsABCD1 and HsABCD2.  相似文献

The sensitivity of Saccharomyces cerevisiae to the redox-active metal copper has recently been found to be influenced by cellular fatty acid composition. This study sought to investigate whether fatty acid composition affected plasma membrane permeabilisation and whole-cell toxicity induced by the redox-inactive metal cadmium. S. cerevisiae NCYC 1383 was enriched with the polyunsaturated fatty acids linoleate (18:2) and linolenate (18:3) by growth in 18:2- or 18:3-supplemented medium. Incorporation of the exogenous fatty acids resulted in them comprising more than 65% of the total fatty acids in plasma membrane lipids. Inhibition of cell division in the presence of Cd(NO3)2 was accentuated by growth in the presence of a polyunsaturated fatty acid. Furthermore, susceptibility to Cd2+-induced plasma membrane permeabilisation increased with the degree of fatty acid unsaturation. Thus, during exposure to Cd2+, K+ efflux from 18:2- and 18:3-enriched cells was up to 2.5-fold or 3-fold greater, respectively than that from unsupplemented cells. In addition, reductions in cell viability during exposure to Cd2+ were most marked in polyunsaturated-fatty-acid-supplemented cells. At certain times, unsupplemented Cd2+-exposed cells displayed up to 7-fold greater viability than supplemented Cd2+-exposed cells. The study demonstrates that the toxicity of the redox-inactive metal Cd2+ towards S. cerevisiae becomes markedly amplified with increased cellular and plasma membrane fatty acid unsaturation. Summary The fatty acid composition of the total lipids and phospholipids of intact and convectively dried yeast has been studied and an increase has been detected in the degree of unsaturation of fatty acids during dehydration. The validity of the inverse relation between the viability of the yeast population during dehydration and rehydration and the degree of unsaturation of the fatty acids has been demonstrated by regression analysis. The activity of thiamine transport in Saccharomyces cerevisiae was decreased by the treatment with tunicamycin without affecting the growth of yeast cells. Although the total activity of a soluble thiamine-binding protein in yeast periplasm, which is known to be a glycoprotein, was decreased by tunicamycin treatment, the activity of thiamine uptake by yeast protoplasts was inhibited as much as by whole cells. Furthermore, tunicamycin decreased the activity of the membrane-bound thiamine-binding protein in a dose dependent way and in parallel with the thiamine transport activity. These findings suggested that the membrane-bound thiamine-binding protein is a glycoprotein which plays a functional role in thiamine transport in S. cerevisiae.  相似文献

Lipoteichoic acid, diglucosyldiacylglycerol, and phosphatidylglycerol isolated from Staphylococcus aureus were embedded in dipalmitoylglycerophosphoglycerol vesicles, and their thermotropic influence on this matrix was studied by differential scanning calorimetry. The natural fatty acids of phosphatidylglycerol effected peak broadening and a decrease in molar heat capacity. These effects were more pronounced with the glycolipid, which also increased the main transition temperature. With the lipoteichoic acid mixtures, two broad main transition peaks were observed, possibly due to different levels of lipoteichoic acid in vesicles. Both peaks showed a further upshift in transition temperatures and a pronounced decrease in molar heat capacity. Since the diacylglycerol moieties of all three amphiphiles were practically identical, the differences in the thermotropic effects have to be ascribed to the different structures of the head groups. Diglucosyldiacylglycerol is proposed to exert an additional effect by hydrogen bonding the hydroxyls of the sugar rings to their phospholipid neighbors. The stronger effect of lipoteichoic acid points to dynamic interactions of the long hydrophilic chain with the vesicle surface, which stabilize the membrane structure.  相似文献

The initiation of growth of a polyaromatic auxotrophic mutant of Saccharomyces cerevisiae was inhibited by several amino acids, whereas growth of the parent prototroph was unaffected. A comparative investigation of amino acid transport in the two strains employing (14)C-labeled amino acids revealed that the transport of amino acids in S. cerevisiae was mediated by a general transport system responsible for the uptake of all neutral as well as basic amino acids. Both auxotrophic and prototrophic strains exhibited stereospecificity for l-amino acids and a K(m) ranging from 1.5 x 10(-5) to 5.0 x 10(-5) M. Optimal transport activity occurred at pH 5.7. Cycloheximide had no effect on amino acid uptake, indicating that protein synthesis was not a direct requirement for amino acid transport. Regulation of amino acid transport was subject to the concentration of amino acids in the free amino acid pool. Amino acid inhibition of the uptake of the aromatic amino acids by the aromatic auxotroph did not correlate directly with the effect of amino acids on the initiation of growth of the auxotroph but provides a partial explanation of this effect.  相似文献

Purified plasma membranes prepared from yeast cells by mechanical agitation with glass beads exhibit no detectable sugar transport activity. However, the addition of phospholipid (asolectin) liposomes to the purified plasma membranes followed by freezing, thawing, and brief sonication produces membrane vesicles which exhibit D-glucose-specific transport activity. The characteristics of zero trans, equilibrium exchange, and influx counterflow exhibited by the membrane vesicles are similar to those of intact cells.  相似文献
