In Argentina, most information on hepatitis C virus (HCV) genotype distribution comes from studies carried out in Buenos Aires (east province). In order to identify HCV subtypes in central Argentina, nucleotide sequencing of core region was performed in samples from 36 patients living in Córdoba, the second most populated province of Argentina. The sequence analysis identified subtype 2c as the most prevalent (50%), followed by subtype 1b (33%) and to a lesser extent by subtypes 1a (11%), 3a (3%) and 4a (3%). This is the first report of circulation of HCV subtype 2c in this region of Argentina and also such high prevalence has never been found before in the genotype distribution of South America.  相似文献

Ras-like (Ral) small GTPases are regulated downstream of Ras and the noncanonical Ral guanine nucleotide exchange factor (RalGEF) effector pathway. Despite RalA and RalB sharing 82% sequence identity and utilization of shared effector proteins, their roles in normal and neoplastic cell growth have been shown to be highly distinct. Here, we determined that RalB function is regulated by protein kinase Cα (PKCα) phosphorylation. We found that RalB phosphorylation on Ser-198 in the C-terminal membrane targeting sequence resulted in enhanced RalB endomembrane accumulation and decreased RalB association with its effector, the exocyst component Sec5. Additionally, RalB phosphorylation regulated vesicular trafficking and membrane fusion by regulating v- and t-SNARE interactions. RalB phosphorylation regulated vesicular traffic of α5-integrin to the cell surface and cell attachment to fibronectin. In summary, our data suggest that phosphorylation by PKCα is critical for RalB-mediated vesicle trafficking and exocytosis.  相似文献

A new heteronuclear NMR pulse sequence for the measurement of nJ(C,H) coupling constants, the /selective HC(C)H-TOCSY, is described. It is shown that the S3E element (Meissner et al., 1997a,b) can be used to obtain spin state selective coherence transfer in molecules, in which adjacent CH moieties are labeled with 13C. Application of the / selective HC(C)H-TOCSY to a 10nt RNA tetraloop 5-CGCUUUUGCG-3, in which the four uridine residues are 13C labeled in the sugar moiety, allowed measurement of two bond and three bond J(C,H) coupling constants, which provide additional restraints to characterize the sugar ring conformation of RNA in cases of conformational averaging.  相似文献

Ghosts prepared from rat basophilic leukemia cells (RBL cell ghosts) and permeabilized with -toxin fromS. aureus are a simplified system for the study of FcRI-mediated activation of phospholipase C (PLC). This activity is dependent upon ATP and magnesium, and is enhanced by the addition of another compound containing an energetic phosphate group, either phosphoenolpyruvate (PEP) or phosphocreatine (PCr). This effect appears to be specific for PEP and PCr in that other compounds with energetic phosphate bonds including fructose 1,6-bisphosphate and additional ATP are not effective. On the contrary, GTP--S, an activator of G proteins, activates PLC in the presence of ATP alone and this is not further enhanced by the addition of PEP. In addition to FcRI and GTP--S, two other stimuli lead to enhanced activity of PLC in permeabilized RBL cell ghosts: 1) an inhibitor of tyrosine phosphatases (Na3VO4) and 2) an analog of adenosine (NECA). Data presented here extend previous results to show that activation of PLC by GTP--S is not enhanced either by the addition of PCr or by the addition of a more MgATP. Further new findings include the observations that activation of PLC by Na3VO4 is augmented by PEP and PCr in a fashion similar to that observed for FcRI-mediated activation of PLC and that activation of PLC by NECA shows even more marked dependency on PEP than does activation by FcRI or Na3VO4. Oxygen inhibition of leaf slice photosynthesis in Panicum milioides increased from 20% to 30% at 21% O2 in the presence of maleate, a phosphoenolpyruvate carboxylase inhibitor. The increased O2 sensitivity was completely reversed by the addition of malate and aspartate, the stable products of the phosphoenolpyruvate carboxylase reaction. The C4 acids, malate and aspartate, also reduced O2 inhibition of photosynthesis by isolated bundle sheath strands, but not mesophyll protoplasts. Similarly, only bundle sheath strands exhibited an active C4 acid-dependent O2 evolution. Compartmentation of C4 cycle enzymes, with pyruvate, Pi dikinase in the mesophyll and NAD-malic enzyme in the bundle sheath, was demonstrated. It is concluded that reduced photorespiration in P. milioides is due to a limited potential for C4 photosynthesis permitting an increase in pCO2 at the site of bundle sheath ribulosebisphosphate carboxylase.  相似文献

A [CO]HN(CA)CB-E.COSY pulse scheme is described for measurement of three-bond couplings, 3JCC, between carbonyl and aliphatic C carbons in ubiquitin, uniformly enriched with 13C and 15N. A Karplus relation, 3JCC = 1.28 cos2( - 120°) -1.02 cos( - 120°) +0.30 Hz, is obtained by correlating the 3JCC values measured for human ubiquitin with backbone angles from its crystal structure. As predicted, the new Karplus parametrization yields 3JCC values slightly larger than previously obtained by quantitative J correlation [Hu, J.-S. and Bax, A. (1997) J. Am. Chem. Soc., 119, 6360-6368], but considerably smaller than what has been reported on the basis of other E.COSY-type measurements carried out on flavodoxin.  相似文献

A series of 4-anilinoquinazolines with C–C multiple bond substitutions at the 6-position were synthesized and investigated for their potential to inhibit epidermal growth factor receptor (EGFR) tyrosine kinase activity. Among the compounds synthesized, alkyne 6d and allenes 7d and 7f significantly inhibited EGFR tyrosine kinase activity. These compounds inhibited EGF-mediated phosphorylation of EGFR in A431 cells, resulting in cell-cycle arrest and apoptosis induction. The C–C multiple bonds substituted at the C-6 position of the anilinoquinazoline framework were essential for the significant inhibitory activity. Compounds with long carbon chains (n = 3–6), such as 6c–f, 7c–f, 11, and 12, displayed prolonged inhibitory activity.  相似文献

The physiological roles of omega-3 fatty acids, eicosapentaenoic acid and docosahexaenoic acid have been investigated in detail and microbial strains producing these polyunsaturated fatty acids have been characterised. It has recently been suggested that docosapentaenoic acid may have an important role, especially in infant nutrition, and that its positive health effects have been overlooked. Docosapentaenoic acid (C22:5, ω-3) production by a strain of Pythium acanthicum ATCC 18660 was thus investigated. Optimum conditions for growth of P. acanthicum ATCC 18660 and docosapentaenoic acid production were: pH 6.0, temperature 20°C and incubation time, 10 days. Among different saccharides and complex nitrogen sources tested, glucose and sodium glutamate were preferred carbon and nitrogen sources, respectively. Maximum biomass content (10.4 g L−1) and docosapentaenoic acid yield (49.9 mg L−1) were obtained in 10 days. An increase in docosapentaenoic acid volumetric yields to 108–110 mg L−1 was obtained when linseed oil was used to supplement glucose or soy flour-containing medium. Batch feeding of additional glucose or linseed oil further enhanced the docosapentaenoic acid volumetric yield to 132 mg L−1 and 125 mg L−1, respectively, in 14 days. The specific production of docosapentaenoic acid in preliminary experiments ranged from 1.0–5.0 mg g−1 biomass. As conditions were optimised, docosapentaenoic acid specific production titers were generally in the range of 4.0–5.5 mg g−1 and increases in docosapentaenoic acid volumetric production could be attributed to increased biomass production. The limited improvement obtained by modifying culture conditions indicates that increasing volumetric yields of docosapentaenoic acid by modifying culture conditions appears to represent a significant barrier to commercialisation of such a process and suggests a more fundamental manipulation of metabolism and physiology is required. -N-oxalyl-l-,-diaminopropionic acid (l-ODAP) toxicity has been associated with lathyrism; a spastic paraparesis caused by excessive dietary intake of the pulse Lathyrus sativus. We investigated the effect of Lathyrus neurotoxin l-ODAP on protein kinase C (PKC) activity under in vitro conditions. l-ODAP activated phosphorylation activity of purified chick brain PKC. Both lysine-rich (histone III-S) and arginine-rich (protamine sulfate) substrate phosphorylation was enhanced in the presence of l-ODAP. The activation is concentration dependent, and maximal activation is observed at 100 M concentration. Protamine sulfate phosphorylation was enhanced by 47%, whereas histone III-S phosphorylation was enhanced by 50% over PS/PDBu/Ca2+ dependent activity. The nontoxic d-isomer (d-ODAP) did not affect both histone III-S and protamine sulfate phosphorylation activity. These results indicate that l-ODAP taken up by neuronal cells could also contribute to PKC activation and so be associated with toxicity.  相似文献

Ginsenoside Re, one of the main constituents of Panax ginseng, possesses novel antioxidant and anti-inflammatory properties. However, the pharmacological mechanism of ginsenoside Re in dopaminergic degeneration remains elusive. We suggested that protein kinase C (PKC) δ mediates methamphetamine (MA)-induced dopaminergic toxicity. Treatment with ginsenoside Re significantly attenuated methamphetamine-induced dopaminergic degeneration in vivo by inhibiting impaired enzymatic antioxidant systems, mitochondrial oxidative stress, mitochondrial translocation of protein kinase Cδ, mitochondrial dysfunction, pro-inflammatory microglial activation, and apoptosis. These protective effects were comparable to those observed with genetic inhibition of PKCδ in PKCδ knockout (?/?) mice and with PKCδ antisense oligonucleotides, and ginsenoside Re did not provide any additional protective effects in the presence of PKCδ inhibition. Our results suggest that PKCδ is a critical target for ginsenoside Re-mediated protective activity in response to dopaminergic degeneration induced by MA.  相似文献

The assimilation of 14CO2 into the C4 acids malate and aspartate by leaves of C3, C4 and C3–C4 intermediate Flaveria species was investigated near the CO2 compensation concentration * in order to determine the potential role of phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) in reducing photorespiration in the intermediates. Relative to air concentrations of CO2, the proportion of CO2 fixed by PEP carboxylase at * increased in all six C3–C4 intermediate species examined. However, F. floridana J.R. Johnston and F. ramosissima Klatt were shown to be markedly less responsive to reduced external CO2, with only about a 1.6-fold enhancement of CO2 assimilation by PEP carboxylase, as compared to a 3.0- to 3.7-fold increase for the other C3–C4 species examined, namely, F. linearis Lag., F. anomala B.L. Robinson, F. chloraefolia A. Gray and F. pubescens Rydb. The C3 species F. pringlei Gandoger and F. cronquistii A.M. Powell exhibited a 1.5- and 2.9-fold increase in labeled malate and aspartate, respectively, at *. Assimilation of CO2 by PEP carboxylase in the C4 species F. trinervia (Spreng.) C. Mohr, F. australasica Hook., and the C4-like species F. brownii A.M. Powell was relatively insensitive to subatmospheric levels of CO2. The interspecific variation among the intermediate Flaverias may signify that F. floridana and F. ramosissima possess a more C4-like compartmentation of PEP carboxylase and ribulose-1,5-bisphosphate carboxylase/oxygenase (EC 4.1.1.39) between the mesophyll and bundle-sheath cells. Chasing recently labeled malate and aspartate with 12CO2 for 5 min at * resulted in an apparent turnover of 25% and 30% of the radiocarbon in these C4 acids for F. ramosissima and F. floridana, respectively. No substantial turnover was detected for F. linearis, F. anomala, F. chloraefolia or F. pubescens. With the exception of F. floridana and F. ramosissima, it is unlikely that enhanced CO2 fixation by PEP carboxylase at the CO2 compensation concentration is a major mechanism for reducing photorespiration in the intermediate Flaveria species. Moreover, these findings support previous related 14CO2-labeling studies at air-levels of CO2 which indicated that F. floridana and F. ramosissima were more C4-like intermediate species. This is further substantiated by the demonstration that F. floridana PEP carboxylase, like the enzyme in C4 plants, undergoes a substantial activation (2.2-fold) upon illuminating dark-adapted green leaves. The biomass production and antioxidant accumulation of the aerial microalga Trentepohlia aurea were examined. Using ammonium chloride as nitrogen source for growth in liquid medium, the growth rate was exponential and accompanied by a marked fall in the pH of the medium. The highest growth rate of 152 mg L-1 day-1 (logarithmic phase) was attained when T. aurea was cultured with aeration of 5% CO2-enriched air in medium buffered with HEPES. The growth rate and antioxidant content were enhanced under 5% CO2 by switching the light intensity from 43 to 143 μmol photon m-2 s-1 for the two-stage culture. As a result, T. aurea cells accumulated 2.1 mg β-carotene, 0.3 mg L-ascorbic acid and 2.4 mg tocopherols, respectively, per g dry cell. The simultaneous production of useful materials, such as β-carotene, vitamin E and vitamin C, was demonstrated. During December and January, dry northeasterly surface winds (the Harmattan)distribute dust over West Africa. Rate of deposition and some chemical andphysical characteristics of Harmattan dust were measured in Taï NationalPark in the southwest corner of Côte d'Ivoire during the 1990–1991 dryseason. The dust deposition was estimated by the classical water-filled basinmethod and by using canopy drip to account for deposition on tree canopies.Contamination by local biotic debris in both, water-filled basin and canopydrip collectors, was corrected for by using Ti (which appears to be wholly ofatmospheric origin) as a reference element. Harmattan dust in Taï consistedmainly of kaolinitic silt finer than that collected in North Nigeria, closer tothe source area in the Chad basin. The estimates of seasonal deposition rateswere 33 to 47 kg ha-1 for the water-filled basin method andaround 80 kg ha-1 for the canopy drip method. The higher valuein canopy drip was in agreement with expected higher deposition of fine duston the canopies than on a water surface, and was therefore considered morereliable to estimate nutrient inputs by Harmattan dust deposition. The seasonalnutrient input by dust was thus estimated to be 0.11 kg ha-1for P, 2.5 kg ha-1 for K, 3.5 kg ha-1 for Caand 0.4 kg ha-1 for Mg.  相似文献

Phosphoenolpyruvate carboxylase (PEPCase; EC 4.1.1.31) from Flaveria trinervia Mohr (C4), F. floridana Johnston (C3–C4), and F. cronquistii Powell (C3) leaves were compared by electrotransfer blotting/enzyme-linked immunoassay (Western-blot analysis), mobility of the native enzyme in polyacrylamide gels and in isoelectric focusing (IEF) gels, peptide mapping, and in-vitro translation of RNA isolated from each plant. The PEPCases from the C3 and C3–C4 plants were very similar to each other in terms of electrophoretic mobilities on gels and isoenzyme patterns on IEF gels, and identical in peptide mapping. Quantitative differences were noted, however, in that the C3–C4 intermediate plant contained more PEPCase overall and that the relative activity of individual isoenzymes shifted between the C3 and C3–C4 intermediate PEPCases. The PEPCase from the C4 plant had a different isoenzyme pattern, a different peptide map, and was far more abundant than the other two enzymes. Western blot analysis demonstrated the cross-reactivity of PEPCases from all three Flaveria species with antibody raised against maize PEPCase. Summary Ammonium and asparagine produced a concentration-dependent reduction of cephamycin C biosynthesis by Streptomyces lactamdurans. Addition of ammonium salts at 1 mM concentration reduced cephamycin biosynthesis by resting cells of S. lactamdurans, whereas concentrations of asparagine above 10 mM were required to get the same effect. High ammonium concentrations decreased glutamine synthetase activity in cell extracts of S. lactamdurans in parallel to the reduction of antibiotic biosynthesis. Ammonium supplementation decreased the pool of glutamic acid and glutamine whereas the intracellular content of ammonium, alanine, and phosphoserine increased significantly. The pool of the tripeptide (l--aminoadipyl)-l-cysteinyl-d-valine, an intermediate in cephamycin biosynthesis, was greatly reduced in ammonium-supplemented cultures. Isopenicillin N synthetase, that converts the tripeptide (l--aminoadipyl)-l-cysteinyl-d-valine to isopenicillin N, isopenicillin N isomerase (that isomerises isopenicillin N to penicillin N) and deacetoxycephalosporin C synthetase (converting penicillin N into deacetoxycephalosporin C) were also reduced in ammonium-supplemented cultures. However, the activities of these enzymes were not inhibited in vitro by 40 mM ammonium, suggesting that the enzymes were repressed but not inhibited by ammonium in vivo.  相似文献
