The cell has been represented as a charged liquid drop. Contrary to the DLVO-theory, the effect of the surface potential upon the value of the interfacial tension of the cell membrane has also been taken into consideration. The cell membrane has visco-elastic properties and its constituents may move against each other. Cell movement is caused by the appearance of a small number of the electrically charged constituents of the cell membrane on the leading edge of the cell. This produces a local decrease in the surface tension and the cell membrane expansion. At the moment of contact between two cells proton transfers occur between the strongly negatively charged microvilli of one cell and the body of the other, analogous to a condenser breakdown. This, through the effect on the surface tension, causes contact inhibition of movement. The distribution of the proton dissociable groups modifies the interaction between the cells (differentiation) and between the cell and the substratum (adhesion). Adsorption of the charged compounds at the surface of the cell membrane, decreasing the surface potential and increasing the surface tension, causes the phenomena of chemotaxis, phagocytosis and pinocytosis. Cell division, considered in the terms of the surface energy, requires an adequate supply of considerable quantities of energy inversely proportional to the surface potential value. In case of a reduction of the distance between the cells, their surface potential and the energetic barrier of the cell division processes increases, and causes contact inhibition of cell division. Due to their high charge, division of neoplastic cells is inhibited much later than division of normal cells, or is completely ininhibited due to geometric conditions. Fusion of the cell membrane in the intra-cellular and intercellular processes is a reverse process in relation to the cell division.  相似文献

A review is presented of issues relevant to the definition, measurement, and classification of stimuli, situations, and environments. Problems such as the lack of adequate definitions of concepts, error and bias in measurement procedures, confusion between measurement of a concept and measurement of its behavioral effects, and the lack of agreement among alternative measures are emphasized. It is suggested that concepts be defined in terms of objective characteristics while allowing for the study of the transactional relationship between organism and environment. The work of the ethologists in defining stimuli while studying their relationship to different organismic states and situational contexts is emphasized in this regard. Following Brunswik, it is also suggested that wherever possible there be a representative sampling of variables in natural settings. Note from the editors: From time to time, Human Ecology will publish a review article. Our first in this series is a review by a psychologist of basic definitional and conceptual problems in environmental studies.This paper was prepared while the author was a Visiting Research Fellow at the Educational Testing Service. The aim of this report has been to present results concerning analytical quality controls of Hg analysis of fish and sediment, analyses of Fe, Ca, total-P, K, pH, alkalinity, conductivity, colour and hardness (Ca + Mg) of lake water samples. Despite the fact that these are standard parameters in many regular water control programs, there are major differences in the reliability with which these parameters can be determined. The focus here is on an overall inter-laboratory comparison between the parameters. Six laboratories have been involved in the analysis. Selected results: pH gives the lowest (average) relative standard deviation (error), about 2 %; conductivity gives an error of about 5–7 %; alkalinity yields an average error of as much as 13–25 %, which is the largest among the parameters studied here; colour also gives a high error, 9–15 %; hardness gives a relative standard deviation of about 6–7 %. Of the other parameters (i. e., Hg, Fe, Ca and P), Hg gives the best reliability and Fe and P the lowest. To have knowledge of the reliability of the analytical data is of paramount importance in most control programs and research projects.  相似文献

A chemiluminescent method is described for the determination of tetracycline, chlortetracycline, minocycline, doxycycline, and demeclocycline. The method is based on the photon counting technique and the spline functions approximation. The simple formula S = b + Ax(2) is proposed for the observed dependency of the integrated number of countings S on the concentration x of a given antibiotic. Phenylpropynal is a specific, irreversible, non-beta-lactam inhibitor of typical beta-lactamases. In the presence of millimolar concentrations of phenylpropynal, the beta-lactamase I of Bacillus cereus and the beta-lactamases of Staphylococcus aureus and Escherichia coli become completely inactivated; the beta-lactamase II of B. cereus is not affected. The E. coli beta-lactamase is considerably more sensitive to the reagent than the gram-positive enzymes. A variety of structural analogs of phenylpropynal are much less effective inhibitors. Bovine alpha-chymotrypsin, bovine carboxypeptidase A, and the D,D-carboxypeptidase/transpeptidase of Streptomyces R-61 were not inactivated by phenylpropynal. The inactivation of the E. coli beta-lactamase can be significantly retarded when the good substrate benzylpenicillin is also present. The development of a characteristic chromophore (lambda max 318 nm) during beta-lactamase inactivation suggests that covalent modification of the enxymes is involved; arginine and/or lysine modification is indicated.  相似文献

The evidence for amphibian population declines is based on count data that were not adjusted for detection probabilities. Such data are not reliable even when collected using standard methods. The formula C = Np (where C is a count, N the true parameter value, and p is a detection probability) relates count data to demography, population size, or distributions. With unadjusted count data, one assumes a linear relationship between C and N and that p is constant. These assumptions are unlikely to be met in studies of amphibian populations. Amphibian population data should be based on methods that account for detection probabilities.  相似文献

Circadian rhythms with a period of ∼24 h, are natural timing machines. They are broadly distributed in living organisms, such as Neurospora, Drosophila, and mammals. The underlying natures of the rhythmic behavior have been explored by experimental and theoretical approaches. However, the global and physical natures of the oscillation under fluctuations are still not very clear. We developed a landscape and flux framework to explore the global stability and robustness of a circadian oscillation system. The potential landscape of the network is uncovered and has a global Mexican-hat shape. The height of the Mexican-hat provides a quantitative measure to evaluate the robustness and coherence of the oscillation. We found that in nonequilibrium dynamic systems, not only the potential landscape but also the probability flux are important to the dynamics of the system under intrinsic noise. Landscape attracts the systems down to the oscillation ring while flux drives the coherent oscillation on the ring. We also investigated the phase coherence and the entropy production rate of the system at different fluctuations and found that dissipations are less and the coherence is higher for larger number of molecules. We also found that the power spectrum of autocorrelation functions show resonance peak at the frequency of coherent oscillations. The peak is less prominent for smaller number of molecules and less barrier height and therefore can be used as another measure of stability of oscillations. As a consequence of nonzero probability flux, we show that the three-point correlations from the time traces show irreversibility, providing a possible way to explore the flux from the observations. Furthermore, we explored the escape time from the oscillation ring to outside at different molecular number. We found that when barrier height is higher, escape time is longer and phase coherence of oscillation is higher. Finally, we performed the global sensitivity analysis of the underlying parameters to find the key network wirings responsible for the stability of the oscillation system.  相似文献

Leucine-rich repeats (LRRs) with 20-30 amino acids in unit length are present in many proteins from prokaryotes to eukaryotes. The LRR-containing proteins include a family of nine small proteoglycans, forming three distinct subfamilies: class I contains biglycan/PG-I and decorin/PG-II; class II: lumican, fibromodulin, PRELP, keratocan, and osteoadherin; and class III: epiphycan/PG-Lb and osteoglycin or osteoinductive factor. Comparative sequence analysis of the 34 available protein sequences reveals that these proteoglycans have two types of LRRs, which we call S and T. The type S LRR is 21 residues long and has the consensus sequence of xxaPzxLPxxLxxLxLxxNxI. The type T LRR has 26 residues; its consensus sequence is zzxxaxxxxFxxaxxLxxLxLxxNxL. In both "x" indicates variable residue; "z" is frequently a gap; "a" is Val, Leu, or Ile; and I is Ile or Leu. These type S and TLRRs are ordered into two super-motifs--STT with about 73 residues in classes I and II and ST with about 47 residues in class III. The 12 LRRs in the small proteoglycans of I and II are best represented as (STT)4; the seven LRRs of class III as (ST)T(ST)2. Our analyses indicate that classes I/II and III evolved along different paths after the establishment of the precursor ST, and classes I and II also diverged after the establishment of the precursor (STT)4.  相似文献

对15a来王港潮滩重金属积累规律和污染状况进行了研究,第1次测到了王港地区210Pb的本底值为1.16dpm/g,校正了前人对王港地区沉积速率的研究。尝试性地把分形理论引入到环境压力的定量研究中。研究表明,王港潮滩现代平均沉积速率为4.13cm/a;从重金属相关性分析得出:王港潮滩Fe、Zn与Li相关系数非常显著,归一化后,Cu、Zn的归一化值在15a内相对稳定,其中Cu的波动范围在0.4~2之间,Zn在1.5~2.6之间狭小的范围内波动。分形回归分析结果表明,王港潮滩重金属Cu、Zn归一化值的信息维数高,系统的结构松散,自组织程度低。Cu的分维值范围在5~6.5,平均值为5.8085,Zn的分维值范围在在3~6.5,平均值为4.68。王港潮滩重金属的人为污染较轻,主要受地球化学元素的控制;同时,Pb的归一化值的分维值较低,其分维值范围在2~5.5,平均值为3.608,自组织程度较高,说明存在一定程度的Pb污染。  相似文献

1. The blood of Chortophaga viridifasciata was analyzed. The average concentrations of inorganic cations expressed as milligrams per cent are: sodium, 250.66; potassium, 13.52; calcium, 11.40; and magnesium, 51.15. The osmotic pressure of the blood at 0°C. is 10.7 atmospheres. Protein and non-protein nitrogen, expressed as milligrams per cent, are 253.4 and 140.0, respectively. 2. The blood of Samia walkeri has an osmotic pressure of 13.36 atmospheres at 0°C. Its protein nitrogen is 628.58, and its non-protein nitrogen, 441.20 milligrams per cent. 3. The effects of isotonic chloride solutions of sodium, potassium, calcium, and magnesium and of distilled water on the heart beat of these two species were determined. The heart of the grasshopper failed to beat in isotonic solutions of KCl, MgCl2, or in distilled water. For both insects, sodium was found to be the least toxic ion. In the case of the grasshopper, calcium ranks next in order. In the case of the moth, potassium ranks next after sodium and is followed by calcium and magnesium. 4. The ratio of sodium to potassium in milligrams per cent, necessary for maintaining the normal heart beat of Chortophaga viridifasciata is 3 to 1, but it may be increased to at least 34 to 1 without any appreciable effects. The ratio of potassium to calcium necessary for maintaining the normal heart beat of this insect is 1 to 1, and may be increased to as much as 3 to 1. 5. The ratio of sodium to potassium, in milligrams per cent, necessary for maintaining the normal heart beat of Samia walkeri was found to be equal to or to exceed 1 to 13.8. The sodium content may be increased so that the ratio of sodium to potassium is 34 to 1 without any toxic effects. The ratios of potassium to calcium required for normal heart beat in this insect may be 1 to 1, 2 to 1, or 3 to 1. 6. The hearts of the grasshoppers beat normally in isotonic solutions having an osmotic pressure of 10.7 atmospheres. They beat equally well in solutions having an osmotic pressure of 13.4 atmospheres. The hearts of the cynthia pupae beat normally in isotonic solutions having an osmotic pressure of 13.36 atmospheres. However, they also beat normally in solutions having an osmotic pressure of 10.02 atmospheres. Therefore, although the blood of the cynthia moth and of the grasshopper have different osmotic pressures, their hearts are tolerant to solutions having the same tonicity. Because of this, and since the ratios of potassium to calcium necessary for maintaining normal heart beats of both insects are the same, solutions favorable to the grasshopper may also be favorable to the cynthia moth.  相似文献

Xishuangbanna of southern Yunnan is biogeographically located at a transitional zone from tropical Southeast (SE) Asia to subtropical East Asia, and is at the junction of the Indian and Burmese plates of Gondwana and the Eurasian plate of Laurasia. The flora of the region consists of a recorded 3336 native seed plant species, belonging to 1140 genera in 197 families, among which 83.5 percent are tropical genera and 32.8 percent are endemic to tropical Asia, suggesting a strong affinity to tropical Asian flora. The vegetation of Xishuangbanna is organized into four forest types: tropical rain forest, tropical seasonal moist forest, tropical montane evergreen broad‐leaved forest, and tropical monsoon forest. The tropical rain forest in Xishuangbanna has the same floristic composition of families and genera as some lowland equatorial rain forests in SE Asia, and is dominated (with a few exceptions) by the same families both in species richness and stem dominance. The exceptions include some deciduous trees in the canopy layer, fewer megaphanerophytes and epiphytes, and a higher abundance of lianas and microphyllic plants. We consider the tropical rain forest of Xishuangbanna as a type of tropical Asian rain forest, based on their conspicuous similarities in ecological and floristic characteristics.  相似文献

The ectodermal eyes, 45–55 μm in diameter, of the cnidarian hydrozoan Cladonema radiatum Dujardin possess a lens approximately 15 μm in diameter enveloped by an eyecup (retina). An overlying layer of intensely vacuolated distal process of the adjoining epithelial cells forms a transparent cornea. The eyecup is composed of three cell types: basal cells, melanin-containing pigment cells, and photoreceptor cells. The last two cell types occur in the ratio of approximately 2:1. Histogenesis of the eye both during ontogeny and regeneration is described from light and electron microscopic investigations. During ontogeny the cell types forming the retina are derived from a compact group of morphologically undifferentiated cells, but during regeneration a primordium is formed by regeneration cells. In both cases the lens is built from distal nonnucleated cytoplasmic portions pinched off from the pigment cells. The cornea is formed by distal lamellar processes of the ocellus adjoining the epithelial cells. Through EM-histochemical methods (silver impregnation and DOPA-oxidase reaction) the pigment of the chromatophores of the retina was identified as melanin.  相似文献

Homogenates of Musca domestica (housefly) larvae contain glycerophosphodiesterase activity, which is found in the supernatant fluid after centrifugation at 88,000 g. The phosphodiesterase is inhibited by EDTA and is stimulated by Mg(2+), Ni(2+), Co(2+), and Mn(2+). The pH optimum is 7.2. The enzyme is stable to heating at 50 degrees C for 15 min and is insensitive to sulfhydryl inhibitors. Glycerophosphoryl diesters of choline, ethanolamine, inositol, serine, glycerol, and beta-methylcholine are hydrolyzed to the common product, l-alpha-glycerophosphate, and the appropriate free alcohol. The rate of glycerophosphorylcholine hydrolysis is 70% greater than the rate of hydrolysis of the other glycerophosphodiesters. Apparent K(m) values for glycerophosphorylcholine, glycerophosphorylethanolamine, and glycerophosphoryl-beta-methylcholine are 2-4 x 10(-4) m, and for glycerophosphorylinositol, 2 x 10(-3) m. Competitive studies using various pairs of substrates, as well as the exchange of free choline into both glycerophosphorylcholine and glycerophosphorylinositol, suggest that a single enzyme cleaves all substrates. Product inhibition and reversal of the reaction were not detected. Choline, but not l-alpha-glycerophosphate, exchanges into glycerophosphorylcholine and glycerophosphorylinositol.  相似文献

Glycerol kinase (EC 2.7.1.30) is a bacterial sugar kinase and a member of the sugar kinase/actin/hsc-70 superfamily of enzymes. The enzyme from Escherichia coli is an allosteric regulatory enzyme whose activity is inhibited by fructose 1,6-bisphosphate (FBP) and the glucose-specific phosphocarrier of the phosphoenolpyruvate:glycose phosphotransferase system, IIA(Glc) (previously termed III(Glc)). Comparison of its primary structure with that of the highly similar Haemophilus influenzae glycerol kinase reveals that the amino acid sequence for the binding site for FBP is conserved while the amino acid sequence for the binding site for IIA(Glc) contains differences that are predicted to prevent its inhibition. To test this hypothesis, the H. influenzae glpK gene was assembled from DNA library fragments and subcloned into pUC18. The enzyme is expressed at high levels in E. coli. It was purified to greater than 90% homogeneity by taking advantage of its solubility behavior in a procedure that requires no column chromatography. The initial-velocity kinetic parameters of the purified enzyme are similar to those of the E. coli glycerol kinase. The H. influenzae glycerol kinase is inhibited by FBP but not by IIA(Glc), in agreement with the prediction based on sequence comparison. Sedimentation velocity experiments reveal that inhibition of HiGK by FBP is associated with oligomerization, behavior which is similar to EcGK. The possibility of utilizing mutagenesis studies to exploit the high degree of similarity of these two enzymes to elucidate the mechanism of allosteric regulation by IIA(Glc) is discussed.  相似文献

Background and aims

The ionome (elemental composition) of grassland species has rarely been studied at the level of individual organs and little is known about effects of soil chemical properties on the ionome. Using the model oxalate plant Rumex obtusifolius, we asked how its biomass production and the distribution of elements between its organs is affected by soil chemical properties.

Methods

We established a pot experiment with R. obtusifolius planted in acidic non-contaminated control and in slightly acidic and alkaline soils anthropogenically contaminated by the risk elements As, Cd, Pb, and Zn. Both contaminated soils were untreated and treated by lime and superphosphate. We determined biomass production and the concentrations of elements in its organs.

Results

Biomass production was negatively related to the mobility of micro- and risk elements. Restricted transport of micro- and risk elements from belowground organs into leaves was recorded in untreated contaminated soils. In both lime-treated soils and in superphosphate-treated alkaline soil, elevated transport of micro- and risk elements from belowground organs into leaves was recorded in comparison to untreated contaminated soils. The lowest concentrations of micro- and risk elements were recorded in stems and seeds, followed by belowground organs and leaves.

Conclusions

R. obtusifolius is an As-, Cd-, Pb-, and Zn-excluder and is sensitive to high availability of micro- and risk elements in the soil. Soil chemical properties affect the distribution of essential elements within the plant greatly.  相似文献

Phytohormones, including auxins, abscisic acid, brassinosteroids, cytokinins, ethylene, gibberellins, and jasmonates, are involved in all aspects of plant growth, and developmental processes as well as environmental responses. However, our understanding of hormonal homeostasis is far from complete. Phytohormone conjugation is considered as a part of the mechanism to control cellular levels of these compounds. Active phytohormones are changed into multiple forms by acylation, esterification or glycosylation, for example. It seems that conjugated compounds could serve as pool of inactive phytohormones that can be converted to active forms by de-conjugation reactions. Some conjugates are thought to be temporary storage forms, from which free active hormones can be released after hydrolysis. It is also believed that conjugation serves functions, such as irreversible inactivation, transport, compartmentalization, and protection against degradation. The nature of abscisic acid, brassinosteroid, ethylene, gibberellin, and jasmonate conjugates is discussed.  相似文献

A rapid and sensitive method for separation and concurrent assay of 14 compounds at the picomole level in individual rat brain parts is described. The putative amino acid neurotransmitters (aspartate, γ-aminobutyric acid, glutamate, and glycine) are extracted from a 20–30 mg portion of the tissue with 5% TCA and assayed as their respective DNP-amino acid methyl ester derivatives by glc. Four other putative neurotransmitters (acetylcholine, dopamine, norepinephrine, and serotonin) and some of their precursors and metabolites (choline, tryptophan, 5-hydroxytryptophan, 5-hydroxyindoleacetic acid, and tyrosine) are extracted in 1n formic acid-acetone (v/v:15/85) from the remaining tissue. The lipids are removed with a heptane-chloroform (v/v:8/1) wash and the aqueous phase is dried at 37°C under N2. The dried extract is dissolved in water (pH 4). With one portion of this solution, acetylcholine and choline are assayed using a radioenzymatic method whereas with the rest, dopamine, norepinephrine, serotonin, 5-hydroxyindoleacetic acid, 5-hydroxytryptophan, tryptophan, and tyrosine are separated with three ion exchange resins arranged in tandem. These compounds are assayed fluorometrically with modified microadaptations of previously reported methods.  相似文献

采用临界值法对尾叶桉幼林材积生长进行叶片营养诊断,试验结果表明,氮,磷,钾,钙,镁和硼等营养元素的临界浓度分别为15.3g/kg,1.2g/kg,4.2g/kg,16.1g/kg,2.5g/kg和0.019g/kg;最适浓度范围分别为15.3-18.1g/kg,1.2-1.7g/kg,4.2-5.6g/kg,16.1-19.8g/kg,2.5-3.0g/kg和0.019-0.031g/kg,而对树高生长进行营养诊断时,上选结果稍微有些变化,试验还得到各营养元素比值的临界值和最适范围。  相似文献

The virion-extracted DNA (Mr5 x 10(6)) of cauliflower mosaic virus (CaMV) has three single-stranded interruptions. The mapping of this DNA using eleven restriction endonucleases (HhaI, SacI, AvaI, PvuII, PstI, XbaI, EcoRI, Bg/II, HincII, HpaII and HindII + III) is reported here. The existence of the three single-stranded breaks complicates the identification and the molecular weight determination of fragments produced by HpaII, HindIII and HindII + III. Indeed the electrophoretic mobility of some fragments in which a single-stranded discontinuity is located is modified, and the fluorescence of ethidium bromide complexed with these fragments is reduced as compared to that observed for the other fragments existing in a molar ratio. These drawbacks were overcome by performing experiments of nick-translation of CaMV DNA with Escherichia coli DNA polymerase I. FRom the data it follows that the CaMV DNA molecule bears bears 1 site for HhaI and SacI, 2 for AvaI and PvuII, 3 for PstI, 4 for XbaI, 5 for EcoRI, 6 for Bg/II and HincII, 11 for HpaII and 15 for HindII + III. The corresponding fragments have all been ordered and precisely located providing a suitable map for further investigations connected with the study of the fine structure and the function of the CaMV genome.  相似文献

Purpose

The purpose of this paper is to provide an improved (up-to-date) insight into the environmental burden of textiles made of the base materials cotton, polyester (PET), nylon, acryl, and elastane. The research question is: Which base material and which life cycle stage (cradle-to-gate as well as cradle-to-grave) have the biggest impact on the environment?

Methods

Life cycle inventory (LCI) data are collected from the literature, life cycle assessment (LCA) databases, and emission registration database of the Dutch government, as well as communications with both manufacturing companies of production equipment and textile companies. The output of the calculations is presented in four single indicators: Eco-costs 2012 (a prevention-based indicator), CO2 equivalent (carbon footprint), cumulative energy demand (CED), and ReCiPe (a damage-based indicator).

Results and discussion

From an analysis of the data, it becomes clear that the environmental burden is not only a function of the base materials (cotton, PET, nylon, acryl, and elastane) but also of the thickness of the yarn (for this research, the range of 50–500 dtex is examined). The authors propose that the environmental burden of spinning, weaving, and knitting is a function of 1/yarn size. The cradle-to-grave analysis from raw material extraction to discarded textile demonstrates that textiles made out of acryl and PET have the least impact on the environment, followed by elastane, nylon, and cotton. The use phase has less relative impact than it is suggested in the classical literature.

Conclusions

The impact of spinning and weaving is relatively high (for yarn thicknesses of less than 100 dtex), and from the environmental point of view, knitting is better than weaving. LCA on textiles can only be accurate when the yarn thickness is specified. In case the functional unit also indicates the fabric per square meter, the density must be known. LCA results of textile products over the whole value chain are case dependent, especially when dyeing and finishing processes and the use phase and end-of-life are included in the analysis. Further LCI data studies on textiles and garments are urgently needed to lower the uncertainties in contemporary LCA of textile materials and products.  相似文献
