Cytochrome systems in cells of a denitrifying bacterium, Pseudomonasstutzeri (VAN NIEL strain), grown under different atmosphericconditions were compared with reference to the effects of nitrateand nitrite on cytochrome synthesis. When a culture was sufficiently aerated (aerobic conditions),synthesis of all cytochrome components was repressed, regardlessof the presence or absence of nitrate and nitrite. When aeratedmoderately (semi-aerobic conditions), both soluble and paniculatecytochromes c-552 and cytochrome b-558 contents markedly increasedeven in the absence of nitrate and nitrite. Under anaerobic or semi-aerobic conditions, nitrite inducedcytochrome a2–c synthesis. This inductive effect of nitritewas counteracted by nitrate. Trichosporon cutaneum WY2-2 was shown to metabolize p-hydroxybenzoatevia protocatechuate and hydroxyquinol. Using superoxide dismutaseas a stabilizer of hydroxyquinol, the conversion of protocatechuateto hydroxyquinol and the ring fission process of hydroxyquinolwere confirmed. Hydroxyquinol was chemically identified as theproduct of protocatechuate hydroxylase reaction. Partially purifiedprotocatechuate hydroxylase was highly specific for protocatechuate;its Km values for protocatechuate and NADH were 17.6 and 12.4µM, respectively. It catalyzed equimolar CO2 formation,NADH oxidation and O2 consumption from protocatechuate. The localization of dissimilatory nitrate and nitrite reductasesof a denitrifying phototrophic bacterium, Rhodopseudomonas sphaeroidesforma sp. denitrificans, was investigated. Nitrate and nitritereductases were located in the periplasmic space of the bacteriumgrown anaerobically in the presence of nitrate either in lightor in darkness. The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. Levels of subunits of two acetyl-coenzyme A carboxylases werehigh in small leaves of Pisum sativum, decreased with growth,and remained constant in fully expanded leaves. Properties of the cell-free extract, prepared from a strainof Thiobacillus thiooxidans by sonic disruption followed byfractionation with centrifugatiori, were investigated with referenceto its sulfite-oxidizing activity. Without the addition of cofactors the particulate fraction(F-P)catalyzed oxidation of sulfite with oxygen or bacterial cytochromec-552 obtained from Pseudomonas stutzeri as electron acceptor.TMPD reduced by ascorbic acid was also oxidized by F-P. Thesoluble fraction(F-S) showed no activity in oxidizing sulfiteand TMPD, but stimulated TMPD oxidation by F-P. Oxygen uptake with either sulfite or TMPD as substrate was inhibitedby KCN, NaN3, CO and c-phenanthroline. CO-Inhibition was reversedby light. Reduction of cytochrome c-552 by sulfite was insensitiveto these agents. Antimycin A markedly inhibited sulfite oxidation with eitheroxygen or cytochrome c-552 as electron acceptor, but was withouteffect on TMPD oxidation. DDC and SAO, both strong inhibitors of sulfur oxidation, didnot affect sulfite and TMPD oxidations. Cytochromes of the a, b and c types were contained in F-P. Thesecytochromes were rapidly reduced when F-P was incubated withsulfite. Elicitor prepared from Phytophthora nicotianae stimulated inositolphospholipid turnover and induced phenylalanine ammonia-lyaseactivity in tobacco suspension culture cells [Kamada and Muto(1994) Plant Cell Physiol. 35: 397]. Protein kinase inhibitors,K252a and staurosporine inhibited both responses. These resultssuggest that inositol phospholipid turnover plays an importantrole in PAL induction through protein kinases. Intact tubers of potato (Solanum tuberosum L. cv. Irish Cobblerand an interspecific hybrid between S. tuberosum and S. demissumcv. Rishiri) contain a very low activity of 3-hydroxy-3-methylglutaryl(HMG)-CoA reductase. The activity increased first in responseto slicing, and again in response to additional treatments suchas inoculation with an incompatible race of Phytophthora infestans,application of a hyphal wall component of the fungus or HgCl2solution, and then decreased. Both the first and the secondincreases in activity in response to slicing and additionaltreatment with a hyphal wall component to elicit phytoalexinproduction were inhibited by blasticidin S. Gibberellin A1, (GA1), GA19, and GA20 in phloem exudates andcotyledons of seedlings of Pharbitis nil cv. Violet, grown underdifferent photoperiodic conditions, were qualitatively and semi-quantitativelyanalyzed by a combination of high performance-liquid chromatography(HPLC) and radioimmunoassays (RIA). The levels of GA19 and GA20were higher in cotyledons from plants grown under dark treatment(DT) conditons of 16 h-light/8 h-dark for 6 days followed by8 h-light/16 h-dark for 3 days than in those grown under continuouslight (CL) for 9 days. This relationship was also observed forthe GAs in phloem exudates, although the levels were much lowerthan in the cotyledons. When GAs were applied to the cotyledons,elongation of the epicotyl was promoted more by GA20 than byGA1 or GA19, especially under the CL treatment. A nitrogen-fixing bacterium Azotobacter vinelandii was successfullygrown in a specially designed system with constant partial pressuresof N2O (0.2 atm) and O2 (0.2 atm) in a nitrogen-free liquidmedium. Reduction of N2O proceeded with the evolution of N2in the gas phage. Large nitrogen isotope fractionation was found for both processes,reduction of N2O to N2 and N2O-fixation. The kinetic isotopefractionation factors of these reactions were at most 1.039and 1.034, respectively. Furthermore, an unexpected inverseisotope effect (organic-N, the end-product, is more enrichedin 15N than N2, the intermediate) strongly suggested that N2Owas directly assimilated within the bacterial cells. Simultaneousassimilation of N2O and N2 was also confirmed by using a 15Ntracer technique. IAA applied simultaneously with osmotica greatly enhanced theadaptive recovery of the elongation growth of segments of Vignahypocotyls during osmotic stress irrespective of whether ornot absorbable solutes were present. IAA stimulated both thesurface pump and the xylem pump, which have been shown to bestimulated by osmotic stress and to control the yielding ofthe cell wall and the absorption of solutes. Thus, wall extensibilityand the effective turgor were further enhanced under osmoticstress in the presence of IAA. These results indicate that thesimultaneous presence of IAA can reduce the inhibition of growthby osmotic stress, and they support numerical predictions basedon the apoplast canal model. Cytochrome b-560 was purified to an electrophoretically homogeneousstate from Nitrosomonas europaea. It showed absorption peaksat 427, 530 and 560 nm in the reduced form. Its molecular weightwas estimated to be 44,000 by SDS-polyacrylamide gel electrophoresisand the same value was obtained on the basis of the contentsof haem and protein. Both cytokinins and fusicoccin (FC) stimulated the transpirationand the amino acid accumulation in leaf discs of Brassica campestrisvar. komatsuna. Enhancement effects were of the same magnitude.Both the accumulation and the transpiration were similarly inhibitedwhen vaseline was smeared on the leaf surface. Abscisic acid(ABA) also inhibited those cytokinin-induced effects. The accumulationof amino acid-14C was at the cytokinin- or FC-treated site unlessthe leaf surface was smeared with vaseline. These facts suggestthat cytokinin- or FC-induced amino acid accumulation in leafis caused by the stimulation of transpiration. A cell extract from acetate-grown Trichosporon cutaneum WY2-2inhibited auto-oxidation of phenolics, especially that of hydroxyquinol.It prevented auto-oxidation of hydroxyquinol without directinteraction with hydroxyquinol. Bovine erythrocyte superoxidedismutase had similar characteristics as the cell extract, andthe elution patterns of superoxide dismutase activity and ofthe inhibitory activity to hydroxyquinol auto-oxidation froma Sephadex G-150 column coincided. These results indicate thatthe inhibitory activity in the cell extract is mainly due tosuperoxide dismutase. The pattern for primary products of CO2-fixation and the chloroplaststructure of Amaranthus retrqflexus L., a species which incorporatescarbon dioxide into C4 dicarboxylic acids as the primary productof photosynthesis, were compared in various chlorophyll containingtissues,i.e., foliage leaves, stems, cotyledons and pale-greencallus induced from stem pith. Despite some morphological differencesin these assimilatory tissues, malate and aspartate were identifiedas the major compounds labelled during a 10 sec fixation of14CO2 in all tissues. Whereas, aspartate was the major componentin C4-dicarboxylic acids formed in foliage leaves, malate predominatedas the primary product in stems, cotyledons and the pale-greencallus. A novel photorespiratory mutant of Arabidopsis thaliana, designatedgld2, was isolated based on a growth requirement for abnormallyhigh levels of atmospheric CO2. Photosynthetic CO2 fixationwas inhibited in the mutant following illumination in air butnot in atmosphere containing 2% O2. Photosynthetic assimilationof 14CO2 in an atmosphere containing 50% O2 resulted in accumulationof 48% of the soluble label in glycine in the mutant comparedto 9% in the wild type. The rate of glycine decarboxylationby isolated mitochondria from the mutant was reduced to 6% ofthe wild type rate. In genetic crosses, the mutant complementedtwo previously described photorespiratory mutants of A. thalianathat accumulate glycine during photosynthesis in air due todefects in glycine decarboxylase (glyD, now designated gld1)and serine transhydroxymethylase (stm). Because glycine decarboxylaseis a complex of four enzymes, these results are consistent witha mutation in a glycine decarboxylase subunit other than thataffected in the gld1 mutant. Crystalline cytochrome c-553 was obtained from Porphyra yezoensisUeda. Chara cells without tonoplasts, prepared by replacing the cellsap with EGTA-containing media, showed essentially the samepattern of light-induced changes in membrane potential and membraneresistance as normal cells although the concentrations of ionsand ATP in the cytoplasm decreased considerably (1/3–1/10)after loss of the tonoplast. Removal of the tonoplast reducedthe rate of photosynthetic O2 evolution to about 50% of thatof normal cells but did not affect the magnitude of light-inducedpotential change.
