表皮生长因子受体(EGFR)是一种具有酪氨酸激酶活性的膜表面受体,其胞内区的3个亚区是其发挥酪氨酸激酶活性、介导信号转导的关键部位。表皮生长因子受体和其他的erbB受体可形成同源和异源的多种二聚体,不同的二聚体与表皮生长因子受体的6种配体形成的不同组合可将不同的细胞外刺激传入胞内。表皮生长因子可激活多种下游信号路径,产生多种生物学效应,ras-raf-MEK-erk/MAPK途径与增殖的激活有关,PI3K-PKC-IKK途径与细胞移动性的增强有关。表皮生长因子受体与肿瘤的发生发展和器官的修复有密切的关系,针对表皮生长因子受体的肿瘤治疗和器官修复具有良好的应用前景。  相似文献

采用免疫组织化学方法观察了表皮生长因子受体在正常大鼠眼组织的表达。结果发现:角膜上皮的深层细胞和角膜缘上皮细胞、部分结膜上皮细胞和结膜下结缔组织内成纤维细胞均强烈表达表皮生长因子受体。结果显示:表皮生长因子受体阳性细胞主要分布于眼表层组织。这些细胞不仅是眼组织损伤后修复、而且是多种手术能否成功和某些疾病形成中起重要作用的细胞  相似文献

阿霉素肾病大鼠表皮生长因子及其受体的表达   总被引:2,自引:0,他引:2  
目的研究阿霉素肾病大鼠肾组织中表皮生长因子(EGF)及其受体EGFR的表达分布以及表达量与尿蛋白之间关系。方法选择第5天、14天、28天作为动态观察的时点,同期设立正常对照。采用荧光定量RT-PCR、免疫组织化学及计算机图像定量分析EGF mRNA以及EGF、EGFR蛋白在肾组织的表达,同时测定24 h尿蛋白定量。WT1和EGFR双重免疫组化确定EGFR在肾小球内确切细胞定位。结果阿霉素注射后第5天,EGFmRNA即较正常增高,28 d明显增高并高于5 d和14 d。正常对照组EGF阳性细胞主要分布于远曲小管和髓袢,阿霉素组EGF还在集合管和近曲小管上表达;EGF阳性表达范围和强度随尿蛋白增加而增加;EGFmRNA表达量以及EGF在肾小管中的表达强度与24 h尿蛋白量呈正相关。肾小管上皮细胞广泛表达EGFR,阿霉素组EGFR在小管表达均高于正常,但组间各时点差异无显著性;随尿蛋白增加EGFR在肾小球内表达逐渐增多。EGFR在肾小球和肾小管中的表达强度均与24 h尿蛋白量呈正相关。WT1和EGFR双重免疫组化显示阿霉素肾病组EGFR可在足突细胞上表达,正常组则无。结论阿霉素肾病大鼠的肾小球脏层上皮有EGFR的表达。EGF/EGFR可能参与了阿霉素肾病的发病过程以及蛋白尿的形成。  相似文献

EGFR主要由三个结构区域组成,即EGF结合区域、跨膜区域和依赖EGF蛋白激酶区域。EGFR被激活后,不仅能使许多底物蛋白磷酸化,自身也发生磷酸化,这些磷酸化作用在信息传导中有重要作用。EGFR与EGF结合后,其复合体以胞饮方式内容形成受体小体,除部分被降解外,大部分在高尔基GERL区加工后到达细胞核调节基因表达。本文通过对受体激活模型和用体外遗传变异方法对变异EGFR的研究阐述了EGFR的结构与功能的关系。  相似文献

为将人表皮生长因子(EGF)受体膜外第Ⅲ功能区(hEGF-RⅢ)表达于大肠杆菌菌膜上,重组构建了EGF-RⅢ表达载体,并转化获得大肠杆菌表达株.放射性受体分析发现125I-EGF可与表达菌特异性结合,其特异性结合量随反应的时间和温度而变化,Scatchard分析显示表达菌表达单一亲和性受体,其解离常数为3.0×10-11 mol/L,每个细菌约有738个结合位点.免疫电镜显示EGF-RⅢ多分布于细菌菌膜上.  相似文献

大鼠脊髓损伤后表皮生长因子受体在脊髓的表达特点   总被引:1,自引:0,他引:1  
目的研究大鼠脊髓损伤(spinal cord injury,SCI)后表皮生长因子受体(epidermal growth factor re-ceptor,EGFR)在脊髓的表达特点及意义。方法健康成年雄性SD大鼠,随机分为4组(每组10只):假手术组,SCI术后3 d、7 d和14 d组。应用Basso Beattie Bresnahan(BBB)评分观察大鼠行为学改变;逆转录-聚合酶链反应(RT-PCR)检测损伤段脊髓组织中EGFR mRNA表达水平;免疫组织化学方法观察损伤段脊髓灰质中EGFR蛋白表达情况;并对EGFRmRNA及蛋白表达情况与BBB评分进行相关性分析。结果行为学观察发现大鼠脊髓损伤后下肢神经功能逐步恢复;RT-PCR结果显示EGFR mRNA在假手术组大鼠脊髓中微量表达,SCI术后3 d表达显著升高,随后趋于下降,14 d时仍高于假手术组(P<0.01);免疫组织化学染色显示损伤段脊髓灰质中EGFR阳性细胞数在损伤后3 d显著高于假手术组(P<0.01),随后趋于下降,但14 d时仍高于假手术组(P<0.01);EGFR mRNA及蛋白的表达均与BBB评分呈显著负相关(r=-0.956,P<0.05;r=-0.966,P<0.05)。结论EGFR在大鼠脊髓损伤后具有时相分布特点,且与动物行为呈负相关,提示其表达可能阻碍损伤后的神经功能恢复。  相似文献

表皮生长因子受体(epidermalgrowthfactorreceptor,EGFR)是ErbB家族成员之一,具有酪氨酸激酶活性,是一种重要的跨膜受体。EGFR被配体激活后启动胞内信号传导,经过细胞质中衔接蛋白、酶的级联反应,调节转录因子激活基因的转录,指导细胞迁移、黏附、增殖、分化、凋亡,且与肿瘤的形成和恶化密切相关。本文对EGFR的结构特性、几种重要的信号通路及各个信号通路之间的交联,以及信号的衰减等方面的研究进展进行了综述。  相似文献

We study a mechanism by which dimerization of the EGF receptor (EGFR) cytoplasmic domain is transmitted to the ectodomain. Therapeutic and other small molecule antagonists to the kinase domain that stabilize its active conformation, but not those that stabilize an inactive conformation, stabilize ectodomain dimerization. Inhibitor-induced dimerization requires an asymmetric kinase domain interface associated with activation. EGF and kinase inhibitors stimulate formation of identical dimer interfaces in the EGFR transmembrane domain, as shown by disulfide cross-linking. Disulfide cross-linking at an interface in domain IV in the ectodomain was also stimulated similarly; however, EGF but not inhibitors stimulated cross-linking in domain II. Inhibitors similarly induced noncovalent dimerization in nearly full-length, detergent-solubilized EGFR as shown by gel filtration. EGFR ectodomain deletion resulted in spontaneous dimerization, whereas deletion of exons 2–7, in which extracellular domains III and IV are retained, did not. In EM, kinase inhibitor-induced dimers lacked any well defined orientation between the ectodomain monomers. Fab of the therapeutic antibody cetuximab to domain III confirmed a variable position and orientation of this domain in inhibitor-induced dimers but suggested that the C termini of domain IV of the two monomers were in close proximity, consistent with dimerization in the transmembrane domains. The results provide insights into the relative energetics of intracellular and extracellular dimerization in EGFR and have significance for physiologic dimerization through the asymmetric kinase interface, bidirectional signal transmission in EGFR, and mechanism of action of therapeutics.  相似文献

Signaling via growth factor receptors, including the epidermal growth factor (EGF) receptor, is key to various cellular processes, such as proliferation, cell survival, and cell migration. In a variety of human diseases such as cancer, aberrant expression and activation of growth factor receptors can lead to disturbed signaling. Intracellular trafficking is crucial for proper signaling of growth factor receptors. As a result, the level of cell surface expression of growth factor receptors is an important determinant for the outcome of downstream signaling. BAR domain-containing proteins represent an important family of proteins that regulate membrane dynamics. In this study, we identify a novel role for the F-BAR protein PACSIN2 in the regulation of EGF receptor signaling. We show that internalized EGF as well as the (activated) EGF receptor translocated to PACSIN2-positive endosomes. Furthermore, loss of PACSIN2 increased plasma membrane expression of the EGF receptor in resting cells and increased EGF-induced phosphorylation of the EGF receptor. As a consequence, EGF-induced activation of Erk and Akt as well as cell proliferation were enhanced in PACSIN2-depleted cells. In conclusion, this study identifies a novel role for the F-BAR-domain protein PACSIN2 in regulating EGF receptor surface levels and EGF-induced downstream signaling.  相似文献

A constitutively active epidermal growth factor receptor (EGFR) mutant, EGFR variant III (EGFRvIII), has been detected at high frequencies in certain human cancers. This study evaluated transactivation and trafficking of erbB family members as a result of constitutive EGFR activity in a cancer cell line. Expression of EGFRvIII modulated erbB family members through different mechanisms; the erbB3 mRNA level was reduced, whereas wild-type EGFR (wtEGFR) and erbB2 protein levels were diminished, with no change in their mRNA levels, and there was no change in the erbB4 expression level. Both EGFR and erbB2 were internalized as a result of EGFRvIII''s activity and redistributed to the cell surface upon addition of AG1478, an inhibitor of wtEGFR/EGFRvIII catalytic activity. Acute activation of EGFRvIII by removing AG1478 from cells increased phosphorylation of both wtEGFR and erbB2 and caused differential trafficking of EGFRvIII''s activation partners; wtEGFR was directed primarily to lysosomal compartments and partially to recycling compartments, whereas erbB2 was directed primarily to recycling compartments and partially to lysosomal compartments. Our data demonstrate that the constitutive activity of EGFRvIII is sufficient to trigger endocytosis and trafficking of wtEGFR and erbB2, which may play a role in activating signaling pathways that are triggered during receptor endocytosis. 目的:探讨雌激素受体(ER)和表皮生长因子受体(EGFR)在分化型甲状腺癌(DTC)中的表达及其意义.方法:用免疫组化SP法检测45例(DTC)组织中ER和EGFR的表达.结果:ER在DTC组中的阳性率为37.3%,明显高于良性腺瘤和正常组(P<0.05);EGFR在DTC组中的阳性率为57.7%,明显高于两对照组(P<0.05).ER在DTC中的表达与性别和年龄有关;EGFR的表达与DTC中的淋巴结转移有关.结论:ER和EGFR的表达可以作为鉴别甲状腺肿瘤良恶性的指标EGFR可作为甲状腺癌患者预后的指标.  相似文献

取花背蟾蜍(Bufo raddei)363、7、38、39期蝌蚪肺组织和幼蟾肺组织,进行常规石蜡切片,用免疫组化SP两步法检测EGFR的表达。观察了表皮生长因子受体(EGFR)在花背蟾蜍胎肺发育过程的表达特征,并探讨表皮生长因子(EGF)和转化生长因子α(TGF-α)通过与EGFR的作用,对花背蟾蜍胎肺形态发生和肺泡上皮成熟分化的作用。结果表明,36期,EGFR在肺网状隔膜上皮细胞处有表达;37期,肺网状隔膜处EGFR阳性表达很明显,在肺泡囊处表达呈弱阳性;38期,肺网状隔膜处EGFR的阳性表达变弱,在远端的肺泡囊上皮细胞处其阳性表达增强;39期,EGFR在肺泡囊上皮细胞处阳性表达最活跃,在网状隔膜处EGFR的表达很弱;幼蟾期,EGFR阳性反应主要定位在肺泡上皮细胞。结论是,在胎肺发育的不同时期,EGFR在上皮细胞的定位有迁移,免疫组化反应强弱也有差异,说明EGFR在胎肺不同发育阶段发挥不同的功能,它对肺泡上皮细胞的成熟分化有重要调节作用。  相似文献

Phosphorylation-induced conformational changes have been well documented with different receptor tyrosine kinases. However, the susceptible epitopes and the tyrosine residue(s) involved in particular structural alteration mostly remain to be determined. Using a conformation-specific anti-peptide antibody, we have not only identified one such domain in the C-terminal tail of the EGF receptor but also identified the phosphate acceptor sites that are involved in the conformational change.  相似文献

目的:随着表皮生长因子受体(epidermal growth factor receptor,EGFR)靶点治疗药物在其它肿瘤治疗中的应用,能否在妇科肿瘤中应用并取得疗效,已经得到学者们的重视。但在临床应用之前,必须明确EGFR与妇科肿瘤发生发展的关系及其机制。因此,我们来探讨表皮生长因子受体EGFR信号通路在维吾尔族不同级别宫颈病变组织中的差异表达及其临床意义。方法:应用免疫组织化学方法检测EGFR、P-EGFR、ERK 1/2、P-ERK1/2在108例子宫颈癌,47例CIN,79例正常宫颈组织中的表达,并分析其与临床病理参数的关系。结果:EGFR在子宫颈癌、CIN及正常宫颈组织中的阳性表达率分别是74.10%、66%、27.80%(P0.01);P-EGFR在三组间的阳性表达率分别是55.60%、74.50%、20.30%(P0.01);ERK1/2在三组间的阳性表达率分别是60.10%、76.60%、25.30%(P0.01);P-ERK1/2在三组间的阳性表达率分别是44.40%、66%、24.10%(P0.01),其在宫颈癌中EGFR与与浸润深度、淋巴结转移有关;ERK与肿瘤的临床分期、远处转移有关,而且在宫颈癌中EGFR与P-EGFR、ERK 1/2、P-ERK1/2的表达成正相关(P0.05)。结论:EGFR、P-EGFR、ERK1/2、P-ERK1/2的表达与宫颈癌的发生、发展相关,促进宫颈癌的浸润和转移,抑制EGFR信号通路可能为多靶点联合治疗宫颈癌提供新的方向。  相似文献

癌组织中表皮生长因子受体(epidermal growth factor receptor,EGFR)基因突变是应用靶向药物EGFR酪氨酸激酶抑制剂(tyrosine kinase inhibitor,TKI)治疗的一个重要相关因素及预测指标。对其突变的检测可以指导TKI类药物(TKIs)的最佳应用。该种突变常出现在非小细胞肺癌(NSCLC)中,尤其是在亚洲女性、肺腺癌、非吸烟者中,与非小细胞肺癌患者对TKIs治疗的敏感性密切相关。本文旨在探讨利用EGFR基因的已知突变热点的相关知识选择适合不同分子遗传学背景的群体或/和个体的"个体化"治疗方案,最终达到延长肺癌患者生存时间和提高生活质量的双重目的。  相似文献

肿瘤靶向分子的筛选一直是肿瘤治疗和早期诊断的研究热点 . 表皮生长因子受体 (EGFR) 在很多肿瘤细胞表面过量表达,是一个理想的药物输送靶点 . 选择了 EGFR 表面不同于表皮生长因子 (EGF) 结合位点的一个凹陷部位作为计算机模拟筛选的结合位点,然后使用 DOCK 软件包对 DTP-Plated 有机小分子数据库进行了两遍筛选,最后选择了 7 个有机小分子作为可能的靶向分子 . BIAcore 体外结合实验对所选择的小分子样品进行了进一步的验证,结果表明,小分子 NSC51186 能特异地与 EGFR 结合 . 小分子 NSC51186 和 EGFR 之间的动力学常数也得到进一步的测定 . 新的靶向分子和药物、纳米粒子或者基因载体相连,将有可能用于靶向于 EGFR 的肿瘤治疗和诊断 .  相似文献
