We investigated whether the relative contribution of the baroreflex and central commands to the control of heart period differs between spontaneously hypertensive rats (SHR) and Wistar-Kyoto normotensive rats (WKY) during physiological behavior. Rats were instrumented with an arterial catheter and with electrodes for discriminating wakefulness, nonrapid eye movement sleep (NREMS), and rapid eye movement sleep (REMS). The cross-correlation function (CCF) between spontaneous fluctuations of heart period and mean arterial pressure was computed at frequencies <0.2 Hz. The baroreflex determines a positive correlation between heart period and previous pressure values. This pattern was observed in the CCF during quiet wakefulness (QW) and NREMS, and in QW, it was accompanied by a pronounced negative correlation between heart period and subsequent pressure values. The relative baroreflex contribution to the control of heart period, estimated from the positive peak value of the CCF, was lower in SHR than in WKY during QW but not during NREMS. During REMS, the CCF showed a negative correlation between heart period and both previous and subsequent pressure values, reflecting the prevalence of central autonomic commands. The relative contribution of central commands to the control of heart period, estimated from the negative peak value of the CCF, was lower in SHR than in WKY during REMS. These results suggest that during QW and REMS, the control of heart period exerted by the baroreflex and central commands, respectively, is less effective in SHR than in WKY. This difference is not apparent in a behavioral state of autonomic stability such as NREMS.  相似文献

Modifications in total amount, frequency and duration of episodes of wakefulness (W), drowsiness (D), slow sleep (SS) and paradoxal sleep (PS) were examined in cats with small unilateral lesion in the locus coeruleus (LC) and/or neighbouring structures. Throughout a 8 week period, 24 h weekly records were obtained from each cat. Control group with lesions in the dorsomedial pontine area showed a noticeable stability in sleep-wakefulness cycle (SWC). Significant decrease of W (with shortening of episodes) occurred during the whole postlesion period in the group with lesions in the lateral vestibular nucleus. The group with small lesions restricted to the dorsal-intermediate part of RPO and RPC nuclei had only acute SWC modifications consisting of a decrease of W and increase of PS. Finally, small lesions in LC complex that encroached to a limited extent neighbouring structures produced a maintained decrease of W (with shortening of episodes) and increase of PS (due to a major number of episodes). Results suggest a different role of dorsopontine region from other pontine areas in SWC mechanisms and give support to the hypothesis of a permissive role of LC in PS mechanisms.  相似文献

The central neural mechanisms underlying differences in cardiovascular variability between wakefulness, non-rapid-eye-movement sleep (NREMS), and rapid-eye-movement sleep (REMS) remain poorly understood. These mechanisms may involve hypocretin (HCRT)/orexin signaling. HCRT signaling is linked to wake-sleep states, involved in central autonomic control, and impaired in narcoleptic patients. Thus, we investigated whether HCRT signaling plays a role in controlling cardiovascular variability during spontaneous behavior in HCRT-deficient mice. HCRT-ataxin3 transgenic mice lacking HCRT neurons (TG), knockout mice lacking HCRT peptides (KO), and wild-type controls (WT) were instrumented with electrodes for sleep recordings and a telemetric blood pressure transducer. Fluctuations of systolic blood pressure (SBP) and heart period (HP) during undisturbed wake-sleep behavior were analyzed with the sequence technique, cross-correlation functions, and coherent averaging of SBP surges. During NREMS, all mice had lower SBP variability, greater baroreflex contribution to HP control at low frequencies, and greater amplitude of the central autonomic and baroreflex changes in HP associated with SBP surges than during wakefulness. During REMS, all mice had higher SBP variability and depressed central autonomic and baroreflex HP controls relative to NREMS. HP variability during REMS was higher than during NREMS in WT only. TG and KO also had lower amplitude of the cardiac baroreflex response to SBP surges during REMS than WT. These results indicate that chronic lack of HCRT signaling may cause subtle alterations in the control of HP during spontaneous behavior. Conversely, the integrity of HCRT signaling is not necessary for the occurrence of physiological sleep-dependent changes in SBP variability.  相似文献

The changes in amplitude of primary responses evoked at cortical level by paired pulses delivered into the optic nerve, have been statistically compared during seven stages of sleep and wakefulness. Results are in agreement with changes in reactivity observed by other authors in the cat; an unexpected but significant decrease in reactivity has however been obtained during eyemovements when compared with other periods of paradoxical sleep.  相似文献

A new multiparameter method for evaluating anxiety-phobic states in rats is elaborated. The method is based on the ranged scale consisting of parameters which characterize the species-specific rat response to the series of ethologically adequate test-stimuli inducing manifestation of the anxiety-phobic states. The method makes it possible to evaluate individual anxiety-phobic level in rats, to form experimental groups with rats of known individual anxiety-phobic level, to conduct repeated observations in the same animals. The method is easy to perform in laboratory and makes it possible to get the results quickly.  相似文献

A daily rhythm of blood pressure (BP), with maximum values in the activity period, carries important prognostic information. The extent to which this rhythm depends on behavioral factors remains debated. Mice are the species of choice for functional genomics. In mice, episodes of wakefulness and sleep are not restricted to particular daily periods, allowing BP in each wake-sleep state to be measured at each time of day. The aim of this study was to investigate whether a circadian rhythm of BP is manifest in each wake-sleep state in mice. Mice with B6 genetic background (n?=?26) were implanted with a telemetric BP transducer and electrodes to discriminate wake-sleep states and recorded while housed under a 12:12?h light-dark period. For each mouse, 8 values of BP were obtained in each wake-sleep state (wakefulness, non-rapid-eye-movement sleep, and rapid-eye-movement sleep) by averaging over successive 3-h time bins. Analysis of variance evidenced a significant time effect in each wake-sleep state as well as a significant wake-sleep state?×?time interaction effect. In an additional group of mice (n?=?3) recorded in constant darkness, the Lomb-Scargle periodogram also revealed a significant circadian rhythm of BP in each wake-sleep state. These findings demonstrate that during each wake-sleep state, mice show daily and circadian rhythms of BP in conditions of entrainment to the light-dark cycle and in free-running conditions of constant darkness, respectively.  相似文献

目的:分析大鼠不同状态脑内递质功率的变化。方法:本研究采用脑涨落图仪。结果:与清醒状态相比,麻醉状态脑内递质的功率显著下降(P<0.05);大鼠死亡后脑内(死亡当时-死亡后48小时)6种递质的功率都降低到10-5级别(P值均大于0.05),说明死亡后不同时间脑内6种递质的功率无显著性差异。结论:从清醒状态到麻醉状态再到死亡状态,大脑的抑制程度逐渐加深。  相似文献

Deformation of the upper airway (UA) by negative transmural pressure alters the activity of UA mechanoreceptors, causing a reflex increase in UA muscle activity. Topical anesthesia of the UA mucosa, which greatly reduces this reflex response, causes an increase in UA resistance during stage 2 sleep. We hypothesized that topical anesthesia of the UA mucosa would predispose to UA instability at sleep onset and, therefore, examined the effect of UA anesthesia on pharyngeal resistance (Rph) in stage 1 sleep. Eleven normal, healthy volunteers were instrumented to record standard polysomnographic variables, respiratory airflow, and UA pressure at the nasal choanae and the epiglottis. Subjects were permitted to sleep until stable stage 2 sleep was reached and were then awoken. This procedure was repeated three times to obtain reproducible wake-sleep transitions. The UA mucosa was then anesthetized with 10% lidocaine to the oropharynx and laryngopharynx, and the pharyngeal mechanics were studied during the subsequent wake-sleep transition. Three subjects were excluded because of failure to resume sleep postanesthesia. Rph was significantly higher after anesthesia during stage 1 sleep [2.88 +/- 0.77 cmH(2)O.l(-1).s (mean +/- SE)] compared with control (0.95 +/- 0.35 cmH(2)O.l(-1).s; P < 0.05), but there was no difference during wakefulness. Furthermore, there was a significant rise in Rph at wake-to-sleep transitions and a significant fall in Rph at sleep-to-wake transitions after anesthesia (P < 0.05) but not in the control condition. We conclude that sensory receptors in the UA mucosa contribute to the maintenance of UA patency at wake-sleep transition in normal humans.  相似文献

An automatically working test arrangement for the permanent analysis of O2 and CO2 in microbiological cultures is described. The measuring principle is based on the paramagnetic properties of oxygen and on the absorption of infrared by carbon dioxide. The preparation of the gas for measuring and the correction of the recording are indicated. The formula of correction was programmed and the values were calculated for a range of 3%. The routine correction of analysis values is done with a nomogram established on the basis of these individual values. The advantages of the described test arrangement are illustrated by two examples of growth experiments on Saccharomyces cerevisiae.  相似文献

Prolactin (PRL) is mitogenic for lymphocytes in vitro , but the responsiveness of lymphocytes depends on the in vivo hormonal status of the rats from which the cells were obtained. Lymphocytes from ovariectomized (OVX) rats, but not from rats in oestrus or from male rats, respond to prolactin; administration of oestradiol to OVX rats diminishes the response. In order to determine if a correlation exists between lymphocyte responsiveness to prolactin and levels of cell surface prolactin receptors (PRL-R) expression, the percentage of splenocytes and each splenocyte subpopulation expressing surface PRL-R from rats of various hormonal states (OVX, oestradiol-injected OVX, oestrus and male) was analysed by single-colour and dual-colour flow cytometric analysis. We found that approximately 20% of splenocytes expressed surface PRL-R regardless of hormonal states ( n =16). The majority (85%) of PRL-R positive splenocytes were B lymphocytes whereas 11.1% and 4.8% of splenocytes expressing the PRL-R were CD4 positive T-helper (TH) and CD8 positive T-cytotoxic (TC) lymphocytes, respectively. B lymphocytes also stained more brightly than T lymphocytes. This distribution of PRL-R expression did not show significant alterations on total splenocytes or TH and TC lymphocytes during various hormonal stages. However, the percentage of PRL-R-positive B lymphocytes increased markedly in OVX rats (twofold), compared to rats at oestrus. In summary, no correlation was found between the responsiveness to prolactin as a mitogen and levels of PRL-R expression by lymphocytes from rats at different hormonal states. This result suggests that sex steroid hormones may control prolactin responsiveness of lymphocytes by affecting the signal transduction pathway through PRL-R rather than by altering the level of the cell surface receptor expression.  相似文献

Isolated liver mitochondria were crosslinked with glutaraldehyde during different respiratory states and then analyzed either by freeze-fracturing, or in thin sections prepared according to a low denaturation embedding technique. In freeze-fractured material, large intracristal spaces were found in 86% of the mitochondria freshly isolated in a sucrose medium. These results were in good agreement with mitochondria analyzed in thin sections, in which case 88% of the mitochondria contained intracristal spaces. In freeze-fractured mitochondria, only 24% of the mitochondria crosslinked after 3 min in respiratory state 4 contained intracristal spaces, while 36% of the mitochondria contained intracristal spaces during state 3. These values compared favorably with those obtained in embedded material in which case 21 and 41% of the mitochondria contained intracristal spaces during states 3 and 4, respectively. The agreement between the observations made with the two methods demonstrated that the disappearance of intracristal spaces is independent of the embedding technique, and is not a special effect caused by dehydration in ethylene glycol.  相似文献

A method is presented which allows to automatically evaluate, by the aid of a small computer, parameters concerning the way a sequence of spikes is organized in time. This method stands on few simple assumptions: Interspike Time Intervals (ITIs) and/or clusters of ITIs that are either "too large" or "too short" so as to be unlikely at 0.05 level on the basis of a Poissonian firing hypothesis are selected and assumed as "Low Firing Periods" (LFP) and "High Firing Periods" (HFP), respectively. General information is obtained about both LFPs and HFPs. Results on real cases are in agreement with previous studies on unit cell activity.  相似文献
