The fungal microflora of orchard (cock's-foot) grass seed (Dactylis glomerata L.) was determined for 140 samples obtained from thirteen countries. The microflora was assessed by visual examination of dry seed (20–50 x), by microscopic examination of centrifuged seed washings and by incubating seed on 2 % malt extract agar (MA) under a 10-day regime of diurnal cycling of temperature (23-9oC/i8-3oC) and near-ultraviolet radiation (12 h NUV/ 12 h dark). More than twenty-five species of potentially pathogenic fungi and forty species of saprophytes were detected. The pathogens included an Ascochyta sp., Botrytis cinerea, nine species of Drechslera including D. dactylitis (infested 23% of seed lots) and D. Useptatum (10% of seed lots), five species oiFusarium including F. avenaceum (5 % of seed lots), Lep-tosphaeria sp., Phleospora sp., Phoma sp., Phomopsis sp., Pyrenochaeta sp., Rhizoctonia sp., Rhynchophoma sp., Septoria sp. and Stagonospora sp. Many more fungi were identified from seed plated on MA and exposed to NUV than by examination of seed washings or visual examination of dry seed. Several of the fungi that were detected are known to produce mycotoxins.  相似文献

Hirsutella thompsonii, a moniliaceous fungus pathogenic to mites, grew and sporulated on sterilised wheat bran. The effects of environmental factors were studied on the fungus grown on potato-dextrose-agar (PDA). The fungus was mesothermophilic. Growth, sporulation and conidial germination were best at 25o-30 oC. Conidia kept at 37 oC for 5 days on PDA died, but those held at 5 oC germinated upon a subsequent removal to 25 oC. Almost all conidial germ tubes survived an 8 h exposure to 3–5% r.h. and to 60% r.h., but subsequently the former grew poorly at 100% r.h. H. thompsonii sporulated equally well in continuous darkness or light, and produced typical chlorinous to light olive-green mycelium and conidia under all conditions. A 2 h exposure of naked mycelium and conidia (which have melanised walls) to u.v. irradiation failed to kill the fungus.  相似文献

Optimising sporulation and virulence in Drechslera avenacea   总被引:1,自引:0,他引:1  
Studies were conducted on agar media to optimise sporulation of Drechslera avenacea, a fungal pathogen being evaluated as a biological control agent for Avena species (wild oats). Conidium production was affected by nutrition, pH, temperature and light conditions. Of the agar media tested, Czapek Dox agar (CZA) and half-strength oatmeal agar (½OMA) were the only media where sporulation occurred at all temperatures tested under a 12-h light:12-h dark photoperiod (L/D). The optimum temperature for conidium production was 20°C on ½OMA, whereas there was no optimum temperature on CZA. Under a 12-h near-ultraviolet (NUV):12-h dark photoperiod (NUV/D), similar numbers of conidia were produced on CZA at 6.66, 14.56, and 22.78 W m?2, whereas on ½OMA conidium production was the highest at 14.56 W m?2. When NUV/D and L/D conditions were compared, similar numbers of conidia where produced on CZA, whereas ½OMA conidium production was superior under the NUV/D photoperiod. Considerable variation in sporulation and degree of virulence of D. avenacea was detected among isolates from different geographic areas. The most virulent conidia were obtained on ½OMA at 20°C incubated under continuous illumination NUV light. Therefore, the most suitable conditions for conidium production of D. avenacea were growth for 1 week on ½OMA at 20°C under continuous NUV at an intensity of 14.56 W m?2. Under these conditions, 1.1×105 conidia mL?1 were produced which is the highest sporulation yet reported for any Drechslera spp., which are traditionally poor sporulators.  相似文献

We compared the size and spatial pattern of the germinable soil seed bank (GSB) of the three dominant perennial-grass species in the arid Patagonian Monte of Argentina. These species differ in plant functional traits, seed morphology, and in their preference by grazers. During three consecutive years, we sampled the soil seed bank at 1 m intervals and the intercepted diameter of perennial-grass species and shrub patches along 50-m linear transects at three sites with different grazing pressure. Soil samples were incubated at field capacity during 12 weeks and emerged seedlings counted. The largest-seeded and most xerophytic Stipa speciosa did not form a GSB. The other two grass species with more mesophytic traits and smaller seeds than S. speciosa formed different GSB. Poa ligularis formed a smaller and more heterogeneous GSB than Stipa tenuis. Grazing had not effect on GSB size of P. ligularis but increased the proportion of seeds under dense shrub canopies. In contrast, grazing reduced the size and also increased the proportion of the GSB of S. tenuis under shrub canopies. We conclude that the size and the spatial pattern of the GSB of the dominant perennial-grass species in the arid Patagonian Monte were related not only to seed and plant traits but also to the spatial distribution of grass plants and their exposure to grazers.  相似文献

Abstract

Light and growth-promoting compounds, such as gibberellic acids (GA3), are among the most important factors that can break physiological seed dormancy. Here, we investigate the effects of GA3 and light on germination of five species of Resedaceae that are known to have different levels of physiological dormancy. Seeds were incubated at 20/30?°C in both 12-hr photoperiod and complete darkness. To study the effect of growth hormone on germination, seeds were soaked for 24?h in different concentrations of GA3 before sowing. The annuals (Reseda aucheri and Oligomeris linifolia) and the perennial Ochradenus arabicus had deep physiological dormancy, and exogenous application of GA3 enhanced their germination in the light, but not in darkness; few or no seeds germinated in the dark in these species. Ochradenus aucheri and O. baccatus had intermediate and non-deep dormancy, respectively, and application of GA3 enhanced their germination in both light and darkness. Germination of the annual species was much slower than that of the shrubby perennials. Overall, these results indicate that conditions under which seed developed, matured and stored on maternal plants as well as incubation conditions should be taken into consideration when assessing germination behavior of the perennial species of Resedaceae.  相似文献

Abstract

The purpose of this research was to explore the effect of temperature, light, and post-harvest period, and their interactions, on seed germination ecology of four common pasture species in the Mediterranean environment. Mature seeds of Diplotaxis erucoides, Hirschfeldia incana, Hyoseris scabra (Mediterranean distribution) and Sonchus oleraceus (cosmopolitan distribution) were subjected to seven constant temperatures (10–40°C, at intervals of 5°C) under continuous darkness, or a 12 h/12 h light/dark photoperiod at 30, 150 and 270 days after harvest (DAH). Cumulative germination and germination speed were determined. In all the tested species, except S. oleraceus, light significantly enhanced germination. S. oleraceus seeds maintained germination values over 90%, in a wide range of temperatures (10–35°C), in the dark as well as in light. Seeds of H. incana germinated well soon after seed dispersal. In D. erucoides and H. scabra, germination increased with storage period, while in S. oleraceus there was no effect of seed age. In all the species, moreover, no germination was recorded at 40°C. Temperature, light, and post- harvest requirement may be regarded as an adaptation strategy to ensure optimal conditions for seedling development and survival in Mediterranean species, while the species with a cosmopolitan distribution germinates under almost all tested conditions.  相似文献

Seedlings of two susceptible sugar-beet stocks were grown either in daylight or in darkness for 24 h before or after inoculation with Peronospora farinosa f.sp. betae. Post-inoculation darkness did not significantly affect the percentage of inoculated plants that became infected but increased the percentage of plants on which the fungus sporulated in both stocks. The effects on sporulation of darkness before inoculation were more variable than those of post-inoculation darkness. In the same stocks removal of all leaves, except those in the bud, before inoculation increased the percentage both of plants which became infected and of those on which P. farinosa sporulated. Infection and sporulation were not significantly affected by defoliation in a more resistant stock. Defoliation or darkness may have affected the concentration of certain sugars or amino acids in the bud leaves thereby changing the expression of polygenically-controlled resistance to downy mildew.  相似文献

A low-temperature basidiomycete (LTB), the causal agent of cottony snow mold, was evaluated as a potential biological control agent for marsh reed grass (Calamagrostis canadensis [Michx.] P. Beauv.), a native grass of the North American boreal forest. Marsh reed grass and 2-year-old white spruce trees (Picea glauca [Moench] Voss) were inoculated with LTB and incubated for 12 weeks in a cold chamber to determine whether the fungus was pathogenic to these species. The fungus was not pathogenic to white spruce but was to marsh reed grass, causing 15% mortality and 64% reduction in dry weight of aboveground plant matter. In a second experiment, two clones of marsh reed grass were inoculated with an isolate of LTB and incubated for 8, 14, and 20 weeks in a cold chamber. Maximum response in the grass to the LTB treatment occurred after 20 weeks of incubation, with decreases of 97, 97, and 63%, respectively, in tiller number, dry weight of aboveground plant matter, and plant height. The LTB is a potential biological control agent of marsh reed grass.  相似文献

Grass shrimp (Palaemonetes pugio) fed liver containing sporulated oocysts of Eimeria funduli permitted development of sporozoites that became infective to a variety of killifishes. The shrimp's gastric mill mechanically ruptured the oocysts. Sporozoites then excysted through an opening in the sporocyst, and by 12 and 13 h postinfection (p.i.) numerous empty sporocysts and free sporozoites occurred extracellularly in the intestine of the grass shrimp. Even at 5, 7, 8, 11, 46, 79, and 83 days p.i., and presumably for many months, numerous sporozoites still occurred free in the alimentary tract or between intestinal cells. The coccidium did not infect killifish at either 2 or 4 days p.i., but did at 5 days; after release from the sporocyst, it became more elongate with a distinct nucleus and two relatively large refractile bodies. Infections of E. funduli resulted in about one half of the fish that were fed either entire hepatopancreas or tips of hepatopancreas from experimentally infected shrimp. Feeding either the entire alimentary tract proximal to the first abdominal segment or any portion of that section from experimentally infected shrimp produced infections in nearly all tested fish. Feeding portions of the cephalothorax without any attached hepatopancreas or alimentary tract failed to produce an infection. Feeding killifish with wild grass shrimp from an enzootic area produced infections in only a fourth of the fish sample; however, feeding experimentally infected wild, laboratory-reared, and juvenile grass shrimp produced infections in nearly all fish. Palaemonid shrimps other than P. pugio also can serve as intermediate hosts for E. funduli, and these shrimps include Palaemonetes vulgaris, P. paludosus, P. kadiakensis, and Macrobrachium ohione. In contrast, a penaeid shrimp, mysidacean, amphipod, and crab fed liver with sporulated oocysts did not produce infections when fed to killifish.  相似文献

Summary The cross-sectional area of rhabdomeres in the compound eye of the blowfly, Lucilia, was found to remain constant under 12 h light/12 h dark cyclic lighting, and 10 days constant light or darkness. It was reduced only slightly during 3 h light after 10 days darkness (by 21%), or on exposure to 2h darkness + 1.5 h light after 10 days light (by 10%). Morphological evidence indicates that shedding of photoreceptor membrane during turnover is achieved by an extracellular route, and by pinocytosis from the bases of the microvilli. The photoreceptor membrane shed by both mechanisms appears to accumulate in multivesicular bodies. The amount of photoreceptor membrane shedding, as indicated by numbers of multivesicular bodies, is constant throughout the day and night on cyclic lighting, decreases in constant darkness, but returns to its normal level after an exposure to 3 h light subsequent to 10 days darkness.  相似文献

Germination occurs usually in response to multiple environmental cues. Seeds with the ecophysiological ability to simultaneously sense the previous presence of fire and appropriate levels of temperature, light and soil nitrate could restrict germination to postfire, winter and competition-free microhabitats, where the potential for seedling survival is enhanced. Germination responses of 16 species with a range of life forms, fire responses and seed weights were determined under controlled conditions of 15°C temperature, a 12 h light cycle, exposure to 1 g L?1 nitrate solution, and six conditions of light quality (white, blue, yellow, red, far-red light and darkness). Germination in Oenothera stricta, a weedy naturalized ephemeral, and two small-seeded indigenous Asteraceae species of mulga woodlands, Leucochrysum fitzgibbonii and Craspedia sp., were enhanced by white, yellow or red light compared with germination achieved in the dark, or under far-red or blue light. In red light, KNO3 further enhanced germination of these positively photoblastic species. The germination response of Trachyandra divaricata, a naturalized herb of sandy, seaside locations, and several native jarrah forest legumes (four Acacia species, Bossiaea aquifolium, Gompholobium marginatum and Sphaerolobium vimineum) proved to be negatively photoblastic. Of these seven negatively photoblastic herb and shrub species, exposure to KNO3 overcame the inhibition of light only in the resprouter species, Acacia lateriticola. In the serotinous, negatively photoblastic tree species, Corymbia calophylla and Eucalyptus marginata, KNO3 seemed to be required before the negative response to light exposure was recorded. A dose–curve experiment on two positively photoblastic and three negatively photoblastic species indicated that although KNO3 exposure affected germination in all species, different concentrations of KNO3 (0, 0.5, 1, 2, and 5 g L?1) produced different levels of response. Detailed studies with additions of KNO3 (1 g L?1) and the growth hormone, gibberellic acid (GA3; 50 mg L?1), showed that increased germination percentages of the positively photoblastic species, Oenothera stricta, occurred in the light, but blocking endogenous gibberellic synthesis with paclobutrazol, or adding exogenous GA3 or KNO3 had no effect on the light-induced germination levels. In the negatively photoblastic species Trachyandra divaricata, additions of KNO3 and GA3 had no influence on the germination inhibition induced by exposure to light nor did blocking endogenous GA synthesis. The 16 species growing naturally in Western Australia, Australia show a range of germination responses to environmental conditions, but depending on their natural habitat, the ecophysiology of each species appears to be optimized for subsequent seedling survival.  相似文献

The simultaneous study of the temporal dynamics of foraging behaviour, diet and seed abundance is essential to assess the way in which resources affect the behaviour and ecology of harvester ants. Here, we evaluate how fluctuations in grass seed abundance during three consecutive growing seasons influenced the foraging behaviour and diet of the harvester ants Pogonomyrmex rastratus, P. mendozanus and P. inermis in the central Monte desert, Argentina. Seed abundance of the most consumed grasses varied greatly through ant activity season, and ants altered their foraging behaviour in response to those changes. Foragers spent more time travelling and searching for food, and their foraging trips took longer during the low seed availability season. Foraging distance was very similar among species and, contrary to our expectations, did not vary between seasons. Foraging success of P. rastratus and P. inermis increased during the high availability season. This matched the seasonal pattern of foraging activity, suggesting that colonies may detect seed abundance and regulate their foraging effort with the rate of forager success. Although grass seeds were the main item in the diet of the three species, P. mendozanus, and to a lesser extent P. rastratus, turned more generalist when grass seeds were scarce. In contrast, P. inermis showed a very narrow diet breadth, only harvesting grass seeds in both seasons. Our results indicate the relevance of seed availability on foraging behaviour of harvester ants, which should be taken into account when predicting and evaluating the effect of ants on seed resources as well as numerical responses of harvester ant populations to the temporal and spatial variations in grass seed abundance.  相似文献

We aimed to determine the ecological role of three seed morphs observed for the first time in a desert population of Lotononis platycarpa (Fabaceae), with respect to their germination requirements. Seeds sorted by seed coat colour (olive green, orange and brown) were germinated under laboratory conditions under two photoperiods (12/12‐h light and continuous dark) and three alternating temperature regimes (15/25, 20/30, 25/35°C). We found that the three distinct seed types differ in their seed mass, germination percentage and speed of germination. Overall, the light‐incubated seeds germinated with higher percentages than seeds in the total darkness. Furthermore, seeds with orange coat germinated with higher percentages at 15/25 and 25/35°C (up to 60%, for both) and significantly faster than the other two colour morphs. Our results suggest an adaptive significance of seed colour heterogeneity in the harsh desert habitat inhabited by the study species.  相似文献

The moniliaceous fungus, Hirsutella thompsoniU grown on potato-dextrose agar (PDA) or on sterile wheat bran, was highly pathogenic to the carmine spider mite, Tetranychus cinnabarinus, and, grown on PDA, to the oriental spider mite, Eutetranychus orientalis. The fungus penetrated the mites' integument mainly through the legs and formed hyphal bodies in chains in the haemolymph. Hyphae, on which the spores were produced, began to emerge through the genital and anal apertures and then all over the body. Various mycophagous mites, a predaceous species and the tick Argas persicus were immune. H. thompsonii grew well on cadavers of diverse insects and mites. The fungus killed most mites and quickest, usually by the 2nd day, at 25o, 27o and 30 oC; least at 13o and 35 oC. It sporulated best on mites at 24o, 27o and 30 oC (full sporulation at 27 oC took place within 12 h after death); it was good, but slower, at 13o and 35 oC. The fungus germinated, penetrated mites and sporulated there very poorly below 100% r.h. When the daily number of hours at 100% r.h. was reduced from 24 to 18 or 6 h, fungus-associated mortality dropped greatly. In the field, use of the fungus for mite control would be suitable particularly in tropical and humid subtropical areas, and its success in Israel would not be assured. In glasshouses, H. thompsonii could induce epizootics in Israel and elsewhere only if humidities were raised to saturation and the daytime temperature kept below 37 oC.  相似文献

Grasses can be important regulators of species diversity and ecosystem processes in prairie systems. Although C4 grasses are usually assumed to be ecologically similar because they are in the same functional group, there may be important differences among species or between seed sources that could impact restorations. I tested whether C4 grass species identity, seed source, or grass species richness scales to influence aboveground net primary productivity (ANPP), resistance to weed invasion, or establishment of subordinate prairie species during restoration. Plots in western Iowa, United States, were planted with equal‐sized transplants of one of five common grass species (Panicum virgatum L., Sorghastrum nutans (L.) Nash, Andropogon gerardii Vitman, Schizachyrium scoparium (Michx.) Nash, and Bouteloua curtipendula (Michx.) Torrey) either from local seed or from cultivar seed sources. These plots were compared to plots containing all five species in mixture and to nonplanted plots. Differences in ANPP were found among species but not between cultivars and noncultivars or between monocultures and mixtures. Panicum virgatum, S. nutans, and S. scoparium were more productive than A. gerardii and B. curtipendula. Weed invasion was much higher when plots were not planted with grasses. Schizachyrium scoparium allowed greater establishment of subordinant prairie species than all other focal grass species. There were two separate mechanisms by which grasses suppressed prairie species establishment either (1) by growing tall and capturing light or (2) by quickly filling in bare space by spreading horizontally through rhizome growth in short species. These results suggest that high ANPP can be found with noncultivar plantings during the first 2 years after planting and that subordinate species establishment is most likely when shorter bunchgrasses such as S. scoparium are dominant.  相似文献

The infection of grasshoppers by naturally occurring, entomopathogenic fungi was monitored at two sites in Malanville, northern Benin, Africa. Grasshoppers were collected and recorded from the sites between June and December 1992 and all of them, barring the first instars, were incubated in ventilated cages. At the first site, 1343 individuals of 35 grasshopper species were incubated, and at the second site, 857 individuals of 36 grasshopper species were incubated. Three hyphomycete fungi (Deuteromycotina: Hyphomycetes), Metarhizium flavoviride Gams and Rozsypal, Beauveria bassiana (Bals.) Vuillemin and Sorosporella sp. were found infecting grasshoppers. The average incidence of M. flavoviride infection was 2.9% and 1.8% at the two sites for all host species. M. flavoviride sporulated on most grasshopper cadavers within 10 days of collection. B. bassiana and Sorosporella sp. were only collected from one and five grasshopper individuals respectively. A significant difference was noted in the time to death of small grasshopper species infected with M. flavoviride compared to larger species. At one site, M. flavoviride infection was positively correlated with rainfall during the 10‐day period in which samples were taken.  相似文献

The Qinghai-Tibetan Plateau is located in the ‘Third Pole’ of the world, characterized by a harsh environment. Despite this, the alpine meadow ecosystem have developed over a wide area but serious grassland degradation is threatening the ecological environment on the Plateau. Recruitment of new plants to the population, via germination and establishment, is vital to plant community survival. Previous work on the seeds in this area has mainly focused on community-wide germination strategies, seed germination characteristics and their correlations with seed size and seed mass. However, there have been no studies on the effects of soaking in rumen contents on the plant seed germination characteristics of alpine meadow species. The present study had two main objectives: (i) to determine the effect of fresh rumen content from yaks on seed germination characteristics and seedling growth of species common to the eastern Tibetan Plateau alpine meadow, and (ii) to develop an effective method to enhance seed germination. Seeds of 11 common species were collected together with fresh rumen content from three yaks that grazed there. Seed germination tests were conducted after they had been soaked in rumen content for one of six soaking periods (12, 24, 36, 48, 60 or 72 h). The seeds were incubated under natural light conditions of 8 h light at 25 °C and 16 h darkness at 15 °C, for the germination period of 34 days. The results showed that seed germination and seedling growth were affected by soaking time, seed coat completeness and seed type. After soaking in rumen content, the germination percentages of scarified (peeled or with the seed coat cut through) seeds of some species (true seeds Oxytropis ochrocephala and Medicago ruthenia var inschanica, nutlet Carex enervis, achenes Anemone rivularis and Polygonum sibiricum) and complete seeds of C. enervis, and A. rivularis were improved but the duration of soaking was also important. Seed germination of caryopsis Achnatherum inebrians (a toxic grass) was significantly inhibited by any exposure to rumen fluids. Scarified seeds generally had higher germination percentages than complete ones after treatment, but with the increase in soaking time, germination percentages declined and scarified seeds were more sensitive to the treatment than the complete seeds. After soaking in yak rumen content, the germination indices of scarified M. ruthenia at 12 h treatment, O. ochrocephala and achene Rumex acetosa at 12–24 h treatment, nutlet Kobresia humilis at 24 h treatment, P. sibiricum at 24–48 h treatment, C. enervis at 12–48 h treatment and A. rivularis at 12–60 h treatment were significantly higher than the control (P < 0.05), while the germination indices of complete C. enervis seeds at 12 h and 36 h treatment, and A. rivularis at 12–60 h treatment were significantly higher compared with the control. The germination indices of other species gradually decreased with the increase in soaking time. We concluded that yak rumen digestion could enhance, inhibit or not affect seed germination and seedling growth of the alpine meadow species, which might influence seedling recruitment, interspecific competition, and the plant community structure of the eastern Tibetan Plateau alpine meadow. Overall, yak digestion has a positive effect on alpine meadow seed germination and seed dispersal.  相似文献

Invasion of alien plant species (IAS) represents a serious environmental problem, particularly in Europe, where it mainly pertains to urban areas. Seed germination traits contribute to clarification of invasion dynamics. The objective of this research was to analyze how different light conditions (i.e., 12-hr light/12-hr darkness and continuous darkness) and temperature regimes (i.e., 15/6°C, 20/10°C and 30/20°C) trigger seed germination of Ailanthus altissima (AA), Phytolacca americana (PA) and Robinia pseudoacacia (RP). The relationship between seed germination and seed morphometric traits was also analyzed. Our findings highlight that temperature rather than light was the main environmental factor affecting germination. RP germinated at all tested temperatures, whereas at 15/6°C seeds of AA and PA showed physiological dormancy. RP had a higher germination capacity at a lower temperature, unlike AA and PA, which performed better at the highest temperatures. Light had a minor role in seed germination of the three species. Light promoted germination only for seeds of PA, and final germination percentage was 1.5-fold higher in light than in continuous darkness. Seed morphometric traits (thickness [T], area [A] and volume [V]) had a significant role in explaining germination trait variations. The results highlight the importance of increasing our knowledge on seed germination requirements to predict future invasiveness trends. The increase in global temperature could further advantage AA and PA in terms of germinated seeds, as well as RP by enhancing the germination velocity, therefore compensating for a lower germination percentage of this species at the highest temperatures.  相似文献

Background and Aims

The Campanulaceae is a large cosmopolitan family, but is understudied in terms of germination, and seed biology in general. Small seed mass (usually in the range 10–200 µg) is a noteworthy trait of the family, and having small seeds is commonly associated with a light requirement. Thus, the purpose of this study was to investigate the effect of light on germination in 131 taxa of the Campanulaceae family, from all five continents of its distribution.

Methods

For all taxa, seed germination was tested in light (8 or 12 h photoperiod) and continuous darkness under constant and alternating temperatures. For four taxa, the effect of light on germination was examined over a wide range of temperatures on a thermogradient plate, and the possible substitution of the light requirement by gibberellic acid and nitrate was examined in ten taxa.

Key Results

For all 131 taxa, seed germination was higher in light than in darkness for every temperature tested. Across species, the light requirement decreased significantly with increasing seed mass. For larger seeded species, germination in the dark reached higher levels under alternating than under constant temperatures. Gibberellic acid promoted germination in darkness whereas nitrates partially substituted for a light requirement only in species showing some dark germination.

Conclusions

A light requirement for germination, observed in virtually all taxa examined, constitutes a collective characteristic of the family. It is postulated that smaller seeded taxa might germinate only on the soil surface or at shallow depths, while larger seeded species might additionally germinate when buried in the soil if cued to do so by fluctuating temperatures.  相似文献

Sporangia of three isolates of Phytophthora ramorum representing three different clonal lineages were subjected to relative humidity (RH) levels between 80 and 100% for exposure periods ranging from 1 to 24 h at 20°C in darkness. Plastic containers (21.5 × 14.5 × 5 cm) were used as humidity chambers with 130 ml of glycerine solution added to each container. Glycerine concentrations corresponded to 100, 95, 90, 85 and 80% RH based on refractive index measurements. Sporangia suspensions were pipeted onto nitrile mesh squares (1.5 × 1.5 cm, 15 micron pore size) which were placed in the humidity chambers and incubated at 20°C in darkness. Following exposure periods of 1, 2, 4, 8, 12 and 24 h, mesh squares were inverted onto Petri dishes of selective medium and sporangia germination assessed after 24 and 48 h. At 100% RH, we observed a mean value of 88% germination after 1 h exposure declining to 18% germination following 24 h incubation. At 95% RH, a steeper decline in germination was noted, with means ranging from 79% at 1 h to less than 1% at 24 h exposure. At 90% RH, no germination was noted after 8 or more h exposure, and values were 57%, 22% and 3% germination for the 1, 2 and 4 h exposures, respectively. Germination was only observed at 1 h exposure for both the 85% RH treatment (52% germination) and the 80% RH treatment (38% germination). The three isolates responded similarly over the range of RH values tested. The germination response of P. ramorum sporangia to RH values between 80% and 100% was comparable to that reported for other Phytophthora species. Knowledge of conditions that affect Pramorum sporangia germination can shed light on pathogenesis and epidemic potential and lead to improved control recommendations.  相似文献
