The heterotrophic plate counts of 15 brands of bottled non-carbonated mineral waters were examined and found to be generally high and variable. Four selective or enrichment media for the enumeration of coliforms (m-Endo LES and m-lauryl sulphate agar) and Pseudomonas aeruginosa (cetrimide-nalidixic acid agar and malachite green broth) were used to isolate several species of Gram-negative bacteria. Strains identified as CDC gr IVc-2 and Comamonas (Ps.) acidovorans were the two most commonly isolated. Considerable variation in populations was seen between the brands, as well as between two batches of the same mineral water.  相似文献

Matric potential evaluations and measurements for gelled substrates   总被引:1,自引:0,他引:1  
A new method for evaluating the matric potential of gelled media has been developed. The method allows the derivation of the matric potential as a limit of a series of measurements of water potential values from gelled media prepared without added components, from agar powders progressively cleaned of mineral impurities. Three commercial agar brands were tested, and for these the matric potential was found to contribute only between 1 and 2% of their total water potential. Thermodynamic features relating matric and osmotic potentials are described. New hypotheses for understanding the water flux mechanism from gel to tissue cultured explants are discussed. Movement of water along polymeric chains is postulated to be a facilitated step in comparison with bulk movement.  相似文献

The microbiological quality of four brands of bottled water sold in retail outlets in Nigeria were assessed by routine methods in 90 samples. Samples of two brands were acidic in the pH range 3.5-5.9. Faecal coliforms and streptococci were not recovered from any sample. Heterotrophic plate counts (HPC) numbered 50-800 cfu/ml in two brands, A and B, and 100-87,000 cfu/ml in C and D. Component colony types among the HPC bacteria in brands C and D produced water-soluble, fluorescent pigments on colony count and other agar media, and occurred in 11 of 16 batches: their numbers varied from 60 to 82,000 cfu/ml. Presumptive antibiotic-resistant proportions of the HPC bacteria were also recovered from brands C and D on agar amended with ampicillin, penicillin or streptomycin. Five isolates from the green pigmented colonies, representing five batches of brands C and D were satisfactorily identified as strains of Pseudomonas aeruginosa. These strains resisted between four and nine of 14 antibiotics tested. Resistance to tetracycline was eliminated in one isolate when it was cured by incubation at 42 degrees C for 100 h, but gel electrophoretic resolution of the DNA did not reveal presence of a plasmid. Strains of Bacillus were the second most commonly isolated bacteria; they were the only colony types in most samples with low HPC counts.  相似文献

Several differences concerning bacterial species, opportunistic pathogens, elements of the resistome as well as variations concerning the CFU/mL counts were identified in some of the five most marketed bottled mineral water from Araraquara city, São Paulo, Brazil. Two out of five brands tested were confirmed as potential source of opportunistic pathogens, including Mycobacterium gordonae, Ralstonia picketti and Burkholderia cepacia complex (Bcc). A total of one hundred and six isolates were recovered from four of these bottled mineral water brands. Betaproteobacteria was predominant followed by Alphaproteobacteria, Gammaproteobacteria and Firmicutes. Ninety percent of the bacteria isolated demonstrated resistance to seventeen of the nineteen antimicrobials tested. These antimicrobials included eight different classes, including 3rd and 4th generation cephalosporins, carbapenems and fluoroquinolones. Multidrug resistant bacteria were detected for fifty-nine percent of isolates in three water brands at counts up to 103 CFU/ml. Of major concern, the two bottled mineral water harboring opportunistic pathogens were also source of elements of the resistome that could be directly transferred to humans. All these differences found among brands highlight the need for continuous bacteriological surveillance of bottled mineral water.  相似文献

Acidity affected which members of an indigenous soil population of Rhizobium trifolii nodulated Trifolium subterraneum L. cv. Mt. Barker. In three experiments involving plants grown either in mineral salts agar adjusted to pH 4.8 or 6.8 and inoculated with a soil suspension or grown directly in samples of unamended soil (pH 4.8) or soil amended with CaCO(3) (pH 6.4), 121 of 151 isolates of R. trifolii were placed into four serogroups. Seventy-nine of these isolates were placed into two serogroups (6 and 36) whose nodulating ability was affected by the pH of the plant root environment. Representatives of serogroup 6 occupied the greatest percentage of the nodules at the low pH in both mineral salts agar (77%) and in unlimed soil (47 and 57%). The same serogroup was a minor nodule occupant at the higher pH in mineral salts agar (0%) and in limed soil (0 and 10%). In contrast, serogroup 36 was virtually absent in nodules formed at the low pH, whereas it was the dominant serogroup at the higher pH in both mineral salts agar (32%) and in limed soil (35 and 49%). Despite the isolates from within each serogroup being antigenically identical, separation of cellular proteins by sodium dodecyl sulfate-polyacrylamide gradient gel electrophoresis revealed four and six different gel types within serogroups 6 and 36, respectively. Isolates represented by one or two gel types dominated the contribution of each serogroup to the nodule population. Further evidence for differences between isolates within each gel type were revealed from measurements of symbiotic effectiveness.  相似文献

Summary The success of in vitro culture is related to several factors. Beside factors associated with the plant material or the medium composition, the physicochemical characteristics of gelled media can play an important role. In this paper, the latter aspect has been considered and the nature of agar powders has been investigated. Moreover, the process of gel formation for three different media and the availability of water and minerals for the corresponding gels have been studied. Analysis of agar powders showed that they can contain different amounts of impurities and the dialysis of these powders suggested that the impurities might be available to the tissues. Thermal analysis on the hygroscopic properties of the agar brands suggest the importance of these data to obtain comparable and reproducible gelled media. The study on the process of formation of gelled media indicates that there is a critical temperature Tss which can be used to control the gel processing. In fact, at this temperature, agar powders in water transform into a sol status through a rapid shift of electrical conductivity. Water potential of the medium, water loss from gels over the culture period, and the ease of releasing liquid from gels under pressure were shown to be different for different agar brands. A different availability of water and minerals in Murashige and Skoog medium was deduced from the gels prepared with three agar brands (Oxoid, Merck, and Roth).  相似文献

A total of 292 imported and domestic bottled mineral waters (90 brands) obtained from consumers and retailers were examined, by eye, for observable microbial foreign bodies. Fungal and bacterial foreign contaminants were found in 45 samples of water (20 brands) and in 14 samples of water (10 brands), respectively. Of the samples of water found to be contaminated, 41 (22 brands) were imported and 18 (8 brands) were produced domestically. Of 22 brands that were contaminated, 20 (91%) had been sterilized by at least one method. Forty-eight (98%) of 49 samples confirmed with foreign bodies were less than 1 year old. Among the moulds isolated the most predominant genus was Penicillium, followed by Acremonium and Cladosporium. The samples that contained fungi were less contaminated by bacteria than those that contained observable bacterial foreign bodies.  相似文献

In tissue-cultured Cynara scolymus L. (globe artichoke) vitrification (hyperhydric transformation) can only be overcome by increasing the concentration of agar. However, with increasing concentrations of Difco Bacto agar (8–15 g/I) the availability of labelled kinetin is decreased. There is some evidence for postulating that cytokinins under inductive conditions of low agar concentration or high matrix potential are evocators of vitrification.
Both the brand and concentration of agar also affect the chemical and physical characteristics of a culture medium.
Impurities introduced with the agar are responsible for significant differences in the concentration of an element in comparable media with different levels of agar, and are easily detected by conductance.
Penetrometer measurements also show large ranges in solidity of media with increasing concentrations, not only within the type of agar, but also for the same concentration within different brands.  相似文献

The organisms from heterotrophic plate counts of four brands of bottled non-carbonated mineral waters were analysed by ribosomal ribonucleic acid gene restriction patterns in addition to identification by customary techniques. Five commercialized bottles of each brand were examined once in the year 1989–90. The total bacterial flora was divided into two main groups: the pseudomonads and the unidentified strains. Whereas phenotypic results did not reveal any significant differences between the four brands, rRNA gene restriction patterns were discriminatory. Among the 73 strains studied, 45 distinct patterns were observed, with only three common to several springs. These results reveal, at one determined time, a remarkable biological specificity of each brand of water.  相似文献

A rapid method is described for the detection of cellulolytic, thermophilic actinomycetes involving the use of a dyed cellulose substrate (cellulose-azure) in an agar medium. The test was performed in test tubes in which agar containing cellulose-azure was layered on top of a basal medium (mineral salts agar) resulting in two distinct layers. After inoculation, and incubation at 50°C, strains which were cellulolytic caused release substrates for such a test.  相似文献

Organic gelling agents such as agar and agarose provide a heterotrophic substrate for growth of illuminated tobacco callus. When green cells are incubated in CO2-free air on a medium lacking sucrose but solidified with 1% agar, an increase in relative dry weight is sustained through two passages. Similar results with different inoculum sources, and with three brands of agar and two forms of agarose, suggest this is a general phenomenon. A fully autotrophic culture system was developed employing polyurethane pads to support cells in a liquid medium lacking sucrose. Growth was negligible in two passages in CO2-free air, and increased with each added increment in CO2 concentration.  相似文献

We surveyed 15 different brands of eye shadow on display for customer use in different retail stores for microbial contamination. This was the first reported microbial surveillance of in-use eye shadow display testers in retail establishments. Cultures were obtained at each retail store. Sterile dacron swabs were rolled and rubbed over the entire used surface of each shadow, and each inoculum was streaked onto the surfaces of blood agar plates. Of the 1,345 individual samples obtained, 67% were contaminated with one or more species of microorganisms representing the genera Staphylococcus, Micrococcus, Corynebacterium, Acinetobacter, Bacillus, and Moraxella. We also purchased two different brands of water-miscible eye shadows in replicate unit containers. Each brand was challenged separately with a few hundred to several thousand colony-forming units of Staphylococcus aureus, Pseudomonas aeruginosa, or Acinetobacter calcoaceticus. Both brands permitted growth of P. aeruginosa but not growth of S. aureus. A. calcoaceticus was inhibited after inoculation into one brand. With the other brand, the inoculum of Acinetobacter multiplied in one of the two different lots tested. This experimental challenge procedure can serve as a useful model system for studying the behavior of microbes in eye shadows and similar matrices.  相似文献

The spores ofBacillus cereus can be germinated in reasonable synchrony in rich media. The survival of germinating cells after UV irradiation is strongly affected by repair. When plating on mineral agar a linear decrease of survival is obtained between 20–60 min of germination, whereas the survival on nutrient agar shows only minor fluctuations. The difference in survival on these two media which reached 1–2 orders of magnitude is due mostly to the shape of the shoulder on the survival curve. Survival on mineral agar may be increased by postincubation in complex media or by preincubation in mineral media. It was concluded that repression and derepression of biosynthetic pathways was responsible for the different efficiency of repair.  相似文献

The influence of the mineral composition and acidity of media, saccharose level, brand and quantity of agar onin vitro shoot rooting of P 60 and P 2 apple and quince S 1 root-stocks were compared. Among the tested salt compositions (full WMP and 1/2 WMP, full MS and 1/2 MS) the most suitable for rooting was composition of WMP. Further modifications in quantity of nitrogen, H3BO3 CaCl2 and MgSO4 of WMP medium did not have a positive effect on rooting. 30 g l?1 saccharose gave better results than 20 g l?1. Acidity of WMP medium tested in a pH range of 4.0 to 6.5 did not affect rooting of P 60 rootstock but media adjusted to pH 4.5 and 5.0 were better for rooting of recalcitrant P 2 rootstock. Among 4 agar brands (Difco Bacto, Oxoid No.1, Japanese commercial fibre and Japanese commercial powder) tested at 6 g l?1, the most effective was the last one, but its quantity had no effect in the range of 3 to 10 g l?1 for rooting of P 60 rootstock.  相似文献

In the present paper the ability of calibration free laser induced breakdown spectroscopy (CF-LIBS) as a quality control tool to monitor the composition of different minerals present in food supplement samples belonging to Indian brands (brand-A and brand-B) has been demonstrated. LIBS spectra of these two food supplements (brand-A and brand-B) available in the form of tablet have been recorded. As reported by manufacturers of these two food supplements, LIBS spectra of brand-A contains the spectral signatures of minerals like Ca, Mg, C, P, Zn, Fe, Cu, and Cr whereas LIBS spectra of brand-B shows the presence of spectral lines like Ca, Mg and C. The spectral signatures of Na and K are also found in both brands whereas spectral signature of Ti is observed only in brand-B but these elements are not mentioned on the nutritional label of the brands. The quantitative analysis of mineral contents in food supplements has been done using CF-LIBS for brand A and brand B to verify the content of the minerals reported by the manufacturer of the food supplements. Our results show that Ca and Mg are the main matrix elements of these brands. The concentration of minor and trace elements estimated using CF-LIBS technique is found in agreement with the reported nutritional values of both the brands. The concentration of major elements Ca and Mg are also estimated from Atomic Absorption Spectroscopy which is in close agreement with CF-LIBS result.  相似文献

Interactions between selenium and sulphur nutrition in Arabidopsis thaliana   总被引:14,自引:0,他引:14  
Selenium (Se) is an essential plant micronutrient, but is toxic at high tissue concentrations. It is chemically similar to sulphur (S), an essential plant macronutrient. The interactions between Se and S nutrition were investigated in the model plant Arabidopsis thaliana (L.) Heynh. Arabidopsis plants were grown on agar containing a complete mineral complement and various concentrations of selenate and sulphate. The Se/S concentration ratio in the shoot ([Se](shoot)/[S](shoot)) showed a complex dependence on the ratio of selenate to sulphate concentration in the agar ([Se](agar)/[S](agar)). Increasing [S](agar) increased shoot fresh weight (FW) and [S](shoot), but decreased [Se](shoot). Increasing [Se](agar) increased both [Se](shoot) and [S](shoot), but reduced shoot FW. The reduction in shoot FW in the presence of Se was linearly related to the shoot Se/S concentration ratio. These data suggest (i) that Se and S enter Arabidopsis through multiple transport pathways with contrasting sulphate/selenate selectivities, whose activities vary between plants of contrasting nutritional status, (ii) that rhizosphere sulphate inhibits selenate uptake, (iii) that rhizosphere selenate promotes sulphate uptake, possibly by preventing the reduction in the abundance and/or activity of sulphate transporters by sulphate and/or its metabolites, and (iv) that Se toxicity occurs because Se and S compete for a biochemical process, such as assimilation into amino acids of essential proteins.  相似文献

Effects of nitric oxide and nitrogen dioxide on bacterial growth   总被引:3,自引:0,他引:3  
The effects of low concentrations of nitric oxide (NO) and nitrogen dioxide (NO2) on actively dividing cultures of Staphylococcus aureus, Micrococcus luteus, Micrococcus roseus, Serratia marcescens, Bacillus subtilis, Bacillus circulans, Bacillus megaterium, and Bacillus cereus were studied. Fresh cultures of each organism were incubated for 24 h at 25 degrees C on both nutrient agar and mineral salts glucose agar plates under atmospheres containing various low concentrations of NO in air (0 to 1.9 ppm [0 to 2.0 micrograms/g of air]), NO2 in air (0 to 5.5 ppm [0 to 8.8 micrograms/g of air]), or NO and NO2 in air. Bacteria grown under air only were used as controls. After incubation, the colonies that developed on the plates were counted. None of the bacteria tested was affected by NO or NO2 at the indicated concentrations while growing on nutrient agar. Serratia marcescens, B. circulans, B. subtilis, B. megaterium, and B. cereus grown on mineral salts glucose agar were not significantly affected by NO or NO2. Low concentrations (0 to 1.9 ppm) of NO were bacteriostatic to log-phase cultures of M. roseus, M. luteus, and Staphylococcus aureus grown on mineral salts glucose agar. Bacteriostatic activity over a 24-h interval was maximal at an initial NO concentration of 1 ppm. Appreciable amounts of NO2 were produced in 24 h at initial NO concentrations greater than 1 ppm. These results suggest that NO2 may reduce the bacteriostatic activity of NO. Low concentrations (0 to 5.5 ppm) of NO2 in air did not affect any of the bacteria tested. At these low concentrations, NO affected bacterial growth, although NO2, NO2-, and NO3- did not. In addition, it was determined that the bacteriostatic activity observed in this study was not due to an increase in the acidity of the medium.  相似文献
