The effects of essential oils isolated from Douglas fir needles on sheep and deer rumen microbial activity were tested by use of an anaerobic manometric technique. Rumen microorganisms were obtained from a sheep which had been fed mainly on alfalfa hay and dried range grass. One deer used in this study had access to Douglas fir trees the year around, whereas the other deer had no access to Douglas fir. All of the monoterpene hydrocarbons isolated from Douglas fir needles—α-pinene, β-pinene, limonene, myrcene, camphene, Δ3-carene, and terpinolene—promoted only slightly or had no effect on deer rumen microbial activity, whereas all of them promoted activity in sheep rumen microbes, except Δ3-carene and terpinolene, which inhibited activity. Of the oxygenated monoterpenes, all monoterpene alcohols—α-terpineol, terpinen-4-ol, linalool, citronellol, and fenchyl alcohol—strongly inhibited the rumen microbial activity of both sheep and deer. Monoterpene esters (bornyl acetate) produced mild inhibition for both sheep and deer microbes, and citronellyl acetate inhibited rumen microbial activity in sheep, whereas it promoted activity in both deer. Monoterpene aldehyde (citronellal) inhibited the activity of rumen microbes from both sheep and deer having no access to Douglas fir from the Hopland Field Station, whereas they produced no effect upon the deer having access to Douglas fir from the Masonite forest. Rumen microbial activity for sheep and deer was promoted slightly with aliphatic ester (ethyl-n-caproate). There was a marked difference between sheep and deer rumen microbes as affected by addition of the various essential oils. The monoterpene hydrocarbons promoted activity more on sheep rumen microbes than on deer, and the monoterpene alcohols inhibited sheep rumen microbial activity more than that of deer. Furthermore, the deer rumen microbes from Hopland Field Station were affected more than the deer from Masonite forest.  相似文献

This experiment studied effects of a mixture of exogenous enzymes (ZADO®) from anaerobic bacteria on ruminal fermentation, feed intake, digestibility, as well as milk production and composition in cows fed total mixed rations (TMRs; 0.7 corn silage and 0.3 of a concentrate mixture). Twenty lactating multiparous Brown Swiss cows (500 ± 12.4 kg live weight) were randomly assigned into two experimental groups of 10 immediately after calving and fed a TMR with or without (CTRL) addition of 40 g/cow/d of enzymes for 12 weeks. Addition of enzymes increased (P<0.05) rumen microbial N synthesis. Intake of dry matter (DM) and organic matter (OM) was positively influenced (P<0.05) by supplementation, and digestibility of all nutrients was higher (P<0.05) in the total tract of supplemented cows, although the magnitude of the improvement varied among nutrients, with the highest improvement in aNDFom and ADFom (418–584 and 401–532 g/kg respectively; P<0.05) than the other nutrients. Supplementation of enzymes also increased (P<0.05) rumen ammonia N and total short chain fatty acid (SCFA) concentrations, and individual SCFA proportions were also altered with an increase in acetate (61.0–64.8 mol/100 mol; P=0.05) before feeding, and acetate and propionate increased 3 h post-feeding (60.0–64.0 and 18.3–20.8 mol/100 mol respectively; P<0.05). Milk and milk protein production was higher (12.8–15.7 and 0.45–0.57 kg/d respectively; P<0.05) for cows fed the ZADO® supplemented diet. This exogenous enzyme product, supplemented daily to the TMR of cows in early lactation, increased milk production due to positive effects on nutrient intake and digestibility, extent of ruminal fermentation and microbial protein synthesis.  相似文献

The Douglas fir terpene α-pinene was shown to inhibit the growth of a variety of bacteria and a yeast. Other terpenes of the Douglas fir, including limonene, camphene, and isobornyl acetate, were also inhibitory to Bacillus thuringiensis. All terpenes were inhibitory at concentrations normally present in the fir needle diet of Douglas fir tussock moth larvae. The presence of such terpenes in the diet of these insects was found to strongly influence the infectivity of B. thuringiensis spores for the Douglas fir tussock moth larvae. The terpene α-pinene destroyed the cellular integrity and modified mitochondrial activity in certain microorganisms.  相似文献

Terpenoids are characteristic constitutive and inducible defense chemicals of conifers. The biochemical regulation of terpene formation, accumulation, and release from conifer needles was studied in Norway spruce [Picea abies L. (Karst)] saplings using methyl jasmonate (MeJA) to induce defensive responses without inflicting physical damage to terpene storage structures. MeJA treatment caused a 2-fold increase in monoterpene and sesquiterpene accumulation in needles without changes in terpene composition, much less than the 10- and 40-fold increases in monoterpenes and diterpenes, respectively, observed in wood tissue after MeJA treatment (D. Martin, D. Tholl, J. Gershenzon, J. Bohlmann [2002] Plant Physiol 129: 1003-1018). At the same time, MeJA triggered a 5-fold increase in total terpene emission from foliage, with a shift in composition to a blend dominated by oxygenated monoterpenes (e.g. linalool) and sesquiterpenes [e.g. (E)-beta-farnesene] that also included methyl salicylate. The rate of linalool emission increased more than 100-fold and that of sesquiterpenes increased more than 30-fold. Emission of these compounds followed a pronounced diurnal rhythm with the maximum amount released during the light period. The major MeJA-induced volatile terpenes appear to be synthesized de novo after treatment, rather than being released from stored terpene pools, because they are almost completely absent from needle oleoresin and are the major products of terpene synthase activity measured after MeJA treatment. Based on precedents in other species, the induced emission of terpenes from Norway spruce foliage may have ecological and physiological significance.  相似文献

Despite their major contribution to the energy supply of ruminants, the production of volatile fatty acids (VFA) in the rumen is still poorly predicted by rumen models. We have developed an empirical approach, based on the interpretation of large bibliographic databases gathering published in vivo measurements of ruminal VFA production rate (PR), rates of duodenal and faecal digestion and molar percentages of VFA in the rumen. These databases, covering a wide range of intake levels and dietary composition, were studied by meta-analysis using within-experiment models. We established models to quantify response laws of total VFA-PR and individual VFA molar percentages in the rumen to variations in intake level and dietary composition. The rumen fermentable organic matter (RfOM) intake, estimated from detailed knowledge of the chemical composition of diets according to INRA Feed Tables, appears as an accurate explanatory variable of measured total VFA-PR, with an average increment of 8.03 ± 0.64 mol total VFA/kg RfOM intake. Similar results were obtained when total VFA-PR was estimated from measured apparent RfOM (total VFA-PR/RfOM averaging 8.3 ± 1.2 mol/kg). The VFA molar percentages were related to dry matter intake and measured digestible organic matter (OM), digestible NDF and rumen starch digestibility, with root mean square error of 1.23, 1.45, 0.88 and 0.41 mol/100 mol total VFA for acetate, propionate, butyrate and minor VFA, respectively, with no effect of pH on the residuals. Stoichiometry coefficients were calculated from the slopes of the relationships between individual VFA production (estimated from measured apparent RfOM and individual VFA molar percentages) and measured fermented fractions. Coefficients averaged, respectively, 66, 17, 14 and 3 mol/100 mol for NDF; 41, 44, 12 and 4 mol/100 mol for starch; and 46, 35, 13 and 6 mol/100 mol for crude protein. Their use to predict VFA molar percentages appear relevant for most dietary conditions, that is, when the digested NDF/digested OM ratio exceeded 0.12. This study provides a quantitative review on VFA yield in the rumen. It contributes to the development of feed evaluation systems based on nutrient fluxes.  相似文献

Selected monoterpenes inhibited methane oxidation by methanotrophs (Methylosinus trichosporium OB3b, Methylobacter luteus), denitrification by environmental isolates, and aerobic metabolism by several heterotrophic pure cultures. Inhibition occurred to various extents and was transient. Complete inhibition of methane oxidation by Methylosinus trichosporium OB3b with 1.1 mM (−)-α-pinene lasted for more than 2 days with a culture of optical density of 0.05 before activity resumed. Inhibition was greater under conditions under which particulate methane monooxygenase was expressed. No apparent consumption or conversion of monoterpenes by methanotrophs was detected by gas chromatography, and the reason that transient inhibition occurs is not clear. Aerobic metabolism by several heterotrophs was much less sensitive than methanotrophy was; Escherichia coli (optical density, 0.01), for example, was not affected by up to 7.3 mM (−)-α-pinene. The degree of inhibition was monoterpene and species dependent. Denitrification by isolates from a polluted sediment was not inhibited by 3.7 mM (−)-α-pinene, γ-terpinene, or β-myrcene, whereas 50 to 100% inhibition was observed for isolates from a temperate swamp soil. The inhibitory effect of monoterpenes on methane oxidation was greatest with unsaturated, cyclic hydrocarbon forms [e.g., (−)-α-pinene, (S)-(−)-limonene, (R)-(+)-limonene, and γ-terpinene]. Lower levels of inhibition occurred with oxide and alcohol derivatives [(R)-(+)-limonene oxide, α-pinene oxide, linalool, α-terpineol] and a noncyclic hydrocarbon (β-myrcene). Isomers of pinene inhibited activity to different extents. Given their natural sources, monoterpenes may be significant factors affecting bacterial activities in nature.  相似文献

Purpose

It is widely accepted that lipopolysaccharide and volatile fatty acids (VFA) accumulate in the digestive tract of ruminants fed diets containing high portions of grain. Compared to the ruminal epithelium, the hindgut epithelium is composed of a monolayer structure that is more “leaky” for lipopolysaccharide and susceptible to organic acid-induced damage. The aim of this study was to investigate changes in epithelial structure, apoptosis and inflammatory response in the hindgut of goats fed a high-concentrate diet for 6 weeks.

Experimental Design

Eight local Chinese goats with rumen cannulas were randomly assigned to two groups: one group was fed a high-concentrate diet (65% concentrate of dry matter, HC) and the other group was fed a low-concentrate diet (35% concentrate of dry matter, LC) for 6 wks. Ruminal fluid, plasma, and hindgut mucosa tissues were collected. Histological techniques, real-time PCR and western blotting were used to evaluate the tissues structure, cell apoptosis and local inflammation in the hindguts.

Results

Feeding HC diet for 6 wks resulted in a significant decrease of ruminal pH (p<0.01), and ruminal lipopolysaccharide concentrations were significantly increased in HC goats (p<0.05). Obvious damage was observed to mucosal epithelium of the hindgut and the intercellular tight junctions in HC, but not in LC, goats. The expression of MyD88 and caspase-8 mRNA was increased in colonic epithelium of HC goats compared to LC (p<0.05), and the expression of TLR-4 and caspase-3 showed a tendency to increase. In the cecum, interleukin-1β mRNA expression was decreased (p<0.05), and caspase-3 showed a potential increase (p = 0.07) in HC goats. The level of NF-κB protein was increased in colonic epithelium of HC goats. Caspase-3 activity was elevated in both colon and cecum, whereas caspase-8 activity was statistically increased only in colon.

Conclusions

Feeding a high-concentrate diet to goats for 6 wks led to hindgut mucosal injuries via activating epithelial cells apoptosis and local inflammatory response.  相似文献

The brown midrib (BMR) gene has been reported to reduce the lignin concentration in plants, which contributed to increased fiber digestion in ruminants. Three studies were completed to compare the digestibility of a BMR mutant of sudangrass (sorghum bicolor subsp. Drummondii) versus a non-BMR (‘Piper’) variety when included in diets fed to sheep (Study 1), to complete a rumen in vitro assessment of sheep and lactating cow diets (Study 2), and to compare digestibility when included in the diet fed to lactating dairy cows (Study 3). Four wether sheep were used in a 2 × 2 Latin square experiment (Study 1) with total fecal collection to determine total tract apparent digestibility of pelleted Piper (P) and BMR (P-BMR) sudangrass hays. Forage pellets consisted of either P-BMR or P hay with added urea to meet the maintenance crude protein (CP) requirement of the sheep. Digestibility of organic matter (OM; P<0.01), dry matter (DM; P<0.01), acid detergent fiber (ADF; P<0.05), and neutral detergent fiber (aNDFom; P<0.07) was higher for P-BMR than P sudangrass. In vitro rumen digestibility of aNDFom using cattle rumen fluid was higher at 24 (P<0.01), 48 (P<0.01) and 72 h (P<0.01) of fermentation for P-BMR versus P (Study 2). Four lactating Holstein dairy cows (251 ± 30 days in milk) and fitted with ruminal and duodenal cannulae were used in a 4 × 4 Latin square experiment. Total mixed rations (TMR) contained 180 g/kg DM shredded sudangrass hay and 180 g/kg sliced alfalfa hay, but the proportion of P to P-BMR sudangrass varied as 100:0, 66:34, 34:66, or 0:100. Yields of milk and milk protein were highest at the 66:34 level (Quadratic: P=0.06 and 0.07, respectively), but composition of milk fat, protein and lactose, as well as DM intake, did not differ (Study 3), probably because forestomach and total tract apparent digestion of aNDFom and OM did not differ due to sudangrass source.  相似文献

Terpenes were analyzed in needles of pollution injured Picea abies (L.) Karst. of the Southern Black Forest (SW Germany) showing symptoms of montane yellowing. Compared to a set of healthy trees, a set of injured trees showed increased terpene hydrocarbon concentrations relative to needle fresh weight. This difference was even more pronounced when the terpene concentrations of green, healthy and yellowing, injured needles from the same internode of individual injured trees were compared. Increased terpene concentrations were observed in the basal parts of injured needles, while their distal parts exhibited a loss of terpenes. In addition, an alteration in the pattern of terpene occurrence was observable. Most striking was the changed ratio of α-pinene to β -pinene. with marked increase of the latter, in injured needles. As indicated by ultratrace analysis of isolated mesophyll. vascular tissue and resin ducts of the needles, mono- and sesquiterpenes were found exclusively in resin ducts. The other tissues were virtually free of terpenes. Isolated resin from a single resin duct contained all terpenes (monoterpene hydrocarbons, alcohols and ketones, and sesquiterpene hydrocarbons) that are typical for the needle. An individual response to air pollution of each separate short resin duct in the needle may be deduced.  相似文献

A simulation rumen model has been developed to function under non-steady state conditions in order to allow prediction of nutrient availability in dairy cows managed under discontinuous feeding systems. The model simulates availability of glycogenic, aminogenic and lipogenic nutrients to lactating dairy cows fed discontinuously. The model structure considers input of up to three different feeds fed independently at any time during the day. Feeds are described by their nitrogen (N), carbohydrate and fatty acid fractions. The N containing feed fractions include ruminally undegraded crude protein (CP), ruminally insoluble but potentially degradable CP, ruminally soluble CP and ammonia N. The feed carbohydrate fractions include ruminally undegradable neutral detergent fibre (NDF), ruminally degradable NDF, ruminally insoluble starch, ruminally soluble starch and sugars. The fatty acids in the feeds are divided between long chain fatty acids and volatile fatty acids (VFA). Additionally four pools were defined representing absorption of amino acids, glucose, long chain fatty acids and volatile fatty acids. The rumen microbial population is represented as a single pool. Besides a flexible structure, new features to the extant model include adoption of the concept of chewing efficiency (or chewing effectiveness) during eating, variable fractional ruminal absorption rates of VFA and variable fractional ruminal degradation rates of NDF as a function of rumen liquid pH, as well as a variable rumen volume which directly affects rumen concentrations of metabolites. The model continuously (i.e., by minute) predicts release of soluble components from the feeds in the rumen, concentration and absorption of fermentation end products in the rumen, rumen pools of nutrients and microbial biomass dynamics, as well as passage of microbial biomass and non-fermented nutrients from the rumen, in response to various feeding strategies. Model evaluation covered a wide range of feeding strategies that included pasture and housed feeding systems. Overall, the mean square prediction error (MSPE) as a percentage of the observed mean was relatively low (<10%) with a high amount of the total variation explained by random variation (>65%). Deviation from unity varied between 23% (rumen dry matter content) and 25% (NDF), indicating some consistent over and/or under prediction. A more detailed evaluation was done based on studies available that reported diurnal behaviour of key model outputs such as rumen pools, rumen pH, and rumen VFA. The predictions broadly simulated the observed values quantitatively, relative to general diurnal patterns, and relative to differences between treatments in the predicted diurnal patterns. Results show that the model provides a tool to assess potential outcomes of changing feeding strategies which may be particularly valuable in assessing selection of feeds, amounts and times of the day to offer the feeds. The continuous nature of the simulated output also allows determination of the time(s) of the day that ruminal (and/or post-ruminal) delivery of nutrients may limit ruminal output of nutrients (and/or availability of nutrients) to support milk nutrient synthesis.  相似文献

Tomato (Lycopersicon esculentum) plants emit a blend of volatile organic compounds, which mainly consists of terpenes. Upon herbivory or wounding, the emission of several terpenes increases. We have identified and characterized the first two tomato monoterpene synthases, LeMTS1 and LeMTS2. Although these proteins were highly homologous, recombinant LeMTS1 protein produced (R)-linalool from geranyl diphosphate (GPP) and (E)-nerolidol from farnesyl diphosphate (FPP), while recombinant LeMTS2 produced β-phellandrene, β-myrcene, and sabinene from GPP. In addition, these genes were expressed in different tissues: LeMTS1 was expressed in flowers, young leaves, stems, and petioles, while LeMTS2 was strongest expressed in stems and roots. LeMTS1 expression in leaves was induced by spider mite-infestation, wounding and jasmonic acid (JA)-treatment, while LeMTS2 did not respond to these stimuli. The expression of LeMTS1 in stems and petioles was predominantly detected in trichomes and could be induced by JA. Because JA treatment strongly induced emission of linalool and overexpression of LeMTS1 in tomato resulted in increased production of linalool, we propose that LeMTS1 is a genuine linalool synthase. Our results underline the importance of trichomes in JA-induced terpene emission in tomato.  相似文献

Eight multiparous periparturient Holstein cows fitted with ruminal cannulas were used in a split plot design to evaluate effects of monensin on ruminal volatile fatty acid (VFA) metabolism. Diets were supplemented with 300 mg/day of monensin, or no monensin, both prepartum and postpartum. Isotopic tracers, Na-1-13C-acetate (Ac), Na-1-13C-propionate (Pr), or Na-1-13C-butyrate (Bu) were used as markers to describe VFA kinetics in the rumen. The Windows version of SAAM software (WinSAAM) was used to develop a steady state VFA model. A 9-compartment model was adequate to comprehensively describe ruminal VFA metabolism. The main VFA compartments consisted of Ac, Pr and Bu. The model estimated lower Bu and Ac interconversions with monensin, postpartum (Bu to Ac; 0.14 versus 0.12; P=0.04, and Ac to Bu; 0.32 versus 0.25; P=0.11) compared to when measured prepartum. Results demonstrate that dilution studies employing stable isotopes of VFA can be used to provide information on VFA metabolism of the periparturient dairy cow. A time frame of 320 min of labeled VFA infusion employing a single injection allows accurate quantification of VFA metabolism in the rumen. Compartmental kinetic analysis of major VFA in the rumen indicate that monensin reduced about 0.125 the portion of the Ac that contributes to Bu by reducing movements of Bu originated carbons to the Ac pool. Monensin may affect certain biochemical pathways of interconversion of Bu and Ac in the rumen. Propionate kinetic data suggests that Pr behaves as a single pool in the rumen. Monensin did not affect Pr production in the rumen suggesting that monensin improves the metabolic status of the transition cow in a way other than increasing Pr production in the rumen.  相似文献

A study was conducted to evaluate soybean hulls (SH) as a replacement of tifton bermudagrass hay (TH) in diets of goats containing high levels of spineless cactus. Ten mature bucks (five ruminally fistulated) were used in a 5 × 5 Latin square experiment with 21-day periods. Soybean hulls replaced 0, 250, 500, 750 and 1000 g/kg of TH in the experimental diets. All diets contained 600 g/kg spineless cactus (dry matter basis). Intakes of dry matter (DM), organic matter (OM) and non-fiber carbohydrates and chewing activities decreased linearly (P < 0.01) as the level SH in the diet increased. In vivo digestibility of DM, OM and neutral detergent fiber increased linearly (P < 0.01) as the level of SH in the diet increased. Addition of SH linearly increased (P < 0.05) N retention without affecting microbial N supply (g/kg of digestible OM intake) to the small intestine. Ruminal pH and NH3-N decreased linearly (P < 0.05) while total volatile fatty acid (VFA) concentration increased linearly (P < 0.05) as proportion of SH in diets increased. It was concluded that inclusion of SH in cactus-based diets had a negative impact on intake and chewing activities but improved total tract nutrient utilization by goats. Replacing TH with SH resulted in a concentrate type ruminal fermentation (i.e., low ruminal pH and high VFA concentration).  相似文献

The objectives of this study were to investigate the effects of rare earth elements (REEs) on in vitro rumen fermentation, gas production, microbial protein synthesis and nutrient digestion using in vitro batch culture and continuous culture technique. A mixture of REE containing (g/kg) 380 g of LaCI3·6H2O, 521 g of CeCI3·6H2O, 30 g of PrCI3·6H2O and 69 g chlorides of other light REEs. The experimental diet consisted of 885 g/kg barley grain, 84 g/kg barley silage and 31 g/kg supplement (dry matter (DM) basis). Diet supplemented with different dosages of REE (control, no additional REE; low, 400 mg/kg REE; and high, 800 mg/kg REE, DM basis) were incubated for 4, 8, 14 and 24 h in diluted rumen fluid. At the end of 24 h of incubation, gas production and concentration of volatile fatty acid (VFA) linearly increased with increasing REE supplementation; whereas, influence of REE supplementation on VFA profile was marginal. Dry matter disappearance was not affected (P>0.10). Six dual-flow continuous culture fermenters were used in a replicated 3 × 3 Latin square with same treatments and same diet used in the batch culture. Mean ruminal pH (5.71) and total VFA (93.6 mM) concentration were not affected by supplementation of REE. The molar proportion (mol/100 mol) of acetate (39.1) and propionate (50.5) was similar among the treatments. However, the proportion (mol/100 mol) of butyrate was higher with the high REE (6.6) than with low REE (5.3) or the control (5.8). Ruminal true digestibilities of organic matter (OM) (0.785, 0.811 and 0.828), acid detergent fibre (0.360, 0.431 and 0.432) and crude protein (0.496, 0.590 and 0.589) for control, low and high REE, respectively, linearly increased with increasing REE supplementation, whereas, the increase in ruminal digestibility from low to high dosage of REE was minimal. Microbial nitrogen (N) production (g/day) and microbial efficiency (g N/kg of truly fermented OM) were not affected by treatments. Improvement of ruminal digestibility of OM due to REE supplementation was attributed to the increase in digestibility of fibre and degradability of protein. The results suggest that REE supplementation improved ruminal fibrolytic and proteolytic activities.  相似文献

To test the hypothesis that the beneficial anthelmintic effect of consuming moderate amounts of tannins may not always be accompanied by anti-nutritional effects in goats, two experiments were conducted. In the first, 48 Cashmere goats were randomly assigned to two treatments: supplementation with tannin-containing heather (6.4% total tannins) and non-supplementation. All goats grazed continuously from May to September under farm conditions in a mountainous area of northern Spain. The mean percentage of heather incorporated into the diet of the supplemented animals was 29.1%. Supplementation reduced the mean number of nematode eggs in faeces (P < 0.001) and the goat mortality rate (P < 0.05). The rumen ammonia concentration was markedly reduced in the goats receiving the heather supplement (160 v. 209 mg/l; P < 0.01), while volatile fatty acid (VFA) concentrations were significantly greater (63.0 v. 53.6 mmol total VFA/l; P < 0.05). The heather-supplemented goats also showed a lower loss of live weight (P < 0.01) and body condition score (P < 0.001). In the second experiment, batch cultures of rumen microorganisms with rumen fluid from nine goats whose diet included 29% heather - or not, were used to incubate three substrates (pasture, pasture + heather and pasture + heather + polyethylene glycol) to investigate in vitro ruminal fermentation. Differences (P < 0.01) among substrates were observed in terms of dry matter disappearance (DMD), in vitro true substrate digestibility (ivTSD), gas production and ammonia concentration, the greatest values always associated with the pasture substrate. Cultures involving rumen inoculum derived from goats receiving the heather-containing diet showed slightly lower DMD (46.9 v. 48.5 g/100 g; P < 0.05), ivTSD (64.6 v. 65.9 g/100 g; P < 0.10) and gas production (105 v. 118 ml/g; P < 0.001) values, but much greater total VFA concentrations (48.5 v. 39.3 mmol/l; P < 0.05), and suggest that the efficiency of ruminal fermentation in these animals was probably improved. Together, the results support the absence of a clear nutritional cost counteracting the beneficial anthelmintic effect of supplementing the diet of grazing goats with tannin-containing heather.  相似文献

Effects of the presence or absence of ciliate protozoa on methanogenesis in the rumen and hindgut were investigated in young calves during a 7-week period. Ten Holstein calves, aged 7 days, were divided in two groups (n = 5) and fed an increasing amount of a commercial milk replacer and small amounts of a calves starter. One group was inoculated with ciliate fauna on two occasions, week 5 and 6, while the second remained ciliate-free. The absence of protozoa in the rumen decreased rumen empty weight ( ? 23%, P < 0.01), and rumen pool size of N ( ? 36%, P < 0.01) and crude fat ( ? 37%, P < 0.05). Rumen bacteria of non-faunated calves contained a higher proportion of total amino acid-N per 16 g N ( + 3%, P < 0.01) and D-alanine-N per 16 g N ( + 13%, P < 0.05) compared to faunated calves. Further results contain a reference for a higher bacterial mass in the ciliate-free rumen with an increased number of bacteria adherent to rumen mucosa. The CH4 production in the rumen increased exponentially with the increase in protozoa population size (R2 = 0.68). In presence of 46 · 104 protozoa per ml rumen fluid, the in vitro CH4 production of rumen fluid per mol total VFA was about 34% higher in faunated than in non-faunated calves (P < 0.001). Hydrogen (2H) recovery of rumen fermentation was positively correlated (R2 = 0.55) to the CH4 production rate. Methanogens were attached on rumen mucosa. Methanogenesis, induced by rumen mucosa attached bacteria, was stimulated by ruminal protozoa. In the absence of protozoa in the rumen, the acetate - propionate ratio and butyrate proportion of VFA were reduced. In vivo in the absence of protozoa not only the whole animal CH4 production ( ? 30%, P < 0.05) but also the digestibility of carbohydrates ( ? 4%, P < 0.05) was reduced. Thereby no difference was observed in the intake of ME per kg DM between the groups. In conclusion, the methanogenesis in the rumen, but not in hindgut, is associated with the development of the ruminal protozoa population. The level of methanogenesis (mol/mol VFA) in the hindgut amounts to 20% of the ruminal methanogenesis.  相似文献

A continuous culture system, inoculated with rumen liquor from goats or sheep, was used to study fermentation characteristics of olive leaves (OL). The effects of adding polyethylene glycol (PEG 4000 MW; 0, 2 or 20 g/100 g OL) and/or supplementing with urea (U) or sunflower meal (SM) (1.0 g N/100 g OM) were also studied. Olive leaf fermentation promoted low VFA production (35.2 mmol/d), predominantly of acetic acid, and low efficiency of VFA production (4.91 mol/kg digestible carbohydrates, DCHO). Both values increased with N supplementation, but effects of PEG were variable. No differences ascribed to the rumen inoculum origin were observed. The ammonia N concentration was increased only by supplementation with U. Total and amino acid N output was low and increased with N addition, but it was not affected by PEG treatment. No differences ascribed to the inoculum origin were observed concerning microbial N production rate or efficiency (g N/kg DCHO). There was no clear difference between sources of supplementary N regarding bacterial protein synthesis. On the basis of PEG results, the effect of tannins on OL fermentation was not important.  相似文献

The objective of this study was to evaluate effects of isobutyrate supplementation on rumen fermentation, lactation performance and plasma characteristics of dairy cows. Twenty multiparous second filial generation (F2) cows of a cross between Chinese Jinnan Yellow and Holstein cows at 148 ± 4.5 days in milk and 22.3 ± 0.81 kg milk production were used in a replicated 4 × 4 Latin square experiment. The treatments were: control (without isobutyrate), low (LIB), medium (MIB) and high (HIB) isobutyrate supplementation of 20, 40 and 60 g per cow per day, respectively. Experimental periods were 30 days with 15 d of adaptation and 15 d of data collection. Dry matter (DM) intake was not affected by increasing isobutyrate supplementation, but milk yields were highest for the 40 g/d isobutyrate supplementation level, where proportion of milk fat, true protein and lactose were minimized. Ruminal pH (6.38–6.24) and ammonia N (13.8–11.1 mg/100 ml) were linearly (P<0.01) decreased, whereas total VFA concentration (124–131 mM) increased at a decreasing rate with increasing isobutyrate supplementation. The ratio of acetate to propionate increased linearly (P<0.01) from 2.77 to 4.43 as isobutyrate supplementation increased due to the increase in acetate production and decrease in propionate production. Digestibilities of OM in the total tract increased linearly (P<0.01) as isobutyrate supplementation increased, digestibilities of DM and EE were highest for the 40 g/d isobutyrate supplementation level, digestibilities of CP, aNDF and ADF increased at a decreasing rate with increasing isobutyrate supplementation. Plasma concentrations of glucose and growth hormone linearly (P<0.03) increased, whereas concentrations of non-esterified fatty acids linearly (P<0.01) decreased. Results indicate that supplementation of this diet with isobutyrate changed the rumen fermentation pattern towards acetate production, improved digestion and modified plasma concentrations of glucose and growth hormone. This suggests that isobutyrate stimulated digestive microorganisms or enzymes in a dose-dependent manner with the optimum isobutyrate dose at about 40 g per cow per day in terms of improved digestion.  相似文献

The objective of this study was to evaluate the effects of LaCl3 supplementation on rumen fermentation, urinary excretion of purine derivatives and feed digestibility in the total tract of steers. Eight ruminally cannulated Simmental steers (420 ± 20 kg) were used in a replicated 4 × 4 Latin square experiment. The treatments were control (without LaCl3); La-low; La-medium and La-high with 450, 900 and 1800 mg LaCl3 per steer per day, respectively. Diet consisted of 600 g/kg corn stover and 400 g/kg concentrate (dry matter [DM] basis). Dry matter intake (averaged 9 kg/day) was restricted to a maximum of 90% of ad libitum intake. Ruminal pH (range of 6.59–6.42) was quadratically (P<0.04) changed, whereas total volatile fatty acids (VFA) concentration (range of 74.16–88.61 mM) was linearly (P<0.01) and quadratically (P<0.01) increased with increasing La supplementation. Ratio of acetate to propionate decreased linearly (P<0.01) from 3.28 to 1.79 as La supplementation increased due to the increased in propionate production. In situ ruminal neutral detergent fibre (aNDF) degradation of corn stover was improved but the crude protein (CP) degradability of soybean meal was decreased with increasing La supplementation. Urinary excretion of purine derivatives was quadratically (P<0.01) changed with altering La supplementation (75.5, 81.0, 82.4 and 70.6 mmol/day for control, low-, medium- and high-LaCl3 supplementation, respectively). Similarly, digestibilities of organic matter, aNDF and CP in the total tract were also linearly and quadratically increased with increasing La supplementation. The present results indicate that supplementation of diet with LaCl3 improved rumen fermentation and feed digestion in beef cattle. It was suggested that the La stimulated the digestive microorganisms or enzymes in a dose-dependent manner. In the experimental conditions of this trial, the optimum La dose was about 900 mg LaCl3 per steer per day.  相似文献

Effects of fatty acids of linseed in different forms, on ruminal fermentation and digestibility were studied in dry cows fitted with ruminal and duodenal cannulas. Four diets based on maize silage, lucerne hay and concentrates (65/10/25 dry matter (DM)) were compared in a 4 × 4 Latin square design experiment where the diets were: control diet (C), diet RL supplied 75 g/kg DM rolled linseeds, diet EL supplied 75 g/kg DM extruded linseeds, and diet LO supplied 26 g/kg DM linseed oil and 49 g/kg DM linseed meal. The diets did not differ in total organic matter (OM) and fibre digestibility, in forestomach and intestinal OM digestibility, and in duodenal N flow. Microbial N duodenal flow tended to be lower for RL versus C diet (P<0.1). Extrusion did not reduce ruminal crude protein (CP) degradation in vivo and in situ. Volatile fatty acid concentration and pattern, and protozoa concentration in the rumen, did not vary among diets. Results confirm the absence of a negative effect of a moderate supply of linseed on rumen function, as well as no effect of extrusion on its ruminal CP degradability.  相似文献
