[目的]考察蝉拟青霉5704s菌丝体提取液的降血糖作用,探索发挥降血糖作用的主要功能组分。[方法]制备蝉拟青霉5704s菌丝体提取液,分离提取液中峰1、峰2蛋白,采用小鼠试验和体外试验测定菌丝体提取液以及不同活性组分的降血糖作用,SDS-PAGE电泳测定主要降血糖组分分子量。[结果]蝉拟青霉5704s菌丝体提取液具有较好的降血糖作用,血糖下降率为36.85%;体外试验中,提取液峰1蛋白的α-糖苷酶抑制率为71.23%,峰2蛋白的抑制率为63.17%;小鼠试验中,提取液峰1蛋白的血糖下降率为40.23%,峰2蛋白的血糖下降率为25.70%;体外试验和小鼠试验均表明提取液峰1蛋白的降血糖效果优于峰2蛋白,峰1蛋白的分子量为57 781 Da。[结论]确定蝉拟青霉5704s菌丝体提取液中发挥主要降血糖功效的组分是峰1蛋白。  相似文献

目的:探讨茶多糖、茶多酚、茶色素对血糖的降低作用.方法:用链脲佐菌素(STZ)建立糖尿病小鼠模型.提取茶多糖,茶多酚,茶色素,作用于糖尿病小鼠.通过葡萄糖氧化酶法(GOD-POD)测定血糖含量,研究并比较各成分的降血糖作用.结果:茶多酚,茶色素具有明显的降血糖作用,茶多糖的降血糖作用不明显.结论:茶多糖,茶多酚,茶色素均具有降血糖的作用.  相似文献

本文用溴化氰活化右旋糖酐对胰岛素进行共价修饰,对修饰胰岛素若干性质作了研究.修饰胰岛素无论对正常小鼠、还是对糖尿病模型小鼠的降血糖活性均较天然胰岛素明显提高;修饰胰岛素对糖尿病模型小鼠,在用药后0.5至6.5小时期间,平均降血糖幅度为70.3%,而天然胰岛素是59.1%,且作用时间延长.本文还探讨了修饰胰岛素的口服降血糖效果.  相似文献

为研究天然虾青素在四氧嘧啶致糖尿病小鼠体内的降血糖作用,将天然虾青素分为高、中、低3个剂量组给四氧嘧啶致糖尿病小鼠灌胃,以0.9%生理盐水为模型对照,以二甲双胍为阳性药物对照进行实验。结果表明:三个剂量的天然虾青素均能较好的降低四氧嘧啶致糖尿病小鼠的血糖水平,并能减缓糖尿病小鼠消瘦和多饮多食的症状,以高剂量组的效果最佳。进一步考察了高剂量的虾青素对正常小鼠的影响,发现其对正常小鼠血糖无显著影响。虾青素对四氧嘧啶致糖尿病小鼠具有明显的降血糖作用。  相似文献

为了研究葡萄籽原花青素提取物(Grape Seed Proanthocyanidin Extracts,GSPE)对糖尿病小鼠的降血糖作用及其机制,本文中采用腹腔注射四氧嘧啶(ALX)构建糖尿病动物模型.造模成功后,将糖尿病小鼠分为模型对照组、格列本脲处理组、葡萄籽原花青素提取物低、中、高三个剂量(50、100、150 mg/kg)处理组,另设正常对照组.连续灌胃给药21天,每天一次,以糖尿病小鼠的体重、血清丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性胰岛素水平、空腹血糖值和糖耐量为测定指标,研究不同剂量葡萄籽原花青素提取物对糖尿病小鼠的降血糖作用及机制.结果显示原花青素高剂量组能促进体重增长,显著降低糖尿病小鼠的血糖水平和糖耐量(P<0.01);同时降低糖尿病小鼠血清MDA水平,提高其SOD活性.通过本实验研究可以看出葡萄籽原花青素提取物具有较强的降血糖作用,降糖机制可能与其提高胰岛素水平和抗氧化能力有关.  相似文献

本文研究了富硒发酵毛头鬼伞(鸡腿菇)Coprinus comatus菌丝体对四氧嘧啶致糖尿病小鼠抗氧化和降血糖的影响。四氧嘧啶建立糖尿病小鼠模型后,将富硒发酵毛头鬼伞菌丝均匀混入饲料中,由小鼠自由取食进行治疗,3周后观察富硒毛头鬼伞菌丝对糖尿病小鼠血糖、MDA、SOD和GSH-Px的影响。结果发现,采用富硒发酵毛头鬼伞菌丝进行治疗后,糖尿病小鼠血糖明显降低;血清和组织中MDA含量显著下降;血清和组织中的SOD和GSH-Px的活力明显的增加。由此推断,富硒发酵毛头鬼伞菌丝对四氧嘧啶致糖尿病小鼠的降血糖效果可能是通过提高机体抗氧化能力来实现的。  相似文献

为了研究血竭超临界提取物的降血糖作用及其机制,用正常小鼠和四氧嘧啶糖尿病小鼠模型,以血糖、糖耐量为指标研究血竭超临界提取物的降糖作用。通过测定体外α-葡萄糖苷酶的活力,观察血竭超临界提取物对α-葡萄糖苷酶的抑制作用。结果发现血竭超临界提取物对正常小鼠空腹血糖无明显降低作用,能改善小鼠对蔗糖的耐受力,能降低四氧嘧啶诱导的糖尿病小鼠的空腹血糖水平,能抑制α-葡萄糖苷酶的活性。表明血竭超临界提取物对糖尿病小鼠有较好的降糖作用,对α-葡萄糖苷酶的抑制作用可能是其降血糖的机制之一。  相似文献

The purpose of this study was to determine the underlying mechanism of the hypoglycemic activity of the aqueous extract perfusion of Spergularia purpurea (SP) in diabetic mice and streptozotocin-induced diabetic rats. The aqueous extract was administered intravenously and the blood glucose levels were determined within 4 hours after starting the treatment. Plasma insulin concentrations and endogenous glucose production were also determined. The aqueous extract at a dose of 10 mg/kg produced a significant decrease in blood glucose levels in normal rats (P < 0.05), and even more in diabetic rats (P < 0.001). This hypoglycemic effect might be due to an extra-pancreatic action of the aqueous extract of SP, since the basal plasma insulin concentrations were unchanged after SP treatment. In diabetic mice, a similar effect was observed and the results showed that aqueous extract of SP caused a potent inhibitor effect on basal endogenous glucose production (p < 0.001). We conclude that aqueous extract perfusion of SP inhibits endogenous glucose production in mice. This inhibition is at least one mechanism explaining the observed hypoglycemic activity of this plant in diabetic animals.  相似文献

本文通过对褐蘑菇提取物的降血糖作用研究,旨在开发降血糖药物新资源.试验数据显示褐蘑菇提取物能显著降低四氧嘧啶所制备的糖尿病小鼠血糖浓度.褐蘑菇提取物高、中、低剂量组对糖尿病小鼠的降糖率分别是16.9%,20.2%和15.1%.阳性对照组的降糖率是25.1%.褐蘑菇提取物中剂量组降糖效果要优于高、低剂量组.  相似文献

The hypoglycemic effects of Eriobotrya japonica seeds were investigated in type 2 diabetic Otsuka Long-Evans Tokushima fatty (OLETF) rats and KK-A(y) mice. The rats and mice were fed on a diet containing 10% powdered Eriobotrya japonica seeds with the coat intact for 4 months. Although the blood glucose concentration in the OLETF rats fed on the control diet without Eriobotrya japonica seeds was increased with time, the concentration in the OLETF rats fed on the diet with Eriobotrya japonica seeds was consistently low throughout the experimental period and was comparable to the level in Long-Evans Tokushima Otsuka (LETO) rats which are normal non-diabetic rats. Serum insulin was significantly lower in the OLETF rats fed on the Eriobotrya japonica seed diet than in those fed on the control diet at the termination of the experimental period. Eriobotrya japonica seeds suppressed the increment of blood glucose for 4 months and also effectively improved the glucose tolerance in the KK-A(y) mice, these actions being mainly exerted by the ethanol extract of the seeds. These results suggest that Eriobotrya japonica seeds had a hypoglycemic property and the effect is attributable to the components extracted by ethanol.  相似文献

Aqueous, ethanolic and hexane fractions of Syzygium alternifolium seeds were prepared and given different doses of these extracts individually to different batches of rats (both normal and alloxan diabetic rats) after an overnight fast. The blood glucose levels were measured at 0, 1, 3, 5 and 7 hours after the treatment. The aqueous extract of Syzygium alternifolium at a dosage of 0.75 g/kg b.w. is showing maximum blood glucose lowering effect in both normal and alloxan diabetic rats. The ethanol and hexane fractions are also showing hypoglycemic and antihyperglycemic activity, but the effect is significantly less than that of aqueous extract. The antihyperglycemic activity of Syzygium alternifolium seed was compared with the treatment of Glibenclamide.  相似文献

The aim of the present study was to determine the in vivo hypoglycemic activity of five organic extracts and enhydrin obtained from yacon leaves. The main constituents of the most active fraction were identified. Five organic extracts and pure crystalline enhydrin were administered to normoglycemic, transiently hyperglycemic and streptozotocin (STZ)-diabetic rats. The fasting and post-prandial blood glucose, and serum insulin levels were estimated and an oral glucose tolerance test (OGTT) was performed for the evaluation of hypoglycemic activity and dose optimization of each extract.We found that the methanol, butanol and chloroform extracts showed effective hypoglycemic activity at minimum doses of 50, 10 and 20 mg/kg body weight, respectively, and were selected for further experiments. Oral administration of a single-dose of each extract produced a slight lowering effect in the fasting blood glucose level of normal healthy rats, whereas each extract tempered significantly the hyperglycemic peak after food ingestion. Daily administration of each extract for 8 weeks produced an effective glycemic control in diabetic animals with an increase in the plasma insulin level. Phytochemical analysis of the most active fraction, the butanol extract, showed that caffeic, chlorogenic and three dicaffeoilquinic acids were significant components. Additionally, enhydrin, the major sesquiterpene lactone of yacon leaves, was also effective to reduce post-prandial glucose and useful in the treatment of diabetic animals (minimum dose: 0.8 mg/kg body weight).The results presented here strongly support the notion that the phenolic compounds above as well as enhydrin are important hypoglycemic principles of yacon leaves that could ameliorate the diabetic state.  相似文献

探讨了湖南安化黑茶的主要品种十两茶水提物的降糖作用及机制.选用6~8周龄db/db自发性糖尿病模型小鼠,每日灌胃给予3个不同剂量的十两茶水提物(100、200、400 mg/kg),连续28 d.每周测定空腹血糖值,实验结束时检测糖耐量和胰岛素水平.研究结果显示,十两茶水提物400 mg/kg给药28 d就能显著降低db/db糖尿病小鼠空腹血糖和改善其对葡萄糖耐受能力.同时,十两茶水提物能显著降低db/db糖尿病小鼠血清胰岛素水平,增加葡萄糖耐量试验胰岛素的释放.结果显示十两茶水提物对2型糖尿病小鼠具有很好的降糖作用,其作用机制与增加胰岛素敏感性有关.  相似文献

Chlorella, a type of unicellular fresh water algae, has been a popular foodstuff in Japan and Taiwan. Chlorella has been shown to produce hypoglycemic effects in alloxan-induced diabetic animals. However, there are no other reports of the effects of this substance in other diabetic animal models. Here we have used streptozocin (STZ)-induced diabetic mice to study the thypoglycemic effects of Chlorella. Diabetes was induced in ICR strain mice by the i.p. injection of STZ. Vehicle-treated ICR mice were used as normal control animals and glibenclamide was used as a positive drug control. The effects of Chlorella on basal blood glucose, exogenous insulin sensitivity test and plasma insulin levels were measured. In normal mice Chlorella produced a transient hypoglycemic effect at 90 min after acute administration; whereas glibenclamide produced a more sustained hypoglycemic effect between 90 min and 180 min after acute administration. Chlorella did not affect the basal blood glucose level in STZ mice. However, Chlorella enhanced and prolonged the hypoglycemic effects of injected insulin in STZ mice for a further 60 min compared to the normal vehicle-treated group. Plasma insulin levels were increased in normal mice after treatment with glibenclamide, whereas Chlorella had no such effect. The current results indicate that Chlorella enhances the hypoglycemic effects of exogenous insulin at a dose which does not produce hypoglycemia in STZ mice, suggesting that insulin sensitivity is increased in these mice.  相似文献

This study researched the effects of chicken meat extract on blood glucose and insulin level, membrane glucose transporter-4 (GLUT4), and tyrosine phosphorylation of insulin receptor substrate-1 (IRS-1) in type 2 diabetic KKAy mice and GK rats. Eight-week-old KKAy mice and GK rats and euglycemic control animals, C57BL/6J mice and Wistar rats, were orally administered a liquid commercial chicken meat extract, BRAND’S Essence of Chicken (BEC), for up to 8 weeks. BEC (1.5 ml/kg) had no effect on blood insulin levels, but significantly lessened the hyperglycemia in the diabetic animals. In the BEC-treated diabetic animals, insulin induced a significant increase in plasma membrane GLUT4 and cytosolic tyrosine-phosphorylated IRS-1, indicating that it attenuates insulin resistance. The present findings are the first demonstration of the hypoglycemic action of a dietary protein, and they lend credence to the reported benefits of using chicken meat as a source of protein in the dietary management of diabetic individuals.  相似文献
