Boron uptake by sunflower, squash and cultured tobacco cells   总被引:5,自引:0,他引:5  
Boron uptake was studied in sunflower ( Helianthus annuus cv. Ha301), squash ( Cucurbita pepo cv. Early prolific straight neck) and cultured tobacco ( Nicotiana tobacum L. cv. TXD Monsanto cell line) cells with the use of stable B isotopes and inductively coupled plasma mass spectrometry. Boron uptake increased linearly with increasing B concentrations in the uptake medium, did not exhibit multiphasic kinetics and was not saturable over a wide concentration range. The addition of respiratory inhibitors to the uptake solution or exposure to low (2°C) or high (42°C) temperatures did not inhibit B uptake. The majority of the B within the plants, including recently absorbed B, was present in a nonexchangeable form and could not be removed by repeated rinsing with deionized water or exchange with B isotope. These results demonstrate that in these species B uptake is a passive, nonmetabolic process and that the formation of nonexchangeable B-complexes within the cytoplasm and cell wall is a key factor in determining the uptake of B by plants.  相似文献

Germ cells in the developing rabbit testis were studied using a modified squash technique. Preleptotene figures present in the postnatal testis were examined and compared with corresponding stages in ovarian germ cells. Severe incidence of a mosaic disease was observed on summer squash (Cucurbita pepo), commonly called pepo, grown in Varanasi during June–September of Khariff season 2007. Symptoms observed were mosaic, puckering on the leaves, wartiness on fruits, general stunting of plants and low yield. PCR amplification with degenerate primers designed to target the conserved sequences of coat protein gene of whitefly transmitted geminiviruses showed ~800 bp fragment in all symptomatic samples tested, indicating the association of a geminivirus with the disease. Nucleotide sequence analysis of the amplified fragment showed 99% identity with pumpkin isolate of squash leaf curl china virus (SLCCNV) from Lucknow. It showed 85–96.7% homology with other isolates of SLCCNV from India and abroad. Phylogenetic analysis revealed the isolate on pepo from Varanasi clustered with SLCCNV isolates on pumpkin from Lucknow and Coimbatore.  相似文献

Aim: To determine the cold tolerance and plant growth promotion potential of Serratia marcescens strain SRM (MTCC 8708). Methods and Results: Serratia marcescens strain SRM was isolated from the flowers of summer squash plants, showing no apparent symptoms of yellow vine disease. It was evaluated for growth and plant growth promotion attributes at 15 and 4°C. At 15°C, the isolate was able to solubilize 76·6 μg ml?1 of P and produce Indole Acetic Acid, IAA (11·1 μg ml?1). HCN and siderophore production were also detected at 15°C. The isolate retained all the plant growth promotion traits at 4°C. Seed bacterization with the isolate significantly enhanced plant biomass and nutrient uptake of wheat seedlings grown in cold temperatures. Conclusion: Serratia marcescens strain SRM is a promising cold‐tolerant isolate that can significantly influence wheat seedling growth at cold temperatures. Significance and Impact of the Study: This strain can be employed as a bioinoculant in cold temperature conditions.  相似文献

Maternal-environmental effects on subsequent progeny life-history traits were evaluated in squash ( Cucurbita pepo L.) in terms of the amount of time available for seed development, and the timing of fruit production. Progeny arising from three kinds of fruit were compared. Plants from which fruits were removed 3 d post-pollination throughout the growing season developed only 'late' fruits (during 10–15 d) at the end of the growing season; on control plants both 'early' and 'late' fruits developed (both types allowed to ripen fully). Seed from each type of fruit was weighed individually and categorized into three size classes, then germinated and raised to maturity, including regular harvesting of all fruits 3 d post-pollination. Maternal effects were evident for both vegetative and reproductive traits and carried over to later stages. In contrast, effects due simply to seed size disappeared by day 30 for leaf variables and day 60 for male flower production. Within a seed-size class, progeny arising from fruit of treated plants produced significantly more leaves, with greater size, and more male flowers than those arising from fruit of control plants, while the reverse was true for fruit number and fruit mass. This result is discussed in terms of possible gibberellic acid involvement. In control plants, progeny arising from seeds in the large, fully mature 'early' fruits produced significantly more, and larger leaves by day 30 than did those from late fruits (suggesting differential provisioning in seeds during development). Male flower production had a highly significant positive correlation with vegetative mass and a significant negative correlation with fruit production.  相似文献

Summary Light- and electron-microscopic observations were made on the stem parenchyma cells of Dalbergia odorifera T. Chen (Papilionaceae), a tropical deciduous tree. In the secondary phloem of branchlet and trunk, all of the parenchyma cells except companion cells contain vacuole proteins. Only the outer secondary xylem of branchlets, but not trunk secondary xylem, has proteins in the ray parenchyma and the vasicentric parenchyma. The xylem vacuole proteins begin to accumulate at the end of the growing period and they disappear after the first flush of growth in spring. The vacuole proteins in phloem cells, particularly in the cells near the cambium, also show seasonal fluctuations. Under the electron microscope, the vacuole proteins appear as fibrous materials in aggregation or in more or less even dispersion, and they occur in the large central vacuoles during both the growth and dormant periods. According to the published studies, the stem storage proteins in the temperate trees appear as small protein-storage vacuoles or protein bodies, and the proteins in the tropical trees occur in large central vacuoles. This distinction is assumed to be related to the differences in the nature of dormancy between temperate and tropical trees.  相似文献

Since Münch in the 1920s proposed that sugar transport in the phloem vascular system is driven by osmotic pressure gradients, his hypothesis has been strongly supported by evidence from herbaceous angiosperms. Experimental constraints made it difficult to test this proposal in large trees, where the distance between source and sink might prove incompatible with the hypothesis. Recently, the theoretical optimization of the Münch mechanism was shown to lead to surprisingly simple predictions for the dimensions of the phloem sieve elements in relation to that of fast growing angiosperms. These results can be obtained in a very transparent way using a simple coupled resistor model. To test the universality of the Münch mechanism, we compiled anatomical data for 32 angiosperm and 38 gymnosperm trees with heights spanning 0.1-50 m. The species studied showed a remarkable correlation with the scaling predictions. The compiled data allowed calculating stem sieve element conductivity and predicting phloem sap flow velocity. The central finding of this work is that all vascular plants seem to have evolved efficient osmotic pumping units, despite their huge disparity in size and morphology. This contribution extends the physical understanding of phloem transport, and will facilitate detailed comparison between theory and field experiments.  相似文献

Phloem parenchyma and companion cells from four species of legumes, Phaseolus vulgaris L., Melilotus alba Desr., Desmodium canadense, L. and Dolichos lablab L., were examined electron microscopically to estabish the presence, structure and development of P-protein. P-protein components consisting of granular, fibrillar, tubular or crystalline structures were found in parenchyma cells of all species and in companion cells of M. alba. The earliest stages of P-protein formation were closely associated with dictyosome cisternae, dictyosome vesicles and/or spiny vesicles. After their formation, the P-protein bodies were frequently transformed into one or more structurally different components. Although these components appeared to be develop-mentally related, their specific associations and transformations differed in each species examined.  相似文献

Summary Changes in the vacuome and cytoplasm of phloem parenchyma cells ofPinus strobus were observed from summer to winter over a three year period. These changes were evidence mainly by aggregation of the vacuolar contents with vital stains in summer but not usually in winter. Gelation of the vacuome could be induced in either season, but in winter the solid material appeared to take up more space in the cell when treated with 2M sucrose than in summer. The same phenomenon of aggregation appeared in fixed material in summer but not in winter in most cases. Changes in the cytoplasm included an apparently greater fluidity in winter, either of whole cytoplasm or a part of it. This was suggested by plasmolysis behavior and starch fallin situ. Use of bromphenol blue stain indicated that there was an increased number of sites for attachment of the stain in winter in the cytoplasm. The seasonal development of phloem in the stems of Siberian larch (Larix sibirica Ldb.) was studied over two seasons on 50–60-year-old trees growing in a natural stand in the Siberian forest-steppe zone. Trees at the age of 20–25 years were used to study metabolites in differentiating and mature phloem elements, cambial zone, and radially growing xylem cells in the periods of early and late wood formation. The development of the current-year phloem in the stems of 50–60-year-old trees started, depending on climatic conditions, in the second-third decades of May, 10–20 days before the xylem formation, and ended together with the shoot growth cessation in late July. Monitoring of the seasonal activity of cambium producing phloem sieve cells and the duration of their differentiation compared to the xylem derivatives in the cambium demonstrated that the top production of phloem and xylem cells could coincide or not coincide during the season, while their differentiation activity was always in antiphase. Sieve cells in the early phloem are separated from those in the late phloem by a layer of tannin-containing cells, which are formed in the period when late xylem formation starts. The starch content in the structural elements of phloem depends on the state of annual xylem layer development. The content of low molecular weight carbohydrates, amino acids, organic acids, and phenols in phloem cells, cambial zone, and xylem derivatives of the cambium depends on the cell type and developmental stage as well as on the type of forming wood (early or late) differing by the cell wall parameters and, hence, by the requirement for assimilates. Significant differences in the dynamics of substances per dry weight and cell were observed during cell development.  相似文献

Abstract. Apical 3-cm root segments excised from 2-d-old squash seedlings ( Cucurbita pepo L. cv. Early Prolific Straightneck) that were germinated and grown between sheets of paper towelling moistened with H2O2 incorporated 144±10 nmol (± SE. n = 15) NaH14CO3 into uridine nucleotides (∑UMP) per gram intact roots during the 3-h incubation period ( Lovatt, Albert & Tremblay, 1979 ). Continued culture of squash seedlings in this manner for an additional 24 or 48 h had no effect or slightly increased (30%) the activity of the orotic acid pathway. However, transfer of 2-d-old seedlings to Shive's nutrient solution reduced the activity of the orotic acid pathway within 15 h to 2.3 nmol NaH14CO3 incorporated into ∑UMP per gram apical 3-cm root segments during the 3-h incubation period. The observed decrease in capacity to synthesize pyrimidine nuclotides de novo paralleled the reduction in glucose content of the roots and was reversed by supplying glucose during hydroponic culture in Shive's nutrient solution. Root elongation was not affected by the reduced activity of the orotic acid pathway nor by the decreased level of available glucose.  相似文献

选用西葫芦(Cucurbita pepo)品种“阿兰”一代为试验材料,研究了外源Ca2+处理对高温强光交叉胁迫下西葫芦幼苗生长特征、光合特性及叶绿素荧光参数的影响.结果表明:高温强光胁迫下,5~20 mmol·L-1 Ca2+处理的西葫芦幼苗具有较高的株高和较大的叶面积,其叶绿素、类胡萝卜素含量及光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)、PSⅡ最大光化学效率(Fv/Fm)、PSⅡ实际光化学效率(ΦPSⅡ)和光化学猝灭系数(qP)均较高,而胞间CO2浓度(Ci)和非光化学猝灭系数(NPQ)较低,其中以10 mmol·L-1Ca2+处理效果最好.说明5~20 mmol·L-1Ca2+处理能有效缓解高温强光对西葫芦光合机构的不可逆伤害,使其保持较快的光合电子传递速率和较高的PSⅡ电子传递活性.Ca2+处理浓度超过40 mmol·L-1时对高温强光胁迫没有缓解效应.  相似文献

Abstract Methods were developed for measuring water content of the free space of suspension-cultured tobacco cells using 3H- or 14C-sorbitol. Sorbitol was not taken up by cells in significant quantities over the 3 min taken to label free space. Free space accounted for 50–60% of the water content of cell pellets irrespective of whether 3H- or 14-C-sorbitol was used. 14C-inulin labelled 13.5% less of the water in cell pellets than 3H-sorbitol, probably due to inadequate penetration by inulin into the free space in the cell wall matrix and within clumps of cells. Measurement of free space is necessary for measuring growth on a fresh or dry weight basis, solute concentrations and parameters of water relations of cells. Techniques for making these measurements on tobacco cells were also developed in this study. Solutes were measured after extraction from cells by expressing sap or by boiling cells in ethanol. Similar solute concentrations were found using both methods of extraction. By expressing sap from cells grown in culture medium with an osmotic pressure of 0.24 MPa, the cells were found to have an internal osmotic pressure of 0.70 MPa. Glucose, fructose, sucrose, amino acids and K+ accounted for 60% of this osmotic pressure. Elastic moduli were estimated using the Boyle-Van't Hoff relationship after suspending cells in solutions with different osmotic pressures and assessing their water content or internal osmotic pressure. For two different lines of tobacco cells, elastic modulus varied between 1 MPa and 5.4 MPa at turgor pressures of 0.15–0.52 MPa (line 1) and between 0.2 MPa and 4.2 MPa at turgor pressures of 0.04–0.26 MPa (line 2).  相似文献

The formation of phloem was studied for two years in stems of 50 to 60 year old trees of Scots pine (Pinus sylvestris L.) growing in nature. The development of phloem of the current year begins 10 to 20 days before the xylem formation and is completed with the termination of shoot growth in the end of June. Observations over the seasonal activity of cambium producing sieve cells of phloem and duration of their differentiation as compared to the xylem derivatives of cambium have shown that the maxima of formation of phloem and xylem cells could coincide or not coincide by season, while the activities of their differentiation were always in antiphase. The sieve cells of early phloem were separated from those of late phloem by a layer of tannin-containing cells, which are formed simultaneously with the formation of late xylem cells by the cambium. Seasonal dynamics of accumulation of starch grain in structural elements of the phloem is related to the xylem development. The content of metabolites in differentiating and mature phloem elements, in the cambium zone, and in the xylem cells growing in the radial direction depended on cell specificity, stage of their development, and type of forming wood, early or late, which differ in the cell wall parameters and, hence, requirement of assimilates. Significant differences were described between the content of low molecular weigh carbohydrates, amino acids, organic acids, and phenol compounds using two methods of calculation: per dry weight and per cell.  相似文献

Transfer cells are specialised transport cells containing invaginated wall ingrowths that generate an amplified plasma membrane surface area with high densities of transporter proteins. They trans‐differentiate from differentiated cells at sites at which enhanced rates of nutrient transport occur across apo/symplasmic boundaries. Despite their physiological importance, little is known of the molecular mechanisms regulating construction of their intricate wall ingrowths. We investigated the genetic control of wall ingrowth formation in phloem parenchyma transfer cells of leaf minor veins in Arabidopsis thaliana. Wall ingrowth development in these cells is substantially enhanced upon exposing plants to high‐light or cold treatments. A hierarchical bioinformatic analysis of public microarray datasets derived from the leaves of plants subjected to these treatments identified GIGANTEA (GI) as one of 46 genes that are commonly up‐regulated twofold or more under both high‐light and cold conditions. Histological analysis of the GI mutants gi‐2 and gi‐3 showed that the amount of phloem parenchyma containing wall ingrowths was reduced 15‐fold compared with wild‐type. Discrete papillate wall ingrowths were formed in gi‐2 plants but failed to develop into branched networks. Wall ingrowth development in gi‐2 was not rescued by exposing these plants to high‐light or cold conditions. In contrast, over‐expression of GI in the gi‐2 background restored wall ingrowth deposition to wild‐type levels. These results indicate that GI regulates the ongoing development of wall ingrowth networks at a point downstream of inputs from environmental signals.  相似文献

Forisomes are protein aggregates found uniquely in the sieve elements of Fabaceaen plants. Upon wounding they undergo a reversible, calcium-dependent conformational switch which enables them to act as cellular stopcocks. Forisomes begin to form in young sieve elements at an early stage of metaphloem differentiation. Genes encoding forisome components could therefore be useful as markers of early sieve element development. Here we present a comprehensive analysis of the developmental expression profile of for1, which encodes such a forisome component. The for1 gene is highly conserved among Fabaceaen species and appears to be unique to this phylogenetic lineage since no orthologous genes have been found in other plants, including Arabidopsis and rice. Even so, transgenic tobacco plants expressing reporter genes under the control of the for1 promoter display reporter activity exclusively in immature sieve elements. This suggests that the regulation of sieve element development is highly conserved even in plants where mature forisomes have not been detected. The promoter system could therefore provide a powerful tool for the detailed analysis of differentiation in metaphloem sieve elements in an unexpectedly broad range of plant species. Collection of cucurbit exudates from cut petioles has been a powerful tool for gaining knowledge on phloem sap composition without full notion of the complex exudation mechanism. Only few publications explicitly mentioned that exudates were collected from the basal side of the cut, which exudes more copiously than the apical side. This is surprising since only exudation from the apical side is supposedly driven by phloem pressure gradients. Composition of carbohydrates and pH values at both wounding sides are equal, whereas protein concentration is higher at the basal side. Apparently, exudation is far more complex than just the delivery of phloem sap. Xylem involvement is indicated by lower protein concentrations after elimination of root pressure. Moreover, dye was sucked into xylem vessels owing to relaxation of negative pressure after cutting. The lateral water efflux from the vessels increases turgor of surrounding cells including sieve elements. Simultaneously, detached parietal proteins (PP1/PP2) induce occlusion of sieve plates and cover wound surface. If root pressure is strong enough, pure xylem sap can be collected after removal of the occlusion plug at the wound surface. The present findings provide a mechanism of sap exudation in Cucurbita maxima, in which the contribution of xylem water is integrated.  相似文献

以西葫芦"晶莹一号"为试材,采用盆栽和人工遮光的方法研究土壤水分和光照强度对西葫芦生长发育和生理特性的影响.结果表明:遮光30%条件下各处理的植株生长较好,其中遮光30%和土壤相对含水量为70%~80%的处理植株生长最好,产量最高.遮光70%条件下,各处理的植株生长受到严重抑制,只开花,不结果,没有经济产量形成.不同处理西葫芦的耗水趋势一致,日耗水量和总耗水量都随遮光程度的增加和土壤含水量的降低而减少.遮光30%和土壤相对含水量为70%~80%的处理水分利用效率和水分产出率最高,分别为2.36和1.57 kg.mm-1·hm-2.西葫芦叶片的净光合速率、蒸腾速率、气孔导度及叶片叶绿素含量随着遮光程度的增加而减小,胞间CO2浓度随着遮光程度的增加而增加.叶片保护酶活性和脯氨酸含量随着遮光程度的增加而降低,丙二醛含量为遮光70%>自然光照>遮光30%.3种光照条件下,随着土壤含水量的增加,西葫芦叶片的光合特性、保护酶活性及脯氨酸和丙二醛含量的变化不同.  相似文献

We performed electron-microscopic examination of structural diurnal changes in the lumen of sieve tubes and the vacuolar system of corresponding companion cells and changes induced by the experimental blockage of assimilate export from the leaf by its cold-girdling. For these investigations, Cucurbita pepo L. and Helianthus annuus L. plants were used, that is, plant species from groups of symplastic and apoplastic plants, which differ in the type of companion cells and a mode of phloem terminal loading. The examinations showed the complete identity of changes in the electron texture of the sieve-tube lumens and companion-cell vacuoles in both plant species in the course of a day, when the level of assimilates changed, or after export blockage. Similar changes in the structure of the vacuolar labyrinths were stated in the companion cells under normal conditions and after cold-girdling, as related to the rate of sieve-tube loading with the vacuolar exudate. Vacuolar expansion and starch accumulation developing in response to changes in the assimilate level in the evening and after cold blockage of the assimilate export occurred in different types of cells, as dependent on their position in the symplast domains. However, the rate of the process similarly depended on the balance between assimilate synthesis and export. Synchronous changes in the texture of the sieve-tube lumen and companion-cell vacuoles were observed within each complex, but asynchronous changes occurred in different complexes. We suggested this phenomenon for recognizing the particular complexes, when they are grouped in a bundle. We observed no signs of cytoplasm or protein synthetic machinery in the sieve tubes. We concluded that the sieve-tube lumen and vacuoles of companion cells are common in nature. Similar electron texture of the images of the companion-cell vacuolar labyrinth and tube lumens, their connection through the lateral sieve fields, morphological modifications of the companion-cell vacuolar system as dependent on the activity of sieve tube loading—all of these facts imply the continuity of these transport compartments and fluxes in them and the similarity in the composition of the exudates from companion-cell vacuoles and phloem tubes.  相似文献
