The possibility of free radical reactions occurring in biological processes led to the development and employment of novel methods and techniques focused on determining their existence and importance in normal and pathological conditions. For this reason the use of nitrones for spin trapping free radicals became widespread in the 1970s and 1980s, when surprisingly the first evidence of their potent biological properties was noted. Since then widespread exploration and demonstration of the potent biological properties of phenyl-tert-butylnitrone (PBN) and its derivatives took place in preclinical models of septic shock and then in experimental stroke. The most extensive commercial effort made to capitalize on the potent properties of the PBN-nitrones was for acute ischemic stroke. This occurred during 1993–2006, when the 2,4-disulfonylphenyl PBN derivative, called NXY-059 in the stroke studies, was shown to be safe in humans and was taken all the way through clinical phase 3 trials and then was deemed to be ineffective. As summarized in this review, because of its excellent human safety profile, 2,4-disulfonylphenyl PBN, now called OKN-007 in the cancer studies, was tested as an anti-cancer agent in several preclinical glioma models and shown to be very effective. Based on these studies this compound is now scheduled to enter into early clinical trials for astrocytoma/glioblastoma multiforme this year. The potential use of OKN-007 in combination with neurotropic compounds such as the lanthionine ketamine esters is discussed for glioblastoma multiforme as well as for various other indications leading to dementia, such as aging, septic shock, and malaria infections. There is much more research and development activity ongoing for various indications with the nitrones, alone or in combination with other active compounds, as briefly noted in this review.  相似文献

Twenty-eight patients with pulmonary tuberculosis in a small, tax-supported sanatorium were treated by primary pulmonary resection. In a comparison of results with those obtained in the same sanatorium by thoracoplasty and extrapleural pneumothorax, it was noted that in general the patients who had resection had earlier conversion of sputum to "negative" and had a shorter stay in hospital. Complications were not of sufficient frequency to contraindicate use of resection in cases in which there was doubt that thoracoplasty would be effective. The cost of hospitalization for surgical treatment and postoperative care was considerably less when resection was done than it was for either three-stage or two-stage thoracoplasty.  相似文献

Summary Chronic treatment of rats with reserpine, isoproterenol, or a combination of these two agents has been suggested as a means to produce an experimental animal model for the chronic exocrinopathy cystic fibrosis. The effect of these treatments on glycoconjugate distribution in rat submandibular gland acinar cells was investigated by quantitative lectin cytochemistry. Significant changes in wheat-germ agglutinin (WGA), soy bean agglutinin (SBA) and concanavalin A (Con A) binding sites in the mucus granules were observed, but peanut agglutinin (PNA) binding was not significantly affected. The quantitative changes in glycoconjugates in the acinar cells of the submandibular gland could be a possible explanation for the increased binding of calcium by the intracellular mucus noted in previous studies on these animal models.  相似文献

Nine hundred and thirty-nine patients admitted to the locked receiving ward in the psychiatric service of the U. S. Naval Hospital, Oakland, over a ten-month period, many of them psychotic and in an acute initial episode, were treated with an intensive group therapy program, which more appropriately should be called a therapeutic community. During this time, the ward medical officer did not put any patients in a seclusion room. Patients who did not require a locked ward were quickly transferred to the open receiving ward which was established five months after this program began. It was possible to greatly diminish the quantity of sleeping medicine prescribed and practically to eliminate the use of barbiturates given parenterally. Restraints were never used. To be dealt with in this atmosphere of candor and relative freedom seemed to evoke a responsive attitude in the patients and many of them benefited from it.  相似文献
