Eight proprietary genotypes of glandular-haired alfalfa, Medicago sativa L., supplied by two different companies, were compared for the degree and types of resistance to the potato leafhopper, Ernpoasca fabae (Harris), and hopperburn. A tube cage no-choice bioassay was developed to test leafhopper mortality, feeding, settling preferences, severity of hopperburn symptoms (in this case, defined as both yellowing and stem growth reduction), and trichome density and type on feeding sites. Leafhopper mortality was both strongly and significantly associated with feeding and leaf trichome density; decreased hopperburn symptom severity was weakly, although significantly, associated with increased mortality. To quantify hopperburn in terms of both yellowing and stem growth reduction, we developed a ranking system that reduces overall hopperburn expression to a single number that considers the varying responses to both types of symptoms. Great variability in leafhopper settling, leafhopper mortality, and stem glandular trichome density was detected among alfalfa genotypes, suggesting that genotypic differences may be based on the concentration and/or chemical constituency of the trichome exudates. We postulate that, among variably resistant genotypes of glandular-haired alfalfa, differences among leafhopper responses and hopperburn severity are linked to forced movement from the stems to the leaves as refuge feeding sites. Principal component analysis was performed to reduce the 10 variables down to five biologically significant factors. Scores for these factors were then used to develop resistance indices for potato leafhopper resistance, hopperburn resistance, and an overall glandular-haired alfalfa resistance index.  相似文献

We examined the effects of potato leafhopper (Empoasca fabae) developmental stage and alfalfa (Medicago sativa) developmental stage on the physiological response of the plant to injury. We used radioactive carbon dioxide to label the photoassimilate stream and evaluate the phloem health of alfalfa. In one experiment, six first instar, four fourth instar, and three adult leafhoppers were caged by stage on single alfalfa stems for approximately one day. Only fourth instar nymphs significantly reduced the amount of label transported to injured tissues above the source of the labeled assimilate. First instar nymphs had no effect and adults reduced assimilate transport to stem tips, but this trend was not significant possibly because of confounding variables. However, injury by both first instar nymphs and adults resulted in greater concentration of labeled assimilate in portions of the stem below the feeding site. In another experiment, the developmental stage of alfalfa stems was central to the physiological response of alfalfa to leafhopper injury. A 20 h exposure to three adult leafhoppers significantly reduced the amount of label translocated to the tip and crown tissues of early vegetative plants, and to the crown tissue only of late vegetative plants. In reproductive plants, assimilate translocation was not affected by leafhopper injury. In a final experiment, we found no evidence of an effect on the photosynthesis of leaves of similar age and position to those used as source leaves in our translocation studies. Our findings contribute to our understanding of the physiological response of plants to injury by sap-feeding insects, and suggest the need for greater refinement of economic injury levels based on leafhopper and plant developmental stage.  相似文献

The potato leafhopper, Empoasca fabae (Harris), is a key pest of alfalfa, Medicago sativa L., in part because of the leafhopper's ability to disrupt upward translocation within phloem tissues. To determine if leafhopper injury also disrupts basal translocation necessary for regrowth and perenniality of alfalfa, we used radiolabeled 14CO2 to measure the basal transport of photoassimilates in injured and healthy plants. In one experiment, less 14C was transported to lower stem tissue of leafhopper-injured plants in comparison to the same tissue of healthy plants in early vegetative and early reproductive stages of alfalfa development. In a second experiment, less 14C was transported to lower stem, crown, and root tissues of injured plants in comparison to the same tissues of healthy, early reproductive plants. The disruption of basal transport caused by potato leafhopper may impact carbon storage and mobilization subsequent to defoliation, winter survival, and nitrogen fixation.  相似文献

New cultivars of alfalfa, Medicago sativa L., have been released with glandular trichomes for resistance to potato leafhopper, Empoasca fabae (Harris). Yet, the impact of the glandular trichomes on the primary natural enemy of the leafhopper, Anagrus nigriventris Girault, is unknown. We compared the host searching behavior of the egg parasitoid on four alfalfa clones varying in trichome characters. Female wasps were videotaped on Ranger, a susceptible clone with relatively sparse trichomes, B14, a resistant clone with dense but nonglandular trichomes, and FG12 and FG18, two resistant clones with glandular trichomes. Although the number of leafhopper eggs per stem exposed to wasps did not significantly differ among the four clones, the frequency of foraging and total foraging time were less on the two clones with glandular trichomes than on the two clones with nonglandular trichomes. In addition, an analysis of covariance demonstrated that, although the number of ovipositional probes increased with egg density on a stem, the number of probes on stems with glandular trichomes was significantly less than that on stems without glandular trichomes. The allocation of time by wasps among drumming, probing, and grooming behaviors was similar among the clones. Wasps tended to fly off of clones with glandular trichomes more often than off of clones with nonglandular trichomes. This study suggests that cultivars with glandular trichomes may interfere with host searching by A. nigriventris.  相似文献

Understanding how changes in plant diversity affect agroecosystem functioning remains a key challenge. We examined how intercropping alfalfa, Medicago sativa, with orchardgrass, Dactylis glomerata, affects the potato leafhopper, Empoasca fabae, its host plant (alfalfa), and the efficiency of a leafhopper predator, Nabis americoferus. In a field experiment, intercropping reduced the reproductive efficiency of the leafhopper. Nabis was more effective at reducing leafhopper abundance, and protecting alfalfa from hopperburn, in the polyculture than in the monoculture of alfalfa. In a series of laboratory experiments, we investigated mechanisms by which intercropping could enhance the efficiency of Nabis. Intercropping resulted in changes in vegetation structure and the spatial distribution of leafhoppers, but there was little evidence that these factors influenced the efficiency of Nabis. Instead, orchardgrass, a nonhost for leafhoppers, increased leafhopper movement, and Nabis captured leafhoppers more efficiently when the herbivores were more mobile. These results indicate that intercropping with nonhost plants promotes leafhopper movement and vulnerability to predation, and reveal a novel mechanism by which plant diversity can reduce herbivory.  相似文献

Plant tolerance to herbivory is a key approach for managing pests. In alfalfa, Medicago sativa, the potato leafhopper, Empoasca fabae, is a major pest as a result of the cascade of plant responses to piercing-sucking injury. To identify tolerance to its injury based on alfalfa physiology, experiments were conducted in the field and greenhouse. In our comparison of the response of field-grown alfalfa cultivars to standardized leafhopper densities, net photosynthesis and transpiration rates of 'Geneva' leaves were reduced by 18 and 21%, respectively, by leafhopper presence compared with a rate change of <1% of resistant 'EverGreen' leaves. Under greenhouse conditions, alfalfa clones varied in their level of gas exchange (net photosynthesis and transpiration) and stem elongation responses to leafhopper injury. For example, in the comparison of seven clones, net photosynthesis declined an average of 40.7% with leafhopper injury, although individual clones varied from 26.6 to 74.3% reduction. Internode elongation after 2 d was 60.3% less on injured stems compared with healthy stems, but again, the individual clones varied from 17.3 to 91.9%. In a time-course study of selected clones, clones varied in their level of injury just after and 3 d after insect removal. Gas exchange responses of all clones recovered by 7 d after cessation of injury. In a choice test, leafhoppers spent similar amounts of time on the susceptible clone and the most tolerant clone; however, their precise feeding behaviors were not measured. Thus, the variable response of clones to injury may be either true physiological tolerance or antixenosis from a change in feeding behavior. This study showed putative tolerance to leafhopper injury among alfalfa genotypes, suggesting that tolerance could be the basis for crop protection in alfalfa from potato leafhopper injury.  相似文献

The feeding behaviors and salivary deposits of the potato leafhopper, Empoasca fabae (Harris), on stems of a susceptible cultivar of alfalfa (Medicago sativa, L.) were studied in relation to the cellular damage and initiation of hopperburn they cause. An AC electronic monitoring system (EMS) was used to quantify the number and duration of probes made on alfalfa stems. Light and electron microscopy were then used to examine the effects of these probes. Most probes were short in duration as determined by EMS and comprised primarily of multiple-cell laceration behavior (represented by the Ia waveform). Probes containing still-stylet ingestion behavior (represented by the Ic waveform) were longer in duration and occurred less frequently. There was a close correspondence between the number of probes containing the Ic waveform and number of salivary deposits. Histological examinations of the stems revealed six types of salivary deposits but only one of these resembled the solid, distinct (or true) salivary sheaths characteristic of sheath-feeders. Less than 25% of E. fabae probes left any salivary deposit, and only 1% left true sheaths. Therefore, it seems unlikely that these deposits could physically block the phloem to cause hopperburn. Cellular disruptions, such as abnormally divided and enlarged cells, were found in the vascular cambium and phloem, as well as other tissues. Enlarged vascular cambial cells appeared to crush the phloem cells, and thus probably caused hopperburn. This microscopic damage was initiated by vigorous, active probing, along with the copious watery saliva that is secreted during the behavior represented by Ia. These observations strongly support Medler's (1941) hypothesis that hopperburn is caused by a substance in the insect's saliva that induces plant cells to enlarge and block the phloem.
Résumé Le comportement alimentaire et les dépôts salivaires d'E. fabae Haris ont été examinés en relation avec les dégâts cellulaires sur tiges de cultivars sensibles de Medicago sativa. Un système de contrôle électronique AC (EMS) a été utilisé pour quantifier le nombre et la durée des sondages dans les tiges de luzerne. Les microscopies optique et électronique ont permis d'examiner les effets de ces condages. EMS a montré que la majorité des sondages étaient brefs (1 à 2 min) et commençait par la lacération multiple de cellules (révélé par un onde Ia). Les sondages comprenant un comportement d'ingestion avec immobilisation des stylets (révélé par une onde Ic) sont plus longs et moins fréquents. Il y a une bonne correspondance entre le nombre de sondages contenant un onde Ic et le nombre de dépôts salivaires. L'association des sondages et des dépôts salivaires montre que la durée du sondage était plus liée à la solidité des dépôts salivaires que la durée des ondes individuelles. L'examen histologique des tiges a révélé 6 types de dépôts salivaires, mais seul l'un d'entre eux ressemblait à celui donnant la gaine solide, individualisée (ou véritable) gaine salivaire caractéristique des consommateurs avec gaines salivaires. Moins de 25% des sondages d'E. fabae laissent un dépôt et seulement 1% laisse une véritable gaine. Ainsi, il semble improbable que ces dépôts puissent bloquer le phloème et provoquer la brûlure des ciccadèles. Des dégâts cellulaires, tels que divisions anormales et cellules hypertrophiées, ont été observés dans la cambium vasculaire et dans le phloème, ainsi que dans d'autres tissus. Dans des tissus fortement abîmés, le phloème est complètement désorganisé et contient des cellules ratatinées et affaissées. Des cellules vasculaires cambiales hypertrophiées paraissaient aplatir les cellules du phloème, et ainsi interférer avec la translocation des photosynthétats. Les dégâts microscopiques sont produits par les sondages actifs et vigoureux, associés à la copieuse détrempe salivaire secrétée pendant le comportement représenter par Ia. Ces observations appuient fortement l'hypothèse de Medler (1941) suivant laquelle la brûlure des ciccadèles est provoquée par une substance de la salive de l'insecte qui induit l'hypertrophie cellulaire et bloque le phloème.

应用可视DC-EPG方法研究了假眼小绿叶蝉(Empoasca vitis Gothe)在9个品种茶苗上的口针刺探行为,共发现并初步确定了7种主要波型,即A波、S波、C波、E波、F波和R波、以及非刺探波NP波。A波为刺探波,S波为口针向韧皮部刺探和进入韧皮部中的分泌唾液波,C波为口针到达韧皮部之前的主动取食波,E波和F波为口针在韧皮部中吸收波,R波为取食间歇波。以该叶蝉在不同品种茶树上的平均刺探次数和各波形平均持续时间为指标,或者以其在不同品种茶树上含有各波形的单次刺探平均持续时间为指标,分别进行聚类分析,均将9个品种分为3个不同的组。S、E和F波对应着假眼小绿叶蝉在茶树上的主要取食活动,可能与茶树抗假眼小绿叶蝉的取食密切相关。以S、E和F各波的平均持续时间、以及含有各波形的单次刺探的平均持续时间为指标,对品种的抗性强弱排序,评判9个茶树品种抗叶蝉取食能力由强至弱的顺序为:龙井长叶、黄旦、政和大白茶、黔湄601、红芽佛手、中茶102、中茶302、龙井43和安吉白茶。该顺序与田间查得的9个品种茶树上假眼小绿叶蝉种群密度由小到大的顺序一致,表明DC-EPG方法简捷、可信度高,可作为检测茶树品种对叶蝉抗性的有效手段之一。  相似文献

We have identified three stem abundantly expressed genes in lucerne (alfalfa, Medicago sativa). A cDNA library, constructed from lucerne stem polyadenylated RNA, was screened by differential hybridization. From this screening, cDNA clones that correspond to genes which are preferentially, or specifically, expressed in the stem were isolated. MsaS1 encodes an unidentified protein, MsaS2 encodes an S-adenosyl-homocysteine hydrolase and MsaS3 encodes an extensin-like protein. Northern blot analysis of RNA isolated from individual stem internodes indicated that the three corresponding genes show differing developmental patterns of expression. The expression of MsaS1 was confined to the youngest stem tissue and may be regulated by sucrose. In stem tissue the level of RNA for the three genes decreased in response to wounding. Tissue print hybridization analysis was used to localize the expression of the genes to the xylem side of vascular bundles in lucerne stems.  相似文献

The 9 tested tea cultivars were categorized into 3 groups by the cluster analysis according to the number of probes per insect, waveform durations, or duration per probe of various waveforms on different tea cultivars. Waveforms S, E and F correlated to the main feeding activity of the leafhopper and may provide valuable information on predicting the resis-tance level of the tea plants to the leafhopper. The resistance level of the 9 tea cultivars to the leafhopper was ranked based on the durations of waveforms S, E and F, as well as the duration per probe including various waveforms. The ranking order of the resis-tance was: Longjingchangye > Hangdan > Zhenghedabaicha > Qianmei 601 > Hongyafoshuo > Zhongcha 102 > Zhongcha 302 > Longjing 43 > Anjibaicha, which corresponded to the resistance level determined by the population density (infestation) of the leaf-hopper on the 9 tea cultivars in the tea fields. Our study suggests that this simple and convenient DC-EPG technique might have great potential as a reliable tool to predict the resistance of tea cultivars to the tea leafhopper.  相似文献

By inserting entomological needles into the lower parts of young inflorescence stems of three-month-old Arabidopsis thaliana (L.) Heynh var. Colombia plants, we studied the process of regenerative xylem production. Regenerative xylem was formed only in one- to two-day-old inflorescence stems but not in older ones. The regenerative vessels originated from re-differentiation of cortical parenchyma. To characterize the process of regenerative xylem formation, we conducted a histological study from the time of wounding to day 30 after wounding. In the first day after wounding the tissues showed no structural responses except for the wounding itself. After six days, regenerative vessel members were already differentiating in a basipetal pattern, forming a vascular bypass around the wound. Regenerative vessel member formation reached a maximal level on the twelfth day after wounding. Sixteen days after wounding the pith parenchyma started to become loose as if indicating tissue senescence. Altogether, vascular regeneration following wounding in inflorescence stems of Arabidopsis thaliana is similar to that in other dicotyledon plants. These findings provide the basis for the use of Arabidopsis thaliana as a model system to study the genetics, physiology and cell biology of wound healing and regenerative vascular tissue formation.  相似文献

The fine structure of the salivary sheaths in plant tissues can provide important information on homopteran probing and ingestion behaviors. Salivary sheaths secreted by the brown planthopper (BPH), Nilaparvata lugens (Stål) (Homoptera: Delphacidae), and their tissue pathway were investigated using light, scanning electron, and transmission electron microscopy. About half of the salivary flanges on the surface of the food substrate were connected with internal salivary sheaths. Only 43% of the salivary sheaths showed side branches. Many sculpture‐like protuberances and small cavities had been formed on the outer surface of the salivary sheath, but the sheath lumen circumferences were sealed. Brown planthoppers showed a preference for probing and leaving salivary sheaths in the susceptible rice variety TN1 rather than in the resistant variety B5 during the first 2 days of the experiments. The salivary sheaths in rice tissues reached the inner tissue layer of the leaf sheaths and stems, but were mostly observed to end in the first and second layer of the leaf sheaths. Brown planthoppers also preferred to probe into the thick segment of the outer leaf sheath. After ingestion by the insect, the cytoplasm in both phloem and companion cells degraded and the main organelles were lost. Numerous small vesicles were found in most of the phloem cells, but cell walls remained intact. Large numbers of symbiont‐like structures were observed inside the salivary sheath lumen. These results indicated that BPH has complicated feeding behaviors, which warrants further investigation.  相似文献

Laboratory experiments were conducted to characterize the trichome-based defense of glandular-haired alfalfa, Medicago sativa L., against the potato leafhopper, Empoasca fabae (Harris). Within-plant variability in leafhopper resistance was examined by caging adult leafhoppers to either basal or apical stem internodes of the leafhopper-resistant, glandular-haired M. sativa genotype G98A or the susceptible, nonglandular-haired M. sativa 'Ranger'. Young, actively secreting glandular trichomes are located on apical internodes of G98A, whereas senesced gland heads are found on older, basal internodes of G98A. After 96 h, the highest cumulative leafhopper mortality and lowest number of excretory droplets were associated with apical internodes of G98A. No difference was detected in mortality and feeding levels among insects caged to basal internodes of G98A and basal and apical internodes of Ranger. The influence of abiotic factors on leafhopper resistance was evaluated by caging adult leafhoppers to either G98A or Ranger under four combinations of low and high light (250 and 1,000 micromol s(-1) m(-2)) and temperature regimes (17 and 30 degrees C). After 96 h, the highest cumulative mortality was associated with leafhoppers confined to G98A under high light and high temperature conditions. Temperature level and plant type also had an effect on the production of excretory droplets, resulting in the highest number of excretory droplets being associated with Ranger under the high temperature regime. These results indicate that certain regions of M. sativa G98A are better protected against the potato leafhopper than others and that temperature influences resistance levels of glandular-haired alfalfa.  相似文献

Cultivars of glandular-haired alfalfa, Medicago sativa L., such as '54H69', are currently available and marketed as being resistant to potato leafhopper, Empoasca fabae (Harris). Between 2000 and 2002, studies were conducted to compare the effects of '54H69' and a standard, nonglandular-haired alfalfa cultivar, 'Choice', on alfalfa weevil, Hypera postica (Gyllenhal), and potato leafhopper populations at Campbell and Montgomery counties, Virginia. '54H69' had no effect on alfalfa weevil populations. At each location, densities of alfalfa weevil in '54H69' and 'Choice' were similar, but pest pressure was higher at Campbell Co. than at Montgomery Co. and always exceeded the economic threshold before insecticide was applied. Densities of potato leafhopper also did not differ between '54H69' and 'Choice' in any year at the two locations. Insecticide treatment effectively reduced potato leafhopper densities in the two cultivars, although populations were below the economic threshold at both locations when the insecticides were applied. Overall, postinsecticide treatment comparisons showed that the densities of alfalfa weevil and potato leafhoppers were similar or higher in untreated '54H69' compared with insecticide-treated 'Choice'. In addition, there were no differences in seasonal dry yields between '54H69' and 'Choice' in any year at either location. Our results indicate that the glandular-haired alfalfa '54H69' does not provide acceptable resistance to potato leafhopper and also does not offer a yield advantage to growers in Virginia.  相似文献

Abstract We report that a chemical stimulus from a herbivore, a galling insect, changes plant morphology and physiology to benefit the herbivore. Previous studies could not determine whether insect galls are induced by mechanical or chemical stimuli because feeding and oviposition both occurred at the site of gall formation. We report that the mouthparts of a spruce‐galling insect, Adelges cooleyi, were inserted in stem phloem cells far from induced galls, that tissues between mouthparts and galls appeared normal, and that the ability to initiate galls was inversely correlated with distance from buds (potential gall sites). Thus the effects of chemical stimuli were unambiguously separated from any mechanical influence of probing stylets or ovipositors. Our results strongly suggest that galls were induced by a chemical stimulus transported to buds via vascular tissue and that its efficacy was dose‐dependent.  相似文献

The fine structure of the mouthparts of the whitefly, Bemisia tabaci Gennadius (Homoptera: Aleyrodidae), was examined by scanning and transmission electron microscopy. Adult whitefly mouthparts are similar to those of other homopterans, especially aphids, being composed of the labrum, the labium, and the stylets. The stylet bundle is the feeding organ of the whitefly and is composed of 2 mandibular stylets and 2 maxillary stylets. Mandibular stylets, which are located on the outer aspect of the stylet bundle, each contain 2 dendrites. The tips of the mandibular stylets are curved inward, and there are barb-like ridges on the lateral aspects, which probably function in piercing and cutting plant tissues and in anchoring the stylets in the tissues. The maxillary stylets are not innervated and are interlocked to form 2 separate compartments, the food canal and salivary canal. At the distal end of the interlocked maxillary stylets, there is a small depression, which may allow for mixing of the salivary canal and food canal components. Movement of the B. tabaci stylets during feeding is discussed in comparison with other homopterans.  相似文献

The ultrastructure of the sensilla, and other structures, within the stylets and precibarium of Macrosteles fascifrons were examined by transmission and scanning electron microscopy. Precibarium is a new term, defined here, for the canal that precedes the cibarium inside the leafhopper head. Within the precibarium are found 20 chemosensilla and a previously undescribed structure, the precibarial valve. Twelve mechanosensilla, three in each stylet, are found within the maxillary and mandibular stylets. The relationship between all of these structures and feeding by the insect is detailed in a feeding mechanism hypothesis. It is concluded that leafhoppers (and probably all homopterans) utilize the precibarial chemosensilla alone for gustatory discrimination, the stylet sensilla for proprioception, and the precibarial valve for regulation of fluid uptake and compartmentalization of the sensilla.  相似文献

A time-dependent correlation of anatomical and chemical defence reactions was shown during the incompatible reaction of tomato against the phanerogamic parasite Cuscuta reflexa. Microscopical analysis of the infection sites at the tomato stem revealed the elongation of epidermal, hypodermal and collenchymatic cells beneath the parasitic prehaustorium. After 9–11 days of infection the elongated cells had collapsed forming a visible brownish plaque at the tomato stem followed by a scalariform tissue with lignified and suberized cell walls. Concomitantly, an enhanced accumulation of soluble phenolic compounds (chlorogenic acid and an unidentified hydroxycinnamic acid derivative), as well as a stimulation of peroxidases, was observed. In contrast, PAL activity was not increased. Whereas the stimulation of phenylpropanoid metabolism could also be induced by artificial wounding, the described anatomical changes were only observed during attack of Cuscuta.  相似文献
