This study is motivated by a real problem encountered in the manufacturing and distribution process at a local electronic manufacturer of security devices. We investigate the impact of operations redesign (i.e., operations merging) on the cost of safety stock in a supply chain. A simple safety stock method is used to derive a model for estimating safety stock levels. Our result shows that operations redesign can have a significant impact on safety stock investment. We extend and complement the existing literature in the following aspects: (i) we address the issue of safety stock deployment, i.e., we not only investigate the problem of how many operations should be delayed, but also determine which operations need to be delayed, (ii) we provide an efficient heuristic algorithm to determine which operations need to be merged, and (iii) we find the optimal operations redesign strategies under some special cases. Our analysis also reveals some important conditions and insights for better operations redesign, which enable us not only to decide when an operations redesign is appropriate, but also to suggest the scale and the format of the operations redesign.  相似文献

The purpose of this paper is to develop optimal tool partitioning policies and strip sequencing strategies for a class of flexible manufacturing problems. The problems under consideration involve a large number of operations to be performed by a series of tools on a two-dimensional object. For example, these operations could consist of drilling holes in a metallic sheet. Tools are arranged in a carousel or along a toolbar according to a predetermined sequence. Operations are performed by repeatedly moving the sheet to bring the hole locations under the tool. During each pass, as all operations involving a series of consecutive tools are executed, two main problems are to be solved: (1) how to move the sheet during each pass, (2) how to partition the tools into blocks of consecutive tools. A strip strategy is used to move the sheet. Given this policy, optimal strip widths and tool partitioning policies are determined jointly. Analytical solutions are derived under two metrics corresponding to different operating modes. A numerical example is provided.  相似文献

补料(流加葡萄糖)操作是谷氨酸发酵过程最重要的操作之一,工业上有各种各样的补料操作方法。控制葡萄糖浓度于一个较为平稳且适中的水平有利于提高谷氨酸发酵的性能指标。通过在线计量谷氨酸发酵中的氨水耗量并据此在线推定发酵液中的葡萄糖浓度,构建了一个谷氨酸发酵自动在线补料系统。使用该控制系统,谷氨酸发酵过程的葡萄糖浓度可以控制在任意水平,平稳、无波动的谷氨酸发酵可以得到实现。  相似文献

The pursuit of mechanistic explanations in biology has produced a great deal of knowledge about the parts, operations, and organization of mechanisms taken to be responsible for biological phenomena. Holist critics have often raised important criticisms of proposed mechanistic explanations, but until recently holists have not had alternative research strategies through which to advance explanations. This paper argues both that the results of mechanistic strategies has forced mechanists to confront ways in which whole systems affect their components and that new representational and modeling strategies are providing tools for understanding these effects of whole systems upon components. Drawing from research on the mechanism responsible for circadian rhythms in mammals, I develop two examples in which mechanistic analysis is being integrated into a more holist perspective: research revealing intercellular integration of circadian mechanisms with those involved in cell metabolism and research revealing that stable?rhythms are dependent on how individual cells in the suprachiasmatic nucleus synchronize with each other to generate regular rhythms. Tools such as network diagramming and computational modeling are providing means to integrate mechanistic models into accounts of whole systems.  相似文献

Selective logging is a major driver of rainforest degradation across the tropics. Two competing logging strategies are proposed to meet timber demands with the least impact on biodiversity: land sharing, which combines timber extraction with biodiversity protection across the concession; and land sparing, in which higher intensity logging is combined with the protection of intact primary forest reserves. We evaluate these strategies by comparing the abundances and species richness of birds, dung beetles and ants in Borneo, using a protocol that allows us to control for both timber yield and net profit across strategies. Within each taxonomic group, more species had higher abundances with land‐sparing than land‐sharing logging, and this translated into significantly higher species richness within land‐sparing concessions. Our results are similar when focusing only on species found in primary forest and restricted in range to Sundaland, and they are independent of the scale of sampling. For each taxonomic group, land‐sparing logging was the most promising strategy for maximizing the biological value of logging operations.  相似文献

A time-optimal strategy for start-up of a chemostat has been established by applying Miele's extremization method based on Green's theorem. The start-up time is always minimized by traveling along the uppermost boundary of the set of admissible domains on the (vx, μ) phase plane. The solutions are batch culture for monotone increasing growth kinetics and exponentially fed-batch culture followed by batch culture for substrate-inhibition kinetics. A time-suboptimal start-up strategy, exponentially fed-batch culture operated the same μ as that of chemostate, is proposed as more feasible than and physiologically preferable to the time-optimal strategies. The complete set of equations of operations necessary for performing each of the optimal and suboptimal start-ups is formulated. Numerical comparisons on the basis of continuous operation giving maximum productivities of cell-mass outputs show that differences in times required for start-up between optimal and suboptimal strategies are small.  相似文献

Abstract When considering the current state of the biorefinery industry, it is readily apparent that industrial cellulose and hemicellulose digestion processes are relatively advanced, whereas enzymatic pre-treatment strategies for biomass delignification and cellulose solubilization are not well developed. The need for efficient biomass pre-treatment strategies presents a significant opportunity for researchers studying lignocellulose digestion in termites and other insects. With an emphasis on industrial biomass pre-treatment, this review provides an overview of: (i) industrial biorefining operations (feedstocks, processing, and economics); (ii) recent findings from termite research that have revealed candidate enzymes; and (iii) research needs and opportunities for consideration by entomologists working in this area. With respect to research findings, recently identified candidate lignases (laccases, catalases, peroxidases, esterases), other potentially important detoxification enzymes (cytochrome P450, superoxide dismutase), and phenolic acid esterases (carboxylesterases) that may assist in hemicellulose solubilization are overviewed. Regarding research needs and opportunities, several approaches for identification of candidate pre-treatment enzymes from upstream, symbiont-free gut regions are also described.  相似文献

Occult coronary artery disease often accompanies symptomatic peripheral vascular disease and has an important effect on survival. Most perioperative and late fatalities after peripheral vascular operations are due to cardiac causes. Noninvasive cardiac testing can identify patients at increased risk for postoperative cardiac complications, although controversy exists regarding the optimal preoperative evaluation. Risk reduction strategies for patients known to be at high risk are also controversial. Some authors advocate coronary revascularization with coronary artery bypass grafting or percutaneous transluminal coronary angioplasty before the vascular procedure. Others believe that the combined morbidity and mortality of 2 operations exceed those of a peripheral vascular operation performed with aggressive monitoring and medical therapy. Continuous electrocardiographic monitoring after an operation has identified silent myocardial ischemia as a powerful predictor of cardiac complications. Ongoing research is likely to provide insights into the pathogenesis of postoperative cardiac complications and may lead to specific therapeutic interventions. Few prospective studies have been done in this area, and the threshold for preoperative and postoperative intervention is unknown. I review the literature and present an algorithm to guide cardiac testing and risk reduction in patients undergoing elective vascular surgical procedures.  相似文献

Aim:  To determine the prevalence of Listeria monocytogenes and associated risk factors among beef operations (cow-calf and feedlot) in central and southern California.
Methods and Results:  A repeated cross-sectional study where faecal and environmental samples were collected from 50 operations three times a year at different seasons was carried out. Samples were tested for presence of L. monocytogenes using a combination of enrichment and polymerase chain reaction tests. Data on putative risk factors were also collected. Listeria monocytogenes was detected in faecal samples from cows, calves and other animals on calf-cow operations at proportions of 3·1%, 3·75% and 2·5%, respectively. The organism was detected in 5·3% of cut-grass, 5·3% of soil, 14·3% of irrigation ditches, 3·1% of the ponds and 6·5% of water troughs samples. Listeria monocytogenes was less common in faecal (0·3%) and soil (0·75%) samples collected from feedlots.
Conclusions:  Listeria monocytogenes was present at a higher proportion among cow-calf operations than feedlots. There was no significant seasonal variation in the occurrence of this pathogen within the two types of operations.
Significance and Impact of the Study:  If risk mitigation strategies were implemented to reduce the public health risk these should focus in cow-calf operations.  相似文献

Owing to certain drawbacks, such as energy-intensive operations in conventional modes of wastewater treatment (WWT), there has been an extensive search for alternative strategies in treatment technology. Biological modes for treating wastewaters are one of the finest technologies in terms of economy and efficiency. An integrated biological approach with chemical flocculation is being conventionally practiced in several-sewage and effluent treatment plants around the world. Overwhelming responsiveness to treat wastewaters especially by using microalgae is due to their simplest photosynthetic mechanism and ease of acclimation to various habitats. Microalgal technology, also known as phycoremediation, has been in use for WWT since 1950s. Various strategies for the cultivation of microalgae in WWT systems are evolving faster. However, the availability of innovative approaches for maximizing the treatment efficiency, coupled with biomass productivity, remains the major bottleneck for commercialization of microalgal technology. Investment costs and invasive parameters also delimit the use of microalgae in WWT. This review critically discusses the merits and demerits of microalgal cultivation strategies recently developed for maximum pollutant removal as well as biomass productivity. Also, the potential of algal biofilm technology in pollutant removal, and harvesting the microalgal biomass using different techniques have been highlighted. Finally, an economic assessment of the currently available methods has been made to validate microalgal cultivation in wastewater at the commercial level.  相似文献

A model for the residual effect of previous years' treatment with the mycoinsecticide Metarhizium anisopliae var. acridum has been constructed based on field data from treatment of two different grasshopper species in east Niger. This model was incorporated in an earlier developed simulation model to evaluate control strategies for the grasshopper Oedaleus senegalensis in West Africa and the model system was used to assess the potential importance of second year's residual effects for the efficiency of different treatment strategies. It has earlier been hypothesized that the persistence of M. anisopliae and the ability of this agent to impose long-term control on grasshopper populations through secondary cycling in some cases might render the use of M. anisopliae more efficient than the use of chemical insecticides like fenitrothion. Results show that this effect is possible if M. anisopliae is used in a repetitive treatment strategy where control operations are concentrated in the main millet production areas.  相似文献

LigBuilder: A Multi-Purpose Program for Structure-Based Drug Design   总被引:5,自引:0,他引:5  
We have developed a new multi-purpose program, LigBuilder, for structure-based drug design. Within the structural constraints of the target protein, LigBuilder builds up ligands step by step using a library of organic fragments. Various operations, such as growing, linking, and mutation, have been implemented to manipulate molecular structures. The user can choose either growing or linking strategies for ligand construction and a genetic algorithm is adopted to control the whole construction process. Binding affinities of the ligands are estimated by an empirical scoring function and the bioavailabilities are evaluated by a set of chemical rules. Using thrombin and dihydrofolate reductase as examples, we have demonstrated that LigBuilder is able to generate chemical structures similar to the known ligands.  相似文献

Fluidized bed adsorption using a high-density synthetic resin has proven to be an invaluable technique for separating novel compounds from unfiltered fermentation broths during the very early stages of fermentation development, where product concentrations are typically in the parts per million range. Previous initial downstream processing strategies consisted of cell separation from whole broth or direct extraction with water-immiscible solvents, both of which resulted in lengthy time cycles, conflicts with existing operations requiring the use of high-cost centrifugal separators, and environmental/solvent recovery concerns. Laboratory and subsequent pilot plant process development work along with concomitant improvements in yield, quality, and time cycles are presented for one of several fluidized bed processes piloted in Merck's Natural Product Isolation facility.  相似文献

The robustness of bioprocesses is becoming increasingly important. The main driving forces of this development are, in particular, increasing demands on product purities as well as economic aspects. In general, bioprocesses exhibit extremely high complexity and variability. Biological systems often have a much higher intrinsic variability compared with chemical processes, which makes the development and characterization of robust processes tedious task. To predict and control robustness, a clear understanding of interactions between input and output variables is necessary. Robust bioprocesses can be realized, for example, by using advanced control strategies for the different unit operations. In this review, we discuss the different biological, technical, and mathematical tools for the analysis and control of bioprocess robustness.  相似文献

Abstract: Production of bulk chemicals by biological processes is presently limited by failure of contemporary biological and bioreactor technology to deliver high product concentrations in high space-time yields in fluids of sufficiently low water content for subsequent down-stream processing operations. Limitations in the bioreactor portion of the process can arise due to failure to process sufficient substrate, substrate inhibition, inadequate rates or yields, and product inhibition. Various process approaches for addressing many of these limitations have been demonstrated or conceptualized. Less developed but potentially effective are genetic strategies addressing these process limitations. Ideally, the most effective combination of genetic and process approaches should be integrated in a synergistic fashion to maximize the economic potential of biological production of chemicals.  相似文献

Next-generation sequencing has allowed identification of millions of somatic mutations in human cancer cells.A key challenge in interpreting cancer genomes is to distinguish drivers of cancer development among available genetic mutations.To address this issue,we present the first webbased application,consensus cancer driver gene caller(C~3),to identify the consensus driver genes using six different complementary strategies,i.e.,frequency-based,machine learning-based,functional bias-based,clustering-based,statistics model-based,and network-based strategies.This application allows users to specify customized operations when calling driver genes,and provides solid statistical evaluations and interpretable visualizations on the integration results.C~3 is implemented in Python and is freely available for public use at http://drivergene.rwebox.com/c~3.  相似文献

Recent psychophysical studies on normal subjects, as well as brain imaging studies, have revised the concepts concerning the mechanisms underlying spatial orientation during navigation tasks. The emphasis has been put on internal models that allow the prediction of a planned trajectory and are essential in the steering of locomotion. Cognitive factors such as strategies and emotional parameters are now starting to be included in the research on spatial orientation. It is obvious that important individual and gender differences exist in the brain operations underlying spatial orientation in humans, which may help to understand the construction of a coherent perception and the organic neural disorders related to the internal representation of space.  相似文献
