Why do fish die after severe exercise?   总被引:7,自引:1,他引:6  
Trout fitted with dorsal aortic cannulae were subjected to 6 min of intensive exercise and monitored over the following 12 h recovery period. Delayed mortality was 40%; the majority of deaths occurred 4–8 h post-exercise. Surviving fish exhibited a short-lived haemoconcentration reflected in increased haematocrit, haemoglobin, plasma protein, Na+ and Ch- levels; an extended rise in plasma [K+]; a quickly corrected respiratory acidosis; and a more prolonged metabolic acidosis in concert with a rise in blood lactate. Dying fish exhibited very similar trends except for a significantly greater metabolic acidosis, lower plasma [Cl-], and the apparent accumulation of an unknown anion in the blood prior to death. Cardiac failure did not occur. Blood metabolic acid levels, while elevated, were only ∼ 50% of peak lactate anion levels and well within the normal range of tolerance, as were all other changes observed in the blood of non-survivors. The hypothesis that post-exercise mortality is due to excessive 'lactic acid' accumulation in the blood is discounted. It is suggested that intracellular acidosis may be the proximate cause of death.  相似文献

Multi-wavelength, differential spectroscopy was used to examine the effects of transient hypoxia on oxygen delivery and intracellular utilization in the brain of developing rats. The in vivo redox status of cytochrome a,a3 was compared simultaneously with changes in relative haemoglobin saturation and blood volume in the cerebral cortex during lowered FiO2. During hypoxia, neonates maintained their intracellular cytochrome a,a3 redox state as well as did adults, but did so through unusual characteristics, including: (1) maintenance of haemoglobin oxygenation at lower FiO2; (2) regulation of cerebral blood volume at blood pressures below the point at which autoregulation would fail in the adult; and (3) the capacity to tolerate a greater reduction of cytochrome a,a3 relative to haemoglobin desaturation at lowered FiO2. These data suggest that mechanisms which protect the neonate from hypoxic insult involve preservation of oxygen delivery, increased respiratory compensation for metabolic acidosis, and maintenance of cellular energy requirements predominantly through anaerobic metabolism.  相似文献

Blood gas and acid-base status was determined in 126 Caesarean-derived calves. The newborn calves were assigned by venous blood pH value at birth to three groups as follows: Group 1 (normal): pH above 7.2; Group 2 (slight acidosis): pH 7.2 to 7.0; and Group 3 (severe acidosis): pH below 7.0. Following Caesarean section births 80 (63.5%) calves had normal acid-base values, while 30 (23.8%) had a slight acidosis, and 16 (12.7%) had severe acidosis. The degree of hypoxia was similar in each group. Six calves (37.5%) in Group 3 died within 48 h of birth. The blood gas and acid-base status of Caesarean-derived. calves was not significantly influenced by any examined parameters with the exception of sex in Groups 1 and 2. The occurrence of meconium-stained calves was 9.1% (n = 11), and only two calves were slightly or severely acidotic immediately after birth.  相似文献

550 blood specimens from the umbilical cord of newborn babies from Apulia have been screened in order to detect Hb Bart's. The electrophoresis of the haemoglobin by Cellogel (Tris Glycine pH 8.6) carried out on the 550 specimens have revealed in 43 variable quantities of Hb Bart's: 34 cases (6,18%) showed non measurable quota, whereas in the other 9 cases (1,63%) Hb Bart's varied from 2,19% to 26%. The haemoglobin biosynthesis "in vitro" of the baby presenting 26% of Hb Bart's has been reported.  相似文献

Summary Strenuous 5-min exercise resulted in a 0.3 unit drop in the dorsal aortic pH of striped bass. The acidosis was metabolic: the blood lactate concentration increased during the exercise, whereas blood CO2 tension decreased. Dorsal aortic oxygen content was maintained despite the acidosis. This was a result of increased blood O2 tension, haemoglobin concentration and red cell volume, decreased cellular nucleoside triphosphate (NTP) concentration, and decreased proton gradient across the red cell membrane. When the fish were treated with the beta-antagonist, propranolol, before the exercise, the arterial oxygen content decreased significantly in the stress. The mean cellular haemoglobin concentration and cellular NTP concentration increased slightly, and the proton gradient across the red cell membrane decreased less than in control exercise. These results show that the beta-adrenergic responses of striped bass red cells play an important role in maintaining the arterial O2 content in stress.Abbreviation NTP nucleoside triphosphates  相似文献

In a prospective audit of the obstetric management of 1210 consecutive deliveries the association was investigated between the need for operative delivery for fetal distress during labour and the condition of the newborn infant. Operative delivery was performed for only 11.5% of the newborn infants with severe acidosis at birth (umbilical artery pH less than 7.12, base deficit greater than 12 mmol (mEq)/1), 24.1% of those with an Apgar score less than 7 at one minute, and 15.8% of those with both severe acidosis and a one minute Apgar score less than 7. Most of the infants delivered operatively were in a vigorous condition at birth and did not have severe acidosis. Fetal blood sampling was done in 4.0% of labours. As none of the fetal blood values were less than 7.20 and only three of the infants sampled in utero suffered severe acidosis at birth, fetal blood sampling would have had to be performed much more often to provide a useful guide to metabolic state at birth. While the large majority of "at risk" fetuses had continuous fetal heart rate monitoring in labour, this had not been provided in 48.7% of the labours of infants with severe acidosis, 38.7% of infants with a one minute Apgar score less than 7, and 47.4% of infants with both severe acidosis and a one minute Apgar score less than 7. Continuous fetal heart rate monitoring was associated with a much higher incidence of operative delivery for fetal distress than was intermittent fetal heart rate auscultation. These results suggest an urgent need to review present methods for assessing the intrapartum condition of the fetus, making the diagnosis of fetal distress, and assessing the condition of the infant at birth.  相似文献

The objective of this study was to determine the effect of sublethal copper and zinc concentrations at a neutral and an acidic pH, on selected haematological parameters as well as on the total osmolality and electrolyte concentrations of Oreochromis mossambicus. In general, at neutral pH copper and zinc caused blood acidosis, increases in circulating white blood cell numbers, causing stimulation of the immune system, and a rapid release of red blood cells from haemopoietic tissue, as reflected in the decreases in mean corpuscular volume. Increases in red blood cells are attributed to an increase in the oxygen-carrying haemoglobin as an adaptation to altered respiratory homeostasis caused by copper and zinc. These increases are therefore a secondary reaction to the metals and not the result of direct stimulation of the haemopoietic tissue. In contrast, at an acidic pH copper and zinc concentrations usually caused blood alkalosis and decreases in white blood cell numbers, due to the bioconcentration of metals which blocks and suppresses the leucopoietic tissue. Decreases were also recorded in red blood cell counts, haemoglobin, haematocrit and mean corpuscular volume, which can be ascribed to anaemic and hypoxic conditions, gill damage and impaired osmoregulation.  相似文献

In a rat model of volume-controlled irreversible haemorrhagic shock, which results in a severe metabolic acidosis and the death of all control animals within 30 min., intracerebroventricular injection of histamine (100 nmol) produces a prompt and long-lasting increase in mean arterial pressure and heart rate, with a 100% survival of 2 h after treatment. Histamine action is accompanied by a decrease in haematocrit value, haemoglobin concentration, erythrocyte and platelet count, and an increase in residual blood volume at the end of the experiment (2 h). Cardiovascular effects are also associated with a long-lasting rise in respiratory rate and biphasic blood acid-base changes - initial increase of metabolic acidosis with the decrease in arterial and venous pH, bicarbonate concentration and base excess, followed by almost a complete recovery of blood gas and acid-base parameters to the pre-bleeding values, with normalisation of arterial and venous pH, Pco2 bicarbonate concentration and base excess at the end of experiment. It can be concluded that in the late phase of central histamine-induced reversal of haemorrhagic hypotension there is almost a complete restoration of blood gas and acid-base status due to circulatory and respiratory compensations, while accompanying haematological changes are the result of the haemodilution and the increase in residual blood volume.  相似文献

The umbilical venous oxygen and carbon dioxide tensions, pH, lactate and glucose concentrations, nucleated red cell (erythroblast) count, and haemoglobin concentration were measured in 38 cases of intrauterine growth retardation in which fetal blood sampling was performed by cordocentesis. The oxygen tension was below the normal mean for gestational age in 33 cases; in 14 it was below the lower limit of the 95% confidence interval for normal pregnancies. The severity of fetal hypoxia correlated significantly with fetal hypercapnia, acidosis, hyperlacticaemia, hypoglycaemia, and erythroblastosis. These findings indicate that "birth asphyxia" is not necessarily due to the process of birth.  相似文献

Whole blood was tapped from the embryos of anaesthetized pregnant New Zealand rats using a micropipette of approximately 250 microm diameter, inserted through the uterine wall via a 5-mm incision in the anterior abdominal wall. The procedure was repeated 3 times (days 15, 17 and 19 of fetal life) in the same pregnant rats, and thereafter, twice in the surviving newborns (days 14 and 30 of neonatal life). Osmotic fragility of the red blood cells (RBC), reticulocyte counts and haemoglobin values of each sample were analysed. The fragility curve of the fetuses showed the presence of 2 types of cell population: a fragile and a more resistant type. This was correlated with the number of immature RBC (reticulocytes) in the samples. Starch gel electrophoresis of the haemoglobin revealed 4 fractions in both the fetuses and newborn animals.  相似文献

目的探讨预负荷/共同负荷联合血管收缩药在剖宫产术中对产妇和新生儿血气的影响。方法将择期剖宫产产妇(ASA I~Ⅱ),在排除酸中毒(pH〈7.35或BE〈-6)或碱中毒(pH〉7.45或BE〉6)后,随机分为麻黄素组(E组)和去氧肾上腺素组(P组),每组30例。两组预负荷晶体液乳酸钠林格氏液6~8ml/kg,穿刺到硬膜外间隙时,分别静脉给予麻黄素5mg、去氧肾上腺素100μg。腰麻注药后,两组均输入羟乙基淀粉130/0.4氯化钠溶液10ml/kg(共同负荷),并分别静脉泵注麻黄素和去氧肾上腺素。观察记录两组产妇的BP、HR、ECG、SpO2,至胎儿娩出即刻查母体和脐动脉血血气,记录新生儿出生后第1min及第5min时的Apgar评分。结果 P组脐动脉血pH、BE值均高于E组(P〈0.05)。结论预负荷/共同负荷联合血管收缩药能有效减少腰麻后低血压的发生,维持产妇血流动力学稳定。去氧肾上腺素比麻黄素能更好地减少新生儿酸中毒的发生。  相似文献

Capture of muskellunge by angling resulted in a reduction of blood pH, elevated lactic acid concentrations, and a drop in total carbon dioxide and bicarbonate concentrations. The acidaemia was most severe immediately after capture and began to decline well before the blood lactate levels rose. Blood lactate levels were not as high as those characterizing fatigue in most other species. Recovery from the acidosis required 12 to 18 h and was accompanied by declines of 22% and 40% in haemoglobin and haematocrit levels respectively. With the exception of dying fish, there were only slight fluctuations in plasma sodium and potassium levels during recovery, indicating that there was no severe ionoregulatory dysfunction.
Thirty per cent of all angled muskellunge died. The last stages immediately preceding death were characterized by declining blood pH and elevated potassium levels.  相似文献

Infection perorally with enteropathogenic E. coli (ST +LT) bacteria in 57 newborn piglets gave rise to watery diarrhoea in 50 (88%) piglets and was lethal in 17 (34 %) cases. The diarrhoea was associated with a progressing partially compensated metabolic acidosis indicated by significantly decreased pH, pCO2 and RE values. The acidosis (BE-values) was significantly correlated with increased blood LA and serum K+ values. The dehydration during the disease was confirmed by increased Hb, Hct, serum protein and urea values as well as loss of weight. The changes were most pronounced in piglets that died and a BE value of —10 mmol/1 seemed to be a critical limit at which the prognosis could be considered poor. The changes in acid-base status and water balance was confirmed in 64 piglets with spontaneous cases of E. coli diarrhoea.  相似文献

A case of Reye''s syndrome in a newborn infant, believed to be the first recorded in the perinatal period, is reported. The clinical, biochemical and histological findings are those previously recognized in older infants. The intractable acidosis, hypoglycemia and extreme degree of lactic acidosis reflect an advanced degree of hepatic involvement, probably initiated in utero, with resultant impairment of glycogenolysis and intermediary metabolism.  相似文献

We hypothesized that part of the newborn tolerance of asphyxia involves strong ion changes that minimize the cerebral acidosis and hasten its correction in recovery. After exposure of newborn puppies to 15 or 30 min experimental asphyxia (inhalation of gas with fractional concentration of CO2 and of O2 in inspired gas = 0.07-0.08 and 0.02-0.03, respectively), blood lactate increased to 13.2 and 23.4 mmol/l, respectively, brain tissue lactate increased to 14.4 and 19.7 mmol/kg, and cerebrospinal fluid (CSF) lactate increased to 7.6 and 14.4 mmol/l. We presume that the tissue lactate increase reflects increases in brain cell and extracellular fluid lactate concentration. The lactate increase, a change that will decrease the strong ion difference (SID), [HCO3-], and pH, was accompanied by increases in Na+ (plasma, CSF, brain), K+ (plasma, CSF), and osmolality without change in Cl-. After 60-min recovery, plasma and brain lactate decreased significantly, but CSF lactate remained unchanged. [H+] recovery was more complete than that of the strong ions due to hyperventilation-induced hypocapnia. We conclude that during asphyxia-induced lactic acidosis, changes in strong ions occur that lessen the decrease in SID and minimize the acidosis in plasma and CSF. To the extent that the increase in brain tissue sodium reflects increases in intra-and extracellular fluid sodium concentration, the decrease in SID will be less in these compartments as well. In recovery, CSF ionic values change little; plasma and brain tissue lactate decrease with a similar time course, and the [H+] is rapidly returned toward normal by hypocapnia even while the SID is below normal.  相似文献

The frequency of haemoglobinopathies among the Hong Kong Chinese newborn was examined by screening 932 consecutive cord blood samples by electrophoresis. The findings indicate that 49 (5.2%) had electrophoretic abnormalities, of which 39 (4.2%) demonstrated the presence of Hb Bart's. 14 of these 39 babies were re-investigated after 1 year and all were proven to carry the alpha-thalassaemia gene. This is the first study in which Chinese babies with Hb Bart's in cord blood had the diagnosis of alpha-thalassaemia trait confirmed in later life. 3 out of 4 other babies had different haemoglobin variants, namely Hb alpha Q, Hb J and Hb New York. 1 out of the 6 babies who had elevated A2 levels at birth was found to carry the beta-thalassaemia trait. None had a serious haemoglobinopathy.  相似文献

Summary The blood oxygen binding properties of rainbow trout responded to environmental hypoxia (the oxygen saturation of water 30% at 11°C) in three ways. The quickest response was a moderate acidosis, leading to slightly lowered blood oxygen loading due to the Bohr effect. The second response, an increase of blood oxygen carrying capacity, was completed with 6 h from the onset of hypoxia. The speed of the response suggests that the formation of new haemoglobin played no practical role, the increase being caused either by a decrease of plasma volume or the liberation of erythrocytes from a storage organ. The slowest response, a 25% increase of the blood oxygen affinity within a week of hypoxia, was probably caused by the concurrent decrease of the erythrocyte ATP concentration from 4.45 to 2.51 mol/ml erythrocytes.  相似文献

The relative amount of regional cerebral oxygen transport was compared between different preterm infants by performing measurements of cerebral blood flow velocity, mean arterial blood pressure, whole blood viscosity and haemoglobin content for each individual. In addition the percentage of fetal haemoglobin was determined. On 25 occasions measurements of fetal haemoglobin and cerebral oxygen transport have been performed prior to and following a blood transfusion with adult red blood cells. Comparison of the data for cerebral oxygen transport suggests that the actual amount of cerebral oxygen transport is lowest at fetal haemoglobin levels below 30% and will increase progressively as soon as the percentage of fetal haemoglobin rises about 30%. Thus, at increasing fetal haemoglobin levels, cerebral haemodynamic mechanisms in the human neonate cause elevations of regional cerebral blood flow and oxygen transport. The found increase of cerebral blood flow and oxygen transport at high fetal haemoglobin levels will minimize the impeded dissociation and delivery of oxygen to brain tissues.  相似文献

Recently attention has been called on the possible role of acidosis in the increased methemoglobin formation in the erythrocyte of newborn infant. In the present paper the relations between acidosis and methemoglobin content in the red cells of newborns has been investigated. No significant differences between the percent of methemoglobin in the normal newborns and percent of methemoglobin in the newborns with acidosis has been found. In addition, no correlations between the base excess and percent of methemoglobin has been observed. On the contrary, two newborns with low glucose-6-phosphate dehydrogenase activity demonstrated a significantly increased methemoglobin content in their red cells. The results of our study do not confirm a key role of acidosis in the mechanism of methemoglobin formation in the neonate. It is likely than impairment of red cell metabolism should be the main factor in the formation of methemoglobin in the first days of life.  相似文献

The present study investigates the integrity of the blood-brain barrier to H+ or HCO3- during acute plasma acidosis in 35 newborn piglets anesthetized with pentobarbital sodium. Cerebrospinal fluid acid-base balance, cerebral blood flow (CBF), and cerebral oxygenation were measured after infusion of HCl (0.6 N, 0.191-0.388 ml/min) for a period of 1 h at a constant arterial PCO2 of 35-40 Torr. HCl infusion resulted in decreased arterial pH from 7.38 +/- 0.01 to 7.00 +/- 0.02 (P less than 0.01). CBF measured by the tracer microsphere technique was decreased by 12% from 69 +/- 6 to 61 +/- 4 ml.min-1.100 g-1 (P less than 0.05). Infusion of 0.6 N NaCl as a hypertonic control had no effect on CBF. Cerebral metabolic rate for O2 and O2 extraction was not significantly changed from control (3.83 +/- 0.20 ml.min-1.100 g-1 and 5.7 +/- 0.6 ml/100 ml, respectively) during acid infusion. Cerebral venous PO2 was increased from 41.6 +/- 2.1 to 53.8 +/- 4.0 Torr by HCl infusion (P less than 0.02) associated with a shift in O2-hemoglobin affinity of blood in vivo from 38 +/- 2 to 50 +/- 1 Torr. Cisternal cerebrospinal fluid pH decreased from 7.336 +/- 0.014 to 7.226 +/- 0.027 (P less than 0.005), but cerebrospinal fluid HCO3- concentration was not changed from control (25.4 +/- 1.0 meq/l). These data suggest that there is a functional blood-brain barrier in newborn piglets, that is relatively impermeable to HCO3- or H+ and maintains cerebral perivascular pH constant in the face of acute severe arterial acidosis.
