对花椒珠心胚及胚乳的发生和发育过程进行了详细的细胞学及细胞学研究。主要研究结果如下;珠心胚发生前,有性胚囊发育过程中从大孢子发生到胚囊形成的各个阶段均可发生退化,退化频率50%,未退化的胚囊发育成熟,成熟胚囊仅含卵器和两个极核。卵器最终退化,极核不经受精自发形成胚肥。当胚乳游离核达到15或32个时,最早的珠心胚原始细胞由靠近胚囊球孔端的珠心细胞分化形成。随着子房生长,多个原始细胞持续不断地从珠孔端  相似文献

柑桔类珠心胚现象对研究其进化和种质保存方面有着重要意义,但是给其有性杂交育种工作造成相当大的麻烦:它不仅使杂交后代难于区分合子苗和珠心苗,而且会使合子胚发育中途天折。解决的途径是适时分离合子胚进行单独培养。为此,本工作于柑桔胚胎发育的不同时期取样、固定、观察,研究其合子胚发育规律,确定珠心胚侵入胚囊的时期。结果表明:一般情况下,授粉后30d,合于处于单细胞状态(Fig.1A);授粉后40d;合子开始分裂(Fig.1B);授粉后45d,合子胚形成8-16个细胞,珠心胚开始分裂(Fig.1C);授粉后50d,合子形成球形胚,珠心胚尚未侵入胚囊(Fig.1D&E);授粉后55d,合子胚为球形胚或早心形胚,开始有少数珠心胚侵入胚囊(Fig.1D&E);授粉后60d,合子胚为心形胚(Fig.1G)。授粉后80d,合子胚为晚心形,珠心胚巨大量侵入并迅速发育(Fig.1H)。杂交的组合对合子,胚珠及果实发育均有一定影响,主要取决于杂交亲本。与种间杂交相比,属间杂交其合子分裂较迟,合子胚发育较慢,珠心胚侵入时期也较迟(Tablel,Fig.2)。这种发育速度的差异,引起所产生的合子胚的大小(Table2)珠心胚侵率(Tab  相似文献

对水稻HDAR胚胎发育过程的进一步研究表明,水稻HDAR中有2.72%(17/434)的不定胚发生和发育。其不定胚起源于胚珠内的珠心细胞。不定胚起始细胞启动分裂时,胚囊发育至2核或4核时期,8核胚囊时期,胚珠内已形成多细胞不定胚结构。随后不定胚细胞不断分裂并逐渐挤进胚囊。开花传粉后,不定胚利用胚乳提供的营养可以继续发育和分化。不定胚可以和合子胚一起发育,有时合子胚败育,不定胚继续发育并分化。讨论了水稻HDAR中不定胚的发生,及其发生远早于合子胚的意义。  相似文献

Cytological and histological studies of seeds from three facultative apomictic Citrus cultivars show that adventive embryos develop, as a rule, from the first few cell layers of the nucellus adjacent to the embryo sac in the micropylar half and occasionally from the chalazal end. The adventive embryos initiated in nucellar tissue away from the embryo sac and most of those initiated from the chalazal end of the nucellus do not develop beyond the one-celled stage. When two or more embryos are developing in the same seed, the successful development of a given embryo depends on its location in relation to access to nutrients from the endosperm. The presence of a zygote and triploid endosperm in seeds with adventive embryos, the abortion of seed when endosperm degenerates, and the lack of seed set without pollination indicate that pollination and fertilization are essential for in vivo adventive embryogenesis.  相似文献

The antipodal cells have been the stepchildren in most investigations of the female gametophyte. In Hordeum vulgare cv. Bomi, three antipodal cells are originally developed chalazally but because of differential growth of the embryo sac they soon become laterally situated and their number increases to 35–50 cells and the shape, size and structure of the cells change in the time before as well as after fertilization. The cells persist until about 60–70 h after pollination. At that time, the embryo consists of about 12–15 cells and a cellularization of the nuclear endosperm has started peripherally. The size of nuclei, and especially nucleoli, in the antipodal cells increases tremendously in the investigated period and the amounts of organelles also change. The walls of antipodals are diversified depending on which cells they are separating, and wall invaginations are developed especially between antipodal cells and surrounding nucellar cells in the placental region. The mitochondria appear in various shapes in section view, very often as cups or dumbbells with a rim in the ends containing cristae and a thin cristae-free base. These bases are sometimes stretched out as thin parts and at last a simple parting occurs. Binary fissions of the plastids happen especially in the hours before and just after egg fertilization. ER is extraordinarily well developed in the whole period of investigation and many ribosomes are attached to the membranes. Dictyosomes form numerous vesicles, especially in the antipodals near the nucellar cells in the placental region. These ultrastructural details support the opinion that antipodal cells may play an important role in the embryo sac and are able to be responsible for the supply of nutrition for the whole gametophyte and take part in the supply of nutrition during embryo formation.  相似文献

The nucellar ultrastructure of apomictic Panicum maximum was analyzed during the meiocytic stage and during aposporous embryo sac formation. At pachytene the megameiocyte shows a random cell organelle distribution and sometimes only an incomplete micropylar callose wall. The chalazal nucellar cells are meristematic until the tetrad stage. They can turn into initial cells of aposporous embryo sacs. The aposporous initials can be recognized by their increased cell size, large nucleus, and the presence of many vesicles. The cell wall is thin with few plasmodesmata. If only a sexual embryo sac is formed, the nucellar cells retain their meristematic character. The aposporous initial cell is somewhat comparable to a vacuolated functional megaspore. It shows large vacuoles around the central nucleus and is surrounded by a thick cell wall without plasmodesmata. In the mature aposporous embryo sac the structure of the cells of the egg apparatus is similar to each other. In the chalazal part of the egg apparatus the cell walls are thin and do not hamper the transfer of sperm cells. Structural and functional aspects of nucellar cell differentiation and aposporous and sexual embryo sac development are discussed.  相似文献

研究了四倍体双穗雀稗(Paspalum distichum L)无孢子生殖胚囊、胚胎发育以及假受精特点。当其大孢子母细胞发育至四分体阶段时,大多数情况下会发生四分体退化,同时有多个特化珠心细胞发育为1—3个无孢子生殖胚囊的现象。成熟无孢子生殖胚囊一般3核,包括1个卵细胞和2个极核。卵细胞在抽穗前就能自发分裂形成原胚团,而极核则在抽穗和传粉后参与假受精形成胚乳。当胚珠内存在多个无孢子生殖胚囊时,只是靠近珠孔端的1个无孢子生殖胚囊内的极核与精核结合,而其它的并不参与。种子成熟后出现很低频率的二胚苗。此外,还能观察到少量的有性生殖胚囊的发育以及有性生殖胚囊和无孢子生殖胚囊在同一胚珠中的发育现象,因此判断该类群为兼性无孢子生殖体。  相似文献

采用半薄切片技术和组织化学染色法对宁夏枸杞大孢子发生和雌配子体发育过程中的细胞结构变化及营养物质积累特征进行了观察。结果表明,(1)宁夏枸杞为中轴胎座,多室子房,倒生胚珠,单珠被,薄珠心类型。(2)位于珠心表皮下的孢原细胞可直接发育为大孢子母细胞,减数分裂后形成直线型大孢子四分体,合点端第一个大孢子发育为功能大孢子,胚囊发育类型为蓼型,具有珠被绒毡层。(3)初形成的胚囊外周组织中没有营养物质积累,成熟胚囊时期出现了大量的淀粉粒且呈珠孔端明显多于合点端的极性分布特征。(4)助细胞的珠孔端具有明显的丝状器结构,呈PAS正反应表现出多糖性质,成熟胚囊具有承珠盘结构。  相似文献

花椒与野花椒的胚囊发育类型属蓼型,成熟胚囊的卵器退化。花椒无雄花,不发生双受精,自发形成胚乳并产生珠心胚。野花椒虽有正常花粉,人工授粉后能萌发,但在花粉管长入胚囊之前卵器已解体,中央细胞中已形成胚乳游离核,因此也不发生双受精,由珠心细胞自发形成胚。这种现象是花椒和野花椒在长期进化过程中形成的一种十分特化的适应。  相似文献

Studies on the formation and development of the embryo sac of the apomictic material of Pennisetum squamulatum Fresen indicated that normal archesporial cell did form with consequent development of a megaspore mother cell and later meiotic division to give rise to a triad. But invariably the megaspore mother cell and the triad underwent degeneration after formation. During the period of formation or degeneration of the megaspore or the triad a number of nucellar cells around the degenerated sexual cell became much enlarged. Frequently, one of the enlarging nucellar cells near the micropylar end became vacuolated and then developed into an aposporous uninucleate embryo sac, which underwent two further mitotic divisions to form an aposporous four-nucleate embryo sac, where the four nuclei remained in the micropylar end. Thus in the mature aposporous embryo sac there were one egg cell, one synergid and one central cell (containing two polar nuclei). Antipodal cells were completely lacking. The pattern of development of the aposporous embryo sac resembles the panicum type. There were two types of embryo formed during apomictic development namely ( 1 ) The pre-genesis embryo--embryo formed without fertilization, 1 to 2 days before anthesis, and (2) The late-genesis embryo--derived from the unfertilized egg cells, 3 to 4 days after anthesis. In the late-genesis embryo type, the egg cell divided after the secondary nucleus has undergone division to form the endosperm nuclei. All egg cells developed vacuoles before they differentiated into embryos. The development of the aposporous embryo followed the sequence of the formation of globular, pearshaped embryo and full stages of differentiation. The unfertilized secondary nucleus divides to form free endosperm nuclei after being stimulated by pollination. The development of the endosperm belongs to the nuclear-type.  相似文献

通过透射电镜对水稻(Oryza sativa L.)功能大孢子形成开始至胚囊成熟期间胚囊壁的形成与发育进行观察,结果表明:水稻胚囊壁是在原有功能大孢子壁的基础上,通过与其周围退化珠心细胞留下的壁相叠合,使壁加厚。功能大孢子近合点端壁存在胞间连丝,其中个别胞间连丝可保留到八核胚囊。胚囊壁上内突最早于四核胚囊近珠孔端发生。八核胚囊形成后,内突的发育在胚囊不同的细胞中表现不同,其中以中央细胞最具特点,表现为先在中央细胞与珠心相接的近珠孔端和近合点端两个区域的胚囊壁上形成,以后近珠孔端胚囊壁上的内突大量增加,而近合点端的却增加不明显,中部胚囊壁上的内突出现的时间相对较晚。到胚囊成熟时,近珠孔端胚囊壁上内突的分布密度最大,中部次之,近合点端的最小,三个区域上内突的形态各异。反足细胞与珠心相接的胚囊壁上内突的形成时间较早,但以后的发育却相对缓慢,数量增加不明显。2个助细胞交界处胚囊壁上的丝状器在胚囊未明显膨大时已形成。卵细胞除在与助细胞交界处的壁外,其它部位不形成明显的内突结构。  相似文献

Cynara cardunculus is a native plant with flowers that are used traditionally in the manufacture of ewe’s cheese in the Iberian Peninsula. Milk clotting ability of the plant is attributed to the high concentrations of aspartic proteinases (APs), named cardosins, found in the flowers. Although these enzymes are well characterised on a molecular and biochemical basis, the biological role of the majority of plant APs is yet unassigned. We suspected APs play an important role in ovule function, and we characterised the maturation of the ovules of C. cardunculus and its Polygonum-type embryo sacs. The internal layer of the integument differentiates into an endothelium as described for other Asteraceae, with differentiation of two nucellar layers, a podium and a hypostase coinciding with the onset of pollen receptivity. In flowering plants, programmed cell death (PCD) events are essential for the success of nucellar maturation and consequent differentiation of a fully functional embryo sac. In C. cardunculus, nucellar PCD is integral to the maturation of the embryo sac, which in turn is closely correlated with the accumulation of the AP cardosin B specifically in the hypostase. The onset of cardosin B expression temporally coincides with the degeneration of nucellar cells. In fully mature embryo sacs, cardosin B is localised in both the hypostase and epistase, two regions that differentiate through PCD. Thus, cardosin B localisations closely correlate with events of PCD in the nucellus of C. cardunculus suggesting involvement in ovule and embryo sac development and further suggest the biological significance of APs like cardosin B, in this particular process. This work contributes new data to the plant AP research field and indicates an involvement of cardosin B in the PCD-dependent degeneration of the nucellus.  相似文献

Light microscopic observations were made on 22 ovules from fertile plants and 108 ovules from sterile plants of the cv. KS synaptic mutant, a highly male-sterile, female-sterile line of soybean [Glycine max (L.) Merr.] (2n = 2x = 40). Ovules of fertile siblings contained normal embryo sacs and embryos. Ovules from sterile plants contained various irregularities. The most consistent abnormality was the failure of the embryo sac to attain normal size. Small megasporocytes of irregular shape were seen; only one megasporocyte of normal shape and size was noted. No linear tetrads were found. However, two ovules contained nonlinear triads. A range from zero to 28 cells and nuclei, of various sizes, were identifiable in small megagametophytes and embryo sacs. Degeneration of these nuclei and cells was noted as early as the four-nucleate gametophyte stage. Other ovules contained degenerated nucellar centers without embryo sacs. Two ovules appeared to be normal. Late postpollination stages were marked by shrunken nucellus and integuments. The presence of pollen tube traces, endosperm, and aborting embryos in ovules of hand-pollinated flowers from sterile plants suggested that no incompatibility was involved. Degeneration of the gametophyte and embryo sac contents at many developmental stages indicated a wide array of effects, possibly resulting from meiotic irregularities similar to those seen in microsporogenesis of this mutant.  相似文献

New combinations for three species ofPolypleurum have been proposed. The Apinagia type of embryo sac is recorded for the first time in a species ofPolypleurum, P. filifolium. The Dicraea type of embryo sac found inP. dichotomum andP. munnarensis has been reinterpreted and renamed as the Polypleurum type. The embryo sac types met with in the family are discussed. Ultrastructure of the embryo sac lacking antipodals in prefertilization stages in Arabidopsis thaliana has been examined 2 hr before and 5 hr after manual cross pollination. The cytoplasm of both synergids before fertilization is rich in ribosomes, mitochondria, and rough endoplasmic reticulum, and also contains several microbodies and spherosomes. The filiform apparatus includes electron-dense material and a fibrous part. Many cortical microtubules appear in the filiform apparatus area. One of the two synergids degenerates before fertilization. The synergids, the egg cell, and central cell have a rich cytoskeleton of microtubules; only the synergids appear to contain microfilaments. At the chalazal end, the antipodals are initially present but degenerate by the time of pollination in most embryo sacs in the starchless line studied. The embryo sac is completely surrounded by a wall containing an electron-dense layer, separating it from the nucellus, including the chalazal end. When the antipodals have degenerated, the electron-dense layer disappears at the chalazal end only, and the wall between the central cell and the nucellus is homogeneous. Between the central cell and nucellar cells no plasmodesmata are found. The membranes of both antipodal cells at the chalazal end of the embryo sac appear sinuous, like those of transfer cells. The central cell has plastids preferentially distributed around the nucleus, but the other organelles are randomly distributed. The central cell in the embryo sac and the adjacent chalazal nucellar cells show a transfer-cell function in the embryo sac after the antipodals degenerate.  相似文献

对水蔗草 (ApludamuticaL .)的生殖方式进行研究 ,结果表明水蔗草进行兼性无融合生殖。胚囊发育分为两种类型 ,即有性生殖的蓼型和无孢子生殖的大黍型。无融合生殖胚囊频率为 6 0 .74%。在大孢子母细胞发育至四分体后 ,珠孔端的 3个大孢子解体。合点端的大孢子未解体时 ,邻近大孢子的 1个珠心细胞开始特化 ,形成无融合生殖的原始细胞 ,由该原始细胞发育形成有 1个卵细胞、1个助细胞和 2个极核的四核胚囊。  相似文献

Calypso bulbosa is a terrestrial orchid that grows in north temperate regions. Like many orchids, the Calypso has ovules that are not fully developed at anthesis. After pollination, the ovule primordia divide several times to produce a nucellar filament which consists of five to six cells. The subterminal cell of the nucellar filament enlarges to become the archesporial cell. Through further enlargement and elongation, the archesporial cell becomes the megasporocyte. An unequal dyad results from the first meiotic division. A triad of one active chalazal megaspore and two inactive micropylar megaspores are the end products of meiotic division. Callose is present in the cell wall of the megaspore destined to degenerate. In the mature embryo sac the number of nuclei is reduced to six when the chalazal nuclei fail to divide after the first mitotic division. The chalazal nuclei join the polar nucleus and the male nucleus near the center of the embryo sac subsequent to fertilization.  相似文献

蚕豆胚珠发育过程中淀粉动态变化如下:1.发育早期,整个胚珠中未见淀粉粒。其后首先在合点区出现淀粉,而后从合点向珠孔逐渐扩大分布范围。2.珠心和内、外珠被中均含有淀粉粒,尤以内珠被的淀粉增长迅速,数量多、个体大。受精后,内珠被解体,淀粉出现在外珠被细胞中,推测营养物质可通过整个胚囊表面进入其中。3.合点与胚囊之间的珠心细胞特化或长形。可能有助于营养物质进入胚囊。4.功能大孢子中贮存丰富的淀粉粒,它和珠心细胞一起是胚囊发育时的营养来源。5.卵细胞受精后,所含淀粉粒的数量和大小明显增长,随着合子和胚细胞的分裂,其中贮存的淀粉逐渐被消耗,到多细胞球形胚时完全消失。6.胚乳核周围始终未出现淀粉粒。7.胚器官分化之后,子叶和胚轴等处逐渐出现淀粉粒,其中生长活跃的结构如生长点、维管束等不贮存淀粉。8.子叶中的淀粉粒含量迅速增加,颗粒特大,是种子内营养物质的最终贮存场所。  相似文献

Summary The present investigation deals with some of the organizational and histochemical aspects of the embryo sac ofScilla sibirica. Both the synergids and egg cell are invested by PAS-positive complete walls. The filiform apparatus comprises an elaborate system of fibrillar projections, showing extensive ramifications. The micropylar region of the embryo sac wall from where the filiform apparatus originates is composed of three distinct layers. On a histochemical basis it may be surmised that, unlike the egg cell, the synergids are metabolically very active. Two kinds of wall ingrowths (i) massive and highly branched very much akin to the filiform apparatus, and (ii) small tuberculate wall projections, are unique to the antipodal cells of S.sibirica. Small tuberculate projections have also been observed along the wall of the central cell adjacent to the nutrient-rich nucellar cells. The antipodals and the central cell show the presence of starch grains and abundant total proteins. All the cell types in the embryo sac ofS. sibirica are structurally so organized as to meet the requirements of its nutrition during pre- and postfertilization development. The presence of abundant PAS-positive granular substance in the cells of nucellar epidermis probably establishes a gradient which assists in the pollen tube growth.  相似文献
