Biofilm formation in an ice cream plant   总被引:1,自引:0,他引:1  
The sites of biofilm formation in an ice cream plant were investigated by sampling both the production line and the environment. Experiments were carried out twice within a 20-day period. First, stainless steel coupons were fixed to surfaces adjacent to food contact surfaces, the floor drains and the doormat. They were taken for the analysis of biofilm at three different production stages. Then, biofilm forming bacteria were␣enumerated and also presence of Listeria monocytogenes was monitored. Biofilm forming isolates were selected on the basis of colony morphology and Gram’s reaction; Gram negative cocci and rod, Gram positive cocci and spore forming isolates were identified. Most of the biofilm formations were seen on the conveyor belt of a packaging machine 8 h after the beginning of the production, 6.5 × 103 cfu cm−2. Most of the Gram negative bacteria identified belong to Enterobacteriaceae family such as Proteus, Enterobacter, Citrobacter, Shigella, Escherichia, Edwardsiella. The other Gram negative microflora included Aeromonas, Plesiomonas, Moraxella, Pseudomonas or Alcaligenes spp. were also isolated. Gram positive microflora of the ice cream plant included Staphyloccus, Bacillus, Listeria and lactic acid bacteria such as Streptococcus, Leuconostoc or Pediococcus spp. The results from this study highlighted the problems of spread of pathogens like Listeria and Shigella and spoilage bacteria. In the development of cleaning and disinfection procedures in ice cream plants, an awareness of these biofilm-forming bacteria is essential for the ice cream plants.  相似文献

Cryopreservation of human umbilical cord blood (UCB) typically involves the cryoprotectant dimethylsulfoxide (DMSO), however, infusional toxicity and reductions in cell viability remain a concern. Ice recrystallization (IR) is an important source of cryopreservation-induced cellular injury and limits the stem cell dose in UCB units. Carbohydrates have wide-ranging intrinsic IR inhibition (IRI) activity related to structural properties. We investigated the impact of carbohydrate IRI on cell viability, induction of apoptosis and hematopoietic progenitor function in cryopreserved UCB. Mononuclear cells (MNCs) from UCB were cryopreserved in storage media containing specific carbohydrates (200 mM) and compared to 5% DMSO. Samples were analyzed under conditions of high IR (‘slow’ thaw) and low IR (‘fast’ thaw). Thawed samples were analyzed for viability and apoptosis by flow cytometry and hematopoietic function using colony-forming unit (CFU) assays. IRI of carbohydrate solutions was determined using the ‘splat cooling’ assay. Greater IRI capacity of carbohydrates correlated with increased yield of viable MNCs (r2 = 0.92, p = 0.004) and CD34(+) cells (r2 = 0.96, p = 0.019) after thawing under conditions of high IR. The correlations were less apparent under conditions of low IR. Carbohydrates with greater IRI modulate the induction of early apoptosis during thawing, especially in CD34+ cells (r2 = 0.96, p = 0.0001) as compared to total mononuclear cells (p = 0.006), and preserve CFU capacity in vitro (r2 = 0.92, p = <0.0001). Our results suggest that carbohydrates with potent IRI increase the yield of non-apoptotic and functional hematopoietic progenitors and provide a foundation for the development of novel synthetic carbohydrates with enhanced IRI properties to improve cryopreservation of UCB.  相似文献

Ice recrystallization, the growth of large ice crystals at the expense of small ones, stresses freeze tolerant organisms and causes spoilage of frozen foods. This process is inhibited by antifreeze proteins (AFPs). Here, we present a simple method for determining the ice recrystallization inhibition (RI) activity of an AFP under physiological conditions using 10microl glass capillaries. Serial dilutions were prepared to determine the concentration below which RI activity was no longer detected, termed the RI endpoint. For type III AFP this was 200nM. The capillary method allows samples to be aligned and viewed simultaneously, which facilitates RI endpoint determination. Once prepared, the samples can be used reproducibly in subsequent RI assays and can be archived in a freezer for future reference. This method was used to detect the elution of type III AFP from a Sephadex G-75 size-exclusion column. RI activity was found at the expected V(e) for a 7kDa protein and also unexpectedly in the void volume.  相似文献

Reliable assessment of the effect of a solute upon ice recrystallization is accomplished with "splat cooling," the impaction of a small solution droplet onto a very cold metal plate. The ice disc has extremely small crystals, and recrystallization can be followed without confusing effects caused by grain nucleation. This method confirms the exceptionally strong recrystallization inhibition effect of antifreeze protein from Antarctic fish and shows that grain growth rate is a sensitive function of both grain size and solute concentration.  相似文献

Extracellular macromolecules associated with Antarctic sea ice diatoms were previously shown to have ice-binding activities. The function of these ice-active substances (IASs) has not been identified. Here we show that two of the IASs have a strong ability to inhibit the recrystallization of ice, possibly signifying a cryoprotectant function. To test this possibility, two species of marine diatom (one Antarctic and one temperate) were subjected to a single freeze-thaw cycle (approximately 20h at -4 or -5 degrees C) in the presence or absence of IAS. Viability, based on a double staining technique, was 15-29% higher in the presence of IAS. Etching of single crystal ice hemispheres grown from dilute IAS solutions indicated that the IASs bind to specific faces of ice and are incorporated into the ice lattice. Together, these results suggest that the IASs acts as a cryoprotectant, probably through some ice-binding mechanism.  相似文献

To understand the molecular basis of freezing tolerance in plants, several low temperature-responsive genes have been identified from wheat. Among these are two genes named TaIRI-1 and TaIRI-2 (Triticum aestivum ice recrystallization inhibition) that are up-regulated during cold acclimation in freezing-tolerant species. Phytohormones involved in pathogen defense pathways (jasmonic acid and ethylene) induce the expression of one of the two genes. The encoded proteins are novel in that they have a bipartite structure that has never been reported for antifreeze proteins. Their N-terminal part shows similarity with the leucine-rich repeat-containing regions present in the receptor domain of receptor-like protein kinases, and their C-terminus is homologous to the ice-binding domain of some antifreeze proteins. The recombinant TaIRI-1 protein inhibits the growth of ice crystals, confirming its function as an ice recrystallization inhibition protein. The TaIRI genes were found only in the species belonging to the Pooideae subfamily of cereals. Comparative genomic analysis suggested that molecular evolutionary events took place in the genome of freezing-tolerant cereals to give rise to these genes with putative novel functions. These apparent adaptive DNA rearrangement events could be part of the molecular mechanisms that ensure the survival of hardy cereals in the harsh freezing environments.  相似文献

We investigated the shade adaptation of a seasonally well-developed ice algal community in thin sea ice at Saroma-Ko Lagoon, Hokkaido, Japan on 3–4 March 2006 and 4–5 March 2007, by examining photosynthetic pigment concentrations, the chlorophyll a-specific light-absorption coefficient (a ph *), and the light-saturation index (E k ). The high proportions of photosynthetic pigments, including chlorophyll a, fucoxanthin, and chlorophyll c, and the low values of a ph *(440) and a ph *(675) suggested that the lagoon’s ice algal community was shade-adapted. The high ratio of E k to total photosynthetically active radiation (PAR) in the ice algal habitat suggested that the degree of shade adaptation is weak. Scaling of E k to total PAR could be extended to studies of the degree of photoadaptive succession of ice algal communities in the Northern Hemisphere. The degree of shade adaptation of ice algal communities in the Northern Hemisphere might be related to ice thickness, regardless of latitude.  相似文献

An infiltration community was the dominating ice algal community in pack-ice off Queen Maud Land, Southern Ocean, in January 1993. The community was dominated by autotrophic processes, and the most common species were the prymnesiophyte Phaeocystis antarctica and the diatoms Chaetoceros neglectus and Fragilariopsis cylindrus. The concentration of chlorophyll a was 1.3–47.9 μg l−1, and the inner part of the community was nitrate depleted. Uptake rates of nitrate, nitrite, ammonium, urea and amino acids were measured using 15N. Nitrate was the major nitrogen source for ice algal growth (67 ± 6% nitrate uptake). It is suggested that % nitrate uptake in the infiltration community decreases during the growth season, from 92% during spring (literature data) to 67% during summer. Scalar irradiance in the infiltration community was high and variable. It reached ca. 2000 μmol m−2 s−1 at some locations, and nitrate uptake rate was potentially photoinhibited at irradiances >500 μmol m−2 s−1. Nitrate uptake rate in an average infiltration community (0.6 m of snow cover) was lowered by 13% over a 2-week period due to photoinhibition. Ice-active substances (IASs), i.e., macromolecular substances that modify the shape of growing ice crystals, were previously found to be associated with various terrestrial and aquatic photosynthetic organisms from Antarctica, but their chemical nature and function are unknown. In this study, we used the ice-binding properties of the IASs to semipurify IASs from a cyanobacterial mat, a eukaryotic green alga (Prasiola sp.), and a moss (Bryum sp.) and examined the ice recrystallization inhibition (RI) activities of the semipure materials. The semipure materials contain both protein and carbohydrate in which the carbohydrate accounted for 73, 52, and 37%, respectively, of the total carbohydrate + protein. The IASs had RI activity at concentrations of 1.4, 0.05, and 0.01 microg ml-1, respectively. RI activity was greatly reduced by heat treatment, suggesting that the IASs inhibit recrystallization through a specific interaction with ice. These results raise the possibility that the IASs increase freezing tolerance of their respective organisms by preventing the recrystallization of ice.  相似文献

The forest canopy is fundamentally important in biodiversity conservation and ecosystem function. Cryptogamic epiphytes are dominant tree bole and canopy elements in temperate and boreal forests, though remain neglected by mainstream forest ecology. This review makes ecological information on cryptogamic epiphytes available to a non-specialist audience, to facilitate their integration in forest biodiversity and ecosystem studies more generally. The review focuses specifically on lichen epiphytes, highlighting their diversity and ecosystem role. A principal task is to explore pattern and process in lichen epiphyte diversity – species composition and richness – therefore demonstrating the utility of lichens as an ecological model system. The review examines key themes in previous research. First, the extensive literature used to resolve species response to, and community turnover along environmental/resource gradients, consistent with the habitat niche. Second, the evidence for dispersal-limitation, which may constrain community composition and richness in isolated habitats. Third, these two processes – the habitat niche and dispersal-limitation – are used to explain stand-scale diversity, in addition to the role of neutral effects (habitat area). Fourth, the review moves from a taxonomic (pattern) to a functional (process) perspective, considering evidence for autogenic succession evidenced by competition and/or facilitation, and non-random trends in life-history traits. This functional approach provides a counter-point to an assumption that lichen epiphyte communities are unsaturated and non-competitive, a situation which would allow the long-term accumulation of species richness with temporal continuity. Finally, the review explores landscape-scale impacts on lichen epiphytes, with recommendations for conservation.  相似文献

Improving aspects of platelet cryopreservation would help ease logistical challenges and potentially expand the utility of frozen platelets. Current cryopreservation procedures damage platelets, which may be caused by ice recrystallization. We hypothesized that the addition of a small molecule ice recrystallization inhibitor (IRI) to platelets prior to freezing may reduce cryopreservation-induced damage and/or improve the logistics of freezing and storage. Platelets were frozen using standard conditions of 5–6% dimethyl sulfoxide (Me2SO) or with supplementation of an IRI, N-(2-fluorophenyl)-d-gluconamide (2FA), prior to storage at −80 °C. Alternatively, platelets were frozen with 5–6% Me2SO at −30 °C or with 3% Me2SO at −80 °C with or without 2FA supplementation. Supplementation of platelets with 2FA improved platelet recovery following storage under standard conditions (p = 0.0017) and with 3% Me2SO (p = 0.0461) but not at −30 °C (p = 0.0835). 2FA supplementation was protective for GPVI expression under standard conditions (p = 0.0011) and with 3% Me2SO (p = 0.0042). Markers of platelet activation, such as phosphatidylserine externalization and microparticle release, were increased following storage at −30 °C or with 3% Me2SO, and 2FA showed no protective effect. Platelet function remained similar regardless of 2FA, although functionality was reduced following storage at −30 °C or with 3% Me2SO compared to standard cryopreserved platelets. While the addition of 2FA to platelets provided a small level of protection for some quality parameters, it was unable to prevent alterations to the majority of in vitro parameters. Therefore, it is unlikely that ice recrystallization is the major cause of cryopreservation-induced damage.  相似文献

The extremely large number of leaves produced by terrestrial and aquatic plants provide habitats for colonization by a diversity of microorganisms. This review focuses on the bacterial component of leaf microbial communities, with emphasis on Pseudomonas syringae-a species that participates in leaf ecosystems as a pathogen, ice nucleus, and epiphyte. Among the diversity of bacteria that colonize leaves, none has received wider attention than P. syringae, as it gained notoriety for being the first recombinant organism (Ice(-) P. syringae) to be deliberately introduced into the environment. We focus on P. syringae to illustrate the attractiveness and somewhat unique opportunities provided by leaf ecosystems for addressing fundamental questions of microbial population dynamics and mechanisms of plant-bacterium interactions. Leaf ecosystems are dynamic and ephemeral. The physical environment surrounding phyllosphere microbes changes continuously with daily cycles in temperature, radiation, relative humidity, wind velocity, and leaf wetness. Slightly longer-term changes occur as weather systems pass. Seasonal climatic changes impose still a longer cycle. The physical and physiological characteristics of leaves change as they expand, mature, and senesce and as host phenology changes. Many of these factors influence the development of populations of P. syringae upon populations of leaves. P. syringae was first studied for its ability to cause disease on plants. However, disease causation is but one aspect of its life strategy. The bacterium can be found in association with healthy leaves, growing and surviving for many generations on the surfaces of leaves as an epiphyte. A number of genes and traits have been identified that contribute to the fitness of P. syringae in the phyllosphere. While still in their infancy, such research efforts demonstrate that the P. syringae-leaf ecosystem is a particularly attractive system with which to bridge the gap between what is known about the molecular biology of genes linked to pathogenicity and the ecology and epidemiology of associated diseases as they occur in natural settings, the field.  相似文献

In polar seas, ice algal communities can acclimate to extremely low light conditions. Reduced acclimation to shade in ice algal communities, as a result of shortened ice seasons at the lower latitude limits of sea ice distribution, has been suggested as advantageous for avoiding strong photoinhibition when cells are released into high light levels at the water’s surface. Thermal dissipation of excess energy by xanthophyll cycle pigments in the de-epoxidated state may occur in ice algal communities released from retreating sea ice. A light exposure experiment was conducted on ice algal communities obtained from sea ice at Saroma-Ko Lagoon in Hokkaido, Japan. Photoprotective responses to direct sunlight were examined through non-photochemical quenching (NPQ) of chlorophyll fluorescence and xanthophyll pigments. De-epoxidation of diadinoxanthin (DD) to diatoxanthin (DT) occurred rapidly, and NPQ showed a dynamic response to high light exposure. The linear relationship between the ratio of DT to chlorophyll a and NPQ followed a steeper slope than previously observed for mesophilic diatoms. The steeper slope could be explained by an apparent increase in DT for the mesophilic diatoms and induction of NPQ in response to low temperatures only in the ice algal communities. Enhanced production of DT in mesophilic diatoms could be the result of de-epoxidation of DD plus de novo synthesis, and the enhancement of NPQ might be caused by low temperature stress in the ice algae. Although the response of NPQ might be related to temperature, NPQ independent of DT synthesis should also be studied.  相似文献

In a recent article published in Cryobiology, Seki and Mazur [9] performed kinetic analysis to investigate the physicochemical mechanism of the intracellular ice formation in mouse oocytes subjected to rapid cooling. Based on their results, the authors calculated the activation energy for the ice recrystallization process to be 27.5 kcal/mol. In this letter, we report our analysis of the result in terms of the transition-state theory to show that the process is unfavorable in terms of enthalpy but favorable in terms of entropy accompanying molecular expansions. This report is expected to evoke interests in applying thermodynamics to the investigation of the intracellular ice formation.  相似文献

The ice recrystallization inhibition activity of various mono- and disaccharides has been correlated with their ability to cryopreserve human cell lines at various concentrations. Cell viabilities after cryopreservation were compared with control experiments where cells were cryopreserved with dimethylsulfoxide (DMSO). The most potent inhibitors of ice recrystallization were 220?mM solutions of disaccharides; however, the best cell viability was obtained when a 200?mM d-galactose solution was utilized. This solution was minimally cytotoxic at physiological temperature and effectively preserved cells during freeze-thaw. In fact, this carbohydrate was just as effective as a 5% DMSO solution. Further studies indicated that the cryoprotective benefit of d-galactose was a result of its internalization and its ability to mitigate osmotic stress, prevent intracellular ice formation and/or inhibit ice recrystallization. This study supports the hypothesis that the ability of a cryoprotectant to inhibit ice recrystallization is an important property to enhance cell viability post-freeze-thaw. This cryoprotective benefit is observed in three different human cell lines. Furthermore, we demonstrated that the ability of a potential cryoprotectant to inhibit ice recrystallation may be used as a predictor of its ability to preserve cells at subzero temperatures.  相似文献

Vascular epiphytes contribute to the structural, compositional, and functional complexity of tropical montane cloud forests because of their high biomass, diversity, and ability to intercept and retain water and nutrients from atmospheric sources. However, human-caused climate change and forest-to-pasture conversion are rapidly altering tropical montane cloud forests. Epiphyte communities may be particularly vulnerable to these changes because of their dependence on direct atmospheric inputs and host trees for survival. In Monteverde, Costa Rica, we measured vascular epiphyte biomass, community composition, and richness at two spatial scales: (1) along an elevation gradient spanning premontane forests to montane cloud forests and (2) within trees along branches from inner to outer crown positions. We also compared epiphyte biomass and distribution at these scales between two different land-cover types, comparing trees in closed canopy forest to isolated trees in pastures. An ordination of epiphyte communities at the level of trees grouped forested sites above versus below the cloud base, and separated forest versus pasture trees. Species richness increased with increasing elevation and decreased from inner to outer branch positions. Although richness did not differ between land-cover types, there were significant differences in community composition. The variability in epiphyte community organization between the two spatial scales and between land-cover types underscores the potential complexity of epiphyte responses to climate and land-cover changes.  相似文献

The role of terrestrial soil nutrient supply in determining the composition and productivity of epiphyte communities has been little investigated. In a montane Hawaiian rainforest, we documented dramatic increases in the abundance and species richness of canopy epiphytes in a forest that had been fertilized annually with phosphorus (P) for 15 years; there was no response in forest that had been fertilized with nitrogen (N) or other nutrients. The response of N-fixing lichens to P fertilization was particularly strong, although mosses and non-N-fixing lichens also increased in abundance and diversity. We show that enhancement of canopy P availability is the most likely factor driving the bloom in epiphytes. These results provide strong evidence that terrestrial soil fertility may structure epiphyte communities, and in particular that the abundance of N-fixing lichens – a functionally important epiphyte group – may be particularly sensitive to ecosystem P availability.  相似文献

Epiphytes in tree canopies make a considerable contribution to the species diversity, aboveground biomass, and nutrient pools in forest ecosystems. However, the nutrient status of epiphytes and their possible adaptations to nutrient deficiencies in the forest canopy remain unclear. Therefore, we analyzed the stoichiometry of five macroelements (C, N, P, K, and Ca) in four taxonomic groups (lichens, bryophytes, ferns, and spermatophytes) to investigate this issue in a subtropical montane moist evergreen broad‐leaved forest in Southwest China. We found that the interspecific variations in element concentrations and mass ratios were generally greater than the intraspecific variations. And there were significant stoichiometric differences among functional groups. Allometric relationships between N and P across the epiphyte community indicated that P might be in greater demand than N with an increase in nutrients. Although canopy nutrients were deficient, most epiphytes could still maintain high N and P concentrations and low N:P ratios. Moreover, ferns and spermatophytes allocated more limited nutrients to leaves than to stems and roots. To alleviate frequent drought stress in the forest canopy, vascular epiphytes maintained several times higher K concentrations in their leaves than in the tissues of lichens and bryophytes. Our results suggest that epiphytes may have evolved specific nutrient characteristics and adaptations, so that they can distribute in heterogeneous canopy habitats and maintain the stability of nutrient metabolism.  相似文献

SUMMARY. 1. We examined the abundance and oxygen metabolism of epiphytic organisms on the dominant macrophyte, Potamogeton pectinatus , in headwaters of the eutrophic River Suså. Microbenthic algae were abundant in the stream during spring and macrophytes during summer.
2. The low macrophyte biomass in spring supported a dense epiphyte cover whereas the high macrophyte biomass during summer had a thin epiphyte cover of 10–100-fold lower abundance per unit area of macrophyte surface. The epiphyte community was dominated by microalgae in spring and by heterotrophs, probably bacteria, during summer. This seasonal shift was shown by pronounced reductions of the chlorophyll a content (from 2–3% to 0.1–0.7% of organic DW), the gross photosynthetic rate (from 20–85 to 3–15 mg O2, g-1 organic DW h−1) and the ratio of gross photosynthesis to dark respiration in the epiphyte community (from 5–18 to 1). The reduced contributions of epiphytic microalgae correlated with reduced light availability during summer.
3. Both the density and the photosynthetic activity of epiphytic algae were low on a stream area basis relative to those of microbenthic algae and macrophytes. Rapid variations in water velocity and extensive light attenuation in water and macrophyte stands probably constrained the development of epiphytic algae. The epiphyte community was more important in overall stream respiration, contributing c. 10% to total summer respiration and c. 20% to summer respiration within the predominantly heterotrophic microbial communities on sediments and macrophyte surfaces.  相似文献
