Treatment of soil with surfactants and chelating agents is used in bioremediation studies to desorb and solubilize contaminants to increase their bioavalability to microorganisms. In the same way that pollutants are made more bioavailable to microorganisms, the procedure can be used to remove potential interfering materials from soil prior to cell lysis and extraction of DNA from indigenous microorganisms. The effect of soil washing was evaluated by extracting DNA from sediments of an intertidal freshwater wetland contaminated with hydrocarbons and from highly contaminated marine sediments from Sydney Harbour, Nova Scotia, Canada. Sediment samples had total organic carbon (TOC) contents that varied between 0.2% and 13%. The chemical lysis technique was also examined by comparison of an ammonium acetate precipitation of proteins and humic acids with a hexadecyltrimethylammonium bromide (CTAB) incubation and phenol:chloroform extraction. In this study, the incorporation of soil washing steps facilitated the desorption of contaminants from sediment surfaces and improved the recovery of DNA of amplifiable quality from both freshwater and marine sediments. CTAB contributed only slightly to the recovery of DNA of higher quality in the most contaminated sample from Sydney Harbour and was concomitant with a decrease in DNA yield in both sediment types. The incorporation of a soil washing step prior to the extraction of DNA from polluted environments may be important to solubilize and remove contaminants when high-quality DNA is required for subsequent analyses.  相似文献

Using sodium dodecyl sulfate--polyacrylamide gel electrophoresis and autoradiography, we have shown that 125I-labeled cholera toxin binds to Newcastle disease virus. Pretreatment of Newcastle disease virus with “cold” cholera toxin (at 37°C for 30 minutes) inhibits the binding of 125I-labeled toxin in a subsequent incubation (at 37°C for 30 minutes). These results suggest that cholera toxin binds to Newcastle disease virus in a specific manner. The precise receptor for toxin is unknown in Newcastle disease virus but it is presumed to be the ganglioside GM1. We have previously shown that the photoreactive probe 12-(4-azido-2-nitrophenoxy)stearoylgucosamine[1-14C] labels the membrane proteins of Newcastle disease virus. Since the reactive group of the probe, ie, N3, resides within the membrane bilayer, studies were initiated to determine which, if any, of the subunits of cholera toxin cross the membrane of Newcastle disease virus and become radioactively labeled upon photoactivation of the probe at 360 nm. After a 15-minute incubation of cholera toxin with Newcastle disease virus containing the photoreactive probe, irradiation effected the 14C-labeling of the active A1 subunit of cholera toxin. Irradiation of cholera toxin in solution with an equivalent amount of probe but without virus resulted in no labeling of toxin subunits.  相似文献

Semen and embryos generated by assisted reproductive techniques (ARTs) may be contaminated with numerous microorganisms. Contamination may arise from systemic or local reproductive tract infections in donors or the inadvertent introduction of microorganisms during ARTs, and may lead to disease transmission. This review describes sanitary procedures which have been investigated to ascertain whether they are effective in rendering semen and embryos free of pathogenic microorganisms, including internationally adopted washing procedures, which can be supplemented by antibiotics and enzymatic treatments. Other methods include treatment with antibodies or ozone, photoinactivation, acidification, and the use of novel antiviral compounds. In conclusion, despite the wide range of antimicrobial procedures available, none can be recommended as a universal disinfection method for rendering semen and embryos free from all potentially pathogenic microorganisms. However, some procedures are unsuitable, as they can compromise the viability of semen or embryos. In humans, washing by the gradient centrifugation method appears to be effective for reducing the microbial population in semen and is harmless to the spermatozoa. A useful procedure for embryos involving multiple washes in sterile medium has much to commend it for the prevention of disease transmission; furthermore, it is recommended by the International Embryo Transfer Society (IETS).  相似文献

Abstract— The interaction of 125I-labelled tetanus toxin with cells in tissue cultures derived from embryonic CNS has been studied.
The optimum toxin binding occurs about 2–3 weeks after transfer of the cells to culture conditions. The amount of label bound per culture was doubled at this time in comparison to the fourth day nfter inoculation.
The amount of toxin bound depends on the concentration applied. It reaches its maximum 8 h after application then decreases slowly. Low amounts of radioactivity were still detectable 97 h after washing off the unbound toxin. Up to 80% of the label can be replaced by simultaneous application of'cold'toxin. Fixation of the toxin is higher at 4°C than at 37°C.
Preincubation of the cultures with neuraminidase prevents about 75% of the binding. The presence of cytochalasin B leads to a small but reproducible decrease of binding, whereas colchicine had no measurable effect.
The radioactive (1251) material was identified by a double-isotope technique in disc gel electrophoresis before and after reductive cleavage of its disulphide bonds. In every test is was indistinguishable from 131I-labelled toxin added as standard.
Our results largely parallel those obtained with synaptosomes and other systems. They suggest that gangliosides might be the acceptor molecules, and that the culture system will be suitable for studying the actions of this toxin in vitro.  相似文献

If Pieris brassicae eggs contaminated with granulosis virus 1 in the form of intact inclusion bodies are exposed to formaldehyde gas under suitable conditions, the virus can be inactivated without preventing the eggs from hatching. The formaldehyde may be derived from formalin solutions, but it is more convenient to enclose the 1-day-old eggs with the diet containing formaldehyde on which the young larvae are intended to feed and to open the jar the day before they hatch. This involves less handling than washing the eggs. As would be expected, transovarially transmitted infection is not reduced.  相似文献

In this study, we identified and quantitated the lipid components in the secretions of human bronchial explants cultured in a serum-free medium over a period of 50 days. Total lipids represented 6% of the dry material. This amount is considerably lower than that reported for 'normal' human sputum and suggests that the latter is contaminated by serum transudates, alveolar secretions, and microorganisms. Such serum-free culture systems are highly suitable to study cell physiology as it relates to human disease.  相似文献

In an experiment with infectious bovine rhinotracheitis (IBR) virus in two bulls, observed over a period of 122 weeks, the pattern of virus release was studied. Recurrent, unprovoked release of virus was demonstrated after one year in a nasal washing from one of the bulls and in preputial washings of both on 13 and 4 occasions, respectively, and finally in weeks 113 and 110, although clinical disease was not observed. During periods of recurrent virus release, concentrations of virus in the prepuce were generally much lower than during the period of primary infection; usually, however, they were not of negligible titer. The frequency of such periods and the virus titers observed strongly suggest that an IBR antibody carrier should always be considered as a potential source of infection to other animals. When virus was demonstrated in semen an almost equal amount was found in the preputial washing (50 ml). In week 120, virus replication in the respiratory tract and prepuce was induced in both bulls by prednisolone injections. It is concluded that antibody carriers will rarely attain a state of absolute immunity.  相似文献

In the present study, the risk of transmission of mouse minute virus (MMV) to recipients of murine embryos arising from in vitro fertilization (IVF) of oocytes with MMV-exposed spermatozoa and to resulting pups was evaluated. Also, the time of seroconversion of recipients and pups was investigated. To achieve this goal, IVF of oocytes with cryopreserved spermatozoa from the inbred C3HeB/FeJ mouse strain was performed, and the resulting embryos were transferred to suitable Swiss recipients. Three groups were investigated: 1) oocytes or the developing embryos were continuously exposed to 10(4) TCID(50) MMVp per milliliter in the fertilization (human tubal fluid [HTF]), culture (KSOM), and embryo transfer (M2) media (positive control); 2) oocytes and spermatozoa were exposed to MMVp in the HTF medium only and transferred after a standard washing procedure with 10 washing steps in virus-free KSOM and M2; and 3) oocytes and spermatozoa were exposed to virus-free HTF, KSOM, and M2 (negative control). To detect antibodies to MMV in recipients and progeny, serological analyses were performed by ELISA on Days 14, 21, 28, and 42, and on Days 42 and 63, respectively, after embryo transfer. The presence of MMV in the washing drops was analyzed by PCR and an in vitro infectivity assay, while organs of some recipients and pups were analyzed by PCR. Using 10(4) of the tissue culture infective dose of MMVp per millilitre in the fertilization medium only, the present results demonstrate that 10 washing steps in the IVF-ET procedure are sufficient to remove the virus to a noninfectious dose, producing MMV-free seronegative recipients and pups. As such, there is minimal risk of transmission of MMV to recipients and pups if spermatozoa become contaminated with such viral loads.  相似文献

Members of the Bacillus genus are ubiquitous soil microorganisms and are generally considered harmless contaminants. However, a few species are known toxin producers, including the foodborne pathogen, B. cereus. This species produces two distinct types of foodborne illness, the emetic (vomit-inducing) syndrome, associated with consumption of toxin in cooked rice dishes, and the diarrheal illness seen occasionally following consumption of contaminated meats, sauces, and certain dairy products. In the latter case, illness results from the production of enterotoxins by vegetative cells in the small intestine of the host. In dairy products, the occurrence of Bacillus spp. is inevitable, and the spore-forming ability of this organism allows it to easily survive pasteurization. Many strains have been shown to grow and produce enterotoxin in dairy products at refrigeration temperatures. Evaluation of toxin gene presence and toxin expression in Bacillus spp. other than B. cereus has not been thoroughly investigated. However, the presence of natural isolates of Bacillus spp. harboring one or more enterotoxin gene(s) and subsequent demonstration of conditions which may support toxin expression holds crucial importance in the food safety arena.  相似文献

When zona pellucida-intact porcine embryos were exposed to 10(7) plaque-forming units (pfu)/ml of swine vesicular disease virus (SVDV) and then washed, infectious virus could be isolated from all of the embryos. Culturing the embryos for 24 or 48 h or treating the embryos with pronase, trypsin, or antiserum after virus exposure and washing reduced the number of embryos carrying virus and lessened the amount of virus on each of the embryos. 研究了斜纹夜蛾幼虫体内的斜纹夜蛾侧沟茧蜂存活率、发育历期、寄主感染病毒时间、病毒浓度之间的关系,并测定了斜纹夜蛾侧沟茧蜂的传毒效率.结果表明,病毒对寄主体内寄生蜂历期无明显影响,寄生在幼虫体内的寄生蜂能在寄主病死前完成发育,存活比例因寄主感染病毒的时间和浓度而异.斜纹夜蛾被寄生后接种病毒(SINPV),距离寄生时间越长,饲毒浓度越低,寄生蜂完成发育的比例越大,但饲毒时间是主要影响因素.从感病幼虫体内发育成的侧沟茧蜂或曾经在感病寄主上产过卵的寄生蜂,以及通过人工方式使产卵器被病毒污染的寄生蜂,均能携带一定数量的病毒.通过产卵活动,侧沟茧蜂成蜂能在寄主幼虫个体间传递病毒.当寄生蜂在感病的寄主幼虫上产卵带毒后,平均可传递病毒给2.14头幼虫;发育于感病幼虫体内的寄生蜂,平均可传递病毒给2.45头幼虫.通过用病毒液浸茧或用混有病毒的蜂蜜饲喂成蜂等方式使产卵器污染病毒的寄生蜂,传毒效率随饲毒浓度增加而提高,平均可传递病毒1.45头和0.94头幼虫  相似文献

The Biosafety Committee of the Japanese Association of Laboratory Animal Facilities of National Universities (JALAN) investigated recent episodes of microbiological contamination in genetically modified mice (GMM), and the countermeasures taken when the contaminated GMM were introduced into animal facilities, by questionnaires addressed to 53 animal facilities belonging to JALAN and serological tests. Although almost all of the contaminated GMM were accepted with conditions such as rederivation after or before reception and housing in designated rooms, contamination with a spectrum of microorganisms was demonstrated in GMM transferred domestically and from abroad. In serological tests, Mycoplasma pulmonis, mouse parvovirus, and mouse encephalomylitis virus were detected in GMM transferred from domestic facilities and from abroad. The present results of the questionnaires and serological tests suggest that GMM are highly and widely contaminated with microorganisms compared with mice from commercial breeders. Thus, we propose a microbiological requirement, including microbiological status--excellent, common, and minimum--as a guide for the transfer and procurement of mice and rats in Japan.  相似文献

The growth and virulence of the bacteria Bacillus thuringiensis depend on the production of Cry toxins, which are used to perforate the gut of its host. Successful invasion of the host relies on producing a threshold amount of toxin, after which there is no benefit from producing more toxin. Consequently, the production of Cry toxin appears to be a different type of social problem compared with the public goods scenarios that bacteria usually encounter. We show that selection for toxin production is a volunteer's dilemma. We make specific predictions that (a) selection for toxin production depends upon an interplay between the number of bacterial cells that each host ingests and the genetic relatedness between those cells; (b) cheats that do not produce toxin gain an advantage when at low frequencies, and at high bacterial density, allowing them to be maintained in a population alongside toxin‐producing cells. More generally, our results emphasize the diversity of the social games that bacteria play.  相似文献

Using the ecological and natural-science approaches, the authors have come to the conclusion that microorganisms, pathogenic for humans (animals), are their parasites for whom the disease of their biological host is the necessary condition of their existence as a biological species. And accordingly, microorganisms, opportunistic in humans (animals) are their parasites and commensals, as well as saprophytes, for whom the disease of their host is not the necessary condition of their existence in nature. The biological host is a symbion necessary for the existence of pathogenic and most opportunistic microorganisms, but for a pathogenic microorganism the disease of the host is the result of symbiotic relationships, while for an opportunistic microorganism the disease of the host is the consequence of disturbances in symbiotic relationships. Such view of pathogenicity is important for creating a scientifically grounded theory of the liquidation of human infectious diseases.  相似文献

Bulk-soap-refillable dispensers are prone to extrinsic bacterial contamination, and recent studies demonstrated that approximately one in four dispensers in public restrooms are contaminated. The purpose of this study was to quantify bacterial hand contamination and transfer after use of contaminated soap under controlled laboratory and in-use conditions in a community setting. Under laboratory conditions using liquid soap experimentally contaminated with 7.51 log(10) CFU/ml of Serratia marcescens, an average of 5.28 log(10) CFU remained on each hand after washing, and 2.23 log(10) CFU was transferred to an agar surface. In an elementary-school-based field study, Gram-negative bacteria on the hands of students and staff increased by 1.42 log(10) CFU per hand (26-fold) after washing with soap from contaminated bulk-soap-refillable dispensers. In contrast, washing with soap from dispensers with sealed refills significantly reduced bacteria on hands by 0.30 log(10) CFU per hand (2-fold). Additionally, the mean number of Gram-negative bacteria transferred to surfaces after washing with soap from dispensers with sealed-soap refills (0.06 log(10) CFU) was significantly lower than the mean number after washing with contaminated bulk-soap-refillable dispensers (0.74 log(10) CFU; P < 0.01). Finally, significantly higher levels of Gram-negative bacteria were recovered from students (2.82 log(10) CFU per hand) than were recovered from staff (2.22 log(10) CFU per hand) after washing with contaminated bulk soap (P < 0.01). These results demonstrate that washing with contaminated soap from bulk-soap-refillable dispensers can increase the number of opportunistic pathogens on the hands and may play a role in the transmission of bacteria in public settings.  相似文献

The soil contaminated by explosive production wastewater was treated by washing using water as solvent. The effect of contact time and temperature, water/soil ratio and washing steps on desorption efficiency was investigated. Six kinetic models—parabolic diffusion model, zero-order equation, pseudo-first-order equation, pseudo-second-order equation, power function equation and Elovich equation—were used to study the desorption kinetics of nitroaromatic compounds from contaminated soil to water. The eluent of contaminated soil before and after washing was characterized by UV–vis analysis. The results showed that the removal rate was fast at the initial stage and then slowed down after 60 min. The desorption of contaminants from soil to water is endothermic. Washing with small quantities of water in high frequency is preferred when water volume is limited. The pseudo-second-order model can be used to describe the desorption process. Soil washing can remove most of the contaminants from the contaminated soil.  相似文献

In this study, the soil washing technique has been used to treat mine tailings contaminated heavily with arsenic and heavy metals at Jingok mine, which is one of the abandoned mines in Korea. The results showed that phosphoric acid, citric acid, oxalic acid, and sodium metabisulfite were highly effective in extracting arsenic and heavy metals. Among them, oxalic acid was the most effective (especially for Pb, Cu, and Zn), as even a residual fraction of arsenic was partly extracted. The optimum concentration of washing reagent and the ratio between the mine tailings and washing reagent were found to be 0.5 M and 1:20, respectively. In addition, the extraction kinetics of arsenic and heavy metals was fast, in which the reaction time of 30 minutes was deemed to be a sufficient contact time. From the results, it may be concluded that the low pH of washing solution and the amount of dissolved Fe may be considered as the most important factors in the extraction of arsenic and heavy metals.  相似文献
