The host response to 3 different larval digeneans affecting Southwestern Atlantic and Magellanic populations of the bivalve Gaimardia trapesina (Lamarck, 1819) (Gaimardiidae) is described. Unencysted metacercariae of 2 species of Gymnophallidae and 1 species of Lepocreadiidae co-exist in the peripheral and general extrapallial spaces. Differences in host responses to stimuli generated by each parasite are described. Infections by Gymnophallidae gen. sp. 1 metacercariae elicit a mantle-shell complex reaction involving both alteration of the mantle epithelium (hyperplasia and metaplasia) and calcium carbonate deposition to form an incomplete calcareous covering of single larvae, comprising individual crystallites in an organic matrix. The calcareous covering covers only the posterior two-thirds of the larval body, having its anterior end free in the extrapallial space, which ensures survival of the larvae. The peculiar features of the host response result from a successful process of adjustment to the parasite's life cycle. Metacercariae of Gymnophallidae gen. sp. 2 occur in one or more small groups of up to 30 individuals, either in the general or in the peripheral extrapallial spaces. The outer mantle reaction includes hyperplasia and metaplasia of the epithelium adjacent to the larvae. Deposition of calcium carbonate did not occur. Lepocreadiidae gen. sp. metacercariae were observed exclusively in the general extrapallial space lodged in single shallow pits whose shape fit the parasite's body shape and size. In simultaneous infections, metacercariae of Gymnophallidae and Lepocreadiidae elicited different responses in an individual, which suggests that each parasitic entity involves a different stimulus to the host.  相似文献

The mortality of Aedes aegypti pre-imagos harboring metacercariae of Plagiorchis noblei Park, 1936, is governed by the stage of development of the host at the time of infection and the location of the parasite in the insect body. First and second instar larvae generally succumbed to infection, regardless of site. Infections of the head and thorax of third and fourth instar larvae were generally lethal or gave rise to imparied adults. However, older instars frequently survived abdominal infections. Pupae showed greater tolerance to cephalic, thoracic and abdominal infections and generally emerged as adults. Again, many such infected adults were impaired.  相似文献

A total of 4,298 snails Bradybaena similaris were collected from the rural area of Rio de Janeiro County and from the Counties of Paracambi and Itaguai, State of Rio de Janeiro. Two thousand and five of these snails had metacercariae in their pericardial cavity. The collecting was done between October 1983 and September 1986. The annual average prevalences were: 42.89% (1st year), 43.42% (2nd year) and 55.86% (3rd year). Experimental infection in chicks allowed the identification of these metacercariae as Postharmostomum gallinum. The small variation in size observed among metacercariae from the same host indicates that recruitment does not occur on a continuous basis. Statistical analysis showed no correlation between the snail size and the number of metacercariae in the pericardical cavity, and that the snail mortality observed in the laboratory was due to the parasitic infection.  相似文献

The subject of the following study was the natural and experimental invasion of trematode larvae in Potamopyrgus antipodarum from Bory Tucholskie National Park (Poland). Only one out of the 14,908 dissected specimens had oval sporocysts and mature cercariae of fish fluke, which belongs to the Sanguinicolidae family. It is the first recorded case in the European population of P. antipodarum living in inland water. The experimental study showed the possibility of native metacercariae (Echinostoma revolutum, Echinoparyphium aconiatum and Hypoderaeum conoideum) settlement in those immigrant snail species; however, exposure to parasites resulted in an increase in snail mortality. The three out of six used cercariae species were able to transform into metacercariae in P. antipodarum as in the second intermediate host, but the exposure to parasitic larvae of four of the used species resulted in an increase in snails’ mortality. It may suggest that not only metacercariae settlement but also the attack of cercariae (Rubenstrema opisthovitellinum at a temperature of 22 °C) affected the low survival of experimental snails in comparison to control animals. The subject of discussion presented in this paper is also the hypothesis on probable effect of the interaction between P. antipodarum and native snail species (as a source of invasive larvae of parasites) living in the same habitat.  相似文献

Histopathological, ultrastructural and immunohistochemical investigations were conducted on 26 specimens of powan Coregonus lavaretus (L.) from Loch Lomond (Scotland). The hearts of all 26 powan (15 females and 11 males) investigated harboured metacercariae of the digenean trematode Ichthyocotylurus erraticus (Rudolphi, 1809). The vast majority of metacercariae were located either singly or as an aggregation of white cysts on the surface of the bulbus arteriosus. The intensity of infection ranged from 2 to 200 larvae heart(-1), although the number of metacercariae found on male powan did not exceed 13. Histochemically, the parasite cyst wall gave a strong positive reaction with periodic acid schiff (PAS) and a faint positive signal with Azan-Mallory stain. All the metacercariae cysts were embedded in a granulomatous proliferation of heart epicardium tissue, forming a reactive fibroconnective capsule around the parasite. The capsule enclosing the parasite (produced by the host's reaction to the parasite) measured 13.57 to 90.20 microm (37.43 +/- 3.56) in thickness. Within the capsule wall, eosinophilic granular cells (EGCs), granulocytes, melanocytes and, in some instances, partially degenerated or vacuolated epithelioid cells were observed in close proximity to the cyst wall. Pigment-bearing macrophages were scattered throughout the granulomatous host-tissue reaction and as macrophage aggregates (MAs) within the capsules surrounding parasites. Immunohistochemical tests were applied to infected heart sections using 12 different antisera. Nerve fibres immunoreactive to bombesin, substance P (SP), and atrial natriuretic peptide (ANP) antisera were observed in close proximity to the parasite larvae. The presence of a serotonin-like substance was also observed within host immune-cells surrounding trematode cysts. Large cells of the epicardium were found to be immunoreactive to met-enkephalin and vasoactive intestinal peptide (VIP) antisera but not immunoreactive to anti-protein gene-product 9.5 (PGP9.5) sera.  相似文献

This study was performed to observe the infection status of sweetfish, caught from Kwangjung-stream and Namdae-stream in Yangyang-gun, Kangwon-do in August 1989, with the metacercariae of Metagonimus yokogawai. All of 28 sweetfish from Kwangjung-stream were infected with the metacercariae of M. yokogawai. The number of metacercariae in a fish ranged 89-521(224 on average), and the number of larvae/g of flesh was 12 approximately 55 (22 on average). On the other hand, no metacercariae were detected in the fish from Namdae-stream. It has been confirmed that the Kwangjung-stream in Yangyang-gun, Kangwon-do is one of the endemic foci of metagonimiasis in Korea.  相似文献

Patterns of parasite load and aggregation of the bird trematode Cardiocephaloides longicollis in its main intermediate host in the Mediterranean, the annular sea bream, Diplodus annularis, were studied in a large sample collected off Valencia (Spain) and are discussed within the context of the parasite induced host mortality hypothesis. The metacercariae were located within large composite cysts of host origin in the ventricles of the optic lobes of the cerebrum. A weak immunological response was detected in older fish, which was significantly associated with the total parasite load. Although the mean abundance of C. longicollis showed a tendency to increase with host size, the infection levels were generally homogeneous with a noticeable plateauing in the intermediate size classes. The distribution of the metacercariae was aggregated and agreed with the negative binomial distribution. There was a marked decline in parasite aggregation in the largest size-class, suggesting parasite-induced host mortality in the oldest fish possibly due to predation by large non-host fish predators. On the other hand, levelling off of abundance and decrease in heterogeneity of parasite distribution within the intermediate age cohort could indicate that these sizes are being rapidly and/or constantly removed from the host population due to by-catch fishing. The overall high infection levels and the continuous recruitment across age cohorts provides evidence that an enhanced parasite transmission is taking place in the Gulf of Valencia due to increased spatial overlap of the hosts involved in the life cycle. We suggest a human-induced facilitation of the digenean life cycle due to the fact that gulls in the area feed extensively on discards, thus indicating the possibility of an unforeseen effect of fishing practices in a marine littoral system.  相似文献

The average quantity of metacercariae in sporocysts is influenced by host species, its syze and by the season. In vitro excretory/secretory products of 4-week (immature) and 8-week-old (mature) Fasciola hepatica parasites, derived from rats, were injected together with adjuvant into naive rats and mice. Resistance to infection was assessed in rats by counting adults in the bile ducts at 9 weeks, or in mice by recording deaths after oral challenge with a high dose of viable metacercariae. Exposure of rats to excretory/secretory products of immature F. hepatica conferred a significant degree of resistance which was comparable to the level of resistance induced following oral administration of a low number of metacercariae. No protection against infection was seen in rats injected with excretory/secretory products from mature, bile duct-derived worms. In mice, no obvious mouse strain variation in susceptibility to first infection existed and hypothymic nude mice were as susceptible to infection as intact mice. As determined by protection against death, vaccination with excretory/secretory products derived from immature F. hepatica was without effect in mice. It is concluded that "host protective antigens", at least for rats, were present in the excretory/secretory products of immature F. hepatica larvae.  相似文献

Cercaria sevillana is the cercaria larval stage that infects the gonads and the digestive gland of its first intermediate host, Nassarius reticulatus. In this study the decapodous crustacean Carcinus maenas was used to determine if it would serve as second intermediate host in the parasite's life cycle. The latter hypothesis was based on the knowledge that C. maenas is the second intermediate host of several other digenean species. After dissection, it was possible to observe encysted metacercariae in the antennal glands of the green crab. After biochemical excystment, the metacercariae were processed for light and scanning electron microscopy. The morphological features observed led us to conclude that this species was a microphallid fluke, and it was identified as Gynaecotyla longiintestinata. To establish a possible relationship between C. sevillana and this metacercariae, the ITS1 region was analyzed. Thus, DNA was extracted from C. sevillana and from the cysts isolated from the antennal glands. The ITS1 region was amplified and sequenced, and the alignment clearly demonstrated that the cercaria and the metacercariae belonged to the same species, G. longiintestinata.  相似文献

The metacercaria of Posthodiplostomum minimum centrarchi Hoffman, 1958, is reported from an unusual host, Roccus saxatilis, from North Carolina. Although few dead or moribund fish were observed, there were many deformed fish, few of which survived. The genetic influence on host susceptibility to Neodiplostomum seoulense infection and fatality of the host was studied in 3 inbred strains of mice (BALB/c [H-2d], C3H/He [H-2k], and C57BL/6 [H-2b]). The survival of the mice, worm expulsion kinetics, worm size, number of eggs produced per day (EPD), and number of uterine eggs were observed from day 1 to day 40 postinfection (PI) with 100 or 200 metacercariae per mouse. Infection with N. seoulense was highly lethal to all 3 strains of mice, but the lethality was dose-dependent and varied according to the genetic backgrounds of the mice. The C3H/He mice exhibited the highest mortality, the lowest worm burdens and EPD, and the quickest expulsion of worms. It is suggested that different genetic backgrounds of mice appear to affect the host's capacity to expel N. seoulense and the fatality of the hosts themselves.  相似文献

The prevalence and intensity of experimental infections of Aedes aegypti with the digenean Plagiorchis noblei increased significantly with the level of trickle exposure to cercariae. Daily exposure to doses of 16 cercariae/day yielded a mean infection intensity of 13.0 metacercariae; doses of 1 cercaria/day resulted in only 2.4 metacercariae per infected mosquito larva. The prevalence of infection rose from 46% at an exposure of 1 cercaria/day to 99% at 16 cercariae/day. Host mortality rose concomitantly from 25% to 88%. Host mortality and parasite acquisition were independent of environmental temperatures (21-29 C), despite the fact that developmental times, and consequently the number of daily exposures, were more than 50% greater at the low end of the temperature scale. This may be attributable to low activity of mosquito larvae and the resulting decrease in the number of encounters with cercariae.  相似文献

The spatial distribution and histopathological changes induced by metacercariae of the digenean trematode Diplostomum phoxini (Faust, 1918) in the brains of European minnows Phoxinus phoxinus (L.) from the River Endrick, Scotland, were studied by light and electron microscopy. Post-mortem examination of a sample of 34 minnows revealed that 50% (n = 17) of the population was infected with 13.7 +/- 2.6 (mean +/- SE; range 1 to 38) metacercariae per infected host. Serial histological sections of the infected minnow brains revealed that the metacercariae were unevenly distributed throughout the brain, with aggregations occurring in the cerebellum, the medulla oblongata and the optic lobes. In fish with highest intensities of infection, over 40% of the cerebellar area and about 30% of the medulla oblongata area were occupied by larvae. Metacercariae disrupt the integrity of brain tissue, with individuals being found in small pockets surrounded by cellular debris. Metacercariae were rarely encountered on the surface of the brain. Electron microscopic examination of infection sites revealed that the granular layer surrounding metacercariae was necrotic, exhibited nuclear degradation and was marked by vacuolation of the cytoplasm. Rodlet cells, the only inflammatory cell types recorded in this study, were found only in parasitized brains and in close proximity to the teguments of metacercariae. It is hypothesised that secretions released from the teguments of metacercariae are a counter response to protect the metacercariae from the fish brain's cellular defence mechanisms.  相似文献

Intraspecific variability in parasite life cycle complexity (number of hosts and species of hosts in the life cycle) may have an impact how parasite genetic variation is partitioned among individual parasites, host individuals or host species within a given area. Among digenean trematodes, a three-host life cycle is common. However, a few species are precocious and may reach sexual maturity in what is typically regarded as the second intermediate host. The objective of this study was to determine whether a precocious life cycle predisposes digeneans to possible inbreeding or genetic subdivision among host species. As a study system, we used the digenean Proctoeces cf. lintoni whose metacercariae precociously mature (facultative) without a cyst wall in the gonads of multiple sympatric species of keyhole limpets (Fissurella spp.), typically regarded as the second intermediate hosts. Genotyped parasites were collected from four species of limpets and the clingfish Sicyases sanguineus, the third and final host where sexual maturity occurs. We found very high microsatellite diversity, Hardy–Weinberg equilibrium over all genotyped individuals, and little to no genetic structuring among parasites collected from the different host species. The fact that metacercariae do not encyst in the keyhole limpets, coupled with the high mixing potential of an aquatic environment, likely promote panmixia in local populations of P. cf. lintoni.  相似文献

A natural mass infection of heterophyid metacercariae in aquacultured Japanese eel Anguilla japonica in Taiwan was observed. Of the 28,000 adult eels in 2 ponds, about 25,000 (90%) showed swollen, cloudy and white eyes. Although morbidity was about 90%, there was no mortality among the affected eels. Histopathological sections showed edema and hemorrhage of the eye. Numerous metacercariae were observed in the muscle tissues around the eyeball, the subcutaneous tissue and even in the cartilage. Of the 6 eels digested with artificial gastric juice, all were found to contain metacercariae in their muscle tissues. The average number of metacercariae recovered from the 6 eels was 1219, with a range of 50 to 3762. These metacercariae, when fed orally to immunodeficient (scid) mice, developed into adult worms which were identified as Procerovum cheni Hsu 1950. The naturally infected eels were transferred to a new pond without snails and their eye lesions were not apparent anymore after 2 wk. In a follow-up investigation, 19 of 20 apparently healthy eels in a nearby aquaculture farm were found to harbour metacercariae in their muscles. However, the number of the metacercariae ranged from 1 to 14, with an average of 4.21. This is the first report of heterophyid metacercariae causing mass morbidity in aquacultured eels.  相似文献

The life cycle of Brachylaima ruminae n.sp. (Trematoda: Brachylaimidae), a duodenal parasite of rodents on the Mediterranean island of Formentera (Spain) is elucidated. The new species follows a terrestrial triheteroxenous life cycle. Eggs passed in the faeces of the definitive host must be ingested by a specific first intermediate host, the land snail Rumina decollata. Branched cercariogenous sporocysts develop in the digestive gland. Microcercous cercariae come out through the terminal birth pores of the branches. Cercariae shed by the snail are terrestrial, crawling on humid substratum. They contact the second intermediate host, another land snail, principally the species R. decollata and less frequently slugs and Helicids. Cercariae enter via the excretory pore and kidney duct to their specific final location, the kidney. Unencysted metacercariae develop in the kidney (also, less frequently, in the pedal glands) to the mature, infective stage. Infective metacercariae infest the definitive host when ingested together with the snail.  相似文献

Clutch size decisions by Aphaereta minuta (Nees) (Hymenoptera: Braconidae), a polyphagous, gregarious, larval-pupal endoparasitoid, were studied under laboratory conditions. This parasitoid attacks larvae of Diptera inhabiting ephemeral microhabitats such as decaying plant and animal material. Females oviposit in young larval stages, but the eventual size of the host pupa determines host food availability for competing offspring. The size of the pupa can differ greatly between host species. We questioned how A. minuta females deal with this delay between the moment of oviposition and eventual host food availability, and whether they make clutch size decisions that benefit their fitness. It was shown that females indeed vary their clutch size considerably and in an adaptive way: (1) females lay larger clutches in larvae of host species that produce larger pupae, even when the larvae are the same size at the moment of oviposition, and (2) females lay larger clutches in larger larvae than in smaller larvae of the same host species. The latter seems functional as larvae parasitized at an older stage indeed developed into larger pupae compared to larvae parasitized at a younger stage. Furthermore, mortality of parasitized young host larvae was greater than that of both unparasitized larvae and parasitized older larvae. Under field conditions the risk of mortality of young host larvae is expected to be even higher due to the limited period of microhabitat (host food) availability, strong scramble type competition between the host larvae, and the longer period of being exposed to predation.  相似文献

Thermal effects on the dynamics of infection with metacercariae of Diplostomum baeri and D. spathaceum were monitored between May and September, in a year-class of perch Perca fluviatilis, in an artificial lake receiving warm water discharges from a nuclear power station and in an unheated reference site, for a two year period. In the heated area the prevalence of infection of the retinal form, D. baeri, was always 100%, whereas in the unheated site there was an increase from 93% in May to 100% in June 1986. The relative density of D. baeri was found to increase gradually during the first summer in both areas, although the accumulation rate of metacercariae was significantly increased in the heated area. In August of the first year the relative density of D. baeri peaked in the heated area, whereas it continued to increase in the unheated control. However, in September of the following year, the relative densities were at the same level in both thermal regimes. A concomitant decline in the degree of overdispersion of metacercariae within the host population was observed in the heated area as the population density of metacercariae decreased, whereas the index of dispersion remained at the same level throughout the study in the unheated area. However, it was not possible to sample perch in the unheated area between October 1986 and May 1987 and changes in the parasite population could have occurred during this period. The prevalence and relative density of D. spathaceum, the lens form, was on the contrary low, especially in the unheated site where it was recorded only occasionally. The infection of D. baeri exhibited a convex pattern in both thermal regimens, although the peak infection was noticed earlier in the heated area. Regulation of the parasite infrapopulation may have been achieved by the combined effects of a decreased transmission rate of cercariae with increasing age of the host, the mortality of metacercariae as a natural termination of the life span and to selective predation of heavily infected hosts. However, regardless of an increased accumulation rate of metacercariae in the heated area, the relative densities of D. baeri became equal in both thermal regimes at the end of the study. The mortality rate of metacercariae in the heated area was therefore presumably increased as compared with the unheated reference site.  相似文献
