The bloom‐forming cyanobacterium Microcystis aeruginosa (Kütz.) Kütz. 854 was cultured with 1.05 W · m?2 ultraviolet‐B radiation (UVBR) for 3 h every day, and the CO2‐concentrating mechanism (CCM) within this species as well as effects of UVBR on its operation were investigated. Microcystis aeruginosa 854 possessed at least three inorganic carbon transport systems and could utilize external HCO3? and CO2 for its photosynthesis. The maximum photosynthetic rate was approximately the same, but the apparent affinity for dissolved inorganic carbon was significantly decreased from 74.7 μmol · L?1 in the control to 34.7 μmol · L?1 in UVBR‐treated cells. At 150 μmol · L?1 KHCO3 and pH 8.0, Na+‐dependent HCO3? transport contributed 43.4%–40.2% to the photosynthesis in the control and 34.5%–31.9% in UVBR‐treated cells. However, the contribution of Na+‐independent HCO3? transport increased from 8.7% in the control to 18.3% in UVBR‐treated cells. The contribution of CO2‐uptake systems showed little difference: 47.9%–51.0% in the control and 49.8%–47.2% in UVBR‐treated cells. Thus, the rate of total inorganic carbon uptake was only marginally affected, although UVBR had a differential effect on various inorganic carbon transporters. However, the number of carboxysomes in UVBR‐treated cells was significantly decreased compared to that in the control.  相似文献

Twelve species, representing 12 families of the chrysophytes sensu lato, were tested for their ability to take up inorganic carbon. Using the pH‐drift technique, CO2 compensation points generally varied between 1 and 20 μmol · L?1 with a mean concentration of 5 μmol · L?1. Neither pH nor alkalinity affected the CO2 compensation point. The concentration of oxygen had a relatively minor effect on CO2‐uptake kinetics, and the mean CO2 compensation point calculated from the kinetic curves was 3.6 μmol · L?1 at 10–15 kPa starting oxygen partial pressure and 3.8 μmol · L?1 at atmospheric starting oxygen partial pressure (21 kPa). Similarly, uptake kinetics were not affected by alkalinity, and hence concentration of bicarbonate. Membrane inlet mass spectrometry (MIMS) in the presence and absence of acetazolamide suggested that external carbonic anhydrase in Dinobryon sertularia Ehrenb. and Synura petersenii Korschikov was either very low or absent. Rates of net HCO3? uptake were very low (~5% of oxygen evolution) using MIMS and decreased rather than increased with increasing HCO3? concentration, suggesting that it was not a real uptake. The CO2 compensation points determined by MIMS for CO2 uptake and oxygen evolution were similar to those determined in pH‐drift and were >1 μmol · L?1. Overall, the results suggest that chrysophytes as a group lack a carbon‐concentrating mechanism (CCM), or an ability to make use of bicarbonate as an alternative source of inorganic carbon. The possible evolutionary and ecological consequences of this are briefly discussed.  相似文献

Migration behaviour and estuarine mortality of cultivated Atlantic salmon Salmo salar smolts in a 16 km long estuary were studied using two methods: (1) acoustic telemetry and (2) group tagging in combination with trap nets. Progression rates of surviving individuals through the estuary were relatively slow using both methods [0·38 LT (total length) s?1 v. 0·25 LT s?1]. In 2012, the progression rate was slow from the river to the estuary (0·55 LT s?1) and the first part of the estuary (0·31 LT s?1), but increased thereafter (1·45–2·21 LT s?1). In 2013, the progression rate was fast from the river to the estuary (4·31 LT s?1) but was slower thereafter (0·18–0·91 LT s?1). Survival to the fjord was higher in 2012 (47%) compared to 2013 (6%). Fast moving individuals were more likely to migrate successfully through the estuary compared to slower moving individuals. Adult recapture of coded‐wire‐tagged S. salar was generally low (0·00–0·04%). Mortality hot spots were related to topographically distinct areas such as the river outlet (in 2012) or the sill separating the estuary and the fjord (in 2013). At the sill, an aggregation of cod Gadus morhua predating on cultivated smolts was identified. The results indicate that slow progression rates through the estuary decreases the likelihood of smolts being detected outside the estuary. The highly stochastic and site‐specific mortality patterns observed in this study highlight the complexity in extrapolating mortality patterns of single release groups to the entire smolt run of wild S. salar.  相似文献

Critical (<30 min) and prolonged (>60 min) swimming speeds in laboratory chambers were determined for larvae of six species of Australian freshwater fishes: trout cod Maccullochella macquariensis, Murray cod Maccullochella peelii, golden perch Macquaria ambigua, silver perch Bidyanus bidyanus, carp gudgeon Hypseleotris spp. and Murray River rainbowfish Melanotaenia fluviatilis. Developmental stage (preflexion, flexion, postflexion and metalarva) better explained swimming ability than did length, size or age (days after hatch). Critical speed increased with larval development, and metalarvae were the fastest swimmers for all species. Maccullochella macquariensis larvae had the highest critical [maximum absolute 46·4 cm s?1 and 44·6 relative body lengths (LB) s?1] and prolonged (maximum 15·4 cm s?1, 15·6 LB s?1) swimming speeds and B. bidyanus larvae the lowest critical (minimum 0·1 cm s?1, 0·3 LB s?1) and prolonged swimming speeds (minimum 1·1 cm s?1, 1·0 LB s?1). Prolonged swimming trials determined that the larvae of some species could not swim for 60 min at any speed, whereas the larvae of the best swimming species, M. macquariensis, could swim for 60 min at 44% of the critical speed. The swimming performance of species with precocial life‐history strategies, with well‐developed larvae at hatch, was comparatively better and potentially had greater ability to influence their dispersal by actively swimming than species with altricial life‐history strategies, with poorly developed larvae at hatch.  相似文献

We report the reappearance of the rare charophyte Nitella translucens in Poland. It was identified in the soft‐water lobelian Lake Jeleń (North Poland) during 2013 and 2018 phytolittoral surveys. This species is considered critically endangered in various European countries and was previously classified as extinct in Poland. Its occurrence was confirmed using morphological and molecular data (ITS1‐18S, ITS2‐28S, rDNA, and rbcL). The Ntranslucens occupied ~20% of the lake bottom, at depths of 1.5–6.5 m, water pH 7.5–8.6, conductivity of 59–66 μS · cm?1, and total nitrogen and phosphorus during growing season in the range of 1.1–1.4 mg · L?1 and 0.07–0.1 mg · L?1, respectively. It co‐occurred mainly with plant species typical for lobelia lakes: Isoetes lacustris, Littorella uniflora, and Myriophyllum alterniflorum, as well as Ceratophyllum demersum and Elodea canadensis, which are characteristic for eutrophicated waters. It appears that N. translucens may thrive in lobelia lakes during their transformation to more eutrophic states.  相似文献

Crustose coralline algae occupied ~1%–2% (occasionally up to 7%) of the sea floor within their depth range of 15–50 m, and they were the dominant encrusting organisms and macroalgae beyond 20 m depth in Young Sound, NE Greenland. In the laboratory, oxygen microelectrodes were used to measure net photosynthesis (P) versus downwelling irradiance (Ed) and season for the two dominant corallines [Phymatolithon foecundum (Kjellman) Düwel et Wegeberg 1996 and Phymatolithon tenue (Rosenvinge) Düwel et Wegeberg 1996] representing> 90% of coralline cover. Differences in P‐Ed curves between the two species, the ice‐covered and open‐water seasons, or between specimens from 17 and 36 m depth were insignificant. The corallines were low light adapted, with compensation irradiances (Ec) averaging 0.7–1.8 μmol photons·m ? 2·s ? 1 and light adaptation (Ek) indices averaging 7–17 μmol photons·m ? 2·s ? 1. Slight photoinhibition was evident in most plants at irradiances up to 160 μmol photons·m ? 2·s ? 1. Photosynthetic capacity (Pm) was low, averaging 43–67 mmol O2·m ? 2 thallus·d ? 1 (~250–400 g C·m ? 2 thallus·yr ? 1). Dark respiration rates averaged ~5 mmol O2·m ? 2 thallus·d ? 1. In ice covered periods, Ed at 20 m depth averaged ~1 μmol photons·m ? 2·s ? 1, with daily maxima of 2–3 μmol photons·m ? 2·s ? 1. During the open water season, Ed at 20 m depth averaged ~7 μmol photons·m ? 2·s ? 1 with daily maxima of ~30 μmol photons·m ? 2·s ? 1. Significant net primary production of corallines was apparently limited to the 2–3 months with open water, and the small contribution of corallines to primary production seems due to low Pm values, low in situ irradiance, and their relatively low abundance in Young Sound.  相似文献

The possibilities for the treatment of low‐temperature mine waste waters have not been widely studied. The amenability of low‐temperature sulfate reduction for mine waste water treatment at 9°C was studied in a bench‐scale fluidized‐bed bioreactor (FBR). Formate was used as the electron and carbon source. The first influent for the FBR was acidic, synthetic waste water containing iron, nutrients, and sulfate, followed by diluted barren bioleaching solution (DBBS). The average sulfate reduction rates were 8 mmol L?1 day?1 and 6 mmol L?1 day?1 with synthetic waste water and DBBS, respectively. The corresponding specific activities were 2.4 and 1.6 mmol SO g VSS?1 day?1, respectively. The composition of the microbial community and the active species of the FBR was analyzed by extracting the DNA and RNA, followed by PCR‐DGGE with the universal bacterial 16S rRNA gene primers and dsrB‐primers specific for sulfate‐reducing bacteria. The FBR microbial community was simple and stable and the dominant and active species belonged to the genus Desulfomicrobium. In summary, long‐term operation of a low‐temperature bioreactor resulted in enrichment of formate‐utilizing, psychrotolerant mesophilic sulfate reducing bacteria. Biotechnol. Aims: To evaluate the antiviral activity of Bignoniaceae species occurring in the state of Minas Gerais, Brazil. Methods and Results: Ethanol extracts of different anatomical parts of bignoniaceous plant species have been evaluated in vitro against human herpesvirus type 1 (HSV‐1), vaccinia virus (VACV) and murine encephalomyocarditis virus (EMCV) by the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide assay. A total of 34 extracts from 18 plant species selected according to ethnopharmacological and taxonomic criteria were screened. Fifteen of the 34 extracts (44·1%) have disclosed antiviral activity against one or more of the viruses assayed with EC50 values in the range of 23·2 ± 2·5–422·7 ± 10·9 μg ml?1. Conclusions: Twelve of the 34 extracts (35·3%) might be considered promising sources of antiviral natural products, as they have shown EC50 ≤ 100 μg ml?1. The present screening discloses the high potential of the Bignoniaceae family as source of antiviral agents. Significance and Impact of the Study: Active extracts were identified and deserve bioguided studies for the isolation of antiviral compounds and studies on mechanism of action.  相似文献

We investigated the contribution of internal water storage and efficiency of water transport to the maintenance of water balance in six evergreen tree species in a Hawaiian dry forest. Wood‐saturated water content, a surrogate for relative water storage capacity, ranged from 70 to 105%, and was inversely related to its morphological correlate, wood density, which ranged between 0·51 and 0·65 g cm?3. Leaf‐specific conductivity (kL) measured in stem segments from terminal branches ranged from 3 to 18 mmol m?1 s?1 MPa?1, and whole‐plant hydraulic efficiency calculated as stomatal conductance (g) divided by the difference between predawn and midday leaf water potential (ΨL), ranged from 70 to 150 mmol m?2 s?1 MPa?1. Hydraulic efficiency was positively correlated with kL (r2 = 0·86). Minimum annual ΨL ranged from ? 1·5 to ? 4·1 MPa among the six species. Seasonal and diurnal variation in ΨL were associated with differences among species in wood‐saturated water content, wood density and kL. The species with higher wood‐saturated water content were more efficient in terms of long‐distance water transport, exhibited smaller diurnal variation in ΨL and higher maximum photosynthetic rates. Smaller diurnal variation in ΨL in species with higher wood‐saturated water content, kL and hydraulic efficiency was not associated with stomatal restriction of transpiration when soil water deficit was moderate, but avoidance of low minimum seasonal ΨL in these species was associated with a substantial seasonal decline in g. Low seasonal minimum ΨL in species with low kL, hydraulic efficiency, and wood‐saturated water content was associated with higher leaf solute content and corresponding lower leaf turgor loss point. Despite the species‐specific differences in leaf water relations characteristics, all six evergreen tree species shared a common functional relationship defined primarily by kL and stem water storage capacity.  相似文献

The influence of surgical implantation of an acoustic transmitter on the swimming performance, growth and survival of juvenile sockeye salmon Oncorhynchus nerka and Chinook salmon Oncorhynchus tshawytscha was examined. The transmitter had a mass of 0·7 g in air while sockeye salmon had a mass of 7·0–16·0 g and Chinook salmon had a mass of 6·7–23·1 g (a transmitter burden of 4·5–10·3% for sockeye salmon and 3·1–10·7% for Chinook salmon). Mean critical swimming speeds (Ucrit) for Chinook salmon ranged from 47·5 to 51·2 cm s?1 [4·34–4·69 body lengths (fork length, LF) s?1] and did not differ among tagged, untagged and sham‐tagged groups. Tagged sockeye salmon, however, did have lower Ucrit than control or sham fish. The mean Ucrit for tagged sockeye salmon was 46·1 cm s?1 (4·1 LF s?1), which was c. 5% less than the mean Ucrit for control and sham fish (both groups were 48·6 cm s?1 or 4·3 LF s?1). A laboratory evaluation determined that there was no difference in LF or mass among treatments (control, sham or tag) either at the start or at the end of the test period, suggesting that implantation did not negatively influence the growth of either species. None of the sockeye salmon held under laboratory conditions died from the influence of surgical implantation of transmitters. In contrast, this study found that the 21 day survival differed between tagged and control groups of Chinook salmon, although this result may have been confounded by the poor health of Chinook salmon treatment groups.  相似文献

Schizochytrium sp. PQ6, a heterotrophic microalga isolated from Phu Quoc (PQ) Island in the Kien Giang province of Vietnam, contains a high amount of docosahexaenoic acid (DHA, C22:6n‐3). In this study, the culture conditions are developed to maximize biomass and DHA production. Nucleotide sequence analysis of partial 18S rRNA gene from genomic DNA showed that PQ6 has a phylogenetic relationship close to Schizochytrium mangrovei Raghu‐Kumar. The highest growth rate and DHA accumulation of this strain were obtained in 6.0% glucose, 1.0% yeast extract, 50% artificial seawater (ASW), and pH 7 at 28°C. In addition, carbon and nitrogen sources could be replaced by glycerol, ammonium acetate, sodium nitrate, or fertilizer N–P–K. Total lipid content reached 38.67% of dry cell weight (DCW), in which DHA and eicosapentaenoic acid (EPA, C20:5n‐3) contents accounted for 43.58% and 0.75% of the total fatty acid (TFA), respectively. In 5 and 10 L fermenters, the cell density, DCW, total lipid content, and maximum DHA yield were 46.50 × 106 cells · mL?1, 23.7 g · L?1, 38.56% of DCW, and 8.71 g · L?1 (in 5 L fermenter), respectively, and 49.71 × 106 cells · mL?1, 25.34 g · L?1, 46.23% of DCW, and 11.55 g · L?1 (in 10 L fermenter), respectively. Biomass of PQ6 strain possessed high contents of Na, I, and Fe (167.185, 278.3, and 43.69 mg · kg?1 DCW, respectively). These results serve as a foundation for the efficient production of PQ6 biomass that can be used as a food supplement for humans and aquaculture in the future.  相似文献

Complex chemistry and biological uptake pathways render iron bioavailability particularly difficult to assess in natural waters. Bioreporters are genetically modified organisms that are useful tools to directly sense the bioavailable fractions of solutes. In this study, three cyanobacterial bioreporters derived from Synechococcus PCC 7942 were examined for the purpose of optimizing the response to bioavailable Fe. Each bioreporter uses a Fe‐regulated promoter (isiAB, irpA and mapA), modulated by distinct mechanisms under Fe deficiency, fused to a bacterial luciferase (luxAB). In order to provide a better understanding of the way natural conditions may affect the ability of the bioreporter to sense iron bioavailability, the effect of relevant environmental parameters on the response to iron was assessed. Optimal conditions (and limits of applicability) for the use of these bioreporters on the field were determined to be: a 12 h (12–24 h) exposure time, temperature of 15°C (15°C–22°C), photon flux density of 100 μmol photons·m?2·s?1 (37–200 lmol photons·m?2·s?1), initial biomass of 0.6–0.8 lg chlorophyll a (chl a)·L?1 (0.3–1.5 lg chl a·L?1) or approximately 105 bioreporter cells·mL?1, high phosphate (10 lM), and low micronutrients (absent). The measured luminescence was optimal with an exogenous addition of 60 lM aqueous decanal substrate allowing a 5 min reaction time in the dark before analysis. This study provides important considerations relating to the optimization in the use of bioreporters under field conditions that can be used for method development of other algal and cyanobacterial bioreporters in aquatic systems.  相似文献

High bulk extracellular phosphatase activity (PA) suggested severe phosphorus (P) deficiency in plankton of three acidified mountain lakes in the Bohemian Forest. Bioavailability of P substantially differed among the lakes due to differences in their P loading, as well as in concentrations of aluminum (Al) and its species, and was accompanied by species‐specific responses of phytoplankton. We combined the fluorescently labeled enzyme activity (FLEA) assay with image cytometry to measure cell‐specific PA in natural populations of three dinophyte species, occurring in all the lakes throughout May–September 2007. The mean cell‐specific PA varied among the lakes within one order of magnitude: 188–1,831 fmol · cell?1 · h?1 for Gymnodinium uberrimum (G. F. Allman) Kof. et Swezy, 21–150 fmol · cell?1 · h?1 for Gymnodinium sp., and 22–365 fmol · cell?1 · h?1 for Peridinium umbonatum F. Stein. To better compare cell‐specific PA among the species of different size, the values were normalized per unit of cell biovolume (amol · μm?3 · h?1) for further statistical analysis. A step‐forward selection identified concentrations of total and ionic Al together with pH as significant factors (P < 0.05, Monte Carlo permutation test), explaining cumulatively 57% of the total variability in cell‐specific PA. However, this cell‐specific PA showed an unexpected reverse trend compared to an overall gradient in P deficiency of the lake plankton. The autecological insight into dinophyte cell‐specific PA therefore suggested other factors, such as light availability, mixotrophy, and/or zooplankton grazing, causing further PA variations among the acidified lakes.  相似文献

A survey of the spatial distribution of benthic macroalgae in a fluvial lake of the St. Lawrence River (Lake Saint‐Pierre, Quebec, Canada) revealed a shift in composition from chlorophytes to cyanobacteria along the flow path of nutrient‐rich waters originating from tributaries draining farmlands. The link between this shift and changes in water quality characteristics was investigated by sampling at 10 sites along a 15 km transect. Conductivity, current, light extinction, total phosphorus (TP; >25 μg P · L?1), and ammonium (8–21 μg N · L?1) remained fairly constant along the transect in contrast to nitrate concentrations, which fell sharply. Filamentous and colonial chlorophytes [Cladophora sp. and Hydrodictyon reticulatum (L.) Bory] dominated in the first 5 km where nitrate concentrations were >240 μg N · L?1. A mixed assemblage of chlorophytes and cyanobacteria characterized a 1 km transition zone where nitrate decreased to 40–80 μg N · L?1. In the last section of the transect, nitrate concentrations dropped below 10 μg N · L?1, and cyanobacteria (benthic filamentous mats of Lyngbya wollei Farl. ex Gomont and epiphytic colonies of Gloeotrichia) dominated the benthic community. The predominance of nitrogen‐fixing, potentially toxic cyanobacteria likely resulted from excessive nutrient loads and may affect nutrient and trophic dynamics in the river.  相似文献

Baseline, post‐angling and maximum attainable blood lactate concentrations were measured for the fishery species redthroat emperor Lethrinus miniatus to gain insight into the condition of fish released following c. 30 s angling and <45 s air exposure. Mean ± s.d . baseline blood lactate was 1·5 ± 0·6 mmol l?1, which increased and plateaued around 6 mmol l?1 at 15–30 min post‐angling. These values were significantly lower than those obtained from fish maximally exhausted with a prolonged chase and air exposure protocol following capture (10·9 ± 1·8 mmol l?1), suggesting that L. miniatus is not maximally exhausted during standard angling practices.  相似文献

Ultrasonic telemetry was used to compare post‐release survival and movements of Atlantic sharpnose sharks Rhizoprionodon terraenovae in a coastal area of the north‐east Gulf of Mexico. Ten fish were caught with standardized hook‐and‐line gear during June to October 1999. Atlantic sharpnose sharks were continuously tracked after release for periods of 0·75 to 5·90 h and their positions recorded at a median interval of 9 min. Individual rate of movement was the mean of all distance and time measurements for each fish. Mean ± s.e . individual rate of movement was 0·45 ± 0·06 total lengths per second (LT s?1) and ranged from 0·28 to 0·92 LT s?1 over all fish. Movement patterns did not differ between jaw and internally hooked Atlantic sharpnose sharks. Individual rate of movement was inversely correlated with bottom water temperature at capture (r2 = 0·52, P ≤ 0·05). No consistent direction in movement was detected for Atlantic sharpnose sharks after release, except that they avoided movement towards shallower areas. Capture‐release survival was high (90%), with only one fish not surviving, i.e. this particular fish stopped movement for a period of 10 min. Total rate of movement was total distance over total time (m min?1) for each Atlantic sharpnose shark. Mean total rate of movement was significantly higher immediately after release at 21·5 m min?1 over the first 1·5 h of tracking, then decreased to 11·2 m min?1 over 1·5–6 h, and 7·7 m min?1 over 3–6 h (P ≤ 0·002), which suggested initial post‐release stress but quick recovery from capture. Thus, high survival (90%) and quick recovery indicate that the practice of catch‐and‐release would be a viable method to reduce capture mortality for R. terraenovae.  相似文献

In the unicellular charophycean alga Closterium peracerosum‐strigosum‐littorale complex, the protoplast‐release‐inducing protein (PR‐IP), a sex pheromone responsible for gametic protoplast release from mating‐type minus (mt) cells, was found to stimulate secretion of mucilage from the cells. Induction of sexual cell division by PR‐IP was also confirmed. Bioassays were used to determine the minimum doses required to induce these functions, revealing that 5 · 10?16 M of PR‐IP stimulated mucilage secretion, and that 5 · 10?10 M of PR‐IP were required for protoplast release. Exposure of the cells to 5 · 10?11 M of PR‐IP resulted in the induction of sexual cell division as well as mucilage secretion. These results strongly suggest that PR‐IP is a multifunctional pheromone that independently promotes multiple steps in conjugation at the appropriate times through different induction mechanisms.  相似文献

Aims: To improve the yield and productivity of docosahexaenoic acid (DHA) by Schizochytrium sp. in terms of the analysis of microbial physiology. Methods and Results: A two‐stage oxygen supply control strategy, aimed at achieving high concentration and high productivity of DHA, was proposed. At the first 40 h, KLa was controlled at 150·1 h?1 to obtain high μ for cell growth, subsequently KLa was controlled at 88·5 h?1 to maintain high qp for high DHA accumulation. Finally, the maximum lipid, DHA content and DHA productivity reached 46·6, 17·7 g l?1 and 111 mg l?1 h?1, which were 43·83%, 63·88% and 32·14% over the best results controlled by constant KLa. Conclusions: This paper described a two‐stage oxygen supply control strategy based on the kinetic analysis for efficient DHA fermentation by Schizochytrium sp. Significance and Impact of the study: This study showed the advantage of two‐stage control strategy in terms of microbial physiology. As KLa is a scaling‐up parameter, the idea developed in this paper could be scaled‐up to industrial process and applied to other industrial biotechnological processes to achieve both high product concentration and high productivity.  相似文献

The removal efficiency of Cu2+ by Spirulina platensis (strain FACHB‐834), in viable and heat‐inactivated forms, was investigated in the presence and absence of linear alkylbenzene sulfonate (LAS). When the initial Cu2+ concentration was in the range of 0.5–1.5 mg · L?1, a slight increase in growth rate of FACHB‐834 was observed. In contrast, when Cu2+ or LAS concentrations were at or higher than 2.0 or 6.0 mg · L?1, respectively, the growth of FACHB‐834 was inhibited and displayed yellowing and fragmentation of filaments. The presence of LAS improved Cu2+ removal by ~20%, and accelerated attainment of Cu2+ retention equilibrium. For the 2‐ mg · L?1 Cu2+ treatments, retention equilibrium occurred within 2 d and showed maximum Cu2+ removal of 1.83 mg · L?1. In the presence of LAS, the ratio of extracellular bound Cu2+ to intracellular Cu2+ taken up by the cells was lower (1.05–2.26) than corresponding ratios (2.46–7.85) in the absence of LAS. The percentages of extracellular bound Cu2+ to total Cu2+ removal (both bound and taken up by cells) in the presence of LAS ranged from 51.2% to 69.3%, which was lower than their corresponding percentages (71.1%–88.7%) in the absence of LAS. LAS promoted biologically active transport of the extracellular bound form of Cu2+ into the cell. In contrast, the addition of LAS did not increase the maximum removal efficiency of Cu2+ (61.4% ± 5.6%) by heat‐inactivated cells compared to that of living cells (59.6% ± 6.0%). These results provide a theoretical foundation for designing bioremediation strategies using FACHB‐834 for use in surface waters contaminated by both heavy metals and LAS.  相似文献

The toxigenic diatom Pseudo‐nitzschia cuspidata, isolated from the U.S. Pacific Northwest, was examined in unialgal batch cultures to evaluate domoic acid (DA) toxicity and growth as a function of light, N substrate, and growth phase. Experiments conducted at saturating (120 μmol photons · m?2 · s?1) and subsaturating (40 μmol photons · m?2 · s?1) photosynthetic photon flux density (PPFD), demonstrate that P. cuspidata grows significantly faster at the higher PPFD on all three N substrates tested [nitrate (NO3?), ammonium (NH4+), and urea], but neither cellular toxicity nor exponential growth rates were strongly associated with one N source over the other at high PPFD. However, at the lower PPFD, the exponential growth rates were approximately halved, and the cells were significantly more toxic regardless of N substrate. Urea supported significantly faster growth rates, and cellular toxicity varied as a function of N substrate with NO3?‐supported cells being significantly more toxic than both NH4+‐ and urea‐supported cells at the low PPFD. Kinetic uptake parameters were determined for another member of the P. pseudodelicatissima complex, P. fryxelliana. After growth of these cells on NO3? they exhibited maximum specific uptake rates (Vmax) of 22.7, 29.9, 8.98 × 10?3 · h?1, half‐saturation constants (Ks) of 1.34, 2.14, 0.28 μg‐at N · L?1, and affinity values (α) of 17.0, 14.7, 32.5 × 10?3 · h?1/(μg‐at N · L?1) for NO3?, NH4+ and urea, respectively. These labo‐ratory results demonstrate the capability of P. cuspidata to grow and produce DA on both oxidized and reduced N substrates during both exponential and stationary growth phases, and the uptake kinetic results for the pseudo‐cryptic species, P. fryxelliana suggest that reduced N sources from coastal runoff could be important for maintenance of these small pennate diatoms in U.S. west coast blooms, especially during times of low ambient N concentrations.  相似文献
