Helianthus annuus L., the common cultivated sunflower, and Helianthus bolanderi Gray subspecies exilis Heiser, a form endemic on serpentine soils, were grown in culture solutions with widely varying Mg levels. For comparable development, H. bolanderi exilis required higher levels of Mg in the solution than H. annuus, both in the range of visual deficiency symptoms and in higher ranges.  相似文献

As a first step in the study of chloroplast genome variability in the genus Helianthus, a physical restriction map of sunflower (Helianthus annuus) chloroplast DNA (cpDNA) has been constructed using restriction endonucleases BamH I, Hind III, Pst I, Pvu II and Sac. I. Sunflower circular DNA contains an inverted repeat structure with the two copies (23 kbp each) separated by a large (86 kbp) and a small (20 kbp) single copy region. Its total length is therefore about 152 kbp. Sunflower cpDNA is essentially colinear with that of tobacco with the exception of an inversion of a 23.5-kbp segment in the large single copy region. Gene localization on the sunflower cpDNA and comparison of the gene map with that from tobacco chloroplasts have revealed that the endpoints of the inversion are located between the trnT and trnE genes on the one hand, and between the trnG and trnS genes on the other hand.Analysis of BamH I restriction fragment patterns of H. annuus, H. occidentalis ssp. plantagineus, H. grossesseratus, H. decapetalus, H. giganteus, H. maximiliani and H. tuberosus cpDNAs suggests that structural variations are present in the genus Helianthus.  相似文献

The phytotoxic compound deoxyradicinin, a metabolite of Alternaria helianthi, was isolated from naturally infected leaves of sunflower (Helianthus annuus). The production of this toxin in liquid culture together with 3-epideoxyradicinol is discussed. Production of deoxyradicinin by liquid cultures was observed to decrease after successive sub-culture of the fungus on solid medium. All 10 accessions of Helianthus tested were sensitive to deoxyradicinin although some quantitative differences between lines were noted. Evidence is presented for the metabolism of deoxyradicinin by host leaf tissue.  相似文献

通过田间选择性试验结合室内黑色素鉴定,研究向日葵螟Homoeosoma nebulellum Denis et Schiffermüller对35个不同向日葵品种的寄主选择性。向日葵螟对不同供试向日葵品种的选择性存在显著差异,选择性最强的是大黑片,其次是S47,再则是5135和RH316,选择性较弱的有T25和天葵503等18个品种,对TO12244和GT110等13个品种无选择性。寄主选择性可以用开花盛期的花盘被害率、每盘可见幼虫数、每盘虫粪指数与籽粒成熟期的籽粒被害率、籽粒被害虫情指数5个指标中的任何一个来评价。相关性分析表明,向日葵螟的寄主选择性与向日葵花盘直径呈显著正相关关系。  相似文献

The expression of genomic instability was studied at the phenotypical (morphological characters, electrophoretic spectra of seed storage proteins) and molecular (DNA amplification products) levels in interspecific hybrids (ISHs) from crosses of inbred lines of cultivated sunflower Helianthus annuus with perennial species of the genus Helianthus and in introgressive lines (ILs) produced on their basis. Unstable state of the locus determining the trait of lower branching was proved by the method of hybridological analysis. It was shown with the use of RAPD markers that the IL genome is characterized by instability even after long-term inbreeding (in generations F8-F12). In progenies of different combinations of interspecific crosses, identical polymorphic variants were revealed for a seed storage protein, helianthinin, and for DNA fragments homologous to structural genes of functionally important proteins, suggesting the nonrandom character of ISH genome variation. This variation may be determined by genome reorganizations under the action of a genome shock induced by interspecific hybridization. The factors inducing reorganizations in the genome include the activity of mobile genetic elements (MGEs). Using primers specific to different MGE families, nucleotide sequences with a high level of homology to the sequences of fragments of the mobile elements MuDR, Far1, CACTA, Stowaway, and Tourist were identified in the sunflower genome. The possibility of using MGE fragments for sunflower genotyping was demonstrated.  相似文献

Summary The restriction map of the rDNA unit of Helianthus annuus was constructed using EcoRI, BamHI, HindIII, KpnI and SacI restriction enzymes. Variations in this map among 61 ecotypes representing 39 species of the genus Helianthus were analyzed. The sizes of the rDNA unit ranged from 9.8 to 11.0 kbp, due to a length-repeat heterogeneity of the external non-transcribed spacer by increments of 200 base pair segments. Lengthrepeat heterogeneity and restriction polymorphism were found to be characteristic of populations or species of Helianthus. Restriction patterns and thermal melting with probes of a cloned H. annuus ENTS segment allowed us to differentiate species from each other. However, most lines of the cultivated sunflower were found to be identical on the basis of the physical properties of their ribosomal DNA.  相似文献

Hybridisation between the annual diploid sunflower (Helianthus annuus) and the perennial diploid species Helianthus mollis and Helianthus orgyalis was obtained by means of a normal crossing procedure or embryo rescue. Hybridisation success was low. All plants examined cytologically appeared to be diploid. However, the phenotypes of these diploids were not intermediate between the parents and, despite great variation, they resembled the female parent-type predominantly. Thirty five percent of plants issued from sunflower pollinated with perennial Helianthus had a phenotype resembling the female sunflower parent. On average, only 5% of the minimum number of expected RAPD and RFLP bands from male parents were recovered in plants produced from mature seeds after pollination of sunflower by H. mollis. More hybrids were found among plants obtained from embryo rescue, with an average of 25% of the male parent bands recovered per plant. Analysis of individual plants indicated the occurrence of various levels of hybridisation. There was a significant positive correlation between the number of phenotype traits related to hybrid status and the number of bands derived from the male parent. A single hybrid plant might possibly represent the product of a ’normal’ hybridisation event. The mechanisms behind these unusual events and the consequences for the breeder are discussed.   • Seed functional traits of native Helianthus species contribute towards ecosystem services but limitations to their use in managed programmes exist. Many perennial Helianthus possess seed dormancy. The ability for germination to occur under different temperature and drought conditions, as well as the capacity of germinated seeds to convert into normal seedlings is rarely considered. Our aim was to identify and quantify these constraints through functional trait analyses.
  • In five seed lots of native Helianthus (four perennial and one annual) and five genotypes of sunflower (H. annuus) for comparison, dormancy, thermal and hydro thresholds and times, morphology, mass, oil content and conversion into normal seedlings were quantified. The influence of the seed collection site environment on these traits was also explored.
  • Seed dormancy of the perennial species was overcome by scarification followed by germination in 5 mm GA3. Thermal and hydro‐time analyses revealed slower germination for the native seed lots (>1350 °Ch) in comparison to the sunflower genotypes (<829.9 °Ch). However, native seed lots had a higher capacity to convert into normal seedlings at high temperatures and low water potentials than sunflower genotypes. For the native seed lots, the average monthly temperature of the collection site was negatively correlated with thermal time.
  • Variability in seed functional traits of native Helianthus and greater capacity for germinated seeds to convert into normal seedlings suggests they are better equipped to cope with high temperature and drought scenarios than sunflower. Effective dormancy alleviation is required to facilitate the use of native Helianthus species.

Assessing levels and patterns of population genetic variation is an important step for evaluating rare or endangered species and determining appropriate conservation strategies. This is particularly important for ensuring the preservation of novel genetic variation in wild relatives of crops, which could provide beneficial alleles for plant breeding and improvement. In this study, we evaluate the population genetics of Helianthus niveus ssp. tephrodes (the Algodones sunflower), which is an endangered, wild relative of cultivated sunflower (H. annuus L.). This rare sunflower species is native to the sand dunes of the Sonoran Desert in southern California, southwestern Arizona, and northern Mexico and is thought to harbor beneficial alleles for traits related to drought tolerance. We genotyped nine populations of this species with a set of simple-sequence repeat markers derived from expressed sequence tags (EST-SSRs) and investigated levels of genetic diversity and population structure, in H. niveus ssp. tephrodes. We also compared our results to findings from five related sunflower species that have been analyzed with these same markers, including annuals and perennials that range from rare to widespread. The Algodones sunflower harbors lower levels of standing genetic variation, but similar levels of population structure as compared to other sunflower species. We also discovered that a disjunct population from northern Mexico was genetically distinct from populations elsewhere in the range. Given the occurrence of such a genetically unique population, our recommendations include population surveys of the southern portion of the range in hopes of bolstering the existing germplasm collection.  相似文献

A highly sensitive gelatin overlay procedure was used to identify inhibitors of serine proteinases and of the cysteine proteinase ficin in seeds and leaves of sunflower. One major and two minor groups of trypsin inhibitors were identified in seeds, the former having a high pI (@10) and also inhibiting chymotrypsin. Three groups of trypsin/subtilisin inhibitors were also present in seeds, together with three inhibitors of ficin. All groups showed polymorphism between lines of Helianthus annuus, while the trypsin and trypsin/subtilisin inhibitors also varied between wild species of Helianthus, with no apparent relationship to growth type (annual or perennial), genome constitution or ploidy level. Genetic analysis showed that the major trypsin inhibitor and three groups of trypsin/subtilisin inhibitors are each controlled by single Mendelian loci, with the three loci for trypsin/subtilisin inhibitors showing recombination values of 0.23–0.40. Purification by RP-HPLC allowed the M r of two trypsin inhibitors to be determined by SDS-PAGE to be about 1,500 and 2,500, while the three trypsin/subtilisin inhibitors varied in M r from about 1,500 to 6,000. From numerous pollinations of Helianthus laciniatus A. Gray sect. Ciliares by H. annuus L. sect. Helianthus (each with n = 17), a single hybrid was grown to maturity. The hybrid was vigorous but only 2.24% pollen-fertile compared to greater than 96% fertility in the parents. Backcrossing to the parental species was unsuccessful, so the possibility of gene flow between the species in nature is remote. Chromosome pairing in the parental plants was normal, but the hybrid was heterozygous for at least one paracentric inversion and numerous translocations. Chiasma frequencies of the parental genotypes were significantly different. The hybrid was significantly different from H. laciniatus and from the midparent value but not from H. annuus. These differences in chiasma frequencies were tested by the way chiasmata were allocated among the different meiotic configurations. Methods of converting multivalents to bivalent equivalents for comparison of chiasmata distribution among species and hybrids are demonstrated.  相似文献

Downy mildew is a fungal disease of sunflower that can lead to severe yield losses. The damage caused by the pathogen can be controlled by growing resistant sunflower varieties. Gene Pl arg was introgressed into cultivated sunflower from the wild species Helianthus argophyllus and provides resistance against all known downy mildew races. In this study, we used a mapping population from the cross-RHA 419/RHA-N-49. We identified a new co-segregating simple sequence repeat marker ORS675 and confirmed the co-segregation of markers ORS716 and ORS662 with Pl arg gene. The markers were validated on two registered resistant inbred lines RHA 443 and RHA 464, as well as on twenty inbred lines RH 1–20 obtained through methods of classical breeding. Molecular marker ORS716 was assessed for usefulness in selecting resistant progeny in 12 BC populations. Markers were found to be valuable for molecular breeding in diverse genetic backgrounds and enabled transfer of the resistance gene in different sunflower genotypes.  相似文献

Sunflower pericarps provide a barrier against seed feeding by larvae of the sunflower moth, Homoeosoma electellum. Pericarp hardening is thought to be accelerated by a phytomelanin layer beneath the hypodermis, but among germplasm with phytomelanin, broad variation in sunflower pericarp strength exists. To facilitate the use of pericarp strength in sunflower breeding, resistance to mechanical puncture was assessed for diverse sunflower germplasm, and feeding tests were used to evaluate whether differences in pericarp strength prevent H. electellum larvae from penetrating achenes. Test on field-grown sunflowers indicates that public restorer lines have lower pericarp strengths compared with maintainer lines and commercial hybrids at 14 days after the start of anthesis. Interspecific crosses or plant introduction (PI) accessions believed to be resistant to H. electellum, including PI 170415, comprised a group with exceptionally high pericarp strength relative to other germplasm. In subsequent tests on greenhouse-grown sunflowers, overall results were similar, but using field-grown plants provided greater statistical power. In choice tests with achenes that differed in pericarp strength, 7- to 9-day-old sunflower moth larvae fed more often on seed protected by a weaker pericarp, at a more than 5-to-1 ratio, while 10-day-old larvae fed indiscriminately. Pericarp strength data contradict previous published results for individual entries and heterotic groups, but support the generalization that improved physical resistance to the sunflower moth is possible. To use pericarp strength in PI 170415 or similar sources, the inheritance of high pericarp strength and potential trade-offs between pericarp strength and other agronomic traits need to be understood.  相似文献

The red sunflower seed weevil, Smicronyx fulvus LeConte (Coleoptera: Curculionidae), is a primary seed-feeding pest of cultivated sunflowers, Helianthus annuus L., in North America. Host plant resistance is one tool available to complement insecticide-based management of S. fulvus. Artificial infestations of 30 adult weevils per head were used to determine whether variation for susceptibility to S. fulvus exists in previously released inbred lines, and how a new weevil-resistant line, HA 488, compares with other putative sources of resistance. Correcting for the number of seeds per head, 13 older inbred lines showed variation in per cent seed damage from 20% to 38%, with two lines (HA 412 HO, HA 821) being more damaged than most of the tested lines. Among four putative resistance sources, HA 488 was significantly less damaged (5%) than two previously identified open-pollinated varieties (PI 170424, PI 253417, with 13%–14% seed damage), while the source of the resistance in HA 488, PI 431542, was statistically intermediate (12%). The resistance available in HA 488 is a marked improvement, potentially reducing damage per weevil by two thirds or more, but additional work on genetic markers for resistance, economic thresholds and basic weevil biology (e.g. degree-day models for adult emergence) is needed to support implementation of integrated pest management for this key sunflower pest.  相似文献

Summary A circular supercoiled mitochondrial DNA plasmid P1 (1.45 kb) is shown in both normal fertile plants of Helianthus annuus, and some cytoplasmic male sterile lines (CMS A and CMS P). In contrast, no plasmid is found in some other types of CMS C, I, B and K. A circular supercoiled DNA (P2) of higher molecular weight (1.8 kb) is observed in CMS F. The mitochondrial plasmid P1 was cloned, nick-translated and hybridized with native mitochondrial DNA from different lines of male fertile, CMS or wild Helianthus. No sequence homology has been detected between plasmid DNA P1 and high molecular weight mitochondrial DNA in any line examined. A slight hybridization occurs between plasmids P1 and P2. Thus, there is no apparent relationship between mitochondrial plasmid DNA and CMS or Helianthus species. On the contrary, each Helianthus CMS and male fertile strain can be characterized by digestion fragment patterns (Sal I and Bgl I). Analysis of mitochondrial DNA from wild Helianthus strains indicated a relation between some CMS and the strain from which they were maternally derived, as for example CMS I and H. annuus ssp lenticularis and CMS F and H. petiolaris fallax. On the basis of restriction endonuclease patterns, a CMS phylogenic tree is proposed which illustrates a molecular polymorphism in the mitochondrial genome of Helianthus.  相似文献

Although the sexual transfer of genetic material between species (i.e. introgression) has been documented in many groups of plants and animals, genome‐wide patterns of introgression are poorly understood. Is most of the genome permeable to interspecific gene flow, or is introgression typically restricted to a handful of genomic regions? Here, we assess the genomic extent and direction of introgression between three sunflowers from the south‐central USA: the common sunflower, Helianthus annuus ssp. annuus; a near‐endemic to Texas, Helianthus debilis ssp. cucumerifolius; and their putative hybrid derivative, thought to have recently colonized Texas, H. annuus ssp. texanus. Analyses of variation at 88 genetically mapped microsatellite loci revealed that long‐term migration rates were high, genome‐wide and asymmetric, with higher migration rates from H. annuus texanus into the two parental taxa than vice versa. These results imply a longer history of intermittent contact between H. debilis and H. annuus than previously believed, and that H. annuus texanus may serve as a bridge for the transfer of alleles between its parental taxa. They also contradict recent theory suggesting that introgression should predominantly be in the direction of the colonizing species. As in previous studies of hybridizing sunflower species, regions of genetic differentiation appear small, whether estimated in terms of FST or unidirectional migration rates. Estimates of recent immigration and admixture were inconsistent, depending on the type of analysis. At the individual locus level, one marker showed striking asymmetry in migration rates, a pattern consistent with tight linkage to a Bateson–Dobzhansky–Muller incompatibility.  相似文献

Background  

Long terminal repeat (LTR) retrotransposons are a class of mobile genetic element capable of autonomous transposition via an RNA intermediate. Their large size and proliferative ability make them important contributors to genome size evolution, especially in plants, where they can reach exceptionally high copy numbers and contribute substantially to variation in genome size even among closely related taxa. Using a phylogenetic approach, we characterize dynamics of proliferation events of Ty3/gypsy-like LTR retrotransposons that led to massive genomic expansion in three Helianthus (sunflower) species of ancient hybrid origin. The three hybrid species are independently derived from the same two parental species, offering a unique opportunity to explore patterns of retrotransposon proliferation in light of reticulate evolutionary events in this species group.  相似文献

Illus. 1960.—The cultivated ornamental sunflower, Helianthus multiflorus L., has been known since 1591 when it was described from Europe by Tabernaemontanus. Both Gray and Bailey concluded that it was a variety of H. decapetalus. The plant is invariably sterile and Dod was first to suggest a hybrid origin from H. annuus and H. decapetalus. Helianthus multiflorus has been found to be triploid (2n = 51) and at meiosis generally shows 17 bivalents and 17 univalents. Morphologically, the plant is similar to H. decapetalus (n = 34) but differs from it in the more hispid stem, broader leaves and phyllaries, and other characters, all of which could have been derived from H. annuus (n = 17). The artificial hybrid between H. decapetalus and H. annuus, while not readily obtained, has been secured, and the hybrid is rather similar to H. multiflorus. It is concluded that this taxon most likely originated in Europe from spontaneous hybridization between H. decapetalus and H. annuus, following the introduction of these species from North America.  相似文献

The relationship between oviposition prefer-ence and offspring performance of a leaf-mining moth (Paraleucoptera sinuella) on four Salicaceae species was investigated in 1997 and 1998. We observed the egg distribution pattern on different plant species in the field and carried out oviposition experiments in the laboratory to determine the preference of ovipositing females. We also examined larval survival, pupal mass, and developmental time to compare larval performance on each plant species. Egg density in the field differed significantly among plant species. However, egg density was not correlated exactly with demonstrated oviposition preference. No larvae could develop on two Salix species. This finding indicated that larval survival is the most critical index of larval performance. Larval performance on each plant species was correlated well with oviposition preference that was revealed by a no-choice experiment in the laboratory. However, this correlation was not found in the field. These results indicate that the preference–performance linkage that was observed under laboratory conditions, was not always maintained in the field.
