荔枝园养鸡配套技术及效益分析   总被引:1,自引:0,他引:1  
果园养鸡是果园立体农业的重要生态模式之一, 其一方面可提高果园的土地利用效率和经济效益; 另一方面可利用鸡的活动减少果园的病虫草害, 并利用鸡粪作为有机肥, 可减少农药和化肥的使用, 从而提高果园的社会效益和生态效益。论文选取华南地区典型的荔枝园养鸡立体农业生态模式为对象, 研究了华南地区荔枝园养鸡模式的配套技术及其效益。结果表明, 荔枝园养鸡可明显地降低农药和化肥的投入成本, 同时该种模式中所增加的鸡收入, 促使整个立体模式经济收益增加, 与传统荔枝园的单一种植相比, 荔枝园养鸡年利润可达到95355 元·hm–2, 约是普通荔枝园经济收益48900元·hm–2 的2 倍。因此, 果园养鸡是一种集社会效益、经济效益和生态效益为一体的立体生态循环农业模式。  相似文献

以海南省文昌市典型的胶茶鸡农林复合系统模式为研究对象,分析了我国热带地区单作胶园、胶茶间作园及养鸡胶园的能流路径、能量输入、输出结构及能流转化效率。结果表明,单作、间作及养鸡胶园的光能利用率逐渐提高,其能量产投比为1.50、1.88、0.20;有机能投入比为0.138、0.119、0.977,养鸡胶园最高;无机能投入比分别为0.862、0.881、0.023;养鸡胶园中人工辅助能投入比相对减少,其产出能最大.  相似文献

枯草芽胞杆菌菌肥对有机冬瓜根区土壤微生态的影响   总被引:4,自引:0,他引:4  
【背景】微生物肥料已广泛应用于我国有机作物的种植,其对有机种植土壤微生态的影响尚需科学评测。【目的】高通量测序技术可用于精确分析土壤微生物群落,从细菌、真菌群落结构和多样性的角度阐释枯草芽胞杆菌菌肥对有机农田根区土壤微生物群落的影响。【方法】在有机农田轮作种植条件下,施用枯草芽胞杆菌菌肥后提取冬瓜根区土壤基因组DNA,通过PCR扩增建立文库,利用IlluminaMiSeq高通量测序技术,并结合相关生物信息学方法分析土壤细菌16SrRNA基因V3-V4区和真菌ITS1区的多样性指数及群落结构;测定根区土壤化学性质及酶活性,分析有机冬瓜果实品质,并作相关分析。【结果】从6个有机冬瓜根区土壤样本中获得14199个细菌操作分类单元(OTU)和3378个真菌OTU,细菌和真菌文库测序覆盖率分别在98%、99%以上。枯草芽胞杆菌菌肥会在一定程度上提高土壤细菌种群多样性而降低真菌种群多样性,丰富了细菌群落结构,但显著降低了真菌群落丰富度(P0.05);并减少了根区土壤特有细菌和真菌物种。变形菌门、厚壁菌门和放线菌门是优势细菌,子囊菌门是优势真菌;枯草芽胞杆菌菌肥会提高绿弯菌门和子囊菌门的相对丰度,比例分别为46.23%、10.01%;降低变形菌门和担子菌门的相对丰度,比例分别为11.14%、74.72%。枯草芽胞杆菌菌肥显著降低了土壤pH,显著提高了有机冬瓜果实总氨基酸、可溶性固形物等营养成分含量(P0.05)。【结论】施用枯草芽胞杆菌菌肥改变有机冬瓜根区土壤细菌和真菌的丰富度和多样性,降低了土壤pH,提高了有机冬瓜果实品质。  相似文献

通过人外文献和市场的调查,并结合科研实践,指出了当前我国微生态制剂发展中存在的一些严峻问题,在介绍了人体肠道微生态规律、微生态制剂应有的质量标准,以及比较了多种产品的活菌含量和“亿活菌价”的基础上,作者认为,提高我国微生态制剂质量的4个关键应该是:选育优良菌种,提高活菌含量,延长保质期和添加有益菌的生长促进剂。为促进我国微生态制剂事业更快、更稳健地发展,作者提出了7项具体建议。  相似文献

好氧反硝化菌强化生态浮床对水体氮与有机物净化机理   总被引:2,自引:0,他引:2  
好氧反硝化菌强化生态浮床通过挂膜将异养硝化-好氧反硝化菌与填料结合,并添加曝气强化措施,提高浮床对富营养化水体中氮和有机物的去除效果。结果表明,在7天内,好氧反硝化菌强化生态浮床在COD_(Cr)/TN=1.7~2.9的进水下,对NH_4~+-N、NO_3~--N、TN和COD_(Cr)的去除率分别为54.5%、100%、59.8%和56.3%,对TN和COD_(Cr)的去除率比传统生态浮床分别提高了47.1%和36.3%;在COD_(Cr)/TN=3.3~5.0的进水下,对NH_4~+-N、NO_3~--N、TN和COD_(Cr)的去除率分别为55.4%、100%、48.9%和65.6%,对TN和COD_(Cr)的去除率比传统生态浮床分别提高了21.0%和20.0%。好氧反硝化菌强化生态浮床中,曝气和异养硝化-好氧反硝化菌的添加提高了浮床对有机物的降解,增强了其在高溶解氧水平下的反硝化作用,克服了曝气生态浮床对NO_3~--N降解效果差的缺点。利用Monod动力学方程对好氧反硝化菌强化生态浮床初期降解氮素和有机物过程进行描述,相关系数为0.9,说明Monod动力学方程适用于描述好氧反硝化菌强化生态浮床的反应动力学。  相似文献

闽江口-平潭海域有机解磷菌多样性及群落特征   总被引:1,自引:0,他引:1  
有机磷的微生物矿化是海洋磷循环的重要环节,开展有机解磷菌研究有助于揭示富营养化海域有机磷矿化的微生物驱动机制。以phoX为标志基因,采用Illumina高通量测序技术分析了闽江口-平潭海域2019年4月(春季)及7月(夏季)有机解磷菌的多样性及群落特征。结果表明: 表层海水样品中有机解磷菌的Shannon指数介于3.21~7.91,各站位多样性春季均大于夏季;沉积物样品中有机解磷菌的Shannon指数介于2.04~8.70,各站位多样性夏季大于春季。春季各站位表层海水有机解磷菌的Shannon指数均高于沉积物,而夏季则相反。表层海水中检测到9个门类的有机解磷菌,主要为变形菌门和蓝细菌门;沉积物中则检测到12个门类,主要为变形菌门和拟杆菌门。在属水平上,有机解磷菌的群落组成呈现时空变异特征。表层海水样品中,春季以雷辛格氏菌属、褐杆菌属、深海球菌属和假单胞菌属等属为主,夏季则以聚球藻属、Halioglobus属、玫瑰变色菌属、褐杆菌属、亚硫杆菌属和生丝单胞菌属等属为主。在沉积物样品中,春季主要菌属包括雷辛格氏菌属、褐杆菌属、弧菌属和亚硫杆菌属;夏季主要菌属包括固氮螺丝菌属、氨基杆菌属、Sulfurifustis属、伯克氏菌属和Thiohalobacter属。此外,在表层海水和沉积物样品中均检测到大量未分类的有机解磷菌。冗余分析显示,溶解氧、水温、pH、可溶性无机氮、亚硝态氮、硝态氮等对闽江口-平潭海域表层海水有机解磷菌群落分布影响较大。表层海水和沉积物中存在的丰富有机解磷菌可能在该海域的磷循环中发挥重要作用。  相似文献

采用物理和化学分组方法研究了不同菌渣施用处理下柑橘果园土壤团聚体有机碳和惰性有机碳的变化, 旨在为以提高有机碳固定为目的的果园土壤管理措施研究提供科学依据。结果表明, 不同施肥对各级别团聚体影响有差异,有机无机配施增加了大团聚体(>250 m)的含量。团聚体内有机碳含量表现为随着粒径增大而增加的趋势, 且大团聚体的有机碳库量表现为随菌渣施用量增加而增加的趋势, 但短期内施用有机肥并没有显著性地提高土壤有机碳库中大团聚体有机碳的比例。施有机肥处理的柑橘园土壤惰性有机碳含量和微团聚体有机碳库量分别比不施肥和单施化肥处理的提高23.77-44.27%和22.78-43.12%、3.67-9.24%和1.80-7.28%, 说明有机无机肥配施可以增加果园土壤有机碳的积累。  相似文献

测定蝇蛆和蝇蛹作为饲料添加剂对清远良种鸡肉质和风味的影响,为家蝇Musca domestica L.作为添加饲料改善鸡的肉质和风味,以及代替鱼粉成为新的蛋白质来源提供数据。150只清远鸡雏鸡随机等分成5组,以鱼粉或商业饲料加入不同比例的家蝇幼虫或蝇蛹作为添加饲料加以饲养。结果显示,饲喂不同食料的清远鸡的增重、脾重和法氏囊重影响差异不显著,但饲喂蝇蛆和蝇蛹的鸡群的死亡率明显低于仅饲喂鱼粉或商业饲料的鸡群;饲喂蝇蛆或蝇蛹的鸡肉的系水力(用失水率表示)和风味评价明显提高,氨基酸含量差异显著。说明家蝇幼虫作为动物蛋白可提高鸡的抗疾病能力及改善鸡肉风味。  相似文献

AIMS: To evaluate an improved bioluminescent enzyme immunoassay (BEIA) using biotinylated firefly luciferase for the rapid detection of Salmonella in naturally contaminated chicken meat samples. METHODS AND RESULTS: Capture agents and lipopolysaccharide (LPS) extraction reagents for Salmonella were investigated to improve the sensitivity of the BEIA. Also, the use of Oxoid SPRINT (Simple Pre-enrichment and Rapid Isolation New Technology) as a pre-enrichment and selective medium for 26-h BEIA detection of Salmonella in chicken meat samples was examined. The use of polymyxin B as a capture agent on solid support and 3-[(3-Cholamidopropyl) dimethylammonio] propanesulfonic acid (CHAPS) for extraction of the LPS facilitated sensitive detection of Salmonella. Of 120 chicken meat samples, 25 samples were positive using the improved BEIA with the SPRINT and 25 samples were positive using the SPRINT followed by the standard isolation methods. CONCLUSIONS: The improved BEIA, in which polymxin B was used as a capture agent and CHAPS was used for extraction of the antigen, had a sensitivity of 96.0% and a specificity of 98.9% for the detection of Salmonella in chicken meat. SIGNIFICANCE AND IMPACT OF THE STUDY: The improved BEIA combined with the SPRINT medium for the detection of Salmonella in chicken meat samples produced comparable results to the culture methods in 26 h.  相似文献

为了建立原代鸡胚细胞的传代培养工艺,探究传代鸡胚细胞对麻疹病毒的敏感性和适应性,本研究将原代鸡胚细胞进行传代培养,分别采用原代鸡胚细胞和传代鸡胚细胞培养麻疹病毒沪-191(Shanghai-191,S-191)株毒种,并对病毒收获液进行滴度检测和基因序列测定。结果显示,原代鸡胚细胞可稳定传代培养至第10代,各代次细胞生长趋势相似;第5代鸡胚细胞染色体检查为正常染色体核型;第8代鸡胚细胞成瘤性检查未见成瘤;采用第3、5代鸡胚细胞制备的麻疹病毒滴度水平高于原代鸡胚细胞,但无显著性差异(n=3,P>0.05),编码病毒核蛋白(nucleoprotein,N)和血凝素蛋白(hemagglutinin,H)的基因序列与S-191株完全一致,未发生变异。本研究证实,原代鸡胚细胞可进行传代培养,各代次鸡胚细胞对麻疹病毒的敏感性不变,产毒水平无显著差异,可用于培养麻疹病毒。  相似文献

Incubation of campylobacter selective broth at 37°C for 48 h followed by selective plating and incubation at 43°C improved significantly the isolation rate of Campylobacter jejuni from naturally contaminated samples of river water and artificially contaminated samples of raw milk. Myostatin (MSTN) is a potent negative regulator for skeletal muscle growth, and binds to activin type IIB receptor (ActRIIB) for its cellular signal transduction. Administration of the extracellular domain of ActRIIB (ActRIIB-ECD) improved skeletal muscle growth in laboratory animals, suggesting that ActRIIB-ECD can be a useful pharmacological agent to improve skeletal muscle growth of meat-producing animals. In the current study, pig and chicken ActRIIB-ECDs were produced in the Pichia pastoris GS115, and the recombinant proteins were purified from induced culture media by Ni-NTA affinity chromatography. The digestion of pig and chicken ActRIIB-ECDs with PNGase F and glycoprotein staining demonstrated an N-linked glycosylation of these recombinant proteins. Glycoprotein staining also indicated an additional presence of glycosylation in chicken ActRIIB-ECD. Both the pig and chicken ActRIIB-ECDs were shown to inhibit MSTN activity in a reporter gene assay system in vitro. When MSTN-inhibitory potencies were compared by analyzing EC50 values, no difference in MSTN-inhibitory potency was observed between the glycosylated and N-deglycosylated forms of pig or chicken ActRIIB-ECD, suggesting that glycosylation does not affect the bioactivity of ActRIIB-ECD. MSTN-inhibitory potency of chicken ActRIIB-ECD was greater (P < 0.01) than that of pig ActRIIB-ECD. Results of this study demonstrate that bioactive pig and chicken ActRIIB-ECDs can be produced from P. pastoris. In addition, the study indicates that the N-glycosylation status of ActRIIB-ECD does not affect its bioactivity in vitro.  相似文献

探讨不同有机肥对西北砂田西瓜生长、产量、品质和养分吸收利用的影响,为砂田西瓜合理施肥、优质高效生产提供理论依据.本试验以单施化肥为对照,在氮、磷、钾等养分条件下,设置了牛粪、鸡粪、羊粪、猪粪4个处理,测定了西瓜不同生育时期生长指标、产量、品质及养分吸收积累量.结果表明: 施用牛粪、鸡粪和猪粪有机肥较单施化肥可促进西瓜营养生长和氮、钾养分的运转,显著提高西瓜产量和养分的吸收积累量,其中西瓜产量较化肥处理分别显著提高了27.4%、31.6%和30.2%,氮素积累量分别提高了26.3%、39.8%和47.4%,磷素积累量分别提高了49.3%、48.3%和55.9%,钾素积累量分别提高了35.8%、41.6%和51.9%.猪粪处理的西瓜品质最优,中心、边缘可溶性固形物含量较化肥处理分别显著提高了5.5%和11.6%,Vc含量提高了19.9%.羊粪处理各指标与化肥处理间差异不显著,甚至略低于化肥处理.综上,砂田西瓜种植应优先选用猪粪和鸡粪有机肥,避免或减少施用羊粪有机肥.  相似文献

1. The effects of avian and mammalian cytokines on avian lipid metabolism were compared using cultured chicken hepatocytes and adipocytes. 2. Conditioned medium from an endotoxin-stimulated chicken macrophage cell line was used as a source of chicken cytokines. Incubation of chicken adipocytes with conditioned medium greatly decreased their lipoprotein lipase activity. 3. Inhibition of lipoprotein lipase synthesis in similar experiments in mammals has been attributed to the effects of TNF-alpha and/or IL-1, but recombinant human TNF-alpha and IL-1 had no effect on lipoprotein lipase activity in chicken adipocytes. 4. Conditioned medium from chicken macrophages produced a 2-fold increase in lipogenesis in chicken adipocytes but had no effect on lipogenesis in chicken hepatocytes. 5. The results point to major differences between mammals and birds in the way that lipid metabolism responds to cytokines and provide further evidence that mammalian cytokines are ineffective in birds.  相似文献

目的:探讨益生菌地衣芽孢杆菌饲喂鸡腹泻的治疗效果以及分子机理。方法:从养鸡场土壤中选择性分离芽孢杆菌,通过优 化条件提高产生芽孢的比例;利用芽孢和大米混合后饲喂腹泻鸡,通过克隆测序分析腹泻鸡肠道微生物群落在饲喂前后的差异。 结果:我们分离到一株地衣芽孢杆菌,通过优化条件使产芽孢的比例提高到60 %。利用芽孢饲喂一周后能够有效地治疗鸡腹泻。 分析鸡肠道微生物群落发现:鸡腹泻后其肠道细菌群落的多样性和均匀度降低。造成腹泻鸡肠道主要菌群为动胶杆菌(Zoogloea sp.);经芽孢饲喂后,主要菌群转变为拟杆菌(Bacteroides sp.),与健康鸡的肠道微生物主要菌群相似。结论:我们筛选到一株可能 治疗鸡腹泻的芽孢杆菌,通过优化培养条件能够提高其产芽孢比例;腹泻鸡经芽孢饲喂后,其肠道中致病菌被益生菌所代替,形 成正常的肠微生物菌群而恢复健康。  相似文献

Little is known about the effects of Cholesterol-Loaded Cyclodextrin (CLC) on post-thaw semen quality in chicken. The aim of the present study is to investigate the efficacy of CLC levels (0, 1, 2 and 3 mg/mL Schramm diluent) on post-thawed semen quality and fertility in two breeds of chicken Pradu Hang Dum (native chicken) and Rhode Island Red. Semen samples of each breed were pooled, divided into 4 aliquots and diluted with Schramm diluents, cooled to 5 °C when DMF was added (6% of final volume). Semen straws were subjected to cryopreservation using the liquid nitrogen vapor method. Post-thawed sperm motility, viability, acrosome integrity, mitochondrial function, and the Malondialdehyde (MDA) level were determined. The fertility of frozen semen was tested by inseminating laying hens. Post-thaw motility between Pradu Hang Dum and Rhode Island Red was no different; but Rhode Island Red had a higher semen viability and live cell intact acrosomes than Pradu Hang Dum (P < 0.05). The percentage of high functioning mitochondria in the Pradu Hang Dum was higher than the Rhode Island Red. CLC at 2 and 3 mg/mL supplementation was associated with improved viability of frozen semen; that is, acrosome integrity and mitochondrial function (P < 0.01), albeit having no effect on MDA levels. The sperm with 1 mg/mL CLC yielded a significantly better fertility (P < 0.01). CLC (1 mg/mL) improved the quality of frozen rooster semen. There was no interaction among breeds and CLC on post-thaw semen quality and fertility.  相似文献

Glutamate Decarboxylase Activities in Single Vertebrate Neurons   总被引:5,自引:3,他引:2  
An enzymatic microassay method for glutamate decarboxylase (GAD) and gamma-aminobutyric acid (GABA) was improved to a degree yielding high sensitivity and low blank. Single cell bodies of anterior horn cells and dorsal root ganglion cells were dissected out from the freeze-dried sections of rabbit and chicken spinal cords and Purkinje cell bodies from those of rabbit cerebellum. A minute amount of GABA, present in single neurons or synthesized by GAD in single neurons, was enzymatically converted to NADPH. The NADPH was amplified 10,000-350,000-fold and measured, using an enzymatic amplification reaction (NADP cycling). GAD was contained in all Purkinje cell bodies and its average activity was four- to fivefold higher than those of the molecular and granular layers of rabbit cerebellum. The GABA concentration was threefold higher in Purkinje cell bodies than in these layers. GAD activity, at a level similar to that in the cerebellar layers, was found in almost all the cell bodies of anterior horn cells from rabbit and chicken. GABA was detected in 40% of rabbit neurons and not in chicken neurons. Dorsal root ganglion cells from both species contained no measurable GAD or GABA.  相似文献
