During metamorphosis of the blowfly Calliphora erythrocephala extracellular ribosomes, in the form of monosomes, appear in the body fluid. The total number of ribosomes, i.e. intracellular and extracellular, remains approximately constant during this period, whereas the proportion of extracellular ribosomes first rises, plateaus, and then declines in a manner suggesting that their appearance is a result of larval tissue breakdown.  相似文献

On the basis of in vitro observation of live cells and examination of stained slides of larval and prepupal Calliphora vicina hemolymph, seven types of hemocytes have been detected: prohemocytes, stable and unstable hyaline cells, thrombocytoids, spindle cells, larval plasmatocytes, and plasmatocytes I-IV, a. The last representing sequential stages of one cell line differentiation. Prohemocytes are basic cells, from which other forms of hemocytes derive outside the hemopoietic tissue, i.e. in free hemolymph. At the last larval instar, three waves of hemopoiesis occur. Either wave tends to increase the general number of cells and to change the quality of hemocyte population. The first wave occurs at the close of larva feeding and is accompanied by increase in the number of hyaline hemocytes, thrombocytoids and larval plasmatocytes. The second wave of hemopoiesis occurs after the larva's crop emptying. In this period the main increase of hemocyte population occurs at the expense of prohemocytes and plasmatocytes I. The most significant (five-fold) explosion of the population of free hemocytes takes place at the onset of pupariation and correlates with the rise of ecdysone titer. At the first stage of this peak, the amount of plasmatocytes I sharply increases. Further on these are rapidly differentiated into plasmatocytes II and III. After the puparium formation, hemocytes are reduced in number. Plasmatocytes III phagocytose fragments of destroyed larval tissues, pass to the stage of plasmatocytes IV (macrophages), and partially settle on tissues.  相似文献

Rapid microspectrofluorometry has been used to evaluate 1-pyrene-butyric acid as an oxygen probe in single living EL2 ascites tissue culture cells. Despite instrumental conditions preventing detection of the pyrene butyric acid maxima at 380 and 400 nm, the probe having penetrated the cell can be easily identified (maximum around 440 nm in unconnected spectra) from the fluorescence emission spectrum, as compared with NAD(P)H emission in controls (maximum around 460 nm). Fluorescence changes during gradually increasing anaerobiosis under nitrogen flow, are compatible with a linear relationship between the reciprocal of the fluorescence intensity and the intracellular oxygen concentration (increase in 430, 434, 442/461 nm ratios at anaerobiosis). The cells having absorbed the probe continue to catabolize glycolytic substrate, but some inhibition is noticeable (e.g. from the amplitude of the NAD(P)H fluorescence increase spectrum due to intracellular addition of glucose-6-P). In principle rapid microspectrofluorometry allows a multiprobe (e.g. 1-pyrene-butyric acid for oxygen, vs NAD(P)H for metabolism) exploration of the living cell.  相似文献

Ecdysteroids, the molting hormones of arthropods, act like vertebrate steroid hormones by binding to an intracellular receptor protein. We have recently isolated a protein from nuclei of blowfly larvae which has satisfied the requirements of an ecdysteroid receptor. The receptor was partially purified and its ecdysteroid-binding properties were characterized. The availability of receptor preparations which have been stabilized by partial purification now enables us to study the general DNA-binding properties of ecdysteroid receptors. DNA-binding characteristics of ecdysteroid receptors were studied with calf thymus DNA. Affinity for DNA was observed both in the presence and in the absence of steroid ligand but the ligand clearly enhanced binding of receptors to DNA. Receptor preparations contained a heterogeneous mixture of receptors; up to 25% of DNA-binding receptors, and nonbinding forms of ecdysteroid receptors. The ability to bind to DNA was subject to inactivation which was not affected by partial purification, but which could be decelerated by dilution of the receptor preparation. Thus, dilution resulted in a spurious activation of DNA binding. A genuine activation, which would have led to an increase in the percentage of the DNA-binding form of the ecdysteroid receptor complex, was not observed.  相似文献

The attraction of the blowfly Calliphora vicina Robineau‐Desvoidy, 1830 (Diptera: Calliphoridae) to single synthetic compounds, blends and authentic odours was investigated in a wind tunnel. A total of 1850 C. vicina (1750 females and 100 males) were tested. A comparison of male and female responses showed significant differences in attraction between the sexes. Females were more attracted than males to liver odour. The attraction of females lay in the ranges of 0–22% for single compounds, 26–64% for synthetic blends and 58–88% for authentic odours. Dimethyl trisulphide was the most attractive single compound. Significant improvement in attraction was achieved with blends and a three‐component lure, consisting of dimethyl trisulphide, mercaptoethanol and o‐cresol, was found to be the best solution for field trapping of C. vicina. Authentic odours from dead fish and mice were significantly more attractive than liver and the three‐component blend, and the blend and liver were similarly effective as attractants. Field tests support the results of the wind tunnel study and a high number of C. vicina were caught in funnel traps. Overall, 99.1% of the specimens caught were females.  相似文献

An improved method for the purification of prophenoloxidase is described. The proenzyme was purified 400 fold in homogenous form. The purity was tested by disc-electrophoresis and the molecular weight was found to be 87 000 in comparison to the mobility of marker enzymes, which were run simultaneously in SDS-gel electrophoresis. The proenzyme was denatured at 80 degrees C and maximum conversion into active state was found between 40 and 50 degrees C.  相似文献

Summary The stage-specific appearance of calliphorin in cuticles of Calliphora vicina was analysed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting. The fate of the protein, injected into last instar larvae, was pursued by autoradiography of histological sections. Fractionation of sclerotized pupal cuticle in buffer-soluble, urea-soluble and NaOH-soluble fractions shows that calliphorin forms covalent and non-covalent links with other cuticle components. Calliphorin traverses the epidermal cells and enters the cuticle in an undegraded state and appears to be an important constituent of the sclerotizing system.  相似文献

Calliphora vicina Robineau‐Desvoidy (Diptera: Calliphoridae) causes yearly losses of 1–2 million Euros to the stockfish industry in Lofoten, Norway. To develop an efficient management program, knowledge of its life cycle and phenology in production areas is needed. Cohort studies in a simulated Lofoten climate showed that field abundance peaks of adults in early spring and midsummer can be explained by a cohort originating from stockfish and its subsequent generations. Laboratory simulations with normal, increased, and decreased Lofoten temperatures indicate that C. vicina overwinter as a mix of larvae, pupae, and adults, and a temperature change of ± 2 °C significantly influences reproductive timing, reproductive output, and female mortality. Flies originating from stockfish reproduced during the first summer when temperatures were increased 2 °C above normal. At lower temperatures, the reproductive investment was low or absent during the first summer and the adult flies entered the winter in a diapausing state. Most offspring produced during the first summer and autumn developed continuously without maternally induced diapause, pupated during the winter, and hatched in the early spring to co‐occur with their parent generation during stockfish production. Calliphora vicina showed flexibility in reproductive efforts and overwintering strategies. The high proportion of adults overwintering compared with the commonly used larval diapause strategy might be interpreted as an adaptation to exploit the stockfish resource. The majority of female C. vicina that cause damage to stockfish likely developed on fish dried the previous year, and a continuous year‐long trapping is recommended to decimate the population.  相似文献

Summary We have used single electrode voltage clamp in the intact animal and whole-cell recording from dissociated cell bodies to investigate the properties of potassium conductances in large monopolar cells (LMCs) of the first optic ganglion of the blowfly Calliphora vicina. Two classes of voltage gated potassium conductances were found: a delayed rectifier current (Kd) with slow inactivation (inac = 1–3 sec), and an A current (Ka) showing both faster inactivation (inac = 21 ms) and also more rapid activation. The reversal potential of both currents is ca. -90 mV with 2 mM [Ko] and 140 mM [Ki], and follows the Nernst slope with increasing [Ko]. The voltage operating range of Ka is unusually negative, with the mid point of the steady-state inactivation curve (V50) at- 101 mV. V50 for Kd is - 84 mV. Although no inward currents were detected, for technical reasons their presence cannot be excluded.In inside-out patches from LMC soma membranes the single channels underlying the currents both have a conductance of ca. 20 pS in symmetrical 140 mM K solutions and channel densities may be as high as 10/m2. Less frequently, inside-out patches contained a large conductance (110 pS) calcium-activated potassium channel which existed almost exclusively in a rapidly flickering mode. Open probability increased with depolarization and Ca concentrations greater than 40 nM.In whole-cell recordings, dissociated LMC cell bodies fall into two classes with respect to their voltage sensitive currents: 37 % of cells only showed Kd; the remainder (63%) were dominated by Ka with a variable (0–30%) contribution from Kd. In the intact animal, intracellular recordings from LMCs, combined with dye-marking, indicate that cells expressing only Kd are type L3 cells, whilst L1 and L2 express predominantly Ka. Since L1 and L2 have resting potentials of ca. - 40 mV and maximum hyperpolarizations reaching -90 mV only transiently, inactivation of Ka is unlikely to be removed under most physiological conditions. In contrast, L3 cells have a more negative resting potential (–60 mV) and Kd should play a significant role in signal-shaping, in particular contributing to the falling phase of a prominent spike-like transient in response to dimming.Abbreviations Ka A current - Kd delayed rectifier - LMC large monopolar cell - L1-L3 classes thereof - TTX tetrodotoxin  相似文献

In late larvae of the blowfly, Calliphora vicina, arylphorin and LSP-2 proteins, which belong to the class of hexamerins, are selectively taken up by the fat body from the haemolymph. Hexamerin endocytosis is mediated by a specific membrane-bound receptor, the arylphorin-binding protein (ABP). Using the two-hybrid technique, we found that the anterior fat body protein (AFP) interacts with the hexamerin receptor. AFP, a homologue of the mammalian calcium-binding liver protein regucalcin (senescence marker protein-30), exhibits a strong binding affinity for a naturally occurring C-terminal cleavage fragment of the hexamerin receptor precursor (the P30 peptide) and other receptor cleavage products that contain P30. Expression of AFP mRNA and protein is restricted to the anterior part of the fat body tissue and to haemocytes in last-instar larvae. AFP mRNA occurs in all postembryonic developmental stages. Our results suggest that AFP plays a role in the regulation of hexamerin uptake by fat body cells along the anterior-posterior axis.  相似文献

S-Antigen (arrestin)-immunoreaction can be considered as a marker for retinal and extraretinal photoreceptors in both vertebrate and invertebrate species. The present immunocytochemical study with the blowfly Calliphora vicina revealed S-antigen immunoreaction in retinal photoreceptors and various groups of neurons bilaterally distributed in the optic lobes and in the proto-, deuto- and tritocerebrum. S-Antigen-immunoreactive processes and terminal formations were found in the lower division of the central body complex and in the neuropil of the mushroom body. Also neuropil regions of the optic lobe, the lamina, medulla and lobula displayed S-antigen-immunoreactive fibers which were arranged in different patterns. These immunocytochemical data suggest that extraocular photoreceptors may be located in various parts of the blowfly brain. They provide a structural basis for further experiments which are needed to identify definitely these elements as extraretinal photoreceptors.  相似文献

The brush border of the larval midgut and hindgut in Calliphora vicina (Diptera : Calliphoridae) shows a positive staining with a rabbit antiserum directed against pig villin. This protein has been extensively studied in chicken and in several mammals; it is known to bind together the actin filaments of the microvillar cytoskeleton. The presence of a villin-like protein in insect microvilli has never been reported. The immunodetection of villin in both hind- and midgut points out that villin exists in ectodermal tissues as well as in endodermal or mesodermal ones. Moreover, the “insect villin” seems to present the polypeptidic sequence of 13 amino acids, which has been found in both mammalian and avian villin. That may indicate that villin is a conserved molecule in several taxa.  相似文献

Blowfly larvae (Diptera: Calliphoridae) fulfil an important ecological function in the decomposition of animal remains. They are also used extensively in forensic entomology, predominantly to establish a minimum time since death, or a minimum post-mortem interval, using the larval length as a 'biological clock'. This study examined the larval growth rate of a forensically important fly species, Calliphora vicina Robineau-Desvoidy (Diptera: Calliphoridae) at temperatures of between 4 degrees C and 30 degrees C, under controlled laboratory conditions. The laboratory flies had been trapped initially in London, U.K. The minimum developmental temperature was estimated to be 1 degrees C and 4700 accumulated degree hours (ADH) were required for development from egg hatch to the point of pupariation. Lines fitted to the laboratory larval growth data were found to adequately explain the growth of larvae in the field. The nature of variation in growth rates from geographically isolated populations is discussed.  相似文献

Field experiments conducted in the environs of St. Petersburg (Russia) with a local population of Calliphora vicina showed that induction of larval diapause under natural conditions was significantly dependent on day lengths and temperature. The maternal photoperiodic response had a distinct threshold: the first diapausing larvae hatched from the eggs laid in the middle of August when the day length was 16 h; at shorter photoperiods, the fraction of diapausing larvae depended only on temperature. At the mean temperature of 16°C, larval diapause was rarely recorded; at 12–13°C, about 50% of the larvae entered diapause; at 7–9°C, nearly all the larvae entered diapause. These results of the field experiments agree well with the parameters of photoperiodic and thermal responses studied in the laboratory at constant temperatures and photoperiods.  相似文献

Open field experiments with the blowfly, Calliphora vicina originating from the environs of St. Petersburg showed that the correlation between the rate of its preimaginal development under the natural conditions and the mean daily temperature could be very closely approximated by linear regression. The sum of effective temperatures required for development from the egg to the puparium constituted ca 140 degree-days and from the egg to the adult, ca 320 degree-days with the lower thresholds of 5.8 and 4.8°C, respectively. The minimum duration of development (6–8 days from the egg to the puparium and 16–20 days from the egg to the adult) was recorded during the period of the highest mean temperatures (22–23°C) from July 8 to August 15 of 2010 and 2011. The rate of development under natural thermorhythms was not siggnificantly different from that under the laboratory conditions at constant temperatures from 12 to 23°C.  相似文献
