Production of the indole alkaloids, ajmalicine or catharanthine, in cell suspension cultures of Catharanthus roseus was enhanced by cerium (CeO2 and CeCl3), yttrium (Y2O3) and neodymium (NdCl3). The yield of ajmalicine in these treated-cultures reached 51 mg l–1 (CeO2), 40 mg l–1 (CeCl3), 41 mg l–1 (Y2O3) and 49 mg l–1 (NdCl3) while catharanthine production reached to 36 mg l–1 (CeO2) and 31 mg l–1 (CeCl3). A major portion of increased alkaloids was released into medium in these treatments. But Sm2O3, SmCl3, La2O3, LaCl3, complex of chromium (III)-titanium (IV) and NaSeO4 treatments had little effect on alkaloid production of C. roseus cell cultures.  相似文献

A two-stage process for enhanced ajmalicine production in elicited Catharanthus roseus cell cultures was developed in shake-flasks and a bioreactor. By using combined elicitor treatment of an Aspergillum niger mycelium and tetramethyl ammonium bromide, yields of ajmalicine were 48 mg l–1, 52 mg l–1 and 33 mg l–1, respectively in 500-ml flasks, 1000-ml flasks and a 20-l airlift bioreactor. The peroxidase and superoxide dismutase activities decreased in elicited cell cultures but catalase and lipoxygenase activities increased in these cultures. The combined elicitor treatment also caused a significant increase of malondialdehyde content in cell cultures.  相似文献

The production of extracellular enzymes by the thermophilic fungus Thermomyces lanuginosus was studied in chemostat cultures at a dilution rate of 0.08 h–1 in relation to variation in the ammonium concentration in the feed medium. Under steady state conditions, three growth regimes were recognised and the production of several extracellular enzymes from T. lanuginosus was recorded under different nutrient limitations ranging from nitrogen limitation to carbon/energy limitation. The range and the production of carbohydrate hydrolysing enzymes and lipase increased from Regime I (NH4Cl 600 mg l–1) to Regime III (NH4CI 1200 mg l–1), whereas production of protease was highest in Regime II (600 mg l–1 < NH4Cl <1200 mg l–1).  相似文献

Saffron callus was grown in a two-stage culture on B5 medium supplemented with casein hydrolysate (300 mg l–1) at 22 °C in dark with naphthalene acetic acid (2 mg l–1) and 6-benzyladenine (1 mg l–1) to give maximum biomass (16 g dry wt l–1), and with indole 3-acetic acid (2 mg l–1) and 6-benzyladenine (0.5 mg l–1) for crocin formation. The maximum crocin production (0.43 g l–1) was achieved by this two-stage culture method, which was three times that by a one-stage method.  相似文献

To investigate the production potential of eicosapentaenoic acid (EPA) by the diatom Nitzschia laevis, the growth characteristics and fatty acid composition of the cells were studied under photoautotrophic, mixotrophic and heterotrophic conditions of growth. The specific growth rate and maximum biomass concentration were respectively 0.466 d–1 and 2.27 g l–1 for mixotrophic culture, 0.344 d–1 and 2.04 g l–1 for heterotrophic culture, and 0.167 d–1 and 0.5 g l–1 for photoautotrophic culture, respectively. As for EPA production, the yield and productivity were respectively 52.32 mg l–1 and 10.46 mg l–1 d for mixotrophic culture, 35.08 mg l–1 and 6.37 mg l–1 d for heterotrophic culture, and 6.78 mg l–1 and 3.39 mg l–1 d for photoautotrophic culture, respectively. Results suggest that mixotrophic culture is the most suitable growth mode for the production of EPA by the diatom Nitzschia laevis. The results are useful for the development of a cost-effective fermentation process for EPA production by Nitzschia laevis.  相似文献

Glycerol at 10–20 g l–1 increased clavulanic acid production by Streptomyces clavuligerus in shake-flask culture. The biosynthesis of clavulanic acid continued for longer by feeding glycerol and production increased to 250 mg l–1 compared with 115 mg l–1 without feeding. In fermenter batch culture, degradation of clavulanic acid began after 72 h. With glycerol feeding in fed-batch culture, clavulanic acid production was not only increased further to about 280 mg l–1 but also remained stable up to 130 h.  相似文献

Spirulina platensis was cultivated, in comparative studies, using several sources of nitrogen. The standard source used (sodium nitrate) was the same as that used in the synthetic medium Zarrouk, whereas the alternative nitrogen sources consisted of ammonium nitrate, urea, ammonium chloride, ammonium sulphate or acid ammonium phosphate. The initial nitrogen concentrations tested were 0.01, 0.03 and 0.05 M in an aerated photobioreactor at 30 °C, with an illuminance of 1900 lux, and 12 h-light/12 h-dark photoperiod over a period of 672 h. Maximum biomass was produced in medium containing sodium nitrate (0.01–0.03–0.05 M), followed by ammonium nitrate (0.01 M) and urea (0.01 M). The final biomass concentrations were 1.992 g l–1 (0.03 M sodium nitrate), 1.628 g l–1 (0.05 M sodium nitrate), 1.559 g l–1 (0.01 M sodium nitrate), 0.993 g l–1 (0.01 M ammonium nitrate) and 0.910 g l–1 (0.01 M urea). This suggested that it is possible to utilize nitrogen sources other than sodium nitrate for growing S. platensis, in order to decrease the production costs of scaled up projects.  相似文献

The sedative triterpene, galphimine B (1), was detected in cell suspension-batch cultures of Galphimia glauca. The effect of inoculum size, growth regulators and different concentrations of sucrose, nitrates and phosphates was evaluated. A two-stage batch process for biomass production and accumulation of compound 1 was established. Major cellular growth (15 g l–1 dry wt) was obtained in the first stage with naphthaleneacetic acid (2 mg l–1) + kinetin (2 mg l–1). Adding 4 mg 2,4-dichlorophenoxyacetic l–1 acid in the second stage resulted in the highest accumulation of 1 (0.21 mg g–1 dry wt) which was 36% higher with respect to calluses and comparable to that obtained from wild plants.  相似文献

Addition of 5 mg abscisic acid l–1 after 12 days' growth of Taxus chinensis suspension culture gave the greatest paclitaxel accumulation at 11 mg l–1, which was almost 5 times that of the control culture. Fungal biotransformation of p-coumaric acid into caffeic acid, potentially a strong antioxidant, was evidenced in Pycnoporus cinnabarinus cultures grown with high feeding of p-coumaric acid. Preliminary experiments showed no toxicity of both p-coumaric and caffeic acids at concentrations ranging from 0 to 500 mg l–1. Feeding 450 mg p-coumaric acid l–1 into P. cinnabarinus cultures grown on 20 g l–1 glucose medium resulted in the production of 257 mg caffeic acid l–1with a molar yield of 21%.  相似文献

An in vitro highly efficient plant regeneration system was established from hypocotyl segments in buckwheat (Fagopyrum esculentum Moench.). Calli were induced on Murashige–Skoog (MS) medium containing 1.0 mg l–1 to 2.0 mg l–1 2,4-dichlorophenoxyacetic acid and 1.5 mg l–1 6-benzylaminopurine. Shoot buds were formed on subcultured pieces of callus. A high frequency (over 80%) of shoot differentiation was obtained on MS medium supplemented with 2.0 mg l–1 6-benzylaminopurine and 1.0 mg l–1 6-furfurylaminopurine. The regenerated shoots rooted readily on MS medium plus 0.2 mg l–1naphthaleneacetic acid and 0.2 mg l–1 indole butyric acid. The regenerated plantlets were acclimatized and successfully transferred to pots. Chromosome examination showed that the regenerated plants had normal chromosome number (2n=16).  相似文献

Pseudomonas oleovorans was cultivated to produce medium chain length polyhydroxyalkanoates (MCL-PHAs) from octanoic acid and ammonium nitrate as carbon and nitrogen source, respectively, by a pH-stat fed-batch culture technique. The octanoate in the culture broth was maintained below 4 g l–1 by feeding the mixture of octanoic acid and ammonium nitrate when the culture pH rose above 7.1. The final cell concentrations of 63, 55 and 9.5 g l–1, PHA contents of 62, 75 and 67% of dry cell wt, and productivities of 1, 0.63 and 0.16 g l–1 h–1 were obtained when the C/N ratios in the feed were 10, 20 and 100 g octanoic acid g–1 ammonium nitrate, respectively.  相似文献

A procedure for rapid in vitro multiplication of Tylophora indica (Burm. f.) Merrill., an important indigenous medicinal plant, has been developed. Addition of ascorbic acid was essential to induce sprouting of axillary buds. Optimum multiplication was observed on MS medium containing 6-benzylamino purine (5.0 mg l–1), -naphathalene-acetic acid (0.5 mg l–1) and ascorbic acid (100 mg l–1). Rooting of in vitro produced shoots was readily achieved with indole-3-acetic acid alone (1.0 mg l–1) in MS. Brevibacterium linens forms hydrolytic enzymes which can be used to accelerate the ripening of cheese without causing bitterness. B. linens ATCC 9172 was grown to a high cell density (50 g dry wt l–1 after 60 h) in a mineral medium containing lactic acid, soy-peptone and ammonium sulphate by applying a continuous feed of nutrients. The maximal activities of l-leucine aminopeptidase and cell-associated proteinase were 286 U l–1 and 202 U l–1, respectively. The cell-associated lipolytic activity exhibited a strong and sudden increase at 46 h, resulting in a maximum of 9.5 U g–1 dry wt; thus the volumetric productivity of proteolytic and lipolytic activity was 4220 U l–1 h–1 and 7.3 U l–1 h–1, respectively.  相似文献

Anthers of Morus indica L., with microspores at the uninucleate stage were cultured; and the influence of temperature and kinetin pretreatment on induction of androgenic calluses was examined. The effects of various pretreatments revealed that 24 h cold pretreatment increased the percentage of cultures inducing callus. First microspore division was observed after 16 to 20 days of culture. Th anthers split and developed embryogenic calluses on MB medium supplemented with NAA (0.5 mg l–1 and BA (1.0 mg l–1)) using 8% sucrose. Rhizogenesis was induced on medium supplemented with NAA and BA (each 0.5 mg l–1) with reduced myo-inositol (75 mg l–1). Cytological study of induced roots confirmed the haploid nature of calluses. Different type of embryos were initiated upon transfer of calluses to medium supplemented with NAA, BA (each 0.5 mg l–1), 2,4-d (1.0 mg l–1) and PVP (600 mg l–1). The influence of maltose and growth regulators on microspore culture response was investigated in japonica rice. High frequency of callus induction of isolated microspores was obtained with liquid medium containing MS salts, 100 mg l–1 myo-inositol, 1 mg l–1 thiamine-HCl, 500 mg l–1 glutamine, 60 g l–1 maltose, and several growth regulators. The effect of maltose on promoting callus formation was associated with keeping a high proportion of swollen microspores after 5 day preculture and increasing the microspore division rate on the 3rd day after culture initiation. No significant effect of maltose in place of sucrose on plantlet regeneration was seen in regeneration medium. Among the growth regulators tested, the combination of auxin 2,4-dichlorophenoxyacetic acid (1 mg l–1), naphthaleneacetic acid (1 mg l–1), and cytokinin (6-benzyl-aminopurine 1 mg l–1) in the medium proved to be much better for callus formation than in the other media, and the percentage of callusing microspores of that medium reached 0.86%. Indole-3-acetic acid (0.5 mg l–1) and kinetin (2 mg l–1) in regeneration medium were beneficial for green plantlet differentiation. The results also showed that the frequencies of microspores initial division, callus formation and green plant regeneration varied among genotypes no matter what kind of growth regulator and sugar were used. Xiushui 117 was the best variety for callusing followed by 02428 & Taipei 309. The effects of concentration of amino acids, nitrate, and ammonium on the growth and taxol production in cultures of cell line TY-21 of Taxus yunnanensis were investigated. Addition of 20 different amino acids each at 15–20 mg l–1 to B5 medium significantly improved callus growth but inhibited taxol formation in the cultures. The optimum nitrate concentration was 20–30 mM for both growth and taxol production. Ammonium greatly suppressed growth but strongly promoted taxol formation in the cells when it was the sole inorganic nitrogen in the medium. Culturing the suspension cells in nitrate-containing medium for 15 days and then in a medium in which ammonium was the sole inorganic nitrogen for 7 days increased taxol yield by 104%, reaching up to 28.1 mg l–1.  相似文献

Taxus chinensis var. mairei was grown in two-liquid-phase culture in a 2.5 l airlift external loop reactor for production of Taxol. A mixture of oleic acid and dibutyl phthalate (1:1, v/v) in a volume ratio of 8:92 to the culture medium gave higher production of Taxol (up to 12 mg l–1) with an aeration rate of 1 vvm. By combination of the in situ extraction, precursor feeding and additional sugar introduction, Taxol production (16.7 mg l–1) was 5 times higher than that in the case without any additives (3.4 mg l–1) and 1.4 times of that in the case with only thein situ extraction (12 mg l–1). Taxol was extracted from the organic solvents with water and methanol (1:1:1.4, by vol.) and lyophilized giving a total yield of ca. 90%.  相似文献

High percentages of micro-calli and micro-derived embryos were produced from isolated asparagus microspores at late uninucleate stage on MS liquid medium supplemented with 1.0 mg l–1 2,4-D and 0.5 mg l–1 BA. Two types of calli, namely compact callus (CC) and loose callus (LC), were found. Plantlets were regenerated via organogenesis, when these calli were transferred onto MS solid medium supplemented with 1.0 mg l–1 BA and 0.2 mg l–1 IBA 6 weeks. Embryos were produced from liquid cultured microspores, or from solid cultured micro-calli. The frequencies of haploid plant production from organogenesis and embryogenesis were compared. Effects of plant growth regulators on callus production, plantlet regeneration, and haploid plant production were tested. Catharanthine production in Catharanthus roseussuspension cell cultures was increased by about 4-fold to 28 mg l–1, 23 mg l–1and 24 mg l–1by adding sodium alginate, mannitol or polyvinyl pyrrolidone, respectively. Sodium alginate and polyvinyl pyrrolidone also enhanced ajmalicine production to 28 mg l–1and 31 mg l–1, respectively. Up to 55–70% of the total alkaloids were released into the medium. These treatments could stimulate higher alkaloid production in C. roseuscell cultures than NaCl and KCl stresses. The possible mechanisms for these treatment effects are discussed.  相似文献
