Young gametophytes of the sensitive fern, Onoclea sensibilis,respond to heat-shock by synthesizing in excess certain proteinsthat are made at normal growth temperature. Enhanced proteinsynthesis occurred during a 2 h heat-shock at a range of temperaturesbetween 38 °C and 50 °C. Although a temperature of 50°C proved lethal, a 5 min pulse at 50 °C resulted inenhanced synthesis of heat-shock proteins which continued forseveral hours at 25 °C. After heat-shock at 50 °C for10 or 15 min, the gametophytes temporarily lost their capacityfor protein synthesis but normal protein synthesis was resumedwithin 24 h of heat-shock. A heat-shock at 38 °C precedingone at 50 °C did not have any protecting effect on the gametophytes.In vitro translation of poly(A)+ RNA isolated from heat-shockedgametophytes yielded several proteins including heat-shock proteins.The results suggest that, rather than activating genes encodingnew messages for the synthesis of stress proteins, heat-shockof gametophytes of O. sensibilis triggers a controlling systemwhich enhances the translation of certain messages that aresynthesized at normal growth temperature. The relationships between ethylene production, aminocyclopropane-1-carboxylicacid (ACC) content and ethylene-forming-enzyme (EFE) activityduring ageing and cold storage of rose flower petals (Rose hybridaL. cv. Gabriella) were investigated. During flower ageing at20 °C there was a climacteric rise in petal ethylene production,a parallel increase in ACC content, but a continuous decreasein EFE activity. Applied ACC increased petal ethylene productionc. 200-fold. During cold storage of flowers at 1 °C therewere parallel increases in petal ethylene production and ACCcontent, to levels greater than those reached in fresh flowersheld at 20 °C. EFE activity decreased during storage. Immediatelyafter cold-stored flowers were transferred to 20 °C ethyleneproduction and ACC levels were c. four times greater than infreshly cut flowers. These levels increased to maximum valuesof two to four times the maximum values reached during ageingof fresh, unstored, flowers. It was concluded that in rose petalsethylene synthesis is probably regulated by ACC levels and thatcold storage stimulates ethylene synthesis because it increasesthe levels of ACC in the petals. The growth of Oikopleura dioica was determined from microcosmsincubated in situ at 29°C in Kingston Harbour, Jamaica.Minimum generation times approached 1 day, with Oikopleura dailyspecific growth rates ranging from 3 to 23 and averaging 10.7-foldincreases in biomass over 24 h. This was an order of magnitudegreater than the copepod Paracalanus crassirostris, whose growthrates ranged from 0.3 to 1.2 and averaged 0.67 day–1 fromthe same experiments A reassessment of previous data indicatesthat larvacean production approaches, and may exceed, the 693kJ m–2 annual production of the copepods. Growth rateand recruitment of Oikopleura decreased as the biomass of thezooplankton community increased; both resource and interferencecompetition are probably occurring. The extraordinary productionpotential of the Larvacea emphasizes their understated importancein marine planktonic systems.  相似文献

Peanut (Arachis hypogaea L. ) seed powder accumulated ATP fromAMP and phosphoenolpyruvate (PEP) at a rate of approx. 100 pmolmin–1mg powder at 35° C. When peanut seed powderwas incubated with various substrates, which may result in PEPor AMP (ADP) synthesis, then ATP accumulated. The best substratecombinations examined so far were AMP + succinate, NADH2, andAMP + malate + NAD, with activities of 33, 12 and 12 pmol minmg–1powder,respectively; AMP + malate showed very low activity. Some combinationsexhibited linear activities with time, while others had an exponential-typeprofile. The temperature dependence of the ATP accumulationdemonstrated by the Ahrrenius plot had a double phase with atransition point at 25° C. The Ea values between 15°C and 25° C were 25 000–50 000 cal/mol, while above25° C the Ea values fluctuated between 6000 and 8000 cal/mol(depending on the substrate). The AMP + PEP combination exhibiteda single-phase profile between 15° C and 40° C, withan Ea value of 22 000 cal/mol. In the presence of some substrates,ethephon (ethylene) had a stimulatory effect and caused an increasein the Ea values at the high temperature phase. A comparisonof seed powder from dormant seeds with that from non-dormantseeds revealed that some substrate combinations accumulate ATPfaster in non-dormant seeds and others do so in dormant seeds. Growth responses of the red tide flagellates, Heterocapsa circularisquama(Dinophyceae) and Chattonella verruculosa (Raphidophyceae),were examined with 36 different combinations of temperature(5–30°C) and salinity (10–35 PSU). Heterocapsacircularisquama did not grow at or below a temperature of 10°C.The maximum growth rate of H.circularisquama (1.3 divisionsday–1) was obtained with a combination of 30°C and30 PSU. In contrast, C. verruculosa did not grow at 10 PSU andat temperatures of 25°C or more. The maximum growth rateof C. verruculosa (1.74 divisions day–1) was obtainedwith a combination of 15°C and 25 PSU. A significant temperature-salinityinteraction on growth was found by factorial analysis. Basedon the physiological characteristics obtained in the presentstudy, these novel flagellates have a potential for future outbreaksof red tides in pre viously unaffected waters.  相似文献

Lolium temulentum plants were grown at 20 °C, under an 8-hdaylength, in a controlled-environment chamber, and the kineticsof leaf expansion were observed by measuring the movement ofan optical grid attached to the fourth leaf. The leaf emerged23–24 d after sowing and was fully expanded 9–10d later. Extension rate was maximal between the second and fifthdays after emergence and declined markedly thereafter. Duringthe rapid growth phase the rate of elongation exhibited a distinctdiurnal rhythm, fluctuating between 1.9 to 2.3 mm h–1in the light period, and 1.3 to 1.7 mm h–1 in the dark.A circadian oscillation with a period of about 27 h was observedin leaves elongating in continuous darkness. When plants weretransferred to 5 °C soon after emergence of the fourth leafthere was an immediate reduction in rate of growth to about22 per cent of the rate at 20 °C: the Q10 for the mean elongationrate in the range 20–5 °C was 3.7. When plants weretransferred from 20 to 2 °C at fourth leaf emergence, meanextension rate declined to less than 5 per cent, correspondingto a Q10 in the range 5–2 °C of more than 300. Furthermore,growth at 2 °C was confined almost entirely to the darkphase of the photoperiod cycle. The responsive tissue was shownto be a small area of expanding leafless than 1.5 cm above theshoot apex and the possible mechanisms underlying low temperatureeffects in this region are discussed. Accumulation of dry weight and leaf plus stem area were measuredin Echinochloa utilis and E. frumentacea grown at temperatureregimes from 15/10°C to 33/28°C (day/night). Tilleringand height were recorded in addition to leaf number which wassubsequently used as a developmental index. In both species shoot dry weight increased with temperatureup to 33/28°C; the increase in relative growth rate (RGR)was negligible above 27/22°C. Below 27/22°C the RGRof E. frumentacea decreased sharply and at 15/10°C it madeno effective growth. At low temperatures the RGR of E. frumentaceawas lower than that of E. utilis due to slow leaf area expansion,and in particular smaller individual leaves. E. frumentaceatillered more than E. utilis. Plant development was retardedat low temperatures but was not as responsive to temperatureas dry weight and leaf area. The different responses to temperatureof the two species were described in equations suitable forinclusion in predictive growth models. The rates of egg production of individually reared females ofCentropager typicus have been observed from the moulting C5-adultto death under constant conditions of food (10 000 cells ml–1of Hymenomonas elongara) and temperature (15 and 20°C).Preliminaiy experiments showed that virgin females could produceunfecunded eggs, but that the distinctive spines of the eggsof C.typicus were only obtained after fecundation occurred.High and continuous egg production needs several matings, andthe following experiments were run with a couple. In all experiments,both males and females fed and produced faecal pellets. Thefemales show a high variability in spawning life periods, inspawning rhythms, in daily rates and, consequently, in cumulatednumbers of eggs spawned during their life. The highest dailyrates of egg production were obtained in the first half of thefemale's life, and were as high at 20°C as at 15°C.Nevertheless, the females survived longer at 15°C than at20°C, and finally produced 47.6% more eggs during theirwhole life at 15°C than at 20°C, which can be partiallydue to the difference in size (almost 10% bigger at 15°Cthan at 20°C). The production of faecal pellets occurredduring the whole of life and was more regular than egg production,with a maximum daily rate in the first part of life, and showeda decrease until death. The decline in daily egg productionwith age was parallel to the decrease in daily faecal pelletproduction. The daily production of faecal pellets was higherat 15°C than at 20°C. At 20°C, a good correlationwas observed between the total egg production per female andthe total faecal pellet production per female. The individual-basedegg production rates of our experiments have been compared withrates obtained in different conditions by several authors, andwere lower than in other published studies, probably due tothe food offered. We emphasize the difficulties of comparingresults which have been obtained for the entire life of thefemales with those based on daily egg production. The totalegg production of a female is an important parameter in regardto the population d because it fixes the maximal rate of losses(mortality plus dispersion) for which a population can continueto develop.  相似文献

Growth (assessed from intermolt period and molt increment) andmetabolism (oxygen consumption) of the post-larva of Euphausiapacifica from the southern Japan Sea were determined at sevengraded temperatures ranging from 1 to 25°C. The intermoltperiod shortened progressively as temperature increased from1 to 20°C, but an effect of temperature on molt-to-moltgrowth increment was not seen. Oxygen consumption rates wereaccelerated by the increase in temperature up to 20°C. Beyond20°C, E.pacifica exhibited reduced oxygen consumption anddied within 1 day without molting. After removing the effectof body size, the relationships between growth rate and temperature,and between oxygen consumption rate and temperature, were established.The carbon budget was calculated as a function of temperature.Because of differential effects of temperature on growth andmetabolism, the net growth efficiency [K2 growthx100/(growth+metabolism)]changed with temperature. The optimum temperature at which E.pacificaattained the maximum K2 was 11.4°C, which was derived fromcalculation of cumulative carbon invested in growth and metabolismin this animal. In an alternative method, the optimum temperaturewas obtained mathematically by solving a set of differentialequations. The biological and ecological significance of theoptimum temperature which leads to the maximum K2 is discussed.  相似文献

The effect of low temperatures on the fatty acid compositionof phosphatidylglycerol (PG) in thylakoid membranes, in particularon the ratios of nmol% 16:1(3t) (mg fresh weight)–1 ofcotyledons and nmol 16:1(3t) (mg chlo rophyll)–1 weremeasured during squash seedling growth. Plants were germinatedand grown for one day at 30°C then were either kept at 30°C(control plants) or trans ferred to low temperatures (18, 14or 10°C). When plant were transferred from 30°C to lowtemperatures, the increase in fresh weight was gradually limited.The lowe the temperature, the smaller was the fresh weight.In contrast, the relative content of 16:1(3t) and 18:3, as wella the ratios of nmol 16:1(3t) (mg chlorophyll)–1 and mol%16:1(3t) (mg cotyledon fresh weight)–1 increased indicatingthat the increase of fresh weight and chlorophyll was mor sensitiveto low temperature than PG desaturation in thyla-koid membranes.Furthermore, low temperatures inducei an increase in 16:1(3t)and 18:3 (the final products of PC synthesis) at the expenseof 16:0 and 18:1 (the initial products of PG synthesis). However,within a range of temperature from 10 to 18°C, the extentof these changes (nmol% of 18:3 or 16:1(3t) per day) was graduallylimited by lower temperatures. We therefore propose that lowtemperature inhibit both fatty acid synthesis and desaturationactivities. However, at low temperatures the fatty acid synthesisis likely to be more strongly inhibited than the desaturationactivities, thus explaining the observed increase in the relativecontent of PG-18:3 and PG-16:l(3t). Sebago) increasedgibberellin activity in crude extracts from buds, but not frommature leaves, as determined by the lettuce hypocotyl bioassay.Changes in the growth of tubers and stolons indicate the occurrenceof basipetal movement of GA3 applied to the terminal bud ora mature leaf. 14C labelling from GA3 or mevalonic acid injectedjust below the terminal bud was recovered in the lower shoot,stolons and tubers, but the amount transported was greater atcool temperatures (20/15°C). Stands of groundnut were grown in controlled environment glasshousesat air temperatures of 19, 22, 25, 28, and 31°C. Leaf areaindex (L) increased with rise of temperature, and after 85 dwas about 10-fold larger at 31°C than 19°C. Over mostof the range of temperature, both L and fractional interceptionof solar radiation (f) were functions of thermal time accumulatedfrom sowing (above a base of 10°C). In this respect, theywere tightly coupled to developmental rate at the main apex.In one experiment, only 38% of seeds emerged at 22°C and21% at 19°C, compared with more than 70% at 25°C and31°C, but the low population density was compensated byfaster leaf expansion by each plant (at 22°C only) and agreater fraction of solar radiation intercepted by unit leafarea. The amount of solar radiation intercepted by stands increasedwith rise in temperature, but the greatest differences betweentreatments occurred before the canopies achieved complete groundcover (i.e.f>0.9) and the relative effect of a rise in temperaturediminished the longer the duration of growth. The dry matterproduced for unit solar radiation intercepted was not stronglyaffected by temperature between 22°C and 31°C, wherethe mean was 2.1 g MJ–1; the value at 19°C was uncertainsince the stands were sparse throughout the experiment. After85 d, the stand at 31°C had produced eight times the drymatter of that at 19°C—a difference caused mainlyby the effect of temperature on the rates of development andexpansion. Triticum aestivumxZea mayscrosses are now widely used in theproduction of wheat doubled haploids to produce homozygous lines.Seasonal effects are known to influence the number of haploidembryos produced through wheatxmaize crosses, but the effectsof temperature and light have not been quantified. This studyinvestigated the effect of temperature and light intensity onhaploid embryo production. New Zealand wheat cultivars weregrown in a glasshouse until booting when they were transferredto growth cabinets at three temperatures (day/night; 17/12,22/17 or 27/22 °C at an irradiance of 250 µmol m-2s-1PAR).In another experiment, wheat lines were transferred to a growthcabinet at one of three light intensities (300, 500 or 1000µmol m-2s-1PAR at 22/17 °C day/night, with a photoperiodof 16 h). The temperature and light intensity at which pollinationswere made and subsequent fertilisation and embryo developmentoccurred, significantly (P<0.01) influenced the frequencyof haploid embryo production. The optimal temperature for embryorecovery was 22/17 °C. The greatest number of embryos wasproduced at a light intensity of 1000 µmol m-2s-1. The onset and rate of infection in root hairs of T. parviflorumand T. glomeratum inoculated with Rhizobium trifolli strain5 varied much with root temperature. At moderate root temperature(18, 24, and 30 °C) infections were initiated earlier andin larger numbers than at low (6 and 12 °C) or moderatelyhigh (36 °C) temperatures. Both species showed a broad temperatureoptimum between 18 and 30 °C. The site of thread initiation(apically or laterally in a hair) was independent of temperature,as was also the proportion of successful threads penetratingthe root cortex, which increased with seedling age. Threadsgrew more slowly at low temperatures. The size of hair nucleinear infection threads remained unaffected by temperature, butnuclei associated with laterally originating threads were largerthan those associated with apical threads. Infection was non-randomly distributed along the main root atall temperatures. More zones of infection were found at moderatetemperatures than at temperature extremes (6–12 or 36°C). Probit plots of numbers of infections for individualplants were steplike, the linear sloping parts correspondingto normal distributions of infection within zones. Between 18 and 30 °C numbers of infections increased exponentiallyin two phases, the first more rapid phase ending at about thetime nodules appeared. A model devised for the infection processand fitted to the data suggested the existence of two kindsof infections: primary ones occuring randomly at a slow rateand probably not affected by temperature and secondary infectionsthat appeared to increase with rising temperatures in the range12 to 30 °C. Nodule numbers were relatively more sensitive to high and lowtemperatures than infection. The numbers of infections and nodulesand the root lengths of T. parviflorum were twice those of T.glomeratum except at the temperature extremes. Numbers of infectionswere otherwise unrelated to root length or cotyledon or leafareas. The development of lateral organs (primordia, lateralroots, and nodules) was reduced at temperatures below 18 °Cand above 30 °C.  相似文献

The regulation of ethylene synthesis in pear fruits was investigated.During storage for 60 d at — 1 °C the rate of ethylenesynthesis increased 100-fold but the concentration of 1-aminocyclopropane-l-carboxylicacid (ACC) increased only 2-fold and ACC synthase activity waslow. On transfer to 15 °C after storage at — 1 °Cethylene synthesis increased 10-fold within 10 h but ACC synthaseactivity only increased rapidly after 24 h; the decline in ACClevels during the first 16 h at 15 °C was insufficient tosustain ethylene synthesis. Ethylene synthesis was further investigatedusing discs cut from the mid cortex of pear fruits. Synthesiswas inhibited by aminoethoxyvinylglycine (AVG) and amino-oxyaceticacid at all stages of ripening. The rate of synthesis and ACCsynthase activity increased rapidly after slicing of pears heldat — 1 °C but more slowly in discs cut from pearsimmediately after harvest. Cycloheximide (CHI) inhibited theseincreases and reversed increases resulting from pre-incubationof discs. A combination of CHI and AVG abolished the capacityof discs to synthesize ACC and ethylene production was curtailed.Cordycepin and actinomycin-D were less effective as inhibitorsof the development of ethylene synthesis and ACC synthase activitythan as inhibitors of incorporation of 5-[3H] uridine into totalRNA or poly A rich RNA. The ability of discs to develop ethylenesynthesis and ACC synthase activity in the presence and absenceof cordycepin increased concurrently during storage of wholefruits at — 1 °C. The growth of three populations of greater lotus (Lotus uliginosusSchkuhrsyn.L. pedunculatusCav.) was compared at photoperiods of 10,12 and 14 h at a maximum day/minimum night temperature of 21/16°C and at maximum day/minimum night temperatures of 27/22,21/16, 18/13 and 15/10 °C at a photoperiod of 12 h. Shortdays (10 h) favoured root and rhizome development compared tolong days (14 h). A temperature regime of 15/10 °C restrictedrhizome development compared to the 18/13 and 21/16 °C regimes.Shoot growth was restricted at the highest temperature regime(27/22 °C). The cultivar Sharnae had fewer, but heavier,rhizomes than Grasslands Maku; this may indicate adaptationto the dry summers at its site of origin (Algarve, Portugal).The response of rhizome growth to temperature and photoperiodexplains part of the performance of greater lotus in the fieldat a wide range of latitudes. Grazing management to encouragethe persistence ofL. uliginosusin pasture in temperate environmentsmay include the exclusion of grazing livestock in autumn. Seasonal changes in the photosynthesis and primary productionof Microcystis aeruginosa Kütz. were investigated in LakeKasumigaura during 1981–1982. Microcystis always showeda light-saturated photosynthesis-light curve. Both Pmax andthe initial slope of the photosynthesis-light curve of Microcystisin early summer were very high, so it was concluded that Microcystisutilized both low and high light intensities efficiently. ThePmax of Microcystis was found to be a function of the watertemperature except in August and September. The linear regressionon the temperature-Pmax relationship discontinued at 11°C,where the Pmax value dropped; Microcystis did not photosynthesizebelow 4°C. The initial slope of the curve was also descendingbelow 11°C. It is suggested that Microcystis changes itsphysiological properties below 11°C. The highest value ofgross production calculated for M. aeruginosa was 5.4 gC m–2d–1 in July; the annual gross production was estimatedto be 300 gC m–2year–1 (i.e., 40% of the total primaryproduction in this lake).  相似文献

Two limpet species occur intertidally on subantarctic SouthGeorgia, the patellid Nacella concinna and the siphonarlid Kerguelenellalateralis. N. concinna is confined to the lower shore closeto LWS; K. lateralis occurs in middle shore pools, so theirdistributions do not overlap. N. concinna has a much narrowerthermal niche (–12.9°C to +15.6°C) than K. lateralis(–17.8°C to +31.8°C). Environmental data are presentedto show that the upper lethal temperature of N. concinna islow enough to prevent the limpet living higher on the shore.Both limpet species are slow-moving, but K. lateralis showsincreasing speed with rising temperature, peaking at 15–20°C.In contrast, N. concinna moves actively down to –1.9°C(when sea water freezes), but there is a steady decrease inspeed of locomotion above +2°C. Locomotion ceases at 14°Cin N. concinna (c.f. 30°C in K. lateralis). Both speciesexhibit very low tenacities, but in N. concinna tenacity decreaseswith increasing shell length. In K. lateralis there is no effectof temperature on tenacity. Both species show a positive allometricrelationship between foot area and shell length. N. concinnafeeds upon microbial films and microepiflora, but K. lateraliseats colonial diatoms and Enteromorpha bulbosa. Observationson shell middens of the kelp gull Larus dominicanus showed thatthe gulls did not eat K. lateralis, though they ate great quantitiesof the less accessible N. concinna. Gulls ate N. concinna assmall as 11 mm shell length (within the size range of K. lateralis).Experiments on gulls demonstrated an unwillingness to eat K.lateralis, probably because the siphonariid extrudes a viscidwhite mucus when the foot is touched. The hydrography, chlorophyll (Chl) a and egg production of thecopepods Paracalanus parvus (Claus), Acartia lilljeborgii Giesbrecht,Acartia clausi Giesbrecht and Centropages furcatus were estimateddaily between 7 February and 5 March 1998 in Bahía Magdalena,Baja California Sur, México. Temperature was homogeneousthroughout the water column during the study (20°C). Positiveanomalies of the sea surface temperature were recorded 10 monthsbefore and during the sampling period compared with a temperature–timeseries, 1982–1989. Chlorophyll a concentration indicatedoligotrophic conditions with <10 Chl a mg m–2 from15 m depth to the surface during the first half of the study,with a pulse of moderate concentration in the second part. Theegg production of these copepod species was usually suboptimal,and not correlated with Chl concentration or temperature. Eachgenus responded differently to Chl a and to environmental variables.The rate of input of turbulent kinetic energy to the ocean bythe winds, indicated by the cube of the wind speed, was negativelycorrelated to copepod egg production, suggesting that turbulencecan disperse phytoplankton patches and may affect the carboninput to these copepod populations. Turbulence and a previouslong warming event observed several months before the winterseason were probably the most important factors in limitingcopepod production and growth rates.  相似文献
